L’implantation retardée ou diapause embryonnaire décrit l'arrêt ou le retardement pendant l'embryogenèse. Chez le vison, la diapause est corrélée avec une sécrétion pituitaire insuffisante de la prolactine, ayant pour résultat la différentiation incomplète du corpus luteum et réduction de la progestérone. Des études antérieures suggèrent que le blastocyste de vison en diapause demeure dans un état de quiescence ou se développe lentement. Pour élucider ceci, la réplication de l'ADN a été étudiée. Les résultats démontrent synthèse de l’ADN et prolifération cellulaire dans les embryons au stade de morula, avant la diapause et dans les blastocystes après la réactivation. La réplication de l'ADN a été également détectée dans des blastocystes en diapause et en diapause prolongée. L'implantation est considérée comme une interaction bidirectionnelle entre le blastocyste et l'utérus. Il a été montré que les prostaglandines sont importantes pour la vascularisation de l’utérus au moment de l’implantation et peuvent réactiver l'utérus des visons après la diapause. La concentration protéinique et la localisation de la phospholipase citosolique A2 (CPLA2) et de la cyclooxygenase 2 (COX2) ont été étudiées dans l'utérus de vison. L’expression de la CPLA2 et COX2 étaient sur-régulées au moment de l'implantation. Il est connu que la prolactine active les corpus luteum des visons. L'idée de un lien entre la prolactine et la voie de signalisation des prostaglandines a été testée en mesurant les récepteurs de prolactine. Les résultats montrent une augmentation de l’expression des récepteurs de prolactine à l'implantation suggérant que la prolactine pourrait activer la voie de prostaglandine à l'utérus par son propre récepteur. La conclusion, les embryons pendant la diapause ne sont pas arrêtées complètement et les protéines liées à la voie de prostaglandine sont implique dans la réactivation de l'utérus.

Mit ansteigenden Jahresmilchleistungen werden die Kühe einer hohen metabolischen Belastung ausgesetzt, die bei einer suboptimalen Gestaltung der Lebensbedingungen mit einer Erhöhung der Inzidenzrate von Faktorenkrankheiten einhergehen kann. Im Vordergrund der Untersuchungen stand die Frage, inwieweit metabolische Belastungszustände von Milchkühen in der Phase des Puerperiums und in Abhängigkeit von der Milchleistung sowie Belastungen durch eine klinische Erkrankung zu Beginn der Laktation mit Hilfe von Parametern der immunologischen Abwehr und der Reaktion des Immunsystems auf eine Challenge dargestellt werden können. In die Untersuchung wurden insgesamt 68 klinisch gesunde sowie 20 klinisch erkrankte Milchkühe der Rasse Holstein Friesian einbezogen. Die Untersuchungen erfolgten in den Zeiträumen 10-21 Tage sowie 14-15 Wochen nach der Kalbung. Dazu wurden an jeweils drei Untersuchungstagen (Tag 0, 2, 7) Blutproben aus der Vena jugularis entnommen. Zur Stimulierung des unspezifischen Immunsystems wurde in beiden Versuchszeiträumen an den Untersuchungstagen 0 und 2 der Paramunitätsinducer Zylexis® appliziert. Ferner wurde den Kühen zu Beginn der Laktation der Tollwutimpfstoff Rabisin® verabreicht. Für die Auswertung wurden die untersuchten Milchkühe in Gruppen unterteilt. Die klinisch gesunden Kühe wurden anhand der nach Fett und Eiweiß korrigierten 305-Tage-Leistung in die Gruppe M = mittleres Leistungsniveau (Milchmengenleistung 6.500-8.990 kg) und die Gruppe H = hohes Leistungsniveau (Milchmengenleistung 9.000-12.500 kg) aufgeteilt. Die klinisch erkrankten Kühe wurden in der Gruppe K zusammengefasst. Zur Beurteilung der Immunabwehr wurden die Parameter Phagozytoseaktivität der isolierten Neutrophilen Granulozyten, die Vollblutbakterizidie, die Lymphozytenproliferation mit den Mitogenen ConA, PHA und PWM sowie die spezifische Antikörperbildung gegen Tollwut untersucht. Anhand der Parameter Phagozytoseaktivität, Vollblutbakterizidie und Lymphozytenproliferation wurde zusätzlich die Reaktion auf die Challenge mit dem Paramunitätsinducer Zylexis® überprüft. Zur Erfassung der metabolischen Belastungszustände wurden verschiedene Stoffwechselparameter und hämatologische Parameter erfasst. Zu Beginn der Laktation konnten bei den Kühen in der mittleren und hohen Leistungsgruppe signifikant höhere Konzentrationen an ß-Hydroxybuttersäure und Freien Fettsäuren im Vergleich zu der Laktationsmitte festgestellt werden. Die Kühe mit einem hohen Leistungsniveau zeigten darüber hinaus eine signifikant höhere Bilirubinkonzentration am Laktationsanfang gegenüber der Laktationsmitte. Die mittlere Konzentration der freien Fettsäuren lag bei den Kühen in beiden Leistungsgruppen oberhalb des Referenzbereiches und wies auf eine negative Energiebilanz zu Laktationsbeginn hin. Die mittleren Konzentrationen bzw. Enzymaktivitäten der Parameter