All medicine, whether allopathic or homeopathic, must go through strict quality control, which must ratify their characteristics throughout the period of validity. During the time of preparation and storage, solutions of the drugs are in permanent contact with packaging materials that can release undesirable substances to the solution. Several factors may influence the release of packing materials, and factorial design (FD) is a useful tool for analyzing the phenomenon. The aim of this study was the determination of quality parameters for Homeopathic solid (globules) and liquid (drops) dosage forms. It was carried out analysis in homeopathic globules for weight variation, mechanical strength, and moisture content uniformity. For liquid preparations, standard solutions were prepared from natural rubber bulbs, which were subjected to exhaustive extraction with two ethanol solutions (30 and 70%) in the ultrasonic bath for 20 minutes at 25°C and 50°C in three successive cycles. Studies of transfer have been made within five days, by spectrophotometric analysis in the UV region at 312 nm with λmáx and 323 nm for samples in 70% ethanol and 30% respectively. PH values were analyzed. We also conducted two FD studies, where the first, the three-level variables were solvent (chloroform, ethanol and nhexane), sample mass (30, 60 and 90mg), particle size (large disk, small disk and powder sample). In the second study, the solvent level variables were different ethanolic degrees (EtOH 30%, 70% and pure). The percentage of lending in the solutions was 5.5%, 12.4%, 24.2% and 41% of the total estimated in the reference solution. The values of rate constants of transfer were determined in the order of 0.0134 days-1 and 0.0232 days-1 in absorbance values, the solutions in ethanol at 30% and 70% respectively. These results suggest that the speed of transfer of materials from rubber is affected both by the nature of the vehicle as by the temperature

In Brazil, several species of scorpions are known to cause accidents which can lead to death, which are mainly belonging to the genus Tityus. The scorpion Tityus serrulatus is the main responsible for more severe cases. Anti-scorpion serums are routinely produced by various institutions, despite their effectiveness, quality and action depends on how quickly treatment is started. Studies have been developed in the search for appropriate technologies to encapsulate and release recombinant or natives proteins capable of inducing antibody production. In this context, chitosan copolymer which can be obtained from the partial deacetylation of chitin or in some microorganisms and it is biocompatible and biodegradable has been widely used for this purpose. This study aimed to search for a system release from chitosan nanoparticles for peptide / protein of the venom of the scorpion T. serrulatus, able to provide a new model of immunization in animals, in order to obtain a potential novel polyclonal serum, anti-venom T. serrulatus. The chitosan nanoparticles were prepared by ionic gelation with polyanion tripolyphosphate (TPP). After standardizing the concentrations of TPP and chitosan was evaluated the efficiency of incorporation of bovine serum albumin (BSA) and scorpion venom, showed particle size compatible with the intended purpose. The particles showed adequate size around 200nm. The crosslinking was confirmed by absorption spectroscopy in the infrared. After verified the high encapsulation efficiency (EE) for acid bicinconínico method (BCA) protein assay and the particle size distribution, the success of the technique was proven and the potential for in vivo application of nanoparticles. Clays are natural materials that have great potential for use as excipients for solid dosage forms. Palygorskite is a type of clay that has hydrophilic properties as well as a large surface area, which could contribute to the dissolution of drugs. Thus, the present study aims to evaluate the use of palygorskite clay, from Piaui (Northeast region of Brazil), as a pharmaceutical excipient for solid dosage forms, using rifampicin and isoniazid as the model drugs. The former is a poorly soluble drug often associated with isoniazid for tuberculosis treatment. Palygorskite was characterized by X-ray diffraction (XRD), X-ray fluorescence (XRF), particle size, transmission electron microscopy (TEM), scanning electron microscopy (SEM) and specific surface area (BET). The rheological and technological properties of palygorskite were determined and compared to those of talc, magnesium stearate and Aersosil 200. Mixtures between drugs and palygorskite were analyzed by differential scanning calorimetry (DSC), thermogravimetric analysis (TG) combined with thermal analysis (DTA) and Fourier Transform Infrared Spectroscopy (FT-IR), where the results were compared with those of the individual compounds. In addition, dissolution studies of solid dispersions and capsules containing the drugs, mixed with either palygorskite or a mixture of talc and magnesium stearate, were performed. The results showed that palygorskite has small particles with a high surface area. Its rheological characteristics were better than those of others commonly used glidants and lubricants. There was no interaction between palygorskite and the drugs (rifampicin and isoniazid). Among the dispersions studied, the mixture with palygorskite (5%) showed the highest drug dissolution when compared to other excipients. The dissolution of the rifampicin capsules containing palygosrkite was faster in higher concentrations. However, these differences were statistically different only in the first minutes of the dissolution experiment. The dissolution profile of isoniazid was also statistically different on the initial part of the experiment. The formulations prepared with isoniazid and palygorskite showed higher drug dissolution, but it was in descending order of concentration. According to these results, the palygorskite clay used in this study has great potential for application as an excipient for solid dosage forms

