The immune system is able to produce antibodies, which have the capacity to recognize and to bind to foreign molecules or pathogenic organisms. Currently, there are a diversity of diseases that can be treated with antibodies, like immunoglobulins G (IgG). Thereby, the development of cost-efficient processes for their extraction and purification is an area of main interest in biotechnology. Aqueous biphasic systems (ABS) have been investigated for this purpose, once they allow the reduction of costs and the number of steps involved in the process, when compared with conventional methods. Nevertheless, typical ABS have not showed to be selective, resulting in low purification factors and yields. In this context, the addition of ionic liquids (ILs) as adjuvants can be a viable and potential alternative to tailor the selectivity of these systems. In this work, ABS composed of polyethylene glycol (PEG) of different molecular weight, and a biodegradable salt (potassium citrate) using ILs as adjuvants (5 wt%), were studied for the extraction and purification of IgG from a rabbit source. Initially, it was tested the extraction time, the effect on the molecular weight of PEG in a buffer solution of K3C6H5O7/C6H8O7 at pH≈7, and the effect of pH (59) on the yield (YIgG) and extraction efficiency (EEIgG%) of IgG. The best results regarding EEIgG% were achieved with a centrifugation step at 1000 rpm, during 10 min, in order to promote the separation of phases followed by 120 min of equilibrium. This procedure was then applied to the remaining experiments. The results obtained in the study of PEGs with different molecular weights, revealed a high affinity of IgG for the PEG-rich phase, and particularly for PEGs of lower molecular weight (EEIgG% of 96 % with PEG 400). On the other hand, the variation of pH in the buffer solution did not show a significant effect on the EEIgG%. Finally, it was evaluated the influence of the addition of different ILs (5% wt) on the IgG extraction in ABS composed of PEG 400 at pH≈7. In these studies, it was possible to obtain EEIgG% of 100% with the ILs composed of the anions [TOS]-, [CH3CO2]-and Cl-, although the obtained YIgG% were lower than 40%. On the other hand, the ILs composed of the anions Br-, as well as of the cation [C10mim]+, although not leading to EEIgG% of 100%, provide an increase in the YIgG%. ABS composed of PEG, a biodegradable organic salt and ILs as adjuvants, revealed to be an alternative and promising method to purify IgG. However, additional studies are still required in order to reduce the loss of IgG.

The main objective of the present thesis consists on the development of an analytical preconcentration technology for the concomitant extraction and concentration of human pollution tracers from wastewater streams. Due to the outstanding tunable properties of ionic liquids (ILs), aqueous biphasic systems (ABS) composed of ILs can provide higher and more selective extraction efficiencies for a wide range of compounds, being thus a promising alternative to the volatile and hazardous organic solvents (VOCs) typically used. For that purpose, IL-based ABS were employed and adequately characterized as an one-step extraction and concentration technique. The applicability of IL-based ABS was verified by their potential to completely extract and concentrate two representative pharmaceutical pollution tracers, namely caffeine (CAF) and carbamazepine (CBZ), from wastewaters. The low concentration of these persistent pollutants (usually found in μg·dm-3 and ng·dm-3 levels, respectively) by conventional analytical equipment does not permit a proper detection and quantification without a previous concentration step. Preconcentration methods commonly applied are costly, timeconsuming, with irregular recoveries and make use of VOCs. In this work, the ABS composed of the IL tetrabutylammonium chloride ([N4444]Cl) and the salt potassium citrate (K3[C6H5O7]) was investigated while demonstrating to be able to completely extract and concentrate CAF and CBZ, in a single-step, overcoming thus the detection limit of the applied analytical equipment. Finally, the hydrotropic effect responsible for the ability of IL-based ABS to extract and concentrate a wide variety of compounds was also investigated. It was shown that the IL rules the hydrotropic mechanism in the solubility of CAF in aqueous solutions, with an increase in solubility up to 4-fold. Moreover, the proper selection of the IL enables the design of the system that leads to a more enhanced solubility of a given solute in the IL-rich phase, while allowing a better extraction and concentration. IL-based ABS are a promising and more versatile technique, and are straightforwardly envisaged as selective extraction and concentration routes of target micropollutants from wastewater matrices.

