Time series of terrigenous source elements (Al, K, Ti, Zr) from core GeoB4901-8 recovered from the deep-sea fan of the Niger River record variations in riverine sediment discharge over the past 245,000 yr. Although the flux rates of all the elements depend on physical erosion, which is mainly controlled by the extent of vegetation coverage in central Africa, element/Al ratios reflect conditions for chemical weathering in the river basin. Maximum sediment input to the ocean occurs during cold and arid periods, when precipitation intensity and associated freshwater runoff are reduced. High carbonate contents during the same periods indicate that the sediment supply has a positive effect on river-induced marine productivity. In general, variations in the terrestrial signals contain a strong precessional component in tune with changes in low-latitude solar radiation. However, the terrestrial signal lags the insolation signal by several thousand years. K/Al, Ti/Al, and Zr/Al records reveal that African monsoonal precipitation depends on high-latitude forcing. We attribute the shift between insolation cycle and river discharge to the frequently reported nonlinear response of African climate to primary orbital configurations, which may be caused by a complex interaction of the secondary control parameters, such as surface albedo and/or thermohaline circulation.

An in situ iron enrichment experiment was carried out in the Southern Ocean Polar Frontal Zone and fertilized a patch of water within an eddy of the Antarctic Circumpolar Current (EisenEx, Nov. 2000). During the experiment, a physical speciation technique was used for iron analysis in order to understand the changes in iron distribution and size-fractionations, including soluble Fe (<200 kDa), colloidal Fe (200 kDa-0.2 µm) and labile particle Fe (>0.2 µm), throughout the development of the phytoplankton bloom. Prior to the first infusion of iron, dissolved (<0.2 µm) iron concentrations in the ambient surface seawater were extremely low (0.06±0.015 nM) with colloidal iron being a minor fraction. For the iron addition, an acidified FeSO4 solution was released three times over a 23-day period to the eddy. High levels of dissolved iron concentrations (2.0±1.1 nM) were measured in the surface water until 4 days after the first iron infusion. After every iron infusion, when high iron concentrations were observed before storm events, there was a significant correlation between colloidal and dissolved iron concentrations ([Colloidal Fe]=0.7627[Dissolved Fe]+0.0519, R2=0.9346). These results indicate that a roughly constant proportion of colloidal vs. dissolved iron was observed after iron infusion (~76%). Storm events caused a significant decrease in iron concentrations (<0.61 nM in dissolved iron) and changed the proportions of the three iron size-fractions (soluble, colloidal and labile particle). The changes in each iron size-fraction indicate that colloidal iron was eliminated from surface mixed layer more easily than particulate and soluble fractions. Therefore, particle and soluble iron efficiently remain in the mixed layer, probably due to the presence of suspended particles and naturally dissolved organic ligands. Our data suggest that iron removal through colloidal aggregation during phytoplankton bloom should be considered in the oceanic iron cycle.

Baeyer–Villiger oxidation of cyclic ketones, using H2O2 as the oxidising agent, was systematically studied using a range of metal chlorides in different solvents, and in neat chlorogallate(III) ionic liquids. The extremely high activity of GaCl3 in promoting oxidation with H2O2, irrespective of solvent, was reported for the first time. The activity of all other metal chlorides was strongly solvent-dependent. In particular, AlCl3 was very active in a protic solvent (ethanol), and tin chlorides, SnCl4 and SnCl2, were active in aprotic solvents (toluene and dioxane). In order to eliminate the need for volatile organic solvent, a Lewis acidic chlorogallate(III) ionic liquid was used in the place of GaCl3, which afforded typically 89–94% yields of lactones in 1–120 min, at ambient conditions. Raman and 71Ga NMR spectroscopic studies suggest that the active species, in both GaCl3 and chlorogallate(III) ionic liquid systems, are chlorohydroxygallate(III) anions, [GaCl3OH]−, which are the products of partial hydrolysis of GaCl3 and chlorogallate(III) anions; therefore, the presence of water is crucial.