The synthetic guanylhydrazones WE010 (3,5-di-tert-butil-4-hidroxibenzaldehyde-guanylhydrazone), WE014 (4-bifenilcarboxialdehydeguanylhydrazone) and WE017 (3,4-diclorobenzaldehydeguanylhydrazone) showed high cytotoxic activity in terms of percentage inhibition of cancer cells growth. However, further progress in the development of these drug candidates requires precise and convenient methods for their qualitative and quantitative analyses. The aim of this study was to develop and validate High Performance Liquid Chromatography with diode-array detection (HPLC-DAD) and Ultra Fast Liquid Chromatography with diode-array detection (UFLC-DAD) methods suitable for as simultaneous as isolated determination of studied guanylhydrazones, based on the optimization of chromatographic parameters and obtaining reduced detection times. The chromatographic analyses of analytes by HPLC were performed on C18 ACE analytical column (150 mm x 4.6 mm), with a particle size of 5.0 μm. Among all the conditions assayed, the best results of separation were obtained with a mixture of methanol:water (60:40, v/v) as the mobile phase at a flow rate 1.5mL/min and pH of 3.5 adjusted at acetic acid. The UFLC method was developed by experimetal desing techniques in order to find optimal chromatographic analytical conditions, which were achieved on XR-ODS analytical column (50 mm x 3.0 mm), with a particle size of 2,2 μm, maintained at 25 ºC. The mobile phase was consisted of methanol:water (65:35, v/v) with 0.1% triethylamine (TEA) and pH of 3.5 adjusted at acetic acid, at a flow rate 0.5 mL/min. The procedure were validated following evaluating parameters such as specificity, linearity, limits of detection (LD) and quantification (LQ), precision, accuracy and robustness, giving results within the acceptable range. Although the UFLC method shows better sensitivity (lower values of LD and LQ), robustness (lower rates of relative standard deviation) and minimize spending time and solvent, both developed methods were adequately applied to the analysis of guanylhydrazones molecules, may be used in routine of quality control laboratories. The present study aimed to characterize the thermal profile of wood fired oven used by the red ceramic industry in Parelhas, in the Seridó region/RN, aiming to propose structural interventions that can contribute to increasing productivity and product quality, optimize wood consumption and mitigate existing losses during the burning process. The study was conducted at Cerâmica Esperança in the city of Parelhas -RN, Brazil, during the period from August 2012 to September 2013. Four treatments were performed with three replicates, ie, with, a total of 12 experimental units (burnings). In the first stage 4 treatments were performed with three replicates, totaling 12 experimental units (firings). In the second stage 2 treatments were performed with three replications, totaling 6 experimental units (firings). The physical characteristics of the wood were analyzed using standard NBR 11941 and NBR 7190 for basic density and moisture, respectively. The clay was used as a reference parameter for distinguishing treatments. For both the analysis and characterization was carried out using techniques of fluorescence X (XRF) rays, X-ray diffraction (XRD) analysis, particle size analysis (FA). In the first and second stages were monitored: the time during the firing process, the amount of wood used at each firing, the number of parts enfornadas for subsequent determination of the percentages of losses, but also product quality. To characterize the thermal profile of the oven, we measured the temperature at 15 points scored in the surface charge put into the oven. Measurements were taken every 30 minutes from preheat until the end of burning, using a pyrometer laser sight sighting from preheating until the end of burning. In the second step 12 metal cylinders distributed on the oven walls, and the cylinder end walls 8 of the furnace 2 and rollers on each side walls are installed equidistant to 17 cm from the soil and the surface 30 of the wall are installed. The cylinders distributed on the front were placed 50 cm above the furnace, and the base of the oven 20 cm distant from the ground. 10 also thermocouples were installed, and five thermocouples distributed 1.77 cm above the combustion chambers, and one thermocouple on each side, and three thermocouples in front of the oven. We carried out the measurements of the temperatures every 1 hour during the burning two hours in cooling the cylinders with a pyrometer and thermocouples for dattaloger. These were fixed with depth of 30 cm from the wall. After statistical analysis it was found that: the thermal profile of the furnace surface and at different heights was heterogeneous; and the ranges of density and moisture content of wood are within recommended for use as an energy source standards. We conclude that even at low temperatures reached during firing there was a significant production of good quality products, this is due to high concentrations of iron oxide and potassium oxide found in clay, which lowers the melting point of the piece. The average burn time for each step varied 650-2100 minutes wood consumption was on average 20 m3, product quality was on average 16% of first quality, 70% second, third and 5% to 10% loss . The distance between the wire and the surface of the oven was a significant parameter for all treatments, but with different variations, meaning that the wire should not be so generic and unique form, used as a criterion for completion of the burn process. The central part of the furnace was the area that reached higher temperature, and in a unified manner, with the highest concentration of top quality products. The ideal temperature curve, which provided the highest quality of ceramic products was achieved in the central part of the furnace

