FUNDAMENTO: Um programa de coaching em pesquisa tem foco em desenvolvimento de habilidades e raciocínio científico. Para os profissionais da saúde, pode ser útil para o incremento no número e na qualidade de projetos e artigos. OBJETIVO: Avaliar os resultados iniciais e a metodologia de implantação do Research and Innovation Coaching Program do grupo Research on Research da Duke University na Sociedade Brasileira de Cardiologia. MÉTODOS: O programa trabalha em duas bases: o treinamento e o coaching. O treinamento é realizado online e aborda conteúdos sobre ideia de pesquisa, busca na literatura, escrita científica e estatística. Após o treinamento, o coaching favorece o estabelecimento de colaboração entre pesquisadores e centros mediante uma rede de contatos. O presente estudo descreve a implementação e os resultados iniciais dos anos 2011-2012. RESULTADOS: Em 2011, 24 centros receberam o treinamento, que consistiu em reuniões online, estudo e prática dos conteúdos abordados. Em janeiro de 2012, foi implementado um novo formato que teve como objetivo atingir mais pesquisadores. Em seis meses foram alocados 52 pesquisadores. No total, 20 artigos foram publicados e mais 49 artigos foram redigidos e aguardam submissão e/ou publicação. De forma adicional, foram elaboradas cinco propostas de financiamento de pesquisa. CONCLUSÃO: O número de artigos e as propostas de financiamento atingiram os objetivos inicialmente propostos. Contudo, os principais resultados desse tipo de iniciativa devem ser medidos em longo prazo, pois a consolidação da produção nacional de pesquisa de alta qualidade é um círculo virtuoso que se autoalimenta e expande com o tempo.

O estudo tem por objetivo caracterizar os Grupos de Pesquisa em Educação em Enfermagem do Brasil quanto à sua organização. Pesquisa documental, descritiva, quantitativa. As informações foram coletadas no Banco de Dados e Estatísticas do Portal Online do CNPq - censo 2006. O Brasil possui 47 Grupos de Pesquisa em Educação em Enfermagem, com 412 pesquisadores, dos quais 91% apresentam título de mestrado, doutorado ou pós-doutorado. Dos 307 estudantes, 92% são graduandos de Enfermagem, porém apenas 9% são bolsistas de iniciação científica. Entre os 112 técnicos, 75% são de Enfermagem, 46% possuem titulação de mestre ou doutor. Há um número expressivo de Grupos que contribuem significativamente para a produção de conhecimento no setor de educação, em nível latino-americano. Todavia, ainda são muitos os desafios a serem superados como a frágil interdisciplinaridade, a limitada integração ensino-serviço, o baixo fomento de bolsas de iniciação científica e as significativas desigualdades no acesso e desenvolvimento de pesquisas nas diferentes regiões do país.

Este artigo é parte de pesquisa que atenta para o peso crescente que a circulação internacional de pessoas, saberes e práticas tem alcançado nos processos de escolarização e formação profissional de determinados setores sociais. Considerando os recursos públicos que a Capes, o CNPq e a Fapesp destinam à formação de pesquisadores com vistas ao fomento à pesquisa e aos investimentos em ciência e tecnologia no horizonte da circulação internacional, recortamos para este artigo a descrição e a análise preliminares da base de dados de bolsistas no exterior dessas agências de fomento, no período de 1970 a 2000. O movimento de fluxos, a partir da metodologia quantitativa de correlação de variáveis, desenha as tendências dos intercâmbios acadêmicos internacionais promovidos pelas três agências e nas diversas áreas do conhecimento, sendo que se procura contextualizá-las nas políticas de desenvolvimento científico-tecnológico desenvolvidas pelo Estado brasileiro no período.

