The peroxisome proliferator-activated receptor gamma (PPARgamma) is highly expressed in the colon mucosa and its activation has been reported to protect against colitis. We studied the involvement of PPARgamma and its heterodimeric partner, the retinoid X receptor (RXR) in intestinal inflammatory responses. PPARgamma(1/)- and RXRalpha(1/)- mice both displayed a significantly enhanced susceptibility to 2,4,6-trinitrobenzene sulfonic acid (TNBS)-induced colitis compared with their wild-type littermates. A role for the RXR/PPARgamma heterodimer in the protection against colon inflammation was explored by the use of selective RXR and PPARgamma agonists. TNBS-induced colitis was significantly reduced by the administration of both PPARgamma and RXR agonists. This beneficial effect was reflected by increased survival rates, an improvement of macroscopic and histologic scores, a decrease in tumor necrosis factor alpha and interleukin 1beta mRNA levels, a diminished myeloperoxidase concentration, and reduction of nuclear factor kappaB DNA binding activity, c-Jun NH(2)-terminal kinase, and p38 activities in the colon. When coadministered, a significant synergistic effect of PPARgamma and RXR ligands was observed. In combination, these data demonstrate that activation of the RXR/PPARgamma heterodimer protects against colon inflammation and suggest that combination therapy with both RXR and PPARgamma ligands might hold promise in the clinic due to their synergistic effects.

A community-based random survey was conducted in a southern Brazilian Amazonian county aiming to investigate hepatitis C virus (HCV) infection prevalence and the association of demographic variables and lifestyle behaviours. Seven hundred eighty individuals were serologically screened with a third generation enzyme-linked immunosorbent assay to detect anti-HCV antibodies between 1994/1995. Positive samples were retested for confirmation with a line immunoassay (LIA, Inno-LIA HCV Ab III). Most of these subjects were low income and came from southern Brazilian states (65.8). Two point four percent (IC 95% 1.2%- 4.6%) of the subjects had LIA-confirmed anti-HCV antibodies reactivity. The age-specific prevalence of HCV antibodies slightly increased with age, with the highest prevalence after the age of 40 years. The results of multivariate analysis indicate a strong association between HCV antibodies and previous surgery and history of intravenous drug use. There were no apparent association with gender, hepatitis B virus markers, blood transfusion, and sexual activity. Mean time living in Amazon did not differ between confirmed and negative anti-HCV individuals. The present data point out an intermediate endemicity of HCV infection among this immigrant community to the Amazon region and that few HCV infected participants presented known risk factors.

Angiostrongylus costaricensis intermediate hosts are terrestrial mollusks mostly belonging to the Veronicellidae family. In the present investigation we focused on the mechanisms of larval expulsion from Sarasinula marginata infected with A. costaricensis. Twenty-five mollusks were individually infected with 5000 L1 and sacrificed at 30 min and 1, 2, 4, 6, and 8 h post-infection and at days 1, 2, 4, 5, 6, 8, 10, 11, 12, 14, 15, 16, 20, 21, 22, 25, 26, 28, and 30 post-infection; the mollusks were then fixed and stained. Diverse organs involved throughout the course of the migratory routes of larvae from oral penetration on were specified and the mechanisms of larval access to the fibromuscular layer through the kidney, rectum, and vascular system were defined. The elimination of L3, derived from oral and/or cutaneous infections, appears to depend on granulomas located close to the excretory ducts of mucous cells.

The present work describes the in vitro infection of a cell line Lulo, derived from Lutzomyia longipalpis embryonic tissue, by Leishmania chagasi promastigotes. This infection process is compared with a parallel one developed using the J774 cell line. The L. chagasi MH/CO/84/CI-044B strain was used for experimental infection in two cell lines. The cells were seeded on glass coverslips in 24-well plates to reach a final number of 2 x 10(5) cells/well. Parasites were added to the adhered Lulo and J774 cells in a 10:1 ratio and were incubated at 28 and 37ºC respectively. After 2, 4, 6, 8, and 10 days post-infection, the cells were extensively washed with PBS, fixed with methanol, and stained with Giemsa. The number of internalized parasites was determined by counting at least 400 cultured cells on each coverslip. The results showed continuous interaction between L. chagasi promastigotes with the cell lines. Some ultrastructural characteristics of the amastigote forms were observed using transmission electron microscopy. The highest percentage of infection in Lulo cells was registered on day 6 post-infection (29.6%) and on day 4 in the J774 cells (51%). This work shows similarities and differences in the L. chagasi experimental infection process in the two cell lines. However, Lulo cells emerge as a new model to study the life-cycle of this parasite.

