A Photocatalyst ceramic powder that presented high photoactivity based on TiO2 modified with 25% molar of SnO2 and up to 5% molar of Ag2O was obtained in the present work. The aforementioned ceramic powder was obtained using all commercial oxides as well as the oxides mixture technique. The powders were ground in high energy mill for one hour with subsequent thermal treatment at 400°C for four hours. They were, furthermore, characterized using surface area of around 6m2/g, where the X-Ray diffraction results provided evidence for the presence of anatase and rutile phases, known to be typical characteristics of both the TiO2 and SnO2 used. During the thermal treatment, Ag2O was reduced to metallic silver. The photodegradation rehearsals were carried out using a 0.01 mmol/L Rhodamine B solution in a 100mg/L photocatalyst suspension in a 500ml beaker, which was irradiated with 4W germicide Ultraviolet light of 254nm. In addition, samples were removed after duration of about 10 minutes to an hour, where they were analyzed thoroughly in UV-vis spectrophotometer. The analysis of the results indicated that for the compositions up to 2.5% molar of Ag2O, the photoactivity was found to be greater than that of Degussa P25 photocatalyst powder, and as such it was then used as a reference. Taking into account 90% degradation of Rhodamine B, a duration period of 11 minutes was obtained for the developed photocatalyst powder compared to the 38 minutes observed for the Degussa P25. FEG-SEM micrographies enabled the verification of the morphology as well as the interaction of the oxide particles with the metallic silver, which led us to propose a model for the increase in photoactivity observed in the photocatalyst powder under investigation.

Nanotechnology, the science of minuscule, has developed products which are able t o manipulate atoms and molecules that could be applied in the sterilization process of dental instruments. Objetives: The objective of the present study was to evaluate the self-cleaning action of TiO2 and Ag nanoparticles coating on dental instruments by the photocataliys process under UV and visible light irradiation. Material and method: Microbiologic tests were done using dental cement spatulas coated with TiO2 and Ag nanoparticles (one or three layers), and contaminated with 10 mcrl of Pseudomonas aeruginosa and Enterococcus faecalis, respectively. After contamination, they were exposed to ultraviolet light and visible light for 120 minutes. Next, they were transferred to and stored in test tubes with BHI (Brain Heart Infusion) and incubated in 35 to 37 °C. Checking times for bacterial growth and for control and retrieval tests were done at: 24, 48, 72 and 96 hours. Result: The Pseudomonas aeruginosa was inactive after 120 minutes of ultraviolet light irradiation, thus confirming the heterogeneous photocatalytic activity of TiO2 and Ag. The Pseudomonas aeruginosa was not inactivated under visible light irradiation and the Enterococcus faecalis was not inactivated under UV and visible light irradiation of the dental cement spatulas coated with TiO2 and Ag nanoparticles in the readings to 96 hours, showing bacterial growth. Conclusion: There were no influence of one or three layers of TiO2 and Ag nanoparticles coating of the spatulas in the results. The heterogeneous photocatalysis activity of TiO2 and Ag under UV light irradiation was confirmed for Pseudomonas aeruginosa but not under visible light. Enterococcus faecalis did not confirmed the photocatalytics activity of TiO2 and Ag under UV light irradiation and visible lights irradiation.

The Sol-gel process has facilitated the processing of Organic-Inorganic Hybrid Materials with several scientific applications and technologies. The hybrid combine the advantages of the Sol-gel process with specific characteristics of organic polymers, allowing processing of thin films without fractures or fissures. The incorporation of azo dyes in polymer matrices has been widely used in the investigation of optical properties. The azo dye Disperse Red (DR1) presents optical alterations when exposed to visible or ultraviolet light. The alterations occur due to transitions of their isomers, trans and cis, caused by photoisomerization, due to electronic transitions of azo group (-N=N-), presenting photochromic and/or photorefractive effects. The hybrid system used in this work is the precursor 3- Glycidoxypropyl-Trimethoxi-silane (GPTS), the Tetraethylorthosilicate (TEOS) and DR1 as a dopant. The characterizations were performed using absorption spectroscopy UV-Vis which allowed the identification of the absorption bands and its variations when the samples were treated thermally and/or illuminated by ultraviolet light

