Sensitive and highly specific quantitative real-time PCR and ELISA for recording a potential transfer of novel DNA and Cry1Ab protein from feed into bovine milk

To address food safety concerns of the public regarding the potential transfer of recombinant DNA (cry1Ab) and protein (Cry1Ab) into the milk of cows fed genetically modified maize (MON810), a highly specific and sensitive quantitative real-time PCR (qPCR) and an ELISA were developed for monitoring suspicious presence of novel DNA and Cry1Ab protein in bovine milk. The developed assays were validated according to the assay validation criteria specified in the European Commission Decision 2002/657/EC. The detection limit and detection capability of the qPCR and ELISA were 100 copies of cry1Ab microL(-1) milk and 0.4 ng mL(-1) Cry1Ab, respectively. Recovery rates of 84.9% (DNA) and 97% (protein) and low (<15%) imprecision revealed the reliable and accurate estimations. A specific qPCR amplification and use of a specific antibody in ELISA ascertained the high specificity of the assays. Using these assays for 90 milk samples collected from cows fed either transgenic (n = 8) or non-transgenic (n = 7) rations for 6 months, neither cry1Ab nor Cry1Ab protein were detected in any analyzed sample at the assay detection limits.

Gegenstand des Werks bilden «Aktuelle Fragen aus dem Erbrecht». Das Erbrecht ist ein eigentliches Kerngebiet notarieller Tätigkeit, und Fragen aus dem Erbrecht stellen sich den Notarinnen und Notaren in mannigfacher Hinsicht und in immer wieder neuer Art und Weise. Prof. Dr. Jolanta Kren Kostkiewicz behandelt das im Zeitalter zunehmender Mobilität bedeutender werdende Internationale Erbrecht. Der Beitrag von Prof. Dr. Stephan Wolf gilt dem Thema Erbrecht in besonderen Situationen; er geht ein auf das Konkubinat, die Ehekrise und das neue Erwachsenenschutzrecht.Rechtsanwalt Dr. Philipp Studhalter befasst sich mit praktischen Aspekten der Nutzniessung gemäss Art. 473 ZGB, einem Klassiker der Rechtsgeschäftsgestaltung unter Ehegatten. Notar und Rechtsanwalt Peter Muntwyler und Notar/Grundbuchverwalter Dr. Roland Pfäffli, beide Mitglieder der Musterurkundensammlungs-Kommission des Verbandes bernischer Notare, widmen sich dem Erbenschein in der Praxis.

Old captains at the helm: chairman age and firm performance

This paper examines whether the chairmen of the boards (COBs) impose their life cycles on the firms over which they preside. Using a large sample of unlisted firms, we find a robust negative relation between COB age and firm performance. COBs age much like ‘ordinary’ people. Their cognitive abilities deteriorate, and they experience significant shifts in motivation. Deteriorating cognitive abilities are the main driver of the performance effect that we observe. The results imply that succession planning problems in unlisted firms are real. Mandatory retirement age clauses cannot solve these problems.

Cone-beam computed tomography-guided stereotactic liver punctures: a phantom study

PURPOSE Images from computed tomography (CT), combined with navigation systems, improve the outcomes of local thermal therapies that are dependent on accurate probe placement. Although the usage of CT is desired, its availability for time-consuming radiological interventions is limited. Alternatively, three-dimensional images from C-arm cone-beam CT (CBCT) can be used. The goal of this study was to evaluate the accuracy of navigated CBCT-guided needle punctures, controlled with CT scans. METHODS Five series of five navigated punctures were performed on a nonrigid phantom using a liver specific navigation system and CBCT volumetric dataset for planning and navigation. To mimic targets, five titanium screws were fixed to the phantom. Target positioning accuracy (TPECBCT) was computed from control CT scans and divided into lateral and longitudinal components. Additionally, CBCT-CT guidance accuracy was deducted by performing CBCT-to-CT image coregistration and measuring TPECBCT-CT from fused datasets. Image coregistration was evaluated using fiducial registration error (FRECBCT-CT) and target registration error (TRECBCT-CT). RESULTS Positioning accuracies in lateral directions pertaining to CBCT (TPECBCT = 2.1 ± 1.0 mm) were found to be better to those achieved from previous study using CT (TPECT = 2.3 ± 1.3 mm). Image coregistration error was 0.3 ± 0.1 mm, resulting in an average TRE of 2.1 ± 0.7 mm (N = 5 targets) and average Euclidean TPECBCT-CT of 3.1 ± 1.3 mm. CONCLUSIONS Stereotactic needle punctures might be planned and performed on volumetric CBCT images and controlled with multidetector CT with positioning accuracy higher or similar to those performed using CT scanners.

The Kinneret Regional Project Excavations of a Byzantine Synagogue at Horvat Kur, Galilee, 2010-2013: A Preliminary Report

The article presents the preliminary results from four seasons of excavations at Horvat Kur in the Galilee. The excavations conducted by the Kinneret Regional Project have exposed the remains of a broad-house synagogue from the Byzantine period. The most important finds include an elevated platform (i. e., a bemah) that supported a chest holding Torah scrolls, an ornamented limestone seat that was probably used by the leader of the congregation, and a basalt stone table that features geometric figures on three sides and figurative representations on one side. The Horvat Kur synagogue represents a valuable example of the diversity of Galilean synagogues that were built or renovated between the 5th and the 7th centuries C.E.
