As células tronco são caracterizadas pela sua capacidade de se diferenciar em várias linhagens de células e exibir um pontente efeito parácrino. O objetivo deste trabalho foi avaliar o efeito da terapia com células da medula óssea (BMCs) na glicose sanguínea, no metabolismo lipídico e remodelamento da parede da aorta em um modelo experimental para aterosclerose. Camundongos C57BL/6 foram alimentados com uma dieta controle (grupo CO) ou uma dieta aterogênica (grupo AT - 60% gordura). Após 16 semanas, o grupo AT foi dividido em quatro sub grupos: grupo AT 14 dias e o grupo AT 21 dias receberam uma injeção de PBS na veia caudal e mortos 14 e 21 dias após respectivamente; grupo AT-BMC 14 dias e AT-BMC 21 dias que receberam uma injeção com BMCs na veia caudal e mortos 14 e 21 dias após, respectivamente. O grupo CO foi sacrificado juntamente com outros grupos. O transplante BMCs reduziu os niveis de glicose, triglicerídeos e colesterol total no sangue. Não houve diferença significativa em relação à massa corporal entre os grupos transplantados e não transplantados, sendo todos diferentes do grupo CO. Não houve diferença significativa na curva glicemica entre os grupos AT 14 dias, AT-BMC 14 dias e AT 21 dias e estes diferentes do grupo CO e do grupo AT-BMC 21 dias. O Qa (1/mm2) foi quantitativamente reduzido no grupo AT 14 dias e AT 21 dias quando comparado ao grupo CO. Este Qa se mostrou elevado no grupo AT-BMC 21 dias quando comparado a todos os grupos. O aumento da expessura da parede da aorta foi observado em todos os grupos aterogênicos, entretanto o aumento da espessura foi significativamente menor no grupo AT-BMC 21 dias em relação ao grupo AT 14 dias e AT 21 dias. A percentagem de fibras elásticas se apresentou significativamente maior no grupo AT 21 dias quando comparado ao CO e AT-BMC 21 dias. Não houve diferença significativa entre o grupo CO e AT-BMC 21 dias. Vacúolos na túnica média, delaminação e o adelgaçamento das lamelas elásticas foram observados nos grupos AT-14 dias e AT-21 dias. O menor número destes foi visualizado no grupo AT-BMC 14 dias e AT-BMC 21 dias. A imunomarcação para alfa actina de músculo liso (α-SMA) e fator de crescimento vascular e endotelial (VEGF) mostrou menor marcação em grupos transplantados com BMCs. A marcação para antígeno nuclear de proliferação celular (PCNA) mostrou-se mais expressiva no grupo AT-BMC 21 dias grupo. Marcação para CD105, CD133 e CD68 foi observada nos grupos AT 14 dias e AT 21 dias. Estas marcações não foram observadas nos grupos AT-BMC 14 dias e AT-BMC 21 dias. Nas eletromicrografias observamos o remodelamento benéfico no grupo AT-BMC14 dias e AT-BMC 21 dias, com a organização estrutural similar ao grupo CO. Vesículas de pinocitose, projeção da célula muscular lisa e a delaminação da lamina elástica interna são observados nos grupos AT 14 dias e AT 21 dias. Célula endotelial preservada, com lamina elástica interna de contorno regular e contínua é observada no grupo CO e nos grupos AT-BMC 14 dias e AT-BMC 21 dias. Como conclusão, os nossos resultados reforçam o conceito de que, em um modelo aterosclerótico utilizando camundongos e dieta aterogênica, a injeção de BMCs melhora os níveis de glicose, metabolismo lipídico e ocasiona um remodelamento benéfico na parede da aorta.

