The aim of this study was to determine the effectiveness of the acaricide tri-n-butyl tin maleate, industrially applied to samples of carpets, mattress foam, and fabrics used for furniture upholstery, soft toys and shoe uppers. Approximately 100 adult house dust mites of the species Dermatophagoides pteronyssinus were inoculated into a Petri dish containing the sample (a piece of carpet, mattress foam, or fabric), treated with the acaricide, randomly collected. Mite-maintenance culture medium was added on top of each sample. After one, two, three, seven and 30 days of incubation at 25 ºC and 75% relative humidity, each dish was examined using a 40X stereoscopic microscope (40X). One hundred percent acaricide effectiveness was obtained in treated materials by the end of the 30th-day postinoculation period, under optimal conditions for mite maintenance.

Introdução: Um terço das mulheres com diabetes gestacional terá o diagnóstico de diabetes ou alteração do metabolismo da glicose no rastreio pós-parto. Objectivo: Avaliar a percentagem de mulheres submetidas a rastreio pós-parto e associar o resultado com a história materna. Métodos: Estudo retrospectivo de 1013 gravidezes com diabetes gestacional (2005-2009). Dividiu-se a população em dois grupos de acordo com o resultado: normal (grupo 1) e com diabetes ou alteração do metabolismo da glicose (grupo 2). Para ambos os grupos foram avaliados: idade materna, índice de massa corporal, ganho de peso na gravidez, idade gestacional do diagnóstico, necessidade de administração de insulina, factores de risco para diabetes gestacional e peso do recém-nascido. Resultados: O rastreio pós-parto foi realizado em 76,8% das mulheres (n=778). O teste foi considerado normal (grupo 1) em 628 mulheres(80,7%) e alterado (grupo 2) em 150 mulheres (19,3%). O Grupo 2 teve mulheres mais velhas (idade média de 34 vs 33 anos;p-value 0,013), com maior índice de massa corporal (28,5 vs 25,8kg / cm2; p-value 0,000), maior número de mulheres com história familiar em primeiro grau de diabetes mellitus (50,3% vs 39,9%; p-value 0,026) e história pessoal de macrossomia prévia (12,1% vs 5,4%; p-value 0,003). O diagnóstico mais precoce da diabetes gestacional foi também feito nesse grupo (27 vs 31 semanas; p-value 0,000) e uma maior percentagem efectuou insulina (41% vs 15%; p-value 0,000), tendo iniciado mais cedo a sua administração (28 vs 30 semanas; p-value 0,010). Verificou-se uma maior percentagem de grávidas multíparas no grupo 2 (64% vs 49,4%; p-value = 0,001) e um maior número de casos de recém-nascidos grandes para a idade gestacional (17,1% vs 8,3%; p-value = 0,001). A história pessoal de diabetes gestacional e ganho de peso durante a gestação foi semelhante nos dois grupos. Conclusões: As mulheres com alteração nos resultados do rastreio pós-parto são geralmente mais velhas, mais pesadas, multíparas, com história familiar em primeiro grau de diabetes Mellitus e história pessoal de macrossomia prévia. O diagnóstico de diabetes gestacional foi mais precoce neste grupo, mais frequentemente necessitaram de terapêutica com insulina com início mais cedo e verificou-se um maior número de recém-nascidos grandes para a idade gestacional.

A Mitomicina C está entre uma variedade de agentes quimioterapêuticos intravesicais actualmente disponíveis para o tratamento do carcinoma superficial da bexiga. Cerca de9% destes doentes desenvolve reacções adversas cutâneas, geralmente dermites de contacto, localizadas nas mãos, pés, genitais, ou erupções mais disseminadas. Descrevem-se 6 casos de dermite de contacto alérgica à Mitomicina C, observados entre Junho/2004 e Março/2005,emcinco doentes do sexo masculino e umdo sexo feminino, com uma idade média de 70 anos.

