I ntroduction: The assessment of respiratory muscle strength is important in the diagnosis and monitoring of the respiratory muscles weakness of respiratory and neuromuscular diseases. However, there are still no studies that provide predictive equations and reference values for maximal respiratory pressures for children in our population. Aim: The purpose of this study was to propose predictive equations for maximal respiratory pressures in healthy school children. Method: This is an observational cross-sectional study. 144 healthy children were assessed. They were students from public and private schools in the city of Natal /RN (63 boys and 81 girls), subdivided in age groups of 7-8 and 9-11 years. The students presented the BMI, for age and sex, between 5 and 85 percentile. Maximal respiratory pressures were measured with the digital manometer MVD300 (Globalmed ®). The maximal inspiratory pressure (MIP) and maximal expiratory pressures (MEP) were measured from residual volume and total lung capacity, respectively. The data were analyzed using the SPSS Statistics 15.0 software (Statistical Package for Social Science) by assigning the significance level of 5%. Descriptive analysis was expressed as mean and standard deviation. T'Student test was used for unpaired comparison of averages of the variables. The comparison of measurements obtained with the predicted values in previous studies was performed using the paired t'Student test. The Pearson correlation test was used to verify the correlation of MRP's with the independent variables (age, sex, weight and height). For the equations analysis the stepwise linear regression was used. Results: By analyzing the data, we observed that in the age range studied MIP was significantly higher in boys. The MEP did not differ between boys and girls aged 7 to 8 years, the reverse occurred in the age between 9 and 11 years. The boys had a significant increase in respiratory muscle strength with advancing age. Regardless sex and age, MEP was always higher than the MIP. The reference values found in this study are similar to a sample of Spanish and Canadian children. The two models proposed in previous studies with children from other countries were not able to consistently predict the values observed in this studied population. The variables sex, age and weight correlated with MIP, whereas the MEP was also correlated with height. However, in the regression models proposed in this study, only gender and age were kept exerting influence on the variability of maximal inspiratory and expiratory pressures. Conclusion: This study provides reference values, lower limits of normality and proposes two models that allow predicting, through the independent variables, sex and age, the value of maximal static respiratory pressures in healthy children aged between 7 and 11 years old

Introduction: The reference values and prediction equations for maximal respiratory pressures (MRP) differ significantly between the available studies. This large discrepancy can be attributed to the different methodologies proposed. Although the importance of MRP is widely recognized, there are no Brazilian studies that provide predictive equations and reference values for PRM adolescents. Objectives: The purpose of this study was to provide normal values and propose predictive equations for maximal static respiratory pressures of Brazilian adolescents. Methods: An observational cross-sectional study, which evaluated 182 adolescents of both sexes aged between 12 and 18 years, enrolled in schools of the state and private in the city of Natal / RN. The selection of schools and participants of the study was randomly through a lottery system. The spirometric evaluation was performed through the digital spirometer One Flow FVC prior to the assessment of respiratory muscle strength. The MICs were measured with MVD digital manometer 300. Statistical analysis was performed using the SPSS 17.0 software STATISTICS, assigning the significance level of 5%. The normality of data distribution was verified using the Kolmogorov-Smirnov (KS). The descriptive analysis was expressed as mean and standard deviation. We used one-way ANOVA test to verify the difference of the averages of MRPs between age and gender and comparing the averages of MRPs between levels of physical activity. The test t'Student unpaired compared the averages of MRPs being ages and sexes. The comparison of mean values obtained in this study PRM with the values predicted using the equations mentioned above was relizada by testing paired t'Student. To verify the correlation between the PRM and the independent variables (age, weight, height) was used Pearson correlation test. Levene's test evaluated the homogeneity of variance. To obtain predictive equations analysis was used stepwise multiple linear regression. Results: There was no significant difference in mean age between the PRM. The male adolescents, regardless of age, showed superiority in MRP values when compared to the opposite sex. Weight, height and sex correlated with the PRM. Regression analysis suggested in this study, pointed out that the weight and sex had an influence in MIP and MEP only in relation to sex influenced. The mean for each PRM adolescents classified as very active were superior to those observed in adolescents classified as irregularly active. Conclusion: This study provides reference values and two models of predictive equations for maximal inspiratory and expiratory pressures, and to establish the lower limits of normality that will serve as an indispensable condition for careful evaluation of respiratory muscle strength in Brazilian adolescents

