Este trabajo de grado busca analizar el papel que juegan los principios en las fallas del Régimen Internacional de Pagos a la hora de responder a situaciones de crisis, específicamente, en la actuación del Fondo Monetario Internacional durante la crisis económica argentina de 2001. El Régimen, atado al cumplimiento de sus principios, actuó de manera tal que produjo unas fallas que terminaron exacerbando la crisis. Mediante el análisis de fuentes primarias y secundarias, y con métodos tanto cualitativos como cuantitativos se busca demostrar que efectivamente, los principios en los que se fundamentan las actuaciones de los regímenes, que son creencias, tienden a tornarse dogmáticos, por lo tanto, se hacen resistentes al cambio y no permiten a las instituciones obrar de manera que se ajuste a la realidad. Es así que los principios llegan agravar las crisis que debían solucionar, tal y como sucedió en Argentina en 2001.

El objetivo de la presente monografía de grado es explicar cómo las reformas del FMI implementadas en el Ecuador desde 1984 generaron una reestructuración de la economía que permitió la articulación organizativa e ideológica del Movimiento de Unidad Plurinacional Pachakutik (MUPP). Se analiza y explica el impacto de las reformas del FMI en la organización del MUPP y su accionar político en el Ecuador, desde la perspectiva de la teoría del neogramscianismo de Robert Cox, para entender el papel que las primeras tuvieron en la conformación y configuración del segundo. Se efectúa una revisión de textos académicos y programáticos del MUPP, entrevistas realizadas por terceros a los dirigentes del movimiento así como entrevistas realizadas por la autora.

Charge transfer properties of DNA depend strongly on the π stack conformation. In the present paper, we identify conformations of homogeneous poly-{G}-poly-{C} stacks that should exhibit high charge mobility. Two different computational approaches were applied. First, we calculated the electronic coupling squared, V2, between adjacent base pairs for all 1 ps snapshots extracted from 15 ns molecular dynamics trajectory of the duplex G15. The average value of the coupling squared 〈 V2 〉 is found to be 0.0065 eV2. Then we analyze the base-pair and step parameters of the configurations in which V2 is at least an order of magnitude larger than 〈 V2 〉. To obtain more consistent data, ∼65 000 configurations of the (G:C)2 stack were built using systematic screening of the step parameters shift, slide, and twist. We show that undertwisted structures (twist<20°) are of special interest, because the π stack conformations with strong electronic couplings are found for a wide range of slide and shift. Although effective hole transfer can also occur in configurations with twist=30° and 35°, large mutual displacements of neighboring base pairs are required for that. Overtwisted conformation (twist38°) seems to be of limited interest in the context of effective hole transfer. The results may be helpful in the search for DNA based elements for nanoelectronics

In this study we report on the electronic and vibrational (hyper)polarizabilities of donor–acceptorsubstituted azobenzene. It is observed that both electronic and vibrational contributions to the electric dipole first hyperpolarizability of investigated photoactive molecule substantially depend on the conformation. The contributions to the nuclear relaxation first hyperpolarizability are found to be quite important in the case of two considered isomers (cis and trans). Although the double-harmonic term is found to be the largest in terms of magnitude, it is shown that the total value of the nuclear relaxation contribution to vibrational first hyperpolarizability is a result of subtle interplay of higher-order contributions. As a part of the study, we also assess the performance of long-range-corrected density functional theory in determining vibrational contributions to electric dipole (hyper)polarizabilities. In most cases, the applied long-range-corrected exchange correlation potentials amend the drawbacks of their conventional counterparts