Toxic effects of ultraviolet (UV) radiation on skin include protein and lipid oxidation, and DNA damage. The latter is known to play a major role in photocarcinogenesis and photoaging. Many plant extracts and natural compounds are emerging as photoprotective agents. Castanea sativa leaf extract is able to scavenge several reactive species that have been associated to UV-induced oxidative stress. The aim of this work was to analyze the protective effect of C. sativa extract (ECS) at different concentrations (0.001, 0.01, 0.05 and 0.1 μg/mL) against the UV mediated-DNA damage in a human keratinocyte cell line (HaCaT). For this purpose, the cytokinesis-block micronucleus assay was used. Elucidation of the protective mechanism was undertaken regarding UV absorption, influence on 1O2 mediated effects or NRF2 activation. ECS presented a concentration-dependent protective effect against UV-mediated DNA damage in HaCaT cells. The maximum protection afforded (66.4%) was achieved with the concentration of 0.1 μg/mL. This effect was found to be related to a direct antioxidant effect (involving 1O2) rather than activation of the endogenous antioxidant response coordinated by NRF2. Electrochemical studies showed that the good antioxidant capacity of the ECS can be ascribed to the presence of a pool of different phenolic antioxidants. No genotoxic or phototoxic effects were observed after incubation of HaCaT cells with ECS (up to 0.1 μg/mL). Taken together these results reinforce the putative application of this plant extract in the prevention/minimization of UV deleterious effects on skin.

Toxocariasis is a worldwide public-health problem that poses major risks to children who may accidentally ingest embryonated eggs of Toxocara. The objectives of this study were to investigate the occurrence of anti-Toxocara spp. antibodies in children and adolescents and the variables that may be involved, as well as environmental contamination by Toxocara spp. eggs, in urban recreation areas of north central mesoregion, Paraná State, Brazil. From June 2005 to March 2007. a total of 376 blood samples were collected by the Public Health Service from children and adolescents one to 12 years old, of both genders. Samples were analyzed by the indirect ELISA method for detection of anti-Toxocara antibodies. Serum samples were previously absorbed with Ascaris suum antigens, and considered positive with a reagent reactivity index >1. Soil samples from all of the public squares and schools located in the four evaluated municipalities that had sand surfaces (n = 19) or lawns (n = 15) were analyzed. Of the 376 serum samples, 194 (51.6%) were positive. The seroprevalence rate was substantially higher among children aging one to five years (p = 0.001) and six to eight years (p = 0.022). The clinical signs and symptoms investigated did not show a statistical difference between seropositive and seronegative individuals (p > 0.05). In 76.5% of the investigated recreation places, eggs of Toxocara were detected in at least one of the five collected samples. Recreation areas from public schools were 2.8 times more contaminated than from public squares. It is important to institute educational programs to inform families and educators, as well as to improve sanitary control of animals and cleaning of the areas intended for recreation in order to prevent toxocariasis.

The in vitro and in vivo activity of diminazene (Dim), artesunate (Art) and combination of Dim and Art (Dim-Art) against Leishmania donovani was compared to reference drug; amphotericin B. IC50 of Dim-Art was found to be 2.28 ± 0.24 µg/mL while those of Dim and Art were 9.16 ± 0.3 µg/mL and 4.64 ± 0.48 µg/mL respectively. The IC50 for Amphot B was 0.16 ± 0.32 µg/mL against stationary-phase promastigotes. In vivo evaluation in the L. donovani BALB/c mice model indicated that treatments with the combined drug therapy at doses of 12.5 mg/kg for 28 consecutive days significantly (p < 0.001) reduced parasite burden in the spleen as compared to the single drug treatments given at the same dosages. Although parasite burden was slightly lower (p < 0.05) in the Amphot B group than in the Dim-Art treatment group, the present study demonstrates the positive advantage and the potential use of the combined therapy of Dim-Art over the constituent drugs, Dim or Art when used alone. Further evaluation is recommended to determine the most efficacious combination ratio of the two compounds.

An efficient method for breeding Biomphalaria tenagophila (Taim lineage/RS) was developed over a 5-year-period (2005-2010). Special facilities were provided which consisted of four cement tanks (9.4 x 0.6 x 0.22 m), with their bottom covered with a layer of sterilized red earth and calcium carbonate. Standard measures were adopted, as follows: each tank should contain an average of 3000 specimens, and would be provided with a daily ration of 35,000 mg complemented with lettuce. A green-house effect heating system was developed which constituted of movable dark canvas covers, which allowed the temperature to be controlled between 20 - 24 ºC. This system was essential, especially during the coldest months of the year. Approximately 27,000 specimens with a diameter of 12 mm or more were produced during a 14-month-period. The mortality rates of the newly-hatched and adult snails were 77% and 37%, respectively. The follow-up of the development system related to 310 specimens of B. tenagophila demonstrated that 70-day-old snails reached an average of 17.0 ± 0.9 mm diameter. The mortality rates and the development performance of B. tenagophila snails can be considered as highly satisfactory, when compared with other results in literature related to works carried out with different species of the genus Biomphalaria, under controlled laboratory conditions.

