The objective of this study was to determine the influence of different ion-exchange temperatures on the biaxial flexural strength (sigma(f)), hardness (HV) and indentation fracture resistance (K(IF)) of a dental porcelain. Disk-shaped specimens were divided into five groups (n = 10) and submitted to an ion-exchange procedure using KNO(3) paste for 15 min in the following temperatures (degrees C); (I) 430; (II) 450; (III) 470; (IV) 490; (V) 510; and control (no ion exchange). The value of sigma(f) was determined in artificial saliva at 37 degrees C. The values of HV and K(IF) were obtained using 3 Vickers indentations in each specimen (19.6 N). Results showed that ion exchange increases significantly the properties of the material as compared to the control and no significant differences were found among the temperatures tested for any of the properties studied. The maintenance of the avulsed teeth in appropriate media for preserving the cellular viability has been important for repairing the periodontal ligament and preventing the root resorption after tooth reimplantation. Propolis is a substance capable of preserving cellular viability. This study aimed to analyze the propolis substance as a storage media for maintaining the avulsed teeth, besides to determine the ideal time period for keeping the tooth inside it. Thus, 60 maxillary right central incisors of rats were extracted and divided into five groups. In groups I and II, teeth were kept in propolis for 60 min and 6 h, respectively; in group III, teeth were kept in milk for 6 h; in group IV, teeth were kept dry for 60 min; and in group V, they were immediately reimplanted. All teeth had their root canals filled with calcium hydroxide paste. Following, teeth were reimplanted in their sockets. After 15 and 60 days, animals were killed and the obtained samples were processed in laboratory for microscopic and morphometric analyzing. The results showed that the occurrence of inflammatory resorption, dental ankylosis and the formation of the connective tissue parallel to the root surface were similar among groups. It could be verified a greater occurrence of replacement resorption in group IV when comparing to other groups. In groups I and IV, the presence of periodontal ligament-like connective tissue was substantially smaller than the other groups. Regarding to the cementum amount over the root, it could be observed that this was present in smaller amount in groups I and IV. Group II was similar to groups III and IV. Therefore, according to the results of this study, the use of propolis as a storage media for maintaining avulsed teeth could be highlighted, and the 6-h period was more appropriate than the 60-min period.

Objective. The aim of this study was to evaluate the biocompatibility of the root canal sealer Epiphany in rat subcutaneous tissues. Study design. Polyethylene tubes were filled with the sealer (I: Epiphany; II: photoactivated Epiphany; III: Epiphany associated with self-etch primer; IV: photoactivated Epiphany associated with primer; and V: control group) and later implanted into 4 different regions of the dorsum of 15 adult male rats (Rattus novergicus, Albinus Wistar). After 7, 21, and 42 days, 5 animals were killed, obtaining 4 samples per group, in addition to the control group, at each analyzed time. Results. In all periods, Epiphany induced a mild inflammatory reaction. However, in group II, in which the primer was not used, extensive necrosis and a moderate to intense inflammatory reaction were observed, mainly after 7 and 21 days. Conclusion. Epiphany sealer appears biocompatible when tested on rat subcutaneous tissues.

In pulmonary hypertension, changes in pulmonary vascular structure and function contribute to the elevation in pulmonary artery pressure. The time-courses for changes in function, unlike structure, are not well characterised. Medial hypertrophy and neomuscularisation and reactivity to vasoactive agents were examined in parallel in main and intralobar pulmonary arteries and salt-perfused lungs from rats exposed to hypoxia (10% O-2) for 1 and 4 weeks (early and established pulmonary hypertension, respectively). After 1 week of hypoxia, in isolated main and intralobar arteries, contractions to 5-hydroxytryptamine and U46619 (thromboxane-mimetic) were increased whereas contractions to angiotensins I and II and relaxations to acetylcholine were reduced. These alterations varied quantitatively between main and intralobar arteries and, in many instances, regressed between 1 and 4 weeks. The alterations in reactivity did not necessarily link chronologically with alterations in structure. In perfused lungs, constrictor responses to acute alveolar hypoxia were unchanged after 1 week but were increased after 4 weeks, in conjunction with the neomuscularisation of distal alveolar arteries. The data suggest that in hypoxic pulmonary hypertension, the contribution of altered pulmonary vascular reactivity to the increase in pulmonary artery pressure may be particularly important in the early stages of the disease.

