This thesis evaluates the rheological behaviour of asphalt mixtures and the corresponding extracted binders from the mixtures containing different amounts of Reclaimed Asphalt (RA). Generally, the use of RA is limited to certain amounts. The study materials are Stone Mastic Asphalts including a control sample with 0% RA, and other samples with RA rates of 30%, 60% and 100%. Another set of studied mixtures are Asphalt Concretes (AC) types with again a control mix having 0% RA rate and the other mixtures designs containing 30%, 60% and 90% of reclaimed asphalt which also contain additives. In addition to the bitumen samples extracted from asphalt mixes, there are bitumen samples directly extracted from the original RA. To characterize the viscoelastic behaviour of the binders, Dynamic Shear Rheometer (DSR) tests were conducted on bitumen specimens. The resulting influence of the RA content in the bituminous binders are illustrated through master curves, black diagrams and Cole-Cole plots with regressing these experimental data by the application of the analogical 2S2P1D and the analytical CA model. The advantage of the CA model is in its limited number of parameters and thus is a simple model to use. The 2S2P1D model is an analogical rheological model for the prediction of the linear viscoelastic properties of both asphalt binders and mixtures. In order to study the influence of RA on mixtures, the Indirect Tensile Test (ITT) has been conducted. The master curves of different mixture samples are evaluated by regressing the test data points to a sigmoidal function and subsequently by comparing the master curves, the influence of RA materials is studied. The thesis also focusses on the applicability and also differences of CA model and 2S2P1D model for bitumen samples and the sigmoid function for the mixtures and presents the influence of the RA rate on the investigated model parameters.

Cette thèse s’inscrit dans le contexte d’une optimisation industrielle et économique des éléments de structure en BFUP permettant d’en garantir la ductilité au niveau structural, tout en ajustant la quantité de fibres et en optimisant le mode de fabrication. Le modèle développé décrit explicitement la participation du renfort fibré en traction au niveau local, en enchaînant une phase de comportement écrouissante suivie d’une phase adoucissante. La loi de comportement est fonction de la densité, de l’orientation des fibres vis-à-vis des directions principales de traction, de leur élancement et d’autres paramètres matériaux usuels liés aux fibres, à la matrice cimentaire et à leur interaction. L’orientation des fibres est prise en compte à partir d’une loi de probabilité normale à une ou deux variables permettant de reproduire n’importe quelle orientation obtenue à partir d’un calcul représentatif de la mise en oeuvre du BFUP frais ou renseignée par analyse expérimentale sur prototype. Enfin, le modèle reproduit la fissuration des BFUP sur le principe des modèles de fissures diffuses et tournantes. La loi de comportement est intégrée au sein d’un logiciel de calcul de structure par éléments finis, permettant de l’utiliser comme un outil prédictif de la fiabilité et de la ductilité globale d’éléments en BFUP. Deux campagnes expérimentales ont été effectuées, une à l’Université Laval de Québec et l’autre à l’Ifsttar, Marne-la-Vallée. La première permet de valider la capacité du modèle reproduire le comportement global sous des sollicitations typiques de traction et de flexion dans des éléments structurels simples pour lesquels l’orientation préférentielle des fibres a été renseignée par tomographie. La seconde campagne expérimentale démontre les capacités du modèle dans une démarche d’optimisation, pour la fabrication de plaques nervurées relativement complexes et présentant un intérêt industriel potentiel pour lesquels différentes modalités de fabrication et des BFUP plus ou moins fibrés ont été envisagés. Le contrôle de la répartition et de l’orientation des fibres a été réalisé à partir d’essais mécaniques sur prélèvements. Les prévisions du modèle ont été confrontées au comportement structurel global et à la ductilité mis en évidence expérimentalement. Le modèle a ainsi pu être qualifié vis-à-vis des méthodes analytiques usuelles de l’ingénierie, en prenant en compte la variabilité statistique. Des pistes d’amélioration et de complément de développement ont été identifiées.

