In recent years, mitochondria have emerged as important targets of agonist-dependent increases in cytosolic Ca2+ concentration. Here, we analyzed the significance of Ca2+ signals for the modulation of organelle function by directly measuring mitochondrial and cytosolic ATP levels ([ATP]m and [ATP]c, respectively) with specifically targeted chimeras of the ATP-dependent photoprotein luciferase. In both HeLa cells and primary cultures of skeletal myotubes, stimulation with agonists evoking cytosolic and mitochondrial Ca2+ signals caused increases in [ATP]m and [ATP]c that depended on two parameters: (i) the amplitude of the Ca2+ rise in the mitochondrial matrix, and (ii) the availability of mitochondrial substrates. Moreover, the Ca2+ elevation induced a long-lasting priming that persisted long after agonist washout and caused a major increase in [ATP]m upon addition of oxidative substrates. These results demonstrate a direct role of mitochondrial Ca2+ in driving ATP production and unravel a form of cellular memory that allows a prolonged metabolic activation in stimulated cells.

Vaccination with synthetic peptides representing cytotoxic T lymphocyte (CTL) epitopes can lead to a protective CTL-mediated immunity against tumors or viruses. We now report that vaccination with a CTL epitope derived from the human adenovirus type 5 E1A-region (Ad5E1A234-243), which can serve as a target for tumor-eradicating CTL, enhances rather than inhibits the growth of Ad5E1A-expressing tumors. This adverse effect of peptide vaccination was rapidly evoked, required low doses of peptide (10 micrograms), and was achieved by a mode of peptide delivery that induces protective T-cell-mediated immunity in other models. Ad5E1A-specific CTL activity could no longer be isolated from mice after injection of Ad5E1A-peptide, indicating that tolerization of Ad5E1A-specific CTL activity causes the enhanced tumor outgrowth. In contrast to peptide vaccination, immunization with adenovirus, expressing Ad5E1A, induced Ad5E1A-specific immunity and prevented the outgrowth of Ad5E1A-expressing tumors. These results show that immunization with synthetic peptides can lead to the elimination of anti-tumor CTL responses. These findings are important for the design of safe peptide-based vaccines against tumors, allogeneic organ transplants, and T-cell-mediated autoimmune diseases.

Objective To assess the level of compliance with the new law in the United Kingdom mandating penalties for rising a hand held mobile phone while driving, to compare compliance with this law with the one on the use of seat belts, and to compare compliance with these laws between drivers of four wheel drive vehicles and drivers of normal cars. Design Observational study with two phases-one within the grace period, the other starting one week after penalties were imposed on drivers using such telephones. Setting Three busy sites in London. Participants Drivers of 38 182 normal cars and 2944 four wheel drive vehicles. Main outcome measures Proportions of drivers seen to be using hand held mobile phones and not using seat belts. Results Drivers of four wheel drive vehicles were more likely than drivers of other cars to be seen using hand held mobile phones (8.2% v 2.0%) and not complying with the law on seat belts (19.5% v 15.0%). Levels of non-compliance with both laws were slightly higher in the penalty phase of observation, and breaking one law was associated with increased likelihood of breaking the other. Conclusions The level of non-compliance with the law on the use of hand held mobile phones by drivers in London is high, as is non-compliance with the law on seat belts. Drivers of four wheel drive vehicles were four times more likely than drivers of other cars to be seen using hand held mobile phones and slightly more likely not to comply with the law on seat belts.

