The study of biological invasions can be roughly divided into three parts: detection, monitoring, mitigation. Here, our objectives were to describe the marine fauna of the area of the port of São Sebastião (on the northern coast of the state of São Paulo, in the São Sebastião Channel, SSC) to detect introduced species. Descriptions of the faunal community of the SSC with respect to native and allochthonous (invasive or potentially so) diversity are lacking for all invertebrate groups. Sampling was carried out by specialists within each taxonomic group, in December 2009, following the protocol of the Rapid Assessment Survey (RAS) in three areas with artificial structures as substrates. A total of 142 species were identified (61 native, 15 introduced, 62 cryptogenic, 4 not classified), of which 17 were Polychaeta (12, 1, 1, 3), 24 Ascidiacea (3, 6, 15, 0), 36 Bryozoa (17, 0, 18, 1), 27 Cmdana (2, 1, 24, 0), 20 Crustacea (11, 4, 5, 0), 2 Entoprocta (native), 16 Mollusca (13, 3, 0, 0). Twelve species are new occurrences for the SSC. Among the introduced taxa, two are new for coastal Brazil. Estimates of introduced taxa are conservative as the results of molecular studies suggest that some species previously considered cryptogenic are indeed introduced. We emphasize that the large number of cryptogenic species illustrates the need for a long-term monitoring program, especially in areas most susceptible to bioinvasion. We conclude that rapid assessment studies, even in relatively well-known regions, can be very useful for the detection of introduced species and we recommend that they be carried out on a larger scale in all ports with heavy ship traffic.

Grünalgen bilden zur Überdauerung schlechter Umweltbedingungen Ruhestadien, die sich durch Ausbildung einer festen Zellwand, die Reduktion des Plastiden und die starke Akkumulation von Speicherfetten und Ketocarotinoiden im Zytosol auszeichnen. Obwohl Ketocarotinoide in Grünalgen seit über vierzig Jahren beforscht werden, gab es hierzu noch wenige molekularbiologische Untersuchungen. Im Vorfeld meiner Promotion wurde durch unsere Arbeitsgruppe entdeckt, dass auch der molekular gut zugängliche Modellorganismus Chlamydomonas reinhardtii im Zygotenstadium große Mengen an Ketocarotinoiden bildet. Neben dem zu erwartenden Ketocarotinoid Astaxanthin fanden wir große Mengen des bisher nur in einer Grünalge beschriebenen 4-Ketoluteins. Vorversuche ließen die Vermutung aufkommen, dass dieses Pigment bei der Untersuchung der Pigmentausstattung in Dauerstadien von vielen Grünalgen bisher übersehen wurde. rnIn der vorliegenden Arbeit wurde daher zunächst die Pigmentzusammensetzung von Dauerstadien der bereits gut untersuchten Grünalgen Muriella zofingiensis und Scenedesmus rubescens durch Vergleich mit dem Ketocarotinoidmuster aus Dauerstadien von C. reinhardtii und Fritschiella tuberosa reevaluiert und dabei erstmals das Vorkommen signifikanter Mengen an 4-Ketolutein nachgewiesen. Außerdem zeigte sich, dass die als bisheriger Modellorganismus der Ketocarotinoidbiosynthese in Grünalgen sehr gut untersuchte Alge Haematococcus pluvialis eher eine Ausnahme darstellt, da ihre Dauerstadien als einzige der hier untersuchten Algen nur minimale Mengen von 4 Ketolutein aufwiesen. Diese Beobachtungen machen es sehr wahrscheinlich, dass die Fähigkeit zur Bildung von 4-Ketolutein unter den Grünalgen wesentlich weiter verbreitet ist als bisher angenommen. Das sekundäre Carotinoid 4-Ketolutein kam in den Dauerstadien der Grünalgen neben seiner freien Form ausschließlich als Monoacylester vor, im Gegensatz zu Astaxanthin, das als mono- und diacylierte Form auftrat. rnÜber die Analyse der Pigmentausstattung hinaus konnten die entscheidenden Schritte des Synthesewegs der Ketocarotinoide in C. reinhardtii durch funktionelle Charakterisierung der beteiligten Enzyme in Bakterien aufgeklärt werden. Als Basis für die Charakterisierungen wurde ein umfangreiches Portfolio von carotinogenen E. coli-Bakterien etabliert, darunter α Carotin und Lutein produzierende Stämme, die bisher nicht zur Verfügung standen. Das wurde durch die Klonierung der Lycopinzyklase (OluLCY) aus der Grünalge Ostreococcus lucimarinus möglich, die eine Sonderolle unter den Zyklasen einnimmt, da sie die Lycopin-β-Zyklase und Lycopin-ε-Zyklase in einem Fusionsenzym vereint. Vorteile dieses Fusionsenzyms sind die Expressionskontrolle durch nur einen Promotor und die weitgehend konstante Stöchiometrie seiner Produkte α-Carotin und β-Carotin, was die OluLCY für die biotechnologische Anwendung prädestiniert.rnDie funktionelle Charakterisierung der Carotinoidbiosyntheseenzyme aus C. reinhardtii umfasste das Schlüsselenzym der Ketocarotinoidbiosynthese, die β-Carotin-Ketolase (BKT), sowie die Carotinoid-Hydroxylasen CHYB, CYP97A5 und CYP97C3. Dabei wurde für das BKT-Enzym aus C. reinhardtii nachgewiesen, dass es nicht nur die Ketolierung von β Carotin zu Canthaxanthin und von Zeaxanthin zu Astaxanthin, sondern auch die Bildung der von α-Carotin abgeleiteten Ketocarotinoide wie 4-Keto-α-Carotin und 4 Ketolutein katalysieren kann.rn

