Os nanomateriais são estruturas com uma ou mais dimensões inferiores a 100 nanómetros. Devido à sua pequena dimensão, as nanopartículas apresentam atributos únicos, tais como a sua elevada área superficial relativamente à sua massa, reactividade ou força tênsil. Estas características influenciam grandemente algumas das propriedades dos nanomateriais, como a sua hidrofobicidade, carga ou toxicidade. As propriedades das nanopartículas tornam-nas também muito úteis para o Homem, sendo aplicadas em medicina, farmácia, electrónica, cosmética, vestuário e biotecnologia, entre outras. O aumento de produção e utilização de nanomateriais tem vindo a aumentar também a possibilidade de exposição humana a este tipo de partículas, levando a preocupações relativas ao risco de toxicidade aguda ou crónica. A exposição humana pode ocorrer por diversas vias, sendo as mais relevantes a via inalatória, ingestão ou contacto com a pele. Dependendo do material e do órgão-alvo, a exposição a nanomateriais pode conduzir a diferentes consequências biológicas: a nível dos órgãos, os nanomateriais podem levar a inflamação ou a supressão do sistema imunitário e, a nível celular e molecular, a perturbações na estrutura e integridade do genoma, assim como a interacções com moléculas biológicas e inibição da actividade proteica, entre outras consequências. Um dos nanomateriais mais utilizados são os nanotubos de carbono. Estes são constituídos por grafite cilíndrica disposta numa única camada (designados nanotubos de carbono de parede simples) ou em várias (nanotubos de carbono de parede múltipla). Os nanotubos de carbono apresentam propriedades como resistência e condutividade que os tornam muito úteis em aplicações como aparelhos electrónicos, vestuário ou biomedicina; cada vez mais, portanto, se torna provável a exposição ocupacional ou ambiental a este material. A semelhança estrutural destas partículas com fibras de amianto conduziu a questões relativas à sua segurança, pelo que já foram elaborados diversos estudos relativos aos seus efeitos biológicos. Alguns trabalhos sugerem que os nanotubos de carbono têm a capacidade de produzir toxicidade associada a lesões físicas, à produção de danos oxidativos por interacção com mecanismos celulares, ou a morte celular. Outros trabalhos defendem que estas partículas não causam toxicidade relevante. O projecto de dimensão europeia “NANoREG” surgiu da necessidade de ser desenvolvida legislação e regulamentação apoiadas em conhecimento científico e adequadas à produção e ao uso actual de nanomateriais. Este trabalho teve como objectivos principais a determinação do potencial cito- e genotóxico de um conjunto de nanotubos de carbono de parede múltipla (designados NM-400 a NM-403), e a consequente tentativa de associar este potencial às características físico-químicas dos nanomateriais. Com este objectivo, a exposição por via inalatória foi analisada, pelo uso de duas linhas celulares in vitro provenientes de tecidos do tracto respiratório: epitélio pulmonar (células A549) e epitélio brônquico (células BEAS-2B). A citotoxicidade dos nanotubos de carbono foi analisada com base em três parâmetros. Em primeiro lugar, as células foram contadas após a exposição aos nanomateriais utilizando o corante azul de tripanao para excluir as células inviáveis; a contagem foi realizada 3 e 24 horas após a exposição das células aos nanotubos. Os resultados deste ensaio apontam para a ausência de citotoxicidade após a exposição mais curta, e dados inconsistentes após a mais longa. Em segundo lugar, foi realizado o ensaio clonogénico, que se baseia na capacidade das células de se dividirem após a exposição ao agente em estudo. Este ensaio só foi realizado nas células A549 pois as BEAS-2B não permitem a formação de colónias. Os resultados apontam para uma citotoxicidade após a exposição a todos os nanomateriais, cuja intensidade