Previously we reported that oxalate oxidase activity increases in extracts of barley (Hordeum vulgare) leaves in response to the powdery mildew fungus (Blumeria [syn. Erysiphe] graminis f.sp. hordei) and proposed this as a source of H2O2 during plant-pathogen interactions. In this paper we show that the N terminus of the major pathogen-response oxalate oxidase has a high degree of sequence identity to previously characterized germin-like oxalate oxidases. Two cDNAs were isolated, pHvOxOa, which represents this major enzyme, and pHvOxOb', representing a closely related enzyme. Our data suggest the presence of only two oxalate oxidase genes in the barley genome, i.e. a gene encoding HvOxOa, which possibly exists in several copies, and a single-copy gene encoding HvOxOb. The use of 3′ end gene-specific probes has allowed us to demonstrate that the HvOxOa transcript accumulates to 6 times the level of the HvOxOb transcript in response to the powdery mildew fungus. The transcripts were detected in both compatible and incompatible interactions with a similar accumulation pattern. The oxalate oxidase is found exclusively in the leaf mesophyll, where it is cell wall located. A model for a signal transduction pathway in which oxalate oxidase plays a central role is proposed for the regulation of the hypersensitive response.

We have characterized a family of repetitive DNA elements with homology to the MgPa cellular adhesion operon of Mycoplasma genitalium, a bacterium that has the smallest known genome of any free-living organism. One element, 2272 bp in length and flanked by DNA with no homology to MgPa, was completely sequenced. At least four others were partially sequenced. The complete element is a composite of six regions. Five of these regions show sequence similarity with nonadjacent segments of genes of the MgPa operon. The sixth region, located near the center of the element, is an A+T-rich sequence that has only been found in this repeat family. Open reading frames are present within the five individual regions showing sequence homology to MgPa and the adjacent open reading frame 3 (ORF3) gene. However, termination codons are found between adjacent regions of homology to the MgPa operon and in the A+T-rich sequence. Thus, these repetitive elements do not appear to be directly expressible protein coding sequences. The sequence of one region from five different repetitive elements was compared with the homologous region of the MgPa gene from the type strain G37 and four newly isolated M. genitalium strains. Recombination between repetitive elements of strain G37 and the MgPa operon can explain the majority of polymorphisms within our partial sequences of the MgPa genes of the new isolates. Therefore, we propose that the repetitive elements of M. genitalium provide a reservoir of sequence that contributes to antigenic variation in proteins of the MgPa cellular adhesion operon.

T-cell receptors (TCRs) recognize peptide bound within the relatively conserved structural framework of major histocompatibility complex (MHC) class I or class II molecules but can discriminate between closely related MHC molecules. The structural basis for the specificity of ternary complex formation by the TCR and MHC/peptide complexes was examined for myelin basic protein (MBP)-specific T-cell clones restricted by different DR2 subtypes. Conserved features of this system allowed a model for positioning of the TCR on DR2/peptide complexes to be developed: (i) The DR2 subtypes that presented the immunodominant MBP peptide differed only at a few polymorphic positions of the DR beta chain. (ii) TCR recognition of a polymorphic residue on the helical portion of the DR beta chain (position DR beta 67) was important in determining the MHC restriction. (iii) The TCR variable region (V) alpha 3.1 gene segment was used by all of the T-cell clones. TCR V beta usage was more diverse but correlated with the MHC restriction--i.e., with the polymorphic DR beta chains. (iv) Two clones with conserved TCR alpha chains but different TCR beta chains had a different MHC restriction but a similar peptide specificity. The difference in MHC restriction between these T-cell clones appeared due to recognition of a cluster of polymorphic DR beta-chain residues (DR beta 67-71). MBP-(85-99)-specific TCRs therefore appeared to be positioned on the DR2/peptide complex such that the TCR beta chain contacted the polymorphic DR beta-chain helix while the conserved TCR alpha chain contacted the nonpolymorphic DR alpha chain.