Le compartiment microvasculaire est une cible importante du stress oxydant qui est un facteur majeur de la dysfonction endothéliale, notamment au cours d’exposition aux rayonnements ionisants. L’altération de l’endothélium induite par le stress oxydant est impliquée dans la toxicité radio-induite des tissus sains. Limiter les dysfonctions endothéliales est donc un enjeu important des traitements radiothérapeutiques actuels. Cet objectif nécessite une meilleure caractérisation de la signalisation du stress oxydant dans les cellules endothéliales. La voie p38 MAPK est incontournable dans la réponse au stress oxydant mais reste encore insuffisamment caractérisée. Par une approche protéomique, nous avons identifié la nucléophosmine (NPM) comme nouveau partenaire de p38 dans le cytoplasme des cellules endothéliales. La phosphatase PP2a est aussi associée à ce complexe NPM/p38. Nos travaux montrent que le stress oxydant (H2O2, 500μM) régule la déphosphorylation de NPM via PP2a, entraine sa dissociation rapide du complexe et favorise sa translocation vers le noyau. De plus, nous montrons que la présence de NPM déphosphorylée au noyau altère la réponse des cellules aux dommages à l’ADN induits par le stress oxydant. Le céramide sphingolipide membranaire est également un facteur important des voies de stress, particulièrement dans les cellules endothéliales. Notre étude aborde donc l’implication de ce sphingolipide dans la régulation de la voie NPM/p38. Une meilleure caractérisation de la voie p38 et de ses acteurs permettra d’identifier de potentielles cibles afin de limiter les dysfonctions endothéliales et leurs conséquences délétères sur les tissus environnants.

The research presented in this dissertation is aimed to the automation of the Fenton process. The Fenton reaction is finalized to the waste water pre-treatment in order to promote the abatement of the organic contaminants and make it more degradable. Reagents adopted are constituted by a mixture of iron ions and hydrogen peroxide and their effect is strictly influenced by several variables, such as: the reagents molar ratio and their quantities counterpoised to the substrate, temperature, pH, agitation, etc. Therefore, the optimization is far from being considered an easy procedure. The research was carried out using a batch configuration, through which the optimal [Fe2+]/[H2O2] and [substrate]/[H2O2] ratios were identified. Then, in order to improve the process, a semibatch configuration was performed. The preliminary results show that is possible to obtain a greater abatement efficiency for high organic burden using the semibatch configuration here proposed.

Neuronal stretching during concussion alters glucose transport and reduces neuronal viability, also affecting other cells in the brain and the Blood Brain Barrier (BBB). Our hypothesis is that oxidative stress (OS) generated in neurons during concussions contributes to this outcome. To validate this, we investigated: (1) whether OS independently causes alterations in brain and BBB cells, namely human neuron-like, neuroblastoma cells (NCs), astrocyte cells (ACs) and brain microvascular endothelial cells (ECs), and (2) whether OS originated in NCs (as in concussion) is responsible for causing the subsequent alterations observed in ACs and ECs. We used H2O2 treatment to mimic OS, validated by examining the resulting reactive oxygen species, and evaluated alterations in cell morphology, expression and localization of the glucose transporter GLUT1, and the overall cell viability. Our results showed that OS, either directly affecting each cell type or originally affecting NCs, caused changes in several morphological parameters (surface area, Feret diameter, circularity, inter-cellular distance), slightly varied GLUT1 expression and lowered the overall cell viability of all NCs, ACs, and ECs. Therefore, we can conclude that oxidative stress, which is known to be generated during concussion, caused alterations in NCs, ACs, and ECs whether independently originated in each cell or when originated in the NCs and could further propagate the ACs and ECs.

Creative ways of utilising renewable energy sources in electricity generation especially in remote areas and particularly in countries depending on imported energy, while increasing energy security and reducing cost of such isolated off-grid systems, is becoming an urgently needed necessity for the effective strategic planning of Energy Systems. The aim of this research project was to design and implement a new decision support framework for the optimal design of hybrid micro grids considering different types of different technologies, where the design objective is to minimize the total cost of the hybrid micro grid while at the same time satisfying the required electric demand. Results of a comprehensive literature review, of existing analytical, decision support tools and literature on HPS, has identified the gaps and the necessary conceptual parts of an analytical decision support framework. As a result this research proposes and reports an Iterative Analytical Design Framework (IADF) and its implementation for the optimal design of an Off-grid renewable energy based hybrid smart micro-grid (OGREH-SμG) with intra and inter-grid (μG2μG & μG2G) synchronization capabilities and a novel storage technique. The modelling design and simulations were based on simulations conducted using HOMER Energy and MatLab/SIMULINK, Energy Planning and Design software platforms. The design, experimental proof of concept, verification and simulation of a new storage concept incorporating Hydrogen Peroxide (H2O2) fuel cell is also reported. The implementation of the smart components consisting Raspberry Pi that is devised and programmed for the semi-smart energy management framework (a novel control strategy, including synchronization capabilities) of the OGREH-SμG are also detailed and reported. The hybrid μG was designed and implemented as a case study for the Bayir/Jordan area. This research has provided an alternative decision support tool to solve Renewable Energy Integration for the optimal number, type and size of components to configure the hybrid μG. In addition this research has formulated and reported a linear cost function to mathematically verify computer based simulations and fine tune the solutions in the iterative framework and concluded that such solutions converge to a correct optimal approximation when considering the properties of the problem. As a result of this investigation it has been demonstrated that, the implemented and reported OGREH-SμG design incorporates wind and sun powered generation complemented with batteries, two fuel cell units and a diesel generator is a unique approach to Utilizing indigenous renewable energy with a capability of being able to synchronize with other μ-grids is the most effective and optimal way of electrifying developing countries with fewer resources in a sustainable way, with minimum impact on the environment while also achieving reductions in GHG. The dissertation concludes with suggested extensions to this work in the future.

