Derris urucu, pertencente à família Leguminosae/Fabaceae, é conhecida popularmente como “timbó”. As raízes desta espécie são utilizadas comumente como pesticidas e veneno para peixe. Do gênero Derris foram relatados muitos estudos fitoquímicos, sendo as raízes a parte mais estudada das plantas desse gênero. A denominação “timbó” é mais generalizada para as espécies Derris urucu e Derris nicou que são as espécies que produzem, nas raízes, substâncias da classe dos rotenóides como a rotenona e a deguelina, de onde deriva a importância dessas plantas. Os extratos, bem como, as substâncias isoladas deste gênero são responsáveis por um vasto conjunto de atividades biológicas, principalmente a atividade inseticida. Do extrato etanólico das folhas de D. urucu, doze substâncias foram isoladas e purificadas por Cromatografia Líquida de Alta Eficiência: cinco estilbenos, seis diidroflavonóis e uma flavanona. A identificação estrutural foi feita com base na análise espectrométrica de massas, de RMN 1H e 13C e técnicas de RMN bidimensionais, além de comparação com dados encontrados na literatura. O extrato etanólico das folhas de D. urucu foi submetido a bioensaios para avaliação do seu potencial alelopático, tendo exibido relevantes percentuais de inibição da germinação de sementes e do desenvolvimento de plantas invasoras de pastagens. Com o objetivo de detectar as substâncias responsáveis pela atividade alelopática, três estilbenos e três diidroflavonóis foram selecionados e submetidos a bioensaios de inibição de germinação e de desenvolvimento da radícula e do hipocótilo de plantas daninhas. Os ensaios alelopáticos foram realizados com as substâncias isoladas e com a combinação delas, visando também avaliar o sinergismo entre elas, no entanto, as inibições observadas foram de magnitudes muito baixas. Por outro lado, quando as substâncias foram testadas em misturas observou-se um aumento significativo dos percentuais de inibição, por isso essas substâncias em misturas, podem ser consideradas promissoras para futuros estudos, envolvendo atividade alelopática. Foram realizados, também, bioensaios de atividade antioxidante com oito substâncias, sendo três estilbenos e cinco diidroflavonóis frente ao radical DPPH. Neste bioensaio, não foi observada uma atividade antioxidante relevante, justificada pela presença de poucas hidroxilas fenólicas nas estruturas das substâncias testadas.

Este trabalho teve como objetivo identificar e quantificar os constituintes dos óleos essenciais de Illicum verum, Ageratum conyzoides, Piper hispidinervum e Ocotea odorifera, bem como avaliar a toxicidade para o pulgão-verde Schizaphis graminum. A qualificação dos constituintes foi realizada por meio de um cromatógrafo gasoso + espectrômetro de massas, e a quantificação, por um cromatógrafo gasoso + detector de ionização de chama, ambos com uma coluna DB5. O método de hidrodestilação promoveu um rendimento (p/p) de 3,81% para I. verum, 0,46% para A. conyzoides, 2,85% para P. hispidinervum e 0,68% para O. odorífera. Já os componentes majoritários foram: precoceno (87,0%) e (E)-cariofileno (7,1%) para A. conyzoides; (E)-anetol (90,4%), limoneno (2,6%) e metil-chavicol (1,3%) para I. verum; metil-eugenol (81,2%) e safrol (10,6%) para Ocotea odorífera; e safrol (82,5%) e α-terpinoleno (13,4%) para P. hispidinervum. Pelos testes de toxicidade aguda (24 horas) com folhas de sorgo ou papel-filtro contaminados, verificou-se que o óleo de A. conyzoides foi o mais tóxico para o pulgão, com CL50 de 7,13 e 7,08 µL óleo/cm2 respectivamente, seguido por O. odorifera com CL50 de 11,80 e 103,00 µL óleo/cm2 respectivamente; I. verum de 51,80 µL óleo/cm2 em ambos os substratos; e o menos tóxico foi o óleo essencial de P. hispidinervum, com CL50 de 62,50 e 143,00 µL óleo/cm2, respectivamente. Dessa maneira, sugere-se que o uso dos óleos essenciais pode representar uma nova ferramenta em programas de manejo integrado de pragas.

