Inflammation is an immune complex-related tissue damage and / or cell caused by chemical, physical, immunological or microbial. The inflammatory process involves a complex cascade of biochemical and cellular events, including awareness and receptor activation, lysis and tissue repair. In general, tissue damage trigger a local inflammatory response by recruiting leukocytes, which release inflammatory mediators. These substances are able to sensitize nociceptors. After synaptic transmission and signal modulation by nociceptive sensory neurons, these signals are perceived as pain. Pain is an experience that involves multiple factors. The route of the supraspinal pain control originates in many brain regions, such as substance periarquedutal gray (PAG), median raphe nucleus and rostral ventromedial medulla (RVM) and have a critical role in determining the chronic and acute pain. Anti-inflammatory drugs (NSAIDs) are used to control inflammation, which inhibit the inflammatory mediators, but can cause side effects such as stomach ulcers and cardiovascular damage. An alternative for the treatment of pain and inflammation is the use of plant species. The genus Eugenia belongs to the family Myrtaceae, one of the largest botanical families of expression in the Brazilian ecosystems. From the pharmacological point of view, studies of similar species crude extracts showed the presence of anti-inflammatory, analgesic, antifungal, hypotensive, antidiabetic and antioxidant activity of some species. As a class of importance in therapeutic phytochemical, the flavonoids has represented an important group with significant anti-inflammatory and gastroprotective, and are present in a significant way in the chemical composition of genus Eugenia. Introduction: Tuberculosis (TB) is a granulomatous disease caused by Mycobacterium tuberculosis. The genus Mycobacteriumhas two different complexes: M. tuberculosis Complex and M. avium Complex. This is a global health epidemic and remains a major global health problem, besides, the clinical severity of TB is significantly higher in transplanted patients. The detection of these mycobacteria complexes in transplanted patients, by molecular methods, is fundamental for quick treatment of patients and can contribute for rapid and accuracy of diagnosis. Objective: To detect mycobacteria DNA of M. tuberculosis and M. avium Complexes in formalin fixed paraffin-embedded samples (FFPE) of two patients groups: non transplanted and transplanted. Materials and Methods: The study includes 40 FFPE biopsies separated in four groups: NTP – presence of epithelioid granuloma and positive ZN, non-transplanted patients – 9 samples; NTN - presence of epithelioid granuloma and negative ZN, non-transplanted patients – 10 samples; TP – positive ZN, transplanted patients – 9 samples; TN – negative ZN, transplanted patients – 7 samples. Sections were cut for DNA extraction. Samples were submitted to PCR for amplification of: a) β-actin, b) IS6110 insertion and c) IS1245 insertion. DNA evaluation was made by spectrophotometry and efficiency and PCR analysis was made by agarose gels under UV light. Results: In all samples processed, 97.1% were positive for human β-actin gene. In22.2% of NTP group were found the IS6110 insertion sequencebut the IS1245 wasn´t. In the NTN group was not found any sequence. In theTP group, 11.1% of the samples were positive for IS6110 and also 11,1% werepositive for IS1245. In the TN group, 14.3% of the samples were positive forIS6110 and for IS1245, 14.3% was also positive. Maternal undernutrition affects the foetal development, promoting renal alterations and adult hypertension. The present study investigates, in adult male rats, the effect of food restriction in utero on arterial blood pressure changes (AP), and its possible association with the number of nephrons, renal function and angiotensin II (AT1R/AT2R), glucocorticoid (GR) and mineralocorticoid (MCR) receptors expression. The daily food supply to pregnant rats was measured and one group (n=5) received normal quantity of food (NF) while the other group received 50% of that (FR50) (n=5). The AP was measured weekly. At 16 weeks of life, fractionator’s method was used to estimate glomeruli number in histological slices. The renal function was estimate by creatinine and lithium clearances. Blood and urine samples were collected to biochemical determination of creatinine, sodium, potassium