Os fungos do gênero Metarhizium são entomopatogênicos e ainda apresentam relações endofíticas e podem viver saprofiticamente no solo. A associação desses microrganismos com insetos é bem conhecida, mas as interações diretas com as plantas ainda são incipientes. Objetivou-se com este estudo, determinar a capacidade de colonização endofítica em raízes de cana-de-açúcar (Saccharum spp.) de M. robertsii, M. anisopliae e três linhagens brasileiras recém-descobertas, bem como revelar o potencial destes fungos como antagonistas a dois fungos fitopatogênicos responsáveis pela podridão-vermelha (Fusarium moniliforme e Colletotrichum falcatum) e no controle de duas importantes pragas, a broca da cana-de-açúcar, Diatraea saccharalis e o nematóide-das-galhas, Meloidogyne javanica. Foram utilizados 24 isolados de Metarhizium spp. nos experimentos de promoção de crescimento de plantas após a inoculação em gemas de cana-de-açúcar em condições de casa-de-vegetação e campo. O efeito da inoculação do fungo em mudas de cana-de-açúcar foi investigado quanto a mortalidade e redução no desenvolvimento de D. saccharalis e redução nas populações de M. javanica. Experimentos de inibição in vitro em cultivo pareado foram conduzidos com os fungos F. moniliforme e C. falcatum e Trichoderma harzianum. Além disto, foram realizados testes qualitativos \"in vitro\" para avaliar a capacidade desses fungos em produzir β-1,3-Glucanase e Sideróforos. A inoculação de Metarhizium de todas as espécies testadas promoveu o crescimento de parte aérea, peso úmido e seco de raízes em relação ao controle (água). Enquanto o tratamento de gemas rotineiramente usado na usina onde se realizou a produção de mudas pré-brotadas com aplicação do fungicida Comet® + o fertilizante Biozyme TF resultou em pegamento de 32% e 59,6%, apenas com o uso de isolados de Metarhizium spp. estes valores foram de até 50% e 86,4%, nos dois experimentos em campo, respectivamente. As mortalidades observadas em lagartas de D. saccharalis que se alimentaram das plantas inoculadas com isolados de Metarhizium spp. atingiram valores de até 80%, sendo a mortalidade confirmada pela esporulação do fungo de até 55%. As lagartas sobreviventes apresentaram menor peso do que aquelas do controle não tratado. A inoculação de Metarhizium spp. em mudas de cana-de-açúcar conferiu proteção a M. javanica, resultando em uma densidade de massa de ovos por grama de raiz nas mudas inoculadas com o fungo até 59,8% menor no primeiro experimento e 64,1% menor no segundo em relação as mudas sem inoculação. Os ensaios de antagonismo mostraram que, todos os isolados de Metarhizium spp. apresentaram alguma forma de inibição tanto para F. moniliforme como para C. falcatum. Dos 24 isolados testados, 20,8% produziram a enzima hidrolítica, β-1,3-glucanase, o que pode estar associado a capacidade de inibição dos fungos fitopatogênicos. Constatou-se que 8 (33,3%) dos isolados produziram sideróforos e sugere que esses fungos apresentam mecanismos de disponibilização do ferro. O conhecimento gerado com este estudo poderá subsidiar novas estratégias de utilização de Metarhizium spp. em campo na produção de MPB de cana-de-açúcar.