Esta dissertação tem como objetivo verificar, de acordo com o conceito de marketing político, como se deu a construção da imagem pública do ex-presidente Fernando Henrique Cardoso na revista Veja durante as eleições presidenciais de 1994. Para tanto, analisou-se 19 edições desse periódico, todas relativas ao período de junho a outubro do referido ano. Estudou-se a propaganda ideológica e suas categorias, como codificação, controle ideológico, contrapropaganda e difusão, presentes no material empírico analisado. Este trabalho desenvolveu-se à luz dos procedimentos metodológicos referentes ao Estudo de Caso enquanto tipo ou estratégia de pesquisa. Entre as técnicas para a coleta de dados, realizou-se a Análise de Conteúdo de natureza quantitativa e qualitativa dos dados pesquisados e entrevista com o próprio ex-presidente. Por meio deste estudo obteve-se os seguintes resultados: Fernando Henrique Cardoso teve o maior volume em centímetro/coluna em relação ao seu opositor, o candidato Luiz Inácio Lula da Silva. Em relação aos códigos utilizados nas matérias analisadas, o volume maior em centímetro/coluna ocorreu no código linguístico, significando que a revista Veja deu mais ênfase às palavras, às frases e aos parágrafos que compõem as estruturas articuladas, segundo os padrões históricos e culturais da língua portuguesa. No que diz respeito aos gêneros informativos, o volume maior em centímetro/coluna foi em reportagem, isso significa que a revista enfatizou os fatos que repercutiram e produziram efeitos na sociedade, e que foram percebidos pela revista. Concluise que as matérias publicadas fortaleceram significativamente a sua imagem perante os leitores da Veja, favorecendo-o em relação ao seu opositor, mas não chegaram a elegê-lo.(AU)
Aplicación de la medicina ancestral en los usuarios del Centro de Salud Carlos Elizalde. Cuenca 2015

ANTECEDENTES: En gran parte del mundo la medicina tradicional es el pilar principal de la prestación de servicios de salud, por ello en algunos países suele denominarse medicina complementaria. La medicina tradicional ha sufrido transformaciones, y los valores de este tipo de medicina están desapareciendo con rapidez a pesar que la OMS dicta estrategias para su implementación y el MSP del Ecuador reconoce su valor. OBJETIVO GENERAL: Caracterización de la aplicación de la medicina ancestral en los usuarios y usuarias del Centro de Salud “Carlos Elizalde”. METODOLOGIA: Es un estudio cuantitativo descriptivo en el que caracteriza la aplicación de la medicina ancestral en los usuarios del Centro de Salud “Carlos Elizalde”. El método utilizado fue la entrevista previo consentimiento informado realizada, según criterios de inclusión y exclusión a un universo de 110 usuarios, en la tabulación de datos se utilizó el programa SPSS Windows y Excel, y estos fueron analizados en tablas simples y gráficos. RESULTADOS: Los resultados obtenidos han servido para identificar la aplicación la Medicina Ancestral en los usuarios que acuden al Centro de Salud “Carlos Elizalde” de la Parroquia Yanuncay, identificando a las personas que más acuden al tratamiento son adultos con 71% y adultos mayores con un 16%, la mayoría son amas de casa en un 38%, con instrucción de primaria incompleta, es las enfermedades más frecuentes son espanto con un 36%, mal aire 18% y 10% caída de zhungo; para el tratamiento utilizan plantas o hiervas en un 54%

De tradição oral e de origem remota, os provérbios circunscrevem a realidade cultural dos seus criadores e utilizadores. A sua riqueza reside, por um lado, na mensagem sintética e por isso, também, pragmática, ao ser utilizada como âncora em contextos específicos; reside, por outro lado, na construção de metáforas conceptuais, tradutoras de verdades e generalizações sedimentadas na observação empírica. É nestas que a língua portuguesa (LP) germina a dimensão cultural e permite a exploração de mundividências partilhadas, transparentes, translúcidas ou opacas. O trabalho que propomos apresentar descreve um conjunto de atividades realizadas com alunos do 5.o ao 8.o ano de escolaridade, no âmbito de um programa desenvolvido por um agrupamento de escolas da região de Viseu e uma instituição de ensino superior durante o ano letivo de 2013-2014. As atividades que propusemos implicaram considerar a LP a partir de diferentes prismas, perspetivando-a não só como código verbal, como herança cultural, mas também como espaço de criação e criatividade. Assim, a sedimentação de significados que foram construindo a LP é apresentada aos alunos como enigma a desvelar. Neste sentido, propomos a) analisar a representação gráfica de provérbios feita por alunos do ensino básico; b) refletir sobre a influência da LP na formação de provérbios em países de expressão portuguesa. A sua interpretação é padronizada, mas permeável a ambiguidades. Neste caso, e a partir da comparação de um exercício de equivalência entre provérbios representados pela variedade do português, revelamos a seleção dos alunos.

