Coffee is a product consumed all around the world, Brazil being the biggest exporter. However, little is known about the difference in composition of the different brands in terms of bioactive substances. In the present study, ten of the most consumed brands of coffee in Rio de Janeiro were analyzed. Caffeine contents, trigonelline and total chlorogenic acid varied from 0.8 g/100g to 1.4 g/100g; 0.2 g/100g to 0.5 g/100g and from 3.5 g kg-1 to 15.9 g kg-1, respectively. The large heterogeneity observed in the amounts of the bioactive compounds can be attributed to different formulations of the various brands, as well as to different roasting conditions.

Coffee is one of the beverages most widely consumed in the world and the "cafezinho" is normally prepared from a blend of roasted powder of two species, Coffea arabica and Coffea canephora. Each one exhibits differences in their taste and in the chemical composition, especially in the caffeine percentage. There are several procedures proposed in the literature for caffeine determination in different samples like soft drinks, coffee, medicines, etc but most of them need a sample workup which involves at least one step of purification. This work describes the quantitative analysis of caffeine using ¹H NMR and the identification of the major components in commercial coffee samples using 1D and 2D NMR techniques without any sample pre-treatment.

Sunflower oil is one of the vegetable oils most susceptible to oxidative reaction. Therefore the presence of natural (tocopherol) and/or synthetic (TBHQ) antioxidants is a prerequisite to secure the stability of this oil during its shelf life, especially with the increased use of PET packaging. On determining the total tocopherol content in 10 samples of deodorized sunflower oils, high losses in total tocopherol during deodorization were identified. In order to facilitate researche involving oxidative stability of vegetable oils, a method for isolated quantification of TBHQ using the isocratic elution system was developed.

This paper reports the use of Raman and infrared techniques for the qualitative and quantitative analysis of plasticizers in polyvinylchloride (PVC) commercial films. FT-Raman marker bands were indentified for di-2-ethyl-hexyl adipate (DEHA) and di-2-ethyl-hexyl phthalate (DEHP), allowing for the rapid identification of these species in the commercial film. Quantitative analysis by FT-IR resulted in plasticizers concentrations ranging from 11 to 27% (w/w). Considering the little sample preparation and the low cost of the techniques, FT-IR and FT-Raman are viable techniques for a first assessment of plasticizers in commercial samples.

Isoamyl butyrate production was investigated using free and immobilized lipases by esterification of butyric acid with isoamyl alcohol in a solvent-free system and in an organic media. Among the enzymes studied, Lipozyme TL IM was found to be the most active catalyst in n-hexane as a solvent. The effects of different solvents and the amount of water added on conversion rates were studied. A maximum conversion yield of 80% in n-hexano at 48 h was obtained under the following conditions: 3 g L-1 of Lipozyme TL IM, 30 ºC, 180 rpm of agitation, isoamyl alcohol to butyric acid molar ratio of 1:1 and acid substrate concentration of 0.06 M.

The amounts of nicotinic acid, trigonelline, 5-CQA, caffeine, kahweol and cafestol in 38 commercial roasted coffees ranged from 0.02 to 0.04; 0.22 to 0.96; 0.14 to 1.20; 1.00 to 2.02; 0.10 to 0.80 and 0.25 to 0.55 g/100 g, respectively. Evaluation of color and content of thermo-labile compounds indicated similarity in roasting degree. Differences in the levels of diterpenes and caffeine, components less influenced by the roasting degree, could be mainly explained by the species used (arabica and robusta). Gourmet coffees showed high concentrations of diterpenes, trigonelline and 5-CQA and low levels of caffeine, indicating high proportion of arabica coffee.

The Neem tree, Azadirachta indica, provides many useful compounds that are used as pesticides. However, the efficiency in field of products like neem oil can be committed because they have not been observed reproductive content of secondary metabolic like azadirachtin. Based on reverse-phase high-performance liquid chromatography (HPLC) a new method was developed to permit the rapid quantitative analysis of azadirachtin from seeds, extracts and oil of Neem. In the present study it was evaluated the azadirachtin quantitative variation among various Neem's extracts and seeds showing the importance of quality control for reproduction of the insecticide efficiency, using S. frugiperda as target insect.

Ophthalmic viscosurgical devices (OVD) are materials injected in intraocular space during cataract removal to reduce trauma in the patient's eye. Three Brazilian commercially available OVDs (Medilon®, Metilcelulose® and Ofthyal®) were evaluated as well as formulations based on Dimorphandra gardneriana galactomannan. Viscosity and viscoelastic parameters, such as viscosity at zero shear, pseudoplasticity index, elastic and viscous moduli, relaxation time, were determined and compared. Characteristics of an effective OVD were proposed. None of the Brazilian devices studied fulfill the rheological requirements. Only the galactomannan at 3% concentration showed potential to be used as effective OVD.

A flow injection spectrophotometric procedure for the determination of glyphosate in commercial formulations of herbicides is proposed. The determination is based on the reaction of glyphosate and p-dimethylaminocinnamaldehyde, in acid medium, yielding a colored compound (l máx = 495 nm). Under optimal conditions, Beer's law is obeyed in a concentration range 40-640 mg mL-1 with a correlation coefficient of 0.9996. The detection limit was 8.60 mg mL-1 for glyphosate. The method was successfully applied for the determination of glyphosate in commercial formulations of herbicides. Recovery of glyphosate from various commercial samples of herbicides range from 91.0 to 110%.

The aim of this paper is to describe a chemometrics experiment for undergraduate chemistry students using basic concepts of experimental design to determine setting-time of commercial gelatins.

