H3K4me3 is a histone modification that accumulates at the transcription-start site (TSS) of active genes and is known to be important for transcription activation. The way in which H3K4me3 is regulated at TSS and the actual molecular basis of its contribution to transcription remain largely unanswered. To address these questions, we have analyzed the contribution of dKDM5/LID, the main H3K4me3 demethylase in Drosophila, to the regulation of the pattern of H3K4me3. ChIP-seq results show that, at developmental genes, dKDM5/LID localizes at TSS and regulates H3K4me3. dKDM5/LID target genes are highly transcribed and enriched in active RNApol II and H3K36me3, suggesting a positive contribution to transcription. Expression-profiling show that, though weakly, dKDM5/LID target genes are significantly downregulated upon dKDM5/LID depletion. Furthermore, dKDM5/LID depletion results in decreased RNApol II occupancy, particularly by the promoter-proximal Pol lloser5 form. Our results also show that ASH2, an evolutionarily conserved factor that locates at TSS and is required for H3K4me3, binds and positively regulates dKDM5/LID target genes. However, dKDM5/LID and ASH2 do not bind simultaneously and recognize different chromatin states, enriched in H3K4me3 and not, respectively. These results indicate that, at developmental genes, dKDM5/LID and ASH2 coordinately regulate H3K4me3 at TSS and that this dynamic regulation contributes to transcription.

Telomerase is an RNA-dependent DNA polymerase that synthesizes telomeric DNA. Its activity is not detectable in most somatic cells but it is reactivated during tumorigenesis. In most cancers, the combination of hTERT hypermethylation and hypomethylation of a short promoter region is permissive for low-level hTERT transcription. Activated and malignant lymphocytes express high telomerase activity, through a mechanism that seems methylation-independent. The aim of this study was to determine which mechanism is involved in the enhanced expression of hTERT in lymphoid cells. Our data confirm that in B cells, some T cell lymphomas and non-neoplastic lymph nodes, the hTERT promoter is unmethylated. Binding sites for the B cell-specific transcription factor PAX5 were identified downstream of the ATG translational start site through EMSA and ChIP experiments. ChIP assays indicated that the transcriptional activation of hTERT by PAX5 does not involve repression of CTCF binding. In a B cell lymphoma cell line, siRNA-induced knockdown of PAX5 expression repressed hTERT transcription. Moreover, ectopic expression of PAX5 in a telomerase-negative normal fibroblast cell line was found to be sufficient to activate hTERT expression. These data show that activation of hTERT in telomerase-positive B cells is due to a methylation-independent mechanism in which PAX5 plays an important role.

Cannabis use has been related to an elevated psychosis risk and attenuated cognitive functioning. Cannabis-related cognitive impairments are also observed in populations along the psychosis dimension. We here investigated whether a potential behavioural marker of the psychosis dimension (attenuated functional hemispheric asymmetry) is even further attenuated in individuals using cannabis (CU) versus those not using cannabis (nCU). We tested 29 patients with first episode psychosis (FEP; 11 CU) and 90 healthy controls (38 CU) on lateralized lexical decisions assessing left hemisphere language dominance. In patients, psychotic symptoms were assessed (PANSS). In controls, self-reported schizotypy was assessed (O-LIFE questionnaire). Results indicated that nCU FEP patients had a relative reduced hemispheric asymmetry, as did controls with increasing cognitive disorganisation scores, in particular when belonging to the group of nCU controls. Positive, disorganised and negative PANSS scores in patients and negative and positive schizotypy in controls were unrelated to hemispheric asymmetry. These findings suggest that cannabis use balances rather than exacerbates uncommon hemispheric laterality patterns. Moreover, in healthy populations, the potential stabilisation of typical hemispheric asymmetry in CU might be most relevant to individuals with elevated cognitive disorganisation. We discuss the potential beneficial and harmful effects of cannabis use along the psychosis dimension together with propositions for future studies that should account for the mediating role of additional substances (e.g. nicotine), cannabis composition (e.g. cannabidiol content), and individual differences (e.g. physical health, or absence of significant polysubstance use).

Common variants at only two loci, FTO and MC4R, have been reproducibly associated with body mass index (BMI) in humans. To identify additional loci, we conducted meta-analysis of 15 genome-wide association studies for BMI (n > 32,000) and followed up top signals in 14 additional cohorts (n > 59,000). We strongly confirm FTO and MC4R and identify six additional loci (P < 5 x 10(-8)): TMEM18, KCTD15, GNPDA2, SH2B1, MTCH2 and NEGR1 (where a 45-kb deletion polymorphism is a candidate causal variant). Several of the likely causal genes are highly expressed or known to act in the central nervous system (CNS), emphasizing, as in rare monogenic forms of obesity, the role of the CNS in predisposition to obesity.