A presente dissertação foi realizada no âmbito do Mestrado de Engenharia e Gestão Industrial da Escola Superior de Estudos Industriais e de Gestão, de Vila do Conde. O projeto desenvolvido tem como tema principal a Otimização de processos de Logística in-house baseado num projeto, em contexto empresarial da empresa cliente, Continental Mabor S.A., da Rangel Distribuição e Logística, S.A. Este projeto tem como objetivo a “aglomeração” de dois armazéns do cliente, devido à necessidade de ocupação do armazém de produto acabado interno, para aumento da área de produção. Inicialmente foi feita uma revisão de literatura sobre os temas mais relevantes de suporte para o projeto, nomeadamente na otimização e melhoria contínua. Seguidamente é apresentado o Grupo Rangel, bem como a Rangel Distribuição e Logística, S.A., onde se enquadra o projeto e para se perceber o enquadramento e objetivo. A metodologia usada, caso de estudo, permitiu a aplicação de conceitos e ferramentas usados na literatura neste contexto, como ferramentas de otimização e melhoria continua como as melhores práticas de Kaizen-Lean. Na fase de diagnóstico do atual sistema, foi realizado um mapeamento de fluxo de processos e uma descrição detalhada do layout dos dois armazéns: Armazém de Produto Acabado (APA) e Armazém de Produto Acabado Externo (APAE), bem como todos os recursos, quer técnicos quer humanos necessários. Verificamos ao longo deste projeto várias limitações, inclusive limitações impostas pelo cliente, tal como não aprovar um estudo para um novo layout do armazém. Foi aprovado apenas a replicação do já existente. Com isto, depararam-se constrangimentos na gestão deste projeto. Os custos aumentaram significativamente, embora estes não sejam apresentados por questões de confidencialidade, principalmente com a necessidade de aquisição de novos equipamentos retráteis, e mais baterias para os mesmos, devido às grandes distâncias que irão ser percorridas. Finalmente foi projetado o sistema futuro, de acordo com as necessidades reais do cliente tendo em consideração a otimização de recursos e uma gestão magra (Lean Management). Foi desenvolvida a implementação da metodologia “Kaizen diário”, a dar início em 2016 juntamente com o novo projeto APAE. Com esta projeção foram identificados problemas e implicações no projeto, bem como possíveis melhorias.

Background: Although most HIV-1 infections in Brazil are due to subtype B, Southern Brazil has a high prevalence of subtype C and recombinant forms, such as CRF31_BC. This study assessed the impact of viral diversity on clinical progression in a cohort of newly diagnosed HIV-positive patients. Methods: From July/2004 to December/2005, 135 HIV-infected patients were recruited. The partial pol region was subtyped by phylogeny. A generalized estimating equation (GEE) model was used to examine the relationship between viral subtype, CD4+ T cell count and viral load levels before antiretroviral therapy. Hazard ratio (Cox regression) was used to evaluate factors associated with viral suppression (viral load < 50 copies/mL at six months). Results: Main HIV-1 subtypes included B (29.4%), C (28.2%), and CRF31_BC (23.5%). Subtypes B and C showed a similar trend in CD4+ T cell decline. Comparison of non-B (C and CRF31_BC) and B subtypes revealed no significant difference in the proportion of patients with viral suppression at six months (week 24). Higher CD4+ T cell count and lower viral load were independently associated with viral suppression. Conclusion: No significant differences were found between subtypes; however, lower viral load and higher CD4+ T cell count before therapy were associated with better response.

Nesta dissertação é apresentado um estudo dos sistemas de processamento automático de imagem em contexto de um problema relacionado com a individualização de neurónios em imagens da nematoda C. elegans durante estudos relacionados com a doença de Parkinson. Apresenta-se uma breve introdução à anatomia do verme, uma introdução à doença de Parkinson e uso do C. elegans em estudos relacionados e também é feita a análise de artigos em contexto de processamento de imagem para contextualizar a situação atual de soluções para o problema de extração de características e regiões específicas. Neste projeto é desenvolvida uma pipeline com o auxilio do software CellProfiler para procurar uma resposta para o problema em questão.