The present study aimed to compare the main zootechnical and economic indicators of farming tilapia in superintensive system developed in two different net cages dimensions: 6 and 18 m(3). Were used 12,960 juveniles male reversed Nile tilapia (Oreochromis niloticus), strain supreme, with an initial average weight of 32.0 g (+/- 0.85) for 6 m(3) net cages and 88.5 g (+/- 0.96) for 18 m(3) net cages. The fish were fed three times a day with commercial extruded ration, crude protein levels from 40 to 32%, particle size from 2 to 8 mm and daily feeding rate starting with 8% of live weight (LW), decreasing up to 1% LW. It was conclued that, although no statistical difference in most indicators zootechnical (P>0.05), the production of tilapias in bigger net cages resulted in lower average total cost and higher rentability per kg of fish produced, 34.4% higher, resulting in better profitability.

This thesis aimed to assess the increase in solubility of simvastatin (SINV) with solid dispersions using techniques such as kneading (MA), co-solvent evaporation (ES), melting carrier (FC) and spray dryer (SD). Soluplus (SOL), PEG 6000 (PEG), PVP K-30 (PVP) e sodium lauryl sulphate (LSS) were used as carriers. The solid dispersions containing PEG [PEG-2(SD)], Soluplus [SOL-2(MA)] and sodium lauryl sulphate [LSS-2(ES)] were presented with a greater increase in solubility (5.02, 5.60 and 5.43 times respectively); analyses by ANOVA between the three groups did not present significant difference (p<0.05). In the phase solubility study, the calculation of the Gibbs free energy (ΔG) revealed that the spontaneity of solubilisation of SINV occurred in the order SOL>PEG >PVP 75%>LSS, always 80%. The phase diagrams of PEG and LSS presented solubilization stoichiometry of type 1:1 (type AL). The diagrams with PVP and SOL tend to 1:2 stoichiometry (type AL + AP). The stability coefficients (Ks) of the phase diagrams revealed that the most stable reactions occurred with LSS and PVP. The solid dispersions were characterized by Fourier transform infrared (FTIR), differential scanning calorimetry (DSC), scanning electron microscopy (SEM), particle size distribution (PSD), near-infrared spectroscopy imaging (NIR-CI) and X-ray diffraction of the powder using the Topas software (PDRX-TOPAS). The solid dispersion PEG-2(SD) presented the greatest homogeneity and the lowest degree of crystallinity (18.2%). The accelerated stability study revealed that the solid dispersions are less stable than SINV, with PEG-2(SD) being the least stable, confirmed by FTIR and DSC. The analyses by PDRX-TOPAS revealed the amorphous character of the dispersions and the mechanism of increasing solubility

