O estudo ora apresentado tem como objetivo analisar o ProJovem enquanto materialização da Política Nacional de Juventude, nos marcos do Governo Lula. O pressuposto central para esta análise está na idéia de que a Política Nacional de Juventude, na forma como foi concebida, materializada quase exclusivamente pelo ProJovem, encontra seus limites no marco das transformações societárias recentes e acaba por reiterar a lógica de constituição histórica das políticas sociais brasileiras, agravada pelos condicionantes políticos e econômicos do pós-1970, comprometendo assim a configuração do seu objetivo, uma vez que nessa conjuntura, a referida política toma por eixo o tripé educação, qualificação profissional e a cidadania, subordinados aos princípios da acumulação capitalista. Para esta análise, optou-se por realizar uma revisão bibliográfica a fim de compreender as principais formulações a respeito da juventude, bem como as suas particularidades na realidade brasileira, tomando como eixo principal para a sua apreensão a perspectiva das expressões da "questão social" sobre condição juvenil. A partir deste debate, a análise da construção da Política Nacional de Juventude toma como pressuposto que esta surge como resposta às manifestações da "questão social", mediada pelas características históricas do processo de constituição das políticas sociais brasileiras e pelas inflexões das novas exigências do modo de produção capitalista, a partir da ofensiva neoliberal e do processo de reestruturação produtiva, no contexto sócio-histórico engendrado no pós-1970. O binômio "inclusão/exclusão social" e o "protagonismo juvenil" são apresentados como os principais eixos que orientam a política, sendo relevante a compreensão do seu significado no contexto das ações voltadas para a juventude. Por fim, buscou-se analisar como os princípios de tal política se materializam no ProJovem, e os limites postos para a efetivação do programa. Para a análise da Política Nacional de Juventude e do ProJovem optou-se por uma pesquisa documental, com base nos instrumentos legais, relatórios e publicações dos profissionais que atuam no programa na realidade do Rio de Janeiro.

A construção de um atendimento voltado exclusivamente para adolescentes não é fortuita. Infância e adolescência são invenções modernas que delimitaram faixas etárias com características próprias e modelos de intervenção específicos que se transformaram historicamente, bem como sendo atravessadas pela própria criação da noção de risco, perigo e vulnerabilidade. A partir de um estudo junto à equipe de uma instituição criada para garantir um tipo de atenção considerado total, voltado para uma clientela designada adolescentes em risco social, busco compreender o modelo de atenção adotado, privilegiando a perspectiva dos profissionais sobre o mesmo. A metodologia empregada consistiu em análise de documentos institucionais e entrevistas com informantes-chaves, terminando por mostrar a relevância de dois tripés estruturantes das práticas institucionais: tratamento da dependência química, prevenção da violência e cuidados básicos de um lado, prevenção aos fatores de risco, acolhimento do paciente e integração dos atendimentos de outro.

O escritor, jornalista e literato Afonso Henriques de Lima Barreto deixou registrados, em praticamente todos os seus escritos, suas impressões sobre as mudanças sociais, políticas e urbanas, sofridas pela cidade do Rio, na Primeira República. A partir de trechos de sua obra e de sua biografia, vão ser analisadas tais transformações à luz da própria avaliação do autor. Serão abordadas as marcas deixadas por modificações urbanas e topográficas ocorridas na então cidade-capital Rio de Janeiro, que se calcaram sobre o velho cenário da cidade, afundando-o, a fim de apagá-lo da memória dos habitantes, ao mesmo tempo em que se erigiam como metáforas de suposto progresso e de modernidade. Metáforas formam em nós, engramas, isto é, marcas que podem ficar retidas em nossa memória pela ação da literariedade que possuem. Literariedade que tem sempre algum espaço nos mais diversos tipos e gêneros de narrativa. Estas marcas de transformações urbanas atingiram de algum modo a rotina diária de parte considerável dos atores que viveram naquele período, entre eles o próprio Lima Barreto. Este trabalho se sustenta, então, no tripé biografia-literatura-metáfora, alinhavado pelas marcas de remodelação da cidade do Rio. Cidade contada por Lima Barreto, durante a Primeira República, numa narrativa que flerta com a etnografia.

Human neutrophils are a type of white blood cell, which forms an early line of defense against bacterial infections. Neutrophils are highly responsive to the chemokine, interleukin-8 (IL-8) due to the abundant distribution of CXCR1, one of the IL-8 receptors on the neutrophil cell surface. As a member of the GPCR family, CXCR1 plays a crucial role in the IL-8 signal transduction pathway in neutrophils. We sequenced the complete coding region of the CXCR1 gene in worldwide human populations and five representative nonhuman primate species. Our results indicate accelerated protein evolution in the human lineage, which was likely caused by Darwinian positive selection. The sliding window analysis and the codon-based neutrality test identified signatures of positive selection at the N-terminal ligand/receptor recognition domain of human CXCR1.

