We studied the expression of arachidonate 5-lipoxygenase (5-LO) in a cell line of human keratinocytes (HaCaT) and in normal human skin keratinocytes in tissue culture. In undifferentiated keratinocytes 5-LO gene expression was low or undetectable as determined by 5-LO mRNA, protein, cell-free enzyme activity, and leukotriene production in intact cells. However, after shift to culture conditions that promote conversion of prokeratinocytes into a more differentiated phenotype, 5-LO gene expression was markedly induced in HaCaT cells and, to a lesser extent, in normal keratinocytes. These results show that 5-LO gene expression is an intrinsic property of human skin keratinocytes.

The epithelial-specific integrin alpha 6 beta 4 is suprabasally expressed in benign skin tumors (papillomas) and is diffusely expressed in carcinomas associated with an increase in the proliferating compartment. Analysis of RNA samples by reverse transcriptase-PCR and DNA sequencing revealed that chemically or oncogenically induced papillomas (n = 8) expressed a single transcript of the alpha 6 subunit, identified as the alpha 6 A splice variant. In contrast, carcinomas (n = 13) expressed both alpha 6A and an alternatively spliced form, alpha 6B. Primary keratinocytes and a number of keratinocyte cell lines that vary in biological potential from normal skin, to benign papillomas, to well-differentiated slowly growing carcinomas exclusively expressed alpha 6A. However, I7, an oncogene-induced cell line that produces highly invasive carcinomas, expressed both alpha 6A and alpha 6B transcript and protein. The expression of alpha 6B in I7 cells was associated with increased attachment to a laminin matrix compared to cell lines exclusively expressing alpha 6A. Furthermore, introduction of an alpha 6B expression vector into a papilloma cell line expressing alpha 6A increased laminin attachment. When a papilloma cell line was converted to an invasive carcinoma by introduction of the v-fos oncogene, the malignant cells expressed both alpha 6A and alpha 6B, while the parent cell line and cells transduced with v-jun or c-myc, which retained the papilloma phenotype, expressed only alpha 6A. Comparative analysis of alpha 6B expression in cell lines and their derived tumors indicate that alpha 6B transcripts are more abundant in tumors than cell lines, and alpha 6B is expressed to a greater extent in poorly differentiated tumors. These results establish a link between malignant conversion and invasion of squamous tumor cells and the regulation of transcript processing of the alpha 6 beta 4 integrin.

This report explores the mechanism of spontaneous closure of full-thickness skin wounds. The domestic pig, often used as a human analogue for skin wound repair studies, closes these wounds with kinetics similar to those in the guinea pig (mobile skin), even though the porcine dermis on the back is thick and nearly immobile. In the domestic pig, as in the guinea pig, daily full-thickness excisions of the central granulation tissue up to but not including the wound edges in both back and flank wounds do not alter the rate or completeness of wound closure or the final pattern of the scar. A purse-string mechanism of closure was precluded by showing that surgical interruption of wound edge continuity does not alter closure kinetics or wound shape. We conclude that "tightness" of skin is not a key factor nor is the central granulation tissue required for normal wound closure. These data imply that in vitro models such as contraction of isolated granulation tissue or of the cell-populated collagen lattice may not be relevant for understanding the cell biology of in vivo wound closure. Implications for the mechanism for wound closure are discussed.

The photoproduction of vitamin D in the skin was essential for the evolutionary development of terrestrial vertebrates. During exposure to sunlight, previtamin D3 formed in the skin is isomerized to vitamin D3 (calciol) by a temperature-dependent process. Since early land vertebrates were poikilothermic, the relatively slow conversion of previtamin D3 to vitamin D3 at ambient temperature put them at serious risk for developing vitamin D deficiency, thus leading to a poorly mineralized skeleton that could have ultimately halted further evolutionary development of vertebrates on land. We evaluated the rate of isomerization of previtamin D3 to vitamin D3 in the skin of iguanas and found the isomerization rate was enhanced by 1100% and 1700% at 25 degrees C and 5 degrees C, respectively. It is likely that the membrane entrapment of previtamin D3 in its s-cis,s-cis conformation is responsible for the markedly enhanced conversion of previtamin D3 to vitamin D3. The membrane-enhanced production of vitamin D3 ensures the critical supply of vitamin D3 to poikilothermic animals such as iguanas.