Background: Indoor air quality (IAQ) is considered an important determinant of human health. The association between exposure to volatile organic compounds, particulate matter, house dust mite, molds and bacteria in day care centers (DCC) is not completely clear. The aim of this project was to study these effects. Methods --- study design: This study comprised two phases. Phase I included an evaluation of 45 DCCs (25 from Lisbon and 20 from Oporto, targeting 5161 children). In this phase, building characteristics, indoor CO2 and air temperature/relative humidity, were assessed. A children’s respiratory health questionnaire derived from the ISAAC (International Study on Asthma and Allergies in Children) was also distributed. Phase II encompassed two evaluations and included 20 DCCs selected from phase I after a cluster analysis (11 from Lisbon and 9 from Oporto, targeting 2287 children). In this phase, data on ventilation, IAQ, thermal comfort parameters, respiratory and allergic health, airway inflammation biomarkers, respiratory virus infection patterns and parental and child stress were collected. Results: In Phase I, building characteristics, occupant behavior and ventilation surrogates were collected from all DCCs. The response rate of the questionnaire was 61.7% (3186 children). Phase II included 1221 children. Association results between DCC characteristics, IAQ and health outcomes will be provided in order to support recommendations on IAQ and children’s health. A building ventilation model will also be developed. Discussion: This paper outlines methods that might be implemented by other investigators conducting studies on the association between respiratory health and indoor air quality at DCC.

ABSTRACT:C-reactive protein (CRP) has been widely used in the early risk assessment of patients with acute pancreatitis (AP), but unclear aspects about its prognostic accuracy in this setting persist. This project evaluated first CRP prognostic accuracy for severity, pancreatic necrosis (PNec), and in-hospital mortality (IM) in AP in terms of the best timing for CRP measurement and the optimal CRP cutoff points. Secondly it was evaluated the CRP measured at approximately 24 hours after hospital admission (CRP24) prognostic accuracy for IM in AP individually and in a combined model with a recent developed tool for the early risk assessment of patients with AP, the Bedside Index for Severity in AP (BISAP). Two single-centre retrospective cohort studies were held. The first study included 379 patients and the second study included 134 patients. Statistical methods such as the Hosmer-Lemeshow goodness-of-fit test, the area under the receiver-operating characteristic curve, the net reclassification improvement, and the integrated discrimination improvement were used. It was found that CRP measured at approximately 48 hours after hospital admission (CRP48) had a prognostic accuracy for severity, PNec, and IM in AP better than CRP measured at any other timing. It was observed that the optimal CRP48 cutoff points for severity, PNec, and IM in AP varied from 170mg/l to 190mg/l, values greater than the one most often recommended in the literature – 150mg/l. It was found that CRP24 had a good prognostic accuracy for IM in AP and that the cutoff point of 60mg/l had a negative predictive value of 100%. Finally it was observed that the prognostic accuracy of a combined model including BISAP and CRP24 for IM in AP could perform better than the BISAP alone model. These results might have a direct impact on the early risk assessment of patients with AP in the daily clinical practice.--------- RESUMO: A proteina c-reactiva (CRP) tem sido largamente usada na avaliação precoce do risco em doentes com pancreatite aguda (AP), mas aspectos duvidosos acerca do seu valor prognóstico neste contexto persistem. Este projecto avaliou primeiro o valor prognóstico da CRP para a gravidade, a necrose pancreática (PNec) e a mortalidade intra-hospitalar (IM) na AP em termos do melhor momento para efectuar a sua medição e dos seus pontos-de-corte óptimos. Em segundo lugar foi avaliado o valor prognóstico da proteína c-reactiva medida aproximadamente às 24 horas após a admissão hospitalar (CRP24) para a IM na AP isoladamente e num modelo combinado, que incluiu uma ferramenta de avaliação precoce do risco em doentes com AP recentemente desenvolvida, o Bedside Index for Severity in Acute Pancreatitis (BISAP). Dois estudos unicêntricos de coorte retrospectivo foram realizados. O primeiro estudo incluiu 379 doentes e o segundo estudo incluiu 134 doentes. Metodologias estatísticas como o teste de Hosmer-Lemeshow goodness-of-fit, a area under the receiver-operating characteristic curve, o net reclassification improvement e o integrated discrimination improvement foram usadas. Verificou-se que a CRP medida às 48 horas após a admissão hospitalar (CRP48) teve um valor prognóstico para a gravidade, a PNec e a IM na AP melhor do que a CRP medida em qualquer outro momento. Observou-se que os pontos de corte óptimos da CRP48 para a gravidade, a PNec e a IM na AP variaram entre 170mg/l e 190mg/l, valores acima do valor mais frequentemente recomendado na literatura – 150mg/l. Verificou-se que a CRP medida aproximadamente às 24 horas após a admissão hospitalar (CRP24) teve um bom valor prognóstico para a IM na AP e que o ponto de corte 60mg/l teve um valor preditivo negativo de 100%. Finalmente observou-se que o valor prognóstico de um modelo combinado incluindo o BISAP e a CRP24 para a IM na AP pode ter um desempenho melhor do que o do BISAP isoladamente. Estes resultados podem ter um impacto directo na avaliação precoce do risco em doentes com AP na prática clínica diária.