El proceso de acondicionamiento consiste en la saturación del sustrato con agua y posteriormente el lavado con un volumen de una disolución acuosa de MA entre 1 y 10 veces igual al volumen de compost. La mezcla peroxiacética (MA) es una disolución acuosa de peróxido de hidrógeno (PH) y ácido peracético (PAA) en distintas proporciones. La concentración de MA en la disolución acuosa que proponemos para su uso como acondicionamiento del compost está comprendida entre 1-10%. La concentración de peróxido de hidrógeno (H2O2) en el MA está comprendida entre 10-40% y la de ácido peroxiacético (C2H4O3) entre 1-10%. El método de acondicionamiento del compost con mezcla peroxiacética (MA) para su uso directo como sustrato agronómico tiene como objeto la obtención de compost de alta calidad, incluso con posibilidad de ser una alternativa a la turba de Sphagnum, presentando como aliciente que el coste medioambiental y económico de este proceso es menor que otras alternativas.

This study compares the antioxidant and antimicrobial transcriptional expression of blue shrimps reared according to two different systems, BioFloc Technology (BFT) and Clear sea Water (CW) and their differential responses when facing an experimental sublethal hydrogen peroxide stress. After 30 days of rearing, juvenile shrimps were exposed to H2O2 stress at a concentration of 30 ppm during 6 hours. The oxidative stress caused by H2O2 was examined in the digestive glands of the shrimp, in which antioxidant enzyme (AOE) and antimicrobial peptide (AMP) gene expression were analysed by quantitative real-time PCR. Results showed that rearing conditions did not affect the expression of genes encoding AOEs or AMPs. However, H2O2 stress induced a differential response in expression between shrimps from the two rearing treatments (BFT and CW). Comparative analysis of the expression profiles indicates that catalase transcripts were significantly upregulated by H2O2 stress for BFT shrimps while no change was observed for CW shrimps. In contrast, H2O2 caused down-regulation of superoxide dismutase and glutathione transferase transcripts and of the three AMP transcripts studied (penaeidin 2 and 3, and crustin) for CW shrimps, while no effect was observed on BFT shrimp transcript levels. These results suggested that BFT shrimps maintained antioxidant and AMP responses after stress and therefore can effectively protect their cells against oxidative stress, while CW shrimp immune competence seems to decrease after stress.

The study of green chemistry is dedicated to eliminating or reducing toxic waste. One route to accomplish this goal is to explore alternative reaction conditions and parameters resulting in the development of more benign synthetic routes and reagents. The primary focus of this research is to find optimal reaction conditions for the oxidation of a primary alcohol to an aldehyde. As a case study, the oxidation of benzyl alcohol to benzaldehyde, a common industrial process, was examined. Traditionally carried out using the Jones Reagent, commonly referred to as chromium (IV) oxide or chromium trioxide (CrO3) in sulphuric acid, a great deal of research went into utilizing less toxic reagents, such as MnO2 or KMnO4 supported on a clay base. This research has led to an improvement on these alternatives, using a lithium chloride (LiCl) catalyst in a montmorillonite K10 clay solid phase, together with the oxidizing agent hydrogen peroxide, as even greener alternatives to these traditional oxidizing agents. Experiments were carried out to determine the lifetime of this LiCl/clay system as compared to MnO2 and KMnO4, to investigate its ability to catalyze the oxidation of other aromatic alcohols (such as 4-methoxybenzyl alcohol and diphenylmethanol), and to further improve the system’s adherence to green chemistry principles. Green solvent alternatives were examined by replacing the toluene solvent with dimethylcarbonate (DMC), and reaction conditions were optimized to improve product yield. It was determined that the LiCl/H2O2 system was, in most cases, equally as effective at catalyzing the oxidation of benzyl alcohol to benzaldehyde. Although the catalyst and oxidizing agent eliminated the toxic waste generated from chromium reagents, it offered significant challenges in product isolation, because of an aqueous-organic phase separation.