Cholesterol, Harnstoff, AST, GLDH, γ-GT, Gesamtprotein, Kalzium und Phosphor wiesen in der Laktationsmitte signifikant höhere Werte als am Laktationsanfang auf. Dabei lagen die mittleren Enzymaktivitäten von AST und GLDH sowie die Konzentrationen von Harnstoff und Gesamtprotein in der Laktationsmitte oberhalb des Referenzbereiches. Die untersuchten Stoffwechselparameter wurden durch die Challenge mit einem Paramunitätsinducer nicht signifikant beeinflusst. Hinsichtlich der Immunparameter zeigten die Milchkühe mit einem mittleren Leistungsniveau in Abhängigkeit vom Laktationszeitpunkt zu Beginn der Laktation eine signifikant höhere Phagozytoseaktivität gegenüber der Laktationsmitte (p < 0,05). Die Gruppe mit einem hohen Leistungsniveau wies dagegen keine signifikanten Unterschiede in der Phagozytoseaktivität zwischen den beiden Untersuchungszeiträumen auf. Bei den Immunparametern Vollblutbakterizidie und Lymphozytenproliferation konnten bei den Kühen beider Gruppen keine signifikanten Unterschiede in Abhängigkeit vom Laktationszeitpunkt festgestellt werden. Im Hinblick auf die Milchleistung wiesen die Kühe in den beiden Leistungsgruppen am Untersuchungstag 0 weder zu Beginn noch in der Mitte der Laktation signifikante Unterschiede bezüglich der untersuchten Immunparameter auf. Nach der Challenge durch den Paramunitätsinducer reagierten einzig die Kühe mit einer mittleren Leistung zu Beginn der Laktation auf die Challenge mit einer signifikanten Reduzierung der mittleren Phagozytoseaktivität von Tag 0 zu Tag 2 (p < 0,05). Bei den weiteren untersuchten Immunparametern konnten weder bei den Kühen mit einer mittleren noch mit einer hohen Leistung eine signifikante Reaktion auf die Challenge nachgewiesen werden. Die klinisch erkrankten Kühe zeigten im Vergleich zu den klinisch gesunden Kühen keine unterschiedlichen Konzentrationen bzw. Aktivitäten der untersuchten Stoffwechselparameter. Auch konnten anhand der immunologischen Parameter und der Reaktion auf die Challenge keine Unterschiede zwischen den klinisch gesunden Kühen festgestellt werden. Die in der vorliegenden Untersuchung geprüften metabolischen Belastungen konnten weder anhand der Immunparameter noch durch die Reaktion der Immunparameter auf eine Challenge dargestellt werden. Es wird geschlussfolgert, dass das immunologische Reaktionsvermögen nicht geeignet ist, um als Indikator für Belastungszustände bei der Milchkuh herangezogen zu werden.

Climate change and variability in sub-Saharan West Africa is expected to have negative consequences for crop and livestock farming due to the strong dependence of these sectors on rainfall and natural resources, and the low adaptive capacity of crops farmers, agro-pastoralist and pastoralists in the region. The objective of this PhD research was to investigate the anticipated impacts of expected future climate change and variability on nutrition and grazing management of livestock in the prevailing extensive agro-pastoral and pastoral systems of the Sahelian and Sudanian zones of Burkina Faso. To achieve this, three studies were undertaken in selected village territories (100 km² each) in the southern Sahelian (Taffogo), northern Sudanian (Nobere, Safane) and southern Sudanian (Sokouraba) zone of the country during 2009 and 2010. The choice of two villages in the northern Sudanian zone was guided by the dichotomy between intense agricultural land use and high population density near Safane, and lower agricultural land use in the tampon zone between the village of Nobere and the National Park Kaboré Tambi of Pô. Using global positioning and geographical information systems tools, the spatio-temporal variation in the use of grazing areas by cattle, sheep and goats, and in their foraging behaviour in the four villages was assessed by monitoring three herds each per species during a one-year cycle (Chapter 2). Maximum itinerary lengths (km/d) were observed in the hot dry season (March-May); they were longer for sheep (18.8) and cattle (17.4) than for goats (10.5, p<0.05). Daily total grazing time spent on pasture ranged from 6 - 11 h with cattle staying longer on pasture than small ruminants (p<0.05). Feeding time accounted for 52% - 72% of daily time on pasture, irrespective of species. Herds spent longer time on pasture and walked farther distances in the southern Sahelian than the two Sudanian zones (p<0.01), while daily feeding time was longer in the southern Sudanian than in the other two zones (p>0.05). Proportional time spent resting decreased from the rainy (June - October) to the cool (November - February) and hot dry season (p<0.05), while in parallel the proportion of walking time increased. Feeding time of all species was to a significantly high proportion spent on wooded land (tree crown cover 5-10%, or shrub cover >10%) in the southern Sahelian