A descontaminação dos explantes é um dos princípios básicos para o sucesso da cultura de tecidos. Um dos problemas diagnosticados na propagação in vitro da figueira, através de gemas apicais, é a contaminação endógena dos explantes por bactérias. Este trabalho teve como objetivo avaliar a eficiência de alguns antibióticos em meio de cultura para o controle de bactérias endógenas em gemas apicais de figueira. Foram avaliados os seguintes tratamentos: T1(sem adição de antibiótico); T2 (30 mg L-1 de cloranfenicol); T3 (250 mg L-1 de ampicilina sódica); T4 (500 mg L-1 de ácido nalidícico); T5 (150 mg L-1 de cefalotina sódica); T6 (500 mg L-1 de tetraciclina), e T7 (400 mg L-1 de norfloxacina). Após coletados em campo, os segmentos de ramos contendo as gemas foram colocados em recipiente com água corrente. Posteriormente, as gemas apicais foram imersas em álcool etílico a 70% e hipoclorito de sódio a 2,5%. Todo procedimento de desinfestação externa dos explantes foi realizado em câmara de fluxo laminar. Os explantes foram inoculados em tubos de ensaio contendo 15 mL de meio básico MS suplementado, após a autoclavagem, com as doses de antibióticos de acordo com os tratamentos estabelecidos. Após a inoculação, os explantes foram mantidos em sala de crescimento por quatro dias no escuro e, em seguida, sob fotoperíodo de 16 horas de luz branca fria e irradiância de 25 µmol m s-1, na temperatura de 22 ± 3ºC. A assepsia realizada externamente nos explantes foi suficiente para o controle de contaminação fúngica, e a adição de antibióticos ao meio, após autoclavagem, foi eficiente para o controle de bactérias endógenas, cujo antibiótico ampicilina sódica proporcionou mais de 90% de explantes sobreviventes.

Different solvents were evaluated for the extraction of jabuticaba anthocyanin pigments, identifying, quantifying and verifying the stability of the anthocyanins, as well as the conduction of three antioxidant activity assays and determination of the vitamin C levels. The maceration with ethanol acidified with HCl 1.5 mol L-1 (85:15) provides better pigment extraction and stability. The skin is anthocyanin rich, presenting 1.59 and 2.06 g 100 g-1 of dry matter in the Paulista and Sabará varieties, respectively. Cyanidin 3-glucoside is the majority pigment of the skins, followed by delphinidin 3-glucoside. The highest level of vitamin C was found in the skins and seeds of both varieties. It was verified that the skins, presented more antioxidant activity, in free radical capture, as well as in retarding the lipid oxidation process.

The close relationship between the chlorophyll-meters readings and the total chlorophyll and nitrogen contents in leaves, has allowed their evaluation both in annual and perennial species. Besides, some physiological events such as the CO2 assimilation have also been estimated by chlorophyll meters. This work was carried out aiming to evaluate the gas exchanges of peach palms as a function of the chlorophyll SPAD-Meter readings. Three year-old peach palms from Yurimaguas, Peru were studied in Ubatuba, SP, Brazil, spaced 2 x 1 m in area under a natural gradient of organic matter which allowed four plots to be considered, according to the peach palms leaves colors, from light yellow to dark green. The SPAD readings and the stomatal frequency of leaflets were evaluated. The photosynthetic photon flux density (PPFD, μmol m-2 s-1), the leaf temperature (Tleaf, ºC), the CO2 assimilation (A, μmol m-2 s-1), the stomatal conductance (g s, mol m-2 s-1), the transpiration (E, mmol m-2 s-1) and the intercellular CO2 concentration (Ci, μmol mol-1) were evaluated with a portable infrared gas analyzer (LCA-4, ADC BioScientific Ltd., Great Amwell, U.K.). A linear increase in the CO2 assimilation as a function of the SPAD readings (y = -0.34 + 0.19x, R² = 0.99), indicates that they can be a rapid and cheap complementary method to evaluate in peach palms some important physiological events, such as CO2 assimilation.

