The effects on mitochondrial respiration and complex I NADH oxidase activity of cubebin and derivatives were evaluated. The compounds inhibited the state 3 glutamate/malate-supported respiration of hamster liver mitochondria with IC50 values ranging from 12.16 to 83.96M. NADH oxidase reaction was evaluated in submitochondrial particles. The compounds also inhibited this activity, showing the same order of potency observed for effects on state 3 respiration, as well as a tendency towards a non-competitive type of inhibition (KI values ranging from 0.62 to 16.1M). A potential binding mode of these compounds with complex I subunit B8, assessed by docking calculations, is proposed.

Aim of the study: In a previous study, our group described the gastric protective effect of the hydroalcoholic extract of Brazilian green propolis. The main compounds found in Brazilian green propolis include phenolic acids, such as: caffeic, ferulic, p-coumaric and cinnamic acids. This study was therefore carried out to evaluate the antiulcerogenic property of the main phenolic acids found in Brazilian Green Propolis. Material and methods: The anti-ulcer assays were performed using the following protocols: nonsteroidal-antinflammatory drug (NSAID)-induced ulcer, ethanol-induced ulcer, and stress-induced ulcer. The effects of the phenolic acids on gastric content volume, pH and total acidity, using the pylorus ligated model, were also evaluated. Results: It was observed that treatment using doses of 50 and 250 mg/kg of caffeic, ferulic, p-coumaric and cinnamic acids and positive controls (omeprazol or cimetidine) significantly diminished the lesion index, the total area of the lesion and the percentage of lesion in comparison with the negative control groups. In addition, the percentage of ulcer inhibition was significantly higher in the groups treated with the different phenolic acids, cimetidine or omeprazol, in all the protocols used, compared with the negative control groups. In the model to determine gastric secretion, using ligated pylorus, treatment with phenolic acids and cimetidine reduced the volume of gastricjuice and total acidity and significantly increased the gastric pH (p < 0.05), compared with the control group, with the exception of the group treated with 50 mg/kg of p-coumaric acid, in which no significant difference was observed, compared with the control. In relation to the acute toxicity, none sign of toxicity was observed when phenolic acids, used in this study, were administered for rats in dose of 2000 mg/kg. Conclusions: In conclusion, the results of this study show that caffeic, ferulic, p-coumaric and cinnamic acids display antiulcer activity. (c) 2008 Elsevier Ireland Ltd. All rights reserved.

In this work, a new weakly hemorrhagic metalloproteinase (BthMP) was purified from Bothrops moojeni snake venom. This enzyme was homogeneous by native and SDS-PAGE. It showed a polypeptide chain of 23.5 kDa, pI=7.1, and N-terminal blocked. BthMP is comprised of high proteolytic activity on casein, fibrin and bovine fibrinogen, with no coagulating, esterase or phospholipase A(2) activities; it was inhibited by EDTA, EGTA and 1,10-phenanthroline and maintained its activity on pH from 7.0 to 9.0 and temperature from 5-40 degrees C. Assays with metal ions showed that Ca(2+) is an activator, whereas Zn(2+) and Hg(2+) inhibited about 50 and 80% of its activity, respectively. The edema evidenced the important role of the toxin in the inflammatory activity of the venom. BthMP also caused unclotting, and provoked histological alterations in the gastrocnemius muscle of mice inducing hemorrhage, necrosis and leukocytic infiltrate. The molecular mass and the inhibition assays suggest that the metal loproteinase BthMP belongs to class P-I of SVMPs. (c) 2008 Elsevier Ltd. All rights reserved.

The koala, Phascolarctos cinereus, is a geographically widespread species endemic to Australia, with three currently recognized subspecies: P.c. adustus, P.c. cinereus, and P.c. victor. Intraspecific variation in the mitochondrial DNA (mtDNA) control region was examined in over 200 animals from 16 representative populations throughout the species' range. Eighteen different haplotypes were defined in the approximate to 860 bp mtDNA control region as determined by heteroduplex analysis/temperature gradient gel electrophoresis (HDA/TGGE). Any single population typically possessed only one or two haplotypes yielding an average within-population haplotypic diversity of 0.180 +/- 0.003, and nucleotide diversity of 0.16%. Overall, mtDNA control region sequence diversity between populations averaged 0.67%, and ranged from 0% to 1.56%. Nucleotide divergence between populations averaged 0.51%, and ranged from 0% to 1.53%. Neighbour-joining methods revealed limited phylogenetic distinction between geographically distant populations of koalas, and tentative support for a single evolutionarily significant unit (ESU). This is consistent with previous suggestions that the morphological differences formalized by subspecific taxonomy may be interpreted as clinal variation. Significant differentiation in mtDNA-haplotype frequencies between localities suggested that little gene now currently exists among populations. When combined with microsatellite analysis, which has revealed substantial differentiation among koala populations, we conclude that the appropriate short-term management unit (MU) for koalas is the local population.

