Understanding the biological activity profile of the snake venom components is fundamental for improving the treatment of snakebite envenomings and may also contribute for the development of new potential therapeutic agents. In this work, we tested the effects of BthTX-I, a Lys49 PLA2 homologue from the Bothrops jararacussu snake venom. While this toxin induces conspicuous myonecrosis by a catalytically independent mechanism, a series of in vitro studies support the hypothesis that BthTX-I might also exert a neuromuscular blocking activity due to its ability to alter the integrity of muscle cell membranes. To gain insight into the mechanisms of this inhibitory neuromuscular effect, for the first time, the influence of BthTX-I on nerve-evoked ACh release was directly quantified by radiochemical and real-time video-microscopy methods. Our results show that the neuromuscular blockade produced by in vitro exposure to BthTX-I (1 μM) results from the summation of both pre- and postsynaptic effects. Modifications affecting the presynaptic apparatus were revealed by the significant reduction of nerve-evoked [3H]-ACh release; real-time measurements of transmitter exocytosis using the FM4-64 fluorescent dye fully supported radiochemical data. The postsynaptic effect of BthTX-I was characterized by typical histological alterations in the architecture of skeletal muscle fibers, increase in the outflow of the intracellular lactate dehydrogenase enzyme and progressive depolarization of the muscle resting membrane potential. In conclusion, these findings suggest that the neuromuscular blockade produced by BthTX-I results from transient depolarization of skeletal muscle fibers, consequent to its general membrane-destabilizing effect, and subsequent decrease of evoked ACh release from motor nerve terminals. © 2012 Elsevier Ltd.

Isoflavones are phenolic compounds widely distributed in plants and found in a high percentage in soybeans. They have important biological properties and are regarded as potential chemopreventive agents. The aim of this study was to verify the preventive effect of two soy isoflavones (genistein and daidzein) by a micronucleus assay, analysis of GST activity, and real-time RT-PCR analysis of GSTa2 gene expression. Mutagens of direct (doxorubicin) and indirect (2-aminoanthracene) DNA damage were used. Hepatoma cells (HTC) were treated with genistein or daidzein for 26 h at noncytotoxic concentrations; 10 μM when alone, and 0.1, 1.0 and 10 μM when combined with genotoxic agents. The micronucleus test demonstrated that both isoflavones alone had no genotoxic effect. Genistein showed antimutagenic effects at 10 μM with both direct and indirect DNA damage agents. On phase II enzyme regulation, the current study indicated an increase in total cytoplasmic GST activity in response to genistein and daidzein at 10 μM supplementation. However, the mRNA levels of GSTa2 isozymes were not differentially modulated by genistein or daidzein. The results point to an in vitro antimutagenic activity of genistein against direct and indirect DNA damage-induced mutagenicity. © 2012 Springer Science+Business Media B.V.

The peptide LYS-[TRP6]-Hy-A1 (Lys-a1) is a synthetic derivative of the peptide Hy-A1, initially isolated from the frog species Hypsiboas albopunctatus. According to previous research, it is a molecule with broad antimicrobial activity. The objective of this study was to evaluate the antimicrobial activity of the synthetic peptide Lys-a1 (KIFGAIWPLALGALKNLIK- NH2) on the planktonic and biofilm growth of oral bacteria. The methods used to evaluate antimicrobial activity include the following: determination of the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) in microtiter plates for growth in suspension and quantification of biomass by crystal violet staining and counting of colony forming units for biofilm growth. The microorganisms Streptococcus oralis, Streptococcus sanguinis, Streptococcus parasanguinis, Streptococcus salivarius, Streptococcus mutans and Streptococcus sobrinus were grown in Brain Heart Infusion broth at 37 °C under atmospheric pressure with 10% CO2. The peptide was solubilized in 0.1% acetic acid (v/v) at various concentrations (500-1.9 μg mL-1). Chlorhexidine gluconate 0.12% was used as the positive control, and BHI culture medium was used as the negative control. The tested peptide demonstrated a remarkable antimicrobial effect, inhibiting the planktonic and biofilm growth of all strains tested, even at low concentrations. Thus, the peptide Lys-a1 is an important source for potential antimicrobial agents, especially for the control and prevention of microbial biofilms, which is one of the most important factors in cariogenic processes. © 2012 Elsevier Inc.