The purpose of this study was to test the hypothesis that athletes having a slower oxygen uptake ( VO(2)) kinetics would benefit more, in terms of time spent near VO(2max), from an increase in the intensity of an intermittent running training (IT). After determination of VO(2max), vVO(2max) (i.e. the minimal velocity associated with VO(2max) in an incremental test) and the time to exhaustion sustained at vVO(2max) ( T(lim)), seven well-trained triathletes performed in random order two IT sessions. The two IT comprised 30-s work intervals at either 100% (IT(100%)) or 105% (IT(105%)) of vVO(2max) with 30-s recovery intervals at 50% of vVO(2max) between each repeat. The parameters of the VO(2) kinetics (td(1), tau(1), A(1), td(2), tau(2), A(2), i.e. time delay, time constant and amplitude of the primary phase and slow component, respectively) during the T(lim) test were modelled with two exponential functions. The highest VO(2) reached was significantly lower ( P<0.01) in IT(100%) run at 19.8 (0.9) km(.)h(-1) [66.2 (4.6) ml(.)min(-1.)kg(-1)] than in IT(105%) run at 20.8 (1.0) km(.)h(-1) [71.1 (4.9) ml(.)min(-1.)kg(-1)] or in the incremental test [71.2 (4.2) ml(.)min(-1.)kg(-1)]. The time sustained above 90% of VO(2max) in IT(105%) [338 (149) s] was significantly higher ( P<0.05) than in IT(100%) [168 (131) s]. The average T(lim) was 244 (39) s, tau(1) was 15.8 (5.9) s and td(2) was 96 (13) s. tau(1) was correlated with the difference in time spent above 90% of VO(2max) ( r=0.91; P<0.01) between IT(105%) and IT(100%). In conclusion, athletes with a slower VO(2) kinetics in a vVO(2max) constant-velocity test benefited more from the 5% rise of IT work intensity, exercising for longer above 90% of VO(2max) when the IT intensity was increased from 100 to 105% of vVO(2max).

