Hsp70 proteins are a family of molecular chaperones that are involved in many aspects of protein homeostasis. In this study, an Hsp70 homologue (SoHsp70) was identified from red drum Sciaenops ocellatus and analyzed at molecular level. The open reading frame of SoHsp70 is 1920 bp and intronless, with a 5'-untranslated region (UTR) of 399 bp and a 3'-UTR of 241 bp. The deduced amino acid sequence of SoHsp70 shares 84-92% overall identities with the Hsp70s of a number of fish species. In silico analysis identified in SoHsp70 three conserved Hsp70 domains involved in nucleotide and substrate binding. The coding sequence of SoHsp70 was subcloned into Escherichia coli, from which recombinant SoHsp70 was purified and, upon ATPase assay, found to exhibit apparent ATPase activity. Expressional analysis showed that constitutive expression of SoHsp70 was detectable in heart, liver, spleen, kidney, brain, blood, and gill. Experimental challenges with poly(I:C) and bacterial pathogens of Gram-positive and Gram-negative nature induced SoHsp70 expression in kidney to different levels. Stress-responsive analysis of SoHsp70 expression in primary cultures of red drum hepatocytes showed that acute heat shock treatment elicited a rapid induction of SoHsp70 expression which appeared after 10 min and 30 min of treatment. Exposure of hepatocytes separately to iron, copper, mercury, and hydrogen peroxide significantly unregulated SoHsp70 expression in time-dependent manners. Vaccination of red drum with a Streptococcus iniae bacterin was also found to induce SoHsp70 expression. Furthermore, recombinant SoHsp70 enhanced the immunoprotective effect of a subunit vaccine. Taken together, these results suggest that SoHsp70 is a stress-inducible protein that is likely to play a role in immunity and in coping with environmental and biological stresses. Web threats are becoming a major issue for both governments and companies. Generally, web threats increased as much as 600% during last year (WebSense, 2013). This appears to be a significant issue, since many major businesses seem to provide these services. Denial of Service (DoS) attacks are one of the most significant web threats and generally their aim is to waste the resources of the target machine (Mirkovic & Reiher, 2004). Dis-tributed Denial of Service (DDoS) attacks are typically executed from many sources and can result in large traf-fic flows. During last year 11% of DDoS attacks were over 60 Gbps (Prolexic, 2013a). The DDoS attacks are usually performed from the large botnets, which are networks of remotely controlled computers. There is an increasing effort by governments and companies to shut down the botnets (Dittrich, 2012), which has lead the attackers to look for alternative DDoS attack methods. One of the techniques to which attackers are returning to is DDoS amplification attacks. Amplification attacks use intermediate devices called amplifiers in order to amplify the attacker's traffic. This work outlines an evaluation tool and evaluates an amplification attack based on the Trivial File Transfer Proto-col (TFTP). This attack could have amplification factor of approximately 60, which rates highly alongside other researched amplification attacks. This could be a substantial issue globally, due to the fact this protocol is used in approximately 599,600 publicly open TFTP servers. Mitigation methods to this threat have also been consid-ered and a variety of countermeasures are proposed. Effects of this attack on both amplifier and target were analysed based on the proposed metrics. While it has been reported that the breaching of TFTP would be possible (Schultz, 2013), this paper provides a complete methodology for the setup of the attack, and its verification.