zone, and on forest land (tree crown cover >10%) in the two Sudanian zones, irrespective of season. It is concluded that with the expansion of cropland in the whole region, remaining islands of wooded land, including also fields fallowed for three or more years with their considerable shrub cover, are particularly valuable pasturing areas for ruminant stock. Measures must be taken that counteract the shrinking of wooded land and forests across the whole region, including also active protection and (re)establishment of drought-tolerant fodder trees. Observation of the selection behaviour of the above herds of cattle and small ruminant as far as browse species were concerned, and interviews with 75 of Fulani livestock keepers on use of browse as feed by their ruminant stock and as remedies for animal disease treatment was undertaken (Chapter 3) in order to evaluate the consequence of climate change for the contribution of browse to livestock nutrition and animal health in the extensive grazing-based livestock systems. The results indicated that grazing cattle and small ruminants do make considerable use of browse species on pasture across the studied agro-ecological zones. Goats spent more time (p<0.01) feeding on browse species than sheep and cattle, which spent a low to moderate proportion of their feeding time on browsing in any of the study sites. As far as the agro-ecological zones were concerned, the contribution of browse species to livestock nutrition was more important in the southern Sahelian and northern Sudanian zone than the southern Sudanian zone, and this contribution is higher during the cold and hot dry season than during the rainy season. A total of 75 browse species were selected on pasture year around, whereby cattle strongly preferred Afzelia africana, Pterocarpus erinaceus and Piliostigma sp., while sheep and goats primarily fed on Balanites aegyptiaca, Ziziphus mauritiana and Acacia sp. Crude protein concentration (in DM) of pods or fruits of the most important browse species selected by goats, sheep and cattle ranged from 7% to 13% for pods, and from 10% to 18% for foliage. The concentration of digestible organic matter of preferred browse species mostly ranged from 40% to 60%, and the concentrations of total phenols, condensed tannins and acid detergent lignin were low. Linear regression analyses showed that browse preference on pasture is strongly related to its contents (% of DM) of CP, ADF, NDF and OM digestibility. Interviewed livestock keepers reported that browse species are increasingly use by their grazing animals, while for animal health care use of tree- and shrub-based remedies decreased over the last two decades. It is concluded that due to climate change with expected negative impact on the productivity of the herbaceous layer of communal pastures browse fodder will gain in importance for animal nutrition. Therefore re-establishment and dissemination of locally adapted browse species preferred by ruminants is needed to increase the nutritional situation of ruminant stock in the region and contribute to species diversity and soil fertility restoration in degraded pasture areas. In Chapter 4 a combination of household surveys and participatory research approaches was used in the four villages, and additionally in the village of Zogoré (southern Sahelian zone) and of Karangasso Vigué (northern Sudanian zone) to investigate pastoralists’ (n= 76) and agro-pastoralists’ (n= 83) perception of climate change, and their adaptation strategies in crop and livestock production at farm level. Across the three agro-ecological zones, the majority of the interviewees perceived an increase in maximum day temperatures and decrease of total annual rainfall over the last two decades. Perceptions of change in climate patterns were in line with meteorological data for increased temperatures while for total rainfall farmers’ views contrasted the rainfall records which showed a slight increase of precipitation. According to all interviewees climate change and variability have negative impacts on their crop and animal husbandry, and most of them already adopted some coping and adaptation strategies at farm level to secure their livelihoods and reduce negative impacts on their farming system. Although these strategies are valuable and can help crop and livestock farmers to cope with the recurrent droughts and climate variability, they are not effective against expected extreme climate events. Governmental and non-governmental organisations should develop effective policies and strategies at local, regional and national level to support farmers in their endeavours to cope with climate change phenomena; measures should be site-specific and take into account farmers’ experiences and strategies already in place.

