The Los Negritos porphyry copper deposit is located ~ 4 km to the northeast of Carmen de Andacollo Mine in the Chilean Cretaceous metallogenic belt. The mineralization is hosted in andesite of the Quebrada Marquesa Formation and a series of at least four early to intramineral porphyry intrusive rock types: plagioclase quartz biotite porphyry (P1b and P1a dated at 109.60± 0.75 Ma and 107.22± 0.40 Ma); plagioclase biotite porphyry (P2: 106.30 ± 0.47 Ma); and quartz plagioclase biotite porphyry (P3: 106.19 ± 0.42 Ma). These units are cut by late‐ to post‐mineral plagioclase‐hornblende porphyritic rocks (P4b: 106.20 ± 0.69 Ma and P4a: 106.50 ± 0.68 Ma). The earliest intrusive units (P1) were affected by an initial stage of K‐feldspar‐biotite alteration, with chalcopyrite, molybdenite (date at 108.5 ± 0.5 Ma) and gold (up to 0.11 ppm), and the surrounding volcanic host rock was overprinted by chlorite‐epidote dominated (propylitic) alteration. Subsequent to the P2 and P3 intrusion, these rocks were affected by albite and then a second stage of potassic alteration. The Ti and Ba contents in hydrothermal biotite are notably lower (typically Ti = 0.100‐0.144 a.p.f.u. and Ba = 0.001‐0.005 a.p.f.u) than in magmatic ones (generally Ti = 0.186‐0.222 a.p.f.u. and Ba = 0.014‐0.023 a.p.f.u.), and constitute an excellent discriminant of the nature of biotite. These early stages of alteration were overprinted by copper‐molybdenum bearing chlorite‐sericite alteration at 106.60 ± 0.5 Ma (Re‐Os age in molybdenite) and by quartz‐sericite‐pyrite veins (phyllic), respectively in the southwest and northeast areas. The average temperature associated with these two alteration facies is estimated around 305 °C. Weak albite‐calcite alteration, spatially associated with sulfosalts and distributed along the margins of P3, overprinted the phyllic facies. The intrusive rock units at the Los Negritos and Carmen de Andacollo deposits are geochemically classified as diorite to granodiorite with a calc‐alkaline magmatic affinity, and formed in a volcanic arc setting from partial melting of a metasomatized mantle wedge. They are interpreted to be cogenetic, and related to a common long‐lived magma chamber that emplaced during a period of tectonic inversion known as the Subhercynian, Peruvian or Pacific event.

Amphibian skin secretions contain biologically-active compounds, such as anti-microbial peptides and trypsin inhibitors, which are used by biomedical researchers as a source of potential novel drug leads or pharmacological agents. Here, we report the application of a recently developed technique within our laboratory to “shotgun” clone the cDNAs encoding two novel but structurally-related peptides from the lyophilized skin secretions of one species of European frog, Rana esculenta and one species of Chinese frog, Odorrana schmackeri. Bioanalysis of the peptides established the structure of a 17-mer with an N-terminal Ala (A) residue and a C-terminal Cys (C) residue with a single disulphide bridge between Cys 12 and 17, which is a canonical Kunitz-type protease inhibitor motif (-CKAAFC-). Due to the presence of this structural attribute, these peptides were named kunitzin-RE (AAKIILNPKFRCKAAFC) and kunitzin-OS (AVNIPFKVHLRCKAAFC). Synthetic replicates of these two novel peptides were found to display a potent inhibitory activity against Escherichia coli but were ineffective at inhibiting the growth of Staphylococcus aureus and Candida albicans at concentrations up to 160 μM, and both showed little haemolytic activity at concentrations up to 120 μM. Subsequently, kunitzin-RE and kunitzin-OS were found to be a potent inhibitor of trypsin with a Ki of 5.56 μM and 7.56 μM that represent prototypes of a novel class of highly-attenuated amphibian skin protease inhibitor. Substitution of Lys-13, the predicted residue occupying the P1 position within the inhibitory loop, with Phe (F) resulted in decrease in trypsin inhibitor effectiveness and antimicrobial activity against Esherichia coli, but exhibits a potential inhibition activity against chymotrypsin.

