A utilização agrícola de biossólidos tem sido muito incentivada, mas, como esses resíduos apresentam composição química variada, o valor agronômico e os efeitos sobre características indicadoras de qualidade do solo precisam ser avaliados caso a caso, a fim de estabelecer normas de segurança para uso desses materiais. Neste trabalho foram avaliadas características biológicas, após a aplicação, por dois anos consecutivos, de doses crescentes (0, 6, 12, 18 e 24 t ha-1 base seca) de um biossólido gerado por uma indústria de fibras e resinas PET e da adubação mineral completa no cultivo de milho, em um Cambissolo distrófico, comparados aos de uma área adjacente, sob Brachiaria sp. e sem cultivo nos últimos 10 anos, usada como referência. Os valores de C e N da biomassa microbiana, a respiração basal e as atividades das enzimas urease e beta-glicosidase e da hidrólise do diacetato de fluoresceína (FDA) aumentaram, enquanto a atividade da fosfatase ácida diminuiu com a elevação das doses de biossólido, porém estas não tiveram efeito sobre o quociente metabólico (qCO2). A diminuição da atividade da fosfatase se deveu ao aumento da disponibilidade de P no solo, não caracterizando efeito adverso da aplicação do biossólido. Com aplicação de 12 t ha-1 de biossólido (recomendação agronômica), a respiração e a hidrólise da FDA foram maiores e a atividade da fosfatase foi menor que a obtida no solo com adubação mineral, mas as demais características avaliadas não diferiram entre estes tratamentos. A colonização micorrízica de Brachiaria sp. não diferiu entre plantas de crescimento espontâneo nas parcelas anteriormente cultivadas com milho e aquelas da área adjacente. Apesar do menor número de esporos, verificou-se enriquecimento de espécies de fungos micorrízicos arbusculares (FMAs) nas parcelas cultivadas. O carbono orgânico (Corg) e a biomassa microbiana apresentaram alta correlação com os demais parâmetros avaliados, indicando que as alterações na quantidade e qualidade da matéria orgânica, promovidas pela aplicação do biossólido, refletiram na dinâmica da microbiota e influenciaram positivamente os parâmetros biológicos de qualidade do solo.

Two cleavage pathways of beta-carotene have been proposed, one by central cleavage and the other by random (excentric) cleavage. The central cleavage pathway involves the metabolism of beta-carotene at the central double bond (15, 15') to produce retinal by beta-carotene 15, 15'-dioxygenase (E.C.888990988). The random cleavage of beta-carotene produces beta-apo-carotenoids, but the mechanism is not clear. To understand the various mechanisms of beta-carotene cleavage, beta-carotene was incubated with the intestinal postmitochondrial fractions of 10-week-old male rats for 1 h and cleavage products of beta-carotene were analyzed using reverse-phase, high-performance liquid chromatography (HPLC). We also studied the effects of alpha-tocopherol and NAD(+)/NADH on beta-carotene cleavage. In addition to beta-carotene, we used retinal and beta-apo-14'-carotenoic acid as substrates in these incubations. Beta-apo-14'-carotenoic acid is the two-carbon longer homologue of retinoic acid. In the presence of alpha-tocopherol, beta-carotene was converted exclusively to retinal, whereas in the absence of alpha-tocopherol, both retinal and beta-apo-carotenoids were formed. Retinoic acid was produced from both retinal and beta-apo-14'-carotenoic acid incubations only in the presence of NAD(+). Our data suggest that in the presence of an antioxidant such as alpha-tocopherol, beta-carotene is converted exclusively to retinal by central cleavage. In the absence of an antioxidant, beta-carotene is cleaved randomly by enzyme-related radicals to produce beta-apo-carotenoids, and these beta-apo-carotenoids can be oxidized further to retinoic acid via retinal. beta-Glucosidase from the fungus Thermoascus aurantiacus grown oil semi-solid fermentation medium (using ground corncob as substrate) was partially purified in 5 steps - ultrafiltration, ethanol precipitation, gel filtration and 2 anion exchange chromatography runs, and characterized. After the first anion exchange chromatography, beta-glucosidase activity was eluted in 3 peaks (Gl-1, Gl-2, Gl-3). Only the Gl-2 and Gl-3 fractions were adsorbed on the gel matrix. Gl-2 and Gl-3 exhibited optimum pH at 4.5 and 4.0, respectively. The temperature optimum of both glucosidases was at 75-80 degreesC. The pH stability of Gl-2 (4.0-9.0) was higher than Gl-3 (5.5-8.5); both enzyme activities showed similar patterns of thermostability. Under conditions of denaturing gel chromatography the molar mass of Gl-2 and Gl-3 was 175 and 157 kDa, respectively. Using 4-nitrophenyl beta-D-glucopyranoside as substrate, K-m, values of 1.17 +/- 0.35 and 1.38 +/- 0.86 mmol/L were determined for Gl-2 and Gl-3, respectively. Both enzymes were inhibited by Ag+ and stimulated by Ca2+.

