In e-health intervention studies, there are concerns about the reliability of internet-based, self-reported (SR) data and about the potential for identity fraud. This study introduced and tested a novel procedure for assessing the validity of internet-based, SR identity and validated anthropometric and demographic data via measurements performed face-to-face in a validation study (VS). Participants (n = 140) from seven European countries, participating in the Food4Me intervention study which aimed to test the efficacy of personalised nutrition approaches delivered via the internet, were invited to take part in the VS. Participants visited a research centre in each country within 2 weeks of providing SR data via the internet. Participants received detailed instructions on how to perform each measurement. Individual’s identity was checked visually and by repeated collection and analysis of buccal cell DNA for 33 genetic variants. Validation of identity using genomic information showed perfect concordance between SR and VS. Similar results were found for demographic data (age and sex verification). We observed strong intra-class correlation coefficients between SR and VS for anthropometric data (height 0.990, weight 0.994 and BMI 0.983). However, internet-based SR weight was under-reported (Δ −0.70 kg [−3.6 to 2.1], p < 0.0001) and, therefore, BMI was lower for SR data (Δ −0.29 kg m−2 [−1.5 to 1.0], p < 0.0001). BMI classification was correct in 93 % of cases. We demonstrate the utility of genotype information for detection of possible identity fraud in e-health studies and confirm the reliability of internet-based, SR anthropometric and demographic data collected in the Food4Me study.

Three generations of settlers, convicts and emigrants occupied the valley of the Molonglo River for eighty years before Canberra was planted here in 1913. Stretched between the great houses of Duntroon and Yarralumla, they lived in some twenty cottages, almost all of which were demolished as the city centre and suburbs developed. This book recreates a lost world via archival sources that describe the land and houses, genealogical records showing the inhabitants and their family relationships around the district, and the work of official photographers and amateur artists who recorded the landscape as the city grew.--Publisher description.

Background: Qualitative research into the effect of school recess on children’s physical activity is currently limited. This study used a write and draw technique to explore children’s perceptions of physical activity opportunities during recess.

Methods: 299 children age 7–11 years from 3 primary schools were enlisted. Children were grouped into Years 3 & 4 and Years 5 & 6 and completed a write and draw task focusing on likes and dislikes. Pen profiles were used to analyze the data.

Results:
Results indicated ‘likes’ focused on play, positive social interaction, and games across both age groups but showed an increasing dominance of games with an appreciation for being outdoors with age. ‘Dislikes’ focused on dysfunctional interactions linked with bullying, membership, equipment, and conflict for playground space. Football was a dominant feature across both age groups and ‘likes/dislikes’ that caused conflict and dominated the physically active games undertaken.

Conclusion:
Recess was important for the development of conflict management and social skills and contributed to physical activity engagement. The findings contradict suggestions that time spent in recess should be reduced because of behavioral issues.

This research is a practice led investigation of large-scale site specific performance installation works that respond to local and historical trauma, place making, and belonging for communities and audiences. The research is part of an ongoing PhD which is primarily questioning how layers of history and lived experience manifest or ‘imprint’ upon natural landscapes and urban sites using the driving concepts of landscape, archaeology, and community immersion to inform the practice. The site of the primary research investigation was Anthology www.anthology.net.au- a major site-specific theatrical journey through Westlake, now known as Stirling Park – Ngunawal land, a traditional pathway and the site of one of the camps created to house the workers building the new city of Canberra. Tents and a hall were erected followed by 61 cottages built in 1923, for married tradesmen building the infrastructure for the new Federal Capital of Australia. These families lived at Westlake for 50 years until the 1960’s when the families were relocated, the houses sold and removed. A community demolished. Westlake is now parkland (and prime real estate), nestled between the lake and the Embassies of Yarralumla. The event took place between 26th November and the 6th December 2014. The performance installation was created and produced over a 3-year period with $45,000.00 in funding provided by ArtsACT and the Centenary of Canberra. Anthology alluded to the power of immersive, site-sympathetic performance as a regenerative force for communities right now. What lies in wait for artists in sites, in places…to be uncovered…with its final form revealed through careful excavation? Anthology centralised rituals of remembrance and the importance of place as vital to the restoration and regeneration of community through processing and transcending what has been lost, hidden, suppressed or in the case of Westlake or Stirling Park ‘vanished’.