En el presente estudio se analizan los efectos de los fotoperiodos ambientales de otoño e invierno y los fotoperiodos experimentales de 24, 12 y 0 horas de luz artificial sobre la calidad del semen de machos reproductores porcinos de raza Landrace. El estudio se realizó sobre 30 machos postpuberales de 8 meses de edad y testados con el fin de comprobar la homogeneidad. Los machos fueron distribuidos aleatóriamente en los 3 grupos de luz artificial durante 3 meses. El tratamiento de 12 horas de luz artificial fue considerado como grupo control. Previamente al inicio de cada tratamiento, se sometió a cada grupo de machos al fotoperiodo ambiental correspondiente a la época del año; así, se caracterizó también la calidad seminal en otoño e invierno, manteniendo la temperatura constante. La nave experimental que acogió a los machos tiene una superficie de 100 m2 y una altura de 3,5 m. Un pasillo central divide la nave en dos hileras de 5 y 6 celdas respectivamente. En una de las celdas pequeñas se instaló el maniquí y fue utilizada para las extracciones de semen. La iluminación artificial se consiguió con la instalación de 6 lámparas fluorescentes en el techo del pasillo central que proporcionaron una luz homogénea superior a 200 lux. Así mismo, la nave se mantuvo en todo momento a 21±1ºC y la humedad relativa osciló entre el 60-75%. A todos los verracos se les proporcionó una dieta nutritiva y equilibrada y se les sometió a un regimen de extracciones de semen de 2 veces por semana, habiendo sido previamente entrenados en la monta del maniquí. Les muestras de semen fueron recogidas según la técnica de la mano enguantada (Martín, 1982; Daza, 1992) y se analizaron los siguientes parámetros: el volumen y el pH seminales, la concentración, la vitalidad y la motilidad espermáticas, la resistencia acrosómica de los espermatozoides, la morfología espermàtica a partir de la frecuencia de los espermatozoides maduros, inmaduros y aberrantes, la producción testicular y el número de dosis seminales. Se analizó, además, bioquímicamente el plasma seminal al principio y al final de cada tratamiento experimental de luz artificial a partir de la concentración de proteína total, de la identificación de residuos fosforilados de proteína y del contenido de azúcares. También se determinaron los índices de fertilidad y prolificidad. El volumen y el pH de los eyaculados se utilizaron como marcadores del estado funcional de las glándulas sexuales accesorias; la concentración espermàtica como un indicador de la actividad testicular (Pinart y col., 1999). La vitalidad y la motilidad espermáticas fueron estimadores del grado de diferenciación del espermatozoide tanto a nivel testicular como epididimario; la resistencia acrosómica fue utilizada para valorar el nivel de diferenciación de la membrana acrosómica durante la espermiogénesis y/o maduración epididimària (Briz i col., 1996; Pinart i col., 1999). Referente a la morfología espermática, los espermatozoides inmaduros fueron marcadores de anomalías en la maduración de éstos a lo largo del conducto epididimario y los espermatozoides aberrantes se utilizaron como marcadores de una diferenciación defectuosa a nivel de testículo (anomalías primarias) y a nivel de conducto epididimario (anomalías secundarias) (Briz i col., 1996). La concentración de proteína total se utilizó para valorar la integridad funcional de las membranas del espermatozoide y la actividad de las glándulas sexuales accesorias. La identificación de proteínas con residuos de tirosina fosforilados fue un estimador de la viabilidad celular y la actividad de las glándulas sexuales, y el contenido de azúcares como un indicador de la producción de las vesículas seminales. La determinación del volumen y el pH de los eyaculados se realizó en las instalaciones de la granja a partir de semen fresco el mismo día de la extracción. El resto de parámetros se analizaron en el laboratorio durante las 48 horas posteriores a la extracción a partir de semen diluido en BTS (diluyente de Bestville) (Daza, 1992) y transportado y conservado a 15ºC. Las muestras fueron previamente filtradas con el fin de eliminar la tapioca. El estudio estadístico de los resultados obtenidos se realizó a partir del análisis de la varianza (ANOVA) con un nivel de significación de =0,05. En cuanto al estudio comparativo de los fotoperiodos ambientales estacionales se ha observado un incremento significativo del pH del eyaculado en los machos expuestos a otoño (P0,0001), mientras que el volumen seminal se mantiene en valores similares en ambos tratamientos (P=0,1650). La concentración espermàtica, la producción espermàtica y el número de dosis seminales que se pueden preparar a partir de un eyaculado se duplica en los verracos sometidos al fotoperiodo de primavera (P0,0001). La vitalidad y la motilidad espermáticas no experimentan cambios significativos entre tratamientos (P=0,3440 y P=0,9220, respectivamente). La resistencia osmótica de los acrosomas desciende únicamente en los machos expuestos a condiciones estacionales de otoño (P0,0001). En referencia a la morfología espermàtica aunque no se observan diferencias entre primavera y otoño (P0,05), sí se detecta un incremento de los porcentajes de espermatozoides inmaduros y aberrantes en ambos fotoperiodos estacionales, y en especial en los machos expuestos a condiciones fotoperiódicas de otoño. Según los resultados obtenidos en este estudio la calidad seminal de los verracos es inferior en el fotoperiodo de otoño debido a un descenso de la concentración y la producción espermáticas, un aumento del pH seminal, una disminución de la resistencia de la membrana acrosómica y a un incremento en la frecuencia de espermatozoides inmaduros y aberrantes. Parece ser, pues, que en el otoño tiene lugar la disminución de la producción testicular, cambios en la actividad de las glándulas sexuales accesorias y disfunciones en el proceso de diferenciación testicular y epididimària de los espermatozoides y especialmente del acrosoma. En relación a los resultados obtenidos en el estudio de los diferentes fotoperiodos artificiales se observa que la iluminación continua provoca un aumento significativo del volumen del eyaculado en el primer y segundo mes de tratamiento (P0,0001), disminuyendo en el tercer mes. La oscuridad absoluta no modifica este parámetro (P0,05). En cuanto al