Kunitz-type (KT) protease inhibitors are low molecular weight proteins classically defined as serine protease inhibitors. We identified a novel secreted KT inhibitor associated with the gut and parenchymal tissues of the infective juvenile stage of Fasciola hepatica, a helminth parasite of medical and veterinary importance. Unexpectedly, recombinant KT inhibitor (rFhKT1) exhibited no inhibitory activity towards serine proteases but was a potent inhibitor of the major secreted cathepsin L cysteine proteases of F. hepatica, FhCL1 and FhCL2, and of human cathepsins L and K (Ki = 0.4 nM - 27 nM). FhKT1 prevented the auto-catalytic activation of FhCL1 and FhCL2 and formed stable complexes with the mature enzymes. Pull-down experiments from adult parasite culture medium showed that rFhKT1 interacts specifically with native secreted FhCL1, FhCL2 and FhCL5. Substitution of the unusual P1 Leu15 within the exposed reactive loop of FhKT1 for the more commonly found Arg (FhKT1Leu15/Arg15) had modest adverse effects on the cysteine protease inhibition but conferred potent activity against the serine protease trypsin (Ki = 1.5 nM). Computational docking and sequence analysis provided hypotheses for the exclusive binding of FhKT1 to cysteine proteases, the importance of the Leu15 in anchoring the inhibitor into the S2 active site pocket, and the inhibitor's selectivity towards FhCL1, FhCL2 and human cathepsins L and K. FhKT1 represents a novel evolutionary adaptation of KT protease inhibitors by F. hepatica, with its prime purpose likely in the regulation of the major parasite-secreted proteases and/or cathepsin L-like proteases of its host.

Purpose: Activation of the transient receptor potential channels, TRPC6, TRPM4, and TRPP1 (PKD2), has been shown to contribute to the myogenic constriction of cerebral arteries. In the present study we sought to determine the potential role of various mechanosensitive TRP channels to myogenic signaling in arterioles of the rat retina.

Methods: Rat retinal arterioles were isolated for RT-PCR, Fura-2 Ca2+ microfluorimetry, patch-clamp electrophysiology, and pressure myography studies. In some experiments, confocal immunolabeling of wholemount preparations was used to examine the localization of specific mechanosensitive TRP channels in retinal vascular smooth muscle cells (VSMCs).

Results: Reverse transcription-polymerase chain reaction analysis demonstrated mRNA expression for TRPC1, M7, V1, V2, V4, and P1, but not TRPC6 or M4, in isolated retinal arterioles. Immunolabeling revealed plasma membrane, cytosolic and nuclear expression of TRPC1, M7, V1, V2, V4, and P1 in retinal VSMCs. Hypoosmotic stretch-induced Ca2+ influx in retinal VSMCs was reversed by the TRPV2 inhibitor tranilast and the nonselective TRPP1/V2 antagonist amiloride. Inhibitors of TRPC1, M7, V1, and V4 had no effect. Hypoosmotic stretch-activated cation currents were similar in Na+ and Cs+ containing solutions suggesting no contribution by TRPP1 channels. Direct plasma membrane stretch triggered cation current activity that was blocked by tranilast and specific TRPV2 pore-blocking antibodies and mimicked by the TRPV2 activator, Δ9-tetrahydrocannabinol. Preincubation of retinal arterioles with TRPV2 blocking antibodies prevented the development of myogenic tone.

Conclusions: Our results suggest that retinal VSMCs express a range of mechanosensitive TRP channels, but only TRPV2 appears to contribute to myogenic signaling in this vascular bed.