Bemisia tabaci (Hemiptera: Aleyrodidae) biotype B is one of the most limiting pests of tomato crops in the world. Tomato yield is currently dependent on the use of pesticides, which are problematic to farmers, consumers and the environment. A promising alternative to reduce the harmful effects caused by the indiscriminated use of synthetic insecticides is the use of insecticides of botanical origin. This study aimed to evaluate the effect of 3% (w/v) aqueous extracts from different structures of thirteen botanical species on the behavior of B. tabaci biotype B adults, as well as insecticidal activity of such aqueous extracts on B. tabaci eggs, nymphs, and adults infesting tomato plants. A distilled water solution was used as a negative control, and thiamethoxam insecticide (18 g/100 L of water) as a positive control. Leaf extract of Toona ciliata was observed to have the most efficient inhibitory effect in tests of extracts on whitefly behavior. Furthermore, the use of leaf extract of Toona ciliata led to the most drastic reduction in the number of adults and eggs on tomato leaflets. Leaf extract of Piper aduncum led to the greatest observed ovicidal effect (78.00% of non-hatched nymphs); however it was not effective against nymphs and adults. The leaf extracts of Trichilia pallida, Trichilia casaretti, and Toona ciliata showed the highest control indexes (67.9, 60.3, and 55.1%, respectively). For adults mortality, T. pallida was the most effective (72.8%). Our results indicate that application of extracts of T. pallida, T. ciliata, and T. casaretti are promising strategies to manage B. tabaci biotype B on tomato.

Photodynamic therapy, term introduced by von Tapeiner in 1900, can be defined as the administration of a non toxic drug, i.e., a dye, known as photosensitizer (FS), which subsequently will be illuminated with light of specific wavelength. PDT is based on the interaction among FS, oxygen and light, which through photochemical reactions cause cell death. The FS molecules must have a high probability to form the singlet state after the excitation, which can induce chemical changes in the neighborhood in two ways, called reactions type I and type II. The type II reaction is based on the exchange of energy to molecular oxygen, exciting it to its state of higher energy (singlet), which is highly reactive. The proposed mechanisms for cell death are linked to damage to the DNA, mitochondria and to the cytoplasmic membrane. Several pre‐clinical and clinical trials have been carried out and the PDT is already used in many countries for treatment mainly against certain types of cancer. The therapy also has been gaining strength in antimicrobial control, since the microorganisms have appeared increasingly resistant to current antibiotics. Another attempt to use the PDT is for the inactivation of macro‐organisms, such as micro‐crustaceans and mosquitoes. To this end I tested whether the photosensitizers methylene blue, rose Bengal and the chlorophyll a has insecticidal activity against the yellow fever and dengue vector mosquito, Aedes aegypti. Since these diseases have no effective treatments, its control is linked to the vector control, which has shown resistance to chemical pesticides used. Based on this, this work shows its importance, because it is a new type of mosquito control since all the photosensitizers used are low cost, do not generate toxic products at the concentrations used and showed good results in mortality. Bacillus thuringiensis (Bt), is an environmental Gram-positive spore-forming bacterium that produces crystalline parasporal protein (Cry) during sporulation. The inclusions often exhibit strong and specific insecticidal activity, making Bt an agent for agricultural controlling insects pest, mites, protozoa and nematodes. Recent studies reported that some of these Crys do not show cytotoxicity against insects but they are capable to kill some human and animal cancer cells. These proteins were denominated parasporins (PS). However, antitumor activity of Bt parasporin on the development of murine colorectal cancer (CT-26), are not well studies and these are no