The overlapping expression profile of MEF2 and the class-II histone deacetylase, HDAC7, led us to investigate the functional interaction and relationship between these regulatory proteins. HDAC7 expression inhibits the activity of MEF2 (-A, -C, and -D), and in contrast MyoD and Myogenin activities are not affected. Glutathione S-transferase pulldown and immunoprecipitation demonstrate that the repression mechanism involves direct interactions between MEF2 proteins and HDAC7 and is associated with the ability of MEF2 to interact with the N-terminal 121 amino acids of HDAC7 that encode repression domain 1. The MADS domain of MEF2 mediates the direct interaction of MEF2 with HDAC7, MEF2 inhibition by HDAC7 is dependent on the N-terminal repression domain and surprisingly does not involve the C-terminal deacetylase domain. HDAC7 interacts with CtBP and other class-I and -II HDACs suggesting that silencing of MEF2 activity involves corepressor recruitment. Furthermore, we show that induction of muscle differentiation by serum withdrawal leads to the translocation of HDAC7 from the nucleus into the cytoplasm. This work demonstrates that HDAC7 regulates the function of MEF2 proteins and suggests that this class-II HDAC regulates this important transcriptional (and pathophysiological) target in heart and muscle tissue. The nucleocytoplasmic trafficking of HDAC7 and other class-II HDACs during myogenesis provides an ideal mechanism for the regulation of HDAC targets during mammalian development and differentiation.

Aim. This study examines the extent to which hospital nurses view their working environment in a positive sense, working as a cohesive group. Background. Despite the fact that nursing in Australia is now considered a profession, it has been claimed that nurses are an oppressed group who use horizontal violence, bullying and aggression in their interactions with one and other. Methods. After ethical approval, a random sample of 666 nurses working directly with patients and all 333 critical care nurses employed in three large tertiary Australian hospitals were invited to participate in the study in the late 1990s. A mailed survey examined the perceptions of interaction nurses had with each other. The hypothesis, that level of employment (either Level I bedside nurses or Level II/III clinical leaders) and area of work (either critical care or noncritical care) would influence perceptions of cohesion, as measured by the cohesion amongst nurses scale (CANS) was tested. Results. In total 555 (56%) surveys were returned. Of these, 413 were returned by Level I and 142 by Level II/III nurses. Of this sample, 189 were critical care and 355 noncritical care nurses. There was no difference between Level I and II/III nurses in mean CANS scores. It is interesting to note that the item rated most positively was, 'nurses on the units worked well together', however, the item rated least positive was 'staff can be really bitchy towards each other' for both Level I and II/III nurses. There was no difference in CANS scores between critical care and noncritical care nurses. Conclusions. Nurses working in Australian hospitals perceived themselves to be moderately cohesive but, as would be expected in other work settings, some negative perceptions existed.

Chemosensory proteins (CSPs) are ubiquitous soluble small proteins isolated from sensory organs of a wide range of insect species, which are believed to be involved in chemical communication. We report the cloning of a honeybee CSP gene called ASP3c, as well as the structural and functional characterization of the encoded protein. The protein was heterologously secreted by the yeast Pichia pastoris using the native signal peptide. ASP3c disulfide bonds were assigned after trypsinolysis followed by chromatography and mass spectrometry combined with microsequencing. The pairing (Cys(I)-Cys(II), Cys(III)-Cys(IV)) was found to be identical to that of Schistocerca gregaria CSPs, suggesting that this pattern occurs commonly throughout the insect CSPs. CD measurements revealed that ASP3c mainly consists of alpha-helices, like other insect CSPs. Gel filtration analysis showed that ASP3c is monomeric at neutral pH. Using ASA, a fluorescent fatty acid anthroyloxy analogue as a probe, ASP3c was shown to bind specifically to large fatty acids and ester derivatives, which are brood pheromone components, in the micromolar range. It was unable to bind tested general odorants and other tested pheromones (sexual and nonsexual). This is the first report on a natural pheromonal ligand bound by a recombinant CSP with a measured affinity constant.

It was previously published by the authors that granules can either coalesce through Type I (when granules coalesce by viscous dissipation in the surface liquid layer before their surfaces touch) or Type II (when granules are slowed to a halt during rebound, after their surfaces have made contact) (AIChE J. 46 (3) (2000) 529). Based on this coalescence mechanism, a new coalescence kernel for population balance modelling of granule growth is presented. The kernel is constant such that only collisions satisfying the conditions for one of the two coalescence types are successful. One constant rate is assigned to each type of coalescence and zero is for the case of rebound. As the conditions for Types I and II coalescence are dependent on granule and binder properties, the coalescence kernel is thus physically based. Simulation results of a variety of binder and granule materials show good agreement with experimental data. (C) 2002 Elsevier Science Ltd. All rights reserved.