BACKGROUND: Methicillin-resistant Staphylococcus aureus (MRSA) is a common cause of complicated skin and skin-structure infection (cSSSI). Increasing antimicrobial resistance in cSSSI has led to a need for new safe and effective therapies. Ceftaroline was evaluated as treatment for cSSSI in 2 identical phase 3 clinical trials, the pooled analysis of which is presented here. The primary objective of each trial was to determine the noninferiority of the clinical cure rate achieved with ceftaroline monotherapy, compared with that achieved with vancomycin plus aztreonam combination therapy, in the clinically evaluable (CE) and modified intent-to-treat (MITT) patient populations. METHODS: Adult patients with cSSSI requiring intravenous therapy received ceftaroline (600 mg every 12 h) or vancomycin plus aztreonam (1 g each every 12 h) for 5-14 days. RESULTS: Of 1378 patients enrolled in both trials, 693 received ceftaroline and 685 received vancomycin plus aztreonam. Baseline characteristics of the treatment groups were comparable. Clinical cure rates were similar for ceftaroline and vancomycin plus aztreonam in the CE (91.6% vs 92.7%) and MITT (85.9% vs 85.5%) populations, respectively, as well as in patients infected with MRSA (93.4% vs 94.3%). The rates of adverse events, discontinuations because of an adverse event, serious adverse events, and death also were similar between treatment groups. CONCLUSIONS: Ceftaroline achieved high clinical cure rates, was efficacious against cSSSI caused by MRSA and other common cSSSI pathogens, and was well tolerated, with a safety profile consistent with the cephalosporin class. Ceftaroline has the potential to provide a monotherapy alternative for the treatment of cSSSI. Se presentan en este reporte algunos resultados obtenidos en el aula de matemática a propósito del desarrollo de la situación problema: ¿Qué relación existe entre el ángulo en posición normal y el cociente del lado opuesto y la hipotenusa del triángulo rectángulo? (Fig. 1) Esta actividad se implementó con el objetivo de contribuir al desarrollo del pensamiento variacional de los alumnos de 10º grado jornada de la tarde del Colegio Nacional Loperena de Valledupar, a través de la mediación instrumental de la calculadora algebraica TI-92+ y el uso de las distintas representaciones semióticas para movilizar el aprendizaje de la red conceptual subyacente a la función Seno.

La introducción a la clase de matemáticas de la calculadora TI 92 Plus y otros dispositivos, tales como el CBR, están generando una nueva cultura matemática, caracterizar algunos rasgos de éste fenómeno educativo en la modelación del movimiento pendular es el propósito central de la presente investigación. El trabajo de los estudiantes permitió observar en la práctica los constitutivos del marco teórico del proyecto de incorporación de nuevas tecnologías al currículo de matemáticas de Colombia, como son: mediación instrumental, representaciones ejecutables, cognición situada, solución de problemas, fluidez algorítmica y fluidez conceptual.

En el trabajo que hemos venido realizando en las pasantías de extensión, pretendemos desarrollar parte de la trigonometría desde la época griega hasta la actualidad; tomando como eje central la proporcionalidad, basados en una metodología de resolución de problemas e implementado la calculadora T.I.- 92 Plus en el aula. Para llevar a cabo este proyecto, diseñamos una serie de actividades enfocadas a desarrollar el concepto de proporcionalidad, trabajando desde la semejanza de triángulos. Este enfoque permite al estudiante, por medio de sus experiencias, construir un conjunto de herramientas que le contribuya no sólo enfrentarse a una situación problema, sino que también le ayude a desarrollar su comprensión y habilidad matemáticas.

Actualmente la influencia que tiene la tecnología en la Educación Matemática como medio facilitador tanto en el proceso de aprendizaje como en el de enseñanza de algunas temáticas de las matemáticas escolares, ha generado su inclusión en las instituciones educativas como es el caso, de las calculadoras graficadoras. No obstante, en general son varios los profesores de matemáticas que aunque cuentan con este tipo de recursos para desarrollar sus clases, se abstienen de utilizarlos porque no saben cómo ni en qué momento hacerlo. Por tanto, se presenta algunas actividades sobre ciertas temáticas de la matemática escolar en donde sea factible el uso de herramientas básicas de la calculadora.

