The effect of DNA cytosine methylation on H-ras promoter activity was assessed using a transient expression system employing the plasmid H-rasCAT (NaeI H-ras promoter linked to the chloramphenicol acetyltransferase (CAT) gene). This 551 bp promoter is 80% GC rich, enriched with 168 CpG dinucleotides, and contains six functional GC box elements which represent major DNA methylation target sites. Prokaryotic methyltransferases HhaI (CGm$\sp5$CG) and HpaII (Cm$\sp5$CGG) alone or in combination with a human placental methyltransferase (HP MTase) were used to introduce methyl groups at different CpG sites within the promoter. To test for functional promoter activity, the methylated plasmids were introduced into CV-1 cells and CAT activity assessed 48 h post-transfection. Methylation at specific HhaI and HpaII sites reduced CAT expression by 70%, whereas more extensive methylation at generalized CpG sites with HP MTase inactivated the promoter $>$95%. The inhibition of H-ras promoter activity was not attributable to methylation-induced differences in DNA uptake or stability in the cell, topological form of the plasmid, or methylation effects in nonpromoter regions. We also observed that DNA cytosine methylation of a 360 bp promoter fragment by HP MTase induced a local change in DNA conformation. Using three independent methodologies (nitrocellulose filter binding assays, gel mobility shifts, and Southwestern blots), we determined that this change in promoter conformation affected the interaction of nuclear proteins with cis-regulatory sequences residing in the promoter region. The results provide evidence to suggest that DNA methylation may regulate gene expression by inducing changes in local promoter conformation which in turn alters the interactions between DNA and protein factors required for transcription. The results provide supportive evidence for the hypothesis of Cedar and Riggs, who postulated that DNA methylation may regulate gene expression by altering the binding affinities of proteins for DNA. ^

Under the process of transition toward a market economy, the economic connections of the Russian Far East (RFE) with external regions changed from a division of labor among the regions of the USSR (Russia) to an international division of labor. This happened due to factors including the liberalization of the trade system away from a state monopoly, the presence of rich natural resources and of developed industries related to these resources, the advantage of geographically proximity to Asia-Pacific countries, and the political and economic division of the once unified national economic space during the process of transition. The economic connections of RFE with external economies changed radically under the transition toward the market economy. First, the value of foreign trade increased dramatically and the importance of foreign trade for the RFE economy increased enormously. Second, however, different territories of RFE traveled along different trajectories, due to factors involving their industrial structure and geographical conditions. Third, in recent years connections with China, in the areas of both exports and imports, have grown. Fourth, the share within exports of "fuel, mineral resources and metal" increased radically from the end of the 1990s, and the share of "machine, facilities and transportation means" increased from 2002 year within imports. Under this situation, especially since 2002, there has been a major change in the structure of foreign trade.

The intermediatebandsolarcell (IBSC) is a photovoltaic device with a theoretical conversion efficiency limit of 63.2%. In recent years many attempts have been made to fabricate an intermediateband material which behaves as the theory states. One characteristic feature of an IBSC is its luminescence spectrum. In this work the temperature dependence of the photoluminescence (PL) and electroluminescence (EL) spectra of InAs/GaAs QD-IBSCs together with their reference cell have been studied. It is shown that EL measurements provide more reliable information about the behaviour of the IB material inside the IBSC structure than PL measurements. At low temperatures, the EL spectra are consistent with the quasi-Fermi level splits described by the IBSC model, whereas at room temperature they are not. This result is in agreement with previously reported analysis of the quantum efficiency of the solarcells

The separation of the lower stage of the ARIANE 5 Vehicle Equipment Bay (VEB) Structure is to be done using a pyrotechnic device. The wave propagation effects produced by the explosion can affect the electronic equipment, so it was decided to analyze, using both physical and numerical modeling, a small piece of the structure to determine the distribution of the accelerations and the relative importance of damping, stiffness, connections, etc. on the response of the equipment.

Current development platforms for designing spoken dialog services feature different kinds of strategies to help designers build, test, and deploy their applications. In general, these platforms are made up of several assistants that handle the different design stages (e.g. definition of the dialog flow, prompt and grammar definition, database connection, or to debug and test the running of the application). In spite of all the advances in this area, in general the process of designing spoken-based dialog services is a time consuming task that needs to be accelerated. In this paper we describe a complete development platform that reduces the design time by using different types of acceleration strategies based on using information from the data model structure and database contents, as well as cumulative information obtained throughout the successive steps in the design. Thanks to these accelerations, the interaction with the platform is simplified and the design is reduced, in most cases, to simple confirmations to the “proposals” that the platform automatically provides at each stage. Different kinds of proposals are available to complete the application flow such as the possibility of selecting which information slots should be requested to the user together, predefined templates for common dialogs, the most probable actions that make up each state defined in the flow, different solutions to solve specific speech-modality problems such as the presentation of the lists of retrieved results after querying the backend database. The platform also includes accelerations for creating speech grammars and prompts, and the SQL queries for accessing the database at runtime. Finally, we will describe the setup and results obtained in a simultaneous summative, subjective and objective evaluations with different designers used to test the usability of the proposed accelerations as well as their contribution to reducing the design time and interaction.