L’objectif principal de cette thèse était d’obtenir, via l’électrophysiologie cognitive, des indices de fonctionnement post-traumatisme craniocérébral léger (TCCL) pour différents niveaux de traitement de l’information, soit l’attention sélective, les processus décisionnels visuoattentionnels et les processus associés à l’exécution d’une réponse volontaire. L’hypothèse centrale était que les mécanismes de production des lésions de même que la pathophysiologie caractérisant le TCCL engendrent des dysfonctions visuoattentionnelles, du moins pendant la période aiguë suivant le TCCL (i.e. entre 1 et 3 mois post-accident), telles que mesurées à l’aide d’un nouveau paradigme électrophysiologique conçu à cet effet. Cette thèse présente deux articles qui décrivent le travail effectué afin de rencontrer ces objectifs et ainsi vérifier les hypothèses émises. Le premier article présente la démarche réalisée afin de créer une nouvelle tâche d’attention visuospatiale permettant d’obtenir les indices électrophysiologiques (amplitude, latence) et comportementaux (temps de réaction) liés aux processus de traitement visuel et attentionnel précoce (P1, N1, N2-nogo, P2, Ptc) à l’attention visuelle sélective (N2pc, SPCN) et aux processus décisionnels (P3b, P3a) chez un groupe de participants sains (i.e. sans atteinte neurologique). Le deuxième article présente l’étude des effets persistants d’un TCCL sur les fonctions visuoattentionelles via l’obtention des indices électrophysiologiques ciblés (amplitude, latence) et de données comportementales (temps de réaction à la tâche et résultats aux tests neuropsychologiques) chez deux cohortes d’individus TCCL symptomatiques, l’une en phase subaigüe (3 premiers mois post-accident), l’autre en phase chronique (6 mois à 1 an post-accident), en comparaison à un groupe de participants témoins sains. Les résultats des articles présentés dans cette thèse montrent qu’il a été possible de créer une tâche simple qui permet d’étudier de façon rapide et peu coûteuse les différents niveaux de traitement de l’information impliqués dans le déploiement de l’attention visuospatiale. Par la suite, l’utilisation de cette tâche auprès d’individus atteints d’un TCCL testés en phase sub-aiguë ou en phase chronique a permis d’objectiver des profils d’atteintes et de récupération différentiels pour chacune des composantes étudiées. En effet, alors que les composantes associées au traitement précoce de l’information visuelle (P1, N1, N2) étaient intactes, certaines composantes attentionnelles (P2) et cognitivo-attentionnelles (P3a, P3b) étaient altérées, suggérant une dysfonction au niveau des dynamiques spatio-temporelles de l’attention, de l’orientation de l’attention et de la mémoire de travail, à court et/ou à long terme après le TCCL, ceci en présence de déficits neuropsychologiques en phase subaiguë surtout et d’une symptomatologie post-TCCL persistante. Cette thèse souligne l’importance de développer des outils diagnostics sensibles et exhaustifs permettant d’objectiver les divers processus et sous-processus cognitifs susceptible d’être atteints après un TCCL.

Im Rahmen dieser Arbeit wurden selektive Inhibitoren der Glutathion-Transferase P1 (GSTP1) mit 1,2,4-Trioxanstruktur als potentielle Wirkstoffe gegen multiresistente Tumore synthetisiert. Die Darstellung dieser Substanzen erfolgte über Typ-II-Photooxygenierung allylischer Alkohole mit anschließender Säure-katalysierter Peroxyacetalisierung unter Verwendung von 4-Nitrobenzaldehyd. Über diesen Syntheseweg konnten unterschiedlich substituierte 1,2,4-Trioxane dargestellt werden. Die höchste biologische Aktivität zeigten Verbindungen mit aromatischen Estersubstituenten am 1,2,4-Trioxanring. Es wurde eine Leitstruktur entwickelt, die einen α,β-ungesättigten aromatischen Estersubstituenten in Position 6 des 1,2,4-Trioxangerüsts und in Position 3 einen 4-Nitrophenylsubstituenten aufweist. Die Verbindungen dieser Substanzklasse zeigen Inhibition der GSTP1 im niedrig mikromolaren Bereich. Durch Aktivitätsstudien an den GST-Klassen A und M konnte gezeigt werden, dass die Verbindungen selektiv GSTP1 inhibieren. Nachdem mittels quantitativer PCR 12 Krebszelllinien, die hohe GSTP1-Expressionsniveaus zeigen, identifiziert worden waren, wurde die Aktivität der 1,2,4-Trioxane gegenüber GST, die in Krebszelllysaten vorkommt, nachgewiesen. Die GST in der Brustkrebsepithelzelllinie HBL100 und der Lungenkarzinomzelllinie SK-MES-1 wird durch 1,2,4-Trioxane noch effektiver inhibiert als aufgereinigte GSTP1 (IC50 im nanomolaren Bereich).