Approaches exploiting trait distribution extremes may be used to identify loci associated with common traits, but it is unknown whether these loci are generalizable to the broader population. In a genome-wide search for loci associated with the upper versus the lower 5th percentiles of body mass index, height and waist-to-hip ratio, as well as clinical classes of obesity, including up to 263,407 individuals of European ancestry, we identified 4 new loci (IGFBP4, H6PD, RSRC1 and PPP2R2A) influencing height detected in the distribution tails and 7 new loci (HNF4G, RPTOR, GNAT2, MRPS33P4, ADCY9, HS6ST3 and ZZZ3) for clinical classes of obesity. Further, we find a large overlap in genetic structure and the distribution of variants between traits based on extremes and the general population and little etiological heterogeneity between obesity subgroups.

Qualitative differences in strategy selection during foraging in a partially baited maze were assessed in young and old rats. The baited and non-baited arms were at a fixed position in space and marked by a specific olfactory cue. The senescent rats did more re-entries during the first four-trial block but were more rapid than the young rats in selecting the reinforced arms during the first visits. Dissociation between the olfactory spatial cue reference by rotating the maze revealed that only few old subjects relied on olfactory cues to select the baited arms and the remainder relied mainly on the visuo-spatial cues.

Background: Air pollution has become an important issue worldwide due to its adverse health effects. Among the different air contaminants, volatile organic compounds (VOCs) are liquids or solids with a high vapor pressure at room temperature that are extremely dangerous for human health. Removal of these compounds can be achieved using nanomaterials with tailored properties such as carbon nanotubes. Methods: Vertically-aligned multiwall carbon nanotubes (CNTs) were successfully grown on quartz filters by means of plasma enhanced chemical vapor deposition (PECVD). Furthermore, a plasma treatment was performed in order to modify the surface properties of the CNTs. The adsorption/desorption processes of three chlorinated compounds (trichloroethylene, 1,2-dichlorobenzene and chloroform) on the CNTs were studied using mass spectrometry measurements with a residual gas analyzer. Results: The adsorption capability of the CNTs increased after functionalization of their surface with a water plasma treatment. In addition, it was found that the presence of aromatic rings, water solubility and polarity of the VOCs play an important role on the adsorption/desorption kinetics at the CNTs surface. Conclusions: This study demonstrates the applicability of CNTs deposited on quartz filters for the removal or selective detection of volatile organic compounds (VOCs). The presence of aromatic rings in VOCs results in π -stacking interactions with a significant increase of their adsorption. On the other hand, it was found that CNTs surface interactions increase with water solubility and polarity of the VOC.

