The objective of this study was to determine the effects of dietary energy and recombinant bovine somatotropin (bST) injection to identify genes that might control mammogenesis. Total RNA was extracted from the parenchymal tissue of 32 heifers randomly assigned to one of four treatments: two diets (a standard diet and a high energy, high protein diet), each with or without bST. To perform microarray experiments, RNA samples were pooled (2 animals/pool) before reverse transcription and labeling with Cy3 or Cy5. A 4-node loop design was used to examine the differential gene expression among treatments using a bovine-specific cDNA micro array (National Bovine Functional Genomics Consortium Library, NBFGC) containing 18,263 unique expressed sequence tags (EST). Significance levels of differential gene expression among treatments were assessed using a mixed model approach. Injection of bST altered the expression of 12 % of the genes on NBFGC slide related to tissue development, whereas 6% were altered by diet. Administration of bST increases the expression of genes positively related to cell proliferation and mammary parenchyma to a greater extent than a high energy diet. © 2013 Sociedade Brasileira de Zootecnia.

Fatores de transcrição desempenham importantes funções em vários processos fisiológicos. Nos últimos anos, muitos fatores de transcrição têm sido isolados de plantas, emergindo como poderosas ferramentas na manipulação de características agronômicas. No presente trabalho, iniciamos estudos para isolar fatores de transcrição de mandioca (Manihot esculenta Crantz), importante cultura tropical e subtropical. Nossos resultados revelaram três tipos de proteínas diferencialmente expressas na raiz de reserva de mandioca (Manihot esculenta Crantz):e imunologicamente relacionadas com o fator de transcrição opaco-2 de milho. Experimentos de Southwestern mostraram duas proteínas capazes de interagir in vitro com uma seqüência de DNA do gene be2S1 de castanha-do-brasil.

O câncer gástrico continua sendo uma importante causa de morte dentre os tipos de câncer no Brasil e no mundo. A origem do câncer de estomago provém, assim como nos demais, de acúmulo de alterações genéticas. Portanto, se faz necessário saber quais alterações genéticas são importantes para desencadear a patogênese do câncer gástrico. O MNU, conhecido carcinógeno, quando ingerido de forma oral em doses determinadas desencadeia o desenvolvimento de adenocarcinoma gástrico do tipo intestinal, com aparecimento de estágios pré definidos característico deste tipo tumoral. Com base nestes conhecimentos, realizamos um experimento com 6 macacos da espécie Cebus apella induzidos à desenvolver câncer gástrico do tipo intestinal. Os animais ingeriam 16mg/kg de peso diariamente da droga, com desenvolvimento de lesões pré-neoplásicas em todos. Infelizmente, devido à toxicidade da droga, somente um animal sobreviveu ao tratamento e desenvolveu desenvolveu uma massa tumoral propriamente dita. Foram feitas avaliações periódicas dos animais, em dias pré-determinados (ao início, no 120º, 150º, 300º, 940º) com coletas de fragmentos da mucosa gástrica. Foram coletadas 20 amostras de tecido, distribuídas entre mucosa normal, gastrite, displasia, metaplasia e tumoral. Destas amostras extraímos DNA para as análises do gene CDH1. Não há na literatura sequencia deste gene para a espécie utilizada no estudo. Com este objetivo, utilizamos iniciadores construídos a partir de sequencias de CDH1 de Callithrix jacchus (espécie filogeneticamente) e seqüenciamos cerca de 342pb da região promotora de CDH1 de C. apella. As análises mostraram uma similaridade de 98% desta região com a dos humanos, com presença de vários sítios de ligação de fatores de transcrição (sp1, Ap2, NF-x, AREB6, Puf e CTF) além da presença do CAAT Box. Esta região ainda possui 30 sitios CpGs, o que indica que ela pode estar sofrendo regulação epigenética. Para se verificar como se encontrava o padrão de metilação desta região realizamos uma análise de MSP com iniciadores específicos e constatamos que houve predominância de alelos não metilados de CDH1 para todas as amostras pré neoplásicas e a amostra de adenocarcinoma encontra-se metilada. O que pode ser constatado com um ensaio de Imunohistoquímica, onde somente a amostra tumoral não expressava a proteína caderina. Desta maneira nossas análises sugerem que a metilação do gene CDH1 desempenha papel importante na tumorigênese gástrica em C. apella, e é um evento tardio, uma vez que não é observado nos outro tipos teciduais não tumorais E, partindo-se da similaridade entre as seqüências, podemos sugerir que o mesmo evento pode está ocorrendo em humanos. Esta metilação do promotor do CDH1 leva a um silenciamento deste gene o que desencadeia o estabelecimento de adenocarcinomas gástricos, fato apoiado pela literatura onde vários trabalhos apontaram que a hipermetilação do promotor da caderina está associada ao desenvolvimento do câncer gástrico.

