We present a feedback control scheme to stabilize unstable cellular patterns during the directional solidification of a binary alloy. The scheme is based on local heating of cell tips which protrude ahead of the mean position of all tips in the array. The feasibility of this scheme is demonstrated using phase-field simulations and, experimentally, using a real-time image processing algorithm, to track cell tips, coupled with a movable laser spot array device to heat the tips locally. We demonstrate, both numerically and experimentally, that spacings well below the threshold for a period-doubling instability can be stabilized. As predicted by the numerical calculations, cellular arrays become stable with uniform spacing through the feedback control which is maintained with minimal heating.

By simulations of the Barkley model, action of uniform periodic nonresonant forcing on scroll rings and wave turbulence in three-dimensional excitable media is investigated. Sufficiently strong rapid forcing converts expanding scroll rings into the collapsing ones and suppresses the Winfree turbulence caused by the negative tension of wave filaments. Slow strong forcing has an opposite effect, leading to expansion of scroll rings and induction of the turbulence. These effects are explained in the framework of the phenomenological kinematic theory of scroll waves.

As expression of Cxs in cells of the immune system increases upon cellular activation, we investigated whether Cxs and especially CxHcs play a major role during T cell-mediated responses. In particular, we studied the expression of Cx43Hc following CD4(+) T cell stimulation using flow cytometry, real-time PCR, and Western blot analysis. We showed that expression of Cx43 and its phosphorylated isoforms increased in response to the engagement of CD3 and CD28. Cx43Hcs were found to be involved in sustaining proliferation of T cells, as assessed by cell cycle staining, thymidine incorporation assays, and CFSE analysis of cells exposed to mimetic peptide inhibitors of the plasma membrane Cx channels and antibodies generated to an extracellular region of Cx. The reduction of T cell proliferation mediated by Cx channel inhibitors suppressed cysteine uptake but not cytokine production. We conclude that upon antigen recognition, T cells require CxHc to sustain their clonal expansion.

The role of the gluco-incretin hormones GIP and GLP-1 in the control of beta cell function was studied by analyzing mice with inactivation of each of these hormone receptor genes, or both. Our results demonstrate that glucose intolerance was additively increased during oral glucose absorption when both receptors were inactivated. After intraperitoneal injections, glucose intolerance was more severe in double- as compared to single-receptor KO mice, and euglycemic clamps revealed normal insulin sensitivity, suggesting a defect in insulin secretion. When assessed in vivo or in perfused pancreas, insulin secretion showed a lack of first phase in Glp-1R(-/-) but not in Gipr(-/-) mice. In perifusion experiments, however, first-phase insulin secretion was present in both types of islets. In double-KO islets, kinetics of insulin secretion was normal, but its amplitude was reduced by about 50% because of a defect distal to plasma membrane depolarization. Thus, gluco-incretin hormones control insulin secretion (a) by an acute insulinotropic effect on beta cells after oral glucose absorption (b) through the regulation, by GLP-1, of in vivo first-phase insulin secretion, probably by an action on extra-islet glucose sensors, and (c) by preserving the function of the secretory pathway, as evidenced by a beta cell autonomous secretion defect when both receptors are inactivated.

En la historiografía de la investigación sobre la cultura ibérica, el poblado de San Antonio el Pobre (Calaceite) va indeleblemente unido a las intervenciones del Institut d¿Estudis Catalans (IEC) y a la figura de Pere Bosch Gimpera, sin duda el prehistoriador español de mayor prestigio hasta el final de la guerra civil. Sin embargo, es significativo que dicho conocimiento se deba más a trabajos aislados y a la recuperación de sus diarios de excavación realizada por Francisca Pallarés Salvador en 1958, que a la publicación de una serie de monografías que recogiera sus trabajos en el área entre 1914 y 1923.

En este artículo se plantea que la planificación y la evaluación pueden y deben reforzar los aspectos democráticos y participativos de cualquier proyecto o programa de animación sociocultural, ya que todo proceso de planificación de servicios y programas sociales en general, y de programas de animación sociocultural en particular, presenta elementos de control y elementos de cambio social, y entre estas dos dimensiones existe una relación dialéctica simétrica inversamente proporcional. Finalmente, se desarrolla un modelo democrático, social y participativo de planificación y evaluación.