Sixty-nine entire male pigs with different halothane genotype (homozygous halothane positive – nn-, n=36; and homozygous halothane negative – NN-, n=33) were fed with a supplementation of magnesium sulphate (Mg) and/or L-tryptophan (Trp) in the diet for 5 days before slaughter. Animals were housed individually and were submitted to stressful ante mortem conditions (mixed in the lorry according to treatments and transported 1h on rough roads). Individual feed intake was recorded during the 5-day treatment. At the abattoir, pig behaviour was assessed in the raceway to the stunning system and during the stunning period by exposure to CO2. Muscle pH, colour, water holding capacity, texture and cathepsin activities were determined to assess meat quality. The number of pigs with an individual feed intake lower than 2kg/day was significantly different among diets (P<0.05; Control: 8.7%; Mg&Trp: 43.5%; Trp: 17.4%) and they were considered to have inadequate supplement intake. During the ante mortem period, 15.2% of pigs included in the experiment died, and this percentage decreased to 8.7% in those pigs with a feed intake > 2kg/day, all of them from the stress-sensitive pigs (nn). In general, no differences were observed in the behaviour of pigs along the corridor leading to the stunning system and inside the CO2 stunning system. During the stunning procedure, Trp diet showed shorter periods of muscular excitation than control and Mg&Trp diets. The combination of a stressful ante mortem treatment and Mg&Trp supplementation led to carcasses with high incidence of severe skin lesions. Different meat quality results were found when considering all pigs or considering only those with adequate supplement intake. In this later case, Trp increased pH45 (6.15) vs Control diet (5.96) in the Longissimus thoracis (LT) muscle (P<0.05) and pH at 24h (Trp: 5.59 vs C: 5.47) led to a higher incidence of dark, firm and dry (DFD) traits in SM muscle (P<0.05). Genotype affected negatively all the meat quality traits. Seventy-five percent of LT and 60.0% of the SM muscles from nn pigs were classified as pale, soft and exudative (PSE), while none of the NN pigs showed these traits (P<0.0001). No significant differences were found between genotypes on the incidence of DFD meat. Due to the negative effects observed in the Mg&Trp group in feed intake and carcass quality, the utilization of a mixture of magnesium sulphate and tryptophan is not recommended

Sixty-one animals with different Halothane genes (homozygous halothane positive, n=34; and homozygous halothane negative, n=27) were fed with three diets (controlgroup, with no supplement; magnesium (Mg) group with 1.28g MgCO3/kg and tryptophan (Trp) group with 5g L-Trp/kg) during the last 5 days before slaughter. Animals were submitted to minimal stress ante mortem conditions. Pig behaviour was recorded at the experimental farm, raceway to the CO2 stunning system and during the stunning period. Corneal reflexes were recorded after stunning as well. There were no differences in feed intake among diets (p>0.05) during the 5 days of treatment. The halothane positive (nn) group had lower intake than the halothane negative (NN) group (p<0.01). The behaviour of the pigs in the raceway did not differ (p>0.05) among treatments or halothane genotype. A significant (p<0.001) interaction diet*halothane was found in the time to appear the first retreat attempt during the exposure to the CO2 system. In the nn group, the time of performing the first retreat attempt was later in the Mg (p<0.05) than the Control group. Moreover, in the Mg group, the nn had a later (p<0.05) first retreat attempt than the NN. Thus, Mg supplementation could have a positive effect on welfare of nn pigs. The nn had a lower proportion of animals that showed corneal reflexes after stunning than NN, indicating a higher effectiveness of the stunning method in nn pigs. Neither Mg nor Trp affected carcass quality and meat quality parameters, although significant differences were found between genotypes

Es presenta el model atòmic a 4.5 Å de DnaB, la principal helicasa replicativa bacteriana, d'Aquifex aeolicus. És un anell hexamèric de 100 Å d'amplada i 80 Å d'alçada amb dues capes de simetria diferenciada, la dels dominis N-terminals en C3 i la dels C-terminals propera a C6. El diàmetre central és de 25 Å al llarg d'ambdues capes, principal diferència amb les estructures prèvies, on era 25 Å més estret a la capa N-terminal. L'estretament s'origina pel trencament d'una de les dues superfícies d'interacció entre monòmers N-terminals, cosa que augmenta la flexibilitat del subdomini implicat. Només l'ssDNA pot atravessar l'anell, quan a les estructures prèvies hi podia passar tant ssDNA com dsDNA. L'estructura aquí presentada és més propera a la conformació funcional de DnaB durant la realització de l'activitat helicasa, mentre que les anteriors correspondrien a la forma inactiva o a la conformació capaç de translocar-se sobre dsDNA.

This PhD thesis is the result of the combination of experimental and computational techniques with the aim of understanding the mechanism of action of de novo cyclic decapeptides with high antimicrobial activity. By experimental techniques the influence of the replacement of the phenylalanine for tryptophan residue in their antimicrobial activity was tested and the stability in human serum was also analyzed, in order to evaluate their potential therapeutic application as antitumor agents. On the other hand, the interaction amongst the peptide BPC194 c(KKLKKFKKLQ), the best candidate from the whole library of cyclic peptides, and a model anionic membrane was simulated. The results showed a structure-function relationship derived from the stable conformation of the peptides involved in the membrane permeabilization. As a result, a rational design was performed being BPC490 the peptide with best antimicrobial activity compared with the best active peptide from the original library.