Commercial Brazilian regular and decaffeinated instant coffees (33 brands) were studied. The levels ranged from 0.47 to 2.15 g 100 g-1 for trigonelline, 0.38 to 2.66 g 100 g-1 for 5-caffeoylquinic acid (5-CQA), 0.24 to 4.08 g 100 g-1 for caffeine, and 0.253 to 0.476 (420 nm) for melanoidins. Variations in bioactive compound levels among batches were observed. There was no relationship between the drying process and the composition of the products. In general, Gourmet and decaffeinated coffees had higher trigonelline and 5-CQA but lower caffeine and melanoidin content than regular products.

The aim of this manuscript was to show the basic concepts and practical application of Partial Least Squares (PLS) as a tutorial, using the Matlab computing environment for beginners, undergraduate and graduate students. As a practical example, the determination of the drug paracetamol in commercial tablets using Near-Infrared (NIR) spectroscopy and Partial Least Squares (PLS) regression was shown, an experiment that has been successfully carried out at the Chemical Institute of Campinas State University for chemistry undergraduate course students to introduce the basic concepts of multivariate calibration in a practical way.

Bico-de-papagaio é uma espécie ornamental, encontrada tanto na forma de arbustos, presente em jardins residenciais e públicos, como comercializada em vasos para decoração de interiores. Com o objetivo de detectar a presença de fitoplasma em plantas mostrando sintomas de proliferação de ramos, amostras foram obtidas de plantas sintomáticas e assintomáticas cultivadas em jardins e de plantas comerciais sintomáticas envasadas. A técnica de duplo PCR, utilizando os iniciadores R16mF2/mR1 e R16F2n/R2, foi empregada para detecção do fitoplasma, a partir de DNA extraído de folhas. Fitoplasma foi detectado consistentemente em todas as amostras sintomáticas, porém não houve detecção em qualquer das amostras assintomáticas. A identificação molecular conduzida através de PCR com iniciadores específicos e por análise de RFLP revelou que o fitoplasma detectado pertence ao grupo 16SrIII. Assim, foi demonstrado que a ocorrência de proliferação de ramos em bico-de-papagaio está associada à presença de fitoplasma. Plantas superbrotadas apresentam um dossel mais compacto, sendo mais atrativas ao consumidor, portanto a presença do fitoplasma é considerada como benéfica para fins ornamentais e comerciais.

Os vírus pertencentes ao subgrupo do Sugarcane mosaic virus (SCMV, gênero Potyvirus, família Potyviridae) infectam e causam mosaico em diferentes espécies botânicas da subfamília Panicoideae (família Poaceae), porém apenas o SCMV e o Sorghum mosaic virus (SrMV) infectam naturalmente cana-de-açúcar. No Brasil, a espécie SCMV parece ser o único agente causal da doença. Embora a maioria das variedades comerciais de cana-de-açúcar seja considerada resistente ou tolerante ao SCMV, relatos de incidência de mosaico em tais variedades têm ocorrido no campo. Amostras com sintomas, de diversos clones e variedades, foram coletadas em campos experimentais e comerciais de cana-de-açúcar. Dentre as variedades, a RB72-454, uma das mais plantadas no país e considerada resistente à doença, também apresentou plantas com sintomas de mosaico. As amostras foram testadas por DAS-ELISA, com anti-soros policlonais para as espécies SCMV, Johnsongrass mosaic virus (JGMV) e Maize dwarf mosaic virus (MDMV), apresentando resultados negativos. Porém, sintomas de mosaico foram observados em mudas de sorgo "Rio" e "TX2786" quando inoculadas mecanicamente com os isolados, indicando tratar-se de infecção pelo SCMV. RNA total foi extraído das folhas de cana e submetido a RT-PCR com oligonucleotídeos específicos para SCMV e SrMV. Fragmentos específicos de aproximadamente 880 pares de bases foram amplificados com os oligonucleotídeos para o SCMV, confirmando os resultados da inoculação mecânica. Os produtos de PCR foram clonados e seqüenciados. Um dos isolados de SCMV encontrado constitui uma nova estirpe, mais severa, capaz de infectar plantas da variedade RB72-454 e de outras variedades, consideradas tolerantes, no campo.

Com o objetivo de quantificar o efeito da cercosporiose do milho (Cercospora zeae-maydis) no rendimento de 12 híbridos comerciais de milho e correlacionar a redução na produtividade de grãos com a severidade da doença, foram conduzidos em duas épocas de semeadura, dois experimentos em área experimental da Universidade Federal de Lavras, em Lavras MG, no ano agrícola de 2005/2006. O primeiro experimento foi instalado em 11/11/2005 e o segundo em 23/12/2005. O controle da doença nas parcelas sadias de todos os híbridos foi obtido através de duas aplicações de epoxiconazol + piraclostrobina (37,5 + 99,75 g i.a.ha-1) em intervalo de 15 dias. Nas parcelas sem controle químico da doença, a epidemia iniciou o seu desenvolvimento com o inóculo natural presente na área. Foram realizadas oito avaliações da severidade da doença em intervalos de sete dias, a partir dos 60 dias após a emergência, por meio de escala de notas variando de 1 (resistente) a 9 (suscetível). Estimou-se a área abaixo da curva de progresso da doença (AACPD) e obteve-se a produtividade de grãos. Com os dados estimou-se a porcentagem de danos e as correlações entre a produtividade de grãos e a porcentagem de danos com as estimativas da AACPD. Constatou-se que: o nível de dano varia entre as épocas de semeadura e híbridos avaliados, sendo em média de 13,3%; a cercosporiose é uma doença que reduz a produtividade de grãos de milho, principalmente para a semeadura tardia e o uso de híbridos resistentes dispensa o controle químico da doença.
