In modern magnetic resonance imaging (MRI), patients are exposed to strong, nonuniform static magnetic fields outside the central imaging region, in which the movement of the body may be able to induce electric currents in tissues which could be possibly harmful. This paper presents theoretical investigations into the spatial distribution of induced electric fields and currents in the patient when moving into the MRI scanner and also for head motion at various positions in the magnet. The numerical calculations are based on an efficient, quasi-static, finite-difference scheme and an anatomically realistic, full-body, male model. 3D field profiles from an actively shielded 4T magnet system are used and the body model projected through the field profile with a range of velocities. The simulation shows that it possible to induce electric fields/currents near the level of physiological significance under some circumstances and provides insight into the spatial characteristics of the induced fields. The results are extrapolated to very high field strengths and tabulated data shows the expected induced currents and fields with both movement velocity and field strength. Influence of magnetically-induced E-fields on cardiac electric activity during MRI: A modeling study

In modern magnetic resonance imaging (MRI), patients are exposed to strong, time-varying gradient magnetic fields that may be able to induce electric fields (E-fields)/currents in tissues approaching the level of physiological significance. In this work we present theoretical investigations into induced E-fields in the thorax, and evaluate their potential influence on cardiac electric activity under the assumption that the sites of maximum E-field correspond to the myocardial stimulation threshold (an abnormal circumstance). Whole-body cylindrical and planar gradient coils were included in the model. The calculations of the induced fields are based on an efficient, quasi-static, finite-difference scheme and an anatomically realistic, whole-body model. The potential for cardiac stimulation was evaluated using an electrical model of the heart. Twelve-lead electrocardiogram (ECG) signals were simulated and inspected for arrhythmias caused by the applied fields for both healthy and diseased hearts. The simulations show that the shape of the thorax and the conductive paths significantly influence induced E-fields. In healthy patients, these fields are not sufficient to elicit serious arrhythmias with the use of contemporary gradient sets. However, raising the strength and number of repeated switching episodes of gradients, as is certainly possible in local chest gradient sets, could expose patients to increased risk. For patients with cardiac disease, the risk factors are elevated. By the use of this model, the sensitivity of cardiac pathologies, such as abnormal conductive pathways, to the induced fields generated by an MRI sequence can be investigated. The effects of the native alpha-conotoxin PnIA, its synthetic derivative [ A10L] PnIA and alanine scan derivatives of [ A10L] PnIA were investigated on chick wild type alpha7 and alpha7-L247T mutant nicotinic acetylcholine receptors (nAChRs) expressed in Xenopus oocytes. PnIA and [A10L] PnIA inhibited acetylcholine (ACh)-activated currents at wtalpha7 receptors with IC50 values of 349 and 168 nM, respectively. Rates of onset of inhibition were similar for PnIA and [ A10L] PnIA; however, the rate of recovery was slower for [ A10L] PnIA, indicating that the increased potency of [ A10L] PnIA at alpha7 receptors is conveyed by its slower rate of dissociation from the receptors. All the alanine mutants of [ A10L] PnIA inhibited ACh-activated currents at wtalpha7 receptors. Insertion of an alanine residue between position 5 and 13 and at position 15 significantly reduced the ability of [ A10L] PnIA to inhibit ACh-evoked currents. PnIA inhibited the non-desensitizing ACh-activated currents at alpha7-L247T receptors with an IC50 194 nM. In contrast, [ A10L] PnIA and the alanine mutants potentiated the ACh-activated current alpha7-L247T receptors and in addition [ A10L] PnIA acted as an agonist. PnIA stabilized the receptor in a state that is non-conducting in both the wild type and mutant receptors, whereas [ A10L] PnIA stabilized a state that is non-conducting in the wild type receptor and conducting in the alpha7-L247T mutant. These data indicate that the change of a single amino acid side-chain, at position 10, is sufficient to change the toxin specificity for receptor states in the alpha7-L247T mutant.