Objective: To assess the (i) benefits, (ii) harms and (iii) costs of continuing mammographic screening for women 70 years and over. Data sources and synthesis: (i) We conducted a MEDLINE search (1966 - July 2000) for decision-analytic models estimating life-expectancy gains from screening in older women. The five studies meeting the inclusion criteria were critically appraised using standard criteria. We estimated relative benefit from each model's estimate of effectiveness of screening in older women relative to that in women aged 50-69 years using the same model. (ii) With data from BreastScreen Queensland, we constructed balance sheets of the consequences of screening for women in 10-year age groups (40-49 to 80-89 years), and (iii) we used a validated model to estimate the marginal cost-effectiveness of extending screening to women 70 years and over. Results: For women aged 70-79 years, the relative benefit was estimated as 40%-72%, and 18%-62% with adjustment for the impact of screening on quality of life. For women over 80 years the relative benefit was about a third, and with quality-of-life adjustment only 14%, that in women aged 50-69 years. (ii) Of 10 000 Australian women participating in ongoing screening, about 400 are recalled for further testing, and, depending on age, about 70-112 undergo biopsy and about 19-80 cancers are detected. (iii) Cost-effectiveness estimates for extending the upper age limit for mammographic screening from 69 to 79 years range from $8119 to $27 751 per quality-adjusted life-year saved, which compares favourably with extending screening to women aged 40-49 years (estimated at between $24 000 and $65 000 per life-year saved). Conclusions: Women 70 years and over, in consultation with their healthcare providers, may want to decide for themselves whether to continue mammographic screening. Decision-support materials are needed for women in this age group.

We have examined melanocortin-1 receptor (MC1R) variant allele frequencies in the general population and in a collection of adolescent dizygotic and monozygotic twins to determine statistical associations of pigmentation phenotypes with increased skin cancer risk. This included hair and skin color, freckling, mole count and sun exposed skin reflectance. Nine variants were studied and designated as either strong R (OR = 63; 95% CI 32-140) or weak r (OR = 5; 95% CI 3-11) red hair alleles. Penetrance of each MC1R variant allele was consistent with an allelic model where effects were multiplicative for red hair but additive for skin reflectance. To assess the interaction of the brown eye color gene BEY2/OCA2 on the phenotypic effects of variant MC1R alleles we imputed OCA2 genotype in the twin collection. A modifying effect of OCA2 on MC1R variant alleles was seen on constitutive skin color, freckling and mole count. In order to study the individual effects of these variants on pigmentation phenotype we have established a series of human primary melanocyte strains genotyped for the MC1R receptor. These include strains which are MC1R wild-type consensus, variant heterozygotes, and homozygotes for strong R alleles Arg151Cys and Arg160Trp. Ultrastructural analysis demonstrated that only consensus strains contained stage III and IV melanosomes in their terminal dendrites whereas Arg151Cys and Arg160Trp homozygous strains contained only immature stage I and II melanosomes. Such genetic association studies combined with the functional analysis of MC1R variant alleles in melanocytic cells should provide a link in understanding the association between pigmentary phototypes and skin cancer risk.

