油蒿(Artemisia ordosica Krasch.)是内蒙古鄂尔多斯高原特有的半灌木,构成该地区沙地优势植被类型。主要分布在固定、半固定沙丘,同时在流动沙丘也有少量分布。它在当地经济价值、防风固沙环保方面均处于无以取代的地位。在毛乌素沙地沙漠化日益扩大的严峻态势下,研究其群落地上、地下过程对生境变化的响应不仅对维持干旱、半干旱区生态系统稳定的管理措施上有所帮助,而且也有助于了解全球变化背景下物种对环境条件的长期变化适应策略。 为此,本项研究以毛乌素沙地为研究区域,利用异速生长关系确立不同生境油蒿生物量最佳回归方程,并调查、比较了毛乌素沙地固定沙丘、半固定沙丘和流动沙丘三个生境油蒿灌丛地的生物量、土壤和植被的碳储量、生产力和细根周转、土壤微生物生物量碳、氮和土壤呼吸。具体结果如下: 1. 建立并比较了油蒿枝、株两个水平上各部分(不含细根)生物量异速生长关系式,其中枝形态指标(枝直径BD、枝长BL、叶枝长LBL)与油蒿叶、枝、果各部分生物量的异速关系最好;株水平上冠层面积CA与其叶、枝干、果、粗根各部分生物量的回归效果较好。不同生境生物量与其生长变量的异速生长关系存在差异。2004年调查的油蒿灌丛生物量从固定沙丘、半固定沙丘到流动沙丘分别是354.8,178.3和30.4 g m-2;各部分(叶、枝干、果、粗根、不同径级细根的)生物量都呈递减趋势。地下根与总生物量比值排序为固定>半固定>流动沙丘。不同生境细根生物量垂直分布存在差异,在固定沙丘根可至100 cm,半固定沙丘达90 cm,而在流动沙丘仅为60 cm,这些结果有助于使了解不同生境中的相同物种如何通过自身形态及其生物量调整来适应生境的差异。 2. 不同生境油蒿灌丛地植被碳储量和土壤碳储量在P < 0.05水平上差异显著,其中固定沙丘植被碳储量和土壤碳储量分别为224.04和7521 g C m-2,半固定沙丘是119.27和3029 g C m-2,流动沙丘是16.83和2300 g C m-2。可见沙区土壤有机碳远大于植被碳量。 3. 利用最大值减最小值方法、标准取样法和内生长土芯法研究了不同生境油蒿灌丛地的地上、粗根生产力和地下细根生产力。发现各生境生产力、细根周转都随着植被盖度增加而增加,地下根生产力与总生产力之比随着植被盖度增加而减少;不同生境油蒿灌丛地生产力在P < 0.05水平上存在显著差异,2005年总生产力范围在18.23-293.82 g m-2 yr-1之间;细根总周转率在0.16-0.54 yr-1之间。 4. 利用异速生产关系确立不同生境不同水平上油蒿叶面积的最佳回归关系式并对不同生境的比叶面积(SLA)进行了比较,其中枝水平上各生境叶面积与枝直径、枝叶长、枝长相关关系在P < 0.001水平上显著;株水平上各生境叶面积与株高、冠层面积相关关系在P < 0.001水平显著;从固定沙丘、半固定沙丘到流动沙丘SLA由大变小,这可能与生境养分差异有关。 5. 不同生境油蒿灌丛地土壤微生物碳、氮和土壤呼吸范围分别在117.99-153.99 mg kg-1、1.49-3.31mg kg-1和0.54-1.96 μmol m-2 s-1之间,它们从固定沙丘、半固定沙丘到流动沙丘依次下降。

Aim: To establish a method for cynomolgus monkey sperm cryopreservation in a chemically defined extender. Methods: Semen samples were collected by electro-ejaculation from four sexually mature male cynomolgus monkeys. The spermatozoa were frozen in straws by liquid nitrogen vapor using egg-yolk-free Tes-Tris (mTTE) synthetic extender and glycerol as cryoprotectant. The effects of glycerol concentration (1%,3%, 5%, 10% and 15% [v/v]) and its equilibration time (10 min, 30 min, 60 min and 90 min) on post-thaw spermatozoa were examined by sperm motility and sperm head membrane integrity. Results: The post-thaw motility and head membrane integrity of spermatozoa were significantly higher (P < 0.05) for 5% glycerol (42.95 +/- 2.55 and 50.39 +/- 2.42, respectively) than those of the other groups (1%: 19.19 +/- 3.22 and 24.84 +/- 3.64; 3%: 34.23 +/- 3.43 and 41.37 +/- 3.42; 10%: 15.68 +/- 2.36 and 21.39 +/- 3.14; 15%: 7.47 +/- 1.44 and 12.90 +/- 2.18). The parameters for 30 min equilibration (42.95 2.55 and 50.39 2.42) were better (P < 0.05) than those of the other groups (10 min: 31.33 +/- 3.06 and 38. 98 +/- 3.31; 60 min: 32.49 +/- 3.86 and 40.01 +/- 4.18; 90 min: 31.16 +/- 3.66 and 38.30 +/- 3.78). Five percent glycerol and 30 min equilibration yielded the highest post-thaw sperm motility and head membrane integrity. Conclusion: Cynomolgus monkey spermatozoa can be successfully cryopreserved in a chemically defined extender, which is related to the concentration and the equilibration time of glycerol.

