Forest fires implications in overland flow and soil erosion have been researched for several years. Therefore, is widely known that fires enhance hydrological and geomorphological activity worldwide as also in Mediterranean areas. Soil burn severity has been widely used to describe the impacts of fire on soils, and has being recognized as a decisive factor controlling post-fire erosion rates. However, there is no unique definition of the term and the relationship between soil burn severity and post-fire hydrological and erosion response has not yet been fully established. Few studies have assessed post-fire erosion over multiple years, and the authors are aware of none which assess runoff. Small amount of studies concerning pre-fire management practices were also found. In the case of soil erosion models, the Revised Universal Soil Loss Equation (RUSLE) and the revised Morgan–Morgan–Finney (MMF) are well-known models, but not much information is available as regards their suitability in predicting post-fire soil erosion in forest soils. The lack of information is even more pronounced as regards post-fire rehabilitation treatments. The aim of the thesis was to perform an extensive research under the post fire hydrologic and erosive response subject. By understanding the effect of burn severity in ecosystems and its implications regarding post fire hydrological and erosive responses worldwide. Test the effect of different pre-fire land management practices (unplowed, downslope plowed and contour plowed) and time-since-fire, in the post fire hydrological and erosive response, between the two most common land uses in Portugal (pine and eucalypt). Assess the performance of two widely-known erosion models (RUSLE and Revised MMF), to predict soil erosion rates during first year following two wildfires of distinctive burn severity. Furthermore, to apply these two models considering different post-fire rehabilitation treatments in an area severely affected by fire. Improve model estimations of post-fire runoff and erosion rates in two different land uses (pine and eucalypt) using the revised MMF. To assess these improvements by comparing estimations and measurements of runoff and erosion, in two recently burned sites, as also with their post fire rehabilitation treatments. Model modifications involved: (1) focusing on intra-annual changes in parameters to incorporate seasonal differences in runoff and erosion; and (2) inclusion of soil water repellency in runoff predictions. Additionally, validate these improvements with the application of the model to other pine and eucalypt sites in Central Portugal. The review and meta-analysis showed that fire occurrence had a significant effect on the hydrological and erosive response. However, this effect was only significantly higher with increasing soil burn severity for inter-rill erosion, and not for runoff. This study furthermore highlighted the incoherencies between existing burn severity classifications, and proposed an unambiguous classification. In the case of the erosion plots with natural rainfall, land use factor affected annual runoff while land management affected both annual runoff and erosion amounts significantly. Time-since-fire had an important effect in erosion amounts among unplowed sites, while for eucalypt sites time affected both annual runoff and erosion amounts. At all studied sites runoff coefficients increase over the four years of monitoring. In the other hand, sediment concentration in the runoff, recorded a decrease during the same period. Reasons for divergence from the classic post-fire recovery model were also explored. Short fire recurrence intervals and forest management practices are viewed as the main reasons for the observed severe and continuing soil degradation. The revised MMF model presented reasonable accuracy in the predictions while the RUSLE clearly overestimated the observed erosion rates. After improvements: the revised model was able to predict first-year post-fire plot-scale runoff and erosion rates for both forest types, these predictions were improved both by the seasonal changes in the model parameters; and by considering the effect of soil water repellency on the runoff, individual seasonal predictions were considered accurate, and the inclusion of the soil water repellency in the model also improved the model at this base. The revised MMF model proved capable of providing a simple set of criteria for management decisions about runoff and erosion mitigation measures in burned areas. The erosion predictions at the validation sites attested both to the robustness of the model and of the calibration parameters, suggesting a potential wider application.