Espécies de Phytophthora tem se destacado ao longo da história devido ao seu potencial destrutivo, se iniciando com a devastadora P. infestans na Irlanda e se estende até os dias de hoje com P. nicotianae em citros e P. plurivora em faia. Uma característica importante deste grupo de patógenos é que as medidas de controle da doença se baseiam na prevenção da entrada do patógeno na área visto que, uma vez instalado, o produtor precisa conviver com o mesmo, pois não se dispõem de métodos efetivos de controle. Neste sentido, a busca por métodos de controle torna-se primordial. O endofítico radicular Piriformospora indica, tem-se destacado em vários patossistemas devido a sua habilidade de induzir resistência contra patógenos, aumentar a tolerância à estresses abióticos e promover o crescimento de plantas. Taxtomina A, produzida por Streptomyces scabies, é capaz de ativar mecanismos de defesa de plantas, os quais são efetivos contra agentes patogênicos. Objetivou-se com este trabalho avaliar o efeito de P. indica e da taxtomina A sobre P. nicotianae em citros e P. plurivora em faia. Ambos foram avaliados quanto ao seu efeito direto sobre os patógenos em questão. O indutor de defesa vegetal Bion® foi utilizado em alguns ensaios para fins de comparação. Plântulas de citros e faia foram tratadas com concentrações crescentes de taxtomina e parâmetros fisiológicos, bioquímicos e de controle da doença foram avaliados. Taxtomina A não apresenta efeito direto sobre os patógenos avaliados. Os dados de incidência da doença em plântulas de faia tratadas com taxtomina A nas concentrações de 10, 25, 50 e 100 μg se mostraram consistentes com a quantidade de DNA do patógeno no sistema radicular, demonstrando que, aparentemente, a toxina induziu suscetibilidade nas plântulas de faia. Em citros, para os parâmetros fisiológicos e bioquímicos avaliados, em linhas gerais, a taxtomina A nas concentrações de 50 e 100 μg demonstrou potencial de aplicação no patossistema citros - P. nicotianae. Quando avaliada a mortalidade de plantas inoculadas com o patógeno e tratadas com taxtomina, bem como, quando quantificado o DNA do oomiceto no sistema radicular, as referidas concentrações também apresentaram os melhores desempenhos. Plântulas das mesmas espécies foram submetidas a inoculação com P. indica, sendo avaliados os efeitos na promoção de crescimento, na atividade de enzimas e de genes relacionados ao processo de defesa, bem como, no controle da doença. Não foi observado efeito direto do endofítico radicular sobre os patógenos avaliados. Quando plântulas de citros foram inoculadas com P. indica e depois com P. nicotianae, não foi observada promoção de crescimento e controle da doença. As análises histológicas e moleculares demonstraram a presença do endofítico no sistema radicular de plântulas de citros e faia. Análises bioquímicas revelaram apenas aumentos pontuais no teor de proteínas e na atividade da β-1,3-glucanase e da peroxidase no tratamento com P. indica + P. nicotianae. Os genes PR-1.4, PR-1.8, PR-β-glucosidase e Hsp70 foram induzidos em plântulas inoculadas com P. indica e com o patógeno, bem como no tratamento com Bion® e patógeno, porém em menor magnitude. O endofítico P. indica ativa o sistema de defesa de plântulas de citros, no entanto, os mecanismos ativados não são efetivos para o controle da doença na interação citros - P. nicotianae.

The fungal species Guignardia citricarpa is an important pathogen in citriculture. Members of the fungal genus Trichoderma are recognized as biocontrol agents but studies on the interactions between both fungi are scarce. This study aimed to identify extracellular proteins secreted by Trichoderma atroviride T17 that are related to the control of G. citricarpa. Two-dimensional gel electrophoresis (2D) was used to study the patterns of proteins secreted by T. atroviride T17 in medium containing glucose (control) and in medium containing G. citricarpa GC3 inactivated mycelium. We identified 59 of the 116 spots differentially expressed (50.86%) by LC–MS/MS. Of these, we highlight the presence of glycoside hydrolases (CAZy families 3, 43, 54, 76 and 93), chitinase, mutanase, a-1,3-glucanase, a-1,2-mannosidase, carboxylic hydrolase ester, carbohydrate-binding module family 13, glucan 1,3-b-glucosidase, a-galactosidase and Neutral protease 2. These proteins are related to mycoparasitism processes, stimuli and therefore to the biological control of pathogens. The results obtained are in agreement with reports describing an increase in the secretion of proteins related to mycoparasitism and biological control and a reduction in the secretion of proteins related to the metabolism of Trichoderma species grown in the presence of the pathogen. Moreover, these results are pioneer in understanding T. atroviride interaction with G. citricarpa. For the first time, we identified potential candidate proteins that may have a role in the antagonism mechanism of G. citricarpa by T. atroviride T17. Thus our results shed a light into the molecular mechanisms that T. atroviride use to control G. citricarpa.