pH seminal la iluminación continua provoca un incremento progresivo del valor del pH a lo largo del periodo experimental (P0,0001), mientras que la oscuridad absoluta tiene un efecto más irregular. La exposición de los machos a iluminación continua y a oscuridad absoluta se manifiesta en un descenso de la concentración y la producción espermáticas que se mantiene hasta el segundo mes de tratamiento (P0,0001), observándose un incremento en el tercer mes de exposición de los machos a oscuridad absoluta (P=0,1010). De todas maneras, este descenso es mas severo en los machos sometidos a iluminación continua ya que no presentan recuperación. La vitalidad y la motilidad espermáticas no se ven alteradas por la iluminación continua y la oscuridad absoluta, ni tampoco el contenido de los azúcares mayoritarios del plasma seminal (P0,005). La glucosa aparece como un azúcar minoritario y sí que presenta concentraciones inferiores en los tratamientos experimentales de luz continua y de oscuridad absoluta (P0,0001 y P=0,0002, respectivamente). La resistencia osmótica de los acrosomas desciende en ambos tratamientos artificiales extremos de luz continua y oscuridad total (P0,0001), aunque en los machos expuestos a iluminación continua se produce una recuperación a partir del segundo mes de tratamiento (P=0,4930). Dado que tampoco se han observado diferencias significativas en las concentraciones de proteína total (P0,05), es probable que las anomalías de la membrana acrosómica se originen durante el proceso de espermiogénesis y/o maduración epididimària. La exposición de los verracos a oscuridad absoluta no altera la morfología espermàtica de los eyaculados, aunque se observa un aumento de la frecuencia de espermatozoides con anomalías en la forma de la cola en el primer mes (P0,0001), y un aumento de la frecuencia de espermatozoides inmaduros con gota distal y de espermatozoides con anomalías en el número de colas en el tercer mes de experimentación (P=0,0030 y P0,0001). La luz continua, sin embargo, provoca un incremento de la frecuencia de espermatozoides inmaduros con gota distal (P0,0001) y de espermatozoides con anomalías en la forma de la cola (P=0,0040) ya en el primer mes. El fotoperiodo provoca un descenso de la fertilidad de los machos expuestos a oscuridad absoluta en el tercer mes de tratamiento (P0,0001) y un incremento de ésta en los machos sometidos a iluminación continua (P=0,0005). La prolificidad no se ve modificada por ambas condiciones extremas de luz artificial (P0,05). Así pues, los resultados obtenidos demuestran que el fotoperiodo afecta la actividad testicular, provoca alteraciones en la actividad de las glándulas sexuales accesorias, altera el proceso de expulsión de la gota citoplasmática y provoca anomalías en el proceso de diferenciación de la cola tanto a nivel testicular como epididimario, siendo los verracos expuestos a luz continua más sensibles a estos parámetros que los verracos sometidos a oscuridad absoluta. El fotoperiodo, sin embargo, no altera de forma esencial la integridad de las membranas del espermatozoide ni la capacidad fecundante de éste.

A fi d'analitzar la contribució de la regió C-terminal proposada com a iniciadora del plegament (CFIS 106-118) a l'estabilitat de l'RNasa A, els residus alifàtics d'aquesta regió es van substituir, mitjançant mutagènesi dirigida, per altres residus en els quals la cadena lateral alifàtica era rogressivament escurçada. La major part de les substitucions projectades suposaven delecions no disruptives de grups metil(è). A més, es va reemplaçar la Tyr115 per un Trp, de manera que, potencialment, s'introduïa una única sonda fluorescent, no desestabilitzant, per tal de seguir els canvis conformacionals que es poguessin generar en la regió durant el procés de legament/desplegament de la proteïna. Tant els paràmetres cinètics, com els espectres d'FTIR i CD, determinats per cadascuna de les ribonucleases variants, indiquen que els reemplaçaments aminoacídics efectuats presenten, en general, poc o cap efecte en l'estructura nativa i en l'activitat de l'enzim. Es va emprar l'espectroscòpia d'absorció a l'ultraviolat de quarta derivada, la fluorescència (per la variant amb Trp) i l'espectroscòpia d'infraroig per transformada de Fourier, per tal de seguir i caracteritzar, en condicions d'equilibri, les transicions conformacionals de cada variant en funció de la pressió i de la temperatura. Els resultats es van comparar amb els que es van obtenir per la proteïna salvatge. Per determinar més a fons les característiques del procés de desplegament de la variant Y115W, les transicions de desnaturalització induïdes per urea d'aquesta variant i de la proteïna salvatge, van ésser examinades per mitjà d'electroforesi en gradient d'urea i espectroscòpia de fluorescència. Curiosament, els canvis conformacionals que resulten de la desnaturalització per pressió són molt semblants als que s'obtenen per temperatura. Enfront d'un augment gradual tant de pressió com de temperatura, l'estructura terciària i els elements d'estructura secundària de les proteïnes estudiades es perden de manera conjunta i reversible. Aquestes variacions estructurals que es promouen descriuen un procés de desplegament molt cooperatiu i en dos estats. Atès que ambdues tècniques (UV i FTIR) utilitzen cadascuna un règim de concentració proteica molt diferent, els resultats indiquen que el procés de desplegament per pressió i per temperatura és intramolecular. Els resultats obtinguts suggereixen que la hidrofobicitat i el volum de les cadenes laterals del CFIS, juntament amb les interaccions de van der Waals entre elements d'estructura secundària intervenen de manera molt notable en l'estabilització de la proteïna. Entre els diferents aminoàcids alifàtics que pertanyen al CFIS C-terminal, la Val108 és el residu més important per tal de preservar la integritat estructural de l'estat natiu. Els reemplaçaments en aquesta posició causen petites alteracions conformacionals i una gran desestabilització de la proteïna (per exemple, el punt mig de la transició de desnaturalització per pressió i per temperatura de la variant V108G disminueix uns 592 MPa i 25ºC, respectivament, respecte a la proteïna salvatge). D'acord amb els resultats obtinguts, la variant Y115W ofereix una sonda útil per tal de seguir la cinètica de plegament/desplegament de l'RNasa A.