Résumé Malgré l'apparition de nouvelles techniques chirurgicales dites « sans tension », l'antalgie post-opératoire après cure de hernie inguinale reste un défi pour les anesthésiologistes. Récemment on a suggéré que l'addition de ketamine ou d'un anti-inflammatoire non-stéroïdien (AINS) à un anesthésique local pourrait améliorer et prolonger l'analgésie postopératoire. Le but de cette étude, à laquelle ont participé 36 patients ASA I-II, était d'évaluer si la coadministration de S(+) ketamine ou de ketorolac renforcerait les effets analgésiques de la bupivacaïne après cure ambulatoire de hernie inguinale sous anesthésie générale. L'analgésie a consisté en une infiltration de la plaie associé à un bloc inguinal avec soit 30 ml de bupivacaïne 0,5 % (n=12), soit 27 ml de bupivacaïne 0,5 % + 3 ml de S(+) ketamine (75 mg) (n=12), soit 28 ml de bupivacaïne 0,5 % + 2 ml de ketorolac (60 mg) (n=12). La prise orale d'antalgique en phase postopératoire était standardisée. L'intensité des douleurs a été évaluée au moyen d'une échelle visuelle analogique (EVA), d'un score verbal d'estimation et par algométrie de pression respectivement 2, 4, 6, 24 et 48 heures après l'intervention. Les trois groupes de patients ont présenté le score de douleur évalué par EVA le plus élevé à 24 heures, score significativement différent de ceux mesurés à 6 et 48 heures (P <0.05). A part une sensation de douleurs significativement moindre (score verbal d'estimation) dans le groupe ketorolac à 24 et 48 heures et seulement à 48 heures dans le groupe ketamine, il n'y avait aucune autre différence entre les groupes pour la durée de l'étude (48 heures) en ce qui concerne les scores de douleur, les seuils de douleur à la pression ou la prise postopératoire d'antalgiques « de secours ». En conclusion, l'addition de S(+) ketamine ou de ketorolac, n'améliore que marginalement l'effet analgésique de la bupivacaïne. Ceci peut-être mis en relation avec la technique de cure de hernie « sans tension » induisant un bas niveau de douleur postopératoire. Abstract Objective: The aim of the study was to assess whether coadministration of S(±) ketamine or ketorolac would enhance or prolong local analgesic effect of bupivacaine after inguinal hernia repair. Design: Prospective double-blind randomized study evaluating pain intensity after surgery under general anesthesia. Setting: Outpatient facilities of the University Hospital of Lausanne. Patient: Thirty-six ASA I-II outpatients scheduled for elective day-case inguinal herniorraphy. Intervention: Analgesia strategy consisted of a wound infiltration and an inguinal field block either with 30 mL bupivacairie (0.5%) or with the same volume of a mixture of 27 mL bupivacaine (0.5%) + 3 mL S(+) ketamine (75 mg) or a 28 mL bupivacaine (0.5%) + 2 ML ketorolac (60 mg). Postoperative analgesic regimen was standardized. Outcome Measures: Pain intensity was assessed with a Visual Analog Seale, a verbal rating score, and by pressure algometry 2, 4, 6, 24, and 48 hours after surgery. Results: The 3 groups of patients experienced the highest Visual Analog Scale pain score at 24 hours, which was different from those at 6 and 48 hours (P < 0.05). Apart from a significantly lower pain sensation (verbal rating score) in the ketorolac group at 24 and 48 hours and only at 48 hours with ketamine, there were no other differences in pain scores, pain pressure thresholds, or rescue analgesic consumption between groups throughout the 48-hour study period. Conclusion: The addition of S (+)-ketamine or ketorolac only minimally improves the analgesic effect of bupivacaine. This may be related to the tension-free hernia repair technique associated with low postoperative pain.

We describe herein the antitrypanosomal activity of 20 novel 1,3-bis(aryloxy)propan-2-amine derivatives. Compounds 2, 4, 6, 12, 15, 16 and 19 were significantly active against amastigote and trypomastigote forms, with half maximal inhibitory concentrationvalues in the range of 6-18 µM. In the cytotoxicity tests against L929 cells, only compound 4 presented selectivity index value above 10, indicating low toxicity.

Background: Inflammatory bowel disease (IBD) is characterized by chronic intestinal inflammation due to dysregulation of the mucosal immune system. The cytokines IL-1β and IL-18 appear early in intestinal inflammation and their pro-forms are processed via the caspase-1-activating multiprotein complex, the Nlrp3 inflammasome. Previously, we reported that the uptake of dextran sodium sulfate (DSS) by macrophages activates the Nlrp3 inflammasome and that Nlrp3(-/-) mice are protected in the acute DSS colitis model. Of note, other groups have reported opposing effects in regards to DSS susceptibility in Nlrp3(-/-) mice. Recently, mice lacking inflammasomes were found to develop a distinct intestinal microflora. Methods: To reconcile the contradicting observations, we investigated the role of Nlrp3 deficiency in two different IBD models: acute DSS colitis and TNBS (2,4,6-trinitrobenzene sulfonic acid)-induced colitis. In addition, we investigated the impact of the intestinal flora on disease severity by performing cohousing experiments of wild-type and Nlrp3(-/-) mice, as well as by antibiotic treatment. Results: Nlrp3(-/-) mice treated with either DSS or TNBS exhibited attenuated colitis and lower mortality. This protective effect correlated with an increased frequency of CD103+ lamina propria dendritic cells expressing a tolerogenic phenotype in Nlrp3(-/-) mice in steady state conditions. Interestingly, after cohousing, Nlrp3(-/-) mice were as susceptible as wild-type mice, indicating that transmission of endogenous bacterial flora between the two mouse strains might increase susceptibility of Nlrp3(-/-) mice towards DSS-induced colitis. Accordingly, treatment with antibiotics almost completely prevented colitis in the DSS model. Conclusions: The composition of the intestinal microflora significantly influences disease severity in IBD models comparing wild-type and Nlrp3(-/-) mice. This observation may - at least in part - explain contradictory results concerning the role of the inflammasome in different labs. Further studies are required to define the role of the Nlrp3 inflammasome in noninflamed mucosa under steady state conditions and in IBD.

