Linking supply to demand: the neuronal monocarboxylate transporter MCT2 and the alpha-amino-3-hydroxyl-5-methyl-4-isoxazole-propionic acid receptor GluR2/3 subunit are associated in a common trafficking process.

MCT2 is the major neuronal monocarboxylate transporter (MCT) that allows the supply of alternative energy substrates such as lactate to neurons. Recent evidence obtained by electron microscopy has demonstrated that MCT2, like alpha-amino-3-hydroxyl-5-methyl-4-isoxazole-propionic acid (AMPA) receptors, is localized in dendritic spines of glutamatergic synapses. Using immunofluorescence, we show in this study that MCT2 colocalizes extensively with GluR2/3 subunits of AMPA receptors in neurons from various mouse brain regions as well as in cultured neurons. It also colocalizes with GluR2/3-interacting proteins, such as C-kinase-interacting protein 1, glutamate receptor-interacting protein 1 and clathrin adaptor protein. Coimmunoprecipitation of MCT2 with GluR2/3 and C-kinase-interacting protein 1 suggests their close interaction within spines. Parallel changes in the localization of both MCT2 and GluR2/3 subunits at and beneath the plasma membrane upon various stimulation paradigms were unraveled using an original immunocytochemical and transfection approach combined with three-dimensional image reconstruction. Cell culture incubation with AMPA or insulin triggered a marked intracellular accumulation of both MCT2 and GluR2/3, whereas both tumor necrosis factor alpha and glycine (with glutamate) increased their cell surface immunolabeling. Similar results were obtained using Western blots performed on membrane or cytoplasm-enriched cell fractions. Finally, an enhanced lactate flux into neurons was demonstrated after MCT2 translocation on the cell surface. These observations provide unequivocal evidence that MCT2 is linked to AMPA receptor GluR2/3 subunits and undergoes a similar translocation process in neurons upon activation. MCT2 emerges as a novel component of the synaptic machinery putatively linking neuroenergetics to synaptic transmission.

Vertebrate and nematode genes coding for yolk proteins are derived from a common ancestor.

One of the most obvious characteristics of the egg cells of oviparous animals is their large size resulting to a major extent from the deposition of nutritional reserves, mainly constituted of yolk proteins. In general, these are derived from a precursor called vitellogenin, which undergoes posttranslational modifications during secretion and during transport into and storage within the oocytes. Comparative analysis of the structural organization of the vitellogenin gene and of its product in different species shows that the vitellogenin gene is very ancient and that in vertebrates the gene may have more resemblance to the earliest gene than in invertebrates.

Abstract

Digital forensic osteology--possibilities in cooperation with the Virtopsy project.

The present study was carried out to check whether classic osteometric parameters can be determined from the 3D reconstructions of MSCT (multislice computed tomography) scans acquired in the context of the Virtopsy project. To this end, four isolated and macerated skulls were examined by six examiners. First the skulls were conventionally (manually) measured using 32 internationally accepted linear measurements. Then the skulls were scanned by the use of MSCT with slice thicknesses of 1.25 mm and 0.63 mm, and the 33 measurements were virtually determined on the digital 3D reconstructions of the skulls. The results of the traditional and the digital measurements were compared for each examiner to figure out variations. Furthermore, several parameters were measured on the cranium and postcranium during an autopsy and compared to the values that had been measured on a 3D reconstruction from a previously acquired postmortem MSCT scan. The results indicate that equivalent osteometric values can be obtained from digital 3D reconstructions from MSCT scans using a slice thickness of 1.25 mm, and from conventional manual examinations. The measurements taken from a corpse during an autopsy could also be validated with the methods used for the digital 3D reconstructions in the context of the Virtopsy project. Future aims are the assessment and biostatistical evaluation in respect to sex, age and stature of all data sets stored in the Virtopsy project so far, as well as of future data sets. Furthermore, a definition of new parameters, only measurable with the aid of MSCT data would be conceivable.

Tanmateix, però, les informacions publicades o emeses venen marcades per les opinions polítiques i periodístiques, que es troben censurades en certa mesura perquè segueixen una determinada línia política i/o editorial. Per això s‟ha elaborat un reportatge digital focalitzat en l‟empresariat català i l‟encaix Catalunya-Espanya que pretén aportar noves valoracions entorn a aquest debat donant veu a empresaris i professionals, entre altres. La part teòrica de la memòria defineix els conceptes bàsics que són de forçat coneixement per realitzar una part pràctica que ha de ser entesa per qualsevol persona i que s‟ha de tractar amb la major objectivitat possible. Per això, el web http://laquestiocatalana.cat aporta un llenguatge planer i les eines bàsiques per donar a conèixer una matèria molt criticada per alguns sectors. A través d‟entrevistes a personatges rellevants, el web introdueix nous punts de vista sobre una qüestió marcada, a simple vista, per la preocupació d‟un possible boicot per part d‟Espanya. Però aquesta no sembla ser la principal inquietud dels entesos, i és que hi ha altres aspectes encara més alarmants que poden influir en l‟empresariat i que no són tractats per cap mitjà de comunicació convencional.