Introduction: The Temporomandibular disorder (TMD) is greatly prevalent in the population and can be associated with bruxism. This disorder produces several signs and symptoms. Among them, pain is one of the most important because it reduces life quality and productivity of people who have such disorder. The aim of this research was to study if massage causes pain relief and/or electromyographic (EMG) changes. Materials and methods: The subjects were chosen by a questionnaire and divided into 2 different groups. Their ages varied from 19 to 22 years. The experimental group consisted of 6 TMD patients, who were submitted to the massage treatment and 4 EMG-sessions (the 1 st EMG-session occurred before the treatment and the others in the 1 st, 15 th and 30 th days after the treatment). The control group consisted of 6 TMD patients, who were submitted to the same 4 EMG-sessions. While EMG activity was recorded, subjects were asked to keep mandibular rest position (MRP) and to perform maximal voluntary clenching (MVC). The treatment consisted of 15 massage-sessions on face and neck and in application of Visual Analogue Scale (VAS) for measuring pain level. The massage sessions had 30 minutes of duration and were performed daily. The EMG data were processed to obtain the Root Mean Square (RMS), which were normalized by MVC. Results: It was demonstrated that (1) RMS-MRP of the right masseter in experimental group at the 1 st EMG-session was higher than at the 2 nd EMG-session and (2) statistically significant reduction was found for VAS values after massage session. Conclusion: Unfortunately the sample is insufficient to draw any conclusions, therefore, more studies regarding the use of massage in the management of myogenic TMD are necessary.

Objective. The myeloid-related proteins 8 and 14 (MRP-8/MRP-14) and neutrophil-derived S100A12 are biomarkers of inflammation. They can be used to determine the relapse risk in patients with juvenile idiopathic arthritis (JIA) after stopping antiinflammatory treatment. In this study, we tested the performance of different enzyme-linked immunosorbent assays (ELISAs) in order to validate systems available for routine use.Methods. MRP-8/MRP-14 and S100A12 serum concentrations of 188 JIA patients in remission were analyzed. Commercially available test systems were compared to experimental ELISAs established in house. The ability of the assays to identify JIA patients at risk for relapse was analyzed.Results. For MRP-8/MRP-14, the PhiCal Calprotectin and Buhlmann MRP8/14 Calprotectin ELISAs revealed hazard ratios of 2.3 and 2.1, respectively. For S100A12, the CircuLex S100A12/EN-RAGE ELISA revealed a hazard ratio of 3.1. The commercial assays allowed a JIA relapse prediction that was at least comparable to the experimental ELISAs.Conclusion. For the prediction of JIA relapse after stopping medication, the biomarkers MRP-8/MRP-14 and S100A12 can be determined by using assays that are available for routine use. The tested commercial MRP-8/MRP-14 and S100A12 ELISAs showed a performance comparable to well-established experimental ELISA protocols when assay-specific cutoffs for the indication of relapse prediction are thoroughly applied.

We compared the effects of medium light roast (MLR) and medium roast (MR) paper-filtered coffee on antioxidant capacity and lipid peroxidation in healthy volunteers. In a randomized crossover study, 20 volunteers consumed 482 +/- 61 ml/day of MLR or MR for four weeks. Plasma total antioxidant status (TAS), oxygen radical absorbance capacity (ORAC), oxidized LDL and 8-epi-prostaglandin F2 alpha, erythrocyte superoxide dismutase (SOD), glutathione peroxidase (GPx), and catalase (CAT) activity were measured at baseline and after the interventions. MLR had higher chlorogenic acids-(CGA; 334 mg/150 mL) and less caffeine (231 mg/150 ml) than MR had (210 and 244 mg/150 ml, respectively). MLR also had fewer Maillard reaction products (MRP) than MR had. Compared with baseline, subjects had an increase of 21 and 26 % in TAS, 13 and 13 % in CAT, 52 and 75 % in SOD, and 62 and 49 % in GPx after MLR and MR consumption (P < 0.001), respectively. ORAC increased after MLR (P = 0.004). No significant alteration in lipid peroxidation biomarkers was observed. Both coffees had antioxidant effects. Although MLR contained more CGA, there were similar antioxidant effects between the treatments. MRP may have contributed as an antioxidant. These effects may be important in protecting biological systems and reducing the risk of diseases related to oxidative stress.