Aims: To describe the drinking patterns and their baseline predictive factors during a 12-month period after an initial evaluation for alcohol treatment. Methods CONTROL is a single-center, prospective, observational study evaluating consecutive alcohol-dependent patients. Using a curve clustering methodology based on a polynomial regression mixture model, we identified three clusters of patients with dominant alcohol use patterns described as mostly abstainers, mostly moderate drinkers and mostly heavy drinkers. Multinomial logistic regression analysis was used to identify baseline factors (socio-demographic, alcohol dependence consequences and related factors) predictive of belonging to each drinking cluster. ResultsThe sample included 143 alcohol-dependent adults (63.6% males), mean age 44.6 ± 11.8 years. The clustering method identified 47 (32.9%) mostly abstainers, 56 (39.2%) mostly moderate drinkers and 40 (28.0%) mostly heavy drinkers. Multivariate analyses indicated that mild or severe depression at baseline predicted belonging to the mostly moderate drinkers cluster during follow-up (relative risk ratio (RRR) 2.42, CI [1.02-5.73, P = 0.045] P = 0.045), while living alone (RRR 2.78, CI [1.03-7.50], P = 0.044) and reporting more alcohol-related consequences (RRR 1.03, CI [1.01-1.05], P = 0.004) predicted belonging to the mostly heavy drinkers cluster during follow-up. Conclusion In this sample, the drinking patterns of alcohol-dependent patients were predicted by baseline factors, i.e. depression, living alone or alcohol-related consequences and findings that may inform clinicians about the likely drinking patterns of their alcohol-dependent patient over the year following the initial evaluation for alcohol treatment.

Phase I was initiated as a result of internal Iowa Department of Transportation (Iowa DOT) studies that raised concerns about the quality of embankments being constructed. Some large embankments have recently developed slope stability problems. In addition, pavement roughness has been noted shortly after roads were opened to traffic. This raised the question as to whether the current Iowa DOT embankment construction specifications are adequate. The primary objective of Phase I was to evaluate the quality of embankments being constructed under the current Iowa DOT specifications. The project was initiated in May 1997 with a tour of several embankment projects being constructed around the state. At each of these projects the resident construction engineer, field inspector, and contractor were interviewed with respect to their opinion of the current specifications. From construction observations and discussion during these visits it became obvious that there were problems with the current embankment construction specifications. Six embankment projects were selected for in-depth analysis and to represent the full range of soil types being used across the state. The results of Phase I field and laboratory construction testing and observations and post construction testing are presented in this report. Overall evaluation of the results of Phase I indicate that Iowa is not consistently obtaining a quality embankment constructed under the current Iowa DOT specifications. Based on these results, recommendations are made for Phase II to evaluate alternative specifications and develop rapid field methods for compaction control and soil identification.

The aim of this Phase I/IIa double-blind controlled trial was to test the efficacy of the sporozoite-based malaria vaccine PfCS 282-383 (PfCS102) to protect against Plasmodium falciparum parasitaemia. 16 volunteers were randomized to receive twice 30 μg of PfCS102 formulated in Montanide ISA 720 or ISA 720 alone (control). Two weeks after 2nd immunization, volunteers were challenged using 5 infected mosquitoes. All vaccinees developed antibodies against PfCS102 versus none control. 8/8 vaccinees and 6/6 controls challenged developed malaria parasitaemia. The duration from infection to onset of patent parasitaemia was similar in both groups (214 h in vaccinees and 216 in controls). PfCS102 is safe and immunogenic but provides no protection against artificial challenge in its current formulation.

Los individuos forman parte de varios grupos a la vez, en primer lugar del familiar y en segundo de todos los demás. La capacidad de cuidado de familiares y profesionales sanitarios así como su credibilidad como promotores de la salud estarán relacionadas con sus creencias y conocimientos sobre la enfermedad. El objetivo fue conocer las creencias y percepciones de los familiares y personal sanitario con relación a las causas, consecuencias y manejo de los factores de riesgo de la enfermedad coronaria, en 4 Hospitales de la Provincia de Barcelona. Muestra: 57 familiares de pacientes ingresados por un primer evento coronario, 65 enfermeras y 38 médicos que estaban trabajando en un momento determinado en todos los turnos y servicios donde se atendían enfermos de cardiopatía isquémica. Para el análisis de datos se utilizó el programa SPSS Vs 9.0.1.Resultados. El 72 % de los familiares opinaban que el paciente realizaba una dieta grasa. No hacen suficiente ejercicio un 78%. Había vivido un suceso importante en los últimos tres años un 61%. Se sentían capaces de cuidar a su familiar un 66%. No se han observado diferencias entre las opiniones de enfermeras y médicos ni entre los diferentes hospitales. El paciente es el máximo responsable del control de su enfermedad.

