Escherichia coli-based bioreporters for arsenic detection are typically based on the natural feedback loop that controls ars operon transcription. Feedback loops are known to show a wide range linear response to the detriment of the overall amplification of the incoming signal. While being a favourable feature in controlling arsenic detoxification for the cell, a feedback loop is not necessarily the most optimal for obtaining highest sensitivity and response in a designed cellular reporter for arsenic detection. Here we systematically explore the effects of uncoupling the topology of arsenic sensing circuitry on the developed reporter signal as a function of arsenite concentration input. A model was developed to describe relative ArsR and GFP levels in feedback and uncoupled circuitry, which was used to explore new ArsR-based synthetic circuits. The expression of arsR was then placed under the control of a series of constitutive promoters, which differed in promoter strength, and which could be further modulated by TetR repression. Expression of the reporter gene was maintained under the ArsR-controlled Pars promoter. ArsR expression in the systems was measured by using ArsR-mCherry fusion proteins. We find that stronger constitutive ArsR production decreases arsenite-dependent EGFP output from Pars and vice versa. This leads to a tunable series of arsenite-dependent EGFP outputs in a variety of systematically characterized circuitries. The higher expression levels and sensitivities of the response curves in the uncoupled circuits may be useful for improving field-test assays using arsenic bioreporters.

A Carnatic music concert is made up of a sequence of pieces, where each piece corresponds to a particular genre and ra¯aga (melody). Unlike a western music concert, the artist may be applauded intra-performance inter-performance. Most Carnatic music that is archived today correspond to a single audio recordings of entire concerts.The purpose of this paper is to segment single audio recordings into a sequence of pieces using thecharacteristic features of applause and music. Spectral flux, spectral entropy change quite significantly from music to applause and vice-versa. The characteristics of these features for a subset of concerts was studied. A threshold based approach was used to segment the pieces into music fragments and applauses. Preliminary resultson recordings 19 concerts from matched microphones show that the EER is about 17% for a resolution of 0.25 seconds. Further, a parameter called CUSUM is estimatedfor the applause regions. The CUSUM values determine the strength of the applause. The CUSUM is used to characterise the highlights of a concert.

MicroRNAs are important regulators of gene expression. The vast majority of the cells in our body rely on hundreds of these tiny non-coding RNA molecules to precisely adjust their protein repertoire and faithfully accomplish their tasks. Indeed, alterations in the microRNA profile can lead to cellular dysfunction that favours the appearance of several diseases. A specific set of microRNAs plays a crucial role in pancreatic beta cell differentiation and is essential for the fine-tuning of insulin secretion and for compensatory beta cell mass expansion in response to insulin resistance. Recently, several independent studies reported alterations in microRNA levels in the islets of animal models of diabetes and in islets isolated from diabetic patients. Surprisingly, many of the changes in microRNA expression observed in animal models of diabetes were not detected in the islets of diabetic patients and vice versa. These findings are unlikely to merely reflect species differences because microRNAs are highly conserved in mammals. These puzzling results are most probably explained by fundamental differences in the experimental approaches which selectively highlight the microRNAs directly contributing to diabetes development, the microRNAs predisposing individuals to the disease or the microRNAs displaying expression changes subsequent to the development of diabetes. In this review we will highlight the suitability of the different models for addressing each of these questions and propose future strategies that should allow us to obtain a better understanding of the contribution of microRNAs to the development of diabetes mellitus in humans.

Objective: Microalbuminuria (MAU) is a marker of early kidney injury and cardiovascular risk. We assessed the association of MAU with plasma adiponectin, leptin, and hsCRP as inflammatory marker, accounting for hypertension, diabetes and obesity. Design and Methods: Population based, cross-sectional study in Caucasian subjects aged 35 to 75 years in Lausanne, Switzerland. MAU, measured by quantitative immunonephelometry on spot morning urine, was used either as a continuous (MAU) or dichotomized variable (MA defined as MAU > 2.5 and >3.5 mg/mmol creatinine in men and women, respectively). Results: The 2955 women (age 53.3_10.7, mean_SD years) had mean body mass index (BMI) 24.9_4.5 kg/m. The 2479 men (age 53.1_10.8 years) hadmean BMI 27.0_3.9 kg/m2.Median hsCRP was 1.3 and 1.3 mg/L, median adiponectin 6.2 and 10.6mg/mL in men and women, respectively. MA prevalence was 4.9% in women and 9.8% in men. In multivariate regression analysis adjusting for potential confounders (age, sex, hypertension, diabetes, eGFR, BMI, percent fat mass, insulin and smoking), logtransformed MAU was positively associated with hsCRP (P<0.001) and adiponectin (P¼0.002), but not with leptin. The association of adiponectin with MAU was stronger in subjects with low hsCRP, and vice versa (P interaction<0.001). Conclusion: Adiponectin and hsCRP are significant positive determinants of MAU, independently of diabetes, hypertension and fat mass. A negative interaction between hsCRP and adiponectin was found for their effect on MAU. Whether hyperadiponectinemia represents an adequate protective response to vascular stress or has negative causal impact on the development of MAU should be assessed in further studies.