Eight Jersey cows were used in two balanced 4 x 4 Latin Squares to investigate the effects of replacement of dietary starch with non-forage fibre on productivity, diet digestibility and feeding behaviour. Total-mixed rations consisted of maize silage, grass silage and a soyabean meal-based concentrate mixture, each at 250g/kg DM, with the remaining 250g consisting of cracked wheat/soya hulls (SH) in the ratios of 250:0, 167:83; 83:167 and 0:250 g, respectively, for treatments SH0, SH83, SH167 and SH250. Starch concentrations were 302, 248, 193 and 140g/kg DM, and NDF concentrations were 316, 355, 394 and 434g/kg DM, for treatments SHO, SH83, SH167 and SH250, respectively. Total eating time increased (p < 0.05) as SH inclusion increased, but total rumination time was unaffected. Digestibility of DM, organic matter and starch declined (p < 0.01) as SH inclusion increased, whilst digestibility of NDF and ADF increased (p < 0.01). Dry-matter intake tended to decline with increasing SH, whilst bodyweight, milk yield and fat and lactose concentrations were unaffected by treatment. Milk protein concentration decreased (p < 0.01) as SH level increased. Feed conversion efficiency improved (p < 0.05) as SH inclusion rose, but it was not possible to determine whether this was due to the increased fibre levels alone, or the favourable effect on rumen fermentation of decreasing starch levels. A study was carried out to determine the influence of fibrolytic enzymes derived from mesophilic or thermophilic fungal sources, added at ensiling, on time-course fermentation characteristics and in vitro rumen degradation of maize silage. The mesophilic enzyme was a commercial product derived from Trichodenna reesei (L), whereas the thermophilic enzyme was a crude extract produced from Thermoascus aurantiacus (Ta) in this laboratory. The fungus was cultured using maize cobs as a carbon source. The resulting fermentation extract was deionised to remove sugars and characterised for its protein concentration, main and side enzymic activities, optimal pH, protein molecular mass and isoelectric point. In an additional study, both enzymes were added to maize forage (333.5 g DM/kg, 70.0, 469.8, 227.1 and 307.5 g/kg DM of CP, NDF, ADF and starch, respectively) at two levels each, normalized according to xylanase activity, and ensiled in 0.5 kg capacity laboratory minisilos. Duplicate silos were opened at 2, 4, 8, 15, and 60 days after ensiling, and analysed for chemical characteristics. Silages from 60 days were bulked and in vitro gas production (GP) and organic matter degradability (OMD) profiles evaluated using the Reading Pressure Technique (RPT), in a completely randomised design. The crude enzyme extract contained mainly xylanase and endoglucanase activities, with very low levels of exoglucanase, which probably limited hydrolysis of filter paper. The extract contained three major protein bands of between 29 and 55 kDa, with mainly acidic isoelectric points. Ensiling maize with enzymes lowered (P < 0.05) the final silage pH, with this effect being observed throughout the ensiling process. All enzyme treatments reduced (P < 0.05) ADF contents. Treatments including Ta produced more gas (P < 0.05) than the controls after 24 h incubation in vitro, whereas end point gas production at 96 h was not affected. Addition of Ta increased (P < 0.01) OMD after 12 h (410 and 416 g/kg versus 373 g/kg), whereas both L and Ta increased (P < 0.05) OMD after 24 h. Addition of enzymes from mesophilic or thermophilic sources to maize forage at ensiling increased the rate of acidification of the silages and improved in vitro degradation kinetics, suggesting an improvement in the nutritive quality. Twenty-eight field experiments on sandy-loam soils in the UK (1982-2003) are reviewed by relating the extension of the green area duration of the flag leaf (GLADF) by fungicides to effects on yield and quality of winter wheat. Over all experiments mean grain yield = 8.85t ha(-1) at 85% DM. With regards quality, mean values were: thousand grain weight (TGW) = 44.5 g; specific weight (SWT) = 76.9 kg hl(-1); crude protein concentration (CP (N x 5.7)) = 12.5 % DM; Hagberg falling number (HFN) = 285 s; and sodium dodecyl sulphate (SDS)-sedimentation volume = 69ml. For each day (d) that fungicides increased GLADF there were associated average increases in yield (0.144 1 ha(-1) d(-1), se 0.0049, df = 333), TGW (0.56 gd(-1), se = 0.017) and SWT (0.22 kg hl(-1) d(-1), se 0.011). Some curvature was evident in all these relationships. When GLADF was delayed beyond 700 degrees Cd after anthesis, as was possible in cool wet seasons, responses were curtailed, or less reliable. Despite this apparent terminal sink limitation, fungicide effects on sink size, eg endosperm cell numbers or maximum water mass per grain, were not prerequisites for large effects on grain yield, TGW or SWT. Fungicide effects on CP were variable. Although the average response of CP was negative (-0.029%DM/d; se = 0.00338), this depended on cultivar and disease controlled. Controlling biotrophs such as rusts, (Puccinia spp.) tended to increase CP, whereas controlling a more necrotrophic pathogen (Septoria tritici) usually reducedCP. Irrespective of pathogen controlled, delaying senescence of the flag leaf was associated with increased nitrogen yields in the grain (averaging 2.24 kg N ha-1 d(-1), se = 0.0848) due to both increased N uptake into the above ground crop, and also more efficient remobilisation of N from leaf laminas. When sulphur availability appeared to be adequate, fungicide x cultivar interactions were similar on S as for CP, although N:S ratios tended to decline (i.e. improve for bread making) when S. tritici was controlled. On average, SDS-sedimentation volume declined (-0. 