Chagas disease is one of the main public health problems in Latin America. Since the available treatments for this disease are not effective in providing cure, the screening of potential antiprotozoal agents is essential, mainly of those obtained from natural sources. This study aimed to provide an evaluation of the trypanocidal activity of 92 ethanol extracts from species belonging to the families Annonaceae, Apiaceae, Cucurbitaceae, Lamiaceae, Lauraceae, Moraceae, Nyctaginaceae, and Verbenaceae against the Y and Bolivia strains of Trypanosoma cruzi. Additionally, cytotoxic activity on LLCMK2 fibroblasts was evaluated. Both the trypanocidal activity and cytotoxicity were evaluated using the MTT method, in the following concentrations: 500, 350, 250, and 100 µg/mL. Benznidazole was used for positive control. The best results among the 92 samples evaluated were obtained with ethanol extracts of Ocotea paranapiacabensis (Am93) and Aegiphila lhotzkiana (Am160). Am93 showed trypanocidal activity against epimastigote forms of the Bolivia strain and was moderately toxic to LLCMK2 cells, its Selectivity Index (SI) being 14.56, while Am160 showed moderate trypanocidal activity against the Bolivia strain and moderate toxicicity, its SI being equal to 1.15. The screening of Brazilian plants has indicated the potential effect of ethanol extracts obtained from Ocotea paranapiacabensis and Aegiphila lhotzkiana against Chagas disease.

Sporotrichosis is a subcutaneous mycosis and is also a zoonosis (sapro- and anthropozoonosis). The objective of the present study was to determine the occurrence of sporotrichosis in domestic cats and in wild or exotic felines in captivity through the isolation of Sporothrix spp. from claw impressions in a culture medium. The samples included 132 felines, of which 120 (91.0 %) were domestic cats, 11 (8.3 %) were wild felines, and one (0.7 %) was an exotic felid. Twenty-one (17.5 %) were outdoor cats. Of the total, 89 (67.4 %) had contact with other animals of the same species. It was possible to isolate Sporothrix schenckii from the claws of one (0.7 %) of the felids probed; this animal exhibited generalised sporotrichosis and had infected a female veterinarian. The potential pathogenic agents Microsporum canis and Malassezia pachydermatis were isolated in 12.1 and 5.3 % of the animals, respectively. The following anemophilous fungi, which were considered to be contaminants, were also isolated: Penicillium sp. (28 or 21.2 %), Aspergillus sp. (13 or 9.8 %), Rhodotorula sp. (5 or 3.8 %), Candida sp. (5 or 3.8 %), Trichoderma sp. (1 or 0.7 %), and Acremonium sp. (1 or 0.7 %). Due to the low magnitude of occurrence (0.7 %) of Sporothrix in feline claws, the potential of the cats evaluated in this study to be sources of infection in the city of São Paulo is considerably low.

The aim of the research project discussed in this thesis was to study the inhibition of aerobic glycolysis, that is the metabolic pathway exploited by cancer cells for the ATP generation. This observation has led to the evaluation of glycolytic inhibitors as potential anticancer agents. Lactate dehydrogenase (LDH) is the only enzyme whose inhibition should allow a blocking of aerobic glycolysis of tumor cells without damaging the normal cells which, in conditions of normal functional activity and sufficient oxygen supply, do not need this enzyme. In preliminar experiments we demonstrated that oxamic acid and tartronic acid, two LDH competitive inhibitors, impaired aerobic glycolysis and replication of cells from human hepatocellular carcinoma. Therefore, we proposed that the depletion of ATP levels in neoplastic cells, could improved the chemotherapeutic index of associated anticancer drugs; in particular, it was studied the association of oxamic acid and multi-targeted kinase inhibitors. A synergistic effect in combination with sorafenib was observed, and we demonstrated that this was related to the capacity of sorafenib to hinder the oxidative phosphorylation, so that cells were more dependent to aerobic glycolysis. These results linked to LDH blockage encouraged us to search for LDH inhibitors more powerful than oxamic acid; thus, in collaboration with the Department of Pharmaceutical Sciences of Bologna University we identified a new molecule, galloflavin, able to inhibit both A and B isoforms of LDH enzyme. The effects of galloflavin were studied on different human cancer cell lines (hepatocellular carcinoma, breast cancer, Burkitt’s lymphoma). Although exhibiting different power on the tested cell lines, galloflavin was constantly found to inhibit lactate and ATP production and to induce cell death, mainly in the form of apoptosis. Finally, as LDH-A is able to bind single stranded DNA, thus stimulating cell transcription, galloflavin effects were also studied on this other LDH function.