se relaciona directamente com o tamanho das partículas, assim como ao seu diâmetro e área de superfície. Em terceiro lugar, foram calculados dois índices de viabilidade no ensaio dos Micronúcleos, cujo objectivo é avaliar se as células se dividiram durante a exposição aos nanomateriais em comparação com o controlo, e cujos resultados apresentam incoerências em relação aos outros já referidos. Estes dados podem ser justificados pelas diferenças existentes entre os ensaios, como o tempo de exposição ou a densidade celular. Os efeitos genotóxicos dos nanomateriais foram avaliados com recurso aos ensaios do cometa e dos micronúcleos. O primeiro detecta lesões pequenas e reversíveis nas cadeias de DNA, ao passo que o segundo detecta efeitos irreversíveis ao nível cromossómico, tais como quebras ou perdas de cromossomas. Os resultados do ensaio do cometa sugerem que nenhum dos nanomateriais testados é genotóxico, uma vez que em ambas as linhas celulares e em ambos os tempos de exposição, os resultados são negativos. O ensaio dos micronúcleos, por outro lado, aponta para existência de genotoxicidade de dois dos nanomateriais (NM-401 e NM-402) nas células A549, mas não em células BEAS-2B. Uma possível explicação para estes dados aparentemente contraditórios pode residir na hipótese de estes nanotubos de carbono serem compostos com efeitos aneugénicos, mas não clastogénicos: o ensaio dos micronúcleos permite a detecção de ambos os mecanismos de acção, ao passo que o ensaio do cometa só revela a quebra de cadeias de DNA. Outra justificação para os resultados é a possível influência da perda de viabilidade das células analisadas. Com base nos dados do ensaio clonogénico, estas partículas apresentam elevada citotoxicidade, pelo que os resultados dos ensaios de genotoxicidade, em particular do Ensaio do Cometa, poderão ser afectados por estes efeitos. O meio de cultura usado para expor as células aos nanomateriais também é um parâmetro muito relevante na sua toxicidade. Neste trabalho, foram usados meios de cultura com proteínas, que podem ser adsorvidas pelas partículas e formar uma “corona” em seu redor; este processo pode alterar propriedades importantes dos nanomateriais, entre os quais o seu potencial efeito biológico. Também o método usado para conseguir uma dispersão homogénea de nanomateriais pode conduzir a diferenças nos resultados dos ensaios de toxicidade. Neste estudo, foram observados alguns problemas relativos à perda de homogeneidade das dispersões de nanotubos de carbono, o que pode ter conduzido a que as células fossem expostas a massas de partículas de grandes dimensões conjuntamente com partículas individualizadas. O período durante o qual as células são expostas ao nanomaterial é também um aspecto essencial na produção de efeitos tóxicos. Resumindo, este projecto forneceu informações relativas à toxicidade dos nanotubos de carbono que, complementadas pelas conclusões dos restantes parceiros do projecto europeu, poderão contribuir significativamente para a avaliação de risco e criação de legislação relativamente à utilização de nanomateriais. Na linha celular BEAS-2B, nenhum destes nanomateriais parece produzir efeitos tóxicos, quer a nível de célula, quer a nível de genoma, nas condições experimentais utilizadas. Nas células A549, por outro lado, os três nanomateriais testados parecem ser acentuadamente citotóxicos, e dois deles (NM-401 e NM-402) são também genotóxicos. Em relação a perspectivas futuras, pode-se concluir que nem todos os ensaios de toxicidade existentes actualmente são adequados à análise de nanopartículas, pelo que novas metodologias devem ser desenvolvidas e complementadas por ensaios in vivo. Todos os estudos envolvendo nanomateriais deverão também descrever as características físico-químicas dos materiais usados, de forma a se poderem comparar os resultados com os de outros trabalhos.