and lithium. At 90th and 23rd days of life, kidneys were also processed to AT1R, AT2R, GR and MCR immunolocalization and for western blotting analysis. FR50 offspring shows a significant reduction in BW (FR50: 5.67 ± 0.16 vs. 6.84 ± 0.13g in NF, P<0.001) and increased AP from 6th to 12nd week (6thwk FR50: 149.1 ± 3.4 vs. 125.1 ± 3.2mmHg in NF, P<0.001and, 12ndwk FR50: 164.4 ± 4.9 vs. 144.0 ± 3.3 mmHg in NF, P=0.02). Expression of AT1R and AT2R were significantly decreased in FR50 (AT1, 59080 ± 2709 vs. 77000 ± 3591 in NF, P=0.05; AT2, 27500 ± 95.50 vs. 67870 ± 1509 in NF, P=0.001) while the expression of GR increased in FR50 (36090 ± 781.5 vs. 4446 ± 364.5 in NF, P=0.0007). The expression of MCR did not change significantly. We also verified a pronounced decrease in fractional urinary sodium excretion in FR50 offspring (0.03 ± 0.02 vs. 0.06 ± 0.04 in NF, p=0.03). A silimarina é um flavonóide polifenólico extraído de frutos e sementes de Silybum marianum, que possui efeitos antiinflamatórios, citoprotetores e anticarcinogênicos. O objetivo deste trabalho foi avaliar o efeito antiinflamatório desse flavonóide sobre linhagens geneticamente selecionadas para máxima e mínima resposta inflamatória aguda, a intensidade dessa resposta foi avaliada através dos seguintes parâmetros: contagem total e diferencial de células, concentração protéica do exsudato infamatório e dosagem de H2O2 através de cultura de células do infiltrado. Para a avaliação do efeito da silimarina, as linhagens foram divididas em dois grupos, grupo AIRmax e grupo AIRmin, e cada um desses grupos foi subdividido em outros três grupos, de acordo com a forma de tratamento determinada (grupo controle, grupo tratado com silimarina 200mg/kg e grupo tratado com silimarina 100mg/kg). Os animais foram tratados durante seis dias consecutivos, recebendo um total de seis doses; ao final do tratamento os animais foram submetidos à indução flogógena pelo composto BIOGEL-P100 por via subcutânea, ao final de 24 horas o exsudato inflamatório foi retirado do dorso dos animais e preparados para análise. Os resultados demonstraram que a silimarina afetou significativamente alguns dos parâmetros avaliados. A contagem total de células mostrou redução significativa no número de células presentes no infiltrado do grupo AIRmax tratado com silimarina 100mg/kg em relação ao seu grupo controle; na contagem diferencial de células houve redução significativa no número de neutrófilos encontrados em amostras do exsudato dos animais AIRmax tratados com silimarina 100mg/kg, assumindo um padrão de resposta semelhante ao da contagem total; a concentração protéica mostrou redução significativa nos grupos AIRmax tratados com 100 e 200mg/kg em relação ao seu controle, indicando um possível efeito modulador da silimarina

A Doença Inflamatória Intestinal (DII) engloba, fundamentalmente, duas doenças distintas: a Doença de Crohn (DC) e a Colite Ulcerativa (CU), ambas caracterizadas por uma inflamação crônica do intestino, com períodos de exacerbação seguidos de intervalos prolongados de remissão dos sintomas. Apesar da DII ser objeto de pesquisa há várias décadas, a sua etiologia ainda é desconhecida e seu tratamento envolve uma série de fármacos com sérios efeitos colaterais, especialmente quando usado cronicamente. As cumarinas representam uma importante classe de compostos fenólicos naturais que apresentam inúmeras propriedades farmacológicas importantes para que um produto seja potencialmente ativo no tratamento da DII. Dentre as inúmeras cumarinas conhecidas e estudadas, dados obtidos em nosso laboratório demonstram que algumas cumarinas apresentam efeitos protetores na fase aguda e preventiva do processo inflamatório intestinal induzido por TNBS (ácido trinitrobenzenosulfônico) em ratos, reduzindo a incidência de diarréia, aderência e obstrução intestinal, além de manterem os níveis de glutationa e reduzirem a atividade da fosfatase alcalina e da mieloperoxidase colônica. Os objetivos do presente projeto são determinar a atividade antiinflamatória intestinal da escopoletina e fraxetina na fase aguda e crônica com recidiva de colite ulcerativa induzida por TNBS tendo-se em vista a determinação de sua atividade curativa. Para tanto, serão avaliados os parâmetros clínicos gerais (consumo de alimento e peso corporal), parâmetros macroscópicos da lesão (determinação de índice de lesão, relação peso-comprimento colônico) e bioquímicos (avaliação das atividades da mieloperoxidase, fosfatase alcalina, níveis de proteínas e glutationa total)