We have examined whether the secretion of erythropoietin (Epo) from genetically modified cells could represent an alternative to repeated injections of the recombinant hormone for treating chronic anemias responsive to Epo. Primary mouse skin fibroblasts were transduced with a retroviral vector in which the murine Epo cDNA is expressed under the control of the murine phosphoglycerate kinase promoter. "Neo-organs" containing the genetically modified fibroblasts embedded into collagen lattices were implanted into the peritoneal cavity of mice. Increased hematocrit (> 80%) and elevated serum Epo concentration (ranging from 60 to 408 milliunits/ml) were observed in recipient animals over a 10-month observation period. Hematocrit values measured in recipient mice varied according to the number of implanted Epo-secreting fibroblasts (ranging from 2.5 to 20 x 10(6)). The implantation of neo-organs containing Epo-secreting fibroblasts appeared, therefore, as a convenient method to achieve permanent in vivo delivery of the hormone. We estimated that the biological efficacy of the approach may be relevant for the treatment of human hemoglobinopathies.

A microwave-assisted extraction (MAE) procedure to isolate phenolic compounds from almond skin byproducts was optimized. A three-level, three-factor Box–Behnken design was used to evaluate the effect of almond skin weight, microwave power, and irradiation time on total phenolic content (TPC) and antioxidant activity (DPPH). Almond skin weight was the most important parameter in the studied responses. The best extraction was achieved using 4 g, 60 s, 100 W, and 60 mL of 70% (v/v) ethanol. TPC, antioxidant activity (DPPH, FRAP), and chemical composition (HPLC-DAD-ESI-MS/MS) were determined by using the optimized method from seven different almond cultivars. Successful discrimination was obtained for all cultivars by using multivariate linear discriminant analysis (LDA), suggesting the influence of cultivar type on polyphenol content and antioxidant activity. The results show the potential of almond skin as a natural source of phenolics and the effectiveness of MAE for the reutilization of these byproducts.

Los estudios epidemiológicos realizados hasta la fecha en neonatos hospitalizados son escasos. En España en las unidades neonatales las cifras de prevalencia se mueven entre un 50% en unidades de cuidados intensivos (UCIN) y un 12,5% en unidades de hospitalización. Desde todas las organizaciones de salud, nacionales (GNEAUPP) e internacionales (EPUAP, EWMA, NPUAP) relacionadas con las heridas, se promociona y potencia la seguridad del paciente mediante la prevención de los efectos adversos hospitalarios. Para conseguirlo es necesario dotar a los profesionales sanitarios de herramientas validadas y adaptadas a la edad neonatal que permitan valorar el riesgo de la población hospitalizada. De esta forma los profesionales sanitarios podrán gestionar de forma eficiente los recursos preventivos y trazar planes de cuidados centrados en el neonato. En la actualidad, en España no existe ninguna escala validada específicamente para neonatos. Por tanto, el objetivo principal de nuestro equipo de investigación fue adaptar transculturalmente al contexto español la escala NSRAS original y evaluar la validez y la fiabilidad de la versión en español. En esta ponencia se presentarán los resultados preliminares de la tesis. Método. El estudio se subdividió en tres fases. En la primera fase se realizó la adaptación transcultural de la escala NSRAS original mediante el método de traducción con retrotraducción. Posteriormente entre un grupo de expertos se calculó la validez de contenido mediante el IVC. La versión de la escala adaptada fue evaluada mediante dos fases de estudio multicéntrico observacional analítico en las unidades neonatales de 10 hospitales públicos del Sistema Nacional de Salud. Se evaluó la fiabilidad interobservadores e intraobservadores, la validez de constructo en la segunda fase y en una tercera fase se evaluó la capacidad predictiva y el punto de corte de la versión en español de la escala NSRAS. Resultados. En la primera fase la validez de contenido evaluada obtuvo un IVC de 0,926 [IC95%0,777-0,978]. En la segunda fase, la muestra evaluada fue de 336 neonatos. La consistencia interna mostró un Alfa de Cronbach de 0,794. Y la fiabilidad intraobservadores fue de 0,932 y la fiabilidad interobservadores fue de 0,969. En la tercera fase la muestra evaluada fue de 268 neonatos. El análisis multivariante de la relación entre los factores de riesgo, las medidas preventivas y la presencia de UPP mostró que 3 variables eran significativas: la puntuación NSRAS, la duración del ingreso y el uso de VMNI. Siendo de esta forma la puntuación NSRAS (debido a que activa las medidas preventivas) un factor protector frente a UPP. Es decir, a mayor puntuación de NSRAS, menor riesgo de UPP. La valoración clinicométrica de la puntuación 17 mostró una sensibilidad del 91,18%, una especificidad de 76,5%, un VPN de 36,05% y un VPP de 98,35%. El área bajo la curva ROC fue de 0,8384 en la puntuación 17. Conclusiones. La versión en español de la escala NSRAS es una herramienta válida y fiable para medir el riesgo de UPP en la población neonatal hospitalizada en el contexto español. Los neonatos hospitalizados con una puntuación igual o menor a 17 están en riesgo desarrollar UPP.

