Background: Duodenal stenting has become a broadly accepted first line of treatment for patients with advanced malignant gastroduodenal obstruction as these patients are difficult to treat and are poor surgical candidates. Aims: To document duodenal stent performance for palliative management of malignant gastroduodenal obstruction. Methods: Multicentre, single arm, prospective registry documenting peroral endoscopic duodenal stenting procedures in 202 patients. Results: Technical success achieved in 98% (CI. 95%, 99%) of stent placements. Increase of Gastric Outlet Obstruction Score by at least 1 point compared to baseline was achieved in 91% (CI, 86%, 95%) of patients persisting for a median of 184 days (CI, 109, 266). By day 5 (CI, 4,6) after stent placement, 50% of patients experienced a score increase of at least 1 point. Improvement from 14% of patients at baseline tolerating soft solids or low residue/normal diet to 84% at 15 days, 86% at 30 days, 81% at 90 days, 79% at 180 days, and 70% at 270 days. Complications included stent ingrowth and/or overgrowth (12.4%), transient periprocedural symptoms (3%), bleeding (3%), stent migration (1.5%), and perforation (0.5%). Conclusions: Safety and effectiveness of duodenal stenting for palliation of malignant gastroduodenal obstruction was confirmed in the largest international prospective series to date. The ALICE experiment at the LHC has studied J/psi production at mid-rapidity in pp collisions at root s = 7 TeV through its electron pair decay on a data sample corresponding to an integrated luminosity L-int = 5.6 nb(-1). The fraction of J/psi from the decay of long-lived beauty hadrons was determined for J/psi candidates with transverse momentum p(t) > 1,3 GeV/c and rapidity vertical bar y vertical bar < 0.9. The cross section for prompt J/psi mesons, i.e. directly produced J/psi and prompt decays of heavier charmonium states such as the psi(2S) and chi(c) resonances, is sigma(prompt J/psi) (p(t) > 1.3 GeV/c, vertical bar y vertical bar < 0.9) = 8.3 +/- 0.8(stat.) +/- 1.1 (syst.)(-1.4)(+1.5) (syst. pol.) mu b. The cross section for the production of b-hadrons decaying to J/psi with p(t) > 1.3 GeV/c and vertical bar y vertical bar < 0.9 is a sigma(J/psi <- hB) (p(t) > 1.3 GeV/c, vertical bar y vertical bar < 0.9) = 1.46 +/- 0.38 (stat.)(-0.32)(+0.26) (syst.) mu b. The results are compared to QCD model predictions. The shape of the p(t) and y distributions of b-quarks predicted by perturbative QCD model calculations are used to extrapolate the measured cross section to derive the b (b) over bar pair total cross section and d sigma/dy at mid-rapidity.

The structural and chemical characteristics of fly ash from coal-fired mineral and fly ash zeolitized are similar to those of zeolites. Urea was added with these materials in the proportions of urea: fly ashes of 100:10, 100:20, 100:50, 100:100, with a control containing just urea. These treatments were applied in soil surface and the experimental design was a randomized block with clay and sandy soil. Nitrogen losses by ammonia volatilization and the chemical characteristics of soil fertility were evaluated. In sandy soil there was reduction of ammonia volatilization for the proportions of 100:10 and 100:20, while fly ash zeolitized and fly ash had no difference.

BACKGROUND AND PURPOSE Independent studies in experimental models of Trypanosoma cruzi appointed different roles for endothelin-1 (ET-1) and bradykinin (BK) in the immunopathogenesis of Chagas disease. Here, we addressed the hypothesis that pathogenic outcome is influenced by functional interplay between endothelin receptors (ETAR and ETBR) and bradykinin B2 receptors (B2R). EXPERIMENTAL APPROACH Intravital microscopy was used to determine whether ETR/B2R drives the accumulation of rhodamine-labelled leucocytes in the hamster cheek pouch (HCP). Inflammatory oedema was measured in the infected BALB/c paw of mice. Parasite invasion was assessed in CHO over-expressing ETRs, mouse cardiomyocytes, endothelium (human umbilical vein endothelial cells) or smooth muscle cells (HSMCs), in the presence/absence of antagonists of B2R (HOE-140), ETAR (BQ-123) and ETBR (BQ-788), specific IgG antibodies to each GPCRs; cholesterol or calcium-depleting drugs. RNA interference (ETAR or ETBR genes) in parasite infectivity was investigated in HSMCs. KEY RESULTS BQ-123, BQ-788 and HOE-140 reduced leucocyte accumulation in HCP topically exposed to trypomastigotes and blocked inflammatory oedema in infected mice. Acting synergistically, ETAR and ETBR antagonists reduced parasite invasion of HSMCs to the same extent as HOE-140. Exogenous ET-1 potentiated T. cruzi uptake by HSMCs via ETRs/B2R, whereas RNA interference of ETAR and ETBR genes conversely reduced parasite internalization. ETRs/B2R-driven infection in HSMCs was reduced in HSMC pretreated with methyl-beta-cyclodextrin, a cholesterol-depleting drug, or in thapsigargin-or verapamil-treated target cells. CONCLUSIONS AND IMPLICATIONS Our findings suggest that plasma leakage, a neutrophil-driven inflammatory response evoked by trypomastigotes via the kinin/endothelin pathways, may offer a window of opportunity for enhanced parasite invasion of cardiovascular cells.