It is understood by Chronic Renal Failure (CRF) or Stage IV Chronic Kidney Disease (CKD) the morbid state in which there is a substantial and irreversible loss of functional capacity, metabolic and endocrine function in both kidneys, leading to a framework of dehydration, azotemia, electrolyte imbalance, anemia, uremic syndrome, secondary hyperparathyroidism, among other conditions that make the patient's life without treatment unfeasible. An adequate nutritional support to patients with CKD at any stage aims to maintain a good clinical condition and an attempt to delay the progression of the disease. The basis of a diet for patients with CKD is the restriction in this protein being of high biological value, low-phosphorus, high energy density, fiber and antioxidants combined with a good dietary practices that allow the patient a good quality of life

The concentration of metal pollutants such as lead (Pb), has grown and developed in populated areas due to pollution and other human activities. Consequently, the potential for achieve this element food chain has also increased. Lead is very toxic to humans, especially to children, and exposure to lead can cause adverse health effects mainly on human nervous system, bone marrow and kidneys, interfering with chromosomal or genetic processes. This paper presents an overview of the main aspects related to environmental contamination by lead from battery plants. The assessment of an area contaminated by lead in Bauru-SP next a battery factory was reported in this work as well as the entire history of monitoring, classification and application of processes applied by CETESB since 2002. Analyzing the issue in its economic aspects, we found that the degradation of the environment is directly related to the development model adopted by the capitalist system, which is based on the law of supply and demand for products and services. The data presented indicate that Brazil still needs a broader policy where government agencies, industries and population through awareness can be united for the same purpose: to preserve life

Cholangiohepatitis is a common hepatic disorder in cats that is second only to hepatic lipidosis in frequency. The cholangitis-cholangiohepatitis complex is known as the diseases that involve the biliar tract inflammation and may extend to the liver. The classification of the cholangiohepatitis is still controversial, in this present study the following classification was used: Acute Cholangiohepatitis, Chronic Cholangiohepatitis, Lymphocitic Cholangitis. The cholangiohepatitis may be associated with pancreatitis and inflammatory bowel disease, this association is known as feline triaditis. The diagnosis is based on the clinical signs, alterations in laboratory exams and ultrasonography alterations, but the definitive diagnosis is only possible through liver biopsy. The treatment for each type of cholangiohepatitis differs, hence the importance of the right diagnosis. As infecções do trato urinário (ITU) são definidas como a invasão e posterior multiplicação de microorganismos nos tecidos das vias urinárias, sendo a bactéria gram-negativa E.coli o principal uropatógeno envolvido nessa infecção. A avaliação para a presença de ITU é feita através do quadro clínico característico desse tipo de paciente e o exame de cultura de urina. As amostras positivas para as culturas são avaliadas quanto ao perfil de sensibilidade aos antimicrobianos dos agentes etiológicos encontrados. Descrever a porcentagem de ocorrência dos agentes etiológicos e o perfil de sensibilidade aos antimicrobianos das Infecções do Trato Urinário em pacientes ambulatoriais de Botucatu e região. Foram incluídas neste estudo culturas de urinas de pacientes atendidos em UBS de Botucatu e região Pólo Cuesta, no período de Janeiro de 2008 a Maio de 2010. As culturas positivas foram avaliadas quanto a porcentagem de ocorrência dos uropatógenos segundo sexo, faixa etária e origem. Foi utilizado o teste do qui-quadrado para verificar associação entre cada antimicrobiano e as porcentagens de sensibilidade e resistência encontradas. Foram avaliadas 11.768 culturas de urina provenientes de 11 municípios da região, sendo consideradas as culturas positivas (28%). Quanto à ocorrência dos agentes etiológicos verificou-se que a E.coli é responsável 58,76% dos