The steady-state negative supercoiling of eubacterial genomes is maintained by the action of DNA topoisomerases. Topoisomerase distribution varies in different species of mycobacteria. While Mycobacterium tuberculosis (Mtb) contains a single type I (Topol) and a single type II (Gyrase) enzyme, Mycobacterium smegmatis (Msm) and other members harbour additional relaxases. Topol is essential for Mtb survival. However, the necessity of Topol or other relaxases in Msm has not been investigated. To recognize the importance of Topol for growth, physiology and gene expression of Msm, we have developed a conditional knock-down strain of Topol in Msm. The Topol-depleted strain exhibited extremely slow growth and drastic changes in phenotypic characteristics. The cessation of growth indicates the essential requirement of the enzyme for the organism in spite of having additional DNA relaxation enzymes in the cell. Notably, the imbalance in Topol level led to the altered expression of topology modulatory proteins, resulting in a diffused nucleoid architecture. Proteomic and transcript analysis of the mutant indicated reduced expression of the genes involved in central metabolic pathways and core DNA transaction processes. RNA polymerase (RNAP) distribution on the transcription units was affected in the Topol-depleted cells, suggesting global alteration in transcription. The study thus highlights the essential requirement of Topol in the maintenance of cellular phenotype, growth characteristics and gene expression in mycobacteria. A decrease in Topol level led to altered RNAP occupancy and impaired transcription elongation, causing severe downstream effects.

This is the first successful attempt to produce Mg-Ce alloys of different texture through different processing routes while keeping the grain size and grain size distribution same. Tensile data shows that contribution of texture to ductility enhancement is primary and that of grain refinement is secondary. The texture resulting from multi-axial forging of extruded billets followed by annealing exhibits the highest ductility (similar to 40%) at room temperature. (C) 2015 Elsevier B.V. Occurrence of the April 25, 2015 (Mw 7.8) earthquake near Gorkha, central Nepal, and another one that followed on May 12 (Mw 7.3), located similar to 140 km to its east, provides an exceptional opportunity to understand some new facets of Himalayan earthquakes. Here we attempt to assess the seismotectonics of these earthquakes based on the deformational field generated by these events, along with the spatial and temporal characteristics of their aftershocks. When integrated with some of the post-earthquake field observations, including the localization of damage and surface deformation, it became obvious that although the mainshock slip was mostly limited to the Main Himalayan Thrust (MHT), the rupture did not propagate to the Main Frontal Thrust (MFT). Field evidence, supported by the available InSAR imagery of the deformation field, suggests that a component of slip could have emerged through a previously identified out-of-sequence thrust/active thrust in the region that parallels the Main Central Thrust (MCT), known in the literature as a co-linear physiographic transitional zone called PT2. Termination of the first rupture, triggering of the second large earthquake, and distribution of aftershocks are also spatially constrained by the eastern extremity of PT2. Mechanism of the 2015 sequence demonstrates that the out-of-sequence thrusts may accommodate part of the slip, an aspect that needs to be considered in the current understanding of the mechanism of earthquakes originating on the MHT. In this paper, we present the solutions of 1-D and 2-D non-linear partial differential equations with initial conditions. We approach the solutions in time domain using two methods. We first solve the equations using Fourier spectral approximation in the spatial domain and secondly we compare the results with the approximation in the spatial domain using orthogonal functions such as Legendre or Chebyshev polynomials as their basis functions. The advantages and the applicability of the two different methods for different types of problems are brought out by considering 1-D and 2-D nonlinear partial differential equations namely the Korteweg-de-Vries and nonlinear Schrodinger equation with different potential function. Atomic force Microscopy (AFM) has become a versatile tool in biology due to its advantage of high-resolution imaging of biological samples close to their native condition. Apart from imaging, AFM can also measure the local mechanical properties of the surfaces. In