Avant-propos : De nombreuses études ont été réalisées sur les inégalités factuelles des structures sociales, comprenant aussi bien l'aspect statique de la stratification sociale que l'aspect dynamique de la mobilité sociale (voir par exemple Levy et Suter, 2002, Lemel, 1991, Erikson et Goldthorpe, 1992, Esping-Andersen, 1993). Par contre, les recherches portant sur la perception, par les acteurs, des inégalités des structures sociales sont en comparaison peu nombreuses en ce qui concerne les représentations de la stratification sociale (Levy et al., 1997, Lorenzi-Cioldi et Joye, 1988, Coxon, Davies et Jones, 1986, Zwicky, 1989) et presque inexistantes dans le cas des représentations de la mobilité sociale (Attias-Donfut et Wolff, 2001). La présente recherche se propose d'étudier simultanément la perception de la stratification sociale et de la mobilité sociale intragénérationnelle par les acteurs en intégrant le caractère multidimensionnel du système d'inégalités. Elle défend la thèse fondamentale d'une double origine des inégalités perçues, qui participeraient à la fois d'aspects macrosociaux et mésosociaux de la stratification sociale, les premiers portant sur la structure sociale dans son ensemble, les seconds sur une partie seulement de celle-ci (voir par exemple Kelley et Evans, 1995, Levy, 2002). Dans une perspective systémique, on se trouverait, à côté de la structure macrosociale, en présence de sous-systèmes mésosociaux, de portée restreinte. La perception de la stratification sociale dépendrait alors du cadre de référence adopté par les acteurs, selon qu'il porte sur le système dans son ensemble ou sur un sous-système. Un des objectifs de cette recherche sera d'établir que la pertinence des cadres de référence macrosocial et mésosocial est étroitement liée à la lecture statique ou dynamique de la stratification sociale. Dans le cas statique, celui du positionnement, les représentations sociales s'articuleraient autour des inégalités macrosociales, tenant compte du système dans son ensemble, tandis que dans le cas dynamique, celui de la mobilité ou de l'évolution du positionnement, les inégalités mésosociales, propres aux sous-systèmes, l'emporteraient. D'une part, la perception du positionnement social dépendrait de l'insertion de l'acteur dans la structure sociale, comprise dans son ensemble, et reproduirait les inégalités factuelles macrosociales, telles qu'elles apparaissent par exemple au travers des catégories socioprofessionnelles. D'autre part, la perception du parcours de mobilité ? conservation, amélioration ou dégradation de la position perçue ? resterait indépendante des changements macrosociaux de l'insertion, mais relèverait avant tout de déterminants propres à l'environnement social immédiat de l'acteur. L'environnement de l'acteur, en tant qu'il s'inscrit dans une partie restreinte de la structure sociale, permettrait de saisir les inégalités mésosociales. L'expérience, par les acteurs, de ces deux aspects de la structure sociale conduirait à la mise en place de deux types d'inégalités perçues irréductibles les unes aux autres dans la mesure où le système macrosocial et les sous-systèmes mésosociaux présentent une certaine autonomie. Cette autonomie peut être vue d'une part en rapport avec l'importance propre des organisations de nature mésosociale - en particulier les entreprises - dans les sociétés contemporaines (Sainsaulieu et Segrestin, 1986, Perrow, 1991), d'autre part en relation avec l'hétérogénéité que ces dernières induisent en termes de segmentation du marché de l'emploi (Baron et Bielby, 1980). Dans une large mesure, les organisations intermédiaires se distinguent ainsi de la structure sociale prise dans son ensemble: plutôt que de reproduire les inégalités macrosociales, elles constitueraient des systèmes d'inégalités indépendants, notamment quant à la régulation des parcours professionnels (Bertaux, 1977). Ainsi, la perception de la structure sociale ne se réduirait pas aux seuls facteurs macrosociaux, mais dépendrait, en l'absence d'un modèle d'organisation mésosocial unique, de la diversité des structures intermédiaires. On peut d'ailleurs supposer que la prise en compte des organisations mésosociales est susceptible de pallier la faiblesse des explications classiques en termes macrosociologiques, relevées par les tenants des thèses avançant le déclin du pouvoir structurant de la stratification sociale ou du travail (voir Levy, 2002 et, sur les thèses citées, par exemple Beck, 1983, Matthes, 1983, Berger et Hradil, 1990, Clark et Lipset, 1991). En effet, dans la mesure où l'acteur serait plus souvent confronté aux structures de son environnement social immédiat plutôt qu'à la structure sociale dans son ensemble, la perception pourrait dépendre en premier lieu de facteurs mésosociaux, susceptibles de supplanter ou, à tout le moins, d'atténuer l'effet des facteurs macrosociaux. Une telle approche permet de conserver une lecture structurelle de la perception du positionnement en enrichissant la relation classique entre structure macrosociale et acteur d'une composante mésosociologique, évitant ainsi le recours à une explication culturelle ad hoc Dès lors, la principale question de recherche s'adresse au lien entre structure sociale factuelle et structure sociale perçue. Dans la perspective statique du positionnement, l'effet des structures mésosociales serait tel qu'il se superposerait à la détermination macrosociale de la perception, sans pour autant subvertir la hiérarchie des positions induites par les catégories socioprofessionnelles. Dans la perspective dynamique, en revanche, les changements liés à l'insertion mésosociale peuvent l'emporter sur l'immobilité ou la mobilité définies en termes macrosociologiques. D'une part, en supposant que les plans mésosocial et macrosocial agissent de manière plus ou moins autonome sur la perception, l'amélioration, la conservation ou la dégradation de la position ne coïncide pas nécessairement selon ces deux plans. D'autre part, l'ampleur de la mobilité perçue due à l'écart entre le positionnement mésosocial passé et actuel peut dépasser celle qui est liée à la mobilité macrosociale, surtout si cette dernière est de faible distance. Le passage de la perspective statique à la perspective dynamique peut dès lors être vu comme un moyen de faire apparaître le rôle fondamental joué par les structures mésosociales au sein de la stratification sociale. L'orientation de la recherche consistera d'abord à mettre en évidence, par-delà les différences macrosociales des représentations des positions professionnelles, les variations de la perception au sein des catégories socioprofessionnelles. Ces étapes montreront, à différents égards, que les représentations se singularisent en relation avec l'insertion mésosociale de l'acteur. On verra également que la perception de la mobilité échappe à une détermination macrosociale, mais qu'elle présente une cohérence mésosociale certaine. Ces résultats, insistant sur la prise en compte des structures mésosociales, nous amèneront enfin à un examen systématique des déterminants de la perception du positionnement et du parcours de mobilité, mettant en oeuvre une variété de facteurs explicatifs dépassant un cadre d'analyse purement structurel. La recherche débute par une discussion de la place qui revient à une étude des représentations du parcours professionnel dans le champ des travaux sur la stratification et la mobilité sociale, en particulier sa justification théorique et empirique, et la formulation des hypothèses de recherche (chapitre 1). Elle se poursuit par la présentation de l'échantillonnage et des variables utilisées (chapitre 2). Le traitement des hypothèses de recherche fait l'objet de trois chapitres distincts. Chaque hypothèse s'accompagne, en plus des développements liés à son examen, d'une introduction et d'une conclusion spécifiques. Le premier (chapitre 3) porte sur la perception de la stratification sociale des positions professionnelles, le second (chapitre 4) sur la perception du parcours de mobilité et le troisième (chapitre 5) sur les déterminants sociologiques de la perception des inégalités liées au positionnement et à la mobilité professionnels. Enfin, au traitement des hypothèses fait suite la conclusion de la recherche (chapitre 6).