The in vitro inhibitory activity of crude EtOH/H(2)O extracts from the leaves and stems of Rosmarinus officinalis L. was evaluated against the following microorganisms responsible for initiating dental caries: Streptococcus mutans, salivarius, S. sobrinus, S. mitts 5 sanguinis, and Enterococcus faecalis. Minimum inhibitory concentrations (MIC) were determined with the broth microdilution method. The bioassay-guided fractionation of the leaf extract, which displayed the higher antibacterial activity than the stem extract, led to the identification of carnosic acid (2) and carnosol (3) as the major compounds in the fraction displaying the highest activity, as identified by HPLC analysis. Rosmarinic acid (1), detected in another fraction, did not display any activity against the selected microorganisms. HPLC Analysis revealed the presence of low amounts of ursolic acid (4) and oleanolic acid (5) in the obtained fractions. The results suggest that the antimicrobial activity of the extract from the leaves of R. officinalis may be ascribed mainly to the action of 2 and 3.

OBJECTIVE To determine the prevalence of erectile dysfunction (ED) in a large cohort of Brazilian men who were screened for prostate cancer, and to determine risk factors in this population, as there are large cultural differences among countries in reporting the frequency of ED, and it is likely that the prevalence of ED among men screened for prostate cancer cannot be generally applied across countries. SUBJECTS AND METHODS The analysis focused on the baseline characteristics of 1008 consecutive South American men from Brazil with no known prostate disease who had routine screening for prostate cancer by urologists. The variables analysed were patient age, urinary symptoms, patient health-related quality of life (HRQL), prostate-specific antigen (PSA) levels, prostate volume and erectile function. To assess lower urinary tract symptoms (LUTS) and HRQL, we used the American Urological Association symptom score and its appended eighth question, respectively. Benign prostatic hyperplasia was defined as a prostate volume of > 30 g. Sexual function was assessed using the five-item version of the International Index of Erectile Function questionnaire. Thus, ED was considered to absent for scores of 22-25, mild for 17-21, mild to moderate for 12-16, moderate for 8-11, or severe for 5-7. Purpose: To define the role of magnetization transfer imaging (MTI) in detecting subclinical central nervous system (CNS) lesions in primary antiphospholipid syndrome (PAPS). Materials and Methods: Ten non-CNS PAPS patients were compared to 10 CNS PAPS patients and 10 age- and sex-matched controls. All PAPS patients met Sapporo criteria. All Subjects underwent conventional MRI and complementary MTI analysis to compose histograms. CNS viability was determined according to the magnetization transfer ratio (MTR) by mean pixel intensity (MPI) and the mean peak height (MPH). Volumetric cerebral measurements were assessed by brain parenchyma factor (BPF) and total/cerebral volume. Results: MTR histograms analysis revealed that MPI was significantly different among groups (P < 0.0001). Non-CNS PAPS had a higher MPI than CNS PAPS, (30.5 +/- 1.01 vs. 25.1 +/- 3.17 percent unit (pu); P < 0.05) although lower than controls (30.5 +/- 1.01 vs. 31.20 < 0.50 pu; P < 0.05). MPH in non-CNS PAPS (5.57 +/- 0.20% (1/pu)} was similar to controls (5.63 +/- 0.20% (1/pu), P > 0.05) and higher than CNS PAPS (4.71 +/- 0.30% (1/pu), P < 0.05). A higher peak location (PL) was also observed in the CNS PAPS group in comparison with the other groups (P < 0.0001). In addition, a lower BPF was found in non-CNS PAPS compared to controls (0.80 +/- 0.03 vs. 0.84 +/- 0.02 units; P < 0.05) but similar to CNS PAPS (0.80 +/- 0.03 vs. 0.79 +/- 0.05 units; P > 0.05). Conclusion: Our findings suggest that non-CNS PAPS patients have subclinical cerebral damage. The long-term-clinical relevance of MTI analysis in these patients needs to be defined by prospective studies.