Neste artigo analisamos as fases evidenciadas no desenvolvimento do Programa de Mentoria do Portal dos Professores da Universidade Federal de São Carlos (SP). O programa se caracteriza por ser online e é dirigido para professores dos anos iniciais do ensino fundamental com até cinco anos de experiência. Foi conduzido por três pesquisadoras e onze mentoras (professoras experientes) que acompanharam individualmente, a distância e por meio de correspondências eletrônicas, 56 professoras iniciantes durante períodos entre seis meses a dois anos e meio. Do ponto de vista teórico consideramos a literatura relativa à aprendizagem e desenvolvimento profissional da docência; início da carreira docente; processos de mentoria e educação a distância. Metodologicamente desenvolvemos uma pesquisa-ação a partir de estratégias construtivo-colaborativas. As principais fontes de dados foram as correspondências trocadas entre mentoras e professoras iniciantes; diários reflexivos de mentoras e professoras iniciantes; redação de casos de ensino pelas professoras iniciantes; registros de observação das reuniões semanais ocorridos entre pesquisadoras e mentoras. Os resultados apontam que os processos de mentoria investigados ocorrem segundo fases distintas (fase inicial ou de aproximação; fase de desenvolvimento ou aprofundamento; fase de desligamento) com características, objetivos e procedimentos próprios.

Com a intensa produção e veiculação de informações científicas, tornou-se difícil para o profissional médico manter-se atualizado com os recursos habituais. A necessidade de conhecer e de participar de processos de formação continuada se impõe. Entre outras iniciativas, a Associação Médica Brasileira e o Conselho Federal de Medicina lançaram o Programa Nacional de Educação Continuada a Distância para médicos, buscando divulgar o conhecimento produzido nos grandes centros para profissionais de áreas mais remotas ou com reduzida disponibilidade de tempo. Tendo como pressu posto que a Sociedade do Conhecimento requer a formação inicial e continuada de profissionais e de cidadãos com um novo conjunto de competências para atuar com eficiência e responsabilidade, esses programas devem ser desenvolvidos com base em abordagens pedagógicas que efetivamente valorizem, além dos conteúdos de ensino, a disposição para a pesquisa, a autonomia na busca da informação, o espírito colaborativo e a postura ética, entre outras. No intuito de contribuir para essa reflexão, este texto tem por objetivos retomar o processo de formalização da educação médica continuada a distância no Brasil em termos didático-pedagógicos e analisar perspectivas dessas ações educativas.

The genus Eigenmannia comprises several species groups that display a surprising variety of diploid chromosome numbers and sex-determining systems. In this study, hypotheses regarding phylogenetic relationships and karyotype evolution were investigated using a combination of molecular and cytogenetic methods. Phylogenetic relationships were analyzed for 11 cytotypes based on sequences from five mitochondrial DNA regions. Parsimony-based character mapping of sex chromosomes confirms previous suggestions of multiple origins of sex chromosomes. Molecular cytogenetic analyses involved chromosome painting using probes derived from whole sex chromosomes from two taxa that were hybridized to metaphases of their respective sister cytotypes. These analyses showed that a multiple XY system evolved recently (<7 mya) by fusion. Furthermore, one of the chromosomes that fused to form the neo-Y chromosome is fused independently to another chromosome in the sister cytotype. This may constitute an efficient post-mating barrier and might imply a direct function of sex chromosomes in the speciation processes in Eigenmannia. The other chromosomal sex-determination system investigated is shown to have differentiated by an accumulation of heterochromatin on the X chromosome. This has occurred in the past 0.6 my, and is the most recent chromosomal sex-determining system described to date. In this study, the effects of nicotine on global gene expression of cultured cells from the brainstem of spontaneously hypertensive rat (SHR) and normotensive Wistar Kyoto (WKY) rats were evaluated using whole-genome oligoarrays. We found that nicotine may act differentially on the gene expression profiles of SHR and WKY. The influence of strain was present in 321 genes that were differentially expressed in SHR as compared with WKY brainstem cells independently of the nicotine treatment. A total of 146 genes had their expression altered in both strains after nicotine exposure. Interaction between nicotine treatment and the strain was observed to affect the expression of 229 genes that participate in cellular pathways related to neurotransmitter secretion, intracellular trafficking and cell communication, and are possibly involved in the phenotypic differentiation between SHR and WKY rats, including hypertension. The OTOF gene encoding otoferlin is associated with auditory neuropathy (AN), a type of non-syndromic deafness. We