O presente trabalho começa por analisar a evolução dos sistemas ao longo de vários períodos históricos, bem como os conceitos e tipologias que os sustentam, no quadro da problemática e da conceptualização teórica do ensino superior. A origem da universidade na idade média, na Europa e em outras partes do mundo, fundamenta-se, principalmente, na procura do saber e nas condicionantes socioeconómicas da época. A evolução da universidade sustentada pela narrativa da modernidade, resultou em modelos diferenciados de ensino superior, que mantinham, no entanto, a razão e a epistemologia do conhecimento académico, como fatores unificadores. O modelo utilizado para traduzir a referida evolução, é proposto por Scott (1995) e configura a relação que se estabelece entre a universidade e outras formas de ensino superior. Na sequência do desenvolvimento dos sistemas, suscitados pelas relações entre os diversos atores envolvidos no ensino superior, procurou-se evidenciar a relação entre o Estado o mercado e os académicos, bem como outros atores sociopolíticos, institucionais e da sociedade civil. O quadro de análise dos mecanismos de coordenação destas envolventes, baseou-se no “Triângulo de Clark”, complementado com o modelo da “Metáfora da Flutuação”. Desde o início da década de 80 do século XX, que a missão, o modo de organização e o funcionamento das IES têm vindo a ser questionadas, como resultado das mudanças económicas de cariz neoliberal. Este cenário, propiciador de crises e de transformações, não tem impedido, porém, de manter o papel fundamental da universidade como produtora e difusora do conhecimento. No contexto da globalização e da cada vez maior influência do mercado no ensino superior, procura-se impor um modelo hegemonizado de racionalidade económica, competitividade e eficiência - o managerialismo. Não obstante alguns êxitos, esta ideologia não tem sido completamente bem-sucedida. Aliás, à tentativa da globalização de gerar uma ordem uniformista, têm sido contrapostos modelos de recontextualização que procuram refletir as realidades locais. Embora num reduzido número de países, África, registou formas de ensino superior endógenas, no período pré-colonial. Depois do espectro colonial que mantém, ainda hoje, a sua influência, os sistemas debatem-se na atualidade com diferentes dilemas, originados pelas correntes da globalização. Porém, o ensino superior em África assume um papel central no contexto do desenvolvimento dos diferentes países contribuindo, igualmente, para a construção da Nação e da sua identidade. Esta dimensão, não é impeditiva do seu envolvimento, nos desafios da sociedade e da economia do conhecimento, buscando, ao mesmo tempo, modelos e práticas equilibradoras, que proporcionem uma resposta satisfatória às necessidades sociais e económicas, nacionais e regionais. Ao enquadrar o sistema de ensino superior em Moçambique merecem destaque as etapas do processo histórico, nomeadamente o surgimento dos estudos gerais universitários como primeira forma de ensino superior mais tarde transformada em universidade, a mudança de paradigma após a independência nacional, a abertura ao setor privado e, ainda, a expansão do sistema. A discussão sobre políticas e estratégias é sustentada pela análise dos respetivos planos estratégicos bem como, pela compreensão das leis fundamentais e respetivos documentos reguladores. Este conjunto de instrumentos concorre para a reforma do sistema, que se procura implementar em Moçambique. Nesta sequência, é de salientar o debate público realizado em torno das qualificações e graus oficialmente estabelecidos, que parece constituir uma problemática ainda não completamente resolvida. Numa outra parte do trabalho, procede-se à apresentação e análise dos resultados de uma investigação sobre o ensino superior em Moçambique. Seguindo uma metodologia de análise qualitativa, foi possível estruturar a informação obtida, em diferentes dimensões e categorias. A informação foi recolhida e tratada, a partir de entrevistas efetuadas a diferentes grupos de atores, direta ou indiretamente relacionados com o ensino superior em Moçambique. Os resultados da análise conduziram à sistematização de um conjunto de linhas de força e ao traçar de conclusões, contributivas para a melhoria do quadro de referência sobre políticas, conceções e práticas do ensino superior em Moçambique.