The HERG K+ channel has very unusual kinetic behavior that includes slow activation but rapid inactivation. These features are critical for normal cardiac repolarization as well as in preventing lethal ventricular arrhythmias. Mutagenesis studies have shown that the extracellular peptide linker joining the fifth transmembrane domain to the pore helix is critical for rapid inactivation of the HERG K+ channel. This peptide linker is also considerably longer in HERG K+ channels, 40 amino acids, than in most other voltage-gated K+ channels. In this study we show that a synthetic 42-residue peptide corresponding to this linker region of the HERG K+ channel does not have defined structural elements in aqueous solution; however, it displays two well defined helical regions when in the presence of SDS micelles. The helices correspond to Trp(585)-Ile(593) and Gly(604)-Tyr(611) of the channel. The Trp(585)-Ile(593) helix has distinct hydrophilic and hydrophobic surfaces. The Gly(604)-Tyr(611) helix corresponds to an N-terminal extension of the pore helix. Electrophysiological studies of HERG currents following application of exogenous S5P peptides show that the amphipathic helix in the S5P linker interacts with the pore region of the channel in a voltage-dependent manner.

The origin of intracellular Ca2+ concentration ([Ca2+](i)) transients stimulated by nicotinic ( nAChR) and muscarinic ( mAChR) receptor activation was investigated in fura-2-loaded neonatal rat intracardiac neurons. ACh evoked [Ca2+](i) increases that were reduced to similar to 60% of control in the presence of either atropine ( 1 muM) or mecamylamine ( 3 muM) and to < 20% in the presence of both antagonists. Removal of external Ca2+ reduced ACh-induced responses to 58% of control, which was unchanged in the presence of mecamylamine but reduced to 5% of control by atropine. The nAChR-induced [Ca2+](i) response was reduced to 50% by 10 μM ryanodine, whereas the mAChR-induced response was unaffected by ryanodine, suggesting that Ca2+ release from ryanodine-sensitive Ca2+ stores may only contribute to the nAChR-induced [Ca2+](i) responses. Perforated-patch whole cell recording at - 60 mV shows that the rise in [Ca2+](i) is concomitant with slow outward currents on mAChR activation and with rapid inward currents after nAChR activation. In conclusion, different signaling pathways mediate the rise in [Ca2+](i) and membrane currents evoked by ACh binding to nicotinic and muscarinic receptors in rat intracardiac neurons.

Di-2-pyridyl ketone isonicotinoyl hydrazone (HPKIH) and a range of its analogues comprise a series of monobasic acids that are capable of binding iron (Fe) as tridentate (N,N,O) ligands. Recently, we have shown that these chelators are highly cytotoxic, but show selective activity against cancer cells. Particularly interesting was the fact that cytotoxicity of the HPKIH analogues is maintained even after complexation with Fe. To understand the potent anti-tumor activity of these compounds, we have fully characterized their chemical properties. This included examination of the solution chemistry and X-ray crystal structures of both the ligands and Fe complexes from this class and the ability of these complexes to mediate redox reactions. Potentiometric titrations demonstrated that all chelators are present predominantly in their charge-neutral form at physiological pH (7.4), allowing access across biological membranes. Keto-enol tautomerism of the ligands was identified, with the tautomers exhibiting distinctly different protonation constants. Interestingly, the chelators form low-spin (diamagnetic) divalent Fe complexes in solution. The chelators form distorted octahedral complexes with Fe-II, with two tridentate ligands arranged in a meridional fashion. Electrochemistry of the Fe complexes in both aqueous and non-aqueous solutions revealed that the complexes are oxidized to their ferric form at relatively high potentials, but this oxidation is coupled to a rapid reaction with water to form a hydrated (carbinolamine) derivative, leading to irreversible electrochemistry. The Fe complexes of the HPKIH analogues caused marked DNA degradation in the presence of hydrogen peroxide. This observation confirms that Fe complexes from the HPKIH series mediate Fenton chemistry and do not repel DNA. Collectively, studies on the solution chemistry and structure of these HPKIH analogues indicate that they can bind cellular Fe and enhance its redox activity, resulting in oxidative damage to vital biomolecules.

A partir dos avan??os alcan??ados desde 1998 no Brasil, conclui-se pela necessidade do aprofundamento da profissionaliza????o das estruturas de comissionamento, tendo em vista a possibilidade de que uma estrutura menos vulner??vel ??s injun????es da pol??tica clientelista possa melhor atender aos princ??pios da efici??ncia e da transpar??ncia.

