La présente thèse doctorale propose l’étude de l’activité théâtrale développée dans les théâtres permanents construits en Amérique Portugaise au long du XVIIIe et dans la première décennie du XIXe siècle, en mettant l’accent sur les idéalisateurs des Casas da Ópera et les moyens de financement utilisés dans l’édification et l’entretien de celles-ci, le répertoire mis en scène, les artistes composant les compagnies théâtrales, les spectateurs et les possibilités de sociabilité établies dans l’espace théâtrale et les modèles architectoniques qui ont inspiré la construction des bâtiments théâtraux au cours de la période mentionnée. Ces cinq axes principaux soulignent l’approche interdisciplinaire qui a orienté nos études, dans un souci de contribution à une plus complète compréhension de notre sujet de recherche. xi RESUMO A acção da insulina no músculo esquelético depende de um reflexo parassimpático hepático que conduz à libertação de uma substância hepática sensibilizadora da insulina, designada por HISS, responsável por cerca de 55% do efeito hipoglicemiante da insulina. A acção da HISS é finamente regulada pelo monóxido de azoto (NO) hepático e pelo estado prandial, aumentando no período pós-prandial imediato e diminuindo progressivamente com as horas de jejum. A secreção da HISS pode ser inibida cirúrgica ou farmacologicamente, quer por desnervação selectiva do plexo anterior hepático, quer por administração de atropina, quer por inibição do sintase do NO (NOS) hepático. O objectivo geral do trabalho apresentado nesta dissertação foi a caracterização da via de transdução de sinal que conduz à libertação da HISS. O modelo utilizado neste estudo foi o rato Wistar. A sensibilidade à insulina foi avaliada através do teste rápido de sensibilidade à insulina (RIST). A primeira hipótese de trabalho testada foi que a sequência de eventos que conduzem à secreção da HISS inicia-se com a activação do sistema parassimpático hepático seguida de activação do NOS hepático com subsequente produção de NO e activação do guanilato ciclase (GC). Observou-se que a administração de um dador de NO reverteu a resistência à insulina induzida, quer por inibição do NOS hepático, quer por antagonismo dos receptores muscarínicos com atropina. Em contraste, a resistência à insulina produzida por inibição do NOS hepático não foi revertida por administração intraportal de acetilcolina (ACh). Constatou-se que a inibição do GC hepático diminuiu a sensibilidade à insulina. Estes resultados sugerem que: a ACh libertada no fígado induz a síntese de NO hepático que conduz à libertação da HISS, que por sua vez é modulada pelo GC hepático. A libertação da HISS em resposta à insulina é regulada pelo estado prandial. Uma vez que os níveis hepáticos de glutationo (GSH) se encontram, tal como a HISS, diminuídos no estado de jejum e aumentados após a ingestão de uma refeição, testou-se a hipótese de que o GSH hepático está envolvido na secreção da HISS. Observou-se que a depleção do GSH hepático induziu resistência à insulina, comparável à obtida após inibição do NOS hepático. Estes resultados suportam a hipótese de que o GSH hepático desempenha um papel crítico na acção periférica da insulina. Considerando que, no estado de jejum, tanto os níveis de GSH hepático como os níveis de NO hepático são baixos, testou-se a hipótese de que a co-administração intraportal de um dador de GSH e de um dador de NO promove um aumento da sensibilidade à insulina no estado de jejum, devido ao restabelecimento do mecanismo da HISS. Observou-se que a administração sequencial de dadores de GSH e de NO no fígado provocou um aumento na sensibilidade à insulina, dependente da dose de dador de GSH administrada. Concluiu-se portanto que ambos, GSH e NO, são essenciais para que o mecanismo da HISS esteja completamente funcional. O GSH e o NO reagem para formar um S-nitrosotiol, o S-nitrosoglutationo (GSNO). Os resultados supra-mencionados conduziram à formulação da hipótese de que a secreção/acção da HISS depende da formação de GSNO. Observou-se que a administração intravenosa de S-nitrosotióis (RSNOs) aumentou a sensibilidade à insulina, em