We present wide-field neutral hydrogen (H I) Lovell telescope multibeam, and Dominion Radio Astrophysical Observatory Hi synthesis observations, of the high velocity cloud (HVC) located in the general direction of the globular cluster M92. This cloud is part of the larger Complex C and lies at velocities between similar to -80 and -130 km s(-1) in the Local Standard of Rest. The Lovell telescope observations, of resolution 12 arcmin spatially and 3.0 km s(-1) in velocity, fully sampling a 3.1 degrees x 12.6 degrees RA-Dec grid, have found that this part of HVC Complex C comprises two main condensations, lying approximately north-south in declination, separated by similar to2 degrees and being parallel to the Galactic plane. At this resolution, peak values of the brightness temperature and Hi column density of similar to1.4 K and similar to5 x 10(19) cm(-2) are determined, with relatively high values of the full width half maximum velocity (FWHM) of similar to 22 km s(-1) being observed, equivalent to a gas kinetic temperature, in the absence of turbulence and geometric effects of similar to 10 000 K. Each of these properties, as well as the sizes of the clouds, are similar in the two components. The DRAO observations, towards the Northern HVC condensation, are the first high-resolution Hi spectra of Complex C. When smoothed to a resolution of 3 arcmin, they identify several Hi intensity peaks with column densities in the range 4-7 x 10(19) cm(-2). Further smoothing of these data to 6 arcmin resolution tentatively indicates that parts of the HVC consist of two velocity components, of similar brightness temperature, separated by similar to7 km s(-1) in velocity, and with FWHM velocity widths of similar to5-7 km s(-1). No IRAS 60 or 100 micron flux is associated with the M92 HVC. Cloud properties are briefly discussed and compared to previous observations of HVCs.

We have studied a solid-to-plasma transition by irradiating Al foils with the FLASH free electron laser at intensities up to 10(16) W/cm(2). Intense XUV self-emission shows spectral features that are consistent with emission from regions of high density, which go beyond single inner-shell photoionization of solids. Characteristic features of intrashell transitions allowed us to identify Auger heating of the electrons in the conduction band occurring immediately after the absorption of the XUV laser energy as the dominant mechanism. A simple model of a multicharge state inverse Auger effect is proposed to explain the target emission when the conduction band at solid density becomes more atomiclike as energy is transferred from the electrons to the ions. This allows one to determine, independent of plasma simulations, the electron temperature and density just after the decay of crystalline order and to characterize the early time evolution.

Geochemical variables (TOC, C/N, TS, delta C-13) and diatom assemblages were analyzed in a lake sediment sequence from Nong (Lake) Han Kumphawapi in northeast Thailand to reconstruct regional climatic and environmental history during the Holocene. By around c. 10,000-9400 cal yr BP, a large shallow freshwater lake had formed in the Kumphawapi basin. Oxygenated bottom waters and a well-mixed water column were characteristic of this early lake stage, which was probably initiated by higher effective moisture and a stronger summer monsoon. Decreased run-off after c. 6700 cal yr BP favored increased aquatic productivity in the shallow lake. Multiple proxies indicate a marked lowering of the lake level around 5900 cal yr BP, the development of an extensive wetland around 5400 cal yr BP, and the subsequent transition to a peatland. The shift from shallow lake to wetland and later to a peatland is interpreted as a response to lower effective moisture. A hiatus at the transition from wetland to peatland suggests very low accumulation rates, which may result from very dry climatic conditions. A rise in groundwater and lake level around 3200 cal yr BP allowed the re-establishment of a wetland in the Kumphawapi basin. However, the sediments deposited between c. 3200 and 1600 cal yr BP provide evidence for at least two hiatuses at c. 2700-2500 cal yr BP, and at c. 1900-1600 cal yr BP, which would suggest surface dryness and consequently periods of low effective moisture. Around 1600 cal yr BP a new shallow lake became re-established in the basin. Although the underlying causes for this new lake phase remain unclear, we hypothesize that higher effective moisture was the main driving force. This shallow lake phase continued up to the present but was interrupted by higher nutrient fluxes to the lake around 1000-600 cal yr BP. Whether this was caused by intensified human impact in the catchment or, whether this signals a lowering of the lake level due to reduced effective moisture, needs to be corroborated by further studies in the region. The multi-proxy study of Kumphawapi's sediment core CP3A clearly shows that Kumphawapi is a sensitive archive for recording past shifts in effective moisture, and as such in the intensity of the Asian summer monsoon. Many more continental paleorecords, however, will be needed to fully understand the spatial and temporal patterns of past changes in Asian monsoon intensity and its ecosystem impacts.
