Lo scopo del presente elaborato è lo studio della regolarità stradale. La regolarità è una caratteristica fondamentale sia dal punto di vista dell’utenza, poiché da essa dipendono il comfort, ma soprattutto, la sicurezza della guida, sia dal punto di vista dell’ente proprietario della strada, perché dalla regolarità dipende la pianificazione della manutenzione della rete viaria, con tutti i pesi economici che essa comporta. Da questa piccola considerazione si comprende perché nell’ultimo secolo l’argomento abbia suscitato sempre maggiore interesse, vedendo la nascita, sin dagli anni ’50, di strumenti sempre più sofisticati, grazie allo sviluppo dell’elettronica, e sempre più veloci nell’acquisizione dei dati, per far fronte al contemporaneo sviluppo della rete viaria. Nel primo capitolo si affronterà la tematica della regolarità stradale, cercando di comprenderne la natura e i suoi livelli di degrado, dei quali viene fatta un’ampia descrizione, comprensiva di cause scatenanti. Inoltre si fa un accenno al Catasto Stradale, poiché costituisce uno strumento essenziale per la gestione dell’infrastruttura viaria e per l’ottimizzazione delle risorse destinate alla sua manutenzione e alla sua sicurezza; infine si parla delle tecnologie finalizzate al rilievo della regolarità. Il secondo capitolo è dedicato al panorama normativo in materia di regolarità stradale. Le varie norme sono commentate nel dettaglio, data l’importanza di una standardizzazione nella calibrazione e nell’uso degli strumenti di rilievo e nelle procedure di stima della regolarità. Il capitolo successivo contiene una trattazione sugli indici di regolarità, ma l’attenzione è in particolar modo rivolta all’IRI, l’indice internazionale di irregolarità. Per le caratteristiche con cui è stato concepito, cioè stabilità temporale, validità e compatibilità, è stato assunto come scala di riferimento internazionale, a cui gli altri indici possono riferirsi. Viene quindi illustrato il significato di IRI, il modello matematico su cui si basa la sua definizione, per passare poi al procedimento analitico e a quello informatico con il quale viene calcolato. Essendo la presente tesi avvenuta in collaborazione con la ditta Siteco Informatica di Bologna, nel capitolo quarto, sono stati analizzati gli strumenti installati sul veicolo ad alto rendimento (VAR) messo a disposizione dalla ditta stessa. Gli strumenti consistono principalmente in dispositivi per il calcolo del posizionamento e dell’assetto del veicolo, in un profilometro laser per il rilievo della regolarità stradale e in un laser scanner, di cui si è tentato di studiarne le potenzialità nell’ambito dell’oggetto di questa tesi. Il percorso di ricerca ha incluso l’esperienza diretta in sito tramite varie campagne di rilievo, per meglio comprendere il mezzo e la natura dei dati ottenuti. Si è quindi ripercorso l’intero processo di elaborazione dei dati grezzi di rilievo, così com’è strutturato per le finalità della ditta collaborante, per utilizzare poi il dato finito per finalità ingegneristiche.

Aim of the research: to develop a prototype of homogeneous high-throughput screening (HTS) for identification of novel integrin antagonists for the treatment of ocular allergy and to better understand the mechanisms of action of integrin-mediated levocabastine antiallergic action. Results: This thesis provides evidence that adopting scintillation proximity assay (SPA) levocabastine (IC50=406 mM), but not the first-generation antihistamine chlorpheniramine, displaces [125I]fibronectin (FN) binding to human a4b1 integrin. This result is supported by flow cytometry analysis, where levocabastine antagonizes the binding of a primary antibody to integrin a4 expressed in Jurkat E6.1 cells. Levocabastine, but not chlorpheniramine, binds to a4b1 integrin and prevents eosinophil adhesion to VCAM-1, FN or human umbilical vein endothelial cells (HUVEC) cultured in vitro. Similarly, levocabastine affects aLb2/ICAM-1-mediated adhesion of Jurkat E6.1 cells. Analyzing the supernatant of TNF-a-treated (24h) eosinophilic cells (EoL-1), we report that levocabastine reduces the TNF-a-induced release of the cytokines IL-12p40, IL-8 and VEGF. Finally, in a model of allergic conjunctivitis, levocastine eye drops (0.05%) reduced the clinical aspects of the early and late phase reactions and the conjunctival expression of a4b1 integrin by reducing infiltrated eosinophils. Conclusions: SPA is a highly efficient, amenable to automation and robust binding assay to screen novel integrin antagonists in a HTS setting. We propose that blockade of integrinmediated cell adhesion might be a target of the anti-allergic action of levocabastine and may play a role in preventing eosinophil adhesion and infiltration in allergic conjunctivitis.