Induction of resistance is defined as the activation of a state of resistance against diseases which is induced systemically in plants by the use of biotic or abiotic agents without any modification of the plant genome, occurring non-specific way, by activating genes coding for various plant defense responses. Chitosan is a polymer derived from the deacetylation of chitin, which is found in large quantities in crustacean shell, and studied with the potential to control plant pathogens, both by its direct fungistatic action, as the ability to induce protection of plants, indicating the presence of molecules of elicitoras characteristics. Three experiments with objective of evaluating the potential of chitosan in the seedling resistance induction were developed, beet (Beta vulgaris) seeds, cucumber (Cucumis sativus) seeds and tomato (Solanum lycopersicum) seeds, and the control of Fusarium sp., Rhizoctonia solani K¨uhn e Pythium sp. in vitro conditions. The experimental design was completely randomized, with four replications. Beet seeds, tomato and cucumber were submerged in chitosan solution for 20 minutes, in concentrations of 0.25, 0.5, 1 and 2% in the control and distilled water. Seeds were sown in trays containing Plantmax Florestalr substrate sterilized and inoculated with Fusarium sp., Rhizoctonia solani K¨unh and Pythium sp., respectively for the three cultures. The experiment was conducted for 14 days in growth chamber with controlled temperature (25 C 2 C), light (12 hour photoperiod) and humidity (70% 10%). The evaluations were seed emergency, seedling damping-off, seedling length, fresh weight and activity of the enzymes phenylalanine amˆonia-liase (PAL), chitinase and b-1,3-glucanase. It was also rated the mycelial growth of Fusarium sp., Pythium sp. and R. solani on P.D.A. (Potato-Dextrose and Agar) culture medium containing chitosan at the same concentrations evaluated in seeds. For beet growing, seed treatment with chitosan presented higher emergence and the length of the seedlings, and reduced the percentage of tipping. Treatment with chitosan activated the systemic acquired resistance with expression of chitinase and b-1,3-glucanase enzymes. For the tomato crop in chitosan concentration of 0.25% favored the emergency of seedlings, reduced the incidence of tipping and activated the PAL enzymes, chitinase and b-1,3-glucanase. In cucumber on the concentration of up 0.5% favored seedlings emergence and reduces the incidence of tipping. Chitosan activated the PAL enzymes and b-1,3-glucanase. Chitosan also presented fungistatic action on the initial growth of Pythium sp. and R. solani in vitro conditions, however, such action did not prevail until the end of the experiment. To Fusarium sp. the concentration of chitosan resulted in the reduction of mycelial growth in vitro.

The use of inputs containing phosphites have been presenting results in many studies, taking on importance to the control of diseases in some cultures and demonstrating the resistance induction in seedlings, with ability to activate defense mechanisms, conferring protection to plants against microorganisms. The soybean crop is recognized for its importance in providing grains and derivatives for human consumption, animal, production of biofuels, pharmaceuticals, among others. Positive results obtained through studies based on resistance inducers in some cultures arouse the interest for further study. The objective of this study was to evaluate the effect of potassium phosphites on the resistance induction and treatment of soybean seeds. Therefore were conducted four laboratory studies at the Federal Technological University of Paraná, Campus of Dois Vizinhos. In the first study it was evaluated the quality attributes of the seeds and the resistance induction as seed treatment. Then it was verified that phosphites have action upon the seedlings metabolism in due to seed treatment, having the phosphite Reforce® contributed to seed quality attributes and phosphites FitofosK® and Fitofos K Plus® induced the resistance increasing the activity of β-1,3-glucanase. In the second study it was evaluated the the resistance induction in soybean cotyledons, in which the phosphites demonstrated induction potential of phytoalexin gliceolin. In the third study It was evaluated the soybean seed health treated with potassium phosphites.. it was observed that the phosphites reduced the incidence of many fungi on seeds, especially of storage fungi like Aspergillus sp. and Fusarium semitectum. In the fourth study it was evaluated the in vitro effect of potassium phosphites on pathogenic fungi of the culture. And it was found direct action of phosphites on the mycelial growth of Fusarium semitectum, Pythium sp. and Sclerotinia sclerotiorum. Based on these results, we concluded that potassium phosphites have potential in seeds treatment, as resistance inducer and on in vitro control of phytopathogens.