The synthesis and optimization of two Li-ion solid electrolytes were studied in this work. Different combinations of precursors were used to prepare La0.5Li0.5TiO3 via mechanosynthesis. Despite the ability to form a perovskite phase by the mechanochemical reaction it was not possible to obtain a pure La0.5Li0.5TiO3 phase by this process. Of all the seven combinations of precursors and conditions tested, the one where La2O3, Li2CO3 and TiO2 were milled for 480min (LaOLiCO-480) showed the best results, with trace impurity phases still being observed. The main impurity phase was that of La2O3 after mechanosynthesis (22.84%) and Li2TiO3 after calcination (4.20%). Two different sol-gel methods were used to substitute boron on the Zr-site of Li1+xZr2-xBx(PO4)3 or the P-site of Li1+6xZr2(P1-xBxO4)3, with the doping being achieved on the Zr-site using a method adapted from Alamo et al (1989). The results show that the Zr-site is the preferential mechanism for B doping of LiZr2(PO4)3 and not the P-site. Rietveld refinement of the unit-cell parameters was performed and it was verified by consideration of Vegard’s law that it is possible to obtain phase purity up to x = 0.05. This corresponds with the phases present in the XRD data, that showed the additional presence of the low temperature (monoclinic) phase for the powder sintered at 1200ºC for 12h of compositions with x ≥ 0.075. The compositions inside the solid solution undergo the phase transition from triclinic (PDF#01-074-2562) to rhombohedral (PDF#01-070-6734) when heating from 25 to 100ºC, as reported in the literature for the base composition. Despite several efforts, it was not possible to obtain dense pellets and with physical integrity after sintering, requiring further work in order to obtain dense pellets for the electrochemical characterisation of Li Zr2(PO4)3 and Li1.05Zr1.95B0.05(PO4)3.