Über die Liniarität der Teichmüllerschen Modulgruppe des Torus mit zwei Punktierungen. In meiner Arbeit beschäftige ich mich mit Darstellungen der Teichmüllerschen Modulgruppe des Torus mit zwei Punktierungen. Mein Ansatz hierbei ist, die Teichmüllersche Modulgruppe in eine p-adische Liegruppe einzubetten. Sei nun F die von zwei Elementen erzeugte freie Gruppe und Aut(F) die Automorphismengruppe von F. Inhalt des ersten Kapitels ist es nun zu zeigen, daß folgende Aussagen äquivalent sind: - Die Teichmüllersche Modulgruppe des Torus mit zwei Punktierungen ist linear, - Aut(F)ist linear, - F besitzt eine p-Kongruenzstruktur, deren Folgen- glieder von Aut(F) festgehalten werden, also charak- teristisch sind. Im zweiten Kapitel wird unter anderem gezeigt, daß es eine Einbettung einer Untergruppe endlichen Indexes der Aut(F) in die Automorphismengruppe einer einfachen p-adischen Liegruppe gibt. Bisher ist unbekannt, ob die Buraudarstellung treu ist.In dieser Arbeit wird ein unendliches, lineares Gleichungssystem, dessen Lösungen gerade die Koeffizienten der Wörter des Kernes der Buraudarstellung sind, vorgestellt.Im dritten Kapitel wird mit den Methoden des 1.Kapitels gezeigt, daß der Torus mit zwei Punktierungen genau dann linear ist, wenn die Teichmüllersche Modulgruppe der Sphäre mit 5 Punktierungen es auch ist. Bekanntlich ist die 4. Braidgruppe linear. Nun ist aber die 4. Braidgruppe letztlich die Teichmüllersche Modulgruppe der abgeschlossenen Kreisscheibe mit 5 Punktierungen. Wenn man nun deren Randpunkte miteinander identifiziert und anschließend wegläßt, erhält man die 5-fach punktiereSphäre.Mit der eben beschriebenen Abbildung kann man zeigen, daß die Teichmüllersche Modulgruppe der fünffach punktierten Sphäre linear ist.

This study compared the efficiency of air abrasion on enamel caries with selective enamel powder (SEP) or with alumina powder and a negative and positive control group. Ninety-three extracted molars with non-cavitated incipient enamel lesions were selected. After embedding the roots in resin, each lesion was sectioned perpendicular to the surface and photographed. Each lesion was classified microscopically as having or not having dentin involvement. The lesions were distributed into four groups with an equal number of enamel caries with or without dentin involvement. Each group was treated differently: Group 1 had SEP abrasion, Group 2 had alumina abrasion, Group 3 had sodium bicarbonate abrasion (negative control) and Group 4 had bur treatment (positive control). The surface was rephotographed after treatment. Superimposition of the photographs identified areas of "correct-excavation," "under-excavation" and "over-excavation." There were no statistical differences between lesions treated with or without dentin involvement for Groups 2 through 4. However, in the SEP group, all measured areas were significantly influenced by dentin involvement. In pairwise comparisons, no statistical differences were found between the alumina and bur groups. The SEP group, however, showed statistically significant differences for each area compared to the alumina group in enamel caries without dentin involvement. SEP performed as well as alumina and bur in lesions with dentin involvement. SEP is different in its ablative properties toward caries with dentin involvement or no dentin involvement. In terms of dental treatment, SEP seems to have a diagnostic potential for enamel lesions before operative intervention in patients with high caries risk.

OBJECTIVE: The aim of this study was to assess the glycosaminoglycan (GAG) content in hip joint cartilage in mature hips with a history of slipped capital femoral epiphysis (SCFE) using delayed gadolinium-enhanced MRI of cartilage (dGEMRIC). METHODS: 28 young-adult subjects (32 hips) with a mean age of 23.8+/-4.0 years (range: 18.1-30.5 years) who were treated for mild or moderate SCFE in adolescence were included into the study. Hip function and clinical symptoms were evaluated with the Harris hip score (HHS) system at the time of MRI. Plain radiographic evaluation included Tonnis grading, measurement of the minimal joint space width (JSW) and alpha-angle measurement. The alpha-angle values were used to classify three sub-groups: group 1=subjects with normal femoral head-neck offset (alpha-angle <50 degrees ), group 2=subjects with mild offset decrease (alpha-angle 50 degrees -60 degrees ), and group 3=subjects with severe offset decrease (alpha-angle >60 degrees ). RESULTS: There was statistically significant difference noted for the T1(Gd) values, lateral and central, between group 1 and group 3 (p-values=0.038 and 0.041). The T1(Gd) values measured within the lateral portion were slightly lower compared with the T1(Gd) values measured within the central portion that was at a statistically significance level (p-value <0.001). HHS, Tonnis grades and JSW revealed no statistically significant difference. CONCLUSION: By using dGEMRIC in the mid-term follow-up of SCFE we were able to reveal degenerative changes even in the absence of joint space narrowing that seem to be related to the degree of offset pathology. The dGEMRIC technique may be a potential diagnostic modality in the follow-up evaluation of SCFE.

