Envenomation via snakebites is an important public health problem in many tropical and subtropical countries that, in addition to mortality, can result in permanent sequelae as a consequence of local tissue damage, which represents a major challenge to antivenom therapy. Venom phospholipases A(2) (PLA(2)s) and PLA(2)-like proteins play a leading role in the complex pathogenesis of skeletal muscle necrosis, nevertheless their precise mechanism of action is only partially understood. Recently, detailed structural information has been obtained for more than twenty different members of the PLA(2)-like myotoxin subfamily. In this review, we integrate the available structural, biochemical and functional data on these toxins and present a comprehensive hypothesis for their myotoxic mechanism. This process involves an allosteric transition and the participation of two independent interaction sites for docking and disruption of the target membrane, respectively, leading to a five-step mechanism of action. Furthermore, recent functional and structural studies of these toxins complexed with ligands reveal diverse neutralization mechanisms that can be classified into at least three different groups. Therefore, the data summarized here for the PLA(2)-like myotoxins could provide a useful molecular basis for the search for novel neutralizing strategies to improve the treatment of envenomation by viperid snakes. Several species of octopus are considered venomous due to toxins present in the glands connected to their "beak", which may be associated with hunt and kill of prey. Herein, we report an accident involving a common octopus (Octopus vulgaris) that injured an instructor during a practical biology lesson and provoked an inflamed infiltrated plaque on the hand of the victim. The lesion was present for about three weeks and was treated with cold compresses and anti-inflammatory drugs. It was healed ten days after leaving a hyperchromic macule at the bite site. The probable cause of the severe inflammation was the digestive enzymes of the glands and not the neurotoxins of the venom.

Scorpions are among the oldest living groups of animals, and are present in almost every continent. Although many studies are made about the toxins present in their venom, little is known about their behavior. The aim of this paper is to investigate the expression of agressiveness in the brazilian yellow scorpion Tityus serrulatus, specifically the relation between agressiveness, motor activity and feeding condition. The study was divided in two parts. First, we used 16 adult individuals, placed in two terrarium, to establish an ethogram. Each group was observed for 36h, and the behaviors displayed were described and categorized as agonistic or non-agonistic. In the second part of the experiment, we used 32 adult animals in three different nutritional states: Feeding (still ingesting food), Sated (1 to 4 days since last meal) and Deprived (14 to 25 days since last meal). The individuals were paired, each pair placed in a terrarium and observed for 30min. Behaviors displayed were timed in seconds. Our results show that only Feeding individuals displayed agonism towards others, and no relation was observed between motor activity and feeding conditions

Throughout the analysis of the collections JJ, CEVAP, IBSP, ZUEC and UEL, a study on the ophiofauna of Botucatu was made. We present a preliminary list of the species of snakes here found, with small comments on their natural history. The number of specimens studied was 943. We registered, for Botucatu, 51 species of snakes, distributed in 31 genera and 6 families. The families here found were Anomalepididae (1 spp.), Boidae (1 spp.), Colubridae (7 spp.), Dipsadidae (32 spp.), Elapidae (3 spp.) and Viperidae (7 spp.). The analysis of the relative abundance shows that Dipsadidae was the most abundant family, with n=425 (44,83%), followed by Viperidae, with n=388 (40,93%), Boidae, n=62 (6,54%), Colubridae, n=57 (6,01%), Elapidae, n=15 (1,6%) and, at last, Anomalepididae, with n=1 (0,1%). The five more representative species were Crotalus durissus (n=135, 14,31%), Oxyrhopus guibei (n=123, 12,8%) Bothropoides jararaca (N=121, 12,6%), Bothropoides neuwiedi (N=95, 9,88%) and Sibynomorphus mikani (N=65, 6,76%). A higher number of individuals collected was registered for the months of january to april that, together, sum up to almost 50% of the total. The months of june to september registered a lower number of individuals. The spacial distribution analysis shows that a higher number of snakes was found on the country areas of Botucatu (n=270, 41,54%), specially on pastures. Due to the lack of studies of Botucatu’s ophiofauna, this list is probably underestimated. It is mandatory that future studies approaching this group and its ecological components on this region are made, using appropriate sampling methodologies, in order to form an accurate list of the species of snakes of Botucatu