A conformação pode modular a predisposição para lesões de um cavalo. Por outro lado, o comportamento dos centros de pressão (CdP) dos cascos do cavalo pode fornecer informação relativa ao desempenho mecânico do seu sistema biológico. O objetivo deste trabalho foi testar a correlação dos CdP dos cascos com os principais defeitos de conformação da população estudada. Oito cavalos Puro Sangue Lusitano (PSL) saudáveis, com a cabeça numa posição neutra foram colocados, durante 15 ensaios de 8 segundos, com os membros anteriores (MA) numa placa de pressão e com os membros posteriores (MP) noutra. Os resultados sincronizados foram usados para caracterizar o deslocamento do CdP de cada casco e estudar a sua correlação com a conformação. Os resultados foram analisados de forma qualitativa, através da interpretação de áreas de contato e estabilogramas, assim como de forma quantitativa utilizando estatística inferencial para determinar uma eventual correlação com a conformação, recorrendo a um índice de conformação (IC). O defeito de conformação mais frequente foi os cavalos serem “esquerdos” e o que menos se observou foi serem “fechados de frente”. O IC variou entre 1 e 7. A média das áreas de contato dos cascos dos 8 cavalos foi maior nos MP do que nos MA, tendo acontecido o mesmo para os valores dos deslocamentos do CdP, que foram significativamente maiores nos MP do que nos MA Não foi detetada correlação entre os deslocamentos totais dos CdP e a conformação. Concluíu-se que o CdP dos PSL em estação não está dependente de variações de conformação em que o IC seja igual ou inferior a 7.

Esta tesis realiza un acercamiento a la situación de la cooperación internacional en materia de lavado de activos, en particular en lo que respecta a la relación entre el Ecuador y el Grupo de Acción Financiera Internacional (GAFI), en el periodo 2010-2014. Este periodo es importante por su inclusión en los listados públicos de países no cooperantes en materia de lavado de activos y financiamiento del terrorismo. En el siglo XXI, se ha vuelto recurrente el pretender entender al sistema de relaciones internacionales a través de la teoría constructivista. Sin embargo, el análisis basado en la teoría realista y en las aristas de poder sigue dominando la relación entre los Estados y su funcionamiento en los organismos internacionales. El análisis plasmado en el presente trabajo se ha elaborado desde la visión realista de Keohane, Nye y otros. Se analizan también las contraposiciones entre las visiones de Escudé y Slaughter sobre las teorías de la cooperación Sur-Sur y la comprensión de los principios de gobernanza global y soberanía, respectivamente. La investigación se centra en el estudio de la documentación oficial, no reservada, de las evaluaciones mutuas en materia de lavado de activos y financiamiento del terrorismo, realizadas por los organismos competentes, de las notas de prensa nacional e internacional; con plena conciencia de que por ser un tema altamente político y delicado, muchas cosas no se dicen o se publican con ligereza. El mal manejo y el desconocimiento del entorno GAFI por parte de las autoridades ecuatorianas designadas para llevar las relaciones del país con los organismos internacionales en materia de lavado de activos, dio como resultado que el Ecuador haya ingresado, retornado y permanecido por tres años en la lista de países con deficiencias serias. El Ecuador ha entrado en un nuevo proceso de revisión, pero deberá cumplir con una serie de requisitos, inclusive más rigurosos que los anteriores, si quiere a futuro redimir su situación en el espacio regional y mundial, en materia de lavado de activos y financiamiento del terrorismo.

There is a recent interest to use inorganic-based magnetic nanoparticles as a vehicle to carry biomolecules for various biophysical applications, but direct attachment of the molecules is known to alter their conformation leading to attenuation in activity. In addition, surface immobilization has been limited to monolayer coverage. It is shown that alternate depositions of negatively charged protein molecules, typically bovine serum albumin (BSA) with a positively charged aminocarbohydrate template such as glycol chitosan (GC) on magnetic iron oxide nanoparticle surface as a colloid, are carried out under pH 7.4. Circular dichroism (CD) clearly reveals that the secondary structure of the entrapped BSA sequential depositions in this manner remains totally unaltered which is in sharp contrast to previous attempts. Probing the binding properties of the entrapped BSA using small molecules (Site I and Site II drug compounds) confirms for the first time the full retention of its biological activity as compared with native BSA, which also implies the ready accessibility of the entrapped protein molecules through the porous overlayers. This work clearly suggests a new method to immobilize and store protein molecules beyond monolayer adsorption on a magnetic nanoparticle surface without much structural alteration. This may find applications in magnetic recoverable enzymes or protein delivery.