casos. A distribuição dos uropatógenos segundo sexo revelou maior ocorrência de E.coli nos dois sexos. O segundo agente etiológico de maior ocorrência foi, nos homens o Proteus mirabilis e nas mulheres, o S. saprophyticus. Dentre as faixas etárias, a E.coli foi o patógeno de maior ocorrência. O perfil de sensibilidade aos antimicrobianos revelou que a E.coli é mais sensível a Ertapenem, Imipenem, Meropenem, Fosfomicina, Amicacina, Ceftriaxona, Nitrofurantoína e Norfloxacina; e mais resistente... Preeclampsia is a specific disorder of pregnancy, characterized by arterial hypertension and proteinuria detected after 20 weeks of gestation. This pathology is associated with hyperuricemia, higher levels of pro-inflammatory cytokines, enhanced leukocyte activation and oxidative stress. Adenosine deaminase (ADA) is an enzyme present in all human tissues and, it is involved with the maturation of the immune system. Although its function is not fully understood, ADA is considered an indicator of cellular inflammation and, its increased serum concentration is observed in inflammatory diseases, such as tuberculosis and rheumatoid arthritis. This study aimed to assess serum ADA levels in preeclamptic patients (PE) compared with normotensive pregnant (NT) and non-pregnant women (NP), and to correlate these values with TNF-α and IL-1β production. Ninety pregnant women were included: 60 were pre-eclamptic and 30 were normotensive matched for gestational age. As control group 20 healthy non-pregnant women matched with pregnant for age were included. Peripheral blood mononuclear cells (PMMC) obtained from the three groups studied were cultured with or without lipopolysaccharide (LPS) for 18h at 37oC, and TNF-α and IL-1β production was assessed in the supernatant of cultures by enzyme immunoassay (ELISA). ADA plasmatic concentration was determined by colorimetric method. The results show that ADA plasma levels were significantly higher in PE group compared with NT and NP groups. A positive correlation between ADA and uric acid levels was detected in preeclamptic women. There was no significant difference in relation to ADA levels when PE patients were classified in early and late-onset PE. The endogenous production of IL-1β and TNF-α by PBMC was significantly higher in PE group than in NT and NP women, showing the activation state of these cells in PE. Pre-eclampsia (PE) is a complication of human pregnancy characterized by hypertension and proteinuria after 20 weeks of gestation. Its incidence varies from 5% to 7% of pregnancies and is a major cause of morbidity and maternal and fetal mortality. This is a multisystemic disease, with focus on vascular dysfunction and is closely related to the exacerbated activation of the immune system. In addition to increased activation of monocytes and granulocytes, there is an elevated production of proinflammatory cytokines in pregnant women with PE. The nuclear transcription factor-kB (NF-kB) is present in the cells of the immune system and is responsible for transcription of genes related to inflammation. Whereas the PE is associated with intense inflammatory response, the use of substances modulating the activity of NF-kB factor could be useful in alleviating the inflammation present in these patients. Silibinin is the main component of silymarin, a polyphenolic extract obtained from fruits and seeds of Sylibum marianum with potent hepatoprotective, anti-inflammatory and anti-fibrotic activities. The silibinin mechanism of action includes the ability to inhibit NF-kB activation and, consequently, its migration to the nucleus. The objective of this study was to assess whether silibinin modulates the activity of NF-kB and the production of inflammatory cytokines in mononuclear cells of patients with PE. We evaluated 34 pregnant women with PE, 20 normotensive pregnant women (GN) and 15 non-pregnant women (NG). Peripheral blood mononuclear cells (PBMC) were obtained from those groups of women and cultured in the presence or absence of silibinin (5 uM or 50 uM) and stimulated or not with lipopolysaccharide (LPS) for 18 h to obtain supernatant for determination of tumor necrosis factor-alpha (TNF-α) and interleukin-1 (IL-1β) by enzyme immunoassay (ELISA). Uremic syndrome, arising from kidney malfunction, consists of a set of systemic changes caused by the accumulation of toxic substances to the body. Since, with the advance of medicine, the animals