O interesse na produção de astaxantina de fontes naturais vem aumentando significativamente, devido principalmente à sua capacidade como potente agente antioxidante. Na obtenção da astaxantina por via biotecnológica, a microalga Haematococcus pluvialis é um dos micro-organismos industrialmente mais interessantes. Entretanto, como a maioria dos carotenoides, a astaxantina é uma molécula altamente insaturada que pode ser facilmente degradada por processos térmicos. Em função desta instabilidade, uma possibilidade que se abre, a fim de proteger sua atividade biológica de fatores ambientais e reforçar a sua estabilidade física, é o encapsulamento. Neste sentido, este trabalho vem contribuir em inovações relacionadas ao desenvolvimento de tecnologia para ruptura celular, extração e nanoencapsulamento de astaxantina produzida por via biotecnológica, mais especificamente de astaxantina obtida através do cultivo de H. pluvialis. Neste estudo, os cultivos foram realizados em meio BBM e acetato de sódio e conduzidos a temperatura constante de 25±1 ºC em fotobiorreatores de 1 L com aeração por borbulhamento de ar de 300 mL.min-1 , agitação manual diária e sob iluminância constante de 444 µmol fótons.m-2 s -1 durante 15 dias, sendo inoculados com suspensão de microalgas previamente preparada, na proporção de 10%, e pH ajustado em 7,0. A biomassa foi recuperada dos cultivos por centrifugação e seca a 35 °C por 48 h. Em seguida, foram empregadas diferentes técnicas de ruptura celular (química, mecânica e enzimática). Após a ruptura, foi realizada a extração dos carotenoides e a quantificação dos carotenoides totais (µg.g-1 ) e da extratibilidade (%). Entre os solventes testados no método de ruptura química, o diclorometano foi o selecionado para a extração dos pigmentos carotenoides. Dentre as técnicas mecânicas de ruptura celular, a maceração da biomassa congelada com terra diatomácea resultou na maior extratibilidade e carotenoides totais (66,01% e 972,35 μg.g-1 ). A melhor condição de lise da parede celular de H. pluvialis, utilizando o preparado enzimático Glucanex® , ocorreu em pH do meio reacional de 4,5 a 55 ºC, com atividade inicial de β-1,3-glucanase de 0,6 U.mL-1 e um tempo de reação de 30 min, alcançando-se 17,73% de atividade lítica relativa. Nestas condições, com a reação enzimática assistida por ultrassom sem congelamento prévio da biomassa, atingiu-se 83,90% e 1235,89 µg.g -1 , respectivamente, para extratibilidade e carotenoides totais. Dentre as técnicas combinadas testadas, a maceração com terra diatomácea associada à lise enzimática apresentou valores de extratibilidade e carotenoides totais de, respectivamente, 93,83% e 1382,12 µg.g-1 . No encapsulamento do extrato contendo astaxantina obtido por lise enzimática associada por ultrassom, envolvendo a coprecipitação com PHBV (poli(3-hidroxibutirato-cohidroxivalerato)) em fluidos supercríticos, o aumento da pressão tendeu a reduzir o diâmetro da partícula formada, enquanto que o aumento da relação biomassa contendo astaxantina:diclorometano usada na etapa de extração incrementou o percentual de encapsulamento e a eficiência de encapsulamento para ambas pressões testadas (80 e 100 bar). Os maiores valores de percentual de encapsulamento (17,06%) e eficiência de encapsulamento (51,21%) foram obtidos nas condições de 80 bar e relação biomassa:diclorometano de 10 mg.mL -1 . Nestas condições, o diâmetro médio de partícula foi de 0,228 µm. Com base nos resultados obtidos, técnicas para a obtenção de astaxantina de H. pluvialis e seu encapsulamento foram desenvolvidas com sucesso, podendo ser extendidas a outros produtos intracelulares de microalgas.