Background: Despite significant advancements in psychopharmacology, treating major depressive disorder (MDD) is still a challenge considering the efficacy, tolerability, safety, and economical costs of most antidepressant drugs. One approach that has been increasingly investigated is modulation of cortical activity with tools of non-invasive brain stimulation - such as transcranial magnetic stimulation and transcranial direct current stimulation (tDCS). Due to its profile, tDCS seems to be a safe and affordable approach. Methods and design: The SELECT TDCS trial aims to compare sertraline vs. tDCS in a double-blinded, randomized, factorial trial enrolling 120 participants to be allocated to four groups to receive sertraline + tDCS, sertraline, tDCS or placebo. Eligibility criteria are moderate-to-severe unipolar depression (Hamilton Depression Rating Scale >17) not currently on sertraline treatment. Treatment will last 6 weeks and the primary outcome is depression change in the Montgomery-Asberg Depression Rating Score (MADRS). Potential biological markers that mediate response, such as BDNF serum levels, Val66Met BDNF polymorphism, and heart rate variability will also be examined. A neuropsychological battery with a focus on executive functioning will be administered. Discussion: With this design we will be able to investigate whether tDCS is more effective than placebo in a sample of patients free of antidepressants and in addition, we will be able to secondarily compare the effect sizes of sertraline vs. tDCS and also the comparison between tDCS and combination of tDCS and sertraline. Background: Blood screening for hepatitis B virus (HBV) is not universally performed for donor selection in human milk banks. Objectives: To evaluate the frequency of detection of HBV surface antigen (HBsAg) and HBV-DNA in colostrum of HBV-infected nursing mothers before and after Holder pasteurization. Study design: Forty-two concentrated breast milk samples were obtained within two postnatal weeks from 24 HBsAg-positive women (4 HBeAg-positive and 20 HBeAg-negative, anti-HBe-positive) were tested for the presence of HBsAg and HBV-DNA before and after Holder pasteurization (30 min at 62.5 degrees C). Results: Before pasteurization, HBsAg and HBV-DNA were found in 14/24 (58%), and 20/24 (75%) first milk samples, respectively, obtained by 4 days after delivery. At least one marker was detected in 20/24 (83%) milk samples. Both markers were identified in milk of HBeAg-positive mothers, and most mothers with anti-HBe in blood had at least one HBV marker. Once detected, viral markers were frequently found in milk samples subsequently obtained from the same woman. Holder pasteurization did not affect the probability of detecting HBsAg (8/18, 44%), HBV-DNA (12/18, 67%). or at least one of them (15118, 83%). Conclusions: Although the biological implications of these findings remain to be determined, considering that HBV is highly contagious and most recipients of banked human milk are preterm infants, these findings should be taken into account when donors are enlisted for human milk banks without serological screening. Yellow fever (YF) vaccines (17D-204 and 17DD) are well tolerated and cause very low rates of severe adverse events (YEL-SAE), such as serious allergic reactions, neurotropic adverse diseases (YEL-AND), and viscerotropic diseases (YEL-AVD). Viral and host factors have been postulated to explain the basis of YEL-SAE. However, the mechanisms underlying the occurrence of YEL-SAE remain unknown. The present report provides a detailed immunological analysis of a 23-year-old female patient. The patient developed a suspected case of severe YEL-AVD with encephalitis, as well as with pancreatitis and myositis, following receipt of a 17D-204 YF vaccination. The patient exhibited a decreased level of expression of Fc-gamma R in monocytes (CD16, CD32, and CD64), along with increased levels of NK T cells (an increased CD3(+) CD16(+/-) CD56(+/-)/CD3(+) ratio), activated T cells (CD4(+) and CD8(+) cells), and B lymphocytes. Enhanced levels of plasmatic cytokines (interleukin-6 [IL-6], IL-17, IL-4, IL-5, and IL-10) as well as an exacerbated ex vivo intracytoplasmic cytokine pattern, mainly observed within NK cells (gamma interferon positive [IFN-gamma(+)], tumor necrosis factor alpha positive [TNF-alpha(+)], and IL-4 positive [IL-4(+)]), CD8(+) T cells (IL-4(+) and IL-5(+)), and B lymphocytes (TNF-alpha(+), IL-4(+), and IL-10(+)). The analysis of CD4(+) T cells revealed a complex profile that consisted of an increased frequency of IL-12(+) and IFN-gamma(+) cells and a decreased percentage of TNF-alpha(+), IL-4(+), and IL-5+ cells. Depressed cytokine synthesis was observed in monocytes (TNF-alpha(+)) following the provision of antigenic stimuli in vitro. These results support the hypothesis that a strong adaptive response and abnormalities in the innate immune system may be involved in the establishment of YEL-AND and YEL-AVD.

