In Bacillus subtilis, parE and parC were shown to be essential genes for the segregation of replicated chromosomes. Disruption of either one of these genes resulted in failure of the nucleoid to segregate. Purified ParE and ParC proteins reconstituted to form topoisomerase IV (topo IV), which was highly proficient for ATP-dependent superhelical DNA relaxation and decatenation of interlocked DNA networks. By immunofluorescence microscopy and by directly visualizing fluorescence by using green fluorescence protein fusions, we determined that ParC is localized at the poles of the bacteria in rapidly growing cultures. The bipolar localization of ParC required functional ParE, suggesting that topo IV activity is required for the localization. ParE was found to be distributed uniformly throughout the cell. On the other hand, fluorescence microscopy showed that the GyrA and GyrB subunits of gyrase were associated with the nucleoid. Our results provide a physiologic distinction between DNA gyrase and topo IV. The subcellular localization of topo IV provides physical evidence that it may be part of the bacterial segregation machinery.

Like all hyperthermophiles yet tested, the bacterium Thermotoga maritima contains a reverse gyrase. Here we show that it contains also a DNA gyrase. The genes top2A and top2B encoding the two subunits of a DNA gyrase-like enzyme have been cloned and sequenced. The Top2A (type II DNA topoisomerase A protein) is more similar to GyrA (DNA gyrase A protein) than to ParC [topoisomerase IV (Topo IV) C protein]. The difference is especially striking at the C-terminal domain, which differentiates DNA gyrases from Topo IV. DNA gyrase activity was detected in T. maritima and purified to homogeneity using a novobiocin-Sepharose column. This hyperhermophilic DNA gyrase has an optimal activity around 82–86°C. In contrast to plasmids from hyperthermophilic archaea, which are from relaxed to positively supercoiled, we found that the plasmid pRQ7 from Thermotoga sp. RQ7 is negatively supercoiled. pRQ7 became positively supercoiled after addition of novobiocin to cell cultures, indicating that its negative supercoiling is due to the DNA gyrase of the host strain. The findings concerning DNA gyrase and negative supercoiling in Thermotogales put into question the role of reverse gyrase in hyperthermophiles.

We describe here a simple and easily manipulatable Escherichia coli-based genetic system that permits us to identify bacterial gene products that modulate the sensitivity of bacteria to tumoricidal agents, such as DMP 840, a bisnaphthalimide drug. To the extent that the action of these agents is conserved, these studies may expand our understanding agents is conserved, these studies may expand our understanding of how the agents work in mammalian cells. The approach briefly is to use a library of E. coli genes that are overexpressed in a high copy number vector to select bacterial clones that are resistant to the cytotoxic effects of drugs. AtolC bacterial mutant is used to maximize permeability of cells to hydrophobic organic molecules. By using DMP 840 to model the system, we have identified two genes, designated mdaA and mdaB, that impart resistance to DMP 840 when they are expressed at elevated levels. mdaB maps to E. coli map coordinate 66, is located between the parE and parC genes, and encodes a protein of 22 kDa. mdaA maps to E. coli map coordinate 18, is located adjacent to the glutaredoxin (grx) gene, and encodes a protein of 24 kDa. Specific and regulatable overproduction of both of these proteins correlates with DMP 840 resistance. Overproduction of the MdaB protein also imparts resistance to two mammalian topoisomerase inhibitors, Adriamycin and etoposide. In contrast, overproduction of the MdaA protein produces resistance only to Adriamycin. Based on its drug-resistance properties and its location between genes that encode the two subunits of the bacterial topoisomerase IV, we suggest that mdaB acts by modulating topoisomerase IV activity. The location of the mdaA gene adjacent to grx suggests it acts by a drug detoxification mechanism.