Food consumption surveys are performed in many countries. Comparison of results from those surveys across nations is difficult because of differences in methodological approaches. While consensus about the preferred methodology associated with national food consumption surveys is increasing, no inventory of methodological aspects across continents is available. The aims of the present review are (1) to develop a framework of key methodological elements related to national food consumption surveys, (2) to create an inventory of these properties of surveys performed in the continents North- America, South-America, Asia and Australasia, and (3) to discuss and compare these methodological properties cross-continentally. A literature search was performed using a fixed set of search terms in different databases. The inventory was completed with all accessible information from all retrieved publications and corresponding authors were requested to provide additional information where missing. Surveys from ten individual countries, originating from four continents are listed in the inventory. The results are presented according to six major aspects of food consumption surveys. The most common dietary intake assessment method used in food consumption surveys worldwide is the 24-HDR (24 h dietary recall), occasionally administered repeatedly, mostly using interview software. Only three countries have incorporated their national food consumption surveys into continuous national health and nutrition examination surveys.

The local inflammatory environment of the cell promotes the growth of epithelial cancers. Therefore, controlling inflammation locally using a material in a sustained, non-steroidal fashion can effectively kill malignant cells without significant damage to surrounding healthy cells. A promising class of materials for such applications are the nanostructured scaffolds formed by epitope containing minimalist self-assembled peptides (SAPs), as they are bioactive on a cellular length scale, whilst presenting as an easily handled hydrogel. Here, we show that the assembly process distributes an anti-inflammatory polysaccharide, fuccoidan, localised to the nanofibers to function as an anti-inflammatory biomaterial for cancer therapy. We show that it supports healthy cells, whilst inducing apoptosis in cancerous endothelial cells, as demonstrated by the downregulation of the proinflammatory gene and protein expression pathways associated with epithelial cancer progression. Our findings highlight an innovative material approach with potential applications as local epithelial cancer immunotherapy and drug delivery vehicles.

Neosporose é uma enfermidade parasitária causada pelo protozoário Neospora caninum reconhecido como importante causa de abortamento bovino e neuropatia canina. Considerando o isolamento de N. caninum e a sorologia freqüente em bovinos em nossa região, os objetivos do presente trabalho foram avaliar a ocorrência de anticorpos anti-N. caninum em cães da Microrregião da Serra de Botucatu, Estado de SãoPaulo, e sua associação ao sexo, idade e procedência quanto à zona urbana (exclusivamente cidade), rural (somente chácaras e sítios) e peri-urbana (acesso à zona urbana e rural) dos cães estudados. Foram analisados 963 cães, com ou sem raça definida, de ambos os sexos e diferentes idades, sem apresentação de qualquer sintomatologia clínica. Os animais foram selecionados aleatoriamente durante a campanha de vacinação anti-rábica da microrregião da Serra de Botucatu, no período de maio a setembro de 1998. O soro obtido dos animais foi avaliado por meio da Reação de Imunofluorescência Indireta (RIFI) utilizando como antígeno a cepa padrão NC-1 deN. caninum. Observaram-se 245 animais reagentes (25,4% de positividade), sendo 161 (27,5%) machos e, 84 (22,3%) fêmeas. Dos animais de zona urbana, rural e mista 223 (25,8%),11(16,9%) e 11(33,3%), respectivamente, foram reagentes à prova de RIFI. Todos os 11 municípios apresentaram cães soropositivos com valores de ocorrência que variaram de 8,9% a 53,5%. Observou-se percentual de positividade menor em cães até um ano (16,2%) quando comparados àqueles entre 1 a 4 anos e superior a 4 anos (28,4 % e 28,0%, respectivamente) que não apresentaram diferença entre si. Os resultados obtidos caracterizaram soropositividade para N. caninum em cães pertencentes a todos os municípios da Microrregião da Serra de Botucatu evidenciando a ampla distribuição do agente na região.

