The objective of this work was to verify the existence of a lethal locus in a eucalyptus hybrid population, and to quantify the segregation distortion in the linkage group 3 of the Eucalyptus genome. A E. grandis x E. urophylla hybrid population, which segregates for rust resistance, was genotyped with 19 microsatellite markers belonging to linkage group 3 of the Eucalyptus genome. To quantify the segregation distortion, maximum likelihood (ML) models, specific to outbreeding populations, were used. These models consider the observed marker genotypes and the lethal locus viability as parameters. The ML solutions were obtained using the expectation‑maximization algorithm. A lethal locus in the linkage group 3 was verified and mapped, with high confidence, between the microssatellites EMBRA 189 e EMBRA 122. This lethal locus causes an intense gametic selection from the male side. Its map position is 25 cM from the locus which controls the rust resistance in this population.

OBJECTIVE: Assertive community treatment (ACT) failed to develop in Europe, and its efficacy is debated. In Lausanne, Switzerland, ACT focuses on difficult-to-engage patients and aims to facilitate linkage with outpatient care through time-limited interventions. This study aimed to evaluate the applicability and efficiency of time-limited ACT. METHODS: We retrospectively assessed social, clinical, and functional outcomes and motivation for treatment in 75 consecutively seen subjects treated between 2000 and 2002. RESULTS: With 70% of the interventions lasting less than 6 months, we observed significant improvement in most clinical and social problems, in collaboration, in motivation for treatment, and in social network support, despite high baseline levels of clinical and social problems. The number of hospitalizations decreased significantly. CONCLUSIONS: Time-limited ACT is a useful treatment for difficult-to-engage patients with severe clinical and social problems, and it facilitates linkage to care. This narrower target for ACT may facilitate its implementation in Europe.

A simple way to quickly optimize microsatellites in nonmodel organisms is to reuse loci available in closely related taxa; however, this approach can be limited by the stochastic and low cross-amplification success experienced in some groups (e.g. amphibians). An efficient alternative is to develop loci from transcriptome sequences. Transcriptomic microsatellites have been found to vary in their levels of cross-species amplification and variability, but this has to date never been tested in amphibians. Here, we compare the patterns of cross-amplification and levels of polymorphism of 18 published anonymous microsatellites isolated from genomic DNA vs. 17 loci derived from a transcriptome, across nine species of tree frogs (Hyla arborea and Hyla cinerea group). We established a clear negative relationship between divergence time and amplification success, which was much steeper for anonymous than transcriptomic markers, with half-lives (time at which 50% of the markers still amplify) of 1.1 and 37 My, respectively. Transcriptomic markers are significantly less polymorphic than anonymous loci, but remain variable across diverged taxa. We conclude that the exploitation of amphibian transcriptomes for developing microsatellites seems an optimal approach for multispecies surveys (e.g. analyses of hybrid zones, comparative linkage mapping), whereas anonymous microsatellites may be more informative for fine-scale analyses of intraspecific variation. Moreover, our results confirm the pattern that microsatellite cross-amplification is greatly variable among amphibians and should be assessed independently within target lineages. Finally, we provide a bank of microsatellites for Palaearctic tree frogs (so far only available for H. arborea), which will be useful for conservation and evolutionary studies in this radiation.

The objective of this work was to propose a way of using the Tocher's method of clustering to obtain a matrix similar to the cophenetic one obtained for hierarchical methods, which would allow the calculation of a cophenetic correlation. To illustrate the obtention of the proposed cophenetic matrix, we used two dissimilarity matrices - one obtained with the generalized squared Mahalanobis distance and the other with the Euclidean distance - between 17 garlic cultivars, based on six morphological characters. Basically, the proposal for obtaining the cophenetic matrix was to use the average distances within and between clusters, after performing the clustering. A function in R language was proposed to compute the cophenetic matrix for Tocher's method. The empirical distribution of this correlation coefficient was briefly studied. For both dissimilarity measures, the values of cophenetic correlation obtained for the Tocher's method were higher than those obtained with the hierarchical methods (Ward's algorithm and average linkage - UPGMA). Comparisons between the clustering made with the agglomerative hierarchical methods and with the Tocher's method can be performed using a criterion in common: the correlation between matrices of original and cophenetic distances.