reports on the in vivo effect of these proteins. Thus, the present study evaluated the in vitro and in vivo anti-tumoral activity of Bt parasporin against the murine colorectal cancer line CT-26. Therefore, Balb/c mice were s.c. inoculated with CT-26 cells and weekly treated with parasporin (i.p.) pre-activated by enzymatic digestion with trypsin or proteinase K. Our results have shown, for the first time, that despite the anti-tumor activity in vitro, parasporin crystals couldn’t combat tumor growth in vivo. Instead, this protein was highly toxic, affecting the liver and spleen, with possible effect on other organs, decreasing the survival of treated animals. The results indicate the need for studies to better detoxification or manipulation of parasporin for therapeutic use and new studies for analysis of toxicological effects of repetitive exposure of farmers to this toxin

Products based on botanical insecticides and entomopathogenic fungi have been widely used in organic farming, especially in southern Brazil. Thus, this study investigated, in vitro, the effect of aqueous extracts and commercial formulations of plants with insecticidal activity on Beauveria bassiana. The treatments comprised the botanical insecticides Neempro (azadiractin +3-tigloylazadirachtol), at the concentrations of 0.25, 0.5, 0.75, and 1.0% (v/v), and DalNeem (neem oil emulsifiable), at 0.5, 1.0, 1.5, and 2.0% (v/v) (both commercial formulations of Azadirachta indica (neem)), and the aqueous extracts, at the concentrations of 2.5, 5.0, 7.5, and 10.0% (w/v), of neem seeds, tobacco powder (Nicotiana tabacum), and catigua leaves (Trichilia clausenii). In potato, dextrose, and agar culture medium, the effects of each product on the mycelial growth and the production and viability of conidia of B. bassiana were estimated. According to the adopted compatibility index, the aqueous extracts of neem seeds and leaves catigua, depending on the concentration used, and the botanical insecticide Neempro, were classified as compatible with the entomopathogen, becoming important alternatives to integrate programmes of integrated pest management, especially in organic farming systems.

In der vorliegenden Arbeit wurde eine neue Strategie zur stereoselektiven Synthese von Tetraponerinen entwickelt. Hierzu wurde die Kohlenhydrat-Auxiliar gesteuerte Tandem-Mannich-Michael-Reaktion als Schlüsselschritt der Totalsynthese eingesetzt. Die synthetisierten Tetraponerine wurden in biologischen Prüfungen auf neurotoxische und insektizide Aktivität untersucht. Zur Darstellung von (+)-Tetraponerin-8 wurde zunächst ausgehend von trans-2-Octensäure in sieben linearen Stufen ein chiraler beta-Amino-Aldehydbaustein synthetisiert. Dieser wurde in einer Tandem-Mannich-Michael-Reaktion mit dem Auxiliar 2,3,4-Tri-O-pivaloyl-alpha-D-arabinopyranosylamin und dem Danishefsky-Dien zur Reaktion gebracht und lieferte stereoselektiv das entsprechende 2-substituierte N-Arabinosyl-dehydropiperidinon. Nach weiterer Umsetzung des Dehydropiperidinons konnte (+)-Tetraponerin-8 in einer Gesamtausbeute von 0.6 % über 17 lineare Stufen dargestellt werden. Der Ringschluss zum tricyclischen Aminal wurde alternativ auch über die Freisetzung eines cyclischen Acetals durch saure Hydrolyse durchgeführt. Diese Syntheseroute lieferte (+)-Tetraponerin-8 in einer Gesamtausbeute von 3 % über 19 lineare Stufen.rnrnUm den Einfluss eines Stereozentrums im Aldehyd auf den stereochemischen Verlauf der Tandem-Mannich-Michael-Reaktion zu untersuchen, wurde auch das spiegelbildliche Enantiomer des chiralen beta-Amino-Aldehydbausteins synthetisiert und zur Darstellung von N-Arabinosyl-dehydropiperidinon herangezogen. Hierbei ergab sich ein „mismatched“-Fall unter Bildung eines Diastereomeren-Gemisches im Verhältnis 2:1. Nach einer Cyclisierung über ein cyclisches Acetal konnte (+)-Tetraponerin-7 über 19 lineare Stufen in einer Gesamtausbeute von 0.6 % dargestellt werden. Darüber hinaus wurde auch die racemische Form von Tetraponerin-8 nach einer literaturbekannten Synthese dargestellt, um dessen biologische Wirksamkeit mit der der enantiomerenreinen Verbindung vergleichen zu können.