Most people get infected with West Nile virus by the bite of an infected mosquito. Mosquitoes become infected when they feed on infected birds. Infected mosquitoes can then spread the virus to humans and other animals. In a very small number of cases, West Nile virus has been spread through blood transfusions, organ transplants, and from mother to baby during pregnancy, delivery, or breastfeeding. Recovery from severe disease may take several weeks or months. Some of the neurologic effects may be permanent. About 10 percent of people who develop neurologic infection due to West Nile virus will die.

O presente estudo está baseado na Metodologia fenomenológica, com pesquisa bibliográfica e de campo. Apresenta como objetivo geral, analisar como se desenvolve o processo de ensino e aprendizagem de leitura, assim como, os estímulos e a prática do ato de ler em alunos de tenra idade em escolas particulares de Belém do Pará, em turmas da 4a Serie, do Ensino Fundamental. Tendo como objetivos específicos averiguar o ponto principal de indagação está centrado na questão se a leitura prática é apenas o desenvolvimento dos códigos linguísticos aprendidos, ou buscar não somente o processo de decodificar, sem considerar os aspectos mais abrangentes como a interpretação do texto, direcionado para o leitor ativo e atuante na compreensão das ideias do autor. Tal questionamento a respeito do processo de aprendizagem da leitura em nível fundamental das escolas particulares de Belém do Pará foi perguntado por meio de pesquisas de campo, a professores, alunos e para a coordenação, utilizando como instrumento de coleta de dados, questionrio com perguntas abertas e fechadas de opinião entrevistas e grupos focais. No decorrer deste estudo, nota-se que para formar leitores mirins, serão exigidos alguns requisitos importantes, como a escolha de bons livros e atividades norteadoras de estimulação em classe e até mesmo fora dela. Afim de que principalmente o aluno em fase inicial de aprendizagem, seja e sinta-se preparado e ainda, conscientizado da importância da leitura para o seu questionamento crítico, como cidadão do futuro. Para tanto, fatores que interferem no processo de leitura, tais como: títulos variados, histórias que versem a respeito do imaginário cultural, textos direcionados ao público infantil, que despertem o interesse do alunado, além de apenas lei os códigos linguísticos na aquisição do texto, sem obtenção dos critérios interpretativos, que este possa evocar. A leitura é tida pela sociedade em geral, como um fator que pode levar o individuo a descobertas de novos patamares e horizontes sociais, mas somente indo além do mecanismo na apresentação dos textos cm sala de aula, é que o professor poderá atingir os níveis de compreensão integral do sistema escrito. Ensinando a ler, com elementos fundamentais para a iniciação de leitura interpretativa e proficiente no ensino fundamental. ABSTRACT: The present study is based upon the Phenomenological Methodology, with field and bibliographic research. It presents as a main objective the analysis on how the teaching and learning educational process develop upon the reading skills, as well as the encouragement and practice of reading in students of young ages in private schools in Belém, state of Pará (Brazil), in fourth grade groups, in elementary school. Having set specific goals, the search is centered in the practical reading ability matter, and if such ability is of linguistic code development solely learned or the search of not only the decoding process, without taking in consideration the broadening aspects, such as text comprehension, targeting the active reader especially when it comes to the author's ideas. Such discussion regarding the reading learning process in elementary levels in private schools in Belém, state of Para (Brazil) is questioned by field research, teachers, students and the coordination, using as data collecting instruments, questionnaires with open and closed questions, interviews and focal groups. Along the study, regarding the formation of young readers, it requires some aspects, such as, the choice of good titles and in-class stimulating activities or even outdoor ones, so the students - especially the ones in the initial phase of learning- are and feel prepared and even conscious of the importance of the reading for their critical thinking- as a future citizen. Therefore, the factors that may interfere in the reading process, for instance, varied titles, stories that verse towards the cultural imaginary, texts directed to the young public, that awaken the interest of the student body, beyond the linguistic codes acquired in the texts, not obtaining the interpreting criteria that it might evoke. The reading is, by the society, a fundamental factor that might lead the human being to discovering new standards and social horizons, but only going further the typical mechanisms in presenting the texts in class, is when the teacher may reach the whole levels of comprehension of the written system, teaching how to read, with fundamental elements to the initial reading in elementary school.