Il teorema del viriale consiste in una relazione tra energia cinetica e energia potenziale totali di un sistema all'equilibrio. Il concetto di Viriale (dal latino vires, plurale di vis, 'forza') è stato introdotto dal fisico e matematico tedesco Rudolf Julius Emanuel Clausius (1822-1888) per indicare la quantità N Fi •xi i=1 che rappresenta la somma, fatta su tutte le N particelle di un sistema, dei prodotti scalari del vettore forza totale agente su ciascuna particella per il vettore posizione della particella stessa, rispetto ad un riferimento inerziale scelto. Tale quantità altro non è che un'energia potenziale. Dire che un sistema di particelle è virializzato equivale a dire che esso è stazionario, cioè all'equilibrio. In questo elaborato sono di nostro interesse sistemi astrofisici gravitazionali, in cui cioè l'energia potenziale sia dovuta solo a campi gravitazionali. Distingueremo innanzitutto sistemi collisionali e non collisionali, introducendo i tempi scala di attraversamento e di rilassamento. Dopo una trattazione teorica del teorema, nell'approssimazione di continuità - per cui sostuiremo alle sommatorie gli integrali - e di non collisionalità, an- dremo a studiarne l'importanza in alcuni sistemi astrofisici: applicazione agli ammassi stellari, alle galassie e agli ammassi di galassie, stima della quantità di materia oscura nei sistemi, instabilità di Jeans in nubi molecolari, rotazione delle galassie ellittiche. Per ragioni di spazio non saranno affrontati altri casi, di cui ne citiamo alcuni: collasso delle stelle, stima della massa dei buchi neri al centro delle galassie, 'mass-to-light ratio' di sistemi sferici. Parleremo in generale di “particelle” costituenti i sistemi per intendere stelle, galassie, particelle di gas a seconda del sistema in esame. Trascureremo in ogni caso le influenze gravitazionali di distribuzioni di densità esterne al sistema.

BACKGROUND: Mechanisms underlying improvement of myocardial contractile function after cell therapy as well as arrhythmic side effect remain poorly understood. We hypothesised that cell therapy might affect the mechanical properties of isolated host cardiomyocytes. METHODS: Two weeks after myocardial infarction (MI), rats were treated by intramyocardial myoblast injection (SkM, n=8), intramyocardial vehicle injection (Medium, n=6), or sham operation (Sham, n=7). Cardiac function was assessed by echocardiography. Cardiomyocytes were isolated in a modified Langendorff perfusion system, their contraction was measured by video-based inter-sarcomeric analysis. Data were compared with a control-group without myocardial infarction (Control, n=5). RESULTS: Three weeks post-treatment, ejection fraction (EF) further deteriorated in vehicle-injected and non-injected rats (respectively 40.7+/-11.4% to 33+/-5.5% and 41.8+/-8% to 33.5+/-8.3%), but was stabilised in SkM group (35.9+/-6% to 36.4+/-9.7%). Significant cell hypertrophy induced by MI was maintained after cell therapy. Single cell contraction (dL/dt(max)) decreased in SkM and vehicle groups compared to non-injected group as well as cell shortening and relaxation (dL/dt(min)) in vehicle group. A significantly increased predisposition for alternation of strong and weak contractions was observed in isolated cardiomyocytes of the SkM group. CONCLUSION: Our study provides the first evidence that injection of materials into the myocardium alters host cardiomyocytes contractile function independently of the global beneficial effect of the heart function. These findings may be important in understanding possible adverse effects.

