We describe methods for obtaining a quantitative description of RNA processing at high resolution in budding yeast. As a model gene expression system, we constructed tetON (for induction studies) and tetOFF (for repression, derepression, and RNA degradation studies) yeast strains with a series of reporter genes integrated in the genome under the control of a tetO7 promoter. Reverse transcription and quantitative real-time-PCR (RT-qPCR) methods were adapted to allow the determination of mRNA abundance as the average number of copies per cell in a population. Fluorescence in situ hybridization (FISH) measurements of transcript numbers in individual cells validated the RT-qPCR approach for the average copy-number determination despite the broad distribution of transcript levels within a population of cells. In addition, RT-qPCR was used to distinguish the products of the different steps in splicing of the reporter transcripts, and methods were developed to map and quantify 3′-end cleavage and polyadenylation. This system permits pre-mRNA production, splicing, 3′-end maturation and degradation to be quantitatively monitored with unprecedented kinetic detail, suitable for mathematical modeling. Using this approach, we demonstrate that reporter transcripts are spliced prior to their 3′-end cleavage and polyadenylation, that is, cotranscriptionally.

Optimistic fair exchange (OFE) allows two parties to exchange their digital items in a fair way. As one of the fundamental problems in secure electronic business and digital rights management, OFE has been studied intensively since its introduction. This paper introduces and defines a new property for OFE: Strong Resolution-Ambiguity. We show that many existing OFE protocols have the new property, but its formal investigation has been missing in those protocols. We prove that in the certified-key model, an OFE protocol is secure in the multi-user setting if it is secure in the single-user setting and has the property of strong resolution-ambiguity. Our result not only simplifies the security analysis of OFE protocols in the multi-user setting but also provides a new approach for the design of multi-user secure OFE protocols. Following this approach, a new OFE protocol with strong resolution-ambiguity is proposed. Our analysis shows that the protocol is setup-free, stand-alone and multi-user secure without random oracles.

We propose a combined character separation and recognition approach for low-resolution images of alphanumeric text. By synthesising the image formation process a set of low-resolution templates is created for each character. Cluster algorithms and normalised cross-correlation are then applied to match these templates and thereby allowing both character separation and recognition to be achieved at the same time. Thus characters are recognised using their low-resolution appearance only without applying image enhancement methods. Experiments showed that this approach is able to recognise low-resolution alphanumeric text of down to 5 pixels in size.

Wide area surveillance requires high-resolution images of the object of interest derived possibly from only low-resolution video of the whole scene. We propose a combined tracking and resolution enhancement approach that increases the resolution of the object of interest during tracking. The key idea is the use of an object-specific 3D mesh model with which we are able to track non-planar objects across a large number of frames. This model is subdivided such that every triangle is smaller than a pixel when projected into the image to facilitate super-resolution on the model rather than on the image. We apply our approach to faces and show that it outperforms interpolation methods by achieving resolution enhancement, while being less blurred.

We describe a novel video player that uses Temporal Semantic Compression (TSC) to present a compressed summary of a movie. Compression is based on tempo which is derived from film rhythms. The technique identifies periods of action, drama, foreshadowing and resolution, which can be mixed in different amounts to vary the kind of summary presented. The compression algorithm is embedded in a video player, so that the summary can be interactively recomputed during playback.

We examine the construction of new filters for computing local energy, and compare these filters with the Gabor filters and the three-point-filter of Venkatesh [l]. Further, we demonstrate that the effect of convolution with complex Gabor filters is to band-pass (with some differentiating effect) and compute the local energy of the result. The magnitude of the resulting local energy is then used to detect features [2], [3] (step features, texture etc.), and the phase is used to classify the detected features [l], [4] or provide disparity information for stereo [5] and motion work [6], [7]. Each of these types of information can be obtained at multiple resolutions, enabling the use of course to fine strategies for computing disparity, and allowing the discrimination of image textures on the basis of which parts of the Fourier domain they dominate [8], [9].

Log polar transformations for space variant systems have been proposed and used in active vision research. The idea is to generate an image with a varying resolution over a wide angle field of view. The fovea is of high resolution and the periphery is of exponentially reduced resolution. The justifications for such a sensor are: (i) it provides high resolution and a wide viewing angle, (ii) feature invariance in the fovea simplifies foveation, and (iii) it allows multiresolution analysis. The receptor density of the human retina is very high, i.e. of the order of 106 receptors at the fovea. The question is, what resolution should space variant active vision systems have? Real visual sensors have been implemented but is the resolution produced high enough? This paper investigates the resolution requirements of a space variant sensor by simulation for a tracking system using raytracing

Numerous authors have expressed concerns that the introduction of the Personally Controlled Electronic Health Record (PCEHR) will lead to an escalation of disputes. Some disputes will concern the accuracy of the record whereas others will arise simply due to greater access to health care records. Online dispute resolution (ODR) programs have been successfully applied to cost-effectively help disputants resolve commercial, insurance and other legal disputes, and can also facilitate the resolution of health care related disputes. However, we expect that health differs from other application domains in ODR because of the emotional engagement patients have with their health and those of loved ones. In this study we will be looking at whether the success of an online negotiation is related to how people recognise and manage emotions, and in particular, their Emotional intelligence score.