A frequência cardíaca (FC) no repouso (FC Rep), no exercício máximo (FC Max) e após o exercício (FC Rec) traz importantes informações para a saúde, e parte dessas respostas é modulada pelo tônus vagal cardíaco (TVC), que também oferece proteção cardíaca. Para uma melhor compreensão dos aspectos prognósticos da FC e de suas interações com o TVC, foram realizados três estudos: dois retrospectivos e um ensaio clínico randomizado. O primeiro testou se o TVC, estimado utilizando o índice vagal cardíaco (IVC), contribui para a FC Max (% do previsto: 208-0,7 x idade (anos)) em 1000 indivíduos saudáveis (39 14 anos; 719 homens). Regressão linear identificou que TVC explica apenas 1% da variabilidade da FC Max (% do previsto), com erro padrão da estimativa alto (~ 6,3%), indicando potencial papel complementar clínico para essas duas variáveis relacionadas ao exercício. O segundo estudo verificou se a análise de mortalidade utilizando FC de reserva (FC Res) e FC Rec de forma combinada descriminaria melhor a mortalidade que a análise de um destes itens e forma isolada. Dados de FC Res e FC Rec de 1.476 indivíduos (41 a 79 anos, 937 homens) foram calculados e divididos em quintis, os quais somados forneceram categorias de 2 a 10, produzindo um gradiente da FC (FC Grad) e refletindo a magnitude dos transientes iniciais e finais do exercício máximo. Análises de sobrevida foram realizadas usando os quintis mais baixos (Q1) dos escores do Grad FC, FC Res e FC Rec. Em um seguimento médio de 7,3 anos, 44 participantes morreram (3,1%). Houve uma tendência inversa entre os escores do Grad FC e a taxa de mortalidade (p<0,05), que passou de 1,2% para 13,5%, respectivamente, para os escores 10 e 2. Uma pontuação no Grad FC de 2 foi melhor preditor de mortalidade por todas as causas, quando comparado ao Q1 da FC Res e da FC Rec, com riscos relativos ajustados pela idade de 3,53 (p=0,01); 2,52 (p<0,05) e 2,57(p<0,05), respectivamente. Conclusão: Grad FC é um preditor de risco de mortalidade por todas as causas com desempenho superior ao das medidas isoladas de FC Res e FC Rec. Por último verificou-se a hipótese do aumento do TVC em participantes de um programa de exercício supervisionado (PES) com IVC baixo (≤ 1,30), através de um treinamento específico, utilizando a transição repouso-exercício no protocolo denominado treinamento vagal (TV). Estes foram randomizados num delineamento cruzado (duas etapas de oito semanas), com ou sem três sessões semanais de TV. Houve discreta melhora no IVC em 16 semanas (1,19 vs 1,22; p=0,02) dos 44 pacientes (64% homens; 65,5 11,4 anos) que finalizaram o estudo, mas não se pôde afirmar que a diferença no IVC se deveu ao período em que foi realizado o TV (p=0,36). Portanto, 16 semanas de PES, incluindo oito semanas com TV, aumentam a resposta vagal à transição repouso-exercício, embora não tenha sido possível atribuir os resultados exclusivamente ao TV. Esta dissertação de mestrado realizou um estudo sobre a identificação de memórias e representações sociais das práticas religiosas de matriz africana na população negra do bairro Dom Bosco situado no município de Juiz de Fora - MG. Foram entrevistados 60 sujeitos, de ambos os sexos, que se auto identificavam como negros, pardos, mulatos e outras denominações que caracterizam a ascendência negra. Através dos dados levantados na pesquisa concluímos que as representações sociais que esta população possui, são de que as práticas de matriz africana são demoníacas, feitiçarias para o mal e ações que causam prejuízo para as pessoas. As lembranças se mesclam com esquecimento, e as representações e memórias dos sujeitos da pesquisa estão associados com as práticas de sincretismo religioso existentes no Brasil. Os entrevistados não possuem lembrança dos líderes religiosos negros e nem dos locais de memória do bairro. Os participantes da pesquisa não se associam com estas práticas religiosas e têm em relação a elas uma visão de distanciamento e desinteresse. Constatamos que isto ocorre devido à dificuldade de aceitação pela sociedade em geral que as desvaloriza e discrimina, sempre atribuindo negatividade a elas e aos praticantes. Estas práticas são estereotipadas, folclorizadas e menosprezadas socialmente. O que faz com que a população afrodescendente não queira ser identificada com as tradições históricas, culturais e religiosos dos seus ancestrais

Biomass estimates of several species of Alaskan rockfishes exhibit large interannual variations. Because rockfishes are long lived and relatively slow growing, large, short-term shifts in population abundance are not likely. We attribute the variations in biomass estimates to the high variability in the spatial distribution of rockfishes that is not well accounted for by the survey design currently used. We evaluated the performance of an experimental survey design, the Trawl and Acoustic Presence/Absence Survey (TAPAS), to reduce the variability in estimated biomass for Pacific ocean perch (Sebastes alutus). Analysis of archived acoustic backscatter data produced an acoustic threshold for delineating potential areas of high (“patch”) and low (“background”) catch per unit of effort (CPUE) in real time. In 2009, we conducted a 12-day TAPAS near Yakutat, Alaska. We completed 59 trawls at 19 patch stations and 40 background stations. The design performed well logistically, and Pacific ocean perch (POP) accounted for 55% of the 31 metric tons (t) of the catch from this survey. The resulting estimates of rockfish biomass were slightly less precise than estimates from simple random sampling. This difference in precision was due to the weak relationship of CPUE to mean volume backscattering and the relatively low variability of POP CPUE encountered. When the data were re-analyzed with a higher acoustic threshold than the one used in the field study, performance was slightly better with this revised design than with the original field design. The TAPAS design could be made more effective by establishing a stronger link between acoustic backscatter and CPUE and by deriving an acoustic threshold that allows better identification of backscatter as that from the target species.