Este estudo foi elaborado a partir da proposta de fortalecimento das relações comunitárias entre a Universidade Federal do Espírito Santo e as comunidades jongueiras e caxambuzeiras. Destina-se a apresentar a pesquisa realizada em territórios negros sob a inspiração do Jongo e do Caxambu, reconhecidos como Patrimônio Imaterial do Brasil pelo Instituto do Patrimônio Histórico e Artístico Nacional (Iphan). A pesquisa foi desenvolvida no norte do Estado do Espírito Santo e tem como recurso analítico e conceitual estudos sobre etnicidade no campo da educação. Sua proposta é ampliar e constituir-se como base para a implementação da Lei nº. 10.639/2003, considerando a descrição das categorias religiosidade, territorialidade, memórias, cultura negra, cultura popular e tradição, com base nas narrativas dos sujeitos. Relaciona as práticas culturais do jongo e do caxambu como elementos importantes para a reconstrução da história do negro no Sudeste brasileiro. O tema de investigação foi construído sob a inspiração teórica dos estudos culturais referenciados em Stuart Hall (2008), Canclini (1997), Santos (2008, 2009), Certeau (2005) e na produção simbólica das interpretações sociais, das fronteiras étnicas para descrever as diferenças percebidas pelos sujeitos. Trabalhou-se basicamente propondo as múltiplas interpretações a partir do vivido. O estudo reforça a importância das africanidades na formação de professores e a discussão do Patrimônio Imaterial do Jongo como possibilidades de saberes-fazeres no campo do currículo escolar. Os caminhos da pesquisa partem de uma base etnográfica, conjugando a metodologia da história oral temática com a pesquisa participante e a pesquisa ação, interligando as memórias dos sujeitos, suas narrativas e vivências ao fazer pedagógico no cotidiano das comunidades. Ressalta a relação intercultural e territorial que identifica jongueiros e caxambuzeiros. Os resultados da pesquisa descrevem as condições dessas práticas, da visibilidade das políticas culturais, da produção das identidades jongueiras no norte do Estado do Espírito Santo, sob o ponto de vista dos sujeitos elencados.

Este trabalho aborda aspectos referente à produção do processo de trabalho no cotidiano da gestão da Estratégia Saúde da Família (ESF) em um município do Espírito Santo, a partir dos discursos dos próprios gestores envolvidos no processo. Valoriza a discussão sobre a produção das relações institucionais neste contexto, assim como questões que dizem respeito ao processo de trabalho no dia a dia deste serviço. O percurso metodológico foi orientado pela abordagem qualitativa, sendo utilizado como instrumento metodológico para a coleta de dados a observação direta de uma equipe de ESF, a construção de um diário de campo e entrevistas individuais semiestruturadas aos gestores (secretário municipal de saúde, coordenador da atenção básica, integrante do núcleo da ESF e coordenador de unidade de saúde da família) da ESF desse município. A partir do trabalho de campo e da análise do material produzido, pode-se apreender um cotidiano marcado pelas subjetividades próprias de cada profissional, um cotidiano construído a partir das relações produzidas em cada espaço de atuação dos gestores com os profissionais de saúde e com os usuários. De maneira geral, os discursos dos gestores evidenciam um dia a dia complexo de operar. Além disso, observa-se as que as atividades voltadas para as tomadas de decisão são centradas no papel do gestor formal da instituição de saúde. O organograma da secretaria de saúde do município reforça a hierarquização das relações, principalmente as que se referem às tomadas de decisão, de forma que, pode-se observar que é um dia a dia marcado por tensões, conflitos e controle. As relações produzidas são baseadas em relações de poder, perpetuando características de uma gestão clássica, apesar da concepção e da tentativa de realizar uma gestão pautada na cogestão.