lived more reaching advanced age and entering thus on track of senility, chronic renal disease, became a common complaint in the routine of the ambulatory. This high rate of morbidity generates an increased need for clarification of pathophysiology involved in this disease. The components of the uremic syndrome include water and sodium imbalance, anemia, intolerance to carbohydrate, neurological disorders, disorders of the gastrointestinal tract, osteoarthritis, immunological incompetence and metabolic acidosis. The clinical manifestations occur in isolation or in combination. In most cases canines patients are subject to an assessment when the kidney disease has evolved to the final stage with uremic syndrome and installed already, under these conditions, the prognosis is reserved

O câncer tem sido alvo de incessantes pesquisas sobre sua etiologia, desenvolvimento, progresso e tratamentos. Sua importância no campo científico se dá pela sua alta taxa de mortalidade e morbidade. O fato de que seja uma doença genética, que pode ter interferência ambiental e dietética, está cada vez mais elucidado, porém ainda existem muitos mecanismos a serem desvendados. As neoplasias ósseas, benignas ou malignas, e processos inflamatórios ósseos acometem desde crianças até adultos e idosos, podendo causar danos físicos, incapacidade motora e até a morte. Portanto, presumir o potencial de transformação maligna das lesões benignas, agressividade tumoral, capacidade de invasão tecidual, probabilidade de recidiva, propensão ao desenvolvimento de metástases e resposta ao tratamento, é um valioso expediente na escolha da proposta terapêutica. Estudos genéticos e citogenéticos têm ajudado a aumentar o entendimento sobre a carcinogênese, progressão tumoral, prognósticos e diagnósticos. Portanto, este estudo teve como objetivo detectar mutações e marcadores cromossômicos consistentes e recorrentes na transformação de tumores benignos em malignos no sistema musculoesquelético, através de análises com citogenética clássica, e relacionar estes achados com o prognóstico e diagnóstico dos pacientes. Dentre os casos coletados e analisados citogeneticamente, foram selecionados três casos de lesões ósseas benignas e três lesões ósseas malignas tidas como a progressão tumoral dos respectivos casos benignos, para discussão e relato de caso. Os achados citogenéticos relataram consistentes alterações numéricas e estruturais clonais em regiões cromossômicas com genes importantes envolvidos na progressão tumoral. Pode-se citar a perda da região contendo o gene TP53, supressor tumoral, tanto em lesões benignas como em maligna, como um dos achados mais relevantes neste estudo. Dessa forma, os resultados ...

As espécies reativas de oxigênio podem participar de mecanismos envolvidos em respostas fisiológicas, como os efeitos hipertensores e dipsogênicos da angiotensina II (ANG II) central, que seriam pelo menos em parte devidos a produção de radical superóxido (O2 −). Por isso, uma superexpressão da enzima superóxido dismutase que reduz a disponibilidade de O2 − convertendo-o em peróxido de hidrogênio (H2O2) inibiria o efeito pressor da ANG II administrada centralmente. Porém, alguns resultados de nosso laboratório demonstram que H2O2 injetado intracerebroventricularmente (icv) reduz a ingestão de água induzida pela ANG II em ratos normotensos. Portanto, no presente trabalho propomos estudar os efeitos de injeções de H2O2 ou de ATZ (inibidor da catalase) sozinhos ou combinados no ventrículo lateral (VL) e também os efeitos do ATZ injetado intravenosamente (i.v.) sozinho ou combinado com H2O2 no VL sobre a resposta pressora da ANG II também injetada no VL em ratos normotensos, espontaneamente hipertensos (SHRs) ou em ratos com hipertensão renovascular do tipo 2 rins 1 clipe (2R1C). Foram utilizados ratos Holtzman normotensos, SHRs e ratos com hipertensão renovascular 2R1C com cânulas de aço inoxidável previamente implantadas no VL. A pressão arterial e a freqüência cardíaca foram registradas em sistema computadorizado diretamente da aorta em ratos não anestesiados por uma cânula de polietileno introduzida pela artéria femoral e a injeção i.v. foi realizada por meio de uma cânula de polietileno introduzida na veia femoral. A pressão arterial e freqüência cardíaca foram registradas continuamente desde 30 minutos antes da primeira injeção (veículo, salina, H2O2 ou ATZ) até 30 minutos após a injeção de ANG II.