Esta investigação visa tomar como ponto de partida a diversidade de línguas faladas em Espanha e que coabitam em muitas das comunidades autónomas (Siguán, 2001) contribuindo, desta forma, para a riqueza em termos linguísticos e culturais da Península Ibérica em geral e de Espanha em particular. Com este projeto é nossa intenção contribuir para a implementação de práticas educativas que promovam, na sala de aula de espanhol como língua estrangeira no ensino secundário português, a (re)construção das representações dos alunos face às línguas românicas faladas em Espanha como línguas oficiais, através do desenvolvimento da competência plurilingue (Alarcão, Andrade, Araújo e Sá, Melo-Pfeifer, & Santos, 2010). Neste estudo pretendemos, através de uma metodologia de índole qualitativa, analisar as representações dos alunos de espanhol iniciação do ensino secundário em Portugal face às línguas oficiais faladas em Espanha e à sua competência plurilingue, recorrendo a inquéritos por questionário. Visa-se, para além disso, evidenciar uma mostra de possíveis práticas didático-pedagógicas e materiais que possibilitem um trabalho sistemático e pró-ativo no âmbito da (re)construção das representações sobre as línguas oficiais faladas em Espanha, que promovam o desenvolvimento da competência plurilingue dos alunos e que sejam potenciadoras de cidadãos conscientes do mundo que os rodeia (Gadotti, 2003). Para tal, serão criados e implementados materiais físicos e digitais com alunos do 10.º e 11.º ano de escolaridade, a frequentar a disciplina de espanhol iniciação, num agrupamento de escolas da região de Aveiro. Os resultados mostram que práticas e materiais desta natureza podem favorecer a (re)construção das representações dos alunos, fomentando o desenvolvimento da sua competência plurilingue.

Bioenergy is one of many contributors to reducing the use of fossil fuels in order to mitigate climate change by decreasing CO2-emissions, and the potential for biofuels are large. The wood fuel pellets are a refined biofuel made of sawdust, which is dried and compressed to achieve improved fuel and transportation properties. In 2007 the amount of wood fuel pellets used for heating purposes in Sweden was 1715000 tons. The aims of this work was: to examine the moisture content and emission of monoterpenes during the drying and pelletising steps of the pellets production (Paper I); to investigate how the recirculation of drying gases affects the energy efficiency of rotary dryers and how the energy efficiency is related to the capacity of the dryer. (Paper II); to analyse the causes of the problems encountered by household end-users of pellets and investigate whether an improved pellet quality standard could reduce these problems (Paper III); to investigate how the energy consumption of the pelletising machine and chosen pellet quality parameters were affected using an increased amount of rapeseed cake in wood fuel pellets (Paper IV); and to identify gaps of knowledge about wood fuel pellet technology and needs for further research on quality, environmental and health aspects throughout the wood fuel pellet chain, from sawdust to heat. A Cultura Digital é uma realidade do século XXI, onde as relações humanas são fortemente mediadas pelas tecnologias e pelo digital. O Digital tem mudado o comportamento das pessoas e influenciado o seu meio cultural. O conceito de Cultura digital inclui saber onde encontrar informação, ferramentas e sistemas, necessários para cumprir uma determinada tarefa de forma não só eficaz, mas também mais eficiente. As competências para explorar as tecnologias que o permitam, são cada vez mais exigidas na interação em sociedade. Tendo em consideração estudos existentes associados com as competências TIC, nomeadamente os propostos pela UNESCO e pelo Executivo Angolano no seu Plano Nacional de Desenvolvimento para 2013-2017, em que está enquadrada a iniciativa da Rede de Mediatecas de Angola, foi tomada a Cultural Digital e a sua promoção no ambito das Mediatecas. Numa primeira avaliação desde da abertura da Mediateca em Fevereiro de 2014, verificamos uma fraca adesão dos professores na utilização dos serviços da Mediateca do Huambo – Angola. Coloca-se neste contexto a questão do porquê que é que os professores não exploram as TIC para as atividades docentes ou da sua aprendizagem? Desta forma, é considerada a questão de investigação: Quais as estratégias a desenvolver para incrementar a exploração das TIC pelos professores e como a Mediateca do Huambo pode servir de espaço para a promoção da Cultura Digital. Foi realizado um estudo de caso, no contexto da Mediateca do Huambo, em que foi efetuada uma recolha de dados pela (a) aplicação de questionários aos professores do 1º e 2º Ciclo do Ensino Secundário do Município do Huambo para mapear as competências TIC dos professores segundo os padrões da UNESCO e o apuramento da formação e uso das TIC em contexto educativo, (b) pela aplicação de um conjunto de tarefas com o objetivo de verificar quais as competências digitais dos professores e por último (c), a implementação do projeto-piloto denominado “ Mediateca +Escola” com o objetivo de levar à Mediateca professores e alunos para desenvolver um projeto TIC onde os professores teriam uma oportunidade de demostrar as competências que afirmavam possuir. Como resultado final obtido neste estudo de caso podemos concluir que 71,27% indicanos que os professores têm a perceção que necessitam de mais formação e desenvolvimento de competências TIC, 27,50% confirma que os professores creem que têm confiança nas TIC. Em face das respostas com valores negativos que apontam para a necessidade de delinear estratégia para capacitar este grupo em competências TIC de forma a incluí-los na sociedade em rede providos de uma Cultura Digital foi proposto o modelo que permite validar os dados obtidos pelo questionário permitindo também delinear estratégias com vista à promoção da Cultura Digital.