investigated the contribution of OTOF mutations to AN and to non-syndromic recessive deafness in Brazil. A test for the Q829X mutation was carried out on a sample of 342 unrelated individuals with non-syndromic hearing loss, but none presented this mutation. We selected 48 cases suggestive of autosomal recessive inheritance, plus four familial and seven isolated cases of AN, for genotyping of five microsatellite markers linked to the OTOF gene. The haplotype analysis showed compatibility with linkage in 11 families (including the four families with AN). Samples of the 11 probands from these families and from seven isolated cases of AN were selected for an exon-by-exon screening for mutations in the OTOF gene. Ten different pathogenic variants were detected, among which six are novel. Among the 52 pedigrees with autosomal recessive inheritance (including four familial cases of AN), mutations were identified in 4 (7.7%). Among the 11 probands with AN, seven had at least one pathogenic mutation in the OTOF gene. Mutations in the OTOF gene are frequent causes of AN in Brazil and our results confirm that they are spread worldwide. In the present work, we sought to mimic the internal state changes in response to a predator threat by pharmacologically stimulating the brain circuit involved in mediating predator fear responses, and explored whether this stimulation would be a valuable unconditioned stimulus (US) in an olfactory fear conditioning paradigm (OFC). The dorsal premammillary nucleus (PMd) is a key brain structure in the neural processing of anti-predatory defensive behavior and has also been shown to mediate the acquisition and expression of anti-predatory contextual conditioning fear responses. Rats were conditioned by pairing the US, which was an intra-PMd microinjection of isoproterenol (ISO; beta-adrenoceptor agonist), with amyl acetate odor-the conditioned stimulus (CS). ISO (10 and 40 nmol) induced the acquisition of the OFC and the second-order association by activation of beta-1 receptors in the PMd. Furthermore, similar to what had been found for contextual conditioning to a predator threat, atenolol (beta-1 receptor antagonist) in the PMd also impaired the acquisition and expression of OFC promoted by ISO. Considering the strong glutamatergic projections from the PMd to the dorsal periaqueductal gray (dPAG), we tested how the glutamatergic blockade of the dPAG would interfere with the OFC induced by ISO. Accordingly, microinjections of NMDA receptor antagonist (AP5, 6 nmol) into the dPAG were able to block both the acquisition, and partially, the expression of the OFC. In conclusion, we have found that PMd beta-1 adrenergic stimulation is a good model to mimic predatory threat-induced internal state changes, and works as a US able to mobilize the same systems involved in the acquisition and expression of predator-related contextual conditioning. A genomic region neighboring the alpha-synuclein gene, on rat chromosome 4, has been associated with anxiety- and alcohol-related behaviors in different rat strains. In this study, we have investigated potential molecular and physiological links between alpha-synuclein and the behavioral differences observed between Lewis (LEW) and Spontaneously Hypertensive (SHR) inbred rats, a genetic model of anxiety. As expected, LEW rats appeared more fearful than SHR rats in three anxiety models: open field, elevated plus maze and light/dark box. Moreover, LEW rats displayed a higher preference for alcohol and consumed higher quantities of alcohol than SHR rats. alpha-Synuclein mRNA and protein concentrations were higher in the hippocampus, but not the hypothalamus of LEW rats. This result inversely correlated with differences in dopamine turnover in the hippocampus of LEW and SHR rats, supporting the hypothesis that alpha-synuclein is important in the downregulation of dopamine neurotransmission. A novel single nucleotide polymorphism was identified in the 30-untranslated region (3`-UTR) of the alpha-synuclein cDNA between these two rat strains. Plasmid constructs based on the LEW 3`-UTR sequence displayed increased expression of a reporter gene in transiently transfected PC12 cells, in accordance with in-vivo findings, suggesting that this nucleotide exchange might participate in the differential expression of alpha-synuclein between LEW and SHR rats. These results are consistent with a novel role for alpha-synuclein in modulating rat anxiety- like behaviors, possibly through dopaminergic mechanisms. Since the behavioral and genetic differences between these two strains are the product of independent evolutionary histories, the possibility that polymorphisms in the alpha-synuclein gene may be associated with vulnerability to anxiety- related disorders in humans requires further investigation. Tumor necrosis factor-related apoptosis-inducing ligand-TNFSF10 (TRAIL), a member of the TNF-alpha family and a death receptor ligand, was shown to selectively kill tumor cells. Not surprisingly, TRAIL is downregulated in a variety of tumor cells, including BCR-ABL-positive