In common with many other plasma membrane glycoproteins of eukaryotic origin, the promastigote surface protease (PSP) of the protozoan parasite Leishmania contains a glycosyl-phosphatidylinositol (GPI) membrane anchor. The GPI anchor of Leishmania major PSP was purified following proteolysis of the PSP and analyzed by two-dimensional 1H-1H NMR, compositional and methylation linkage analyses, chemical and enzymatic modifications, and amino acid sequencing. From these results, the structure of the GPI-containing peptide was found to be Asp-Gly-Gly-Asn-ethanolamine-PO4-6Man alpha 1-6Man alpha 1-4GlcN alpha 1-6myo-inositol-1-PO4-(1-alkyl-2-acyl-glycerol). The glycan structure is identical to the conserved glycan core regions of the GPI anchor of Trypanosoma brucei variant surface glycoprotein and rat brain Thy-1 antigen, supporting the notion that this portion of GPIs are highly conserved. The phosphatidylinositol moiety of the PSP anchor is unusual, containing a fully saturated, unbranched 1-O-alkyl chain (mainly C24:0) and a mixture of fully saturated unbranched 2-O-acyl chains (C12:0, C14:0, C16:0, and C18:0). This lipid composition differs significantly from those of the GPIs of T. brucei variant surface glycoprotein and mammalian erythrocyte acetylcholinesterase but is similar to that of a family of glycosylated phosphoinositides found uniquely in Leishmania.

1 Abstract Sleep is a vital necessity, yet its basic physiological function is still unknown, despite numerous studies both in healthy humans and animal models. The study of patients with sleep disorders may help uncover major biological pathways in sleep regulation and thus shed light on the actual function of sleep. Narcolepsy is a well defined but rare sleep disorder characterized by excessive daytime sleepiness and cataplexy, thought to be caused by a combination of genetic and environmental factors. The aim of this work was to identify genes or genetic variants, which contribute to the pathogenesis of sporadic and familial narcolepsy. Sporadic narcolepsy is the disorder with the strongest human leukocyte antigen (HLA) association ever reported. Since the associated HLA-DRB1 *1501-DQB1 *0602 haplotype is common in the general population (15-25%), it has been suggested that it is necessary but not sufficient for developing narcolepsy. To further define the genetic basis of narcolepsy risk, we performed a genome-wide association study (GWAS) in 562 European individuals with narcolepsy (cases) and 702 ethnically matched controls, with independent replication in 370 cases and 495 controls, all heterozygous for DRB1*1501-DQB1*0602. We found association with a protective variant near HLA-DQA2. Further analysis revealed that the identified SNP is strongly linked to DRB1*03-DQB1*02 and DRBΠ 301-DQB1*0603. Cases almost never carried a trans DRB1*1301-DQB1*0603 haplotype. This unexpected protective HLA haplotype suggests a causal involvement of the HLA region in narcolepsy susceptibility. Familial cases of narcolepsy account for 10% of all narcolepsy cases. However, due to low number of affected family members, narcolepsy families are usually not eligible for genetic linkage studies. We identified and characterized a large Spanish family with 11 affected family members representing the largest ever reported narcolepsy family. We ran a genetic linkage analysis using DNA of 11 affected and 15 unaffected family members and hereby identified a chromosomal candidate region on chromosome 6 encompassing 163 kb with a maximum multipoint LOD score of 5.02. The coding sequences of 4 genes within this haplotype block as well as 2 neighboring genes were screened for pathogenetic mutations in 2 affected and 1 healthy family members. So far no pathogenic mutation could be identified. Further in-depth sequencing of our candidate region as well as whole genome exome sequencing are underway to identify the pathogenic mutation(s) in this family and will further improve our understanding of the genetic basis of narcolepsy. 2 Résumé Le sommeil est un processus vital, dont la fonction physiologique est encore inconnue, malgré de nombreuses études chez des sujets humains sains ainsi que dans des modèles animaux. L'étude de patients souffrant de troubles du sommeil peut permettre la découverte de voies biologiques jouant un rôle majeur dans la régulation du sommeil. L'un de ces troubles, la narcolepsie, est une maladie rare mais néanmoins bien définie, caractérisée par une somnolence diurne excessive accompagnée de cataplexies. Les connaissances actuelles suggèrent qu'une combinaison de facteurs génétiques et environnementaux en est à l'origine. Le but du présent travail était d'identifier !e(s) gène(s) ou les polymorphismes constituant des facteurs de risque dans les formes sporadique et familiale de narcolepsie. La narcolepsie sporadique est la maladie possédant la plus forte association avec le complexe majeur d'histocompatibilité humain (HLA) jamais reportée. La fréquence au sein de la population générale de l'haplotype associé HLA-DRB1*1501- DQB1*0602 (15-25%) suggère que ce dernier est nécessaire, mais pas suffisant, pour (e développement de la maladie. Nous avons voulu approfondir la recherche de facteurs génétiques augmentant le risque de la narcolepsie. A cette fin, nous avons entrepris une étude d'association à l'échelle du génome (genome-wide association study, GWAS) parmi 562 sujets narcoleptiques européens (cas) et 702 individus contrôle de même origine ethnique et nous avons trouvé une association avec un variant protecteur près du gène HLA- DQA2. Ce résultat a été répliqué indépendamment dans 370 cas et 495 contrôles, tous hétérozygotes au locus DRB1*1501-DQB1*0602. Une analyse plus fine montre que le polymorphisme identifié est fortement lié aux allèles DRB1*03-DQB1*02 et DRB1*1301-DQB1*0603. Nous notons que seul un cas était porteur d'un haplotype en trans DRB1*1301-DQBr0603. La découverte de cet allele HLA protecteur suggère que la région HLA joue un rôle causal dans la susceptibilité à la narcolepsie. Dix pourcents des cas de narcolepsie sont familiaux. Cependant, le faible nombre de membres affectés rend ces familles inéligibles pour des études de liaison génétique. Nous avons identifié et caractérisé une grande famille espagnole, dont 11 membres sont atteints par la maladie, ce qui représente la plus grande famille narcoleptique rapportée jusqu'à ce jour. A partir de l'ADN de 11 membres atteints et 15 non- atteints, nous avons identifié par étude de liaison une région candidate de 163 kîlobases (kb) sur le chromosome 6, correspondant à un LOD score multipoints de 5.02. Nous avons cherché, sans succès, des mutations pathogéniques dans la séquence codante de deux gènes situés à l'intérieur de ce segment, ainsi que 4 gènes adjacents. Un séquençage plus approfondi de la région ainsi que le séquençage des exons de tout le génome est en cours et doit s'avérer plus fructueux et révéler la ou tes mutation(s) pathogénique(s) dans cette famille, ce qui contribuerait à une meilleure compréhension des causes génétiques de la narcolepsie. 3 Résumé pour un large public Le sommeil est une nécessité vitale, dont le rôle physiologique exact reste inconnu malgré de nombreuses études sur des sujets humains sains ainsi que sur des modèles animaux. C'est pourquoi les troubles du sommeil intéressent les chercheurs, car l'élucidation des mécanismes responsables peut permettre de mieux comprendre le fonctionnement du sommeil normal. La narcolepsie est une maladie du sommeil caractérisée par une somnolence diurne excessive. Les personnes atteintes peuvent s'endormir involontairement à tout moment de la journée, et souffrent également de pertes du tonus musculaire (cataplexie) lors de fortes émotions, par exemple un fou rire. La narcolepsie est une maladie rare, apparaissant dans 1 personne sur 2000. Les connaissances actuelles suggèrent qu'une combinaison de facteurs génétiques et environnementaux en est à l'origine. Nous avons voulu identifier les facteurs génétiques influençant le déclenchement de la maladie, d'abord dans sa forme sporadique, puis dans une famille comptant de nombreux membres atteints. En comparant les variations génétiques de près de 1000 sujets narcoleptiques européens avec ceux de 1200 individus sains, nous avons trouvé chez 30% de ces derniers un variant protecteur, qui diminue de 50 fois le risque de développer la maladie, ce qui constitue le plus puissant facteur génétique protecteur décrit à ce jour. Nous avons ensuite étudié une grande famille espagnole comptant une trentaine de membres, dont 11 sont atteints de narcolepsie. De nouveau, nous avons comparé les variations génétiques des membres atteints avec ceux des membres sains. Nous avons ainsi pu identifier une région dans le génome où se trouverait le(s) gène(s) impliqué(s) dans la maladie dans cette famille, mais n'avons pas encore trouvé le(s) variant(s) exact(s). Une étude plus approfondie devrait permettre de P(les) identifier et ainsi contribuer à l'élucidation des mécanismes menant au développement de la narcolepsie.