The diet of Common Chiffchaffs Phylloscopus collybita wintering in a Mediterranean wetland (El Hondo Natural Park, SE Spain) was studied by analysing the gizzard content of 17 individuals that died accidentally when trapped for ringing. Prey availability was assessed via water-trap sampling over two winters. The bulk of the diet was composed of midges (Chironomidae), which were found in all the gizzards and represented approximately 95% of the prey. Brachycera flies were one of the most captured taxa in the water-traps but represented less than 0.5% of the total number of prey consumed. Compositional analysis revealed very strong prey selection, with Chiffchaffs selecting clumped and less mobile prey, such as chironomids, and avoiding abundant but fast-escaping prey, such as Brachycera.

El principal objetivo de esta investigación es conocer aspectos ecológicos y distribución del arruí — Ammotragus lervia (Pallas, 1777)— dentro de la sierra de Mariola. El área de estudio es un parque natural de 17.500 hectáreas situado en el sur de la Comunidad Valenciana. Un mejor conocimiento de su distribución será de interés para la definición de medidas de gestión de fauna del parque. En 2009, utilizando técnicas de fototrampeo, se recopilaron 29.941 imágenes con algún contacto animal. De estas imágenes, el 0,09% de las fotografías registradas son de arruí y se ha detectado su presencia en 7 de las 63 cuadrículas (2 × 2 km) del Parque Natural de la Sierra de Mariola (el 11,11%). El periodo de muestreo se prolongó desde agosto de 2008 hasta mayo de 2010. Este estudio ha permitido integrar la información recopilada en campo con las bases de datos existentes para confirmar la colonización y la expansión del arruí en la sierra de Mariola.

T2K reports its first measurements of the parameters governing the disappearance of νµ in an off-axis beam due to flavor change induced by neutrino oscillations. The quasimonochromatic νµ beam, produced with a peak energy of 0.6 GeV at J-PARC, is observed at the far detector SuperKamiokande, 295 km away, where the νµ survival probability is expected to be minimal. Using a dataset corresponding to 4.01×10²⁰ protons on target, 34 fully contained µ-like events were observed. The best-fit oscillation parameters are sin²(θ₂₃) = 0.45 and |∆m^2_32| = 2.51 × 10⁻³ eV² with 68% confidence intervals of 0.38 - 0.64 and 2.26 - 2.80 ×10⁻³ eV² respectively. These results are in agreement with existing antineutrino parameter measurements and also with the νµ disappearance parameters measured by T2K.

We report a measurement of the νµ-nucleus inclusive charged current cross section (=σ cc) on ironusing data from exposed to the J-PARC neutrino beam. The detector consists of 14 modules in total, which are spread over a range of off-axis angles from 0◦ to 1.1◦. The variation in the neutrino energy spectrum as a function of the off-axis angle, combined with event topology information, is used to calculate this cross section as a function of neutrino energy. The cross section is measured to be σcc(1.1 GeV) = 1.10±0.15 (10^−38cm^2/nucleon), σcc(2.0 GeV) = 2.07±0.27 (10^−38cm^2/nucleon), and σcc(3.3 GeV) = 2.29 ± 0.45 (10^−38cm^2/nucleon), at energies of 1.1, 2.0, and 3.3 GeV, respectively. These results are consistent with the cross section calculated by the neutrino interaction generators currently used by T2K. More importantly, the method described here opens up a new way to determine the energy dependence of neutrino-nucleus cross sections.

The periphrasis made of aller + infinitive has been very productive in Romance languages. If in French, Spanish or Portuguese, it refers to future, it refers to a simple past in Catalan. fr.: je vais aller au cinéma esp.: voy a ir al cine pt. The same structure seems therefore to have grammaticalised in diverging and even diametrically opposite ways in different Romance languages. In this talk, we shall try to explain how the phrase aller + infinitive is able to function both as a future and a past. We will especially concentrate on the case of contemporary French where, while the future interpretation has obtained the status of tense as futur proche or périphrastique, a past interpretation is now commonplace. Our reflection will be supported by a personal corpus of authentic examples.