Die hochspezifische Funktionalisierung von Proteinen und Peptiden kann durch milde reduktive Spaltung der lösungsmittelzugänglichen Disulfidbrücken und anschließende Rückverbrückung durch den Einbau sogenannter Linkermoleküle über einen konsekutiven Eliminierungs-Additionsprozess verwirklicht werden. Die Erweiterung des Linkerportfolios stellte in erster Instanz die Entwicklung von verschieden funktionalisierten Systemen dar, welche als hochflexible Kernbausteine für den Aufbau komplexer Architekturen dienten. Das Verständnis für die Reaktivität und Reversibilität der Thioladdition an die Mono-und Bissulfone in Abhängigkeit des Substituenten in p-Position konnte durch Variation von Parametern wie Lösungsmittel oder pH-Wert für intelligentes Produktdesign genutzt werden. Heterokonjugate zweier Biomoleküle mit ungepaartem Cystein wurden durch die Kombination von Maleinimid- und Bissulfonchemie innerhalb eines Linkermoleküls realisiert. Polymer-Peptid-Konjugate wurden einerseits über die grafting to Methode durch Modifizierung von Somatostatin mit PEGbissulfonen und anderseits durch grafting from unter Verwendung eines zuvor synthetisierten ATRP-Makroinitiators dargestellt. Multivalente Konjugate konnten durch die Synthese von hochsymmetrischen Tetra- sowie Hexasulfonen und anschließende Umsetzung mit Somatostatin erhalten werden. Die Polyinterkalatorpolymere, die durch lebende radikalische Polymerisation eines Bissulfidmonomers generiert wurden, wurden mit Glutathion umgesetzt. Durch die Interkalation von p-Ethinyl sowie p-Iodmonosulfon in die Disulfidbrücke von Somatostatin konnte erfolgreich gezeigt werden, dass die Rückverbrückung unter Rezyklisierung gelang. Die biologische Integrität wurde durch die Modifikation nicht beeinträchtigt und die erfolgreiche Aufnahme wurde nur bei den rezeptorpositiven Zellen (CAPAN-2) beobachtet. Das artifizielle Iodderivat im Vergleich zum nativen Somatostatin ein erhöhtes Potential zur Apoptoseinduktion. Die Somatostatinderivate präsentierten sich somit als attraktive potentielle Therapeutika.