A study was conducted to determine the possibility of cashew (Anacardium occidentale) cloning by air-layering and influence of IBA (indol-butyric acid) on this process. It was adopted a completely randomized design with 4 treatments, 10 air layers each and 4 replications, reaching 160 air layers. The IBA levels on the treatments were, as follow: 0, 1000, 3000 and 5000 mg.kg-1. It was evaluated: survival, callus and rooting percentage, average number and length of roots. The highest survival rate (67.5%) was registered with no growth regulator and IBA at 1000 mg.kg-1, while the best rooting percentage (82%) referred to 1000 mg.kg-1. In spite of average number and length of roots, the highest results were observed with IBA at 5000 mg.kg-1. IBA concentrations had no influence on cashew air-layering formation.

The work had as objective the physico-chemical characterization of yellow mangosteen fruits. Six samples of 25 fruits were harvested in yellow mangosteen plants of the Active Germoplasm Bank of São Paulo State University and characterized by evaluation of length and width, weight, percentage and number of seeds per fruit, peel and pulp percentage, soluble solid (SS), titratable acidity (TA), vitamin C and SS/TA rate. Yellow mangosteen fruit is an intermediate vitamin C source with an average content 120.33 mg/100g of fresh fruit and has good technological quality.

O trabalho foi desenvolvido com o objetivo de avaliar os efeitos do tempo e ambiente de armazenamento na emergência de plântulas de doviális (Dovyalis caffra), espécie frutífera arbustiva de sementes recalcitrantes. Foi adotado o delineamento experimental inteiramente casualizado, em esquema fatorial 3 x 5, correspondendo às condições ambientais de armazenamento: câmara seca (18&plusmn;2ºC e 60% de UR), em condição ambiente (sem controle) e emmara fria (10&plusmn;2ºC e 70% de UR); e aos períodos de armazenamento 0; 7; 14; 21 e 28 dias. Esses efeitos foram avaliados para o cálculo da percentagem de emergência (%E) e índice de velocidade de emergência (IVE). Os ambientes de armazenamento não exerceram efeito estatístico significativo nas variáveis estudadas, diferentemente dos períodos de armazenamento, onde as mais altas %E e IVE foram observadas ao 14º dia de armazenamento. A partir do 14º dia de armazenamento, ocorreu uma redução na %E e IVE.