Pituitary adenylate cyclase-activating polypeptide (PACAP) is a neuropeptide abundantly expressed in the central nervous system and involved in regulating neurogenesis and neuronal signal transduction. The amino acid sequence of PACAP is extremely conserved across vertebrate species, indicating a strong functional constraint during the course of evolution. However, through comparative sequence analysis, we demonstrated that the PACAP precursor gene underwent an accelerated evolution in the human lineage since the divergence from chimpanzees, and the amino acid substitution rate in humans is at least seven times faster than that in other mammal species resulting from strong Darwinian positive selection. Eleven human-specific amino acid changes were identified in the PACAP precursors, which are conserved from murine to African apes. Protein structural analysis suggested that a putative novel Deuropeptide might have originated during human evolution and functioned in the human brain. Our data suggested that the PACAP precursor gene underwent adaptive changes during human origin and may have contributed to the formation of human cognition.

MRGX2, a G-protein-coupled receptor, is specifically expressed in the sensory neurons of the human peripheral nervous system and involved in nociception. Here, we studied DNA polymorphism patterns and evolution of the MRGX2 gene in world-wide human populations and the representative nonhuman primate species. Our results demonstrated that MRGX2 had undergone adaptive changes in the path of human evolution, which were likely caused by Darwinian positive selection. The patterns of DNA sequence polymorphisms in human populations showed an excess of derived substitutions, which against the expectation of neutral evolution, implying that the adaptive evolution of MRGX2 in humans was a relatively recent event. The reconstructed secondary structure of the human MRGX2 revealed that three of the four human-specific amino acid substitutions were located in the extra-cellular domains. Such critical substitutions may alter the interactions between MRGX2 protein and its ligand, thus, potentially led to adaptive changes of the pain-perception-related nervous system during human evolution. Background: The emergence of agriculture about 10,000 years ago marks a dramatic change in human evolutionary history. The diet shift in agriculture societies might have a great impact on the genetic makeup of Neolithic human populations. The regionally restricted enrichment of the class I alcohol dehydrogenase sequence polymorphism (ADH1BArg47His) in southern China and the adjacent areas suggests Darwinian positive selection on this genetic locus during Neolithic time though the driving force is yet to be disclosed. Results: We studied a total of 38 populations (2,275 individuals) including Han Chinese, Tibetan and other ethnic populations across China. The geographic distribution of the ADH1B*47His allele in these populations indicates a clear east-to-west cline, and it is dominant in south-eastern populations but rare in Tibetan populations. The molecular dating suggests that the emergence of the ADH1B*47His allele occurred about 10,000 similar to 7,000 years ago. Conclusion: We present genetic evidence of selection on the ADH1BArg47His polymorphism caused by the emergence and expansion of rice domestication in East Asia. The geographic distribution of the ADH1B*47His allele in East Asia is consistent with the unearthed culture relic sites of rice domestication in China. The estimated origin time of ADH1B*47His allele in those populations coincides with the time of origin and expansion of Neolithic agriculture in southern China.

Recent studies showed that nonhuman primate TRIM5 alpha can efficiently block HIV-1 infection in human cell lines. It can also restrict other retroviruses, therefore, suggested as a general defender against retrovirus infection. Here, we present an evolutionary analysis of TRIM5 alpha in primates. Our results demonstrated that TRIM5a has been evolving rapidly in primates, which is likely caused by Darwinian positive selection. The SPRY domain of TRM5 alpha, which may be responsible for recognition of incoming viral capsids showed higher nonsynonymous/synonymous substitution ratios than the non-SPRY domain, indicating that the adaptive evolution of TRIM5a ill primates might be an innate strategy developed in defending retrovirus infection during primate evolution. In addition, the comparative protein sequence analysis suggested that the amino acid substitution pattern at a single site (344R/Q/P) located in the SPRY domain may explain the differences in Susceptibilities of HIV-1 infection in diverse primate species. Antimicrobial peptides secreted by the skin of many amphibians play an important role in innate immunity. From two skin cDNA libraries of two individuals of the Chinese red belly toad (Bombina maxima), we identified 56 different antimicrobial peptide cDNA sequences, each of which encodes a precursor peptide that can give rise to two kinds of antimicrobial peptides, maximin and maximin H. Among these cDNA, we found that the mean number of nucleotide substitution per non-synonymous site in both the maximin and maximin H domains significantly exceed the mean number of nucleotide substitution per synonymous site, whereas the same pattern was not observed in other structural regions, such as the signal and propiece peptide regions, suggesting that these antimicrobial peptide genes have been experiencing rapid diversification driven by Darwinian selection. We cloned and sequenced seven genes amplified from skin or liver genomic DNA. These genes have three exons and share the same gene structure, in which both maximin and maximin H are encoded by the third exon. This suggests that alternative splicing and somatic recombination are less likely to play a role in creating the diversity of maximins and maximin Hs. The gene trees based on different domain regions revealed that domain shuffling or gene conversion among these genes might have happened frequently.