Line converters have become an attractive AC/DC power conversion solution in industrial applications. Line converters are based on controllable semiconductor switches, typically insulated gate bipolar transistors. Compared to the traditional diode bridge-based power converters line converters have many advantageous characteristics, including bidirectional power flow, controllable de-link voltage and power factor and sinusoidal line current. This thesis considers the control of the lineconverter and its application to power quality improving. The line converter control system studied is based on the virtual flux linkage orientation and the direct torque control (DTC) principle. A new DTC-based current control scheme is introduced and analyzed. The overmodulation characteristics of the DTC converter are considered and an analytical equation for the maximum modulation index is derived. The integration of the active filtering features to the line converter isconsidered. Three different active filtering methods are implemented. A frequency-domain method, which is based on selective harmonic sequence elimination, anda time-domain method, which is effective in a wider frequency band, are used inharmonic current compensation. Also, a voltage feedback active filtering method, which mitigates harmonic sequences of the grid voltage, is implemented. The frequency-domain and the voltage feedback active filtering control systems are analyzed and controllers are designed. The designs are verified with practical measurements. The performance and the characteristics of the implemented active filtering methods are compared and the effect of the L- and the LCL-type line filteris discussed. The importance of the correct grid impedance estimate in the voltage feedback active filter control system is discussed and a new measurement-based method to obtain it is proposed. Also, a power conditioning system (PCS) application of the line converter is considered. A new method for correcting the voltage unbalance of the PCS-fed island network is proposed and experimentally validated.