The trajectory of the somatic membrane potential of a cortical neuron exactly reflects the computations performed on its afferent inputs. However, the spikes of such a neuron are a very low-dimensional and discrete projection of this continually evolving signal. We explored the possibility that the neuron's efferent synapses perform the critical computational step of estimating the membrane potential trajectory from the spikes. We found that short-term changes in synaptic efficacy can be interpreted as implementing an optimal estimator of this trajectory. Short-term depression arose when presynaptic spiking was sufficiently intense as to reduce the uncertainty associated with the estimate; short-term facilitation reflected structural features of the statistics of the presynaptic neuron such as up and down states. Our analysis provides a unifying account of a powerful, but puzzling, form of plasticity.

Band-edge liquid crystal lasers are of interest for a number of applications including laser projection displays. Herein, we demonstrate simultaneous red-green-blue lasing from a single liquid crystal sample by creating a two-dimensional laser array fabricated from dye-doped chiral nematic liquid crystals. By forming a pitch gradient across the cell, and optically pumping the sample using a lenslet array, a polychromatic laser array can be observed consisting simultaneously of red-green-blue colors. Specifically, the two-dimensional polychromatic array could be used to produce a laser-based display, with low speckle and wide color gamut, whereby no complex fabrication procedure is required to generate the individual 'pixels'.

Many aspects of human motor behavior can be understood using optimality principles such as optimal feedback control. However, these proposed optimal control models are risk-neutral; that is, they are indifferent to the variability of the movement cost. Here, we propose the use of a risk-sensitive optimal controller that incorporates movement cost variance either as an added cost (risk-averse controller) or as an added value (risk-seeking controller) to model human motor behavior in the face of uncertainty. We use a sensorimotor task to test the hypothesis that subjects are risk-sensitive. Subjects controlled a virtual ball undergoing Brownian motion towards a target. Subjects were required to minimize an explicit cost, in points, that was a combination of the final positional error of the ball and the integrated control cost. By testing subjects on different levels of Brownian motion noise and relative weighting of the position and control cost, we could distinguish between risk-sensitive and risk-neutral control. We show that subjects change their movement strategy pessimistically in the face of increased uncertainty in accord with the predictions of a risk-averse optimal controller. Our results suggest that risk-sensitivity is a fundamental attribute that needs to be incorporated into optimal feedback control models.

Computer Aided Control Engineering involves three parallel streams: Simulation and modelling, Control system design (off-line), and Controller implementation. In industry the bottleneck problem has always been modelling, and this remains the case - that is where control (and other) engineers put most of their technical effort. Although great advances in software tools have been made, the cost of modelling remains very high - too high for some sectors. Object-oriented modelling, enabling truly re-usable models, seems to be the key enabling technology here. Software tools to support control systems design have two aspects to them: aiding and managing the work-flow in particular projects (whether of a single engineer or of a team), and provision of numerical algorithms to support control-theoretic and systems-theoretic analysis and design. The numerical problems associated with linear systems have been largely overcome, so that most problems can be tackled routinely without difficulty - though problems remain with (some) systems of extremely large dimensions. Recent emphasis on control of hybrid and/or constrained systems is leading to the emerging importance of geometric algorithms (ellipsoidal approximation, polytope projection, etc). Constantly increasing computational power is leading to renewed interest in design by optimisation, an example of which is MPC. The explosion of embedded control systems has highlighted the importance of autocode generation, directly from modelling/simulation products to target processors. This is the 'new kid on the block', and again much of the focus of commercial tools is on this part of the control engineer's job. Here the control engineer can no longer ignore computer science (at least, for the time being). © 2006 IEEE.

CMOS nanocrystalline silicon thin film transistors with high field effect mobility are reported. The transistors were directly deposited by radio-frequency plasma enhanced chemical vapor deposition at 150°C The transistors show maximum field effect mobility of 450 cm2/V-s for electrons and 100 cm2/V-s for holes at room temperature. We attribute the high mobilities to a reduction of the oxygen content, which acts as an accidental donor. Indeed, secondary ion mass spectrometry measurements show that the impurity concentration in the nanocrystalline Si layer is comparable to, or lower than, the defect density in the material, which is already low thanks to hydrogen passivation.

