The purpose of the present work was to study various aspects of the germination of seeds of Senna occidentalis Link, which had presented promising results in biological activity against the etiological agent of malaria. These aspects were dormancy, temperatures of germination and photoblastic response. In the dormancy studies, the treatments used were: unscarified seed (control); tegument puncture with sharp point; and scarification with sand-paper and immersion in concentrated sulfuric acid during 5, 10, 15 and 20 minutes. For the study of temperature and photoblast, the seeds were immersed in sulfuric acid for 20 minutes, submitted to temperatures of 10 to 45degreesC, at intervals of 5degreesC, both under light and in the dark. The seeds presented dormancy related to tegument, the best treatments for breaking dormancy were immersion in sulfuric acid for 15 and 20 minutes. At temperatures 25 and 30degreesC, the best results of percentage and index of velocity of germination were observed, at which the seeds behaved as neutral photoblastic; there was no germination at 10 or at 45degreesC.

The mycelia-to-yeast (M-Y) transition, thermal tolerance and virulence profiles were evaluated for nine isolates of Paracoccidioides brasiliensis, including samples from two of the three recently discovered cryptic species, as well as their relation to the partial sequence and transcription of the hsp70 gene. The isolates Bt84 and T10 (from PS2 species) took more time to convert to yeast form and presented elongated yeast cells at 36 degrees C. Arthroconidia production was also observed during the M-Y transition for some isolates. Our data confirm that the hsp70 transcription may be associated with thermal tolerance, but this does not seem to be directly related to high virulence profiles. The partial sequencing of this gene allowed the separation of our isolates into two clusters that correspond to the two sympatric cryptic species occurring in an area hyperendemic for PCM (Botucatu, SP, Brazil).

Caesalpinia echinata and C ferrea var. ferrea have different seed behaviours and seed and fruit types. Comparison of the seed ontogeny and anatomy partly explained the differences in seed behaviour between these two species of Brazilian legumes; some differences were also related to fruit development. The seed coat in C. ferrea consisted of two layers of osteosclereids, as well as macrosclereids and fibres, to form a typical legume seed coat, whereas C. echinata had only macrosclereids and fibres. In C. echinata, the developing seed coat had paracytic stomata, a feature rarely found in legume seeds. These seed coat features may account for the low longevity of C. echinata seeds. The embryogeny was similar in both species, with no differences in the relationship between embryo growth and seed growth. The seeds of both species behaved as typical endospermic seeds, despite their different morphological classification (exendospermic orthodox seeds were described for C. echinata and endospermic orthodox seeds for C. ferrea). Embryo growth in C. ferrea accelerated when the sclerenchyma of the pericarp was developing, whereas embryonic growth in C. echinata was associated with the conclusion of spine and secretory reservoir development in the pericarp. Other features observed included an endothelial layer that secreted mucilage in both species, a nucellar summit, which grew up into the micropyle, and a placental obturator that connected the ovarian tissue to the ovule in C. ferrea. A espécie Passiflora cincinnata Mast., pertencente à família Passifloraceae, é silvestre e popularmente conhecida como maracujá-do-mato, sendo considerada importante na produção de porta-enxertos, uma vez que é tolerante à seca, a doenças causadas por bactérias e a nematóides, além de poder ser utilizada em programas de melhoramento genético. O trabalho teve como objetivos estudar o efeito da luz e da temperatura e a interação entre temperatura e reguladores vegetais na germinação de sementes de Passiflora cincinnata Mast. Foi constituído de três experimentos: no primeiro, estudou-se o efeito da luz e da temperatura na germinação de sementes; no segundo, o efeito de diferentes concentrações dos reguladores vegetais GA4+7 + N-(fenilmetil)-aminopurina na germinação das sementes e, no terceiro, a interação entre temperatura e reguladores vegetais na germinação das sementes. O delineamento experimental foi inteiramente casualizado para todos os experimentos e os dados foram submetidos à análise de variância e comparação das médias pelo teste Tukey, a 5% de probabilidade. É possível observar que a luz exerce efeito inibitório sobre a germinação das sementes, e que os reguladores vegetais, GA4+7 + N-(fenilmetil)-aminopurina, são eficientes na superação da dormência, além de ampliarem os limites de temperatura da germinação. A temperatura alternada 20-30ºC mostra-se a mais adequada para a germinação de sementes dessa espécie.