En la presente investigaci?n se estudi? la din?mica de la generaci?n y captura de las emisiones de gases de efecto invernadero (GEI) en un humedal natural eutrofizado bajo condiciones tropicales denominado El Madrigal, ubicado en la vereda El Madrigal, del municipio de Riofr?o, Valle del Cauca. Con tal prop?sito se evalu? la variaci?n espacial de las emisiones de los GEI tales como metano (CH4), di?xido de carbono (CO2) y ?xido nitroso (N2O) y se estableci? su posible correlaci?n con par?metros de calidad de agua como pH, ox?geno disuelto, potencial de ?xido reducci?n, alcalinidad, fosfatos, nitratos entre otros y a su vez con el contenido de materia org?nica y nutrientes presente en los sedimentos. En este sentido, se determinaron y compararon las emisiones en cuatro puntos de muestreo establecidos en el humedal con caracter?sticas diferentes: P1 (Zona de intercambio de agua y sedimentos con R?o Cauca), P2 (Zona influenciada por actividades pisc?colas), P3 (Zona influenciada por vertimientos de aguas residuales y presencia de vegetaci?n) y P3A (Zona sin cobertura vegetal). En cada punto se realizaron mediciones de gases de efecto invernadero, par?metros in situ, calidad de agua y, sedimentos. Los valores de flux obtenidos evidenciaron que el humedal El Madrigal actu? como fuente y sumidero de gases de efecto invernadero durante todo el per?odo de muestreo. El punto P1 present? flux promedios de -1,96 ? 3,10 g.CH4.m-2 d-1; -0,12 ? 7,63 g.CO2.m-2 d-1 y 1,73 ? 5,75 mg.N2O.m-2 d-1. Para el caso del punto P2 se obtuvieron flux promedios de 0,898 ? 1,27 g.CH4.m-2 d-1; 1,03 ? 9,31 g.CO2.m-2 d-1 y -1,19 ? 5,08 mg.N2O.m-2 d-1. Por su parte el punto P3 evidenci? flux promedios de 3,08 ? 3,99 g.CH4.m-2 d-1; -2,74 ? 9,83 g.CO2.m-2 d-1 y 2,06 ? 5,20 mg.N2O.m-2 d-1. Por ?ltimo los flux promedios obtenidos para el punto P3A fueron 1,80 ? 6,89 g.CH4.m-2 d-1; 1,40 ? 8,40 g.CO2.m-2 d-1 y -1,24 ? 6,43 mg.N2O.m-2 d-1. Las pruebas estad?sticas indicaron que los flux de los gases de efecto invernadero estudiados presentaron una variaci?n espacial significativa entre los puntos muestreados. CH4 present? variaci?n entre los puntos P1, P2 y P3 (p<0.001 Mann-Whitney) y evidenci? similitudes entre los puntos P3A-P3 y P3A-P2 (p>0.05 Mann-Whitney). Para CO2 se evidenci? similitud para los flux obtenidos en los cuatro puntos (p>0.05 Mann-Whitney). Por su parte, para el ?xido nitroso tambi?n se present? un comportamiento similar en los cuatro puntos sin variaci?n significativa (p>0.05 Mann-Whitney). Con respecto a las relaciones entre los par?metros de calidad de agua, sedimentos y los flux de gases de efecto invernadero, se determin? que los par?metros con mayor influencia sobre la producci?n y consumo de CH4 CO2 y N2O fueron hierro f?rrico (Fe+3) Coeficiente Spearman = -73%), nitratos (NO3) (Coeficiente Spearman = -65%) y los nitritos (NO2) en sedimentos Coeficiente Spearman = 86%), respectivamente. Los resultados obtenidos en este estudio evidenciaron que la intervenci?n antr?pica contribuye al estado eutr?fico del ecosistema y este a su vez influencia la din?mica de producci?n y consumo de GEI favoreciendo el incremento en la generaci?n de estos, demandando medidas de manejo y control que permitan mitigar el impacto frente al cambio clim?tico

La détermination de la structure d’une molécule dans une cellule est souvent la première étape à la compréhension de son mode d’action. Un exemple flagrant est le riborégulateur, qui est un ARN hautement structuré capable de lier un ligand et de modifier la régulation génique chez les bactéries. Cette étude se penche sur la structure de l’aptamère d’un riborégulateur pouvant se lier au métabolite S-adénosylméthionine (SAM), qui est impliqué dans des réactions de méthylations essentielles dans une cellule. Plus précisément, le modèle sélectionné pour ces travaux provient de la famille SAM-I et est composé de quatre tiges (P1 à P4). En utilisant des techniques biophysiques telles le FRET et le single-molecule FRET (sm-FRET), nous pouvons comprendre plus en détails le dynamisme structural du repliement de l’aptamère qui se passe en deux étapes. La première étape de repliement de l’aptamère est constituée par la formation de l’état intermédiaire induite par la liaison du magnésium. Cet état a été caractérisé grâce à des mutations nucléotidiques bloquant des changements conformationnels spécifiques. Aussi, cette étude a permis d’ajouter des détails biophysiques sur la dernière étape de repliement de l’aptamère induite par la liaison de SAM. La réorganisation finale du site de liaison implique une rotation de la tige P1. D’ailleurs, la formation de la tige P1 permet le contrôle direct de l’expression génétique. Ces deux étapes permettent à l’aptamère du riborégulateur SAM d’adopter sa structure native essentielle à l’arrêt de transcription. En bref, nous décrivons en détail les mécanismes structuraux expliquant le fonctionnement du riborégulateur SAM. Certains de ces mécanismes pourraient être communs à plusieurs classes de riborégulateurs, d’où l’intérêt de prospecter d’autres cibles.