The management of wound bioburden has previously been evaluated using various antimicrobial wound dressings on bacterial pathogens isolated from various wounds. In this present study, the antimicrobial effect of silver-impregnated dressings (Acticoat and Silvercel) and honey-impregnated dressing (Medihoney™ Apinate) on both planktonic bacteria and quasi-biofilms by Staphylococcus aureus and Proteus mirabilis were assessed using a 6-well plate and standard agar technique. In the 6-well plate assay, a bacterial suspension of 108 colony forming unit (CFU)/mL was inoculated on each dressing in excess Luria-Bertani broth and incubated at 35 – 37°C for 30 and 60 minutes and 24 hours. After each incubation time, bacteria were recovered in sodium thioglycolate solution (STS) and the CFU/mL determined on LB agar. Dressings were cut into circular shapes (2cm diameter and placed on Mueller Hinton agar plates pre-inoculated with bacterial suspensions to determine their zones of inhibition (ZOI) after 24 hours incubation. None of the dressings was effective to significantly inhibit bacterial growth or biofilm formation at all the times tested. Acticoat and Medihoney™ Apinate produced ZOIs between 1.5 – 15 mm against both Staphylococcus aureus and Proteus mirabilis. It is possible that, dressings augmented with antibiotics can significantly reduce quasi-biofilms on standard agar.

The impact of biofilm in the effective control of wound microbiome is an ongoing dilemma which has seen the use of different treatment strategies. The effects of wound dressings and antibiotics on both planktonic bacteria and biofilms have been separately evaluated in previous studies. In this current study, the combined antimicrobial effects of some selected wound dressings (silver-impregnated: Acticoat and Silvercel; and honey-impregnated: Medihoney™ Apinate) and antibiotics (ceftazdime and levofloxacin) on Klebsiella pneumoniae and Proteus mirabilis in their quasi-biofilm state were assessed using zone of inhibition (ZOI) test. Before the addition of the wound dressings, bacterial suspension of 108 colony forming units per mL and different concentrations of ceftazidime and levofloxacin (256, 512, 1024 and 5120µg/mL) of a final volume of 1mL were inoculated on Mueller Hinton agar and allowed to dry. Wound dressings cut into circular shapes (2cm diameter) were aseptically placed on the agar plates and incubated at 35 – 37°C for 24 hours. ZOIs associated with Acticoat, Silvercel and Medihoney™ Apinate dressings were compared with that of Atrauman (non-medicated control) dressing. All three dressings showed significant (p < 0.05) biofilm-inhibiting activity against both bacteria at antibiotic concentrations of 1024 and 5120µg/mL with ZOI between 17.5 and 35mm.

Background: The increasing resistance of Gram-negative bacteria isolated from nosocomial infections and chronic wounds, such as diabetic foot ulcers has renewed research interests in the use of polymyxins in the treatment of multidrug resistant infections. The added resistance conferred by biofilm development in such infections and the absence of novel antibiotics presuppose that polymyxins are the likely drugs of choice in spite of their nephrotoxicity. The effects of PMB and PMBN have been previously assessed on planktonic bacteria isolated from various infections. Methods: This current study assessed the synergy between a PMB/PMBN and two antibiotics (ceftazidime and levofloxacin) in an attempt to develop a strategy for biofilm disruption using the Minimum Biofilm Eradication Concentration Physiology and Genetic assay (MBEC™ P & G, Innovotech Inc, Edmonton, Alberta, Canada) according to manufacturer’s instructions. Klebsiella pneumoniae (K. pneumoniae) and Proteus mirabilis (P. mirabilis) biofilms of initial broth suspensions of 108 colony forming units per mL, cultivated on the pegs of the MBEC device were challenged with 5120 µg/mL of both ceftazidime and levofloxacin in a ten-fold dilution assay and in the presence of 100 and 500 µg/mL PMB and PMBN. Results: From table of results (Table 1), it can be deduced that both ceftazidime and levofloxacin are very effective in inhibiting biofilm development (as shown by percentage inhibition (PI)) when augmented with PMB and PMBN. This is about 100-fold increase in efficacy when compared to the antibiotics used on their own. The percentage reduction (PR) in biofilm was also increased considerably when PMB and PMBN concentrations were increased to 500 µg/mL. PMB was more effective than its less antibacterial derivative PMBN. Levofloxacin was also found to be more effective than ceftazidime when combined with both PMB and PMBN due to its enhanced cell-membrane permeability and as an anti-DNA replication uncoupling agent. Conclusion: The above results indicate that the synergy between antibiotics and cell membrane permeabilising agents may provide alternate strategies towards biofilm eradication