Abstract Findings We set out to analyse the gene expression profile of pre-osteoblastic C2C12 cells during osteodifferentiation induced by both rhBMP2 and rhBMP7 using DNA microarrays. Induced and repressed genes were intercepted, resulting in 1,318 induced genes and 704 repressed genes by both rhBMP2 and rhBMP7. We selected and validated, by RT-qPCR, 24 genes which were upregulated by rhBMP2 and rhBMP7; of these, 13 are related to transcription (Runx2, Dlx1, Dlx2, Dlx5, Id1, Id2, Id3, Fkhr1, Osx, Hoxc8, Glis1, Glis3 and Cfdp1), four are associated with cell signalling pathways (Lrp6, Dvl1, Ecsit and PKCδ) and seven are associated with the extracellular matrix (Ltbp2, Grn, Postn, Plod1, BMP1, Htra1 and IGFBP-rP10). The novel identified genes include: Hoxc8, Glis1, Glis3, Ecsit, PKCδ, LrP6, Dvl1, Grn, BMP1, Ltbp2, Plod1, Htra1 and IGFBP-rP10. Background BMPs (bone morphogenetic proteins) are members of the TGFβ (transforming growth factor-β) super-family of proteins, which regulate growth and differentiation of different cell types in various tissues, and play a critical role in the differentiation of mesenchymal cells into osteoblasts. In particular, rhBMP2 and rhBMP7 promote osteoinduction in vitro and in vivo, and both proteins are therapeutically applied in orthopaedics and dentistry. Conclusion Using DNA microarrays and RT-qPCR, we identified both previously known and novel genes which are upregulated by rhBMP2 and rhBMP7 during the onset of osteoblastic transdifferentiation of pre-myoblastic C2C12 cells. Subsequent studies of these genes in C2C12 and mesenchymal or pre-osteoblastic cells should reveal more details about their role during this type of cellular differentiation induced by BMP2 or BMP7. These studies are relevant to better understanding the molecular mechanisms underlying osteoblastic differentiation and bone repair.

Abstract Background Bone fractures and loss represent significant costs for the public health system and often affect the patients quality of life, therefore, understanding the molecular basis for bone regeneration is essential. Cytokines, such as IL-6, IL-10 and TNFα, secreted by inflammatory cells at the lesion site, at the very beginning of the repair process, act as chemotactic factors for mesenchymal stem cells, which proliferate and differentiate into osteoblasts through the autocrine and paracrine action of bone morphogenetic proteins (BMPs), mainly BMP-2. Although it is known that BMP-2 binds to ActRI/BMPR and activates the SMAD 1/5/8 downstream effectors, little is known about the intracellular mechanisms participating in osteoblastic differentiation. We assessed differences in the phosphorylation status of different cellular proteins upon BMP-2 osteogenic induction of isolated murine skin mesenchymal stem cells using Triplex Stable Isotope Dimethyl Labeling coupled with LC/MS. Results From 150 μg of starting material, 2,264 proteins were identified and quantified at five different time points, 235 of which are differentially phosphorylated. Kinase motif analysis showed that several substrates display phosphorylation sites for Casein Kinase, p38, CDK and JNK. Gene ontology analysis showed an increase in biological processes related with signaling and differentiation at early time points after BMP2 induction. Moreover, proteins involved in cytoskeleton rearrangement, Wnt and Ras pathways were found to be differentially phosphorylated during all timepoints studied. Conclusions Taken together, these data, allow new insights on the intracellular substrates which are phosphorylated early on during differentiation to BMP2-driven osteoblastic differentiation of skin-derived mesenchymal stem cells.