this study, we explore the possibility of using AFM to quantify the rough eye phenotype of Drosophila melanogaster through mechanical properties. We have measured adhesion force, stiffness and elastic modulus of the corneal lens using AFM. Various parameters affecting these measurements like cantilever stiffness and tip geometry are systematically studied and the measurement procedures are standardized. Results show that the mean adhesion force of the ommatidial surface varies from 36 nN to 16 nN based on the location. The mean stiffness is 483 +/- 5 N/m, and the elastic modulus is 3.4 +/- 0.05 GPa (95% confidence level) at the center of ommatidia. These properties are found to be different in corneal lens of eye expressing human mutant tau gene (mutant). The adhesion force, stiffness and elastic modulus are decreased in the mutant. We conclude that the measurement of surface and mechanical properties of D. melanogaster using AFM can be used for quantitative evaluation of `rough eye' surface. (C) 2015 Elsevier Ltd. This work intends to demonstrate the effect of geometrically non-linear cross-sectional analysis of certain composite beam-based four-bar mechanisms in predicting the three-dimensional warping of the cross-section. The only restriction in the present analysis is that the strains within each elastic body remain small (i.e., this work does not deal with materials exhibiting non-linear constitutive laws at the 3-D level). Here, all component bars of the mechanism are made of fiber-reinforced laminates. They could, in general, be pre-twisted and/or possess initial curvature, either by design or by defect. Each component of the mechanism is modeled as a beam based on geometrically non-linear 3-D elasticity theory. The component problems are thus split into 2-D analyses of reference beam cross-sections and non-linear 1-D analyses along the three beam reference curves. The splitting of the three-dimensional beam problem into two- and one-dimensional parts, called dimensional reduction, results in a tremendous savings of computational effort relative to the cost of three-dimensional finite element analysis, the only alternative for realistic beams. The analysis of beam-like structures made of laminated composite materials requires a much more complicated methodology. Hence, the analysis procedure based on Variational Asymptotic Method (VAM), a tool to carry out the dimensional reduction, is used here. The representative cross-sections of all component bars are analyzed using two different approaches: (1) Numerical Model and (2) Analytical Model. Four-bar mechanisms are analyzed using the above two approaches for Omega = 20 rad/s and Omega = pi rad/s and observed the same behavior in both cases. The noticeable snap-shots of the deformation shapes of the mechanism about 1000 frames are also reported using commercial software (I-DEAS + NASTRAN + ADAMS). The maximum out-of-plane warping of the cross-section is observed at the mid-span of bar-1, bar-2 and bar-3 are 1.5 mm, 250 mm and 1.0 mm, respectively, for t = 0:5 s. (C) 2015 Elsevier Ltd. La Brucelosis Bovina es una enfermedad infectocontagiosa que afecta al hombre, animales domésticos y salvajes la cual es producida por una Bacteria del genero Brucella. Es una zoonosis por excelencia ya que se trasmite en forma natural de los animales vertebrados al nombre ocasionando pérdidas directas reduciendo la producción. Con el objetivo de determinar la prevalencia en el departamento de León se desarrolló un "ESTUDIO EPIDEMIOLOGICO SOBRE LA PREVALENCIA DE BRUCELOSIS BOVINA 2000-2006 EN MUNICIPIOS DEL DEPARTAMENTO DE LEON" Por lo que se realizó un sangrado (Sangre sin anticoagulante) a una población total de 161,522 Bovinos distribuidos en las 3 fincas de referencia del Departamento de León, para este estudio diagnóstico se utilizó el método de Rosa de Bengala y un método confirmativo Rivanol. Los resultados del diagnóstico serológico presentaron una prevalencia global de 0.16%, presentando la finca de referencia "La Base" una prevalencia de 0.18%, La Garrapata 0.22% y Sán José del Fortín 0.02% y una prevalencia de 0% para los municipios El Jicaral, Nagarote y Quesalguaque, 0.18% para la Paz Centro , 0.02% para León, 0.10% para el Sauce, 0.41% para Telica y el municipio con mayor nivel de prevalencia fue Malpaisillo con 0.49% de una población total de 2,831 animales, por lo que se concluye en este trabajo de tesis que se obtuvo resultados con prevalencia de 0.16 %, lo cual es muy baja en novillos analizados por lo que se recomienda un estricto control y vigilancia para mantener el nivel bajo de prevalencia del Departamento de León y resto del país.