In order to investigate the effect of folate depletion, lymphocyte sister-chromatid exchange (SCE) rates were compared among homozygous β-thalassaemic patients with low folic acid levels, heterozygous β-thalassaemic patients with normal folate levels and healthy persons with normal haemoglobin, in cultures with both normal and depleted folate conditions. Significantly higher SCE rates were found in homozygous patients in all assays, but the in vitro folate depletion did not induce an increase in SCE frequency in any group.

Microbial β-glucosidases have been used for the enhancement of wine aroma. Nevertheless, few enzymes are active in the conditions of winemaking. In this work, the production of a β-glucosidase by an Aureobasidium pullulans strain (Ap-β-gl) isolated from grape ecosystems was evaluated. The maximum enzymatic synthesis using submerged fermentation was after 96 h of growth in complex media containing 20 g/L of cellobiose as the sole carbon source. The crude enzyme (Ap-β-gl) showed optimal pH at 5.5 and two peaks of optimum temperature (at 45 and 70 C). It showed a wide range of pH stability, stability at low temperatures, and tolerance to ethanol, showing suitable characteristics for winemaking conditions. The hydrolysis of glycosidic terpenes by Ap-β-gl was studied, and its ability to efficiently release free terpenols was demonstrated by gas chromatography/mass spectrometry. The enzymatic treatment notably increased the amount of monoterpenes, showing good prospects for its potential application for the development of aroma in wines. © 2012 Springer Science+Business Media New York.

Os óleos vegetais vêm sendo estudados, por alguns anos, de forma intensiva. Entretanto o estudo dessas substâncias apresenta certa dificuldade pelo fato destes óleos apresentarem composições químicas muito complexas. Neste estudo apresentado em forma de dissertação, apresentaremos algumas das formas variadas do espectro Raman do Beta-caroteno e do óleo de buriti em diferentes concentrações e em diferentes meios. O óleo de buriti é uma substância que possui muitas propriedades, entre elas propriedades ópticas e medicinais. É formado por uma composição de várias substâncias graxas e não graxas. Duas das substâncias encontradas no OB também fazem parte de nosso estudo que são o beta-caroteno e o ácido oleico. O beta-caroteno é um carotenoide precursor da vitamina A, muito encontrado em frutos e verduras que apresentam coloração vermelho-alaranjado. O ácido oleico é um ácido graxo muito importante e está presente, quase sempre em grande concentração, na maioria dos óleos vegetais. Com o intuito de estudarmos os espectros Raman desses materiais foram dissolvidos 6,1 mg de beta-caroteno em 50 ml de ácido oléico chamado de solução base. A partir dessa solução, foram misturadas diferentes quantidades de ácido oleico a fim de obtermos soluções mais diluídas, gerando assim soluções de concentrações menores que a solução base, até ser atingido uma concentração próxima a 0% de beta-caroteno. Todas as soluções foram submetidas a agitação mecânica. Uma em especial foi submetida a agitação tanto mecânica quanto ao Ultra-Som. As blendas de PMMA modificados com óleo de buriti foram fabricadas no Laboratório de Químico Física da UNB, numa parceria do Grupo de Polímeros da UNB e do Grupo de Física de Materiais da Amazônia da UFPA. Os resultados mostram que as intensidades Raman aumentam ou diminuem, conforme a concentração de beta-caroteno na solução e de óleo de buriti na blenda. Mostra que em 1656 cm-1 aparece um pico referente a presença de óleo de buriti na blenda e o deslocamento de alguns modos em algumas regiões do espectro da blenda. Com relação ao espectro da solução foi identificado mudança na solução submetida ao ultra-som com o desaparecimento de alguns modos refrentes ao ácido oleico e ao beta caroteno.