As guidelines de cardiologia nuclear europeia e americanas não são específicas na escolha dos melhores parâmetros de reconstrução de imagem a utilizar na Cintigrafia de Perfusão do Miocárdio (CPM). Assim, o presente estudo teve como objectivo estabelecer e comparar o efeito dos parâmetros quantitativos dos métodos de reconstrução: Retroprojecção Filtrada (FBP) e Ordered ‑Sub‑set Expectation Maximization (OSEM). Métodos: Foi utilizado um fantoma cardíaco, cujos valores do volume telediastólico (VTD), volume telesistólico (VTS) e fracção de ejecção ventricular esquerda (FEVE) eram conhecidos. O software Quantitative Gated SPECT/Quantitative Perfusion SPECT foi utilizado em modo semi‑automático, a fim de obter esses parâmetros quantitativos. O filtro Butterworth foi usado no FBP com as frequências de corte entre 0,2 e 0,8 ciclos/pixel combinadas com as ordens de 5, 10, 15 e 20. Na reconstrução OSEM, foram utilizados os subconjuntos 2, 4, 6, 8, 10, 12 e 16, combinados com os números de iterações de 2, 4, 6, 8, 10, 12, 16, 32 e 64. Durante a reconstrução OSEM efectuou‑se uma outra reconstrução baseada no número de iterações equivalentes - Expectation‑Maximization (EM) 12, 14, 16, 18, 20, 22, 26, 28, 30 e 32. Resultados: Após a reconstrução com FBP verificou‑se que os valores de VTD e VTS aumentavam com o aumento da frequência de corte, enquanto o valor da FEVE diminui. Esse mesmo padrão é verificado na reconstrução OSEM. No entanto, com OSEM há uma estimativa mais precisa dos parâmetros quantitativos, especialmente com as combinações 2I × 10S e 12S × 2I. Conclusão: A reconstrução OSEM apresenta uma melhor estimativa dos parâmetros quantitativos e uma melhor qualidade de imagem do que a reconstrução com FBP. Este estudo recomenda o uso de 2 iterações com 10 ou 12 subconjuntos para a reconstrução OSEM e uma frequência de corte de 0,5 ciclos/pixel com as ordens 5, 10 ou 15 para a reconstrução com FBP como a melhor estimativa para a quantificação da FEVE através da CPM.

The State of Iowa currently has approximately 69,000 miles of unpaved secondary roads. Due to the low traffic count on these unpaved o nts as ng e two dust ed d roads, paving with asphalt or Portland cement concrete is not economical. Therefore to reduce dust production, the use of dust suppressants has been utilized for decades. This study was conducted to evaluate the effectiveness of several widely used dust suppressants through quantitative field testing on two of Iowa’s most widely used secondary road surface treatments: crushed limestone rock and alluvial sand/gravel. These commercially available dust suppressants included: lignin sulfonate, calcium chloride, and soybean oil soapstock. These suppressants were applied to 1000 ft test sections on four unpaved roads in Story County, Iowa. Tduplicate field conditions, the suppressants were applied as a surface spray once in early June and again in late August or early September. The four unpaved roads included two with crushed limestone rock and two with alluvial sand/gravel surface treatmewell as high and low traffic counts. The effectiveness of the dust suppressants was evaluated by comparing the dust produced on treated and untreated test sections. Dust collection was scheduled for 1, 2, 4, 6, and 8 weeks after each application, for a total testiperiod of 16 weeks. Results of a cost analysis between annual dust suppressant application and biennial aggregate replacement indicated that the cost of the dust suppressant, its transportation, and application were relatively high when compared to that of thaggregate types. Therefore, the biennial aggregate replacement is considered more economical than annual dust suppressant application, although the application of annual dust suppressant reduced the cost of road maintenance by 75 %. Results of thecollection indicated that the lignin sulfonate suppressant outperformed calcium chloride and soybean oil soapstock on all four unpavroads, the effect of the suppressants on the alluvial sand/gravel surface treatment was less than that on the crushed limestone rock, the residual effects of all the products seem reasonably well after blading, and the combination of alluvial sand/gravel surface treatment anhigh traffic count caused dust reduction to decrease dramatically.