目的:研究异丙酚对大鼠海马CA1区自发性兴奋性突触后电流(sEPSC)的影响。方法:断头法分离Wistar大鼠(13~19 d)海马半脑,用切片机切出400μm厚度的海马脑片,全细胞膜片钳记录CA1区锥体神经元sEPSC。20张脑片分为两组:脂肪乳剂组(n=10)和异丙酚组(n=10)。两组细胞稳定10~15 min后,加入90μl脂肪乳剂或异丙酚(相当于100μmol/L),记录40 min sEPSC。膜钳制电压为-70 mV。结果:100μmol/L异丙酚降低sEPSC的频率达68.1%,降低sEPSC的幅值达29.1%,缩短sEPSC的半衰期达49.3%;另外,异丙酚缩短sEPSC的上升时间达29.1%,减少曲线下面积达74.7%。结论:异丙酚通过影响突触前膜递质释放和突触后膜受体功能两个因素抑制兴奋性突触活动

Many ionotropic receptors are modulated by extracellular H+. So far, few studies have directly addressed the role of such modulation at synapses. In the present study, we investigated the effects of changes in extracellular pH on glycinergic miniature inhibitory postsynaptic currents (mIPSCs) as well as glycine-evoked currents (I-Gly) in mechanically dissociated spinal neurons with native synaptic boutons preserved. H+ modulated both the mIPSCs and I-Gly, biphasically, although it activated an amiloride-sensitive inward current by itself. Decreasing extracellular pH reversibly inhibited the amplitude of the mIPSCs and I-Gly, while increasing external pH reversibly potentiated these parameters. Blockade of acid-sensing ion channels (ASICs) with amiloride, the selective antagonist of ASICs, or decreasing intracellular pH did not alter the modulatory effect of H+ on either mIPSCs or I-Gly, H+ shifted the EC50 of the glycine concentration-response curve from 49.3 +/- 5.7 muM at external pH 7.4 to 131.5 +/- 8.1 muM at pH 5.5, without altering the Cl- selectivity of the glycine receptor (GlyR), the Hill coefficient and the maximal I-Gly, suggesting a competitive inhibition of I-Gly by H+. Both Zn2+ and H+ inhibited I-Gly. However, H+ induced no further inhibition of I-Gly in the presence of a saturating concentration of Zn2+. In addition, H+ significantly affected the kinetics of glycinergic mIPSCs and I-Gly. It is proposed that H+ and/or Zn2+ compete with glycine binding and inhibit the amplitude of glycinergic mIPSCs and I-Gly. Moreover, binding of H+ induces a global conformational change in GlyR, which closes the GlyR Cl- channel and results in the acceleration of the seeming desensitization of IGly as well as speeding up the decay time constant of glycinergic mIPSCs. However, the deprotonation rate is faster than the unbinding rate of glycine from the GlyR, leading to reactivation of the undesensitized GlyR after washout of agonist and the appearance of a rebound I-Gly. H+ also modulated the glycine cotransmitter, GABA-activated current (I-GABA). Taken together, the results support a 'conformational coupling' model for H+ modulation of the GlyR and suggest that W may act as a novel modulator for inhibitory neurotransmission in the mammalian spinal cord.

Twenty strains of Microcystis Kutz were isolated from different freshwater bodies in China to analyze the diversity, geographical distribution and toxin profiles. Based on whole-cell polymerase chain reaction of cpcBA-IGS nucleotide sequence, the derived neighbor-joining (NJ) and maximum parsimony (MP) trees indicate that these strains of Microcystis can be divided into four clusters. The strains from south, middle and north region of China formed distinct lineages, suggesting high diversity and a geographical distribution from south to north locations. Moreover, the results being indicating high variable genotypes of the strains of the Microcystis strains from the same lake show that there is high diversity of Microcystis within a water bloom population. Comparing the results of the present study with those reported for compared with 43 strains of Microcystis from other locations, also reveals Chinese strains have high similarity with those from regions in the North Hemispherical. This suggests that the Microcystis strains in the world might have a geographical distribution. Analysis of 30 strains using the primers MCF/TER and TOX2P/TOX2M showed that there was no correlation between the gene of cpcBA-IGS and the presence of mcy. Toxic strains were founded to be predominant in different water bodies throughout China.