Portuguese chestnut (Castanea sativa) is used for forest products, raw nuts and processed flour, paste, and candy. We studied the influence on germination efficiency of seed with and without partial tegument removal combined with different substrate composition (coconut fiber, pine compost and vermiculite) at São Paulo State University, Campus of Botucatu, São Paulo, Brazil. Randomized blocks were used in a factorial scheme 3x2 (three substrates x two seed types) consisting of six treatments with five replications (twelve seeds). Seeds were sowed in expanded polystyrene trays, with 72 cells, and maintained at 25 C under controlled environment. Rate, time elapsed and speed of seed germination was submitted to ANOVA and the average compared using Tukey's 5% test of probability and curves adjusted based on Gompertz regression. Coconut fiber or vermiculite associated with seed without partial tegument removal showed the highest germination rate, and coconut fiber with or without partial tegument removal displayed the fastest speed of seed germination.

center dot Background and Aims Drought is a major environmental constraint affecting growth and production of Coffea canephora. Selection of C. canephora clones has been largely empirical as little is known about how clones respond physiologically to drought. Using clones previously shown to differ in drought tolerance, this study aimed to identify the extent of variation of water use and the mechanisms responsible, particularly those associated morphological traits.center dot Methods Clones (14 and 120, drought-tolerant; 46 and 109A, drought-sensitive, based on their abilities to yield under drought) were grown in 120-L pots until they were 12-months old, when an irrigation and a drought treatment were applied; plants were droughted until the pressure potential (Psi(x)) before dawn (pre-dawn) reached -3.0 MPa. Throughout the drought period, Psi(x) and stomatal conductance (g(s)) were measured. At the end of the experiment, carbon isotope ratio and parameters from pressure-volume curves were estimated. Morphological traits were also assessed.center dot Key Results and Conclusions With irrigation, plant hydraulic conductance (K-L), midday Psi(x) and total biomass were all greater in clones 109A and 120 than in the other clones. Root mass to leaf area ratio was larger in clone 109A than in the others, whereas rooting depth was greater in drought-tolerant than in drought-sensitive clones. Predawn Psi(x) of -3.0 MPa was reached fastest by 109A, followed progressively by clones 46, 120 and 14. Decreases in g(s) with declining Psi(x), or increasing evaporative demand, were similar for clones 14, 46, and 120, but lower in 109A. Carbon isotope ratio increased under drought; however, it was lower in 109A than in other clones. For all clones, Psi(x), g(s) and KL recovered rapidly following re-watering. Differences in root depth, KL and stomatal control of water use, but not osmotic or elastic adjustments, largely explained the differences in relative tolerance to drought stress of clones 14 and 120 compared with clones 46 and 109A.

An Arabidopsis thaliana cDNA clone encoding a plant uncoupling mitochondrial protein (AtPUMP1) was overexpressed in transgenic tobacco plants. Analysis of the AtPUMP1 mRNA content in the transgenic lines, determined by Northern blot, revealed variable levels of transgene expression. Antibody probing of Western blots of mitochondrial proteins from three independent transgenic lines showed significant accumulation of AtPUMP1 in this organelle. Overproduction of AtPUMP1 in transgenic tobacco plants led to a significant increase in tolerance to oxidative stress promoted by exogenous hydrogen peroxide as compared to wild-type control plants. These results provide the first biological evidence for a role of PUMP in protection of plant cells against oxidative stress damage.