A crucial method for investigating patients with coronary artery disease (CAD) is the calculation of the left ventricular ejection fraction (LVEF). It is, consequently, imperative to precisely estimate the value of LVEF--a process that can be done with myocardial perfusion scintigraphy. Therefore, the present study aimed to establish and compare the estimation performance of the quantitative parameters of the reconstruction methods filtered backprojection (FBP) and ordered-subset expectation maximization (OSEM). METHODS: A beating-heart phantom with known values of end-diastolic volume, end-systolic volume, and LVEF was used. Quantitative gated SPECT/quantitative perfusion SPECT software was used to obtain these quantitative parameters in a semiautomatic mode. The Butterworth filter was used in FBP, with the cutoff frequencies between 0.2 and 0.8 cycles per pixel combined with the orders of 5, 10, 15, and 20. Sixty-three reconstructions were performed using 2, 4, 6, 8, 10, 12, and 16 OSEM subsets, combined with several iterations: 2, 4, 6, 8, 10, 12, 16, 32, and 64. RESULTS: With FBP, the values of end-diastolic, end-systolic, and the stroke volumes rise as the cutoff frequency increases, whereas the value of LVEF diminishes. This same pattern is verified with the OSEM reconstruction. However, with OSEM there is a more precise estimation of the quantitative parameters, especially with the combinations 2 iterations × 10 subsets and 2 iterations × 12 subsets. CONCLUSION: The OSEM reconstruction presents better estimations of the quantitative parameters than does FBP. This study recommends the use of 2 iterations with 10 or 12 subsets for OSEM and a cutoff frequency of 0.5 cycles per pixel with the orders 5, 10, or 15 for FBP as the best estimations for the left ventricular volumes and ejection fraction quantification in myocardial perfusion scintigraphy.

A calagem tem sido considerada uma prática suficiente para o suprimento de molibdênio (Mo) para as culturas, pelo aumento do pH que o torna mais disponível no solo. A cultura da soja exige doses elevadas de calcário para a otimização da produtividade, o que também está relacionado com a demanda adicional de Mo para o complexo da nitrogenase, atuante na fixação do nitrogênio. Assim, o presente trabalho foi planejado para estudar as interações entre calagem e Mo, nas culturas de soja e sorgo, em um Podzólico Vermelho-Amarelo da Estação Experimental de Mococa (IAC), de 1985 a 1989. Os tratamentos foram arranjados no delineamento experimental de blocos ao acaso, com parcelas subdivididas e quatro repetições. Nas parcelas principais, foram aplicadas as doses de calcário 0, 2, 4, 6, e 8 t ha-1 (PRNT = 126%) e, nas subparcelas, as doses de Mo de 0, 50 e 100 g ha-1, aplicadas às sementes, na forma de molibdato de amônio. Foram realizados três cultivos de soja, cultivar IAC 11, e um de sorgo granífero, híbrido DK 64. Em todos os cultivos, as respostas à calagem foram acentuadas, porém reduzidas com a aplicação de molibdênio, tanto para a soja como para o sorgo. Tais resultados demonstraram a relação de substituição de calcário por Mo. A resposta da soja ao Mo foi mais acentuada na ausência do calcário, enquanto a do sorgo, mais sensível à acidez do solo, foi mais acentuada nas doses intermediárias de calcário. De modo geral, as respostas ao Mo ocorreram até o valor de pH (CaCl2 ) do solo igual a 5,2. Concluiu-se que altas produtividades de soja exigem níveis mais elevados de correção de acidez de solo, e que é possível reduzir a necessidade de calagem, para atingir a produtividade máxima, mediante a aplicação de Mo nas sementes, para ambas as culturas, principalmente num solo ácido com pouco alumínio e manganês trocáveis.