Fragaria vesca L., morango silvestre, pertence à família das Rosaceae e é comumente encontrada nas bermas e taludes [1]. Os seus frutos pequenos e de sabor doce podem ser consumidos em fresco como uma fonte de vitamina C, ou em infusões muito utilizadas no tratamento de vários transtornos intestinais [2]. No presente trabalho, os frutos de F. vesca silvestre foram caracterizados em termos de valor nutricional (hidratos de carbono, proteínas, gordura, cinzas e valor energético), teor em fibra alimentar e perfil em ácidos gordos. Para além disso, os frutos e respetivas infusões foram também caracterizados pelo seu conteúdo em açúcares solúveis, ácidos orgânicos, folatos e tocoferóis por técnicas de HPLC acoplada a detectores de índice de refração, fotodíodos e fluorescência, respetivamente, e também pela sua composição mineral avaliada por espectroscopia de absorção atómica. Os hidratos de carbono foram os macronutrientes maioritários nos frutos, seguidos pela gordura total, cinzas e proteínas. Também demonstraram teores elevados em fibra alimentar, maioritariamente fibra solúvel. Os ácidos linolénico (C18:3n3) e γ-linolénico (C18:3n6) foram os ácidos gordos maioritários, havendo uma prevalência de ácidos gordos polinsaturados. Os frutos e as respetivas infusões apresentaram sacarose, seguida da frutose, como sendo os açúcares maioritários. O ácido cítrico foi o ácido orgânico mais abundante em ambas as amostras, enquanto os ácidos oxálico e ascórbico foram detetados nas infusões em concentrações vestigiais. Em termos de microelementos, o manganês foi o mais abundante em ambas as amostras; o potássio e o cálcio foram os macroelementos maioritários encontrados nos frutos e nas suas infusões, respetivamente. Em termos de vitaminas, ambas as amostras apresentaram folatos (vitamina B9) e tocoferóis (vitamina E), sendo o γ-tocoferol a isoforma mais abundante, seguido de α-tocoferol. É de referir que nas infusões foi somente encontrada a isoforma de α-tocoferol. Apesar dos frutos de F. vesca silvestre serem normalmente consumidos em fresco, este estudo prova a potencialidade das suas infusões como uma fonte de moléculas bioativas.

A concentração na castanha de minerais como o cálcio, potássio, o magnésio e outros tem sido estudada tradicionalmente por absorção atómica ou por espectrofotometria de UV-VIS [1-2]. Neste trabalho foi estudada a composição elementar de duas variedades (Longal e Judia) de castanha europeia (Castanea sativa Mill.) com a técnica PIXE (Proton Induced X-ray Emission). Os resultados preliminares mostram que a composição difere de acordo com a variedade, nos elementos maioritários e nos minoritários. Este estudo é parte de um projecto mais amplo, que visa avaliar o efeito do processamento pós-colheita de alimentos com feixe de electrões e raios-X.

Existe por parte dos consumidores uma tendência crescente na escolha de alimentos designados por mais saudáveis em que a presença em aditivos sintéticos é reduzida ou até mesmo ausente. Para melhorar a aparência e/ou propriedades dos alimentos a indústria recorre ao uso de aditivos sintéticos [1], no entanto, alguns autores têm apresentado alguma relação entre o consumo excessivo de alguns desses aditivos com efeitos adversos para a saúde do consumidor [2]. Para contornar esta problemática e ir ao encontro das expectativas dos consumidores, têm sido considerados os extratos naturais obtidos a partir de plantas como excelentes ingredientes naturais para a indústria alimentar como alternativas aos aditivos sintéticos [3]. Este trabalho teve como objetivo comparar os efeitos de antioxidantes naturais (extratos aquosos de Foeniculum vulgare Mill., funcho, e Matricaria recutita L., camomila, obtidos por decocção) com um aditivo sintético (sorbato de potássio, E202) utilizado em iogurtes. Neste trabalho, as amostras de Foeniculum vulgare Mill. (funcho) e Matricaria recutita L. (camomila) foram submetidas a uma extração por decocção. A sua caracterização química foi feita por HPLC-DAD-ESI/MS. As propriedades antioxidantes foram avaliadas através de diferentes ensaios in vitro (efeito captador de radicais livres, poder redutor e inibição da peroxidação lipídica), tal como as propriedades antimicrobianas (contra bactérias e fungos). A incorporação dos extratos foi feita em iogurtes e desta forma, foram preparados quatro grupos de amostras: iogurtes controlo (sem adição de qualquer aditivo), iogurtes com decocção de funcho, iogurtes com decocção de camomila e iogurtes com E202. As amostras foram avaliadas quanto à cor, pH e ao seu valor nutricional e potencial antioxidante. O estudo foi feito no tempo zero e após sete e catorze dias de armazenamento a 4ºC. Tal como podemos observar na Figura 1, a incorporação dos aditivos quer naturais quer sintéticos, não provocou alteração no aspeto visual quando comparado com a amostra controlo sem aditivos (A). Os resultados demonstram ainda que a introdução dos aditivos não provocou alterações significativas no pH e no valor nutricional dos iogurtes quando comparados com o controlo (Tabela 1). No entanto, esta incorporação conferiu propriedades antioxidantes aos iogurtes principalmente, pela adição do extrato de camomila (Figura 2). Estes resultados permitem-nos concluir que os extratos aquosos de funcho e camomila ricos em compostos fenólicos [4,5] podem representar uma alternativa aos conservantes sintéticos melhorando desta forma as propriedades funcionais dos iogurtes sem, no entanto, provocar alterações no perfil nutricional dos mesmos.