Larven der Eulenfalter, Gattung Agrotis (Lepidoptera: Noctuidae), sind Schädlinge in der Landwirtschaft, welche gravierende Fraßschäden an bodennahen Pflanzenteilen verursachen. Häufig kommt es zum Absterben der noch jungen Pflanzen oder zu Beschädigungen der pflanzlichen Produkte, was zu finanziellen Ertragsverlusten führt. Zwei der wichtigsten landwirtschaftlichen Schädlinge der Gattung Agrotis sind die Larven der Saateule (Agrotis segetum) und der Ypsiloneule (Agrotis ipsilon), welche bisher überwiegend mittels chemischer Pestizide bekämpft werden. Als eine umweltfreundliche, nachhaltige und vielversprechende Alternative in der Bekämpfung wird der Einsatz von Baculoviren berücksichtigt. Baculoviren zeichnen sich durch eine hohe Virulenz und einem sehr engen Wirtsbereich aus. Häufig werden nur wenige nah verwandte Arten der gleichen Gattung infiziert. Aus der Gattung Agrotis wurden bisher mindestens vier Baculoviren isoliert und charakterisiert, welche als potentielle biologische Pflanzenschutzmittel in Frage kommen; sie gehören zu zwei Gattungen der Baculoviren: rnAlphabaculovirusrn(i) Agrotis segetum nucleopolyhedrovirus A (AgseNPV-A)rn(ii) Agrotis segetum nucleopolyhedrovirus B (AgseNPV-B)rn(iii) Agrotis ipsilon nucleopolyhedrovirus (AgipNPV)rnBetabaculovirusrn(i) Agrotis segetum granulovirus (AgseGV).rnDie Genome der AgseNPV-A, AgipNPV sowie des AgseGV wurden in vorherigen Studien bereits vollständig sequenziert und publiziert. In der vorgelegten Dissertation wurde das AgseNPV-B sequenziert und umfassend mit AgseNPV-A und AgipNPV verglichen. Das Genom von AgseNPV-B ist 148981 Kbp groß und kodiert ….. offene Leseraster. Phylogenetische Analysen zeigen eine enge Verwandtschaft dieser drei Viren und klassifizieren AgseNPV-B als eine neue Art innerhalb der Gattung Alphabaculovirus. Auf Basis der vorhandenen Genomsequenzen konnte eine PCR-basierende Methode zur Detektion und Quantifizierung on AgseNPV-A, AgseNPV-B, AgipNPV und AgseGV etabliert werden. Dises Verfahren ermöglichte die Quantifizierung von AgseNPV-B und AgseGV in Larven von A. segetum, die von beiden Viren zeitgleichinfiziert waren. Durch das gemeinsame Auftreten dieser beiden Wiren innerhalb eines Wirtsindividuums stellte sich die Frage, welche Art der Interaktion bei einer Ko-Infektion vorliegt. Durch Mischinfektionsversuche von AgseNPV-B und AgseGV konnte gezeigt werden, dass beide Viren um die Ressourcen der Larven konkurrieren. Eine für landwirtschaftliche Zwecke vorteilige Interaktion, wie das vorzeitige Verenden der Larven, das bereits für andere interagierende Baculoviren nachgewiesen wurde, konnte ausgeschlossen werden. Neben den Mischinfektionsversuchen wurden auch AgseGV und AgseNPV-B einzeln auf ihre Eignung als biologisches Pflanzenschutzmittel getestet. AgseGV zeigte in den Laborversuchen eine relativ langsame Wirkung, während AgseNPV-B durchaus Potential für ein rasche Abtötung besitzt. rnDie durchgeführten Aktivitätsstudien und die Charakterisierung von AgseNPV-B als neue Art erlauben ein vertieftes biologisches und molekulares Verständnis des Virus legen den Grundstein für und eine mögliche spätere Zulassung als Pflanzenschutzmittel. Die Methode zur Identifizierung und Quantifizierung der Agrotis-Baculoviren stellt ein wichtiges Instrument in der Qualitätskontrolle für Produzenten dar und ermöglicht zudem weitere Untersuchungen von Agrotis-Baculoviren in Mischinfektionen.

OBJECTIVE: Patients in the stomatology service of the Department of Oral Surgery and Stomatology who were clinically and histopathologically diagnosed with oral lichen planus (OLP) in the years 1995 to 2001 were examined for a possible malignant transformation of a previously biopsied OLP site. METHOD AND MATERIALS: For the 145 patients included, the recordings were searched for initial localization and type of OLP lesion, potential noxious agents, distribution between symptomatic and asymptomatic OLP types, and for a malignant transformation of a known OLP site during the follow-up period up to December 2003. RESULTS: The group comprised 47 men and 98 women with a mean age of 56.3 years. Of the 497 lesions, almost half were classified as reticular or papular, predominantly located on the buccal mucosa, gingiva, and borders of the tongue. Four patients did not adhere to their scheduled control visits and were dropped from the study. During the follow-up period 4 patients developed malignant transformation of OLP. In 3 of these cases, dysplasia was present at the initial diagnosis of OLP. This results in a malignant transformation rate of 2.84% among the remaining 141 patients; if the 3 patients with initial dysplasia are excluded, the rate drops to 0.71%. CONCLUSIONS: Until further knowledge is derived from large prospective studies, the data supporting or negating a potential malignant character of OLP lesions remains inconclusive. Special emphasis has to be directed toward unified inclusion and exclusion criteria regarding clinical and histologic findings and identifiable risk factors to allow the comparison of different studies.

