Considering the increasing consumption of saturated fat and glucose in diets worldwide and its possible association to carcinogenesis, this investigation analysed the proliferation profile of nonmalignant human prostate epithelial cells after exposure to elevated levels of fat and glucose. PNT1A cells were cultured with palmitate (100 or 200 mu M) and/or glucose (450mg/dl) for 24 or 48 h. Treated cells were evaluated for viability test and cell proliferation (MTS assay). AKT and AMPK phosphorylation status were analysed by Western blotting. After 24 h of high-fat alone or associated with high-glucose treatment, there was an increase in AMPK and AKT activation associated to unchanged MTS-cell proliferation. Following 48 h of high-fat but not high-glucose alone, cells decreased AMPK activation and maintained elevated AKT levels. These data were associated to increased cell proliferation after further high-fat treatment. After longer high-fat exposure, MTS revealed that cells remained proliferating. High-glucose alone or associated to high-fat treatment was not able to increase cell proliferation and AKT activation. A high-fat medium containing 100 mu M of palmitate stimulates proliferation in PNT1A cells by decreasing the activation of AMPK and increasing activation of AKT after longer exposure time. These findings improve the knowledge about the negative effect of high levels of this saturated fatty acid on proliferative disorders of prostate.

The oral cancer model in hamsters shows many simílarities with developmental oral cancer in humans. The proliferating capacity is one the most characteristics of neoplásica ce/Is and detection of these ce/Is allow us, throughout of its counting, to achieve an estimated tumour growing index, with a consequent repercussion about prognostic and in the treatment of those lesions. 40 golden Syrian hamsters, both genders, aged between two to six mouth and weight 150g in average were used. The left síde of tangue of each animal was painted for eight consecutive weeks, with a solution of DMBA. Tongues were removed, fixed in 10% buffered fonnal solution. The histological slides were stained by AgNOR technique and by PCNA and β-catenin immunohistochemical antibodies. Statistical analyzes were performed by ANOVA one-way test and Tukey test. We may conclude that an association between AgNOR and PCNA might indicate the higher proliferating activity of the analyzed celIs. The experimental carcinogenesis model in hamster tongue is an available methodology for immunohistochemistry study. And finally, PCNA and β-catenin immunohistochemical antibodies may be used to analyze possible premalignant areas in oral leukoplakia

Mammary cancer is a multifactorial disease that is believed to be caused by genetic and environmental factors. Among the environmental factors, pyrethroids appear to be able to participate in carcinogenesis through several mechanisms, and have been shown to be associated to mammary tumors in canines. In order to investigate the possible rule of pyrethroid on DNA lesion in mammary tissue we compare the comet assay results between mammary tumor bearing dogs with and without pyrethroid associated to the peri mammary adipose tissue or the tumor itself. The pyrethroids presence was assessed by High Performance Liquid Chromatography (HPLC) and the DNA damage was assessed by the comet assay as previously described. Despite of correlation between DNA damage and tumor histologic aggressiveness, association between the severity of DNA damage and different types of mammary carcinoma was not found. Although pyrethroids were present in 22% of tumors and peritumoral adipose tissue, no difference in the degree DNA damage between the exposed and non exposed cells to pyrethroids were found. As future perspectives for this work, our group will evaluate the relationship of pyrethroids presence in tumors with its angiogenic potential. The use of pesticides in Brazil has intensified over the years. And since 2009 it was ranked as the largest market for pesticides worldwide. The consequential diffuse contamination of the environment is therefore due to the increasing number of chemicals of different classes, origins and modes of action. Little is known about the action of pesticides on human health in situations of co-exposure. Possible toxic effects are not restricted to agricultural and industrial workers, but also the general population that may be exposed continuously to its residues in food and water. Although these pesticides are mostly present in the environment at low doses, it must be considered that possible cumulative or synergistic effects may occur when there are concurrent or sustained exposure for two or more of these agents, which can lead to late manifestation of subclinical damages, sometimes irreversible. Thus, the specific objective of this study was to assess the effect of carcinogenesis promotion of a mixture of pesticides at low doses and analyze the phenomena of cell proliferation and apoptosis in rat liver. A total of 50 male Lewis rats was separated into 5 groups for 8 weeks in a medium term hepatocarcinogenesis model. The three different classes of pesticides (dieldrin, dicofol, endosulfan, dichlorvos and permethrin), whose residues were detected by ANVISA during the period from 2001 to 2005 in tomatoes cultures, were added to the feed of rats initiated to hepatocarcinogenesis with diethylnitrosamine (DEN- 200mg/kg ip). We used two different mixtures, one with no toxic effects at doses (MEX1) referring to the NOEL (no-observed-effect level) and another at doses LOEL / LEL / LOAEL (Lowest-observed-effect level / Lowest-effect level / Lowest -observed-adverse-effect level), to the installation of adverse effects (MEX2), derived from chronic studies. Helicobacter pylori (H. pylori) is a common gastric pathogen that has infected more than 50% of the population of the world and it has been associated with chronic gastritis, gastric ulcers, duodenal ulcer, and gastric cancer. Although, almost all infected people develop gastritis, there is a variety of clinical outcomes, and only a minority (<1%) of infected individuals develop gastric cancer. There are evidences which suggest that the chronic inflammatory reaction caused by the bacterial infection may be involved in the production of reactive oxygen species or reactive nitrogen species. It may lead to DNA damage, which together with the cellular response could lead to gene mutations, chromosomal aberrations characterizing genomic instability that may represent the early step in gastric carcinogenesis. The extent and severity of gastric mucosal inflammation, as well as the clinical outcome of the infection, depend on a number of factors, including the host genetic susceptibility such SNP T3801 CYP1A1, immune response, age at which the infection was acquired, environmental factors, especially dietary and bacterial virulence factors. Due to the risk of developing gastric cancer in humans infected by H. pylori, we used the Comet Assay to investigate the influence of the SNP T3801C CYP1A1 on levels of oxidative DNA damage in gastric epithelial cells. The study was conducted with biopsies from the gastric antrum and corpus of 103 H. pylori-infected patients and 24 uninfected control patients. Genotype of SNP T3801C CYP1A1 was determined by PCR-RFLP and DNA damage levels were measured in gastric mucosal cells from antrum and corpus by the Comet assay. Levels of DNA damage in gastric mucosa cells from antrum and corpus of H. pylori-infected patients with mild, moderate, severe gastritis, and gastric cancer were significantly higher compared to uninfected normal mucosa cells.