Lipopolysaccharide and beta-1,3-glucan-binding protein (LGBP) play a crucial role in the innate immune response of invertebrates as a pattern recognition protein (PRP). The scallop LGBP gene was obtained from Chlamys farreri challenged by Vibrio anguillarum by randomly sequencing cDNA clones from a whole body cDNA library, and by fully sequencing a clone with homology to known LGBP genes. The scallop LGBP consisted of 1876 nucleotides with a canonical polyadenylation signal sequence AATAAA and a poly(A) tail, encoding a polypeptide of 440 amino acids with the estimated molecular mass of 47.16 kDa and a predicted isoelectric point of 5.095. The deduced amino acid sequence showed a high similarity to that of invertebrate recognition proteins from blue shrimp, black tiger shrimp, mosquito, freshwater crayfish, earthworms, and sea urchins, with conserved features including a potential polysaccharide-binding motif, a glucanase motif, and N-glycosylation sites. The temporal expression of LGBP genes in healthy and V. anguillarum-challenged C farreri scallop, measured by real-time semiquantitative reverse transcription polymerase chain reaction (PCR), showed that expression was up-regulated initially, followed by recovery as the stimulation cleared. Results indicated that scallop LGBP was a constitutive and inducible acute-phase protein that could play a critical role in scallop-pathogen interaction. (C) 2004 Elsevier B.V. All rights reserved.

The codon usage of a hybrid bacterial gene encoding a thermostable (1,3-1,4)-beta-glucanase was modified to match that of the barley (1,3-1,4)-beta-glucanase isoenzyme EII gene. Both the modified and unmodified bacterial genes were fused to a DNA segment encoding the barley high-pI alpha-amylase signal peptide downstream of the barley (1,3-1,4)-beta-glucanase isoenzyme EII gene promoter. When introduced into barley aleurone protoplasts, the bacterial gene with adapted codon usage directed synthesis of heat stable (1,3-1,4)-beta-glucanase, whereas activity of the heterologous enzyme was not detectable when protoplasts were transfected with the unmodified gene. In a different expression plasmid, the codon modified bacterial gene was cloned downstream of the barley high-pI alpha-amylase gene promoter and signal peptide coding region. This expression cassette was introduced into immature barley embryos together with plasmids carrying the bar and the uidA genes. Green, fertile plants were regenerated and approximately 75% of grains harvested from primary transformants synthesized thermostable (1,3-1,4)-beta-glucanase during germination. All three trans genes were detected in 17 progenies from a homozygous T1 plant.

Mutations in exon 3 of the CTNNB1 gene encoding beta-catenin have been reported in colorectal cancer cell lines and tumours. Although one study reported mutations or deletions affecting beta-catenin in 20% of melanoma cell lines, subsequent reports detected a much lower frequency of aberrations in uncultured melanomas. To determine whether this difference in mutation frequency reflected an in vitro culturing artefact, exon 3 of CTNNB1 was screened in a panel of 62 melanoma cell lines. In addition, reverse transcription-polymerase chain reaction (RT-PCR) was performed to detect intragenic deletions affecting exon 3. One out of 62 (1.6%) cell lines was found to carry a mutation, indicating that aberration of the Wnt-1/wingless pathway through activation of beta-catenin is a rare event, even in melanoma cell lines.

Migraine is a common neurological disorder characterised by temporary disabling attacks of severe head pain and associated disturbances. There is significant evidence to suggest a genetic aetiology to the disease however few causal mutations have been conclusively linked to the migraine subtypes Migraine with (MA) or without Aura (MO). The Potassium Channel, Subfamily K, member 18 (KCNK18) gene, coding the potassium channel TRESK, is the first gene in which a rare mutation resulting in a non-functional truncated protein has been identified and causally linked to MA in a multigenerational family. In this study, three common polymorphisms in the KCNK18 gene were analysed for genetic variation in an Australian case-control migraine population consisting of 340 migraine cases and 345 controls. No association was observed for the polymorphisms examined with the migraine phenotype or with any haplotypes across the gene. Therefore even though the KCNK18 gene is the only gene to be causally linked to MA our studies indicate that common genetic variation in the gene is not a contributor to MA.