To find the most reliable screening method for Trypanosoma cruzi infection in blood banks. Epidemiological data, lymphoproliferation assay, parasitological, conventional serological tests: immunofluorescence, haemagglutination, ELISA with epimastigote and trypomastigote antigens and reference serological tests: trypomastigote excreted-secreted antigens (TESA) blot and chemiluminescent ELISA assay with mucine from trypomastigote forms were applied to individuals with inconclusive serology, non-chagasic individuals and chronic chagasic patients. TESA blot had the best performance when used as a single test in all the groups. In the inconclusive group 20.5% of individuals were positive for TESA blot, 23.3% for either lymphoproliferation or TESA blot, and 17.8% for lymphoproliferation only. Positive lymphoproliferation without detectable antibodies was observed in 5.47% of all inconclusive serology cases. Analysis of six parameters (three serological assays, at least one parasitological test, one lymphoproliferation assay and epidemiological data) in the inconclusive group showed that diagnosis of Chagas` disease was probable in 15 patients who were positive by two or more serological tests or for whom three of those six parameters were positive. TESA blot is a good confirmatory test for Chagas` disease in the inconclusive group. Although lymphoproliferation suggests the diagnosis of Chagas` disease in the absence of antibodies when associated with a high epidemiological risk of acquiring Chagas` disease, the data from this study and the characteristics of the lymphoproliferation assay (which is both laborious and time-consuming) do not support its use as a confirmatory test in blood-bank screening. However, our findings underscore the need to develop alternative methods that are not based on antibody detection to improve the diagnosis when serological tests are inconclusive.

Introduction: The aim was to investigate retrospectively use of Complementary and Alternative Medicine (CAM) in the treatment of patients with organ transplantation in Switzerland. Methods: Members of the Swiss transplant association completed a questionnaire about CAM use retrospectively. Five different stages were differentiated: CAM usage (1) during underlying disease, (2) before transplant, (3) during hospitalisation/ rehabilitation from transplant, (4) for transplant complications and (5) after transplant for other diseases. Results: Of the 267 patients contacted, 124 (46%) completed the questionnaire, and data of 118 (44%) participants could be analyzed: 55 women (47%), mean age 56 years. Overall, 64 (54%) indicated CAM use, with about 30% usage at every stage (except during hospitalization with only 10%). Different methods were most common: during underlying disease classical homeopathy (15% of all participants), before transplant dietary supplements (13%), during hospitalization meditation (3%), for transplantation complications dietary supplements (10%), and after transplant for other diseases massage (11%). Among the 64 CAM-users, the most important reasons for the usage were improvement of general condition (36%) and abatement of adverse effects of conventional treatment (25%). Among the 54 non-CAM-users, most frequent reasons for not choosing CAM were insecurity about interactions with conventional treatment (46%), and ignorance of this option (28%). About 35% of the CAM-users reported an improved general condition, while 30% noticed an abatement of side effects of conventional treatment. Conclusions: To prevent dangerous interactions with conventional treatment, more information on possibilities of CAM in the treatment of patients with transplantations is needed for doctors and patients.