BACKGROUND: Contagious bovine pleuropneumonia (CBPP) caused by Mycoplasma mycoides subsp. mycoides small-colony type (SC) is among the most serious threats for livestock producers in Africa. Glycerol metabolism-associated H2O2 production seems to play a crucial role in virulence of this mycoplasma. A wide number of attenuated strains of M. mycoides subsp. mycoides SC are currently used in Africa as live vaccines. Glycerol metabolism is not affected in these vaccine strains and therefore it does not seem to be the determinant of their attenuation. A non-synonymous single nucleotide polymorphism (SNP) in the bgl gene coding for the 6-phospho-beta-glucosidase (Bgl) has been described recently. The SNP differentiates virulent African strains isolated from outbreaks with severe CBPP, which express the Bgl isoform Val204, from strains to be considered less virulent isolated from CBPP outbreaks with low mortality and vaccine strains, which express the Bgl isoform Ala204. RESULTS: Strains of M. mycoides subsp. mycoides SC considered virulent and possessing the Bgl isoform Val204, but not strains with the Bgl isoform Ala204, do trigger elevated levels of damage to embryonic bovine lung (EBL) cells upon incubation with the disaccharides (i.e., beta-D-glucosides) sucrose and lactose. However, strains expressing the Bgl isoform Val204 show a lower hydrolysing activity on the chromogenic substrate p-nitrophenyl-beta-D-glucopyranoside (pNPbG) when compared to strains that possess the Bgl isoform Ala204. Defective activity of Bgl in M. mycoides subsp. mycoides SC does not lead to H2O2 production. Rather, the viability during addition of beta-D-glucosides in medium-free buffers is higher for strains harbouring the Bgl isoform Val204 than for those with the isoform Ala204. CONCLUSION: Our results indicate that the studied SNP in the bgl gene is one possible cause of the difference in bacterial virulence among strains of M. mycoides subsp. mycoides SC. Bgl does not act as a direct virulence factor, but strains possessing the Bgl isoform Val204 with low hydrolysing activity are more prone to survive in environments that contain high levels of beta-D-glucosides, thus contributing in some extent to mycoplasmaemia.

There is an increasing demand for novel metal-based complexes with biologically relevant molecules in technology and medicine. Three new Cu(II) coordination compounds with antifungal agent isoconazole (L), namely mononuclear complexes CuCl2(L)(2) (1), and Cu(O2CMe)(2)(L)(2)center dot 2H(2)O (2) and coordination polymer Cu(pht)(L)(2)(n) (3) (where H(2)pht - o-phthalic acid) were synthesized and characterized by IR spectroscopy, thermogravimetric analysis and X-ray crystallography. X-ray analysis showed that in all complexes, the isoconazole is coordinated to Cu(II) centres by a N atom of the imidazole fragment. In complex I, the square-planar environment of Cu(II) atoms is completed by two N atoms of isoconazole and two chloride ligands, whereas the Cu(II) atoms are coordinated by two N atoms from two isoconazole ligands and two O atoms from the different carboxylate residues: acetate in 2 and phthalate in 3. The formation of an infinite chain through the bridging phthalate ligand is observed in 3. The biosynthetic ability of micromycetes Aspergillus niger CNMN FD 10 in the presence of the prepared complexes 1-3 as well as the antifungal drug isoconazole were studied. Complexes 2 and 3 accelerate the biosynthesis of enzymes (beta-glucosidase, xylanase and endoglucanase) by this fungus. Moreover, a simplified and improved method for the preparation of isoconazole nitrate was developed.

In the present study, we report the results of comprehensive amino acid (AA) analyses of four Indian lakes from different climate regimes. We focus on the investigation of sediment cores retrieved from the lakes but data of modern sediment as well as vascular plant, soil, and suspended particulate matter samples from individual lakes are also presented. Commonly used degradation and organic matter source indices are tested for their applicability to the lake sediments, and we discuss potential reasons for possible limitations. A principal component analysis including the monomeric AA composition of organic matter of all analysed samples indicates that differences in organic matter sources and the environmental properties of the individual lakes are responsible for the major variability in monomeric AA distribution of the different samples. However, the PCA also gives a factor that most probably separates the samples according to their state of organic matter degradation. Using the factor loadings of the individual AA monomers, we calculate a lake sediment degradation index (LI) that might be applicable to other palaeo-lake investigations.