Microsecond long Molecular Dynamics (MD) trajectories of biomolecular processes are now possible due to advances in computer technology. Soon, trajectories long enough to probe dynamics over many milliseconds will become available. Since these timescales match the physiological timescales over which many small proteins fold, all atom MD simulations of protein folding are now becoming popular. To distill features of such large folding trajectories, we must develop methods that can both compress trajectory data to enable visualization, and that can yield themselves to further analysis, such as the finding of collective coordinates and reduction of the dynamics. Conventionally, clustering has been the most popular MD trajectory analysis technique, followed by principal component analysis (PCA). Simple clustering used in MD trajectory analysis suffers from various serious drawbacks, namely, (i) it is not data driven, (ii) it is unstable to noise and change in cutoff parameters, and (iii) since it does not take into account interrelationships amongst data points, the separation of data into clusters can often be artificial. Usually, partitions generated by clustering techniques are validated visually, but such validation is not possible for MD trajectories of protein folding, as the underlying structural transitions are not well understood. Rigorous cluster validation techniques may be adapted, but it is more crucial to reduce the dimensions in which MD trajectories reside, while still preserving their salient features. PCA has often been used for dimension reduction and while it is computationally inexpensive, being a linear method, it does not achieve good data compression. In this thesis, I propose a different method, a nonmetric multidimensional scaling (nMDS) technique, which achieves superior data compression by virtue of being nonlinear, and also provides a clear insight into the structural processes underlying MD trajectories. I illustrate the capabilities of nMDS by analyzing three complete villin headpiece folding and six norleucine mutant (NLE) folding trajectories simulated by Freddolino and Schulten [1]. Using these trajectories, I make comparisons between nMDS, PCA and clustering to demonstrate the superiority of nMDS. The three villin headpiece trajectories showed great structural heterogeneity. Apart from a few trivial features like early formation of secondary structure, no commonalities between trajectories were found. There were no units of residues or atoms found moving in concert across the trajectories. A flipping transition, corresponding to the flipping of helix 1 relative to the plane formed by helices 2 and 3 was observed towards the end of the folding process in all trajectories, when nearly all native contacts had been formed. However, the transition occurred through a different series of steps in all trajectories, indicating that it may not be a common transition in villin folding. The trajectories showed competition between local structure formation/hydrophobic collapse and global structure formation in all trajectories. Our analysis on the NLE trajectories confirms the notion that a tight hydrophobic core inhibits correct 3-D rearrangement. Only one of the six NLE trajectories folded, and it showed no flipping transition. All the other trajectories get trapped in hydrophobically collapsed states. The NLE residues were found to be buried deeply into the core, compared to the corresponding lysines in the villin headpiece, thereby making the core tighter and harder to undo for 3-D rearrangement. Our results suggest that the NLE may not be a fast folder as experiments suggest. The tightness of the hydrophobic core may be a very important factor in the folding of larger proteins. It is likely that chaperones like GroEL act to undo the tight hydrophobic core of proteins, after most secondary structure elements have been formed, so that global rearrangement is easier. I conclude by presenting facts about chaperone-protein complexes and propose further directions for the study of protein folding.

Le présent travail explore le phénomène de la rivalité interétatique1 et les mesures de son intensité. Chacune des définitions existantes de la rivalité fait la lumière sur l’une de ses caractéristiques. La rivalité durable met l’accent sur la compétition militarisée, la rivalité stratégique accorde une importance particulière à la perception de l’ennemi, tandis que la rivalité interétatique est axée sur les questions autour desquelles la compétition se déroule. Ces visions différentes du phénomène de la rivalité laissent leur empreinte sur son opérationnalisation et sur le choix des paramètres visant à le capter. Nous réunissons ces trois interprétations dans une seule définition de rivalité interacteur, en proposant une nouvelle classification des actions hostiles, ainsi que la mesure alternative d’hostilité fondée sur la fréquence de ces dernières. Les quatre études de cas suivantes nous ont permis d’atteindre ces objectifs : les relations de rivalité entre l’Afghanistan et le Pakistan, entre l’Arménie et l’Azerbaïdjan, entre le Bélize et le Guatemala, ainsi qu’entre la Somalie et l’Éthiopie.