O stress oxidativo está associado ao envelhecimento e a inúmeras patologias, nomeadamente a doenças neurodegenerativas e cardiovasculares, e a diversos outros fatores. O stress oxidativo leva à oxidação de importantes biomoléculas como os lípidos e, ao contrário da maior parte dos produtos de oxidação de fosfolípidos e ácidos gordos insaturados (PUFAS), os produtos de oxidação de glicosfingolípidos (GSLs) têm sido escassamente estudados. Os glicosfingolípidos são moléculas muito diversificadas estruturalmente e com importantes funções, essencialmente no sistema nervoso central (SNC) onde estão localizados maioritariamente. Deste modo, alterações na estrutura dos GSLs conduzirão a consequente comprometimento das suas funções e ao possível desenvolvimento de patologias. Assim para identificar as modificações oxidativas que ocorrem em glicosfingolípidos e pressupor consequentes efeitos biológicos nas células sob stress oxidativo, prepararamse sistemas modelo biomiméticos com diferentes GSLs os quais foram expostos a radicais hidroxilo gerados sob condições da reação de Fenton (H2O2 e Fe2+) e as reações foram monitorizadas por diferentes metodologia utilizando a espectrometria de massa. Os resultados obtidos com este estudo permitiram-nos identificar vários produtos de oxidação produzidos durante a oxidação desta classe de lípidos. Os produtos de oxidação observados em comum, em todos os GSLs estudados (C16:0GalCer, C24:1GalCer, C24:1LacCer e GM1) foram as suas correspondentes ceramidas. Estas atuam como agentes pro-apoptóticos e podem in vivo promover a neurodegeneração nas células sob stress oxidativo. Também foi possível observar produtos com inserção de oxigénio junto às duplas ligações ou na cadeia de esfingosina (no caso do GM1) ou na cadeia de ácido gordo monoinsaturada (no caso da C24:1GalCer, C24:1LacCer), corroborando o facto de que ácidos gordos saturados não são susceptíveis à oxidação por radicais. Interessantemente em ambos os GSLs de cadeias glicosiladas compostas com mais de um açúcar (C24:1LacCer e GM1) observou-se a despolimerização oxidativa da porção glicosilada por quebra das correspondentes ligações glicosídicas. Esta degradação leva à formação de GlcCer no caso de oxidação de LacCer ou na formação de outros gangliósidos (GM2, GM3, asialoGM1 e asialoGM2) e glicolípidos (LacCer e GlcCer), no caso de oxidação de GM1. A formação por via radicalar não enzimática destes GSLs leva a distúrbios no perfil lipídico. Previamente, em certas doenças, foram observadas variações na concentração do perfil de gangliósidos e de ceramidas. Estes dados permitem sugerir que em células em condições de stress oxidativo, a acumulação de gangliósidos mais simples e ceramidas poderá ter uma contribuição de produtos da degradação oxidativa dos gangliósidos e GSLs mais complexos. Este trabalho contribui assim para uma melhor compreensão das modificações estruturais que ocorrem em alguns glicosfingolípidos em condições de stress oxidativo. Os produtos de oxidação aqui identificados suportam a sua possível futura deteção em sistemas biológicos.