Background: Inspiratory muscle training (IMT) has been shown to increase diaphragm thickness. We evaluated the effect of IMT on mid-respiratory pressure (MRP) in patients with gastroesophageal reflux disease (GERD) and hypotensive lower esophageal sphincter (LES), and compared the results with a sham group. Methods: Twenty consecutive patients (progressive loading group) and 9 controls (sham group) were included. All patients had end expiratory pressure (EEP) between 5 and 10 mmHg, underwent esophageal manometry and pulmonary function tests before and after 8 weeks of training, and used a threshold IMT twice daily. The threshold IMT was set at 30% of the maximal inspiratory pressure for the progressive loading group; while, the threshold for sham-treated patients was set at 7 cmH(2)O for the whole period. Results: There was an increase in MRP in 15 (75%) patients in the progressive loading group, with an average gain of 46.6% (p<0.01), and in six (66%) patients in the sham group with a mean increase of 26.2% (p<0.01). However, there was no significant difference between the groups (p = 0.507). The EEP also increased compared with measurements before training (p<0.01), but it did not differ between groups (p = 0.727). Conclusion: IMT increased LES pressure in patients with GERD, in both the treatment and sham groups, after an eight-week program. Although there was no statistically significant difference between groups, suggesting the pressure increase in LES occurs regardless of the resistance load of the threshold IMT. Riba Composites, azienda specializzata nella lavorazione della fibra di materiali compositi avanzati, si trova in una vantaggiosa situazione di sviluppo e ampliamento del proprio raggio d’azione, e dove le informazioni da gestire sono sempre più numerose. E’ quindi risultato necessario un supporto informativo che gestisca le informazioni. Dal punto di vista produttivo, l’introduzione del sistema informativo ha l’obiettivo di rispondere alle problematiche legate alla gestione dei materiali, sia a livello di materie prime, che di semilavorati e prodotti finiti in modo tale da gestirli con efficienza ed evitando le rotture di stock. L’obiettivo di fondo che Riba vuole perseguire é di crescere e svilupparsi in logica di lean production che, nell’ottica della gestione dei magazzini significa “approvvigionare i materiali solamente nel momento in cui si manifesta un fabbisogno”. Quest’approccio abbandona la attuale logica di pianificazione “a spinta” (push) che prevedeva la programmazione degli approvvigionamenti e la produzione di semilavorati e prodotti finiti attraverso previsioni basate sull’analisi di dati storici o di mercato, e non attraverso gli ordini effettivamente acquisiti su cui si basa la logica di produzione “snella” (pull). L’implementazione di un sistema ERP ha richiesto un’analisi approfondita dell’azienda in cui si opera così come del prodotto finito e del processo produttivo, a tal punto da poter riconoscere le esigenze e le necessità a cui dovrà rispondere il sistema informativo. A questa fase di analisi e raccolta dati segue un momento di assestamento del sistema informativo, in cui solo una parte di articoli viene gestita dal sistema per poter procedere contemporaneamente con la graduale formazione del personale. La durata del progetto in questione è stata stimata di circa 20 mesi, tempo necessario per poter adattare il sistema e le diverse personalizzazioni ad un processo così complesso come la lavorazione della fibra di carbonio. In cycling cells positive stimuli like nutrient, growth factors and mitogens increase ribosome biogenesis rate and protein synthesis to ensure both growth and proliferation. In contrast, under stress situation, proliferating cells negatively modulate ribosome production to reduce protein synthesis and block cell cycle progression. The main strategy used by cycling cell to coordinate cell proliferation