Com o objetivo de diminuir a incidência das leveduras encapsulas em alginato se aglomerarem na segunda fermentação em garrafa e reduzir custos associados à sua eliminação, pretendeu-se utilizar a metodologia NIR para determinar a quantidade de cálcio no vinho. Foram quantificados outros minerais para avaliar hipotéticos antagonismos iónicos. Observou-se também que este controlo de qualidade poderá ser efetuado no mosto. Atualmente este controlo é realizado por metodologias de análise de referência bastante morosas como EAA. Contudo a metodologia NIR demonstrou ser uma boa alternativa no controlo dos parâ-metros de qualidade na produção de vinho espumante, permitindo a diminuição do tempo de análise e de resíduos. No desenvolvimento dos modelos matemáticos para a calibração do NIR utilizaram-se 79 vinhos brancos e 60 amostras de mosto. Foram desenvolvidos 11 modelos de calibração onde o coeficiente de correlação foi, em aproximadamente 58% dos casos, maior que 0,99; ABSTRACT: In order to reduce the incidence of alginate beads aggregation during the second fermentation in the bottle and to reduce costs associated with their disposal, it was used the NIR technology to quantify calcium content in the wine. Other minerals were also quantified in order to evaluate possible ionic antagonisms. Quality control is usually done in the base wine but it can also be evaluated in the must. Currently this control was carried out by reference analysis methodologies generally rather slow as AAS. However NIR proved to be a good alternative technique in the control of quality parameters of wine production, allowing the reduction of the analysis time and waste. In order to develop mathematical models for the calibration of NIR it was used 79 white wine samples and 60 must samples. Eleven calibration models have been developed, where the correlation coeffi-cient was, in approximately 58% of cases, greater than 0,99.

Oil wells subjected to cyclic steam injection present important challenges for the development of well cementing systems, mainly due to tensile stresses caused by thermal gradients during its useful life. Cement sheath failures in wells using conventional high compressive strength systems lead to the use of cement systems that are more flexible and/or ductile, with emphasis on Portland cement systems with latex addition. Recent research efforts have presented geopolymeric systems as alternatives. These cementing systems are based on alkaline activation of amorphous aluminosilicates such as metakaolin or fly ash and display advantageous properties such as high compressive strength, fast setting and thermal stability. Basic geopolymeric formulations can be found in the literature, which meet basic oil industry specifications such as rheology, compressive strength and thickening time. In this work, new geopolymeric formulations were developed, based on metakaolin, potassium silicate, potassium hydroxide, silica fume and mineral fiber, using the state of the art in chemical composition, mixture modeling and additivation to optimize the most relevant properties for oil well cementing. Starting from molar ratios considered ideal in the literature (SiO2/Al2O3 = 3.8 e K2O/Al2O3 = 1.0), a study of dry mixtures was performed,based on the compressive packing model, resulting in an optimal volume of 6% for the added solid material. This material (silica fume and mineral fiber) works both as an additional silica source (in the case of silica fume) and as mechanical reinforcement, especially in the case of mineral fiber, which incremented the tensile strength. The first triaxial mechanical study of this class of materials was performed. For comparison, a mechanical study of conventional latex-based cementing systems was also carried out. Regardless of differences in the failure mode (brittle for geopolymers, ductile for latex-based systems), the superior uniaxial compressive strength (37 MPa for the geopolymeric slurry P5 versus 18 MPa for the conventional slurry P2), similar triaxial behavior (friction angle 21° for P5 and P2) and lower stifness (in the elastic region 5.1 GPa for P5 versus 6.8 GPa for P2) of the geopolymeric systems allowed them to withstand a similar amount of mechanical energy (155 kJ/m3 for P5 versus 208 kJ/m3 for P2), noting that geopolymers work in the elastic regime, without the microcracking present in the case of latex-based systems. Therefore, the geopolymers studied on this work must be designed for application in the elastic region to avoid brittle failure. Finally, the tensile strength of geopolymers is originally poor (1.3 MPa for the geopolymeric slurry P3) due to its brittle structure. However, after additivation with mineral fiber, the tensile strength became equivalent to that of latex-based systems (2.3 MPa for P5 and 2.1 MPa for P2). The technical viability of conventional and proposed formulations was evaluated for the whole well life, including stresses due to cyclic steam injection. This analysis was performed using finite element-based simulation software. It was verified that conventional slurries are viable up to 204ºF (400ºC) and geopolymeric slurries are viable above 500ºF (260ºC)