Der Suche nach neuen Wirkstoffen für den chemischen Pflanzenschutz kommt insbesondere vor dem Hintergrund der steigenden Weltbevölkerung und weniger zur Verfügung stehenden kulturfähigen Ackerflächen eine stetig wachsende Bedeutung zu. Ziel dieser Arbeit war die Synthese von cyclischen Peptiden und Depsipeptiden, die aufgrund ihrer biologischen Aktivität als potentielle Insektizide für den chemischen Pflanzenschutz in Frage kommen. Darüber hinaus sollten von Kohlenhydraten abgeleitete Katalysatoren zur enantioselektiven Cyanhydrinsynthese entwickelt werden, um einen leichten Zugang zu den Bausteinen der Depsipeptide zu ermöglichen. Als vielversprechender Naturstoff mit insektiziden Eigenschaften gilt das cyclische Pentapeptid Cycloaspeptid E, dessen Totalsynthese in 10 Stufen mit einer Gesamtausbeute von 25% erreicht wurde, sodass die Verbindung für biologische Tests bereitgestellt werden konnte. Zusätzlich gelang die Kristallisation der Verbindung, was eine Röntgenstrukturanalyse ermöglichte. Ein Derivat von Cycloaspeptid E sollte 2-Aminonicotinsäure anstelle von Anthranilsäure enthalten. Die Synthese dieser Verbindung wurde auf drei Wegen versucht. Dabei zeigte sich, dass es bei einer zur Totalsynthese des Naturstoffs analogen Strategie zur quantitativen Bildung eines Diketopiperazins kommt. Auf den anderen Routen ließ sich entweder ein Kupplungsschritt nicht realisieren, oder die Verbindung erwies sich unter den gewählten Bedingungen als instabil. Die Darstellung eines 2-Aminonicotinsäure-Derivats von Cycloaspeptid E bleibt daher weiterhin ein ungelöstes Problem, das weiterer Forschung bedarf. Verticilid A1 ist ein cyclisches Depsipeptid, das aufgrund seiner Bindungsfähigkeit an den Ryanodinrezeptor von Insekten, als Leitstruktur für die Suche nach neuen Insektiziden von Interesse ist. Um zu untersuchen, wie wichtig die Esterbindungen im Molekül für die biologische Aktivität sind, sollte das entsprechende Amid-Derivat und das Cyclodepsipeptid mit nur zwei statt vier Esterbindungen hergestellt werden. Hierbei zeigte sich, dass eine zur Darstellung von Verticilid A1 analoge Syntheseroute zu einer ausgeprägten Epimerisierung führt. Eine lineare Synthese der Derivate endet in der Bildung des Diketopiperazins. Weiterhin wurden zwei neue, zueinander pseudoenantiomere Vanadium(IV)-Katalysatoren auf Basis von D-Glucose einerseits und L-Xylose andererseits dargestellt. Diese lassen sich in fünf bzw. 14 Stufen synthetisieren und liefern in der enantioselektiven Katalyse von Mandelsäurenitril Enantiomerenüberschüsse von 89% bzw. 91% bei hohen Ausbeuten. Zusammenfassend lässt sich feststellen, dass im Rahmen dieser Arbeit die Totalsynthese von Cycloaspeptid E erfolgreich durchgeführt wurde, und die Syntheseversuche von weiteren cyclischen Peptiden wichtige Erkenntnisse für weitere Synthesen lieferten. Mit den beiden hergestellten Vanadium(IV)-Komplexen wurden zwei potente, auf Kohlenhydraten basierende Katalysatoren für die enantioselektive Synthese von Cyanhydrinen entwickelt.