and ribosome biogenesis is to share regulatory elements, which participate directly in ribosome production and in cell cycle regulation. In fact, there is evidence that stimulation or inhibition of cell proliferation exerts direct effect on activity of the RNA polymerases controlling the ribosome biogenesis, while several alterations in normal ribosome biogenesis cause changes of the expression and the activity of the tumor suppressor p53, the main effector of cell cycle progression inhibition. The available data on the cross-talk between ribosome biogenesis and cell proliferation have been until now obtained in experimental model in which changes in ribosome biogenesis were obtained either by reducing the activity of the RNA polymerase I or by down-regulating the expression of the ribosomal proteins. The molecular pathways involved in the relationship between the effect of the inhibition of RNA polymerase III (Pol III) activity and cell cycle progression have been not yet investigated. In eukaryotes, RNA Polymerase III is responsible for transcription of factors involved both in ribosome assembly (5S rRNA) and rRNA processing (RNAse P and MRP).Thus, the aim of this study is characterize the effects of the down-regulation of RNA Polymerase III activity, or the specific depletion of 5S rRNA. The results that will be obtained might lead to a deeper understanding of the molecular pathway that controls the coordination between ribosome biogenesis and cell cycle, and might give useful information about the possibility to target RNA Polymerase III for cancer treatment.

L’elevato consumo e il limitato tasso di rimozione dei farmaci favoriscono l’entrata di questi composti, biologicamente attivi, nell’ambiente acquatico. Essendo continuamente aggiunti, più velocemente di quanto si degradino, tali sostanze diventano pseudo-persistenti nell’ambiente, e gli organismi acquatici possono risultarne esposti a lungo termine. Per limitare gli effetti avversi di ambienti inquinanti, gli organismi più resistenti hanno sviluppato vari meccanismi di protezione cellulare, fra cui il sistema denominato Multixenobiotic resistance (MXR). Tale meccanismo agisce da sistema di detossificazione attraverso l’azione di specifici trasportatori transmembrana quali la P-glicoproteina (Pgp) e la Multidrug Resistance-associated Protein (MRP). Questi, codificati rispettivamente dai prodotti genici ABCB e ABCC, favorendo l’efflusso attivo di composti tossici, prevengono il loro accumulo cellulare e, dunque, i loro potenziali effetti dannosi. Tuttavia, alcune sostanze possono influenzare questo delicato meccanismo, esponendo gli organismi acquatici agli effetti tossici degli inquinanti. In questo lavoro di Tesi è stato condotto uno studio sperimentale in vitro volto a determinare i possibili effetti dell’antidepressivo fluoxetina (FX), dell’antiepilettico carbamazepina (CBZ) e del β-bloccante propranololo (PROP) sul meccanismo MXR, in emociti di Mytilus galloprovincialis. In particolare si è valutata l’azione di questi farmaci sulla funzionalità dei trasportatori e/o sulla modulazione trascrizionale. I risultati ottenuti hanno dimostrato l’importante effetto del PROP, della CBZ e della FX sul sistema MXR. In particolare, il PROP e la CBZ alterano tale sistema, agendo, principalmente, sui livelli di espressione del prodotto genico ABCB, nonostante influenzino anche i livelli di ABCC. La FX, invece, non porta ad alterazioni dell’attività MXR totale, sebbene induca variazioni sui livelli di espressione di ABCB. In conclusione, le analisi dei profili trascrizionali dimostrano che la proteina predominante nell’efflusso di tali contaminanti negli emociti di M. galloprovincialis è la P-glicoproteina e che le variazioni dell’attività MXR osservate in questa Tesi sono il risultato combinato dell’azione dei farmaci analizzati come modulatori trascrizionali e come substrati da parte dei trasportatori MXR.