animais submetidos a um período de jejum, ao contrário da administração intraportal destes fármacos, o que RSNOs têm uma acção periférica, mas não hepática, na sensibilidade à insulina. Os resultados obtidos conduziram à reformulação da hipótese da HISS, sugerindo que a ingestão de uma refeição activa os nervos parassimpáticos hepáticos levando à libertação de ACh no fígado que, por sua vez activa o NOS. Simultaneamente, ocorre um aumento dos níveis de GSH hepático que reage com o NO hepático para formar um composto nitrosado, o GSNO. Este composto mimetiza a acção hipoglicemiante da HISS no músculo esquelético. SUMMARY Insulin action at the skeletal muscle depends on a hepatic parasympathetic reflex that promotes the release of a hepatic insulin sensitizing substance (HISS) from the liver, which contributes 55% to total insulin action. HISS action is modulated by hepatic nitric oxide (NO) and also by the prandial status so as to, in the immediate ostprandial state, HISS action is maximal, decreasing with the duration of fasting. HISS secretion may be inhibited by interruption of the hepatic parasympathetic reflex, achieved either by surgical denervation of the liver or by cholinergic blockade with atropine, or by prevention of hepatic NO release, using NO synthase (NOS) antagonists. The main objective of this work was to characterize the signal transduction pathways that lead to HISS secretion by the liver. Wistar rats were used and insulin sensitivity was evaluated using the rapid insulin sensitivity test (RIST). The first hypothesis tested was that the sequence of events that lead to HISS secretion starts with an increase in the hepatic parasympathetic tone, followed by the activation of hepatic NOS and subsequent triggering of guanylate cyclase (GC). We observed that insulin resistance produced either by muscarinic receptor antagonism with atropine or by hepatic NOS inhibition was reversed by the intraportal administration of an NO donor. In contrast, intraportal acetylcholine (ACh) did not restore insulin sensitivity after NOS inhibition. We also observed that GC inhibition lead to a decrease in insulin sensitivity.These results suggest that the release of ACh in the liver activates hepatic NO synthesis in order to allow HISS secretion, through a signaling pathway modulated by GC. HISS release in response to insulin is controlled by the prandial status. The second hypothesis tested was that glutathione (GSH) is involved in HISS secretion since the hepatic levels of GSH are, like HISS action, decreased in the fasted state and increased after ingestion of a meal. We observed that hepatic GSH depletion led to insulin resistance of the same magnitude of that observed after inhibition of hepatic NOS. These results support the hypothesis that hepatic GSH is crucial in peripheral insulin action. Since, in the fasted state, both hepatic GSH and NO levels are low, we tested the hypothesis that intraportal o-administration of a GSH donor and an NO donor enhances insulin sensitivity in fasted Wistar rats, by restoring HISS secretion. We observed that GSH and NO increased insulin sensitivity in a GSH dose-dependent manner. We concluded that HISS secretion requires elevated levels of both GSH and NO in the liver. GSH and NO react to form a S-nitrosothiol, S-nitrosoglutathione (GSNO). The last hypothesis tested in this work was that HISS secretion/ action depends on the formation of GSNO. We observed that intravenous administration of -nitrosothiols (RSNOs) increased insulin sensitivity in animals fasted for 24 h, in contrast with the intraportal administration of the drug. This result suggests that RSNOs enhanced insulin sensitivity through a peripheral, and not hepatic, mechanism. The results obtained led to a restructuring of the HISS hypothesis, suggesting that the ingestion of a meal triggers the hepatic parasympathetic nerves, leading to the release of Ach in the liver, which in turn activates NOS. Simultaneously, hepatic GSH levels increase and react with NO to form a nitrosated compound, GSNO. S-nitrosoglutathione mimics HISS hypoglycaemic action at the skeletal muscle.