O cultivo de milho e soja em solos ácidos requer a aplicação de calcário, para elevar o pH, reduzir a toxidez de alumínio e fornecer os nutrientes cálcio e magnésio. O calcário pode ter efeito residual prolongado, sendo necessário quantificar a durabilidade e a eficiência da calagem no solo por um período longo de cultivos sucessivos. Neste trabalho, deu-se continuidade a um experimento instalado na EMBRAPA Cerrados, em 1986, em solo Glei Pouco Húmico argiloso, com doses iniciais de 0, 2, 4, 6 e 8 t ha-1 de calcário dolomítico a lanço. A partir de 1990, foi cultivado o milho em rotação com a soja, em um total de sete cultivos sucessivos. Os dados de produtividade mostraram que o calcário aplicado em 1986 tem apresentado efeito residual. A resposta das duas culturas até o quarto cultivo e da soja, no sexto cultivo, foi significativamente maior a partir da primeira dose de 2 t ha-1 de calcário. No quinto e no sétimo ano de cultivo, a produtividade do milho foi significativamente maior, respectivamente, a partir das doses de 4 e 6 t ha-1 de calcário. A produtividade relativa máxima, para a média de quatro cultivos de milho e três cultivos de soja em sucessão, foi obtida com a saturação por bases do solo de 42% para o milho e 38% para a soja. A partir do quarto ano de aplicação do calcário, a perda anual média de cálcio + magnésio trocáveis do solo nos tratamentos com calcário foi de 2 mmol c dm-3.

Com o objetivo de avaliar a influência da relação entre Ca e Mg na CTC do solo sobre o rendimento do feijoeiro, foi realizado um experimento em casa de vegetação, em vasos com 3,5 dm³ de amostras de um Latossolo Vermelho distrófico psamítico-LVdp e um Latossolo Vermelho aluminoférrico típico-LVat. Os tratamentos, dispostos em blocos casualizados, com seis repetições, constaram das seguintes relações molares entre Ca e Mg: 1/2, 1, 2, 4, 6 e 8. Três repetições foram colhidas no florescimento e as demais mantidas até à colheita dos grãos. Os rendimentos da matéria seca do feijoeiro obtida no estádio de florescimento foram semelhantes nos dois solos, mas os rendimentos de grãos obtidos no LVat foram 40 % superiores aos obtidos no LVdp. Entretanto, estas variáveis não foram significativamente influenciadas pelas diferentes relações Ca:Mg no solo. As concentrações de Ca e Mg e suas relações no tecido da matéria seca do feijoeiro, obtida no florescimento, apresentaram estreita relação com os teores e relações destes elementos no solo. Os resultados obtidos mostraram que em solos com teores adequados de Ca e Mg trocáveis, a relação entre eles é de importância secundária para o desenvolvimento e rendimento do feijoeiro.

Pesquisas sobre a disponibilidade de B em solos são necessárias para avaliar o comportamento das culturas sob diferentes teores disponíveis desse elemento no solo. Com essa finalidade, foi realizado um experimento em casa de vegetação para avaliar a resposta de milho (Zea mays) a doses de boro (0, 2, 4, 6 e 12 mg dm-3) em dez solos de Pernambuco. O B disponível foi determinado por três extratores: água quente, ácido clorídrico 0,05 mol L-1 e Mehlich-1. Foram ainda determinados os níveis críticos (90 e 95 % da produção máxima de matéria seca) e tóxicos (equivalente à redução de 10 % da produção máxima) nos solos e na planta, além da descrição de sintomas de toxidez de B em milho. Os resultados demonstraram que os níveis críticos e tóxicos de B nos solos variaram de 0,4 a 2,3 mg kg-1 e de 1,8 a 8,3 mg kg-1, respectivamente. Em plantas de milho, os níveis críticos variaram de 13,8 a 129,6 mg kg-1, enquanto teores tóxicos foram obtidos entre 43,3 e 372,2 mg kg-1. O extrator que obteve a melhor correlação com o teor de B na planta foi o HCl 0,05 mol L-1, seguido pelo Mehlich-1 e pela água quente. A disponibilidade do B no solo pelo extrator HCl e os teores de B na folha foram afetados negativamente pelo aumento no teor de argila e matéria orgânica dos solos. Os solos que apresentaram plantas com maior teor de B no tecido vegetal mostraram sintomas mais graves de toxidez, fator associado à maior disponibilidade de boro em solos com textura mais arenosa e, ou, baixos teores de matéria orgânica.