In vitro-produced bovine embryos become infected after exposure to bovine Herpesvirus type 5 (BoHV-5), yet no changes in developmental rates, mitochondrial activity and inhibition of apoptosis are detected in comparison to unexposed embryos. Thus, the aim of the present study was to assess the transcription of mitochondria-mediated apoptosis genes using TaqMan real-time polymerase chain reaction. Transcripts of mcl-1, caspase-2, -3, Apaf-1 and Bax genes were measured after exposure to BoHV-5 in vitro. Mitochondrial dehydrogenase activity was evaluated by MIT test and compared between groups of exposed and unexposed embryos, at day 7 of development. The rate of oocyte maturation was assessed by the extrusion of the first polar body. In summary, BoHV-5 exposed embryos retained their viability, mitochondrial dehydrogenase activity and displayed up-regulation of transcription of survival mcl-1 gene and down-regulation of Bax transcription in relation to mitochondria-mediated pathway which might improve embryo viability. These findings demonstrate that BoHV-5 exposed embryos maintain their viability and mitochondrial dehydrogenase activity with no compromise of embryos produced in vitro. (c) 2013 Elsevier Ltd. HLA-G has a relevant role in immune response regulation. The overall structure of the HLA-G coding region has been maintained during the evolution process, in which most of its variable sites are synonymous mutations or coincide with introns, preserving major functional HLA-G properties. The HLA-G promoter region is different from the classical class I promoters, mainly because (i) it lacks regulatory responsive elements for IFN-gamma and NF-kappa B, (ii) the proximal promoter region (within 200 bases from the first translated ATG) does not mediate transactivation by the principal HLA class I transactivation mechanisms, and (iii) the presence of identified alternative regulatory elements (heat shock, progesterone and hypoxia-responsive elements) and unidentified responsive elements for IL-10, glucocorticoids, and other transcription factors is evident. At least three variable sites in the 3' untranslated region have been studied that may influence HLA-G expression by modifying mRNA stability or microRNA binding sites, including the 14-base pair insertion/deletion, +3142C/G and +3187A/G polymorphisms. Other polymorphic sites have been described, but there are no functional studies on them. The HLA-G coding region polymorphisms might influence isoform production and at least two null alleles with premature stop codons have been described. We reviewed the structure of the HLA-G promoter region and its implication in transcriptional gene control, the structure of the HLA-G 3' UTR and the major actors of the posttranscriptional gene control, and, finally, the presence of regulatory elements in the coding region.

It is believed that epigenetic mechanisms such as DNA methylation are important for the tumorigenesis and maintenance of the altered state of tumor cells. DNA methylation occurs by the addition of a methyl group to carbon 5 of cytosine, catalyzed by the enzyme DNA methyl-transferase, which can change the expression of a gene, including the tumor suppressor genes. In human squamous cell carcinoma, several features have shown the etiological role of genes in tumor development. Among them, FOXE1 gene (forkhead box E1 - thyroid transcription factor) is presented with an important role in susceptibility to disease. Similarly the FOXE1 methylation pattern could alter the expression of this gene in dogs and predisposed to tumor on. Therefore, this study aims to investigate in dogs, the validity of the strategy employed in humans to analyze the FOXE1 methylation status. DNA extraction from fresh frozen tumoral samples was performed by Wizard Genomic® DNA Purification Kit. The methylation status was determined by MSP-PCR (methylation-specific polymerase chain reaction), using 2.0 ng of DNA treated with sodium bisulphate. One hundred micrograms of bisulphite-modified DNA was amplified using primers specific for either methylated or unmethylated DNA (primers sequences are available at http://pathology2.jhu.edu/pancreas/primer.pdf). The analysis of fragments was loaded on to 7% polyacrylamide gels and silver nitrate staining. In this stage of technical approach, 60% were FOXE1 hypermethylated. In conclusion, it was observed that the standard technique for assessing the methylation pattern of gene FOXE1 in humans can be used for the same evaluation in dogs. The correlation of these molecular data with clinical and histopathological parameters may have diagnostic and prognostic value and still be used as a tumor marker for therapeutic decision and surgical approach