OBJECTIVE To evaluate the frequency and clinical features of endemic and other opportunistic infections in liver or kidney transplant recipients in four transplant centres in different geographical areas of Brazil. METHODS Retrospective analysis of medical and laboratory records of four transplant centres on endemic and other opportunistic infections in liver or kidney transplant recipients. Analyses were performed with spss statistical software. RESULTS From 2001 to 2006, 1046 kidney and 708 liver transplants were registered in all centres. The average age was 42 years. Among 82 (4.7%) cases with infections, the most frequent was tuberculosis (2.0%), followed by systemic protozoal infections (0.7%), toxoplasmosis (0.4%) and visceral leishmaniasis (0.3%). Systemic fungal infections occurred in 0.6%, of which 0.4% were cryptococcosis and 0.2% were histoplasmosis. Dengue was the only systemic viral infection and was registered in two cases (0.1%), of which one was classified as the classic form and the other as dengue haemorrhagic fever. Nocardiosis was described in one case (0.05%). The infectious agents most frequently associated with diarrhoea were Blastocystis sp., Schistosoma mansoni and Strongyloides stercoralis. CONCLUSIONS Opportunistic Infections in transplant patients have a wide spectrum and may vary from asymptomatic to severe infections with high mortality. A better understanding of the epidemiology of endemic pathogens and clinical manifestations can contribute to the establishment of an early diagnosis as well as correct treatment aimed at decreasing morbidity and mortality.
Connecting free volume with shape memory properties in noncytotoxic gamma-irradiated polycyclooctene

The free volume holes of a shape memory polymer have been analysed considering that the empty space between molecules is necessary for the molecular motion, and the shape memory response is based on polymer segments acting as molecular switches through variable flexibility with temperature or other stimuli. Therefore, thermomechanical analysis (TMA) and positron annihilation lifetime spectroscopy (PALS) have been applied to analyse shape recovery and free volume hole sizes in gamma irradiated polycyclooctene (PCO) samples, as a non-cytotoxic alternative to more conventional PCO crosslinked via peroxide for future applications in medicine. Thus, a first approach relating structure, free volume holes and shape memory properties in gamma irradiated PCO is presented. The results suggest that free volume holes caused by gamma irradiation in PCO samples facilitate the recovery process by improving movement of polymer chains and open t possibilities for the design and control of the macroscopic response.

PURPOSE: Postmortem computed tomography angiography (PMCTA) was introduced into forensic investigations a few years ago. It provides reliable images that can be consulted at any time. Conventional autopsy remains the reference standard for defining the cause of death, but provides only limited possibility of a second examination. This study compares these two procedures and discusses findings that can be detected exclusively using each method. MATERIALS AND METHODS: This retrospective study compared radiological reports from PMCTA to reports from conventional autopsy for 50 forensic autopsy cases. Reported findings from autopsy and PMCTA were extracted and compared to each other. PMCTA was performed using a modified heart-lung machine and the oily contrast agent Angiofil® (Fumedica AG, Muri, Switzerland). RESULTS: PMCTA and conventional autopsy would have drawn similar conclusions regarding causes of death. Nearly 60 % of all findings were visualized with both techniques. PMCTA demonstrates a higher sensitivity for identifying skeletal and vascular lesions. However, vascular occlusions due to postmortem blood clots could be falsely assumed to be vascular lesions. In contrast, conventional autopsy does not detect all bone fractures or the exact source of bleeding. Conventional autopsy provides important information about organ morphology and remains the only way to diagnose a vital vascular occlusion with certitude. CONCLUSION: Overall, PMCTA and conventional autopsy provide comparable findings. However, each technique presents advantages and disadvantages for detecting specific findings. To correctly interpret findings and clearly define the indications for PMCTA, these differences must be understood.

The increasing number of pertussis cases reported on the last twenty years and the existence of new acellular vaccines reinforce the need of research for experimental models to assure the quality of available pertussis vaccines. In this study, allotments of whole-cell and acellular pertussis vaccines were tested through the Intranasal Challenge Model (INM) using conventional NIH mice. The results have been compared to those achieved by the "Gold standard" Intracerebral Challenge Model (ICM). In contrast to ICM, INM results did not show intralaboratorial variations. Statistical analysis by Anova and Ancova tests revealed that the INM presented reproducibility and allowed identification and separation of different products, including three-component and four-component accellular pertussis vaccines. INM revealed differences between pertussis vaccines. INM provides lower distress to the mice allowing the reduction of mice number including the possibility of using conventional mice (less expensive) under non-aseptic environment. Thus, INM may be used as an alternative method of verifying the consistence of allotment production, including acellular pertussis vaccines.