This present study analyses the relationship between national culture and entrepreneurship, in order to contribute in understanding the impact and the influence of Brazilian culture under the business practices of foreign entrepreneur on the tourism area of Rio Grande do Norte state. Researchers in entrepreneurship field, such as Schumpeter (2005), Weber (2006), McClelland (1972), Murphy, Liao, Welsch (2006), Peyrefitte (1999), Blanchflower (1988), Filion (2000), among others, and in the national culture, high-lighting Caldas and Woods (1999), Hofstede (1997) and Barros and Prates (1996), are put together to create a starting point to this present research, that reaches out for a differential approach, in order to create a correlation between Brazilian culture and the foreign entrepreneur, who chose Rio Grande do Norte state to live and invest. It was observed that in the case of the Switzerland entrepreneur, where the cultural difference is more strong, either in Hofstede's perspective than in Brazilian s authors, the management practices are more negatively affected. The rationalization adopted by the entrepreneur and the employee s logic are in battle in a day-to-day operation of the company. In the case of the Argentinean entrepreneur, the cultural difference exists, but it does not impact as much, because of her personal characteristics and the similarities of Latin culture than surfaces the differences. Finally, this study showed the necessity of analyzing the enterprising regarding the cultural issue, in a way to highlight the different rationalities in different atmospheres where it is practiced the enterprise

In this paper agricultural waste; Canarium schweinfurthii was explored for the sequestering of Fe and Pb ions from wastewater solution after carbonization and chemical treatment at 400oC. Optimum time of 30 and 150 min with percentage removal of 95 and 98% at optimum pH of 2 and 6 was obtained for Fe and Pb ions. Kinetics model followed pseudofirst order as sum of absolute error (EABS) between Qe and Qc greater than that of pseudo second order. Parameters evaluated from isothermal equation (Freundlich and Langmuir) showed that KL and QO for Fe > Pb and R2 for Langmuir> Freundlich. The study reveals the suitability of the adsorbent for sequestering of Fe and Pb ions from industrial wastewater.

Cells adapt to their changing world by sensing environmental cues and responding appropriately. This is made possible by complex cascades of biochemical signals that originate at the cell membrane. In the last decade it has become apparent that the origin of these signals can also arise from physical cues in the environment. Our motivation is to investigate the role of physical factors in the cellular response of the B lymphocyte. B cells patrol the body for signs of invading pathogens in the form of antigen on the surface of antigen presenting cells. Binding of antigen with surface proteins initiates biochemical signaling essential to the immune response. Once contact is made, the B cell spreads on the surface of the antigen presenting cell in order to gather as much antigen as possible. The physical mechanisms that govern this process are unexplored. In this research, we examine the role of the physical parameters of antigen mobility and cell surface topography on B cell spreading and activation. Both physical parameters are biologically relevant as immunogens for vaccine design, which can provide laterally mobile and immobile antigens and topographical surfaces. Another physical parameter that influences B cell response and the formation of the cell-cell junction is surface topography. This is biologically relevant as antigen presenting cells have highly convoluted membranes, resulting in variable topography. We found that B cell activation required the formation of antigen-receptor clusters and their translocation within the attachment plane. We showed that cells which failed to achieve these mobile clusters due to prohibited ligand mobility were much less activation competent. To investigate the effect of topography, we use nano- and micro-patterned substrates, on which B cells were allowed to spread and become activated. We found that B cell spreading, actin dynamics, B cell receptor distribution and calcium signaling are dependent on the topographical patterning of the substrate. A quantitative understanding of cellular response to physical parameters is essential to uncover the fundamental mechanisms that drive B cell activation. The results of this research are highly applicable to the field of vaccine development and therapies for autoimmune diseases. Our studies of the physical aspects of lymphocyte activation will reveal the role these factors play in immunity, thus enabling their optimization for biological function and potentially enabling the production of more effective vaccines.

O presente Trabalho de Projecto assenta na análise da programação e da evolução espacial e temporal do Festival Internacional de Teatro de Almada, de modo a perceber quais as linhas gerais em que se baseia a sua programação. Procura-se perceber, ao longo do trabalho, de que forma o Festival se ligou à cidade de Almada, contribuindo para a sua evolução ao nível de equipamentos culturais, recuperação urbanística e reestruturação da esfera pública. A par destes assuntos, dá-se conta igualmente da expansão geográfica do Festival de Almada para cidades como Lisboa e Porto, tentando-se perceber o que esteve nas bases da mesma e de que forma o Festival equilibrou esta necessidade de crescimento com a manutenção do vínculo com Almada.