The present study was aimed to evaluate different (semi-solid) media for the production of Metarhizium anisopliae and Beauveria bassiana propagules, and to evaluate the tolerance of these propagules to ultraviolet radiation and temperature. The experiments were performed at the Biological Control Laboratory of the Instituto Biológico at Campinas, São Paulo, Brazil. For both fungi, 6 repetitions were performed for each of the 17 treatments: corn starch, full rice, parboiled rice, type-1 rice, type-2 rice, oat flakes, canjiquinha [grits], wheat flour, raw cassava flour, yellow corn flour, special wheat flour, corn flour, corn in grains, cassava starch, soy in grains, crushed wheat, and turf. The viability analysis was done in plastic plates containing BDA. For the bioassays involving exposure to ultraviolet light and temperature, BDA was also used for viability analysis, and each treatment was exposed to the UV radiation for 0, 25 and 50 seconds, the temperature exposure being at 20, 25, 30 and 35º C. Using a Potter tower, 2 mL of fungus suspension from each treatment was inoculated to the Diatraea saccharalis caterpillars. Regarding the sporulation, the largest concentrations of M. anisopliae and B. bassiana were found for the treatments with parboiled rice, type-1 rice, type-2 rice, yellow corn flour, corn flour and crushed wheat. The viability of all treatments was superior to 94.00%. Also, the longer the duration of the exposition to the UV, the smaller the number of fertile conidia. At 35o C, a significant loss of conidia viability was observed, and all the treatments presented some level of virulence.

This thesis presents and uses the techniques of computational chemistry to explore two different processes induced in human skin by ultraviolet light. The first is the transformation of urocanic acid into a immunosuppressing agent, and the other is the enzymatic action of the 8-oxoguanine glycosylase enzyme. The photochemistry of urocanic acid is investigated by time-dependent density functional theory. Vertical absorption spectra of the molecule in different forms and environments is assigned and candidate states for the photochemistry at different wavelengths are identified. Molecular dynamics simulations of urocanic acid in gas phase and aqueous solution reveals considerable flexibility under experimental conditions, particularly for for the cis isomer where competition between intra- and inter-molecular interactions increases flexibility. A model to explain the observed gas phase photochemistry of urocanic acid is developed and it is shown that a reinterpretation in terms of a mixture between isomers significantly enhances the agreement between theory and experiment , and resolves several peculiarities in the spectrum. A model for the photochemistry in the aqueous phase of urocanic acid is then developed, in which two excited states governs the efficiency of photoisomerization. The point of entrance into a conical intersection seam is shown to explain the wavelength dependence of photoisomerization quantum yield. Finally some mechanistic aspects of the DNA repair enzyme 8-oxoguanine glycosylase is investigated with density functional theory. It is found that the critical amino acid of the active site can provide catalytic power in several different manners, and that a recent proposal involving a SN1 type of mechanism seems the most efficient one.

The nucleus of a eukaryotic cell contains both structural and functional elements that contribute to the controlled operation of the cell. In this context, functional components refers to those nuclear constituents that perform metabolic activities such as DNA replication and RNA transcription. Structural nuclear components, designated nuclear matrix, organize the DNA into loops or domains and appear to provide a framework for nuclear DNA organization. However, the boundary between structural and functional components is not clear cut as evinced by reports of associations between metabolic functions and the nuclear matrix. The studies reported here attempt to determine the relationship of another nuclear function, DNA repair, to the nuclear matrix.^ One objective of these studies was to study the initiation of DNA repair by directly measuring the UV-incision activities in human cells and determine the influence of various extractable nuclear components on these activities. The assay for incision activities required the development of a nuclear isolation protocol that produced nuclei with intact DNA; the conformation of the nuclear DNA and its physical characteristics in response to denaturing conditions were determined.^ The nuclei produced with this protocol were then used as substrates for endogenous UV-specific nuclease activities. The isolated nuclei were shown to contain activities that cause breaks in nuclear DNA in response to UV-irradiation. These UV-responsive activities were tightly associated with nuclear components, being unextractable with salt concentration of up to 0.6 M.^ The tight association of the incision activities with salt-extracted nuclei suggested that other repair function might also be associated with salt-stable components of the nucleus. The site of unscheduled DNA synthesis (UDS) was determined in salt-extracted nuclei (nucleoids) using autoradiography and fluorescent microscopy. UDS was found to occur in association with the nuclear matrix following low-doses (2.55 J/M('2)) of ultraviolet light, but the association became looser after higher doses of ultraviolet light (10-30 J/m('2)). ^