Il presente lavoro di tesi si occupa dell’analisi delle caratteristiche costituenti un sistema informativo destinato alla gestione della logistica e della produzione di piccole realtà imprenditoriali e, successivamente, della definizione di una possibile struttura modulare dello stesso sistema. Il lavoro, frutto di una collaborazione (tirocinio formativo) presso una società di consulenza organizzativa e informatica, prende le mosse dallo studio di una serie di trattazioni riguardanti i seguenti temi: gestione delle scorte e della produzione, lean production, cambiamento organizzativo e reingegnerizzazione dei processi aziendali, sistemi informativi e informatici, rapporto tra piccole-medie imprese e tecnologie dell’informazione. Da un iniziale introduzione sulle tematiche legate alla gestione dell’informazione in ambito aziendale, si procede ad una descrizione dell’utilizzo delle informazioni, in particolare in riferimento alle tecniche di reingegnerizzazione dei processi aziendali. In seguito, viene analizzato il più ampio concetto delle tecnologie a supporto dell’informazione, e della loro relazione con le piccole e medie imprese italiane. Successivamente, si offre una panoramica dei metodi più utilizzati per la pianificazione e programmazione della produzione e per la gestione delle scorte, differenziandoli tra metodi a fabbisogno e metodi a ripristino. Infine, si procede alla presentazione di una configurazione originale di un sistema informativo gestionale, tramite descrizione approfondita dei moduli di base costituenti, anche attraverso l’ausilio di diagrammi esplicativi, ed il confronto tra il proprio metodo di programmazione materiali ed il più famoso metodo MRP (Material Requirements Planning), diffuso nella maggior parte dei software gestionali in commercio; quest’ultimi verranno confrontati con la soluzione presentata tramite mappa di posizionamento. In conclusione, vengono esposte le ragioni di possibile successo del sistema presentato, mettendo in evidenza l’ormai appurata imprescindibilità dei sistemi informativi gestionali, sottolineata dalla crescita costante della loro adozione da parte delle imprese italiane. In particolare, viene posto l’accento sul fatto che il bacino di mercato costituito dalle piccole imprese sia ancora in parte insoddisfatto. Sono proprio le piccole imprese, come verrà spiegato nel dettaglio, le beneficiarie del nuovo sistema progettato, grazie a determinate caratteristiche studiate ad hoc per questa cospicua fascia di mercato.

La presenza dei farmaci in ambiente sta assumendo sempre più rilevanza come problematica da affrontare e a cui tentare di porre rimedio. Anche se le concentrazioni nei corpi idrici risultano piuttosto basse (μg/L o ng/L), i farmaci hanno proprietà diverse dagli inquinanti convenzionali e possono ugualmente indurre effetti dannosi nella fauna acquatica. In questo lavoro sono stati valutati gli effetti del propranololo (PROP, farmaco β-bloccante) e della carbamazepina (CBZ, farmaco anticonvulsivante), su di uno stadio larvale del mitilo Mytilus galloprovincialis (larva veliger – 48 ore post fecondazione). L’esposizione a concentrazioni crescenti dei due farmaci induce un progressivo aumento della anormalità delle larve, che mostrano malformazioni o ritardi nello sviluppo. Sia PROP che CBZ aumentano l’attività dei trasportatori implicati nella Multixenobiotic Resistance (MXR), indicando che il sistema di estrusione degli xenobiotici viene attivato già in questa fase dello sviluppo larvale. Analogamente, PROP aumenta l’espressione dei geni che codificano per le proteine della famiglia MXR, P-gp ed MRP. Aumenta anche l’espressione dei geni che codificano per catalasi e GST, suggerendo un aumento delle risposte antiossidanti/detossificanti. PROP aumenta i trascritti dell’anidrasi carbonica, mentre al contrario riduce i trascritti per la proteina extrapalliale: non è chiaro quindi l’effetto del farmaco sulla regolazione della espressione di proteine coinvolte nella formazione della conchiglia. CBZ aumenta l’espressione dei trascritti per P-gp ma diminuisce quella relativa ad MRP. Inoltre, riduce l’espressione dei geni codificanti per catalasi e GST, e della proteina extrapalliale, senza avere effetto sui trascritti dell’anidrasi carbonica. In generale possiamo dire che i due farmaci inducono evidenti alterazioni a livello biochimico e molecolare che possono influenzare sia la detossificazione che la biomineralizzazione, con conseguenze rilevanti per lo sviluppo larvale.