The principal aim of this study was to investigate the possibility of transference to Escherichia coli of β-lactam resistance genes found in bacteria isolated from ready-to-eat (RTE) Portuguese traditional food. From previous screenings, 128 β-lactam resistant isolates (from different types of cheese and of delicatessen meats), largely from the Enterobacteriaceae family were selected and 31.3% of them proved to transfer resistance determinants in transconjugation assays. Multiplex PCR in donor and transconjugant isolates did not detect bla CTX, bla SHV and bla OXY, but bla TEM was present in 85% of them, while two new TEMs (TEM-179 and TEM-180) were identified in two isolates. The sequencing of these amplicons showed identity between donor and transconjugant genes indicating in vitro plasmid DNA transfer. These results suggest that if there is an exchange of genes in natural conditions, the consumption of RTE foods, particularly with high levels of Enterobacteriaceae, can contribute to the spread of antibiotic resistance.

The ruthenium(II)-cymene complexes [Ru(eta(6)-cymene)(bha)Cl] with substituted halogenobenzohydroxamato (bha) ligands (substituents = 4-F, 4-Cl, 4-Br, 2,4-F-2, 3,4-F-2, 2,5-F-2, 2,6-F-2) have been synthesized and characterized by elemental analysis, IR, H-1 NMR, C-13 NMR, cyclic voltammetry and controlled-potential electrolysis, and density functional theory (DFT) studies. The compositions of their frontier molecular orbitals (MOs) were established by DFT calculations, and the oxidation and reduction potentials are shown to follow the orders of the estimated vertical ionization potential and electron affinity, respectively. The electrochemical E-L Lever parameter is estimated for the first time for the various bha ligands, which can thus be ordered according to their electron-donor character. All complexes exhibit very strong protein tyrosine kinase (PTK) inhibitory activity, even much higher than that of genistein, the clinically used PTK inhibitory drug. The complex containing the 2,4-difluorobenzohydroxamato ligand is the most active one, and the dependences of the PTK activity of the complexes and of their redox potentials on the ring substituents are discussed. Bone is constantly being molded and shaped by the action of osteoclasts and osteoblasts. A proper equilibrium between both cell types metabolic activities is required to ensure an adequate skeletal tissue structure, and it involves resorption of old bone and formation of new bone tissue. It is reported that treatment with antiepileptic drugs (AEDs) can elicit alterations in skeletal structure, in particular in bone mineral density. Nevertheless, the knowledge regarding the effects of AEDs on bone cells are still scarce. In this context, the aim of this study was to investigate the effects of five different AEDs on human osteoclastic, osteoblastic and co-cultured cells. Osteoclastic cell cultures were established from precursor cells isolated from human peripheral blood and were characterized for tartrate-resistant acid phosphatase (TRAP) activity, number of TRAP+ multinucleated cells, presence of cells with actin rings and expressing vitronectin and calcitonin receptors and apoptosis rate. Also, the involvement of several signaling pathways on the cellular response was addressed. Osteoblastic cell cultures were obtained from femur heads of patients (25-45 years old) undergoing orthopaedic surgery procedures and were then studied for cellular proliferation/viability, ALP activity, histochemical staining of ALP and apoptosis rate. Also the expression of osteoblast-related genes and the involvement of some osteoblastogenesis-related signalling pathways on cellular response were addressed. For co-cultured cells, osteoblastic cells were firstly seeded and cultured. After that, PBMC were added to the osteoblastic cells and co-cultures were evaluated using the same osteoclast and osteoblast parameters mentioned above for the corresponding isolated cell. Cell-cultures were maintained in the absence (control) or in the presence of different AEDs (carbamazepine, gabapentin, lamotrigine, topiramate and valproic acid). All the tested drugs were able to affect osteoclastic and osteoblastic cells development, although with different profiles on their osteoclastogenic and osteoblastogenic modulation properties. Globally, the tendency was to inhibit the process. Furthermore, the signaling pathways involved in the process also seemed to be differently affected by the AEDs, suggesting that the different drugs may affect osteoclastogenesis and/or osteoblastogenesis through different mechanisms. In conclusion, the present study showed that the different AEDs had the ability to directly and indirectly modulate bone cells differentiation, shedding new light towards a better understanding of how these drugs can affect bone tissue.