Se realizó un estudio seroepidemiológico de Toxoplasma gondii en cuatro Municípios de la Provincia La Habana en el periodo comprendido entre el mes de Octubre de 1990 y Abril de 1991, utilizando una técnica de ultramicro ELISA de 10 µL. Se examinaron un total de 362 muestras de mujeres embarazadas y se obtuvo una prevalencia de infección de 71,0%. La infección toxoplásmica fue más frecuente en las embarazadas que residían en zona rural y convivian con gatos. Se estudió la relación entre la infección toxoplásmica y el antecedente de abortos espontáneos sin encontrar diferencias estadísticamente significativas.

In order to learn the prevalence of Chagas' infection among students from Santa Cruz de la Sierra's universities, a random sample of 372 new students was drawn. All participants have had electrocardiograms (EKG) and serologic analysis (IHAT). 64/372 (17.2%) had serologic evidence of Chagas' infection, and from those, 10/64 (15.6%) had some EKG alterations. Among students presenting negative serologic test, 31/308 (10.1%) had EKG alterations. There was no statistical association between Chagas' infection and EKG alterations (X2=1.67, p=0.2). There was a positive association between Chagas' infection and intraventricular conduction defects and this association was higher among the students of 19 years of age or less (O.R. 10.4, p<0.05).

Se comunica el hallazgo de Dracunculus sp. en un canino proveniente de la localidad de Fontana, Departamento Patiño, Provincia de Formosa, Argentina. El hallazgo constituye el cuarto caso de Dracunculosis registrado en animales en una misma área geográfica de Formosa.

Este texto parte de la caracterización del concepto de multiculturalismo en general y específicamente en el contexto de la UE. A continuación, intentamos demostrar el carácter multicultural de una red de información, la red de los CDE, presente en todo el território de la EU, a partir de los resultados de encuestas distribuídas a los responsables, a sus utilizadores y en base a los datos recogidos durante visitas a algunos de ellos. Efectivamente, se verifica la existência de diferencias substanciales Finlandia, Irlanda, Hungría y Portugal en lo que respecta a la localización de los CDE, las tipologías de utilizadores, al uso de lenguages sistemáticos y alfabéticos para la organización de la información. Estas diferencias son menos marcadas pero también existen en lo que respecta a las áreas temáticas mas buscadas y los aspectos que los utilizadores de los CDE consideran que facilitan su acceso a la información. Por lo que se refiere a las formas de búsqueda en el catálogo y a los aspectos que se apuntan como obstáculos en el acceso a la información parece haber más coincidencia entre los diferentes países.

En este artículo tratamos la relación interdisciplinar entre las Ciencias de la Documentación y las Ciencias de la Educación, principalmente en lo que se refiere al papel que la alfabetización informacional puede desempeñar en la ejecución de modelos educativos orientados al currículum integrado o al aprendizaje basado en recursos, teniendo en cuenta el desarrollo de competencias transdisciplinares necesarias para el aprendijaze a lo largo de la vida. Intentamos demostrar que la alfabetización informacional presenta una relación muy estrecha con el aprendizaje, ya que una persona competente, en términos de información és aquélla que ha aprendido a aprender, sabe encontrar la información, sabe utilizar la información y sabe cómo organizarla. En este sentido, presentamos un estudio de caso, centrado en el análisis de las actitudes, de los hábitos de lectura y de las prácticas informaciónales de los alumnos del municipio de Vila do Conde, con base en la utilización de las collecciones disponibles en el espacio de la biblioteca escolar y de los recursos tecnológicos, principalmente las formas de localización y uso de los documentos de libre acceso, así como la busquéda de información. Presentamos una breve caracterización generale de esta red municipal de bibliotecas escolares, pero centramos el análisis del universo de la investigación en tres escuelas del 1º Ciclo, una EB 2,3 y una Secundaria, con la aplicación de una encuesta. Para una mejor contextualización de nuestro estudio haciemos también la caracterizacion y el encuadrimiento de cada biblioteca escolar en el medio en que está inserto (urbano, rural y pesquero), a fin de realizar puntualmente un análisis comparativo de los resultados.