Les études typologiques (Hagège 1993, Bybee et al.1994, Dahl 2000, Bourdin 2008 notamment) ont montré de façon consistante que les indications spatiales (notamment les verbes de mouvement) tendaient à se grammaticaliser en expressions temporelles. La forme itive (en français aller) dans les langues romanes a fait l’objet de ce processus linguistique , et avec succès, puisque, en tant qu’auxiliaire d’un verbe à l’infinitif ou au participe présent (et moins fréquemment au participe passé), elle est à même d’offrir, au cours de son histoire, pas moins de onze emplois grammaticalisés (Bres et Labeau à paraître). Nous nous intéresserons dans cet article, qui ne portera que sur le français, à l’emploi que nous nommerons, avec Larreya (2005) et Lansari (2009), narratif: il apparaît en textualité narrative, dont les propositions du premier plan sont régies par la relation de progression (Labov 1972/1978). Cet emploi que le français a connu jusqu’au début du XVIIème (1) tend, sur des bases peut-être différentes, à se répandre aujourd’hui (2): (1) Sur ces propos, feirent leur accord, et, en regardant le lieu le plus propre pour faire ceste belle oeuvre, elle vat dire qu'elle n'en sçavoit poinct de meilleure ne plus loing de tout soupson, que une petite maison qui estoit dedans le parc, où il y avoit chambre et lict tout à propos. Fossil associations from the middle and upper Eocene (Bartonian and Priabonian) sedimentary succession of the Pamplona Basin are described. This succession was accumulated in the western part of the South Pyrenean peripheral foreland basin and extends from deep-marine turbiditic (Ezkaba Sandstone Formation) to deltaic (Pamplona Marl, Ardanatz Sandstone and Ilundain Marl formations) and marginal marine deposits (Gendulain Formation). The micropalaeontological content is high. It is dominated by foraminifera, and common ostracods and other microfossils are also present. The fossil ichnoasssemblages include at least 23 ichnogenera and 28 ichnospecies indicative of Nereites, Cruziana, Glossifungites and ?Scoyenia-Mermia ichnofacies. Body macrofossils of 78 taxa corresponding to macroforaminifera, sponges, corals, bryozoans, brachiopods, annelids, molluscs, arthropods, echinoderms and vertebrates have been identified. Both the number of ichnotaxa and of species (e. g. bryozoans, molluscs and condrichthyans) may be considerably higher. Body fossil assemblages are comparable to those from the Eocene of the Nord Pyrenean area (Basque Coast), and also to those from the Eocene of the west-central and eastern part of South Pyrenean area (Aragon and Catalonia). At the European scale, the molluscs assemblages seem endemic from the Pyrenean area, although several Tethyan (Italy and Alps) and Northern elements (Paris basin and Normandy) have been recorded. Palaeontological data of studied sedimentary units fit well with the shallowing process that throughout the middle and late Eocene occurs in the area, according to the sedimentological and stratigraphical data.

An ethnobotanical survey of medicinal plants was carried out in the Central Middle Atlas in the years 2013 and 2014 to establish the catalog of medicinal plants used in traditional medicine in the treatment of diabetes. Thus, 1560 people were interviewed, using questionnaires. The latter enabled us to gather information on traditional healing practices of the local population including scientific name, French name, vernacular name, plant parts used , therapeutic indications , revenues and mode of administration. The results show that 76 medicinal species were inventoried in the study area. These plant species are included into 67 genus and 40 families. The most represented families are: Lamiaceae (12 species), Asteraceae and Brassicaceae species with 14 each. Of 76 medicinal species found in the region, four species are reported for the first time in the traditional treatment of diabetes in Morocco. They are Pistacia atlantica, Ptychotis verticillata, Anacyclus pyrethrum, Alyssum spinosum, Cistus albidus, Juniperus thurifera, Ephedra nebrodensis, Thymus algeriensis, Th. munbyanus, Th. zygis, Abelmoschus esculentus, Fraxinus augustifolia, Sorghum vulgare and, Eriobotrya japonica. The leaves are the most used organs (38%). The decoction is the dominant mode of preparation (50%) and administration is mostly for oral use (97%).