leukemia. Although we know much about the molecular basis of TRAIL-mediated cell killing, the mechanism responsible for TRAIL inhibition in tumors remains elusive because (a) TRAIL can be regulated by retinoic acid (RA); (b) the tumor antigen preferentially expressed antigen of melanoma (PRAME) was shown to inhibit transcription of RA receptor target genes through the polycomb protein, enhancer of zeste homolog 2 (EZH2); and (c) we have found that TRAIL is inversely correlated with BCR-ABL in chronic myeloid leukemia (CML) patients. Thus, we decided to investigate the association of PRAME, EZH2 and TRAIL in BCR-ABL-positive leukemia. Here, we demonstrate that PRAME, but not EZH2, is upregulated in BCR-ABL cells and is associated with the progression of disease in CML patients. There is a positive correlation between PRAME and BCR-ABL and an inverse correlation between PRAME and TRAIL in these patients. Importantly, knocking down PRAME or EZH2 by RNA interference in a BCR-ABL-positive cell line restores TRAIL expression. Moreover, there is an enrichment of EZH2 binding on the promoter region of TRAIL in a CML cell line. This binding is lost after PRAME knockdown. Finally, knocking down PRAME or EZH2, and consequently induction of TRAIL expression, enhances Imatinib sensibility. Taken together, our data reveal a novel regulatory mechanism responsible for lowering TRAIL expression and provide the basis of alternative targets for combined therapeutic strategies for CML. Focal and segmental glomerulosclerosis (FSGS) is one of the most important causes of end-stage renal failure. The bradykinin B1 receptor has been associated with tissue inflammation and renal fibrosis. To test for a role of the bradykinin B1 receptor in podocyte injury, we pharmacologically modulated its activity at different time points in an adriamycin-induced mouse model of FSGS. Estimated albuminuria and urinary protein to creatinine ratios correlated with podocytopathy. Adriamycin injection led to loss of body weight, proteinuria, and upregulation of B1 receptor mRNA. Early treatment with a B1 antagonist reduced albuminuria and glomerulosclerosis, and inhibited the adriamycin-induced downregulation of podocin, nephrin, and alpha-actinin-4 expression. Moreover, delayed treatment with antagonist also induced podocyte protection. Conversely, a B1 agonist aggravated renal dysfunction and even further suppressed the levels of podocyte-related molecules. Thus, we propose that kinin has a crucial role in the pathogenesis of FSGS operating through bradykinin B1 receptor signaling. This paper presents an approach for assisting low-literacy readers in accessing Web online information. The oEducational FACILITAo tool is a Web content adaptation tool that provides innovative features and follows more intuitive interaction models regarding accessibility concerns. Especially, we propose an interaction model and a Web application that explore the natural language processing tasks of lexical elaboration and named entity labeling for improving Web accessibility. We report on the results obtained from a pilot study on usability analysis carried out with low-literacy users. The preliminary results show that oEducational FACILITAo improves the comprehension of text elements, although the assistance mechanisms might also confuse users when word sense ambiguity is introduced, by gathering, for a complex word, a list of synonyms with multiple meanings. This fact evokes a future solution in which the correct sense for a complex word in a sentence is identified, solving this pervasive characteristic of natural languages. The pilot study also identified that experienced computer users find the tool to be more useful than novice computer users do.

For many learning tasks the duration of the data collection can be greater than the time scale for changes of the underlying data distribution. The question we ask is how to include the information that data are aging. Ad hoc methods to achieve this include the use of validity windows that prevent the learning machine from making inferences based on old data. This introduces the problem of how to define the size of validity windows. In this brief, a new adaptive Bayesian inspired algorithm is presented for learning drifting concepts. It uses the analogy of validity windows in an adaptive Bayesian way to incorporate changes in the data distribution over time. We apply a theoretical approach based on information geometry to the classification problem and measure its performance in simulations. The uncertainty about the appropriate size of the memory windows is dealt with in a Bayesian manner by integrating over the distribution of the adaptive window size. Thus, the posterior distribution of the weights may develop algebraic tails. The learning algorithm results from tracking the mean and variance of the posterior distribution of the weights. It was found that the algebraic tails of this posterior distribution give the learning algorithm the ability to cope with an evolving environment by permitting the escape from local traps.