18 ml/d, se = 0.027) with increased GLADF, broadly commensurate with the average effect on CP. Hagberg falling number decreased as fungicide increased GLADF (-2.73 s/d, se = 0.178), indicating an increase in alpha-amylase activity.

The effects of applying nitrogen (30 or 40 kg N/ha) to wheat crops at and after anthesis, after 200 kg N/ha had already been applied to the soil during stem extension, were studied in field experiments comprising complete factorial combinations of different cultivars, fungicide applications and nitrogen treatments. Actual recoveries of late-season fertilizer nitrogen (LSFN), as indicated by N-15 studies, interacted with cultivar and fungicide treatment, and depended on nitrogen source (Urea applied as a solution to the foliage, or as ammonium nitrate applied to the soil) and year. These interactions, however, were not reflected in apparent fertilizer recoveries ((N in grain with LSFN - N in grain without LSFN)/N applied as LSFN), or in the crude protein concentration. Apparent fertilizer recovery was always lower than actual recoveries, and declined during grain filling. Fertilizer treatments with higher actual fertilizer recoveries were associated with lower net renlobilisation of non-LSFN (net remobilised N = N in above ground crop at anthesis - N in non-grain, above ground crop at harvest). LSFN also increased mineral nitrogen in the soil at harvest even when applied as a solution to the foliage. These effects are discussed in relation to potential grain N demand. (c) 2006 Elsevier B.V. The effects of intercropping wheat with faba bean (Denmark, Germany, Italy and UK) and wheat with pea (France), in additive and replacement designs on grain nitrogen and sulphur concentrations were studied in field experiments in the 2002/03, 2003/04 and 2004/05 growing seasons. Intercropping wheat with grain legumes regularly increased the nitrogen concentration of the cereal grain, irrespective of design or location. Sulphur concentration of the cereal was also increased by intercropping, but less regularly and to a lesser extent compared with effects on nitrogen concentration. Nitrogen concentration (g/kg) in wheat additively intercropped with faba bean was increased by 8% across all sites (weighted for inverse of variance), but sulphur concentration was only increased by 4%, so N:S ratio was also increased by 4%. Intercropping wheat with grain legumes increased sodium dodecyl sulphate (SDS)-sedimentation volume. The effect of intercropping on wheat nitrogen concentration was greatest when intercropping had the most deleterious effect on wheat yield and the least deleterious effect on pulse yield. Over all sites and seasons, and irrespective of whether the design was additive or replacement, increases in crude protein concentration in the wheat of 10 g/kg by intercropping with faba bean were associated with 25-30% yield reduction of the wheat, compared with sole-cropped wheat. It was concluded that the increase in protein concentration of wheat grain in intercrops could be of economic benefit when selling wheat for breadmaking, but only if the bean crop was also marketed effectively.

Sixteen multiparous Holstein cows were used to determine the effects of 2-hydroxy-4-(methylthio) butanoic acid isopropyl ester (HMBi: 0 vs. 1.26 g/kg of total ration dry matter (DM) and dietary crude protein (CP) concentration [14.7% (low) vs. 16.9% (standard), DM basis] on milk yield and composition using a replicated 4 x 4 Latin square design experiment with 4-wk periods. Cows were fed ad libitum a total mixed ration with a 1: 1 forage-to-concentrate ratio (DM basis), and diets provided an estimated 6.71 and 1.86% lysine and methionine, respectively, in metabolizable protein for the low-protein diet and 6.74 and 1.82% in the standard protein diet. Dry matter intake, milk yield, and composition were measured during wk 4 of each period. There were no effects on DM intake, which averaged 24.7 kg/d. There was an interaction between dietary CP and HMBi for milk yield and 3.5% fat-corrected milk (FCM). Feeding HMBi decreased milk and FCM yield when fed with the low-CP diet but did not affect milk or FCM yield when fed with the standard CP diet. Feeding HMBi increased milk protein concentration regardless of diet CP concentration and increased milk protein yield when added to the standard CP diet but not the low-CP diet. The positive effect of HMBi on milk protein yield was only observed at the standard level of dietary CP, suggesting other factors limited the response to HMBi when dietary protein supply was restricted.