A Baía de Vitória é um sistema estuarino da costa leste do Brasil que apresenta grande importância econômica e características ímpares, já que se desenvolveu entre três unidades geomorfológicas distintas. Apesar disto, este ambiente apresenta poucos estudos relacionados às suas características geológicas e oceanográficas. O presente trabalho se focou no preenchimento desta lacuna através da descrição da morfologia, da distribuição sedimentar e dos padrões sonográficos da Baía de Vitória, buscando relações entre os mesmos e inferindo os processos sedimentares dominantes em cada trecho do estuário. A análise integrada mostrou boa correlação entre os métodos e revelou a grande complexidade deste sistema estuarino. Três diferentes regiões foram identificadas mostrando processos distintos: o estuário superior apresentou input sedimentar fluvial e processos estuarinos; um largo trecho da região central do sistema apresentou características erosivas, relacionadas a adaptações morfológicas e ao aumento das correntes provocadas pelos estreitamentos artificiais da baía, propiciando a formação de formas de fundo; e a boca do estuário se apresentou dominada por processos marinhos. Parte da região central do estuário mostrou características mascaradas pela atividade antrópica através da instalação de pontes, aterros e dragagens.

Levantamentos sonográficos desenvolvidos em duas áreas da plataforma interna adjacente ao estado do Espírito Santo revelam formas de fundo cuja origem é fortemente relacionada a eventos de tempestades. Estas são caracterizadas por alternâncias de bandas de areias finas sobrepostas, de forma alternada, a um fundo de areias grossas, gerando manchas de areias grossas intercaladas abruptamente com faixas de areias finas a profundidades de 25-30 m para GUA e de 05-08 m para BES. Em ambas as áreas as faixas de areias grossas apresentam marcas de ondulação geradas por ondas com orientação levemente paralela a linha de costa. A sedimentologia de GUA é composta por areias finas a muito finas lamosas carbonáticas com cascalhos siliciclásticos e a de BES por areias grossas e médias com cascalhos biodetríticos. Desta forma, é clara a ação de correntes/ondas de tempestade no fundo marinho de GUA e de BES; porém, a classificação quanto ao comportamento hidrodinâmico e ao transporte de sedimento é necessária para as estimativas sobre a formação, transporte e manutenção das feições encontradas.

A teoria dos atos de fala, de John Austin, apresenta a linguagem a partir de uma perspectiva pragmática, ou seja, quando os signos têm um compromisso com a produção de realidades, de mundo e de sujeitos. A linguagem possui – nessa perspectiva – força de criação. Em sintonia com os estudos da pragmática da linguagem, defenderemos a ideia que uma classificação não é mera descrição neutra acerca dos fatos, e Ian Hacking será a ponte entre a linguagem (enquanto fonte de produção de mundo) e as classificações (como produtoras de sujeitos). Para esse autor, as classificações tanto transformam quanto são transformadas. Elas, de diversas formas, interagem entre si, com os sujeitos, com as instituições, com os saberes, enfim, com tudo aquilo a que faz referência. A partir de uma revisão, análise e composição bibliográfica, o trabalho se dedicará a produzir um elo entre a problemática da linguagem-ato, tal como postulada por John Austin, e o caráter produtor de realidade das classificações. Dentre as classificações, daremos importância particular ao diagnóstico (enquanto uma espécie de estudo de caso na teoria dos enunciados) e seus efeitos pragmáticos, a partir dos elementos que lhe dão força de eficácia e existência em nosso mundo contemporâneo, tendo em vista o aumento do número e da proliferação de categorias nosológicas no corpo social. Trataremos, em particular, as classificações da medicina psiquiátrica. Abordaremos, por um lado, as condições que contribuem na emergência e na potência de um diagnóstico e, por outro, seus efeitos. Dentre os efeitos que daremos importância, encontramos a formação de novos sujeitos e modos de vida. Sendo assim, tanto as condições de eficácia de um enunciado quanto seus efeitos são estendidos à noção de diagnóstico enquanto uma classificação numa condição sociohistórica particular.