Polarization-insensitivity is achieved in a reflective spatial light modulator by laying a quarter-wave plate (QWP) at the incident wavelength directly over the mirror pixels of a silicon backplane, and forming a nematle Fréedrickcz cell over the QWP to modulate the reflected phase. To achieve the highest drive voltage from the available silicon process, a switched voltage common front electrode design is described, with variable amplitude square wave drive to the pixels to maintain constant root-mean-square drive and minimize phase fluctuations during the dc balance refresh cycle. The silicon has been fabricated and liquid-crystal-on-silicon cells both with and without the QWP assembled; applications include optically transparent switches for optical networks, beam steering for add-drop multiplexers for wavelength-division- multiplexing telecommunications, television multicast, and holographic projection.

Horseflies are economically important blood-feeding arthropods and also a nuisance for humans, and vectors for filariasis. They rely heavily on the pharmacological propriety of their saliva to get blood meat and suppress immune reactions of hosts. Little information is available on horsefly immune suppressants. By high-performance liquid chromatography (HPLC) purification coupling with pharmacological testing, an immunoregulatory peptide named immunoregulin HA has been identified and characterized from salivary glands of the horsefly of Hybomitra atriperoides (Diptera, Tabanidae). Immunoregulin HA could inhibit the secretion of interferon-gamma (IFN-gamma) and monocyte chemoattractant protein (MCP-1) and increase the secretion of interteukin-10 (IL-10) induced by lipopolysaccharide (LIPS) in rat splenocytes. IL-10 is a suppressor cytokine of T-cell proliferative and cytokine responses. IL-10 can inhibit the elaboration of pro-inflammatory cytokines. Immunoregulin HA possibly unregulated the IL-10 production to inhibit IFN-gamma and MCP-1 secretion in the current experiments. This immunosuppression may facilitate the blood feeding of this horsefly. The current works will facilitate to understand the molecular mechanisms of the ectoparasite-host relationship. 2008 Elsevier Ltd. All rights reserved.

Horseflies are economically important blood-feeding arthropods and also a nuisance for humans and vectors for filariasis. They rely heavily on the pharmacological properties of their saliva to get a blood meal and suppress immune reactions of hosts. Little information is available on antihemostatic substances in horsefly salivary glands; especially no horsefly immune suppressants have been reported. By proteomics or peptidomics and coupling transcriptome analysis with pharmacological testing, several families of proteins or peptides, which act mainly on the hemostatic system or immune system of the host, were identified and characterized from 30,000 pairs salivary glands of the horsefly Tabanus yao (Diptera, Tabanidae). They are: (i) a novel family of inhibitors of platelet aggregation including two members, which possibly inhibit platelet aggregation by a novel mechanism and act on platelet membrane, (ii) a novel family of immunosuppressant peptides including 12 members, which can inhibit interferon-gamma production and increase interleukin-10 secretion, (iii) a serine protease inhibitor with 56 amino acid residues containing anticoagulant activity, (iv) a serine protease with anticoagulant activity, (v) a protease with fibrinogenolytic activity, (vi) three families of antimicrobial peptides including six members, (vii) a hyaluronidase, (viii) a vasodilator peptide, which is an isoform of vasotab identified from Hybomitra bimaculata, and interestingly (ix) two metallothioneins, which are the first metallothioneins reported from invertebrate salivary glands. The current work will facilitate the understanding of the molecular mechanisms of the ectoparasite-host relationship and help in identifying novel vaccine targets and novel leading pharmacological compounds.

Lobster analogues were prepared with lobster base flavour (paste),h lobster cook water (whole lobster omogenate:water, 1:1 and 1:0.5) and lobster meat mince. In another experiment, different combinations of ginger-garlic paste and lobster base flavour, i.e., 1:3, 1:4, 1:6, 3:3, 3:4, 3:6, 5:3, 5:4 and 5:6 were added to the lobster analogue paste. It was observed that lobster analogues prepared with lobster base flavour (paste) are suitable organoleptically. The combination of ginger garlic paste and lobster base flavour in the ratio of 3:4 was found to be suitable organoleptically. Lobster analogues coloured with annatto seed colour at 1:2 (annatto seed:water) concentration had high values for the colour attribute as compared to orange-red synthetic colour, beetroot colour, caramel colour and paprika colour. It was observed that come-up-time to achieve a temperature of850°C was 28 minutes with a processing period of 11 minutes.