Recent changes in regulatory requirements and social views on animal testing have incremented the development of reliable alternative tests for predicting skin and ocular irritation potential of products based on new raw materials. In this regard, botanical ingredients used in cosmetic products are among those materials, and should be carefully reviewed concerning the potential presence of irritant constituents. In particular, cosmetic products used on the face, in vicinity of the eyes or that may come in contact with mucous membranes, should avoid botanical ingredients that contain, or are suspected to contain, such ingredients. In this study, we aimed to evaluate the effect of a new cosmetic ingredient, namely, coffee silverskin (CS), with an in vitro skin and ocular irritation assay using reconstructed human epidermis, EpiSkin™, and human corneal epithelial model, SkinEthics™ HCE, and an in vivo assay. Three different extracts of CS were evaluated. The histology of the models after extracts applications was analysed. The in vitro results demonstrated that extracts were not classified as irritant and the histological analyses proved that extracts did not affect both models structure. The content of caffeine, 5-hydroxymethyl furfural and chlorogenic acid was quantified after the epidermal assay. The in vivo test carried out with the most promising extract (hydroalcoholic) showed that, with respect to irritant effects, these extracts can be regarded as safe for topical application.

As leishmanioses são um grupo de doenças causadas pelo parasita protozoário Leishmania sp. Na Bacia mediterrânica, Leishmania infantum, é a principal espécie causadora de leishmaniose visceral, a forma mais severa da doença, sendo L. major um dos agentes etiológicos da leishmaniose cutânea. Apesar de se considerar que estes parasitas têm uma reprodução essencialmente clonal, nos últimos 20 anos tem vindo a ser descrita a recombinação genética entre diferentes estirpes e espécies, com ocorrência de híbridos naturais, quer no Velho quer no Novo Mundo. Recentemente, em Portugal, foram isoladas e identificadas pela primeira vez, estirpes híbridas de L. infantum/L. major. O presente estudo teve como principais objetivos, a pesquisa de “novas espécies” de Leishmania e a análise do comportamento “in vitro” de estirpes parentais e híbridas de L. infantum e L. major. Numa primeira parte do trabalho efetuou-se a cultura e pesquisa de DNA de Leishmania sp., em amostras de sangue medular de 229 cães provenientes de uma região endémica de Portugal, utilizando diferentes marcadores moleculares (kDNA, ITS1 e SSU rRNA) e protocolos de PCR. Não foi encontrado DNA de espécies híbridas, tendo-se no entanto, identificado DNA de Leishmania sp. em 45,85% (105/229) das amostras, incluindo cães sem sinais clínicos. Na segunda parte do trabalho, realizaram-se diversos ensaios “in vitro” com estirpes híbridas naturais L. infantum/L. major e parentais L. infantum e L. major. Em condições normais de crescimento, observou-se um padrão de crescimento distinto para cada estirpe estudada. Em condições de “stress” oxidativo, destacou-se uma diferença significativa entre as duas estirpes híbridas estudadas. Em condições de “stress” nutricional, as estirpes não apresentaram diferenças entre si. Após avaliação da suscetibilidade das estirpes na presença de Anfotericina B, todas se mostraram suscetíveis, com concentrações inibitórias (CI50) entre 0.21 e 1.15 μg/mL. Após infeção em linhas celulares monocíticas, não se verificaram diferenças estatisticamente significativas na taxa e intensidade de infeção das estirpes híbridas em comparação às putativas parentais. Os resultados obtidos, contribuíram para um melhor conhecimento sobre o comportamento biológico destas estirpes híbridas naturais L. infantum/L. major. Estas demonstraram um comportamento “in vitro” intermédio, relativamente às estirpes parentais. Estes resultados poderão servir de base para o desenvolvimento de outros estudos com estas “novas espécies”, nomeadamente estudos de patogenicidade “in vivo” e o papel de biomarcadores de virulência, que permitam um potencial prognóstico da infeção e avaliação do seu risco epidemiológico.