A triple cell co-culture model was recently established by the authors, consisting of either A549 or 16HBE14o- epithelial cells, human blood monocyte-derived macrophages and dendritic cells, which offers the possibility to study the interaction of xenobiotics with those cells. The 16HBE14o- containing co-culture model mimics the airway epithelial barrier, whereas the A549 co-cultures mimic the alveolar type II-like epithelial barrier. The goal of the present work was to establish a new triple cell co-culture model composed of primary alveolar type I-like cells isolated from human lung biopsies (hAEpC) representing a more realistic alveolar epithelial barrier wall, since type I epithelial cells cover >93% of the alveolar surface. Monocultures of A549 and 16HBE14o- were morphologically and functionally compared with the hAEpC using laser scanning microscopy, as well as transmission electron microscopy, and by determining the epithelial integrity. The triple cell co-cultures were characterized using the same methods. It could be shown that the epithelial integrity of hAEpC (mean ± SD, 1180 ± 188 Ω cm(2)) was higher than in A549 (172 ± 59 Ω cm(2)) but similar to 16HBE14o- cells (1469 ± 156 Ω cm(2)). The triple cell co-culture model with hAEpC (1113 ± 30 Ω cm(2)) showed the highest integrity compared to the ones with A549 (93 ± 14 Ω cm(2)) and 16HBE14o- (558 ± 267 Ω cm(2)). The tight junction protein zonula occludens-1 in hAEpC and 16HBE14o- were more regularly expressed but not in A549. The epithelial alveolar model with hAEpC combined with two immune cells (i.e. macrophages and dendritic cells) will offer a novel and more realistic cell co-culture system to study possible cell interactions of inhaled xenobiotics and their toxic potential on the human alveolar type I epithelial wall.

Melatonin is an important endocrine signal for darkness in mammals. Transcriptional activation of the arylalkylamine-N-acetyltransferase gene encoding for the penultimate enzyme in melatonin synthesis drives the daily rhythm of the hormone in the pineal gland of rodents. Rhythmic arylalkylamine-N-acetyltransferase expression is controlled by the cAMP-signal transduction pathway and involves the activation of ?-adrenergic receptors and the inducible cAMP early repressor. In addition, the rat arylalkylamine-N-acetyltransferase promoter contains an E-box element which can interact with clock proteins. Moreover, the pineal gland of mice shows a circadian rhythm in clock proteins such as the transcriptional repressor Period1, which has been shown to control rhythmic gene expression in a variety of tissues. However, the role of Period1 in the regulation of pineal melatonin synthesis is still unknown. Therefore, circadian rhythms in arylalkylamine-N-acetyltransferase, ?-adrenergic receptor, and inducible cAMP early repressor mRNA levels (real time PCR), arylalkylamine-N-acetyltransferase enzyme activity (radiometric assay) and melatonin concentration radio immuno assay (RIA) were analyzed in the pineal gland of mice with a targeted deletion of the Period1 gene (Per1-/-) and the corresponding wildtype. In Per1-/- the amplitude in arylalkylamine-N-acetyltransferase expression was significantly elevated as compared to wildtype. In contrast, ?-adrenergic receptor and inducible cAMP early repressor mRNA levels were not affected by the Period1-deficiency. This indicates that the molecular clockwork alters the amplitude of arylalkylamine-N-acetyltransferase expression. In vitro, pineal glands of Per1-/- mice showed a day night difference in arylalkylamine-N-acetyltransferase expression with high levels at night. This suggests that a deficient in Period1 elicits similar effects as the activation of the cAMP-signal transduction pathway in wildtype mice.

Rapid speciation in Lake Victoria cichlid fish of the genus Pundamilia may be facilitated by sexual selection: female mate choice exerts sexual selection on male nuptial coloration within species and maintains reproductive isolation between species. However, declining water transparency coincides with increasingly dull coloration and increasing hybridization. In the present study, we investigated the mechanism underlying this pattern in Pundamilia nyererei, a species that interbreeds with a sister species in turbid but not in clear water. We compared measures of intraspecific sexual selection between two populations from locations that differ in water transparency. First, in laboratory mate-choice experiments, conducted in clear water and under broad-spectrum illumination, we found that females originating from turbid water have significantly weaker preferences for male coloration than females originating from clear water. Second, both the hue and body coverage of male coloration differ between populations, which is consistent with adaptation to different photic habitats. These findings suggest that the observed relationship between male coloration and water transparency is not mediated by environmental variation alone. Rather, female mating preferences are indicated to have changed in response to this variation, constituting the first evidence for intraspecific preference-trait co-evolution in cichlid fish.