Famille de pasteurs, politiciens et entrepreneurs de Zofingue, attestée pour la première fois en 1527, lorsque leur aïeul Jean, tonnelier originaire de Nîmes, obtint la bourgeoisie de Zofingue. Michael (1521-1605), avoyer, l'un de ses cinq fils, est l'ancêtre de la branche des imprimeurs et éditeurs. Après lui, de nombreux R. consolidèrent durablement l'influence de la famille. A partir du XVIIIe s., divers membres firent des carrières politiques, tels Samuel (1706-1786), avoyer, et Rudolf Friedrich (1805-1886), président de la ville. Les R. furent aussi très liés à l'Eglise. Le fils de Michael, Moritz (1557-1615), fut pasteur et doyen à Zofingue. Jusqu'au XIXe s., la famille compta une trentaine d'ecclésiastiques, essentiellement des pasteurs officiant sur le territoire bernois, tels Johann Heinrich ( -> 3) et Michael ( -> 8). Les conseillers Beat (1712-1778) et Niklaus (1734-1766) furent les premiers R. actifs dans la production protoindustrielle de drap. D'autres négociants suivirent jusqu'au milieu du XIXe s. L'architecte Niklaus Emanuel (1744-1815) construisit l'hôtel de ville de Zofingue (1792-1795) de style baroque tardif. Johann Rudolf ( -> 4) se distingua sous la République helvétique (1798-1803). Samuel (1767-1826), conseiller municipal de Zofingue, créa les armoiries du canton d'Argovie en 1803. Les R. s'affirmèrent sur le plan cantonal avec Karl Ludwig ( -> 6), chancelier, et Arnold ( -> 1), conseiller d'Etat et plusieurs fois landamman, et sur le plan fédéral avec Johann Rudolf ( -> 5), conseiller national, et Gottlieb ( -> 2), conseiller aux Etats et chancelier de la Confédération. Johann Rudolf (1803-1874) fonda, en 1833, l'imprimerie Ringier à Zofingue, reprise par son fils Franz Emil (1837-1898). A partir de 1898, Paul August ( -> 9), représentant de la troisième génération d'imprimeurs, agrandit l'entreprise dont il fit la principale imprimerie et maison d'édition de Suisse. Cette expansion se poursuivit après 1960 sous son fils Hans (1906-2003). Avec les fils de celui-ci, Christoph (naissance1941, dans la firme jusqu'en 1991) et Michael (naissance1949), Ringier devint, à partir de 1985, une entreprise multinationale et multimédia. We report on a 74-year-old male patient who presented with progressive neuroophthalmologic symptoms soon after the administration of a long-acting gonadotropin-releasing hormone agonist for treatment of a prostate cancer. Imaging revealed a destructively growing and extensively calcified sellar mass inconsistent with a pituitary adenoma. A transseptal transsphenoidal tumor mass reduction yielded a histological diagnosis of a collision tumor comprised of a gonadotroph adenoma intermingled with osteochondroma. We discuss a potential causal relationship between the administration of the long-acting gonadotropin-releasing hormone agonist and the sudden appearance of the previously unsuspected sellar lesion. Although the association of these two tumors is very likely coincidental, the possibility of causal relationship is addressed.

Intramyocellular lipids (IMCL) are flexible fuel stores that are depleted by physical exercise and replenished by fat intake. IMCL or their degradation products are thought to interfere with insulin signaling thereby contributing to insulin resistance. From a practical point of view it is desirable to deplete IMCL prior to replenishing them. So far, it is not clear for how long and at which intensity subjects have to exercise in order to deplete IMCL. We therefore aimed at developing a standardized exercise protocol that is applicable to subjects over a broad range of exercise capacity and insulin sensitivity and allows measuring reliably reduced IMCL levels.Twelve male subjects, including four diabetes type 2 patients, with wide ranges of exercise capacity (VO(2)peak per total body weight 27.9-55.8 ml x kg(-1) x min(-1)), insulin sensitivity (glucose infusion rate per lean body mass 4.7-15.3 mg x min(-1) x kg(-1)), and BMI (21.7-31.5 kg x m(-2)), respectively, were enrolled. Using (1)H magnetic resonance spectroscopy ((1)H-MRS), IMCL was measured in m.tibialis anterior and m.vastus intermedius before and during a depletion protocol of a week, consisting of a moderate additional physical activity (1 h daily at 60% VO(2)peak) and modest low-fat (10-15%) diet.Absolute IMCL-levels were significantly reduced in both muscles during the first 3 days and stayed constant for the next 3 days of an identical diet/exercise-scheme. These reduced IMCL levels were independent of insulin sensitivity, yet a tendency to lower depleted IMCL levels has been observed in subjects with higher VO(2)peak.The proposed protocol is feasible in subjects with large differences in exercise capacity, insulin sensitivity, and BMI, leading to reduced IMCL levels that neither depend on the exact duration of the depletion protocol nor on insulin sensitivity. This allows for a standardized preparation of IMCL levels either for correlation with other physiological parameters or for replenishment studies.
Internalized somatostatin receptor subtype 2 in neuroendocrine tumors of octreotide-treated patients

Somatostatin receptor subtype 2 (sst(2)) is widely expressed in neuroendocrine tumors and can be visualized immunohistochemically at the cell membrane for diagnostic purposes. Recently, it has been demonstrated in animal sst(2) tumor models in vivo that somatostatin analog treatment was able to induce a complete internalization of the tumor sst(2).