The massive adoption of sophisticated mobile devices and applications led to the increase of mobile data in the last decade, which it is expected to continue. This increase of mobile data negatively impacts the network planning and dimension, since core networks are heavy centralized. Mobile operators are investigating atten network architectures that distribute the responsibility of providing connectivity and mobility, in order to improve the network scalability and performance. Moreover, service providers are moving the content servers closer to the user, in order to ensure high availability and performance of content delivery. Besides the e orts to overcome the explosion of mobile data, current mobility management models are heavy centralized to ensure reachability and session continuity to the users connected to the network. Nowadays, deployed architectures have a small number of centralized mobility anchors managing the mobile data and the mobility context of millions of users, which introduces issues related to performance and scalability that require costly network mechanisms. The mobility management needs to be rethought out-of-the box to cope with atten network architectures and distributed content servers closer to the user, which is the purpose of the work developed in this Thesis. The Thesis starts with a characterization of mobility management into well-de ned functional blocks, their interaction and potential grouping. The decentralized mobility management is studied through analytical models and simulations, in which di erent mobility approaches distinctly distribute the mobility management functionalities through the network. The outcome of this study showed that decentralized mobility management brings advantages. Hence, it was proposed a novel distributed and dynamic mobility management approach, which is exhaustively evaluated through analytical models, simulations and testbed experiments. The proposed approach is also integrated with seamless horizontal handover mechanisms, as well as evaluated in vehicular environments. The mobility mechanisms are also speci ed for multihomed scenarios, in order to provide data o oading with IP mobility from cellular to other access networks. In the pursuing of the optimized mobile routing path, a novel network-based strategy for localized mobility is addressed, in which a replication binding system is deployed in the mobility anchors distributed through the access routers and gateways. Finally, we go further in the mobility anchoring subject, presenting a context-aware adaptive IP mobility anchoring model that dynamically assigns the mobility anchors that provide the optimized routing path to a session, based on the user and network context. The integration of dynamic and distributed concepts in the mobility management, such as context-aware adaptive mobility anchoring and dynamic mobility support, allow the optimization of network resources and the improvement of user experience. The overall outcome demonstrates that decentralized mobility management is a promising direction, hence, its ideas should be taken into account by mobile operators in the deployment of future networks.

Esta dissertação tem como objetivo a divulgação de temas e de algoritmos presentes nos tratados de aritmética prática do século XVI. Pretendemos delinear a atuação dos seus autores face aos desafios do mundo mercantil envolvente. Sendo o nosso «ator principal» Ruy Mendes, esta escolha deve-se a dois motivos: entre os três autores é aquele que tem sido menos mencionado e estudado; os interesses de Mendes parecem-nos mais distantes do mundo mercantil. Assim, no desenrolar deste estudo apresentaremos a Prática do ponto de vista da estrutura e organização, contemplando os seguintes pontos: uma Matemática básica; a Matemática pour elle Même; uma Matemática para o comércio. Neste último ponto incluiremos as regras locais do comércio português: a regra de quarto e vintena e a regra da conta de Flandres. Para cada assunto é realizado um estudo comparativo com os dois tratados da mesma época: o Tratado da Pratica d'Arismetica de Gaspar Nicolas e o Tratado da Arte d'Arismetica de Bento Fernandes. Apesar de se tratar de autores já referidos por alguns historiadores, consideramos que não foram ainda estudados do ponto de vista do interesse intrínseco presente no conhecimento histórico da Matemática, bem como na sua atuação relativamente à divulgação do cálculo aritmético e do seu contributo para o desenvolvimento da Matemática através de problemas práticos.