This paper addresses the issue of restructuring the information professional in the global economy. This restructuring is justified by a tripartite explanation: the redesign understood as innovation and disciplinary change, the reorganization of institutions conditioned by the development of information technology and economic deflation interpreted as eliminating jobs. To understand this restructuring scenario multigenerational work contexts information services are also addressed. These work contexts represent a search for understanding and to overcome common referential differences, by baby boomer practitioners and digital natives. The issue of tension and challenge between the specific technical skills and flexibility to adapt to ever-changing organizations is another topic for reflection and serves to point out new paths for the restructuring of the information professional.

Evaluation of TNF-alpha in patients with Kala-azar has drawn increasing interest due to its regulatory role on the immune system, in addition to its cachetizing activity. The objective of this study was to examine the association between plasma levels of TNF-alpha, measured by immunore-activity (ELISA) and bioactivity (cytotoxicity assay with L-929 cells), and clinical manifestations of visceral leishmaniasis. Plasma samples from 19 patients with Kala-azar were obtained before, during and at the end of antimonial therapy. TNF-alpha determinations was done by using the cytotoxicity assay (all patients) and the enzyme-linked immunoassay (ELISA - 14 patients). A discrepancy between results obtained by ELISA and cytotoxicity assay was observed. Levels of circulating TNF-alpha, assessed by ELISA, were higher in patients than in healthy controls, and declined significantly with improvement in clinical and laboratory parameters. Plasma levels before treatment were 124.7 ± 93.3 pg/ml (mean ± SD) and were higher than at the end of therapy 13.9 ± 25.1 pg/ml (mean ± SD) (p = 0.001). In contrast, plasma levels of TNF-alpha evaluated by cytotoxicity assay did not follow a predicted course during follow-up. Lysis, in this case, might be not totally attributed to TNF-alpha. The discrepancy might be attributed to the presence of factor(s) known to influence the release and activity of TNF-alpha.

A 73 year-old white male, living in the interior of the state of Mato Grosso do Sul, in central Bra­zil, after an initial diagnosis of sinusitis was transferred to the neurology service with a 3-day evolution of intracranial hypertension. Exams showed lymphocytic leukemia and a tumor-like lesion, either an expanding inflammatory process such as an abscess or a neoplasm. Treatment with Ceftriaxone and Decadron was started and intracranial hypertension was controlled. Methotrexate was injected on the occasion of the next puncture considering a possible leukemia infiltration. Flagellate forms of T. cruzi were observed in the CSF and treatment with Benznidazole was started. After 4 days the CSF presented fractionated forms of trypomastigotes. The protein level was 27%. Signs of intracranial hypertension ceased. Tomography and magnetic resonance images showed an important reduction of the tumor-like lesion. The clinical condition of the patient improved.

Experimental murine L. major infection is characterized by the expansion of distinct CD4+ T cell subsets. The Th1 response is related to production of IFN-g and resolution of infection, whereas Th-2 response with production of IL-4 and IL-10 and dissemination of infection. The objective of this study was to measure the circulating levels of IFN-g, IL-10 and TNF-a in patients with visceral leishmaniasis (VL) before, during and at the end of therapy and to examine the association between cytokine levels and activity of VL. Fifteen patients with VL were evaluated. The cytokine determinations were done by using the enzyme-linked immunoassay (ELISA) before, during and at the end of therapy. At baseline, we detected circulating levels of IFN-g in 13 of 15 patients (median = 60 pg/ml); IL-10 in 14 of 15 patients (median = 141.4 pg/ml); and TNF-a in 13 of 14 patients (median = 38.9 pg/ml). As patients improved, following antimonial therapy, circulating levels of IL-10 showed an exponential decay (y = 82.34 e–0,10367x, r = –0.659; p < 0.001). IFN-g was no longer detected after 7/14 days of therapy. On the other hand, circulating levels of TNF-a had a less pronounced decay with time on therapy, remaining detectable in most patients during the first seven days of therapy (y = 36.99-0.933x, r = –0.31; p = 0.05). Part of the expression of a successful response to therapy may, therefore, include reduction in secretion of inflammatory as well as suppressive cytokines. Since IL-10 and IFN-g are both detected prior to therapy, the recognized cellular immune depression seen in these patients may be due to biological predominance of IL-10 (type 2 cytokine), rather than lack of IFN-g (type 1 cytokine) production.