Absence of association between specific common variants of the obesity-related FTO gene and psychological and behavioral eating disorder phenotypes.

Extensive population-based genome-wide association studies have identified an association between the FTO gene and BMI; however, the mechanism of action is still unknown. To determine whether FTO may influence weight regulation through psychological and behavioral factors, seven single-nucleotide polymorphisms (SNPs) of the FTO gene were genotyped in 1,085 individuals with anorexia nervosa (AN) and 677 healthy weight controls from the international Price Foundation Genetic Studies of Eating Disorders. Each SNP was tested in association with eating disorder phenotypes and measures that have previously been associated with eating behavior pathology: trait anxiety, harm-avoidance, novelty seeking, impulsivity, obsessionality, compulsivity, and concern over mistakes. After appropriate correction for multiple comparisons, no significant associations between individual FTO gene SNPs and eating disorder phenotypes or related eating behavior pathology were identified in cases or controls. Thus, this study found no evidence that FTO gene variants associated with weight regulation in the general population are associated with eating disorder phenotypes in AN participants or matched controls. © 2011 Wiley-Liss, Inc.

La preservació digital (PD) s'ha convertit en un problema persistent per a tots els que vulguin conservar la seva informació digital, garantir el seu estat i consultar aquest informació en el transcurs del temps. Fins ara només grans institucions amb coneixement expert i eines especialitzades han pogut fer front a aquest problema, però la preservació digital no pot ser abordada per una sola institució o nació. Les biblioteques, arxius i altres institucions de conservació de la memòria comparteixen aquest repte de la mateixa manera que els col•leccionistes i creadors, que ho fan a títol individual.L’objectiu del projecte és crear l'aplicació Pyramid que està concebuda com una eina de suport orientada a l'usuari domèstic (sense coneixements tècnics ni de preservació) per a la preservació a mig i llarg termini de col•leccions digitals, texts i vídeos, tal que funcioni com un antivirus (en BackGround) i preservi la informació sense requerir un cost addicional a l'ordinador i que l'usuari no noti cap molèstia a l'hora de fer les seves tasques diàries

Flea infestation reduces the life span of the common vole.

Parasitism is often a source of variation in host's fitness components. Understanding and estimating its relative importance for fitness components of hosts is fundamental from physiological, ecological and evolutionary perspectives. Host-parasite studies have often reported parasite-induced reduction of host fecundity, whereas the effect of parasitism on host survival has been largely neglected. Here, we experimentally investigated the effect of infestation by rat fleas (Nosopsyllus fasciatus) on the life span of wild-derived male common voles (Microtus arvalis) bred in captivity. We found that the mean life span of parasitized voles was reduced by 36% compared to control voles. Parasitized voles had a smaller body size, but a relatively larger heart and spleen than control voles. These results indicate an effect of flea infestation on host life span and our findings strongly suggest that ectoparasites should be taken into account in the studies of host population dynamics.

De lo analógico a lo digital. El futuro de la enseñanza de la composición

Las prácticas lectoras están cambiando a causa de factores múltiples: leemos otro tipo de textos, con objetivos también más ambiciosos, en contextos nuevos, que nunca antes habían existido. Analizamos algunos de los matices que ha adoptado la práctica de la lectura en este inicio del siglo XXI, desde esta perspectiva sociocultural.

El portafolios digital como herramienta para el desarrollo de competencias transversales en el alumnado.

Partiendo de un enfoque sobre las relaciones entre las tecnologías digitales como herramientas de mediación, se analiza el papel de un caso concreto, los portafolios electrónicos, para la mejora de competencias transversales en el ámbito universitario. Los resultados indican que se obtienen mejoras, como mínimo desde la percepción de los propios estudiantes, en algunas dimensiones ligadas a la capacidad para seleccionar y organizar información, así como a la planificación del aprendizaje.

Microsatellites for genetic studies and breeding programs in common bean

Twenty microsatelitte loci were identified and characterized in common bean. Microsatellites were tested in 14 genotypes. The allele number ranged from 1 to 3, and the polymorphism information content (PIC) was between 0.14 and 0.65. These polymorphic markers are available to be used for breeding programs.