La presente ricerca si focalizza su alcuni processi chiave dello sviluppo larvale del mitili M. galloprovincialis: la biomineralizzazione, in quanto elemento fondamentale per la formazione della conchiglia; lo sviluppo ed il mantenimento della struttura cellulare, in quanto il progredire dello sviluppo comporta molteplici cambiamenti a tutti i livelli; la detossificazione, in quanto capace di fornire l’adeguata protezione. Lo studio ha visto l’impiego di tecniche molecolari (RT-qPCR) al fine di valutare l’espressione di geni codificanti per l’enzima anidrasi carbonica, la proteina extrapalliale, il recettore 5-HT1, la proteina actina, i trasportatori di membrana P-gp e Mrp, e gli enzimi catalasi e glutatione S-transferasi. È stata poi esaminata l’attività dell’enzima protein-chinasi A e si è caratterizzato il sistema MXR. Inoltre si è cercato di valutare il coinvolgimento di meccanismi cAMP/PKA-dipendenti nella regolazione dell’espressione di geni sopra menzionati. A tale scopo, sono stati utilizzati i composti farmaceutici propranololo e carbamazepina, che, da studi precedenti, sono noti interferire nella regolazione della via di trasduzione cAMP/PKA dipendente nel mitilo. I risultati di questa ricerca permettono di affermare che lo sviluppo larvale è un processo continuo in cui vi sono costanti cambiamenti fisiologici. L’espressione basale dei trascritti studiati ha mostrato evidenti variazioni nel tempo tra uova fertilizzate, trocofore e veliger, con un trend generale di sovraregolazione. E’ emerso anche che durante il suo sviluppo, il mitilo mediterraneo non presenta lo stesso tipo di regolazione della trascrizione riscontrabile nell’esemplare adulto. La modulazione dell’espressione da parte dei farmaci utilizzati ha comportato sovra- o sotto-espressione dei geni esaminati, indicando il possibile coinvolgimento di vie di regolazione della trascrizione affidate alla trasduzione del segnale neuroendocrino e del pathway cAMP/PKA dipendente

Trypanosoma brucei and related pathogens transcribe most genes as polycistronic arrays that are subsequently processed into monocistronic mRNAs. Expression is frequently regulated post-transcriptionally by cis-acting elements in the untranslated regions (UTRs). GPEET and EP procyclins are the major surface proteins of procyclic (insect midgut) forms of T. brucei. Three regulatory elements common to the 3' UTRs of both mRNAs regulate mRNA turnover and translation. The glycerol-responsive element (GRE) is unique to the GPEET 3' UTR and regulates its expression independently from EP. A synthetic RNA encompassing the GRE showed robust sequence-specific interactions with cytoplasmic proteins in electromobility shift assays. This, combined with column chromatography, led to the identification of 3 Alba-domain proteins. RNAi against Alba3 caused a growth phenotype and reduced the levels of Alba1 and Alba2 proteins, indicative of interactions between family members. Tandem-affinity purification and co-immunoprecipitation verified these interactions and also identified Alba4 in sub-stoichiometric amounts. Alba proteins are cytoplasmic and are recruited to starvation granules together with poly(A) RNA. Concomitant depletion of all four Alba proteins by RNAi specifically reduced translation of a reporter transcript flanked by the GPEET 3' UTR. Pulldown of tagged Alba proteins confirmed interactions with poly(A) binding proteins, ribosomal protein P0 and, in the case of Alba3, the cap-binding protein eIF4E4. In addition, Alba2 and Alba3 partially cosediment with polyribosomes in sucrose gradients. Alba-domain proteins seem to have exhibited great functional plasticity in the course of evolution. First identified as DNA-binding proteins in Archaea, then in association with nuclear RNase MRP/P in yeast and mammalian cells, they were recently described as components of a translationally silent complex containing stage-regulated mRNAs in Plasmodium. Our results are also consistent with stage-specific regulation of translation in trypanosomes, but most likely in the context of initiation.