An experimental investigation of Benard-Marangoni convection has been performed in double immiscible liquid layers of rectangular configuration. The two kinds of liquid are 10cst silicon oil and FC-70 respectively. The velocity fields in the vertical cross-section are obtained by PIV. Flow patterns and/or temperature distributions on the horizontal interface are displayed by using thermal color liquid crystal (TLC), and the velocity distributions on the interface were also obtained with the help of the serial particle image of TCL. The evolution processes of convection are observed in the differential thickness ratio of two liquid layers, and the convection styles are discussed.

El presente trabajo se planificó con el propósito de determinar la influencia de diferentes dosis (a2,a3 y a4) de humus de lombriz y dos momentos de aplicación (b1 y b2) sobre el crecimiento y rendimiento del cultivo del ajonjolí(Sesamum indicun L), variedad Cuyumaqui, bajo las condiciones ecológicas de la finca La Concepción, Nagarote, León. El ensayo se estableció en la época de primera del año 2004, utilizándose un diseño de bloques completos al azar con arreglos en parcelas divididas con cuatro repeticiones. Se encontró que las variables número de semillas por cápsulas y peso de mil semillas resultaron no significativas, pero si se encontró diferencias estadísticas para la altura de planta, el diámetro del tallo, el número de hojas por planta, el número de cápsulas por planta y el rendimiento de grano los que mostraron un efecto significativo ante los niveles del Factor A (dosis de humus de lombriz), del Factor B (momento de aplicación) y la interacción dosis de humus de lombriz por momento de aplicación. Para el rendimiento de grano, los niveles nivel a2,a3 y a4 del Factor A, el nivel b2 del Factor B y las interacciones a3 b2 y a2 b2 indujeron a obtener los mayores rendimiento de grano de 1 161.70 y 1 158.70 kg-1.

Con la finalidad de conocer el estado de la aplicación de Buenas Prácticas Agrícolas (BPA) y obtener información básica para diseñar un Manual de BPAs en el cultivo de la fresa, se realizó un diagnóstico en 3 fincas productoras de fresa de uso industrial del departamento de Jinotega, Nicaragua. Dicho diagnóstico se llevó a cabo el 1 de noviembre de 2005. Las 3 fincas en estudio fueron Sta Carmela, San Carlos y Jardines Las Pilas. Para realizar el diagnóstico se uso la metodología que emplea el departamento de inspección a fincas y trazabilidad del Ministerio Agropecuario y Forestal. Los resultados del estudio indican que ninguna de las tres fincas alcanzó el puntaje mínimo (85%) requerido para alcanzar la certificación BPA, dentro del marco de los requisitos exigidos por el MAG-FOR. La finca que obtuvo mayor puntaje fue La finca San Carlos (201; 50 %); la finca Jardines Las Pilas obtuvo el segundo lugar en puntaje (181; 45 %) y la finca Santa Carmela obtuvo el puntaje mas bajo (161;40%). Entre los aspectos con una calificación de menor puntaje figuran: 1. Estiércol y biosólidos, 2. Producto, 3. Almacenamiento y 4. Organismos genéticamente modificados, este ultimo no aplica para las tres fincas en su totalidad. Estos aspectos deberán ser los prioritarios en un plan de trabajo para corregir inconformidades a fin de alcanzar la certificación de BPA. Después de concluido el diagnóstico, se realizó un manual de BPA para este cultivo, para el cual se empleo el contenido mínimo recomendado por el MAG-FOR. El contenido del manual fue de 10 puntos que se detallan a continuación: 1. Buenas prácticas agrícolas para el manejo del agua, 2. buenas prácticas agrícolas para el manejo de suelos, 3. Buenas prácticas agrícolas para la fertilización de cultivos, 4. Buenas prácticas agrícolas para la protección de cultivos, 5. Buenas prácticas agrícolas para la recolección y el manejo postcosecha, 6. Buenas practicas agrícolas en los elementos de apoyo para las labores productivas, 7. Buenas prácticas agrícolas en la salud, seguridad y bienestar de los trabajadores, 8. Buenas practicas agrícolas para la trazabilidad y los registros, 9. Glosario y 10 bibliografía.