A presente dissertação teve como objetivo o desenvolvimento de uma aplicação informática para automatização do dimensionamento de elementos de betão armado com GFRP, através da utilização de guias e normas existentes. O documento elaborado apresenta uma breve síntese da situação atual do estado do conhecimento (estado da arte) nesta temática, uma abordagem aos conceitos de dimensionamento, de elementos estruturais baseados em documentos normativos, e alguns conceitos sobre a programação efetuada, quer através da descrição das metodologias quer por esquematização em figuras. Neste documento é, também, descrito o funcionamento geral do programa, exemplos de aplicação prática e breves considerações sobre o trabalho desenvolvido. A aplicação efetua o cálculo de vigas e lajes de acordo com o guia do Committee 440 do American Concrete Institute, ACI 440.1R-06 – “Guide for the Design and Construction of Structural Concrete Reinforced with FRP Bars”, bem como através de uma análise não linear, como descrito na norma NP EN 1992: Eurocódigo 2 – “Projecto de estruturas de betão”. Foram ainda realizados cálculos para determinação de diagramas de interação de flexão composta, com pares de esforços axiais e de flexão, utilizados para o dimensionamento de colunas. No final do desenvolvimento da aplicação o programa foi testado com exemplos de dimensionamento, para que fosse possível realizar uma análise crítica aos resultados obtidos e ao próprio modo de funcionamento da aplicação.

Este trabalho consiste na apresentação de técnicas de reabilitação e reforço e a sua adaptação ao GFRP (Glass Fiber Reinforced Polymer). O foco incidiu sobre as estruturas degradadas pela corrosão das armaduras, e apresenta-se uma vasta gama de técnicas de intervenção. Encamisamento, proteção eletroquímica, adição de armaduras são as técnicas mais desenvolvidas ao longo do trabalho. São ainda abordados os tipos de produtos de recobrimento, as técnicas de aplicação do recobrimento, bem como as ferramentas com maior utilidade para a reabilitação e reforço. De seguida faz-se um breve resumo das características e potencialidades do material GFRP, aborda-se alguns dos principais fornecedores, fazendo uma descrição dos produtos comercializados por estes, e ainda uma análise de preços por metro linear de varão GFRP. Por fim apresenta-se um levantamento fotográfico realizado na Região Autónoma da Madeira, de estruturas que apresentem sinais claros de corrosão das armaduras, com o objetivo de perceber a aplicabilidade dos varões de GFRP na reabilitação das mesmas. Apresenta-se ainda, uma possível técnica de reabilitação/reforço para todos os casos apresentados, utilizando os varões de GFRP. O levantamento fotográfico tem ainda o objetivo de perceber as principais causas de deterioração de estruturas na região, e de criar uma base de estruturas a necessitar de intervenção urgente.

BACKGROUND Stiff skin syndrome and systemic or localized scleroderma are cutaneous disorders characterized by dermal fibrosis and present clinically with induration of the skin, with or without joint, internal organ or vascular involvement. OBJECTIVES To provide clinical, histological and preliminary genetic analysis of two West Highland white terrier siblings presenting with indurated skin resembling stiff skin syndrome in humans. ANIMALS Two client owned full sibling West Highland white terriers from two different litters. METHODS Clinical examination, histopathological examination and whole genome sequencing analysis of affected and unaffected West Highland white terriers. RESULTS Affected dogs exhibited markedly indurated skin that was attached firmly to the underlying tissue and incomplete closure of the mouth and eyes. No abnormalities were found by neurological or orthopaedic examination, radiographs of the head or whole body computed tomography. Histologically, the dermis and pannicular septa were thickened by a marked increase in coarse collagen fibres and a mild to moderate increase in collagen fibre diameter. The syndrome most likely follows an autosomal recessive mode of inheritance. The sequence analysis did not reveal any obvious causative variant in the investigated candidate genes ADAMTSL2 and FBN1. CONCLUSION AND CLINICAL IMPORTANCE The clinical phenotype and histopathological features of two West Highland white terrier siblings resembled stiff skin syndrome in humans. Unlike in humans, or previously described beagles with stiff skin, there was no restriction of joint mobility. Genetic analysis did not detect a candidate causative variant and warrants further research.