Cancer är en av de sjukdomar som ökar mest för tillfället. En av orsakerna till ökningen är att medellivslängden blivit högre. Den ökade medellivslängden är något positivt i sig, men det innebär också att sjukdomen hinner drabba fler människor. Cancer är en sjukdom som påverkar många personer direkt eller indirekt och det är inte bara de som har cancer som är sjuka, utan hela familjen blir sjuk. Syftet med denna fallstudie var att belysa en persons upplevelse av att vara närstående till en svårt sjuk familjemedlem. Metoden som användes var manifest innehållsanalys av en skönlitterär självbiografisk bok, skriven av Conny Palmqvist med titeln ”Hejdå, allihopa!”. Analysen resulterade i fyra kategorier; maktlöshet, relationer, kärlek och sorg. Det är svårt att stå bredvid och se på när en familjemedlem sakta tynar bort och att inte kunna göra något för att förhindra detta. Att som patient känna kärlek och närhet är en förutsättning för att kunna känna hopp och orka kämpa. Efter att ha analyserat boken har vi fått en ökad förståelse för den närståendes upplevelse vid palliativ vård. Att kunna bemöta och ge god omvårdnad till patienter och närstående anser vi är en förutsättning vid arbete inom vård.

Membrane proteins are a large and important class of proteins. They are responsible for several of the key functions in a living cell, e.g. transport of nutrients and ions, cell-cell signaling, and cell-cell adhesion. Despite their importance it has not been possible to study their structure and organization in much detail because of the difficulty to obtain 3D structures. In this thesis theoretical studies of membrane protein sequences and structures have been carried out by analyzing existing experimental data. The data comes from several sources including sequence databases, genome sequencing projects, and 3D structures. Prediction of the membrane spanning regions by hydrophobicity analysis is a key technique used in several of the studies. A novel method for this is also presented and compared to other methods. The primary questions addressed in the thesis are: What properties are common to all membrane proteins? What is the overall architecture of a membrane protein? What properties govern the integration into the membrane? How many membrane proteins are there and how are they distributed in different organisms? Several of the findings have now been backed up by experiments. An analysis of the large family of G-protein coupled receptors pinpoints differences in length and amino acid composition of loops between proteins with and without a signal peptide and also differences between extra- and intracellular loops. Known 3D structures of membrane proteins have been studied in terms of hydrophobicity, distribution of secondary structure and amino acid types, position specific residue variability, and differences between loops and membrane spanning regions. An analysis of several fully and partially sequenced genomes from eukaryotes, prokaryotes, and archaea has been carried out. Several differences in the membrane protein content between organisms were found, the most important being the total number of membrane proteins and the distribution of membrane proteins with a given number of transmembrane segments. Of the properties that were found to be similar in all organisms, the most obvious is the bias in the distribution of positive charges between the extra- and intracellular loops. Finally, an analysis of homologues to membrane proteins with known topology uncovered two related, multi-spanning proteins with opposite predicted orientations. The predicted topologies were verified experimentally, providing a first example of "divergent topology evolution".

Then there is a part of the talk about the agreed facts of climate change, and what this means for us all. The end of the talk brings together these two sections in a summary.

Die künstlichen Elemente Rutherfordium, Dubnium und Seaborgium werden an Schwerionenbeschleunigern erzeugt. Ihre chemische Untersuchung erfolgt mit computergesteuerten Apparaturen, z.B. ARCA (Automated Rapid Chemistry Apparatus). Ziel der Untersuchungen ist die Einordnung derElemente ins Periodensystem. Im einzelnen beschreibt die vorliegende Arbeit Experimente zur Fluorid- und Chlorid-Komplexierung von Rutherfordium und Dubnium, zum ionischen Zustand von Seaborgium in Nitratlösungen, zur Komplexierung von Seaborgium mit alpha-HIB sowie Vorexperimente zur Reduktion von Seaborgium.