Atualmente há uma crescente preocupação quanto à qualidade dos recursos hídricos, e essa notória inquietação se deve em parte ao aumento na demanda e consumo de água a nível global, uma vez que o crescimento econômico, juntamente com o desenvolvimento tecnológico, agrícola e industrial conduz a poluição ambiental. Neste contexto, os agrotóxicos, apresentam um alto fator de risco para a qualidade dos recursos hídricos, pois essas substâncias geralmente são tóxicas e não biodegradáveis. Diante deste cenário, um dos maiores desafios nos dias atuais é a eliminação de uma parte significativa, dessa poluição, a qual é causada por esses contaminantes orgânicos tóxicos. O processo clássico de oxidação biológica falha na eliminação de compostos tóxicos, bem como de contaminantes orgânicos recalcitrantes. Além disso, os processos físico-químicos, como a adsorção em carvão ativado, floculação, e a filtração por membranas, apenas transferem esses contaminantes de fase, sem que ocorra a sua destruição. Sendo assim, surge a necessidade da adoção de técnicas que possam ser destrutivas a essas espécies. Os Processos Oxidativos Avançados, surgem como um caminho alternativo para diminuir e eliminar os resíduos desses contaminantes orgânicos. Por tanto, tiveram-se como objetivos neste trabalho: (i) desenvolver e otimizar um processo Fenton empregando ferro zero e peróxido de hidrogênio (Fe0 /H2O2) (ii) utilizar como fonte de ferro zero para o processo Fenton limalha de ferro, a qual é um resíduo da atividade metalúrgica na cidade de Rio Grande (iii) otimizar o tempo de reação (TR) e tempo de homogeneização (TH) para a Fotocatálise Heterogênea empregando um catalisador de sílica dopada com dióxido de titânio e dicloreto de tris (2,2-bipiridina) rutênio II (iv) desenvolver um método cromatográfico empregando LC-DAD para realizar o monitoramento da degradação dos agrotóxicos (v) empregar a LC-MS/MS para confirmar a degradação dos agrotóxicos e monitorar a formação de produtos (vi) empregar IC para monitorar a formação de espécies iônicas (vii) realizar a determinação de COT para estimar a mineralização do carbono orgânico dissolvido. Para tanto, um total de seis agrotóxicos foram selecionados para a o estudo de degradação: bentazona, carbofurano, clomazona, diurom, tebuconazole e piraclostrobina. O sistema Fe0 /H2O2 desenvolvido para a degradação dos agrotóxicos mostrou ser eficiente. A eficiência de degradação foi fortemente afetada pelo pH, massa de limalha de ferro e concentração de peróxido de hidrogênio. As melhores condições para a degradação foram: pH 2,0, 5 mmol L-1 H2O2 e 2,0 g de limalha de ferro. No tempo total de 20 min o carbono orgânico total foi reduzido levando a mineralização de 55%, com taxas de degradação que variaram de 51 a 100%. A utilização de recirculação no sistema Fe0 /H2O2 elevou a taxa de degradação dos agrotóxicos bentazona e carbofurano. As taxas variaram de 93 a 100% de degradação, com um tempo total de reação de 120 min, chegando a 63% de mineralização. O sistema TiO2/UV também mostrou-se adequado a degradação dos agrotóxicos. As condições ótimas foram 20 mg de catalisador, pH 7, TH de 15 min e TR de 110 min. O sistema apresentou taxas de degradação que variaram de 71 a 99,98%. Levando a mineralização de 97,60% da demanda de carbono orgânico.

El cianuro es el compuesto químico empleado por excelencia para la lixiviación de oro en la industria minera. Sin embargo, es altamente tóxico para los organismos que se desarrollan alrededor de las industrias mineras, y para el medio ambiente. Con el fin de reducir los niveles de cianuro libre en efluentes provenientes de la minería, el trabajo se enfocó en determinar las condiciones óptimas para la degradación de cianuro empleando compuestos químicos y un consorcio microbiano. Los ensayos químicos y biológicos se realizaron por separado, utilizando muestras de efluentes provenientes de la minería a diferentes concentraciones de cianuro (280 y 10 mg/l CN-). Para la degradación química se utilizó tres oxidantes diferentes: hipoclorito de sodio, peróxido de hidrógeno y ácido de caro en diferentes concentraciones, pH (10-11) y tiempos de degradación (4,71 y 20,75 h). Para los ensayos de biodegradación se empleó un consorcio microbiano en matraces que contenían el efluente cianurado y medio líquido a pH (11), agitación (200 rpm) y temperatura (20±5°C). Se midió la concentración de cianuro libre, pH y la concentración de biomasa. Los resultados del tratamiento químico mostraron que el mejor compuesto oxidante fue el peróxido de hidrógeno (8:1 gH2O2/gCN-) a pH (10), obteniendo un 92,7% remoción de cianuro libre en 45 minutos (280 mg/l CN-) y un 91,0% de remoción en 25 minutos (10 mg/l CN-). Mientras que en la degradación biológica en matraces la remoción fue del 73,7% (280 mg/l CN-) en 384 h y de 78,6% (10 mg/l CN-) en 240 h.