Tämän diplomityön tarkoituksena oli tutkia pitkäketjuisten rasvahappokloridien ja alkyyliketeenidimeerien (AKD) valmistusreaktioiden kinetiikkaa. Työn tavoitteena oli saada mittaustuloksia, joiden perusteella voitaisiin kehittää reaktioille kineettinen kineettinen malli ja suorittaa valmistusprosessien alustava optimointi. Teoreettisessa osassa on selvitetty rasvahappokloridien ja alkyyliketeenidimeerien (AKD) eri valmistustapoja. Lisäksi on perehdytty tarkemmin rasvahappokloridien dehydrohalogenointireaktioiden reaktiomekanismeihin ja valmistusprosessin problematiikkaan. Kokeellisessa osassa tutkittiin rasvahappokloridien valmistusta klooraamalla öljyhappoa 30 mol-% fosforitrikloridiylimäärällä lämpötiloissa 45, 50 ja 55OC. Alkyyliketeenidimeerien valmistusta tutkittiin dehydrohalogenoimalla palmitiini- ja öljyhappokloridia 0-30 mol-% trietyyliamiiniylimäärällä inertissä liuottimessa, lämpötiloissa 45 ja 50OC. Reaktiot toteutettiin puolipanosreaktiona, joissa reaktioastiassa olevaan reagenssin ja liuottimen seokseen lisättiin lähtöaine tasaisena massavirtana. Reaktioiden etenemistä seurattiin FT-IR- ja GC-analyysien avulla. Kalorimetrisilla kokeilla tutkittiin öljyhappokloridin dimeroitumisreaktion reaktiolämmön muodostumista ja UV-VIS-analyyseillä seurattiin öljyhappokloridin vanhenemista. Öljyhappokloridin valmistusreaktiolle saatiin hyvä kineettinen malli. Kineettisen mallin puutteena voidaan pitää sitä, ettei kokeissa saatu tietoa mahdollisten sivutuotteiden muodostumisesta. Sovitusohjelmalla saatiin sovitettua estimaatit reaktionopeusvakioille lämpötiloissa 45, 50 ja 55OC. Happokloridien dimeroitumisreaktioiden kineettisen mallin sopivuudelle saatiin suhteellisen hyvä kuva Kuten öljyhappokloridin tapauksessa, myöskään AKD:n valmistusreaktioissa syntynyt sivutuotteita, joten niiden osuutta oletettuun kineettiseen malliin ei tunneta. Sovitusohjelmalla saatiin sovitettua estimaatit reaktionopeusvakioille lämpötiloissa 45 ja 50OC.

Wheat plants (Triticum durum Desf., cv. Regallo) were grown in the field to study the effects of contrasting [CO2] conditions (700 versus 370 μmol mol−1) on growth, photosynthetic performance, and C management during the post-anthesis period. The aim was to test whether a restricted capacity of sink organs to utilize photosynthates drives a loss of photosynthetic capacity in elevated CO2. The ambient 13C/12C isotopic composition (δ13C) of air CO2 was changed from-10.2 in ambient [CO2] to-23.6 under elevated [CO2] between the 7th and the 14th days after anthesis in order to study C assimilation and partitioning between leaves and ears. Elevated [CO2] had no significant effect on biomass production and grain filling, and caused an accumulation of C compounds in leaves. This was accompanied by up-regulation of phosphoglycerate mutase and ATP synthase protein content, together with down-regulation of adenosine diphosphate glucose pyrophosphatase protein. Growth in elevated [CO2] negatively affected Rubisco and Rubisco activase protein content and induced photosynthetic down-regulation. CO2 enrichment caused a specific decrease in Rubisco content, together with decreases in the amino acid and total N content of leaves. The C labelling revealed that in flag leaves, part of the C fixed during grain filling was stored as starch and structural C compounds whereas the rest of the labelled C (mainly in the form of soluble sugars) was completely respired 48 h after the end of labelling. Although labelled C was not detected in the δ13C of ear total organic matter and respired CO2, soluble sugar δ13C revealed that a small amount of labelled C reached the ear. The 12CO2 labelling suggests that during the beginning of post-anthesis the ear did not contribute towards overcoming flag leaf carbohydrate accumulation, and this had a consequent effect on protein expression and photosynthetic acclimation.