A growth trial was conducted at 30 degrees C to investigate the effect of body size on growth and energy budget of Nile tilapia. The average initial body weights of the four size groups tested were 9.3, 34.1, 80.3 and 172.4 g, respectively. Fish were fed to satiation twice a day with a diet containing 35.6% crude protein. Food consumption (C-max: kJ/day) increased with body size (W: g) according to the relationship: Ln C-max = 1.45 + 0.42 LnW. The final body contents of dry matter, crude protein and ash per unit body weight increased with increasing body size while contents of fat and energy were independent of body size. Specific growth rates of wet weight, dry weight, protein and energy decreased as the fish increased in size. Feed efficiencies in wet weigh, dry weight and crude protein decreased with increasing body size, while that of energy remained unchanged. The proportions of energy intake allocated to the various components (faecal energy, excretory energy, heat production and recovered energy) of the energy budget were not significantly affected by body size, and the average budget was: 100IE-18.5(+/- 1.33)FE + 5.9 (+/- 3.09)(ZE + UE) + 49.3(+/- 3.77)HE + 26.3(+/- 6.23)RE, where IE, FE, (ZE + UE), HE and RE represent gross energy intake, faecal energy, excretory (non-faecal) energy loss, heat production and recovered energy (growth), respectively. It is suggested that the decrease in growth rate in larger fish is mainly due to the decrease in relative food intake. (C) 1997 Elsevier Science B.V.

Based oil rare equations of semiconductor laser, a symbolically-defined model for optical transmission system performance evaluation and network characterization in both time- and frequency domains is presented. The steady-state and small-signal characteristics, such as current-photon density curve, current-voltage curve, and input impedance, call be predicted from this model. Two important dynamic characteristics, second-order harmonic distortion and two-tone third-order intermodulation products, are evaluated under different driving conditions. 1985年贲昆龙等首次发现人和猴T淋巴细胞能与树鼩(Tupaia belangeri)的红细胞(TRBC)形成亲和力极强的玫瑰花结,它与绵羊红细胞(SRBC)玫瑰花结(E花结)明显不同。例如,TRBC经神经氨酸酶处理之后,结花率明显下降,TRBC与T淋巴细胞所形成的玫瑰花结,经45℃保温,仍不受影响,为了进一步探讨TRBC受体和SRBC受体(CD2),以及与其它T细胞表面分化搞原(CD)的系。我用某些抗人T细胞CD的单克隆抗体(McAb)对人和猴淋巴细胞进行玫瑰花结抑制试验,抗原调变和共调变(Antigenic modulation or co-modulation)实验,并且研究了TRBC受体在其它免疫细胞和某些人类和长臂猿细胞系的分布。结果表明,TRBC与外周血E~+-PBL形成玫瑰花结的百分率为88.8%。而E~--PBL仅为4.16%。TRBC受体存在于所有被试T细胞系(CEM, H33JHJA1, Jurkat, MLA-144, Molt-3, Molt-4, Molt-4 clone-8 和PEER),但不存在于外周血粒细胞,B细胞,以及B细胞系的绝大多数细胞表面(Daudi, Raji和Reh)。分布于全T细胞的CD3, TCR, CD5, CD6和CD7的相McAb OKT3, T108(F1), T136(F101-15), T149(M-T604)和T152(7G5)均不能调变和共调变TRBC受体。猴和人外周血淋巴细胞与TRBC玫瑰花结的形成,不被T11.1 McAb OKT11所阻断,相反,OKT11显著地阻断猴和人外周血淋巴细胞E玫瑰花结的形成,最大抑制率分别为49.3%和77.7%。在世界各地10个实验室送交第四次国际人白细胞化化抗原讨论会待鉴定的13个CD2 McAb中,除T089 (39C1.5)因抗体量不够未作实验外,对其他12种McAb都进行试验,T081 (x/3),T082 (GLB-T11.2/1),T083 (GLB-T11.1/1),T085 (RPA-2.10),T1088 (0-275)和T092 (M-T201)对TRBC玫瑰花结和E玫瑰花结都呈现明显的阻断作用,T084 (F110.08),T091 (AICD2.1)以及已知参数CD2 McAb-T086 (D-66 clonel)和T087 (GT-2)都不阻TRBC玫瑰花结的形成,亦不调变TRBC受体,而对E玫瑰花结则有不同程度阻断效应,并且调变E变体,使E玫瑰花结形成细胞百分率下降。T090 (6F10.3)和T198 (JOR-T2)即不阻TRBC玫瑰花结的形成,也不抑制E玫瑰花结的形成。由此可见,TRBC受体分布于全T细胞,它不同于已知的全T细胞表面分化抗原CD2 (gp50), CD3/TCR复合物,CD5, CD6和CD7。CD2分子不与TRBC玫瑰花结的形成,也不是介导E玫瑰花结的唯一分子。至少有二个或三个以上的蛋白质与E玫瑰花结和TRBC玫瑰花结的形成有关,其中有的分子为E受体和TRBC受体所共有。TRBC受体很有可能包括新的T淋巴细胞分化抗原。