Electrospinning is a method used to produce nanoscale to microscale sized polymer fibres. In this study we electrospin 1:1 blends of deuterated and hydrogenated atactic-Polystyrene from N,N-Dimethylformamide for small angle neutron scattering experiments in order to analyse the chain conformation in the electrospun fibres. Small angle neutron scattering was carried out on randomly orientated fibre mats obtained using applied voltages of 10kV-15kV and needle tip to collector distances of 20cm and 30cm. Fibre diameters varied from 3mm - 20mm. Neutron scattering data from fibre samples were compared with bulk samples of the same polymer blend. The scattering data indicates that there are pores and nanovoiding present in the fibres; this was confirmed by scanning electron microscopy. A model that combines the scattering from the pores and the labelled polymer chains was used to extract values for the radius of gyration. The radius of gyration in the fibres is found to vary little with the applied voltage, but varies with the initial solution concentration and fibre diameter. The values for the radius of gyration in the fibres are broadly equivalent to that of the bulk state.

Research into transmissible spongiform encephalopathy (TSE) diseases has become a high priority worldwide in recent years yet remarkably little is known about the behaviour of TSE infectivity in the environment. The resilience and stability of prion proteins could lead to soils becoming a potential reservoir of TSE infectivity as a result of contamination from activities such as infected carcass burial or the dispersion of effluents from slaughter houses, or by contamination of pastures by infected animals, (e.g. scrapie in sheep). Knowledge of the fate of prion proteins in soils, and associated physico-chemical conditions which favour migration, can be used to help prevent re-infection of animals through grazing, to protect watercourses and develop good management practices. In two consecutive experiments of 9 and 6 months, the migration of recombinant ovine PrP (recPrP) in soil columns was followed under contrasting levels of microbial activity (normal versus reduced), under varying regimes of soil water content and redox potential, and in two different soil types (loamy sand and clay loam). At each analysis time (1, 3, 6 or 9 months), in both soil types, full-length recPrP was detected in the original contaminated layer, indicating the resilience and stability of recPrP under varied soil conditions, even in the presence of active soil microbial populations. Evidence of protein migration was found in every soil column at the earliest analysis time (1 or 3 months), but was restricted to a maximum distance of 1 cm, indicative of limited initial mobility in soils followed by strong adsorption over the following days to weeks. The survival of recPrP in the soil over a period of at least 9 months was demonstrated. In this study, recPrP was used as an indicator for potential TSE infectivity, although infectivity tests should be carried out before conclusions can be drawn regarding the infection risk posed by prions in soil. However, it has been demonstrated that soil is likely to act as a significant barrier to the dispersion of contaminated material at storage or burial sites. (c) 2007 Elsevier Ltd. The recently determined apucker(xz) = ∂Ixz/∂Qpucker constants for five isotopic species of oxetane have been interpreted quantitatively by allowing the ring-puckering normal coordinate to contain a small amount of methylene rocking and twisting motion. The observed data are most closely reproduced by assuming a methylene rock angle of about 3° and a twist angle for the -methylene groups of about 1° at the equilibrium conformation of the ring.

The low molecular weight glutenin subunits (LMW-GS) are major components of the glutenin polymers which determine the elastomeric properties of wheat (Triticum aestivum L.) gluten and dough. They comprise a complex mixture of components and have proved to be difficult to purify for detailed characterisation. The mature LMW subunit proteins comprise two structural domains, with one domain consisting of repeated sequences based on short peptide motifs. DNA sequences encoding this domain and a whole subunit were expressed in Escherichia coli and the recombinant proteins purified. Detailed comparisons by spectroscopy (CD, FT-IR) and dynamic light scattering indicated that the repetitive and non-repetitive domains of the proteins formed different structures with the former having an extended conformation with an equilibrium between poly-L-proline II-like structure and type II’ b-turns, and the latter a more compact globular structure rich in a-helix. Although the structures of these two domains appear to form independently, dynamic light scattering of the whole subunit dissolved in trifluoroethanol(TFE) suggested that they interact, leading to a more compact conformation. These observations may have relevance to the role of the LMW-GS in gluten structure and functionality.