Evolutionary developmental biology has grown historically from the capacity to relate patterns of evolution in anatomy to patterns of evolution of expression of specific genes, whether between very distantly related species, or very closely related species or populations. Scaling up such studies by taking advantage of modern transcriptomics brings promising improvements, allowing us to estimate the overall impact and molecular mechanisms of convergence, constraint or innovation in anatomy and development. But it also presents major challenges, including the computational definitions of anatomical homology and of organ function, the criteria for the comparison of developmental stages, the annotation of transcriptomics data to proper anatomical and developmental terms, and the statistical methods to compare transcriptomic data between species to highlight significant conservation or changes. In this article, we review these challenges, and the ongoing efforts to address them, which are emerging from bioinformatics work on ontologies, evolutionary statistics, and data curation, with a focus on their implementation in the context of the development of our database Bgee (http://bgee.org). O objetivo deste trabalho foi determinar o comportamento reológico do suco de abacaxi- pérola natural e tratado com enzimas pectinolíticas. As condições de tratamento enzimático foram otimizadas através de um planejamento experimental do tipo fatorial completo 2k, com três repetições do ponto central. Na avaliação do comportamento reológico foram utilizadas duas amostras submetidas a peneiramento (N e D), analisadas em quatro diferentes temperaturas (10; 25; 50 e 65 °C). As análises reológicas foram realizadas utilizando um viscosímetro de cilindros concêntricos Brookfield e os dados experimentais foram ajustados ao modelo de Mizrahi-Berk. A relação entre temperatura e viscosidade aparente foi descrita por uma equação tipo Arrhenius. A otimização da atividade enzimática indicou, através da análise de variância e da metodologia de superfície de resposta, que as variáveis temperatura e tempo de tratamento exerceram efeito estatisticamente significativo (p<0,05) sobre a concentração de pectina presente na amostra. O modelo utilizado mostrou-se adequado para descrever o comportamento reológico dos sucos de acordo com os parâmetros R², χ² e Bf. Os baixos valores obtidos para o índice de comportamento indicaram um comportamento pseudoplástico (n<1). A temperatura exerceu influência sobre a tensão de cisalhamento e a viscosidade aparente dos sucos analisados, sendo os menores valores observados nas amostras analisadas a 65 °C. A equação tipo Arrhenius descreveu de modo satisfatório o efeito da temperatura sobre a viscosidade aparente. Os valores da energia de ativação (Eat) foram de 4,54 Kcal.g.mol-1 e 4,89 Kcal.g.mol-1, respectivamente, para as amostras do suco de abacaxi natural e despectinizado, aumentando com o tratamento enzimático. A atividade enzimática proporcionou uma redução nos valores dos parâmetros de comportamento reológico das amostras, bem como na viscosidade aparente, em todas as temperaturas utilizadas, sendo o maior percentual de redução observado a 65 °C (41,66%).

Objective: Imipenem is a broad spectrum antibiotic used to treat severe infections in critically ill patients. Imipenem pharmacokinetics (PK) was evaluated in a cohort of neonates treated in the Neonatal Intensive Care Unit of the Lausanne University Hospital. The objective of our study was to identify key demographic and clinical factors influencing imipenem exposure in this population. Method: PK data from neonates and infants with at least one imipenem concentration measured between 2002 and 2013 were analyzed applying population PK modeling methods. Measurement of plasma concentrations were performed upon the decision of the physician within the frame of a therapeutic drug monitoring (TDM) programme. Effects of demographic (sex, body weight, gestational age, postnatal age) and clinical factors (serum creatinine as a measure of kidney function; co-administration of furosemide, spironolactone, hydrochlorothiazide, vancomycin, metronidazole and erythromycin) on imipenem PK were explored. Model-based simulations were performed (with a median creatinine value of 46 μmol/l) to compare various dosing regimens with respect to their ability to maintain drug levels above predefined minimum inhibitory concentrations (MIC) for at least 40 % of the dosing interval. Results: A total of 144 plasma samples was collected in 68 neonates and infants, predominantly preterm newborns, with median gestational age of 27 weeks (24 - 41 weeks) and postnatal age of 21 days (2 - 153 days). A two-compartment model best characterized imipenem disposition. Actual body weight exhibited the greatest impact on PK parameters, followed by age (gestational age and postnatal age) and serum creatinine on clearance. They explain 19%, 9%, 14% and 9% of the interindividual variability in clearance respectively. Model-based simulations suggested that 15 mg/kg every 12 hours maintain drug concentrations over a MIC of 2 mg/l for at least 40% of the dosing interval during the first days of life, whereas neonates older than 14 days of life required a dose of 20 mg/kg every 12 hours. Conclusion: Dosing strategies based on body weight and post-natal age are recommended for imipenem in all critically ill neonates and infants. Most current guidelines seem adequate for newborns and TDM should be restricted to some particular clinical situations.