No Brasil, existem diversas fruteiras nativas com potencial de exploração comercial, especialmente na Amazônia, local de origem do bacuripari. Neste sentido, realizou-se um trabalho com o objetivo de avaliar a clonagem dessa espécie pelo processo da estaquia, mediante uso de ácido indolil-3-butírico (AIB), em condições de nebulização intermitente. O delineamento experimental foi o inteiramente casualizado, com 5 tratamentos, caracterizados pelas concentrações de AIB (0; 1.000; 3.000; 5.000 e 7.000 mg.L-1), com 4 repetições e 10 estacas por parcela. Foram avaliados na porcentagem de estacas enraizadas, o número médio de folhas e o comprimento de raízes. O uso de AIB não influenciou na porcentagem de enraizamento.

Foram conduzidos dois experimentos com a finalidade de determinar a possibilidade de clonagem da variedade de abacateiro "Duke 7", por alporquia e a influência do AIB (ácido indol-3-butírico) no processo. Experimento 1 - Alporque em plantas - O delineamento experimental utilizado foi o inteiramente casualizado, em esquema fatorial 2L x 4N x 2E, correspondendo à manutenção ou não das plantas à ausência de luz (L), níveis de AIB (N) e tipo de estrutura (E). Nos quatro dias antecedentes à realização da alporquia, 50% das plantas permaneceram na ausência total de luz (L1), e as demais, em condições normais de ripado, 50% de luminosidade (L2). No local anelado, foram aplicadas as concentrações (N) de AIB (ácido indolbutírico): 0; 1.000, 3.000 e 5.000 mg kg-1. O experimento foi realizado em duas estruturas diferenciadas pelo tipo de cobertura: a estrutura um (E1) e a estrutura dois (E2), diferenciadas pela temperatura e intensidade luminosa. Experimento 2- Alporque em plantas adultas após poda drástica - Os alporques foram realizados cinco meses após a poda drástica, quando os ramos possuíam entre 1,5 e 2,0 cm de diâmetro. O delineamento experimental utilizado foi o inteiramente casualizado em esquema fatorial, com 4 tratamentos, caracterizados pelas concentrações de AIB (0; 1.000; 3.000 e 5.000 mg kg-1), com quatro repetições, e cada parcela composta por 10 alporques. Em nenhum dos experimentos, houve enraizamento dos alporques, consequentemente há necessidade de maiores estudos, quanto à clonagem da variedade "Duke 7" para viabilizá-la como porta-enxerto.

BACKGROUND: A few studies have suggested an association between prenatal exposure to methylmercury and decreased heart rate variability (HRV) related to autonomic heart function, but no study has examined this association using baroreflex sensitivity (BRS). In this study we assessed the distribution of BRS and immediate orthostatic hypotension (IOH) in young Seychellois adults and their associations with exposure to prenatal and recent postnatal methylmercury. METHODS: Subjects in the Seychelles Child Development Study (SCDS) main cohort were evaluated at age 19 years. Non-invasive beat-to-beat blood pressure (BP) monitoring (Finapres, Ohmeda) was performed at rest and during active standing in 95 consecutive subjects. Recent postnatal mercury exposure was measured in subjects' hair at the age of 19 years and prenatal exposure in maternal hair grown during pregnancy. BRS was estimated by sequence analysis to identify spontaneous ascending and descending BP ramps. HRV was estimated by the following markers: PNN50 (relative numbers of normal-to-normal intervals which are shorter by more than 50 ms than the immediately following normal-to-normal intervals); rMSSD (root mean of the squared sum of successive interval differences); LF/HF (low frequency/high frequency component ratio); ratio of the mean expiratory/inspiratory RR intervals (EI ratio); and the ratio between the longest RR interval 30 s after active standing and the shortest RR interval at 15 s (Max30/Min15). IOH was estimated by the deepest BP fall within the first 15 s after active standing up. RESULTS: Prenatal MeHg exposures were similar in boys and girls (6.7±4.3, 6.7±3.8 ng/g) but recent postnatal mercury levels were higher in males than females (11.2±5.8 vs 7.9±4.3 ng/g, p=0.003). Markers of autonomic heart rate control were within the normal range (BRS: 24.8±7 ms/mm Hg, PNN50: 24.9±6.8%, rMSSD: 68±22, LF/HF: 0.61±0.28) in both sexes. After standing, 51.4% of subjects had a transient systolic BP drop>40 mm Hg, but only 5.3% reported dizziness or had syncope. Prenatal and recent postnatal MeHg levels, overall, were not associated with BRS, E/I ratio, PNN50, rMSSD, LF/HF ratio, Max30/Min15 ratio, and IOH. CONCLUSIONS: This study provides no support for the hypothesis that prenatal or recent postnatal MeHg exposure from fish consumption is associated with impaired autonomic heart rate control.