A pesquisa analisa da constituição histórica da disciplina História da Educação ministrada na Faculdade de Filosofia Ciências e Letras do Estado do Espírito Santo, posteriormente incorporada a Universidade Federal do Espírito Santo entre os anos de 1951 e 2000. Investiga a constituição histórica da disciplina, as transformações programáticas, legais e institucionais referentes à disciplina de História da Educação, como também as abordagens historiográficas, periodizações e os conceitos de tempo, história e educação. A fundamentação teórica e metodológica articula-se dialogicamente a partir das construções conceituais e metodológicas de Carlo Ginzburg e Mikhail Bakhtin. A partir dos conceitos de polifonia e dialogismo, comum a ambos, investigou-se as vozes e diálogos impressos nas narrativas da disciplina de História da Educação e seu ensino, sejam em camadas mais superficiais ou profundas, encontradas no corpus documental consultado e analisado, que correspondem a: programas de ensino, transparências, leis, estruturas curriculares, documentos de departamento; resenhas e fichamentos de textos, bibliografia obrigatória e complementar, avaliações e entrevistas. Procurou-se no corpus documental dados aparentemente negligenciáveis – pistas, indícios e sinais – remontar uma realidade histórica complexa e não experimentável diretamente. Ao investigar historicamente a trajetória da disciplina História da Educação e seu ensino a partir dos parâmetros legais, programáticos e institucionais, foi possível perceber que as mudanças mais profundas operadas na disciplina não se originam das legislações e reestruturações curriculares, mas dos locais de produção e socialização do conhecimento histórico. Durante o período analisado, as duas esferas de produção historiográficas que mais influenciaram nas abordagens, periodizações e conceitos de tempo, história e educação da disciplina História da Educação do curso de pedagogia pesquisado foram: a editora responsável pela publicação e divulgação dos Manuais de História da Educação da coleção Atualidades Pedagógicas (1951-1979) e os Programas de Pós-graduação em Educação e História (1980 - 2000). Entre 1951 e finais de 1970 observa-se a influência dos Manuais de História da Educação, na organização e programação do ensino de História da Educação e uma abordagem filosófica voltada para a história das ideias pedagógicas e análises do pensamento de filósofos e educadores sobre a educação e respectivas inserções em doutrinas filosóficas europeias. A partir de 1980 as abordagens de cunho econômico, político e ideológico dos contextos históricos educativos passaram a predominar nos programas de ensino das disciplinas de História da Educação I e II, e vigoraram até meados nos anos de 1990. Na disciplina de História da Educação I a abordagem é marcada por análises do contexto de produção e organização das classes sociais; com relação à disciplina História da Educação II, até meados de 1995, trata da educação brasileira. A partir da abordagem fundamentada na Teoria da Dependência após 1995, os documentos consultados começam a mostrar outras marcas que sugerem uma abordagem voltada para a dimensão política e social, abordando a História da Educação Brasileira, a partir dos movimentos sociais e seus respectivos projetos educacionais.

O zinco é um oligoelemento envolvido em múltiplas reações orgânicas, sendo um componente essencial na síntese de proteínas e um provável fator na estabilização de membranas celulares. Na cóclea, é tido como o principal componente na defesa contra a ação de radicais livres, agindo também como modulador sináptico em alguns sistemas glutamatérgicos. Sua participação em sinapses no sistema auditivo e no tratamento do zumbido tem sido muito estudada nos últimos anos. OBJETIVO: O presente estudo procurou avaliar os níveis séricos de zinco em portadores de zumbido, antes e após o tratamento com compostos com zinco, comparando-os a um grupo normal. FORMA DE ESTUDO: Clínico prospectivo. CASUÍSTICA E MÉTODO: Foram avaliados 41 pacientes (22 com zumbido e 19 normais), que constituíram dois grupos (I e II), respectivamente. Ambos foram submetidos à avaliação audiológica e exames metabólicos e bioquímicos, que procuraram atender a critérios de inclusão e exclusão, além de dosagem sérica de zinco. Os pacientes foram tratados com compostos com zinco por 90 dias, após o que novamente foram submetidos à dosagem sérica do oligoelemento. RESULTADOS: Não foi encontrada uma relação estatisticamente significante entre níveis séricos de zinco em pacientes com zumbido e indivíduos normais, mas o tratamento aumentou os níveis do oligoelemento no grupo sintomático, tornando esse mais próximo ao grupo normal. Houve melhora do zumbido em 45,5% dos pacientes tratados. CONCLUSÕES: Pacientes com zumbido não apresentam níveis séricos de zinco distintos aos da população normal neste estudo.

Avaliar o significado prognóstico dos níveis linfáticos acometidos por metástases nos carcinomas epidermóides da região tonsilar. OBJETIVO: Definir o tipo de esvaziamento cervical eletivo mais apropriado. FORMA DE ESTUDO: Série de casos. MATERIAL E MÉTODO: 51 pacientes com tumor de região tonsilar tratados entre 1992 e 2001. A incidência de metástases foi avaliada nos diferentes níveis linfonodais, bem como sua relação com a extensão do tumor primário para os subsítios anatômicos adjacentes. RESULTADOS: Nos pacientes cN0 foram encontradas metástases apenas nos níveis I e II. Entre os pacientes pN+ com metástases no nível I, 6/7 apresentavam extensão da lesão para a cavidade oral. CONCLUSÃO: O esvaziamento supraomohioideo (níveis I, II e III) mostrou ser o mais adequado para o tratamento eletivo do pescoço nos tumores da região tonsilar que se estendem para a cavidade oral. Nos casos N0 restritos à orofaringe, o esvaziamento apenas dos níveis II e III mostrou-se a melhor indicação terapêutica.