O Cerrado brasileiro possui uma riqueza de espécies frutíferas que ainda não foram suficientemente estudadas em relação às suas características físicas, químicas e funcionais. O presente estudo teve o objetivo de mensurar as características físicas de frutos de gabirobeira e analisar a composição centesimal e de minerais, o teor de compostos fenólicos totais e a atividade antioxidante da polpa e do resíduo de gabiroba. As características físicas de maior variabilidade foram massa da polpa e do fruto, destacando-se o elevado rendimento de polpa (46,24%). A polpa e o resíduo de gabiroba contêm altos teores de umidade e fibra alimentar e quantidades consideráveis de ferro. O resíduo de gabiroba apresentou maior conteúdo de compostos fenólicos (1.787,65 mg AGE.100g-1) e atividade antioxidante (197,13 µmol TE.g-1) em relação à polpa. Contudo, os valores constatados na polpa de gabiroba (1.222,59 mg AGE.100g-1 e 107,96 µmol TE.g-1, respectivamente) são superiores aos de muitos frutos consumidos tradicionalmente. O teor de fenólicos totais apresentou forte correlação (r= 0,9723) com a atividade antioxidante. Os resultados indicam perspectivas promissoras para o aproveitamento integral do fruto da gabirobeira, visto seu conteúdo apreciável de nutrientes e de compostos fenólicos, e sua elevada atividade antioxidante.

As indústrias de frutas destacam-se pelo volume de resíduo gerado e, principalmente, pela composição dos mesmos. O Brasil vem-se consolidando como um importante produtor mundial de suco de uva, gerando grande quantidade de resíduos diversos, como o bagaço, que corresponde a aproximadamente 20% da fruta processada. O bagaço constitui-se de casca e semente, e apresenta uma composição rica e heterogênea em compostos fenólicos (flavonoides e não flavonoides), razão pela qual foi empregado como matéria-prima no presente trabalho para a obtenção de um extrato bioativo. As extrações enzimática e etanólica foram avaliadas para a recuperação destes compostos para fins alimentícios. Dentre as condições avaliadas nos dois planejamentos experimentais, a extração hidroetanólica foi mais eficiente na recuperação dos compostos bioativos (p< 0,01), cujo extrato apresentou uma capacidade antioxidante de 1.965 µmol Trolox por 100 g de bagaço, 25% superior à do extrato obtido com a melhor condição enzimática (1.580 µmol Trolox por 100 g de bagaço).

RESUMO Com o conhecimento de que a aplicação exógena de giberelinas pode alterar os processos fisiológicos, modificando o crescimento e o desenvolvimento das plantas, vistoque funcionam como regulador da divisão e do alongamento das células, objetivou-se com estetrabalho avaliar o crescimento e o desenvolvimento de Passiflora edulis Sims f. flavicarpa Degener cultivadosob luminosidade de 650 µmol m-2 s-1, por nove meses, e submetido a pulverizações semanais com GA3. Odelineamento experimental foi inteiramente casualizado, com cinco tratamentos (0,0; 1,5; 3,0; 15,0 e 30,0 mgL-1 de GA3) e dez repetições. O substrato utilizado foi constituído por uma mistura (1:1:1, em volume) de areia grossa lavada, moinha de carvão lavado e Latossolo Vermelho distroférrico disposto em vasos com capacidadepara 5L. As concentrações de GA3 influenciaram (p<0,05) na maioria das variáveis analisadas, com exceção do comprimento do caule principal até a inserção do primeiro caule lateral, do número de cauleslaterais, do número de nós do caule principal até à inserção do primeiro caule lateral, do número de nós do caule principal até a inserção da primeira gavinha e da primeira folha trilobada, do início da floração, donúmero de caules laterais emitidos pela planta no início da floração e do número de nós da base do caule principal até à inserção do primeiro botão floral. Concentração em torno de 15 mg L-1 propiciou maior comprimento da planta, maior número de nós da planta e maior massa fresca e seca das plantas. A utilização de 30 mg L d-1e GA3 antecipou a emissão da primeira gavinha e da primeira folha trilobada, e produziu maior número de botões florais. A utilização exógena de GA3 em concentrações de até 30 mg L-1 foi mais eficaz na indução do que na evocação floral dePassiflora edulis Sims f. flavicarpa Degener.