Although the genetic code is generally viewed as immutable, alterations to its standard form occur in the three domains of life. A remarkable alteration to the standard genetic code occurs in many fungi of the Saccharomycotina CTG clade where the Leucine CUG codon has been reassigned to Serine by a novel transfer RNA (Ser-tRNACAG). The host laboratory made a major breakthrough by reversing this atypical genetic code alteration in the human pathogen Candida albicans using a combination of tRNA engineering, gene recombination and forced evolution. These results raised the hypothesis that synthetic codon ambiguities combined with experimental evolution may release codons from their frozen state. In this thesis we tested this hypothesis using S. cerevisiae as a model system. We generated ambiguity at specific codons in a two-step approach, involving deletion of tRNA genes followed by expression of non-cognate tRNAs that are able to compensate the deleted tRNA. Driven by the notion that rare codons are more susceptible to reassignment than those that are frequently used, we used two deletion strains where there is no cognate tRNA to decode the rare CUC-Leu codon and AGG-Arg codon. We exploited the vulnerability of the latter by engineering mutant tRNAs that misincorporate Ser at these sites. These recombinant strains were evolved over time using experimental evolution. Although there was a strong negative impact on the growth rate of strains expressing mutant tRNAs at high level, such expression at low level had little effect on cell fitness. We found that not only codon ambiguity, but also destabilization of the endogenous tRNA pool has a strong negative impact in growth rate. After evolution, strains expressing the mutant tRNA at high level recovered significantly in several growth parameters, showing that these strains adapt and exhibit higher tolerance to codon ambiguity. A fluorescent reporter system allowing the monitoring of Ser misincorporation showed that serine was indeed incorporated and possibly codon reassignment was achieved. Beside the overall negative consequences of codon ambiguity, we demonstrated that codons that tolerate the loss of their cognate tRNA can also tolerate high Ser misincorporation. This raises the hypothesis that these codons can be reassigned to standard and eventually to new amino acids for the production of proteins with novel properties, contributing to the field of synthetic biology and biotechnology.

Candida albicans is the major fungal pathogen in humans, causing diseases ranging from mild skin infections to severe systemic infections in immunocompromised individuals. The pathogenic nature of this organism is mostly due to its capacity to proliferate in numerous body sites and to its ability to adapt to drastic changes in the environment. Candida albicans exhibit a unique translational system, decoding the leucine-CUG codon ambiguously as leucine (3% of codons) and serine (97%) using a hybrid serine tRNA (tRNACAGSer). This tRNACAGSer is aminoacylated by two aminoacyl tRNA synthetases (aaRSs): leucyl-tRNA synthetase (LeuRS) and seryl-tRNA synthetase (SerRS). Previous studies showed that exposure of C. albicans to macrophages, oxidative, pH stress and antifungals increases Leu misincorporation levels from 3% to 15%, suggesting that C. albicans has the ability to regulate mistranslation levels in response to host defenses, antifungals and environmental stresses. Therefore, the hypothesis tested in this work is that Leu and Ser misincorporation at CUG codons is dependent upon competition between the LeuRS and SerRS for the tRNACAGSer. To test this hypothesis, levels of the SerRS and LeuRS were indirectly quantified under different physiological conditions, using a fluorescent reporter system that measures the activity of the respective promoters. Results suggest that an increase in Leu misincorporation at CUG codons is associated with an increase in LeuRS expression, with levels of SerRS being maintained. In the second part of the work, the objective was to identify putative regulators of SerRS and LeuRS expression. To accomplish this goal, C. albicans strains from a transcription factor knock-out collection were transformed with the fluorescent reporter system and expression of both aaRSs was quantified. Alterations in the LeuRS/SerRS expression of mutant strains compared to wild type strain allowed the identification of 5 transcription factors as possible regulators of expression of LeuRS and SerRS: ASH1, HAP2, HAP3, RTG3 and STB5. Globally, this work provides the first step to elucidate the molecular mechanism of regulation of mistranslation in C. albicans.

O regulamento técnico relativo ao projecto, construção, exploração e manutenção de redes de gás combustível canalizado tem sofrido algumas alterações ao longo dos últimos treze anos. Este facto está directamente relacionado com o projecto de introdução do gás natural (GN) no território nacional, considerado como uma das prioridades na política energética dos últimos governos. A área geográfica de concessão de serviço público de importação de gás natural e do seu transporte e fornecimento através da rede de alta pressão encontra-se actualmente alargada a todos os municípios do continente, por força da Portaria n.º 2/2001, de 2 de Janeiro.