This paper analyzes the behavior of international capital flows by foreigners and domestic agents, especially during financial crises. We show that gross capital flows by foreigners and domestic agents are very large and volatile, especially relative to net capital flows. This is because when foreigners invest in a country domestic agents tend to invest abroad and vice versa. Gross capital flows are also pro-cyclical. During expansions, foreigners tend to bring in more capital and domestic agents tend to invest more abroad. During crises, there is retrenchment, i.e. a reduction in capital inflows by foreigners and an increase in capital inflows by domestic agents. This is especially true during severe crises and during systemic crises. The evidence can shed light on the nature of shocks driving international capital flows. It seems to favor shocks that affect foreigners and domestic agents asymmetrically -e.g. sovereign risk and asymmetric information- over productivity shocks.

OBJECTIVES: (1) To evaluate the changes in surface roughness and gloss after simulated toothbrushing of 9 composite materials and 2 ceramic materials in relation to brushing time and load in vitro; (2) to assess the relationship between surface gloss and surface roughness. METHODS: Eight flat specimens of composite materials (microfilled: Adoro, Filtek Supreme, Heliomolar; microhybrid: Four Seasons, Tetric EvoCeram; hybrid: Compoglass F, Targis, Tetric Ceram; macrohybrid: Grandio), two ceramic materials (IPS d.SIGN and IPS Empress polished) were fabricated according to the manufacturer's instructions and optimally polished with up to 4000 grit SiC. The specimens were subjected to a toothbrushing (TB) simulation device (Willytec) with rotating movements, toothpaste slurry and at three different loads (100g/250g/350g). At hourly intervals from 1h to 10h TB, mean surface roughness Ra was measured with an optical sensor and the surface gloss (Gl) with a glossmeter. Statistical analysis was performed for log-transformed Ra data applying two-way ANOVA to evaluate the interaction between load and material and load and brushing time. RESULTS: There was a significant interaction between material and load as well as between load and brushing time (p<0.0001). The microhybrid and hybrid materials demonstrated more surface deterioration with higher loads, whereas with the microfilled resins Heliomolar and Adoro it was vice versa. For ceramic materials, no or little deterioration was observed over time and independent of the load. The ceramic materials and 3 of the composite materials (roughness) showed no further deterioration after 5h of toothbrushing. Mean surface gloss was the parameter which discriminated best between the materials, followed by mean surface roughness Ra. There was a strong correlation between surface gloss and surface roughness for all the materials except the ceramics. The evaluation of the deterioration curves of individual specimens revealed a more or less synchronous course suspecting hinting specific external conditions and not showing the true variability in relation to the tested material. SIGNIFICANCE: The surface roughness and gloss of dental materials changes with brushing time and load and thus results in different material rankings. Apart from Grandio, the hybrid composite resins were more prone to surface changes than microfilled composites. The deterioration potential of a composite material can be quickly assessed by measuring surface gloss. For this purpose, a brushing time of 10h (=72,000 strokes) is needed. In further comparative studies, specimens of different materials should be tested in one series to estimate the true variability.

A Cultura pode ser definida de várias maneiras e cada autor tem uma visão própria do termo mas, genericamente, cultura pode ser entendida como o conjunto de características humanas que não são inatas e que se criam, preservam ou aprimoram com o convívio na sociedade, com a comunicação e cooperação entre indivíduos. Os membros de uma sociedade não estão conscientes dos seus valores culturais e somente percebem estes valores quando entram em “choque” com uma cultura diferente. Estas diferenças culturais podem criar confusões e mal-entendidos. As culturas Portuguesa e a Brasileira parecem ser bastante semelhantes: a mesma língua, as mesmas crenças, religião predominantemente católica e alimentos similares. Porém, apesar das semelhanças, e após entrevistas de carácter narrativo, observou-se nestas duas culturas características próprias e que muitas vezes foram necessárias adaptações para uma melhor integração na sociedade. Esta tese de mestrado é uma análise das diferenças culturais entre Portugal e Brasil e tem por objectivo encontrar standards culturais de ambos os países podendo ser um instrumento de ajuda para aqueles que vêm do Brasil para Portugal, e vice-versa, para trabalharem ou abrirem uma empresa. Os standards culturais encontrados, após análise comparativa, estão relacionados com a maneira de ver a vida, de ultrapassar os problemas e pressões do dia a dia, com colectivismo, hierarquia, simpatia, preocupação com o cliente, ansiedade face à incerteza e profissionalismo. Este trabalho termina com a identificação dos standards culturais e com dicas para aqueles que saem do seu país e recomeçam a sua vida noutro com cultura diferente