Although an excitotoxic mechanism of neuronal injury has been proposed to play a role in chronic neurodegenerative disorders such as Alzheimer’s disease, and neurotrophic factors have been put forward as potential therapeutic agents, direct evidence is lacking. Taking advantage of the fact that mutations in the presenilin-1 (PS1) gene are causally linked to many cases of early-onset inherited Alzheimer’s disease, we generated PS1 mutant knock-in mice and directly tested the excitotoxic and neurotrophic hypotheses of Alzheimer’s disease. Primary hippocampal neurons from PS1 mutant knock-in mice exhibited increased production of amyloid β-peptide 42/43 and increased vulnerability to excitotoxicity, which occurred in a gene dosage-dependent manner. Neurons expressing mutant PS1 exhibited enhanced calcium responses to glutamate and increased oxyradical production and mitochondrial dysfunction. Pretreatment with either basic fibroblast growth factor or activity-dependent neurotrophic factor protected neurons expressing mutant PS1 against excitotoxicity. Both basic fibroblast growth factor and activity-dependent neurotrophic factor stabilized intracellular calcium levels and abrogated the increased oxyradical production and mitochondrial dysfunction otherwise caused by the PS1 mutation. Our data indicate that neurotrophic factors can interrupt excitotoxic neurodegenerative cascades promoted by PS1 mutations.

The telomeric G-rich single-stranded DNA can adopt in vitro an intramolecular quadruplex structure, which has been shown to directly inhibit telomerase activity. The reactivation of this enzyme in immortalized and most cancer cells suggests that telomerase is a relevant target in oncology, and telomerase inhibitors have been proposed as new potential anticancer agents. In this paper, we describe ethidium derivatives that stabilize G-quadruplexes. These molecules were shown to increase the melting temperature of an intramolecular quadruplex structure, as shown by fluorescence and absorbance measurements, and to facilitate the formation of intermolecular quadruplex structures. In addition, these molecules may be used to reveal the formation of multi-stranded DNA structures by standard fluorescence imaging, and therefore become fluorescent probes of quadruplex structures. This recognition was associated with telomerase inhibition in vitro: these derivatives showed a potent anti-telomerase activity, with IC50 values of 18–100 nM in a standard TRAP assay.

The reactivation of telomerase activity in most cancer cells supports the concept that telomerase is a relevant target in oncology, and telomerase inhibitors have been proposed as new potential anticancer agents. The telomeric G-rich single-stranded DNA can adopt in vitro an intramolecular quadruplex structure, which has been shown to inhibit telomerase activity. We used a fluorescence assay to identify molecules that stabilize G-quadruplexes. Intramolecular folding of an oligonucleotide with four repeats of the human telomeric sequence into a G-quadruplex structure led to fluorescence excitation energy transfer between a donor (fluorescein) and an acceptor (tetramethylrhodamine) covalently attached to the 5′ and 3′ ends of the oligonucleotide, respectively. The melting of the G-quadruplex was monitored in the presence of putative G-quadruplex-binding molecules by measuring the fluorescence emission of the donor. A series of compounds (pentacyclic crescent-shaped dibenzophenanthroline derivatives) was shown to increase the melting temperature of the G-quadruplex by 2–20°C at 1 μM dye concentration. This increase in Tm value was well correlated with an increase in the efficiency of telomerase inhibition in vitro. The best telomerase inhibitor showed an IC50 value of 28 nM in a standard telomerase repeat amplification protocol assay. Fluorescence energy transfer can thus be used to reveal the formation of four-stranded DNA structures, and its stabilization by quadruplex-binding agents, in an effort to discover new potent telomerase inhibitors.