O Sistema único de Saúde (SUS) pode ser considerado uma das maiores conquistas sociais consagradas na Constituição de 1988, representando a materialização de uma nova concepção acerca da saúde no Brasil. As diretrizes do SUS são importantes instrumentos indutores de mudanças e modernização da gestão, incluindo aspectos relativos à organização da assistência, como sua humanização e também a busca de maiores níveis de desempenho e responsabilidade institucional para com os resultados alcançados. Diversos autores tem se debruçado sobre o tema gestão, porém, qual o modelo de gestão que conseguiria combinar um papel ativo, de liderança e de coordenação para gestores com autonomia? Este estudo objetiva analisar a prática da gestão nos serviços de saúde do município de Itapemirim/ES, buscando compreender as múltiplas e complexas dimensões que orientam essa prática, tendo como principal fonte as entrevistas de seus gestores. O município adota uma política de gestão participativa na saúde com várias instâncias formais e pratica um investimento em saúde acima do preconizado pela Constituição Federal o que permite um investimento em estruturação e oferta de serviços públicos. Do ponto de vista metodológico, o estudo adota a abordagem qualitativa através de entrevistas semiestruturadas, focalizando a gestão como um mundo social e expressão dos processos presentes nas organizações e que atravessam os relatos coletados. Foram entrevistados três gestores, sendo esses, Secretário Municipal de Saúde, Diretora da Atenção Primária em Saúde e Coordenadora de Estratégia Saúde da Família do município de Itapemirim. A análise do material empírico teve como um de seus eixos o estudo do percurso profissional dos gestores, especialmente os processos que os levaram à função de gestão. A contingência marca essas trajetórias que se desenrolam em um contexto em que, em alguns dos casos, percebe-se que não existe qualquer exigência quanto à formação de gestão. Outro eixo abordado são as práticas de gestão onde são examinados os sentidos que o exercício da função de gestor tem para os sujeitos, como também as suas estratégias de trabalho, planejamento e ferramentas de uso. O exame das práticas de gestão norteia-se pela análise das possibilidades e limites para desencadear processos de mudança. Os depoimentos apontam as características dessas experiências de gestão, que procuram construir condições para processos de mudança, seja através das experiências anteriores desses gestores ou das estratégias de trabalho e do sistema de gestão articulado que procuram desenvolver. Os projetos profissionais dos gestores entrevistadas vão claramente se definindo no âmbito da gestão: se veem, não mais como profissionais de sua área de formação original (enfermeiro, farmacêutico ou cirurgião dentista), mas acima de tudo como gestores, alimentados pelo reconhecimento de suas capacidades de mobilização e de desenvolvimento de mudanças relativas a outras realizações no campo da gestão.

O Acidente Vascular Cerebral (AVC) é uma das principais causas de prejuízos ao sistema neuromuscular. Dispositivos Robóticos vêm sendo amplamente desenvolvidos e estudados com a finalidade de serem utilizados na assistência à marcha e para o treinamento da marcha durante a reabilitação. O objetivo deste trabalho é avaliar a marcha assistida pelo AROW (Assistive Robotic Walker) em indivíduos hemiparéticos pós-AVC, através da análise de sinais de acelerometria e sinais mioelétricos de superfície (sEMG) provenientes dos músculos vasto medial (VM), bíceps femoral (BF), tibial anterior (TA) e gastrocnêmio medial (GM), e também utilizando os métodos de avaliação GAS (Goal Attainment Scaling) e SUS (System Usability Scale). Nove indivíduos hemiparéticos participaram dos testes. A velocidade da marcha foi reduzida com o uso do AROW e, consequentemente, houve algumas alterações na duração das fases da marcha, por exemplo, uma maior duração da fase de apoio (p = 0,0174). O padrão de ativação muscular para o grupo analisado não apresentou diferença estatisticamente significativa (início da ativação VM: p= 0,4999; término da ativação VM: p= 0,5647; início BF: p= 0,1186; término BF: p= 0,7823; início TA: p= 0,5833; término TA: p= 0,8393; início GM: p= 0,6077; término GM: p= 0,1429). Entretanto, avaliando o padrão de ativação muscular individualmente, podem-se notar algumas alterações benéficas, por exemplo, redução da coativação dos músculos tibial anterior e gastrocnêmio medial. Os resultados das avaliações através do GAS (54,8) e SUS (81,4) sobre o uso do AROW mostraram boa aceitação pelos usuários, e os objetivos esperados durante o uso do andador foram atingidos. A adaptação rápida, facilidade de utilização e sentimento de segurança ao usar o dispositivo são pontos positivos obtidos com o uso do AROW.