OBJECTIVE: Altered microbiota composition, changes in immune responses and impaired intestinal barrier functions are observed in IBD. Most of these features are controlled by proteases and their inhibitors to maintain gut homeostasis. Unrestrained or excessive proteolysis can lead to pathological gastrointestinal conditions. The aim was to validate the identified protease IBD candidates from a previously performed systematic review through a genetic association study and functional follow-up. DESIGN: We performed a genetic association study in a large multicentre cohort of patients with Crohn's disease (CD) and UC from five European IBD referral centres in a total of 2320 CD patients, 2112 UC patients and 1796 healthy controls. Subsequently, we did an extensive functional assessment of the candidate genes to explore their causality in IBD pathogenesis. RESULTS: Ten single nucleotide polymorphisms (SNPs) in four genes were significantly associated with CD: CYLD, USP40, APEH and USP3. CYLD was the most significant gene with the intronically located rs12324931 the strongest associated SNP (pFDR=1.74e-17, OR=2.24 (1.83 to 2.74)). Five SNPs in four genes were significantly associated with UC: USP40, APEH, DAG1 and USP3. CYLD, as well as some of the other associated genes, is part of the ubiquitin proteasome system (UPS). We therefore determined if the IBD-associated adherent-invasive Escherichia coli (AIEC) can modulate the UPS functioning. Infection of intestinal epithelial cells with the AIEC LF82 reference strain modulated the UPS turnover by reducing poly-ubiquitin conjugate accumulation, increasing 26S proteasome activities and decreasing protein levels of the NF-κB regulator CYLD. This resulted in IκB-α degradation and NF-κB activation. This activity was very important for the pathogenicity of AIEC since decreased CYLD resulted in increased ability of AIEC LF82 to replicate intracellularly. CONCLUSIONS: Our results reveal the UPS, and CYLD specifically, as an important contributor to IBD pathogenesis, which is favoured by both genetic and microbial factors.

Gastrointestinal (GI) protein loss, due to lymphangiectasia or chronic inflammation, can be challenging to diagnose. This study evaluated the diagnostic accuracy of serum and fecal canine α1-proteinase inhibitor (cα1PI) concentrations to detect crypt abscesses and/or lacteal dilation in dogs. Serum and fecal cα1PI concentrations were measured in 120 dogs undergoing GI tissue biopsies, and were compared between dogs with and without crypt abscesses/lacteal dilation. Sensitivity and specificity were calculated for dichotomous outcomes. Serial serum cα1PI concentrations were also evaluated in 12 healthy corticosteroid-treated dogs. Serum cα1PI and albumin concentrations were significantly lower in dogs with crypt abscesses and/or lacteal dilation than in those without (both P <0.001), and more severe lesions were associated with lower serum cα1PI concentrations, higher 3 days-mean fecal cα1PI concentrations, and lower serum/fecal cα1PI ratios. Serum and fecal cα1PI, and their ratios, distinguished dogs with moderate or severe GI crypt abscesses/lacteal dilation from dogs with only mild or none such lesions with moderate sensitivity (56-92%) and specificity (67-81%). Serum cα1PI concentrations increased during corticosteroid administration. We conclude that serum and fecal α1PI concentrations reflect the severity of intestinal crypt abscesses/lacteal dilation in dogs. Due to its specificity for the GI tract, measurement of fecal cα1PI appears to be superior to serum cα1PI for diagnosing GI protein loss in dogs. In addition, the serum/fecal cα1PI ratio has an improved accuracy in hypoalbuminemic dogs, but serum cα1PI concentrations should be carefully interpreted in corticosteroid-treated dogs.