Tests used to simulate the separation of the lower stage of the Ariane Vehicle Equipment Bay (VEB) were carried out on a flat full scale model. Theoretical studies carried out prior to testing are described. Three different mathematical methods, finite element, component element, and wave propagation, were used. Comparison of the predicted theoretical results with the actual test results is planned.

A new method to analyze the influence of possible hysteresis cycles in devices employed for optical computing architectures is reported. A simple full adder structure is taken as the basis for this method. Single units, called optical programmable logic cells, previously reported by the authors, compose this structure. These cells employ, as basic devices, on-off and SEED-like components. Their hysteresis cycles have been modeled by numerical analysis. The influence of the different characteristic cycles is studied with respect to the obtained possible errors at the output. Two different approaches have been adopted. The first one shows the change in the arithmetic result output with respect to the different values and positions of the hysteresis cycle. The second one offers a similar result, but in a polar diagram where the total behavior of the system is better analyzed.

Various researchers have developed models of conventional H2O–LiBr absorption machines with the aim of predicting their performance. In this paper, the methodology of characteristic equations developed by Hellmann et al. (1998) is applied. This model is able to represent the capacity of single effect absorption chillers and heat pumps by means of simple algebraic equations. An extended characteristic equation based on a characteristic temperature difference has been obtained, considering the facility features. As a result, it is concluded that for adiabatic absorbers a subcooling temperature must be specified. The effect of evaporator overflow has been characterized. Its influence on cooling capacity has been included in the extended characteristic equation. Taking into account the particular design and operation features, a good agreement between experimental performance data and those obtained through the extended characteristic equation has been achieved at off-design operation. This allows its use for simulation and control purposes.

The theoretical improvements performed since the last spacecraft and mechanical testing conference on the study of the pyrotechnic shock phenomena produced during the separation of the lower stage of the Ariane 5 Vehicle Equipment Bay (VEB) structure are described. The first theoretical approach used was based on the wave propagation method, including axial and shear waves. The method was changed, in order to capture the bending effects, as well as the influence of the frequency dependent damping values. In addition to the development of the theoretical method, efforts were made to improve the criteria used to model the structure. Comparison of the theoretical predictions with the test results of a flat test sample 1 m width, as well as a preliminary test performed on a small sample, are presented.

We study the evolution of a finite size population formed by mutationally isolated lineages of error-prone replicators in a two-peak fitness landscape. Computer simulations are performed to gain a stochastic description of the system dynamics. More specifically, for different population sizes, we compute the probability of each lineage being selected in terms of their mutation rates and the amplification factors of the fittest phenotypes. We interpret the results as the compromise between the characteristic time a lineage takes to reach its fittest phenotype by crossing the neutral valley and the selective value of the sequences that form the lineages. A main conclusion is drawn: for finite population sizes, the survival probability of the lineage that arrives first to the fittest phenotype rises significantly

We can say without hesitation that in energy markets a throughout data analysis is crucial when designing sophisticated models that are able to capture most of the critical market drivers. In this study we will attempt to investigate into Spanish natural gas prices structure to improve understanding of the role they play in the determination of electricity prices and decide in the future about price modelling aspects. To further understand the potential for modelling, this study will focus on the nature and characteristics of the different gas price data available. The fact that the existing gas market in Spain does not incorporate enough liquidity of trade makes it even more critical to analyze in detail available gas price data information that in the end will provide relevant information to understand how electricity prices are affected by natural gas markets. In this sense representative Spanish gas prices are typically difficult to explore given the fact that there is not a transparent gas market yet and all the gas imported in the country is negotiated and purchased by private companies at confidential terms.

The interest for modelling of human actions acting on structures has been recurrent since the first accidents on suspension bridges in the nineteenth century such as Broughton (1831) in the U.K. or Angers (1850) in France. Stadiums, gymnasiums are other types of structure where human induced vibration is very important. In these structures a particular phenomenon appears such as the interaction personstructure (lock-in), the person-person synchronization, and the influence of the mass and damping of the people in the structural behaviour. This paper focuses on the latter topic. In order to evaluate these property modifications several tests have been carried out on a stand-alone building. For the test an electro-dynamic shaker was installed at a fixed point of the gym slab and different groups of people were located around the shaker. The dynamic characteristics of the structure without people inside have been calculated by two methods: using a three-dimensional finite element model of the building and by operational modal analysis. These calculated experimental and numerical values are the reference values used to evaluate the modifications in the dynamic properties of the structure.