Lactate may represent a supplemental fuel for the brain. We examined cerebral lactate metabolism during prolonged brain glucose depletion (GD) in acute brain injury (ABI) patients monitored with cerebral microdialysis (CMD). Sixty episodes of GD (defined as spontaneous decreases of CMD glucose from normal to low [<1.0 mmol/L] for at least 2 h) were identified among 26 patients. During GD, we found a significant increase of CMD lactate (from 4±2.3 to 5.4±2.9 mmol/L), pyruvate (126.9±65.1 to 172.3±74.1 μmol/L), and lactate/pyruvate ratio (LPR; 27±6 to 35±9; all, p<0.005), while brain oxygen and blood lactate remained normal. Dynamics of lactate and glucose supply during GD were further studied by analyzing the relationships between blood and CMD samples. There was a strong correlation between blood and brain lactate when LPR was normal (r=0.56; p<0.0001), while an inverse correlation (r=-0.11; p=0.04) was observed at elevated LPR >25. The correlation between blood and brain glucose also decreased from r=0.62 to r=0.45. These findings in ABI patients suggest increased cerebral lactate delivery in the absence of brain hypoxia when glucose availability is limited and support the concept that lactate acts as alternative fuel.

This paper reports the method development for the simultaneous determination of methylmercury MeHgþ) and inorganic mercury (iHg) species in seafood samples. The study focused on the extraction and quantification of MeHgþ (the most toxic species) by liquid chromatography coupled to on-line UV irradiation and cold vapour atomic fluorescence spectroscopy (LC-UV-CV-AFS), using HCl 4 mol/L as the extractant agent. Accuracy of the method has been verified by analysing three certified reference materials and different spiked samples. The values found for total Hg and MeHgþ for the CRMs did not differ significantly from certified values at a 95% confidence level, and recoveries between 85% and 97% for MeHgþ, based on spikes, were achieved. The detection limits (LODs) obtained were 0.001 mg Hg/kg for total mercury, 0.0003 mg Hg/kg for MeHgþ and 0.0004 mg Hg/kg for iHg. The quantification limits (LOQs) established were 0.003 mg Hg/kg for total mercury, 0.0010 mg Hg/kg for MeHgþ and 0.0012 mg Hg/kg for iHg. Precision for each mercury species was established, being 12% in terms of RSD in all cases. Finally, the developed method was applied to 24 seafood samples from different origins and total mercury contents. The concentrations for Total Hg, MeHg and iHg ranged from 0.07 to 2.33, 0.003-2.23 and 0.006-0.085 mg Hg/kg, respectively. The established analytical method allows to obtain results for mercury speciation in less than 1 one hour including both, sample pretreatment and measuring step.

Here, we report the culture and characterization of an alphaproteobacterium of the order Rhizobiales, isolated from the gut of the honey bee Apis mellifera. Strain PEB0122T shares >95 % 16S rRNA gene sequence similarity with species of the genus Bartonella, a group of mammalian pathogens transmitted by bloodsucking arthropods. Phylogenetic analyses showed that PEB0122T and related strains from the honey bee gut form a sister clade of the genus Bartonella. Optimal growth of strain PEB0122T was obtained on solid media supplemented with defibrinated sheep blood under microaerophilic conditions at 35-37 °C, which is consistent with the cultural characteristics of other species of the genus Bartonella. Reduced growth of strain PEB0122T also occurred under aerobic conditions. The rod-shaped cells of strain PEB0122T had a mean length of 1.2-1.8 μm and revealed hairy surface structures. Strain PEB0122T was positive for catalase, cytochrome c oxidase, urease and nitrate reductase. The fatty acid composition was comparable to those of other species of the genus Bartonella, with palmitic acid (C16 : 0) and isomers of 18- and 19-carbon chains being the most abundant. The genomic DNA G+C content of PEB0122T was determined to be about 45.5 mol%. The high 16S rRNA gene sequence similarity with species of Bartonella and its close phylogenetic position suggest that strain PEB0122T represents a novel species within the genus Bartonella, for which we propose the name Bartonella apis sp. nov. The type strain is PEB0122T ( = NCIMB 14961T = DSM 29779T).