To produce porcelain tiles fluxing agents are used in order to obtain a liquid phase during firing. This liquid phase fills the pores decreasing porosity, water absorption and contributes to material densification. In the porcelain tiles industry, feldspar is the main flux material used, with quantities ranging between 35 and 50%. Studies focus on the discovery of materials with flux characteristics that can reduce the consumption of feldspar by porcelain tiles industry. In this context, the coffee husk ashes, a residue obtained when coffee husks are burned to produce heat for the dryers during the processing of the green fruit, have as main chemical constituents potassium, calcium and magnesium, giving them characteristics of fluxing material. Brazil is the largest coffee producer in the world and is responsible for over 30% of the world s production. In this work a physical treatment of coffee husk ash was carried out in order to eliminate the organic matter and, after this, two by-products were obtained: residual wastes R1 and R2. Both residues were added separately as single fluxes and also in association with feldspar in mixtures with raw materials collected in a porcelain industry located in Dias d Ávila-Ba. The addition of these residues aimed to contribute to the reduction of the consumption of feldspar in the production of porcelain tiles. Specimens were produced with dimensions of 60 mm x 20 mm x 6 mm in an uniaxial die with compacting pressure of 45 MPa. The samples were heated to a temperature of 1200 °C, for 8 minutes. Tests were performed to characterize the raw materials by XRF, XRD, particle size analysis, DTA and TGA and, additionally, the results of the physical properties of water absorption, apparent porosity, linear shrinkage, density, dilatometry, flexural strength and SEM of sintered body were analyzed. Additions of less than 8% of the residue R1 contributed to the decrease of porosity, but the mechanical strength of the samples was not satisfactory. Additions of 5% the R2 residue contributed significantly to decrease the water absorption and apparent porosity, and also to increase the mechanical strength. Samples with addition of feldspar associated with the R2 residue, in proportions of 6.7% of R2 and 6.7% of feldspar, led to results of water absorption of 0.12% and mechanical strength of 46 MPa, having parameters normalized to the manufacture of porcelain stoneware tiles

The development of activities in the oil and gas sector has been promoting the search for materials more adequate to oilwell cementing operation. In the state of Rio Grande do Norte, the cement sheath integrity tend to fail during steam injection operation which is necessary to increase oil recovery in reservoir with heavy oil. Geopolymer is a material that can be used as alternative cement. It has been used in manufacturing of fireproof compounds, construction of structures and for controlling of toxic or radioactive waste. Latex is widely used in Portland cement slurries and its characteristic is the increase of compressive strength of cement slurries. Sodium Tetraborate is used in dental cement as a retarder. The addition of this additive aim to improve the geopolymeric slurries properties for oilwell cementing operation. The slurries studied are constituted of metakaolinite, potassium silicate, potassium hydroxide, non-ionic latex and sodium tetraborate. The properties evaluated were: viscosity, compressive strength, thickening time, density, fluid loss control, at ambient temperature (27 ºC) and at cement specification temperature. The tests were carried out in accordance to the practical recommendations of the norm API RP 10B. The slurries with sodium tetraborate did not change either their rheological properties or their mechanical properties or their density in relation the slurry with no additive. The increase of the concentration of sodium tetraborate increased the water loss at both temperatures studied. The best result obtained with the addition of sodium tetraborate was thickening time, which was tripled. The addition of latex in the slurries studied diminished their rheological properties and their density, however, at ambient temperature, it increased their compressive strength and it functioned as an accelerator. The increase of latex concentration increased the presence of water and then diminished the density of the slurries and increased the water loss. From the results obtained, it was concluded that sodium tetraborate and non-ionic latex are promising additives for geopolymer slurries to be used in oilwell cementing operation