The vomeronasal receptor 1 (V1R) are believed to be pheromone receptors in rodents. Here we used computational methods to identify 95 and 62 new putative V1R genes from the draft rat and mouse genome sequence, respectively. The rat V1R repertoire consists of 11 subfamilies, 10 of which are shared with the mouse, while rat appears to lack the H and I subfamilies found in mouse and possesses one unique subfamily (M). The estimations of the relative divergence times suggest that many subfamilies originated after the split of rodents and primates. The analysis also reveals that these clusters underwent an expansion very close to the split of mouse and rat. In addition, maximum likelihood analysis showed that the nonsynonymous and synonymous rate ratio for most of these clusters was much higher than one, suggesting the role of positive selection in the diversification of these duplicated V1R genes. Because V1R are thought to mediate the process of signal transduction in response to pheromone detection, we speculate that the V1R genes have evolved under positive Darwinian selection to maintain the ability to discriminate between large and complex pheromonal mixtures.

We study the origin of robustness of yeast cell cycle cellular network through uncovering its underlying energy landscape. This is realized from the information of the steady-state probabilities by solving a discrete set of kinetic master equations for the network. We discovered that the potential landscape of yeast cell cycle network is funneled toward the global minimum, G1 state. The ratio of the energy gap between G1 and average versus roughness of the landscape termed as robustness ratio ( RR) becomes a quantitative measure of the robustness and stability for the network. The funneled landscape is quite robust against random perturbations from the inherent wiring or connections of the network. There exists a global phase transition between the more sensitive response or less self-degradation phase leading to underlying funneled global landscape with large RR, and insensitive response or more self-degradation phase leading to shallower underlying landscape of the network with small RR. Furthermore, we show that the more robust landscape also leads to less dissipation cost of the network. Least dissipation and robust landscape might be a realization of Darwinian principle of natural selection at cellular network level. It may provide an optimal criterion for network wiring connections and design.

We uncover the underlying potential energy landscape for a cellular network. We find that the potential energy landscape of the mitogen-activated protein-kinase signal transduction network is funneled toward the global minimum. The funneled landscape is quite robust against random perturbations. This naturally explains robustness from a physical point of view. The ratio of slope versus roughness of the landscape becomes a quantitative measure of robustness of the network. Funneled landscape is a realization of the Darwinian principle of natural selection at the cellular network level. It provides an optimal criterion for network connections and design. Our approach is general and can be applied to other cellular networks.

A comprehensive field study has been undertaken to investigate sediment resuspension dynamics in the Moreton Bay, a large semi-enclosed bay situated in South East Queensland, Australia. An instrumented tripod, which housed three current meters, three OBS sensors and one underwater video camera, was used to collect the field data on tides, currents, waves and suspended sediment concentrations at four sites (Sites 1, 2, 4, and 5) in the bay. Site I was located at the main entrance, Site 2 at the central bay in deep water, and Sites 4 and 5 at two small bays in shallow water. The bed sediment was fine sand (d(50) = 0. 2 mm) at Site 1, and cohesive sediment at the other three sites. Based on the collected field data, it is found that the dominant driving forces for sediment resuspension are a combination of ocean swell and tidal currents at Site 1, tidal currents at Site 2, and wind-waves at Sites 4 and 5. The critical bed shear stress for cohesive sediment resuspension is determined as 0. 079 Pa in unidirectional flow at Site 2, and 0. 076 Pa in wave-induced oscillatory flow at Site 5.

以钢坯修磨为实用目的研制的一台机器人化三腿机床,具有刚度大、负载能力强、机构简单、工作空间大、无运动耦合及奇异位形等优点,首次解决了钢坯局部修磨自动化问题,配以其它加工执行器还可实现多种工艺要求。建立了该机床的运动学逆、正解方程,对其工作空间和受力分析进行了介绍,并介绍了局部修磨自动化控制系统