It was aimed to evaluate the spectrum and uniformity of droplets in function of sprayers, spraying volumes and rates of chemical insecticide on the mortality of Pseudoplusia includens in laboratory. The work was carried out at UNESP, Jaboticabal, SP, Brazil. The following treatments were applied over the caterpillars: two equipments (atomizer and hydraulic nozzle); two spray volumes (17 and 50 L ha(-1) for the atomizer and 50 and 100 L ha(-1) for the hydraulic nozzle); and two dosages of the insecticide endosulfan (0.5 and 1.0 L pc ha(-1)), in fully randomized plots, in a factorial scheme 2x2x2 and 1 untreated check. The caterpillars mortality was evaluated until the 6(th) day after the application of treatments. The spectrum of the droplets was evaluated in a particle size analyzer in real time determining the diameter and spectrum of droplets sprayed through the laser beam of the analyser. It was verified that the spray volume can be reduced by 17 L ha(-1) without losses on the P. includens control. The doses of 0.5 L pc ha(-1) (recommended for Anticarsia gemmatalis) did not satisfactorily control the caterpillar soybean looper. The atomizer produces droplets of higher uniformity (SPAN: 0.52) and lower percentage of droplets susceptible to drift (3.3%) compared to hydraulic nozzle (SPAN: 1.34 e % droplets <= 100 mu m: 15.2).

Os objetivos deste trabalho foram avaliar o desempenho germinativo de sementes de palmeira-australiana em diferentes substratos e estabelecer a granulometria e intensidade de umedecimento adequado. A semeadura foi realizada com quatro repetições de 25 sementes em solo, areia, rolo de papel e em vermiculita de diferentes granulometrias: mícron (0,15-0,20 mm), superfina (0,21-0,30 mm), fina (0,30-0,50 mm) e média (1,19-0,50 mm), umedecidas com 0,5; 1,0; 1,5; e 2,0 vezes o seu peso em água. O teste de germinação foi conduzido a 20-30 °C, avaliando-se a primeira contagem do teste aos sete dias após a semeadura e, semanalmente, a germinação (plântulas normais) até os 35 dias, quando foram contabilizadas, também, as plântulas anormais e as sementes mortas. Calcularam-se o tempo médio e frequência relativa de germinação. A vermiculita mícron umedecida com uma vez o seu peso em água apresentou o melhor desempenho como substrato para o teste de germinação de sementes de palmeira-real-australiana, por possibilitar a máxima germinação das sementes (90%) e velocidade de germinação, demandando tempo médio de 15,3 dias nesse processo.

Vermiculite is a substrate used in seedling production of forest species and can be an option for the germination test of ipe seeds. The objective of this research was to evaluate the use of vermiculite in the germination test of Tabebuia chrysotricha seeds and to stablish appropriate particle size and moisture for this substrate. Four replications of 20 seeds were sown in soil, sand, paper rolls, and vermiculite with different particle sizes: micron (0.15-0.20 mm), superthin (0.21-0.30 mm), thin (0.30-0.50 mm) and medium (0.50-1.19 mm) moistened with water equivalent to 0.5, 1.0, 1.5 and 2.0 times the weight of the dry subtrate. Germination test was carried out at 30 degrees C. The percentage of normal seedlings was obtained seven days after sowing (first count) and then weekly until 28 days, when abnormal seedlings and dead seeds were also counted. It was concluded that the germination test of ipe seeds can be carried out with thin or medium vermiculite moistened with water equivalent to 1.5 times the dry weight of the substrate. These treatments resulted in higher and faster germination (21 days).