Neurological disorders are a major concern in modern societies, with increasing prevalence mainly related with the higher life expectancy. Most of the current available therapeutic options can only control and ameliorate the patients’ symptoms, often be-coming refractory over time. Therapeutic breakthroughs and advances have been hampered by the lack of accurate central nervous system (CNS) models. The develop-ment of these models allows the study of the disease onset/progression mechanisms and the preclinical evaluation of novel therapeutics. This has traditionally relied on genetically engineered animal models that often diverge considerably from the human phenotype (developmentally, anatomically and physiologically) and 2D in vitro cell models, which fail to recapitulate the characteristics of the target tissue (cell-cell and cell-matrix interactions, cell polarity). The in vitro recapitulation of CNS phenotypic and functional features requires the implementation of advanced culture strategies that enable to mimic the in vivo struc-tural and molecular complexity. Models based on differentiation of human neural stem cells (hNSC) in 3D cultures have great potential as complementary tools in preclinical research, bridging the gap between human clinical studies and animal models. This thesis aimed at the development of novel human 3D in vitro CNS models by integrat-ing agitation-based culture systems and a wide array of characterization tools. Neural differentiation of hNSC as 3D neurospheres was explored in Chapter 2. Here, it was demonstrated that human midbrain-derived neural progenitor cells from fetal origin (hmNPC) can generate complex tissue-like structures containing functional dopaminergic neurons, as well as astrocytes and oligodendrocytes. Chapter 3 focused on the development of cellular characterization assays for cell aggregates based on light-sheet fluorescence imaging systems, which resulted in increased spatial resolu-tion both for fixed samples or live imaging. The applicability of the developed human 3D cell model for preclinical research was explored in Chapter 4, evaluating the poten-tial of a viral vector candidate for gene therapy. The efficacy and safety of helper-dependent CAV-2 (hd-CAV-2) for gene delivery in human neurons was evaluated, demonstrating increased neuronal tropism, efficient transgene expression and minimal toxicity. The potential of human 3D in vitro CNS models to mimic brain functions was further addressed in Chapter 5. Exploring the use of 13C-labeled substrates and Nucle-ar Magnetic Resonance (NMR) spectroscopy tools, neural metabolic signatures were evaluated showing lineage-specific metabolic specialization and establishment of neu-ron-astrocytic shuttles upon differentiation. Chapter 6 focused on transferring the knowledge and strategies described in the previous chapters for the implementation of a scalable and robust process for the 3D differentiation of hNSC derived from human induced pluripotent stem cells (hiPSC). Here, software-controlled perfusion stirred-tank bioreactors were used as technological system to sustain cell aggregation and dif-ferentiation. The work developed in this thesis provides practical and versatile new in vitro ap-proaches to model the human brain. Furthermore, the culture strategies described herein can be further extended to other sources of neural phenotypes, including pa-tient-derived hiPSC. The combination of this 3D culture strategy with the implemented characterization methods represents a powerful complementary tool applicable in the drug discovery, toxicology and disease modeling.