The extensive development of the ruminant forestomach sets apart their N economy from that of nonruminants in a number of respects. Extensive pregastric fermentation alters the profile of protein reaching the small intestine, largely through the transformation of nitrogenous compounds into microbial protein. This process is fueled primarily by carbohydrate fermentation and includes extensive recycling of N between the body and gut lumen pools. Nitrogen recycling occurs via blood and gut lumen exchanges of urea and NH3, as well as endogenous gut and secretory N entry into the gut lumen, and the subsequent digestion and absorption of microbial and endogenous protein. Factors controlling urea transfer to the gut from blood, including the contributions of urea transporters, remain equivocal. Ammonia produced by microbial degradation of urea and dietary and endogenous AA is utilized by microbial fermentation or absorbed and primarily converted to urea. Therefore, microbial growth and carbohydrate fermentation affect the extent of NH3 absorption and urea N recycling and excretion. The extensive recycling of N to the rumen represents an evolutionary advantage of the ruminant in terms of absorbable protein supply during periods of dietary protein deficiency, or asynchronous carbohydrate and protein supply, but incurs a cost of greater N intakes, especially in terms of excess N excretion. Efforts to improve the efficiency of N utilization in ruminants by synchronizing fermentable energy and N availability have generally met with limited success with regards to production responses. In contrast, imposing asynchrony through oscillating dietary protein concentration, or infrequent supplementation, surprisingly has not negatively affected production responses unless the frequency of supplementation is less than once every 3 d. In some cases, oscillation of dietary protein concentration has improved N retention compared with animals fed an equal amount of dietary protein on a daily basis. This may reflect benefits of Orn cycle adaptations and sustained recycling of urea to the gut. The microbial symbiosis of the ruminant is inherently adaptable to asynchronous N and energy supply. Recycling of urea to the gut buffers the effect of irregular dietary N supply such that intuitive benefits of rumen synchrony in terms of the efficiency of N utilization are typically not observed in practice.

Field experiments were conducted over 3 years to study the effect of applying triazole and strobilurin fungicides on the bread-making quality of Malacca winter wheat. Averaged over all years the application of a fungicide programme increased yields, particularly when strobilurin fungicides were applied. Reductions in protein concentration, sulphur concentration, Hageberg failing number and loaf volumes also occurred as the amount of fungicide applied increased. However, there were no deleterious effects of fungicide application on sodium dodecyl sulphate (SDS) sedimentation volumes, N:S ratios or dough theology. Effects of fungicide application on bread-making quality were not product specific. Therefore, it appears that new mechanisms to explain strobilurin effects on bread-making quality do not need to be invoked. Where reductions in protein concentration did occur they could be compensated for by a late-season application of nitrogen either as granular ammonium nitrate at flag leaf emergence or foliar urea at anthesis. These applications, however, sometimes increased the N:S ratio of the extracted flour and failed to improve loaf volume. Multiple regression analysis revealed that main effects of year, flour protein concentration and N:S ratio could explain 93% of the variance in loaf volume caused by season, fungicide and nitrogen treatments. Field experiments were conducted over 3 years to assess the effect of a triazole fungicide programme, and additions of strobilurin fungicides to it, on nitrogen uptake, accumulation and partitioning in a range of winter wheat cultivars. Commensurate with delayed senescence, fungicide programmes, particularly when including strobilurins, improved grain yield through improvements in both crop biomass and harvest index, although the relationship with green area duration of the flag leaf (GFLAD) depended on year and in some cases, cultivar. In all years fungicide treatments significantly increased the amount of nitrogen in the above-ground biomass, the amount of nitrogen in the grain and the nitrogen harvest index. All these effects could be linearly related to the fungicide effect on GFLAD. These relationships occasionally interacted with cultivar but there was no evidence that fungicide mode of action affected the relationship between GFLAD and yield of nitrogen in the grain. Fungicide treatments significantly reduced the amount of soil mineral N at harvest and when severe disease had been controlled, the net remobilization of N from the vegetation to the grain after anthesis. Fungicide maintained the filling of grain with both dry matter and nitrogen. The proportionate accumulation of nitrogen in the grain was later than that of dry matter and this difference was greater when fungicide had been applied. Effects of fungicide on grain protein concentration and its relationship with GFLAD were inconsistent over year and cultivar. There were several instances where grain protein concentration was unaffected despite large (1(.)5 t/ha) increases in grain yield following fungicide use. Dilution of grain protein concentration following fungicide use, when it did occur, was small compared with what would be predicted by adoption of other yield increasing techniques such as the selection of high yielding cultivars (based on currently available cultivars) or by growing wheat in favourable climates.