Em um segmento de vertente com substrato de arenito em contato com basalto, regionalmente muito freqüente, pretendeu-se não só relacionar as superfícies geomórficas com os atributos físicos, químicos e mineralógicos dos Latossolos nelas encontrados, mas também testar métodos geoestatísticos para localização de limites dessas superfícies. Usando critérios geomorfológicos, três superfícies foram identificadas e topograficamente caracterizadas. Os solos foram amostrados, a intervalos regulares de 25 m, na profundidade de 0,6 a 0,8 m (topo do horizonte B), em uma transeção de 1.700 m perfazendo 109 pontos. Nas amostras, foram analisados: densidade de partículas, granulometria, CTC do solo, CTC da argila, Fe total da argila (ataque por H2SO4) e óxidos de Fe livres (por dissolução seletiva). A fração argila desferrificada foi analisada por difração de raios X. Com base na estratigrafia e variações do relevo local, foram identificadas e diferenciadas, no campo, três superfícies geomórficas. Analisaram-se também o perfil altimétrico e o modelo de elevação digital do terreno. Observou-se que as três diferentes superfícies estão bem relacionadas com os atributos físicos, químicos e mineralógicos dos seus respectivos solos. Na parte inferior desta vertente, superfície mais recente e sobre basalto, em Latossolo Vermelho eutroférrico típico, foram encontradas as maiores variabilidades da declividade, da argila e de Fe. As variações da inclinação do terreno, quando analisadas sistematicamente pelo split moving windows dissimilarity analysis (análise estatística de dissimilaridade, em segmentos móveis), mostraram que este método estatístico pode ser usado para ajudar a localizar os limites entre superfícies geomórficas. As variações dos solos da transeção, e arredores, mostraram-se relacionadas com idade, inclinação do terreno e litologia. O trabalho geomórfico detalhado forneceu importantes informações para subsidiar os trabalhos de levantamento de solos e de pedogênese.

A condução das operações de preparo de forma inadequada ocasiona sérios problemas de conservação do solo, destacando-se a compactação, que acarreta a redução do espaço poroso, principalmente dos macroporos, e altera os atributos físico-hídricos. Este trabalho teve como objetivo verificar a influência dos diferentes sistemas e tempos de adoção de manejos em Latossolo Vermelho de Jaboticabal, Estado de São Paulo, por meio da densidade máxima, e correlacioná-la com a produtividade da soja, a densidade relativa e a umidade crítica de compactação. O delineamento experimental foi o inteiramente casualizado com parcelas subdivididas (cinco sistemas de uso e três camadas), com quatro repetições. Os cinco sistemas de uso foram: plantio direto por cino anos (SPD5), plantio direto por sete anos (SPD7), plantio direto por nove anos (SPD9), preparo convencional (SPC) e uma área adjacente de mata nativa (MN). As camadas do solo avaliadas foram as de 0-0,10, 0,10-0,20 e 0,20-0,30 m, nas quais foram determinados a densidade máxima do solo (Ds máx), a umidade crítica de compactação (Ugc), a densidade relativa do solo (Dsr), a composição granulométrica, a porosidade e o teor de matéria orgânica do solo. Os resultados mostraram que o comportamento das curvas de compactação do solo foi o mesmo em todas as camadas dos diferentes manejos e que os teores de matéria orgânica não justificaram as pequenas alterações da Ds máx. Para o Latossolo Vermelho, as operações mecanizadas nos sistemas de manejo podem ser executadas na faixa de 0,13 a 0,19 kg kg-1 de umidade sem causar degradação física. Verificou-se que a Dsr ótima e a umidade crítica de compactação foram de 0,86 e 0,15 kg kg-1, respectivamente, embora os diferentes sistemas e tempos de adoção de manejo tenham apresentado comportamento semelhante quanto à produtividade da soja.

O objetivo deste trabalho foi estudar a influência das superfícies geomórficas na variação espacial da perda de solo por erosão na região de Pereira Barreto, São Paulo (SP). Os solos foram amostrados nos pontos de cruzamento de uma malha, georreferenciados, com intervalos de 350m, na profundidade de 0,0-0,2m, totalizando 67 pontos. Foram feitas determinações da composição granulométrica e do conteúdo de matéria orgânica. Foram avaliados os fatores de erosão locais, tais como erosividade (R), erodibilidade (K), fator topográfico (LS), uso e manejo (C), práticas conservacionistas (P), potencial natural de erosão (PNE), perda de solo com e sem práticas conservacionistas (A e *A), tolerância de perda de solo (T) e risco de erosão (RE). As variáveis A, PNE e RE apresentaram forte correlação espacial com o fator topográfico (LS), indicando a forte relação do relevo sobre os fatores de erosão. As perdas de solo (A e *A) apresentaram comportamento coerente com a conceituação de superfícies geomórficas, evidenciando as relações de dependência do processo erosivo do solo aos ambientes geomórficos.