The tubular transport of [3H]methotrexate was studied in isolated nonperfused and perfused superficial proximal tubular segments of rabbit kidneys. Reabsorption represented only 5% of perfused methotrexate, and appeared to be mostly of passive nature inasmuch as it was not modified by reducing the temperature or by ouabain. Cellular accumulation in nonperfused segments and secretion in perfused tubules were highest in the S2 segment and lower in the S3 and S1 segments. Secretion against a bath-to-lumen concentration gradient was observed only in S2 segments (with a maximum methotrexate secretory rate of 478 +/- 48 fmol/mm.min and an apparent Km of transport of 363 +/- 32 microM), and was inhibited by probenecid and folate. The low capacity for methotrexate secretion may be explained by a low capacity of transport across the basolateral membrane of the proximal cell as methotrexate was accumulated only to a low extent in nonperfused tubules (tissue water to medium concentration ratio of 8.2 +/- 1 in S2 segments). During secretion a small amount of methotrexate was metabolized; the nature of the metabolite(s) remains to be defined.

OBJECTIVE: : To determine the influence of nebulizer types and nebulization modes on bronchodilator delivery in a mechanically ventilated pediatric lung model. DESIGN: : In vitro, laboratory study. SETTING: : Research laboratory of a university hospital. INTERVENTIONS: : Using albuterol as a marker, three nebulizer types (jet nebulizer, ultrasonic nebulizer, and vibrating-mesh nebulizer) were tested in three nebulization modes in a nonhumidified bench model mimicking the ventilatory pattern of a 10-kg infant. The amounts of albuterol deposited on the inspiratory filters (inhaled drug) at the end of the endotracheal tube, on the expiratory filters, and remaining in the nebulizers or in the ventilator circuit were determined. Particle size distribution of the nebulizers was also measured. MEASUREMENTS AND MAIN RESULTS: : The inhaled drug was 2.8% ± 0.5% for the jet nebulizer, 10.5% ± 2.3% for the ultrasonic nebulizer, and 5.4% ± 2.7% for the vibrating-mesh nebulizer in intermittent nebulization during the inspiratory phase (p < 0.01). The most efficient nebulizer was the vibrating-mesh nebulizer in continuous nebulization (13.3% ± 4.6%, p < 0.01). Depending on the nebulizers, a variable but important part of albuterol was observed as remaining in the nebulizers (jet and ultrasonic nebulizers), or being expired or lost in the ventilator circuit (all nebulizers). Only small particles (range 2.39-2.70 µm) reached the end of the endotracheal tube. CONCLUSIONS: : Important differences between nebulizer types and nebulization modes were seen for albuterol deposition at the end of the endotracheal tube in an in vitro pediatric ventilator-lung model. New aerosol devices, such as ultrasonic and vibrating-mesh nebulizers, were more efficient than the jet nebulizer.

In previous work we found that mezerein, a C kinase activator, as well as basic fibroblast growth factor (FGF-2) induce demyelination and partial oligodendrocyte dedifferentiation in highly differentiated aggregating brain cell cultures. Here we show that following protein kinase C activator-induced demyelination, effective remyelination occurs. We found that mezerein or FGF-2 caused a transient increase in DNA synthesis following a pronounced decrease of the myelin markers myelin basic protein and 2',3'-cyclic nucleotide 3'-phosphohydrolase. Both oligodendrocytes and astrocytes were involved in this mitogenic response. Within 17 days after demyelination, myelin was restored to the level of the untreated controls. Transient mitotic activity was indispensable for remyelination. The present results suggest that myelinating oligodendrocytes retain the capacity to reenter the cell cycle, and that this plasticity is important for the regeneration of the oligodendrocyte lineage and remyelination. Although it cannot be excluded that a quiescent population of oligodendrocyte precursor cells was present in the aggregates and able to proliferate, differentiate and remyelinate, we could not find evidence supporting this view.