Air pollution can threat the environment and public health, and is assess by pollutant ́s concentration measurements in order to verify whether the limits set by environmental agencies are being respected. However, these measures do not indicate immediately the impacts to living beings. To faced this problem, plants are been investigated as potential bioindicators of air pollution and, among them, stand out bromeliads Tillandsia genus which colonize various substrates,. obtaining water and nutrients from the atmosphere directly. In this context, this research assessed the potential of epiphytic bromeliad Tillandsia recurvata (L.) L. found in urbanized areas of the city of Curitiba - PR as a bioindicator of urban air pollution. According to vehicle traffic, five sample points were selected and classified. Points P1 and P2 were classified as high-traffic vehicle due presenting trucks and urban transport; point P3 was classified as moderate traffic due the predominance of private vehicles and urban transport; and points P4 and P5 were classified as low-traffic, presenting circulation of private vehicles only. There were analyzed the abundance of T. recurvata, morphophysiological parameters (leaf area, leaf specific area, sclerophylly index, percentage dry weight / fresh weight, chlorophyll (a + b), analysis of structural mesophyll organization) and the heavy metals accumulation (Fe, Cd, Cr, Cu, Pb and Zn). The abundance analysis and the results obtained for metals analysis were correlated with the intensity of vehicular traffic, directing the sampling points P1 > P2 = P3 > P4 = P5. This result demonstrate that the abundance of T. recurvata is greater in urban air pollution impacted areas, thus indicating that T. recurvata absorbs and accumulates metals and can be used in biomonitoring of urban air pollution in areas impacted by vehicular traffic. Morphophysiological parameters analyzed shows that the internal plant ́s structure is not significantly impacted by urban air pollution due plant ́s adptations. The presence of absorbing scales, the CAM metabolism pathway and it ́s store water ability, among other features, demonstrate their potential as bio-indicator in urban areas, especially regarding heavy metals accumulation .

Recent research on affective processing has suggested that low spatial frequency information of fearful faces provide rapid emotional cues to the amygdala, whereas high spatial frequencies convey fine-grained information to the fusiform gyrus, regardless of emotional expression. In the present experiment, we examined the effects of low (LSF, <15 cycles/image width) and high spatial frequency filtering (HSF, >25 cycles/image width) on brain processing of complex pictures depicting pleasant, unpleasant, and neutral scenes. Event-related potentials (ERP), percentage of recognized stimuli and response times were recorded in 19 healthy volunteers. Behavioral results indicated faster reaction times in response to unpleasant LSF than to unpleasant HSF pictures. Unpleasant LSF pictures and pleasant unfiltered pictures also elicited significant enhancements of P1 amplitudes at occipital electrodes as compared to neutral LSF and unfiltered pictures, respectively; whereas no significant effects of affective modulation were found for HSF pictures. Moreover, mean ERP amplitudes in the time between 200 and 500ms post-stimulus were significantly greater for affective (pleasant and unpleasant) than for neutral unfiltered pictures; whereas no significant affective modulation was found for HSF or LSF pictures at those latencies. The fact that affective LSF pictures elicited an enhancement of brain responses at early, but not at later latencies, suggests the existence of a rapid and preattentive neural mechanism for the processing of motivationally relevant stimuli, which could be driven by LSF cues. Our findings confirm thus previous results showing differences on brain processing of affective LSF and HSF faces, and extend these results to more complex and social affective pictures.