La thèse propose un regard en profondeur sur les fondements de la connaissance en phonologie structurale. Elle s articule autour de cinq objectifs. Tout d'abord, un cadre précis de discussion métathéorique en phonologie est développé, admis qu'il n'existe tout simplement pas de méthode du genre permettant une critique systématique du contenu de programmes de recherche différents dans ce domaine. Cette méthodologie est ensuite appliquée à la phonologie fonctionnelle, celle issue de renseignement de Nikolaï S. Troubetz- koy et du fonctionnalisme d'André Martinet. De plus, elle est appliquée à un second programme de recherche, en marge de la réflexion centrale. Il s'agit de la phonologie générative et, plus spécifiquement, d'une théorie contemporaine à l'intérieur de celle-ci, la théorie de loptimalité, dont le caractère représentatif du cadre général est également abordé. Cet exercice comparatif permet de dégager les relations entre les contenus respectifs des deux programmes à l'étude pour, enfin, évaluer le statut épistémologique de la phonologie fonctionnelle.Il s'avère que la connaissance de la langue issue de la phonologie fonctionnelle se distingue clairement du domaine de la phonologie générative dégagé à travers l'étude de la théorie de loptimalité (et vice versa). Le partage des mêmes fondements phonétiques, d'une terminologie parfois trompeusement homogène et de la notion d'explication fonctionnelle masque la profonde opposition de valeur épistémologique accordée à des prémisses acceptées d'emblée par tous (l'opposition significative, par exemple, ou la réalité psychologique).En somme, ce travail remet en question une vision cumulative et rectiligne de l'histoire de la phonologie par l'épistémologie. Porter un regard critique sur la motivation des théories et sur les choix terminologiques permet de dégager les fondements de deux traditions de pensée qui ne se chevauchent pas là où celles-ci se définissent respectivement en tant que phonologie. Ainsi, du point de vue de la connaissance, la phonologie américaine n'inclut jamais ni ne peut contredire le fonctionnalisme européen, qui dès lors représente un courant parallèle plutôt que primitif.

I develop an overlapping-generations framework in which changes in lending standards generateendogenous cycles. In my economy, entrepreneurs who are privately informed about thequality of their projects need to borrow funds. Intermediaries screen entrepreneurs both throughthe amount of investment undertaken and through the level of entrepreneurial net worth.I show that endogenous regime switches in financial contracts from pooling to separatingand vice-versa may generate fluctuations even in the absence of exogenous shocks. Whenthe economy is in the pooling (separating) regime, lending standards seem lax ( tight ) andinvestment is high (low). Differently from the existing literature, my model does not requireentrepreneurial net worth to be counter cyclycal or inconsequential for determining aggregateinvestment.

A assinatura digital é um processo de assinatura electrónica baseado no sistema criptográfico assimétrico composto por um algoritmo ou série de algoritmos, mediante o qual é gerado um par de chaves assimétricas exclusivas e complementares. Tal como a assinatura manuscrita, a assinatura digital não pode ser falsificada, pelo que se adiciona uma função hash ao valor da chave gerada, para garantir a segurança do sistema. A assinatura digital permite, ao mesmo tempo, a identificação positiva do autor de uma mensagem (ou do signatário de um documento) e a verificação da integridade da mesma. Refira-se, porém, que a assinatura digital pode ser anexa a qualquer mensagem, seja esta decifrada ou não, apenas para que o receptor tenha a certeza da identidade do emissor e de que a mensagem chegou intacta ao destino. Baseia-se na criptografia da chave pública, que usa um algoritmo de duas chaves, a privada e a pública, diferentes mas matematicamente associadas: a primeira cria a assinatura digital e decifra os dados; a segunda verifica a assinatura e devolve-a ao formato original. O autor da mensagem mantém secreta a sua chave privada e divulga a chave pública, e vice-versa. A identidade do dono da chave pública é atestada pelo certificado digital emitido por uma entidade certificadora (uma espécie de notário). Independentemente da forma que assume, a assinatura electrónica, e a digital em particular, deve cumprir três funções: a autenticação (tal como acontece com o habitual gatafunho a que chamamos assinatura, apenas o indivíduo deve ser capaz de a reproduzir); não repudiação (quando o indivíduo assina não pode voltar atrás); e integridade (é necessário assegurar que os documentos assinados não são alterados sem o consentimento dos signatários).