A bothropstoxina-I (BthTX-I) é uma fosfolipase A2 (PLA2) Lys49 miotóxica isolada do veneno da Bothrops jararacussu. Embora seja desprovida de atividade neurotóxica in vivo, esta toxina bloqueia a transmissão neuromuscular in vitro. A relação entre as atividades miotóxica e paralisante da BthTX-I ainda não está esclarecida. A crotapotina corresponde à subunidade não-enzimática da crotoxina, principal fração tóxica do veneno da Crotalus durissus terrificus. Isoladamente a crotapotina é atóxica, porém atua como carreadora da PLA2 Asp49 da crotoxina, potencializando sua ação neurotóxica. Esta proteína também é capaz de se complexar com outras PLA2s (Asp49 ou Lys49) de venenos ofídicos, alterando suas toxicidades. Neste trabalho avaliamos a influência da crotapotina sobre o bloqueio neuromuscular e a atividade miotóxica da BthTX-I in vitro. Preparações do nervo frênico-músculo diafragma de camundongos machos foram montadas em cubas para o registro das contrações musculares evocadas direta e indiretamente. Cortes transversais do músculo foram submetidos à coloração por hematoxilina e eosina para a avaliação do padrão morfológico. A BthTX-I (1 μM) isoladamente, ou pré-incubada com crotapotina (2 M) à 35 ºC por 30 minutos, foram adicionadas às preparações. A análise dos dados foi realizada por testes não paramétricos (p<0.05). A BthTX-I induziu bloqueio irreversível e tempo-dependente das contrações musculares diretas e indiretas. O tempo para o bloqueio de 50% das contrações indiretas (18,98 ± 1,94 min, n=4) foi significativamente menor que o das diretas (45,97 ± 5,61 min, n=5). A pré-incubação com a crotapotina não alterou de forma significativa o bloqueio das contrações diretas ou indiretas induzidos pela BthTX-I. Isoladamente, a crotapotina não afetou as contrações... The Natural Rubber Latex (NRL) from Hevea brasiliensis has shown promise in biomedical applications due to its low cost, easy handling, mechanical properties and biocompatibility, being used for bone regeneration and wound healing due to its natural stimulus to angiogenesis. The aim of this work was to incorporate Casearia sylvestris Sw. extract in NRL biomembranes and study its release behavior. The complex membrane-extract has as object of study a new approach of using C. sylvestris extract in the treatment of wounds, for possessing antiseptic activity, anti-inflammatory and analgesic properties. The C. sylvestris species (Salicaceae), popularly known as "guaçatonga", presents great distribution and is used in folk medicine as antiulcer, wound healing, anti-snake venom, properties which have been proven and related to clerodane diterpenes (casearins A-X). The release rate of C. sylvestris compounds from extract-membrane complex was monitored and analyzed using the method of optical spectroscopy (UV-VIS). The release varied with temperature ranging from 14 to 33 days, releasing more than 90%, with an interesting and promising biomedical application, such as wound healing and burns.

Tissue engineering has special interest in bone tissue aiming at future medical applications Studies have focused on recombinant human bone morphogenetic protein-2 (rhBMP-2) and natural latex proteins due to the osteogenic properties of rhBMP-2 and the angiogenic characteristic of fraction 1 protein (P-1) extracted from the rubber tree Hevea brasiliensis. Furthermore, heterologous fibrin sealant (FS) has been shown as a promising alternative in regenerative therapies. The aim of this study was to evaluate these substances for the repair of bone defects in rats. A bone defect measuring 3 mm in diameter was created in the proximal metaphysis of the left tibia of 60 rats and was implanted with rhBMP-2 or P-1 in combination with a new heterologous FS derived from snake venom. The animals were divided into six groups: control (unfilled bone defect), rhBMP-2 (defect filled with 5 mu g rhBMP-2), P-1 (defect filled with 5 mu g P-1), FS (defect filled with 8 mu g FS), FS/rhBMP-2 (defect filled with 8 mu g FS and 5 mu g rhBMP-2), FS/P-1 (defect filled with 8 mu g FS and 5 mu g P-1). The animals were sacrificed 2 and 6 weeks after surgery. The newly formed bone projected from the margins of the original bone and exhibited trabecular morphology and a disorganized arrangement of osteocyte lacunae. Immunohistochemical analysis showed intense expression of osteocalcin in all groups. Histometric analysis revealed a significant difference in all groups after 2 weeks (p < 0.05), except for the rhBMP-2 and FS/rhBMP-2 groups (p > 0.05). A statistically significant difference (p < 0.05) was observed in all groups after 6 weeks in relation to the volume of newly formed bone in the surgical area. In conclusion, the new heterologous fibrin sealant was found to be biocompatible and the combination with rhBMP-2 showed the highest osteogenic and osteoconductive capacity for bone healing. These findings suggest a promising application of this combination in the regeneration surgery.

A myographic study was performed to compare the neuromuscular effects of venoms and crotoxin-like proteins from Crotalus durissus ruruima and Crotalus durissus cumanensis in mice phrenic-diaphragm preparation. It was concluded that both venoms present neurotoxic activity as a consequence of their crotoxin content. Furthermore, crotoxin from C.d. cumanensis is more potent than that from C.d. ruruima venom. At the concentration range in which both venoms express neurotoxic activity, only C.d. cumanensis venom also manifest a direct myotoxic effect that probably involves the synergic participation of other components than crotoxin.