Pituitary apoplexy in pregnancy is rare. Its clinical features may range from unspecific complaints to panhypopituitarism resulting even in coma and death. Therefore, alertness to signs and symptoms of acute loss of pituitary function in pregnancy is mandatory. We report a woman in her 7th week of her first gestation presenting with sudden coma due to severe hyponatremia. Secondary adrenal insufficiency could be identified as the underlying cause. Panhypopituitarism including central diabetes insipidus and spontaneous abortion developed during the follow-up. Magnetic resonance imaging showed pituitary apoplexy without a pre-existing pituitary mass. The clinical course was notable for severe complications, including neurological deficits through cerebral ischemia, but eventual recovery could be achieved. We discuss the diagnostic difficulties in the evaluation of pituitary disease in pregnancy.

Patients with GH deficiency (GHD) are insulin resistant with an increase in visceral fat mass (FM). Whether this holds true when sedentary control subjects (CS) are matched for waist has not been documented. GH replacement therapy (GHRT) results in a decrease in FM. Whether the decrease in FM is mainly related to a reduction in visceral FM remains to be proven. The aim was to separately assess visceral and subcutaneous FM in relation to insulin resistance (IR) in GHD patients before and after GHRT and in sedentary CS.

Glucagonlike peptide-1 receptors (GLP-1R) play an increasingly important role in endocrine gastrointestinal tumor management. In particular, virtually all benign insulinomas express GLP-1R in high density. Exendin-4 is a GLP-1 analog that has a longer half-life than GLP-1. Targeting GLP-1R by (111)In-DOTA-exendin-4 or (111)In-DPTA-exendin-4 offers a new approach that permits the successful localization of small benign insulinomas. It is likely that this new noninvasive technique has the potential to replace the invasive localization by selective arterial stimulation and venous sampling.

Intramyocellular acetylcarnitine (IMAC) is involved in exercise-related fuel metabolism. It is not known whether levels of systemic glucose influence IMAC levels in type 1 diabetes.

We hypothesized that network analysis is useful to expose coordination between whole body and myocellular levels of energy metabolism and can identify entities that underlie skeletal muscle's contribution to growth hormone-stimulated lipid handling and metabolic fitness. We assessed 112 metabolic parameters characterizing metabolic rate and substrate handling in tibialis anterior muscle and vascular compartment at rest, after a meal and exercise with growth hormone replacement therapy (GH-RT) of hypopituitary patients (n = 11). The topology of linear relationships (| r | ≥ 0.7, P ≤ 0.01) and mutual dependencies exposed the organization of metabolic relationships in three entities reflecting basal and exercise-induced metabolic rate, triglyceride handling, and substrate utilization in the pre- and postprandial state, respectively. GH-RT improved aerobic performance (+5%), lean-to-fat mass (+19%), and muscle area of tibialis anterior (+2%) but did not alter its mitochondrial and capillary content. Concomitantly, connectivity was established between myocellular parameters of mitochondrial lipid metabolism and meal-induced triglyceride handling in serum. This was mediated via the recruitment of transcripts of muscle lipid mobilization (LIPE, FABP3, and FABP4) and fatty acid-sensitive transcription factors (PPARA, PPARG) to the metabolic network. The interdependence of gene regulatory elements of muscle lipid metabolism reflected the norm in healthy subjects (n = 12) and distinguished the regulation of the mitochondrial respiration factor COX1 by GH and endurance exercise. Our observations validate the use of network analysis for systems medicine and highlight the notion that an improved stochiometry between muscle and whole body lipid metabolism, rather than alterations of single bottlenecks, contributes to GH-driven elevations in metabolic fitness.

The role of exercise testing in the assessment of GH deficiency (GHD) in adult patients is currently unclear. This study aimed at evaluating the diagnostic value of exercise-induced GH levels in the detection of severe GHD in adult patients.

Conventional MRI may still be an inaccurate method for the non-invasive detection of a microadenoma in adrenocorticotropin (ACTH)-dependent Cushing's syndrome (CS). Bilateral inferior petrosal sinus sampling (BIPSS) with ovine corticotropin-releasing hormone (oCRH) stimulation is an invasive, but accurate, intervention in the diagnostic armamentarium surrounding CS. Until now, there is a continuous controversial debate regarding lateralization data in detecting a microadenoma. Using BIPSS, we evaluated whether a highly selective placement of microcatheters without diversion of venous outflow might improve detection of pituitary microadenoma.