Zusammenfassung Die vorliegenden Arbeit beschäftigt sich mit den Synthesen und Charakterisierungen multifunktioneller, Arylamin-haltiger Polymere, welche sich als photorefraktive(PR)-Materialien eignen. Die Glastemperaturen (Tg) der angestrebten Materialien liegen deutlich über Raumtemperatur, um so den Pockels-Mechanismus zum Aufbau des PR-Effektes zu favorisieren. Hierzu sind zwei synthetische Konzepte, basierend auf Maleinimid-Methylvinylisocyanat-Reaktiv-Polymeren und Triphenylamin-haltigen Polymeren, entwickelt worden. Im Rahmen des Reaktiv-Polymer-Konzeptes konnten PR-Materialien mit den bisher größten Beugungs-Effizienzen sowie den schnellsten Ansprechzeiten für multifunktionelle hoch-Tg-Polymere dargestellt werden. Hierfür wurden Maleinimid-Methylvinylisocyanat-Reaktiv-Polymere synthetisiert welche an der Imid-Position über Spacer-Gruppen mit Carbazol-Einheiten funktionalisiert sind. Die Tg´s der Polymere konnten zwischen 60°C und 194°C eingestellt werden. Die Isocyanat-Gruppen wurden dann polymeranlog mit hydroxyalkyl-funktionalisierten Chromophoren umgesetzt. Die Kinetik des PR-Effektes dieser Materialien wird durch die Ladungsträger-Beweglichkeiten in den Proben bestimmt. Eine Steigerung der Farbstoff-Konzentrationen erhöht die PR-Leistungen der Materialien, behindert jedoch deren Kinetik.Das Triphenylamin-Polymer-Konzept verwendet Triphenylamine als Lochleiter. Hierfür wurden die radikalischen Polymerisations-Verhalten der Monomere p-Diphenylaminostyrol (DPAS) und erstmals p-Ditoluylaminostyrol (DTAS) untersucht. Die Monomere wurden durch spontane, freie und kontrollierte radikalische Verfahren polymerisiert. Mittels eines TEMPO-Derivates gelang der Aufbau von Block-Copolymeren. Poly-DPAS konnte, im Gegensatz zu Poly-DTAS, polymeranlog tricyanovinyliert werden. Dadurch lassen sich PDPAS-block-PTPAS-Copolymere selektiv im PDPAS-Block tricyanovinylieren. Diese Materialien weisen eine Tendenzen zur Mikro-Phasen-Separation auf.Die Strukturierung von PDPAS konnte durch Photo-Polymerisation mit einer Auflösung von wenigen mm demonstriert werden. Carbazol und Triphenylamin-haltige Materialien wurden mittels Cyclo-Voltametrie untersucht.

Das experimentelle Studium der 1966 von Gerasimov, Drell undHearn unabhängig voneinander aufgestellten und als GDH-SummenregelbezeichnetenRelation macht die Vermessung totalerPhotoabsorptionswirkungsquerschnitte von zirkular polarisierten Photonen an longitudinalpolarisierten Nukleonen über einen weiten Energiebereich notwendig. Die im Sommer1998 erfolgte Messung am Mainzer Mikrotron stellt das erste derartigeExperiment mit reellen Photonen zur Messung des GDH-Integrals am Protondar. Die Verwendung eines Frozen-Spin-Butanoltargets, das eingesetzt wurde, umeinen möglichst hohen Proton-Polarisationsgrad zu erreichen, hat diezusätzliche experimentelle Schwierigkeit zur Folge, daß die imButanoltarget enthaltenen Kohlenstoffkerne ebenfalls Reaktionsprodukte liefern, diezusammen mit den am Proton erzeugten nachgewiesen werden.Ziel der Arbeit war die Bestimmung von Wirkungsquerschnittenam freien Proton aus Messungen an einem komplexen Target (CH2) wie esbeim polarisiertenTarget vorliegt. Die hierzu durchgeführten Pilotexperimentedienten neben der Entwicklung von Methoden zur Reaktionsidentifikation auchder Eichung des Detektorsystems. Durch die Reproduktion der schon bekanntenund vermessenen unpolarisierten differentiellen und totalenEin-Pion-Wirkungsquerschnitte am Proton (gamma p -> p pi0 und gamma p -> n pi+), die bis zueiner Photonenergievon etwa 400 MeV den Hauptbeitrag zum GDH-Integralausmachen, konnte gezeigt werden, daß eine Separation der Wasserstoff- vonKohlenstoffereignissen möglich ist. Die notwendigen Techniken hierzu wurden imRahmen dieser Arbeit zu einem allgemein nutzbaren Werkzeug entwickelt.Weiterhin konnte gezeigt werden, daß der vom Kohlenstoffstammende Anteil der Reaktionen keine Helizitätsabhängigkeit besitzt. Unterdieser Voraussetzung reduziert sich die Bestimmung der helizitätsabhängigenWirkungsquerschnittsdifferenz auf eine einfacheDifferenzbildung. Aus den erhaltenen Ergebnissen der intensiven Analyse von Daten, diemit einem unpolarisierten Target erhalten wurden, konnten so schnellerste Resultate für Messungen, die mit dem polarisierten Frozen-Spin-Targetaufgenommen wurden, geliefert werden. Es zeigt sich, daß sich dieseersten Resultate für polarisierte differentielle und totale (gammaN)-Wirkungsquerschnitte im Delta-Bereich in guter Übereinstimmung mit theoretischenAnalysen befinden.