Desde, pelo menos, os finais da década de 60, que por todo o Planeta se verifica um agravamento progressivo dos problemas ambientais. Muitos são os motivos apontados para este facto, mas na sua base encontra-se o comportamento do quotidiano do homem que, desprovidos de uma educação firme e consciente, sem uma cultura de utilização adequada, conservação e reutilização, não faz à correcta gestão dos recursos naturais, abriu um caminho para uma crise quase irreversível do ambiente. Quase irreversível porque ainda estamos a tempo de ‘reciclar’ os nossos comportamentos e aplicá-los às necessidades de conservação da qualidade ambiental, sem a qual o futuro do homem entra no princípio do terceiro instruído (incerteza), sem viabilidade possível. Nestes termos acreditamos que, uma educação apropriada poderemos promover uma Cultura Ambiental e mudar o rumo dos acontecimentos se for globalmente pensada (associacionismo) e localmente praticada por todos que fazem parte de uma comunidade (associativismo). Essa educação terá como objectivo o desenvolvimento humano de modo a torná-lo consciente de que o mundo é uma só ‘casa’ e que os problemas ambientais é uma só ‘causa’. Não interessa os protagonistas, mas sim, que todos envoltam para a aquisição de conhecimentos, capacidades práticas, atitudes, motivações e compromissos que sejam necessários para a luta contra a pobreza (seja ela, material, espiritual ou intelectual), oferecendo assim, soluções para os problemas existentes e prevenir as gerações vindouras. Reconhecemos pois, o grande papel que os movimentos associativos, a OSC e as ONG’s poderão desempenhar junto das comunidades (ou localidades) para a formação, divulgação, sensibilização (…) e combater os problemas ambientais localizadas. Estas acções educativas enformam aquilo que designamos por «Educação Ambiental para o Desenvolvimento Sustentável» (EADS), aplicada a realidade do Concelho de Santa Cruz, e serve como pano de fundo desta dissertação. Assim, todas as estratégias pedagógicas e metodológicas partirão de práticas vividas nos meio associativos e conduzirão à criação de espíritos adequados ao compromisso na solução dos problemas encontrados.

This paper analyzes the behavior of international capital flows by foreign and domestic agents,dubbed gross capital flows, over the business cycle and during financial crises. We show thatgross capital flows are very large and volatile, especially relative to net capital flows. Whenforeigners invest in a country, domestic agents invest abroad, and vice versa. Gross capital flowsare also pro-cyclical. During expansions, foreigners invest more domestically and domesticagents invest more abroad. During crises, total gross flows collapse and there is a retrenchmentin both inflows by foreigners and outflows by domestic agents. These patterns hold for differenttypes of capital flows and crises. This evidence sheds light on the sources of fluctuations drivingcapital flows and helps discriminate among existing theories. Our findings seem consistent withcrises affecting domestic and foreign agents asymmetrically, as would be the case under thepresence of sovereign risk or asymmetric information.

This paper reports an analysis of income related health inequalities at the AutonomousCommunity level in Spain using the self assessed health measure in the 2001 edition of theEncuesta Nacional de Salud. We use recently developed methods in order to cardinalise andmodel self assessed health within a regression framework, decompose the sources ofinequality and explain the observed differences across regions. We find that the regions with the highest levels of mean health tend to enjoy the lowest degrees of income related health inequality and vice-versa. The main feature characterizing regions where income related health inequality is low is the absence of a positive gradient between income and health. In turn, the regions where income related health inequality is greater are characterized by a strong and significant positive gradient between health and income. These results suggest that policies aimed at eliminating the gradient between health and income can potentially lead to greate r reductions in socio-economic health inequalities than policies aimed at redistributing income.

A acessibilidade é um tema que, apesar de ser discutido desde décadas retrógradas, foi retomado recentemente como um assunto de extrema importância para o planeamento urbano, com o intuito de promover uma boa qualidade de vida aos cidadãos. É impossível falar de acessibilidade sem a ergonomia, ou vice-versa, pois um conceito remete ao outro. A acessibilidade cumpre os espaços, os mobiliários e os equipamentos urbanos, as edificações, os transportes e os meios de comunicação, que podem ser utilizados com segurança e autonomia por toda e qualquer pessoa, tenha ela deficiência ou não ou mobilidade reduzida. Por sua vez, ergonomia é o estudo da adaptação do trabalho ao trabalhador, promovendo-lhe o bem-estar e reduzindo ou evitando eventuais acidentes. Este trabalho tem como objectivo descobrir se existe acessibilidade para as pessoas com deficiência na UniPiaget e identificar as barreiras físicas. Num primeiro momento do estudo prático utilizou-se um questionário com catorze perguntas fechadas e uma aberta, em que fizeram parte da amostra 6 alunos com deficiência da UniPiaget, sendo este o seu universo na dita universidade. No seu segundo momento, foi feito o levantamento das medidas físicas, das portas, dos sanitários, dos balcões de atendimento, das rampas, das escadas e do estacionamento com o intuito de identificar as barreiras físicas.Chegou-se a conclusão de que a UniPiaget da Praia ainda não está totalmente adaptada para receber pessoas com deficiência.