During the last decade, the discovery that astrocytes possess a nonelectrical form of excitability (Ca21-excitability) that leads to the release of chemical transmitters, an activity called ''gliotransmission'', indicates that these cells may have additional important roles in brain function. Elucidating the stimulus-secretion coupling leading to the exocytic release of chemical transmitters (such as glutamate, Bezzi et al., Nature Neurosci, 2004) may therefore clarify i) whether astrocytes represent in full a new class of secretory cells in the brain and ii) whether they can participate to the fast brain signaling in the brain. Here by using a recently developed approach for studying vesicle recycling dynamics at synapses (Voglmaier et al., Neuron, 2006; Balaji and Ryan, PNAS, 2007) combined with epifluorescence and total internal reflection fluorescence (TIRF) imaging, we investigated the spatiotemporal characteristics of stimulus-secretion coupling leading glutamate exocytosis of synaptic-like microvesicles (SLMVs) in astrocytes. We performed the analysis at both the whole-cell and single-vesicle levels providing the first system for comparing exo-endocytic processes in astrocytes with those in neurons. Both the time course and modalities of secretion in astrocytes present more similarities to neurons then previously expected. We found that 1. the G-protein-coupled receptor (GPCR)-evoked exocytosis reached the maximum on a ms time scale and that 2. ER tubuli formed sub-micrometer domains beneath the plasma membrane in close proximity to exocytic vesicles, where fusion events were spatiotemporally correlated with fast Ca21 events.

The impact of host genetic variation on determining the differential outcomes after HIV infection has been studied by two approaches: targeting of candidate genes and genome-wide association studies (GWASs). The overlap in genetic variants that has been identified by these two means has essentially been restricted to variants near to the human leukocyte antigen (HLA) class I genes, although variation in the CCR5 locus, which was first shown to have an effect on HIV outcomes using the candidate gene approach, does reach significance genome-wide when very large samples sizes (i.e. thousands) are used in GWAS. Overall, many of the variants identified by the candidate gene approach are likely to be spurious, as no additional variants apart from a novel variant near the HLA-C gene have been consistently identified by GWAS. Variants with low frequency and/or low impact on HIV outcomes are likely to exist in the genome and there could be many of them, but these are not identifiable, given current GWAS sample sizes. Several loci centrally involved in the immune response, including the immunoglobulin genes, T-cell receptor loci, or leukocyte receptor complex, are either poorly covered on the GWAS chips or difficult to interpret due to their repetitive nature and/or the presence of insertion/deletion polymorphisms in the region. These loci warrant further interrogation, but genetic characterization of these regions across a range of individuals will first be required. Finally, synergistic interactions between loci may affect outcome after infection, as suggested by associations of specific, functionally relevant HLA and killer cell immunoglobulin-like receptor variants with HIV disease outcomes, and these require further consideration as well.

El presente trabajo propone una reflexión sobre la relación entre el sistema de enseñanza-aprendizaje y el grado de presencialidad tomando como punto de partida la experiencia desarrollada en tres asignaturas obligatorias de la licenciatura de Ciencias Actuariales y Financieras (CAF) de la Universidad de Barcelona: Matemática Actuarial Vida (MAV), Matemática Actuarial No Vida (MANV) y Análisis Numérico (AN). Todas son asignaturas obligatorias en la licenciatura y constan de 9 créditos en el caso de las dos primeras (6 teóricos y 3 prácticos) y de 6 créditos el Análisis Numérico (3 teóricos y 3 prácticos). La implantación de un sistema de evaluación continuada a partir del curso 2004-05 ha supuesto un cambio de enfoque del proceso de enseñanza-aprendizaje que ha afectado tanto al profesorado implicado como al alumnado, se ha basado en el control de la asistencia del estudiante, en la valoración conjunta teoría-práctica, en el trabajo en grupo y en el portafolio docente como herramientas de trabajo del estudiante y de evaluación para el profesor. A partir del curso 2007-08 la Universidad de Barcelona establece un cambio en las normas reguladoras de la evaluación y de la calificación de los aprendizajes, de manera que se considerará que la evaluación será continuada como norma general. Debido a este cambio y a la valoración que habían realizado los alumnos respecto al sistema de evaluación continuada aplicado en cursos anteriores, mediante la realización de encuestas de opinión, se toma la decisión de introducir modificaciones en dicho sistema. Al finalizar el primer semestre del curso 2007-08 se analizan cómo han afectado estos cambios en los resultados académicos y nos planteamos el enfoque que hay que dar a estas asignaturas que, debido a sus contenidos, exigen un alto grado de presencialidad de los alumnos. Debido a que muchos estudiantes están compaginando trabajo y estudios, los profesores implicados se plantean si sería conveniente tender hacia un nuevo sistema de enseñanza-aprendizaje en el cual el grado de presencialidad requerido no fuese tan elevado.