Macrophage migration inhibitory factor (MIF) is an abundantly expressed proinflammatory cytokine playing a critical role in innate immunity and sepsis and other inflammatory diseases. We examined whether functional MIF gene polymorphisms (-794 CATT(5-8) microsatellite and -173 G/C SNP) were associated with the occurrence and outcome of meningococcal disease in children. The CATT(5) allele was associated with the probability of death predicted by the Pediatric Index of Mortality 2 (P=0.001), which increased in correlation with the CATT(5) copy number (P=0.04). The CATT(5) allele, but not the -173 G/C alleles, was also associated with the actual mortality from meningoccal sepsis [OR 2.72 (1.2-6.4), P=0.02]. A family-based association test (i.e., transmission disequilibrium test) performed in 240 trios with 1 afflicted offspring indicated that CATT(5) was a protective allele (P=0.02) for the occurrence of meningococcal disease. At baseline and after stimulation with Neisseria meningitidis in THP-1 monocytic cells or in a whole-blood assay, CATT(5) was found to be a low-expression MIF allele (P=0.005 and P=0.04 for transcriptional activity; P=0.09 and P=0.09 for MIF production). Taken together, these data suggest that polymorphisms of the MIF gene affecting MIF expression are associated with the occurrence, severity, and outcome of meningococcal disease in children.

False identity documents represent a serious threat through their production and use in organized crime and by terrorist organizations. The present-day fight against this criminal problem and threats to national security does not appropriately address the organized nature of this criminal activity, treating each fraudulent document on its own during investigation and the judicial process, which causes linkage blindness and restrains the analysis capacity. Given the drawbacks of this case-by-case approach, this article proposes an original model in which false identity documents are used to inform a systematic forensic intelligence process. The process aims to detect links, patterns, and tendencies among false identity documents in order to support strategic and tactical decision making, thus sustaining a proactive intelligence-led approach to fighting identity document fraud and the associated organized criminality. This article formalizes both the model and the process, using practical applications to illustrate its powerful capabilities. This model has a general application and can be transposed to other fields of forensic science facing similar difficulties.

Kilpailun kiristyminen on pakottanut ydinvoimalaitoksia parantamaan tehokkuuttaan etsimällä uusia toimintatapoja. Heräte työn teettämiseen syntyi tutkimuksen case-kohteen Loviisan voimalaitoksen kiinnostuksesta Balanced Scorecard (BSC) -johtamisjärjestelmää,sen käyttöönoton mahdollisia vaikutuksia sekä BSC:n ja prosessiajattelun yhdistämistä kohtaan. Tutkimuksen tavoitteena on rakentaa Loviisan voima-laitoksen tuotannon ylläpitoprosessille BSC-mittaristo. Tämä edellyttää selvitystä siitä, mitä erityispiirteitä ydinvoimalaitoksiin liittyy strategisen suorituskyvyn mittaamisen kohteena. Lisäksi tavoitteena on selvittää, mikä tulisi prosessikohtaisten tuloskorttien rooli olla Loviisan voimalaitoksen BSC-järjestelmässä. Tavoitteenaon myös muodostaa suositus toimintamallista, jolla BSC voitaisiin ottaa käyttöön Loviisan voimalaitoksella, sekä selvittää, mitä vaikutuksia käyttöönotolla voiolla. Ydinvoimalaitoksen erityispiirteitä ovat muutokset toimintaympäristössä, viranomais-valvonta, toiminnan pitkäjänteisyys, laaja osaamis- ja tietotarve sekä turvallisuuden ja tiettyjen sidosryhmäsuhteiden merkityksen korostuminen. Johtuen erityispiirteistä Kaplanin ja Nortonin alkuperäistä asiakasnäkökulmaa muutetaan kattamaan sidosryhmät laajemmin. Tuotannon ylläpitoprosessin tuloskortissa vähiten painottuva näkö-kulma on henkilöstön ja uudistumisen näkökulma. Osa laitostason kriittisistä menestystekijöistä todetaan prosessin kannalta epäolennaisiksi. Prosessikohtaiset tuloskortit osoittautuvat vaikeasti hyödynnettäviksi linjaorganisaation ohjaamisessa. Strategiakartta todetaan hyväksi työvälineeksi BSC:nlaadinnassa. Toivasen projektimalli arvioidaan sopivaksi välineeksi mahdolliseen BSC:n käyttöönottoon Loviisan voimalaitoksella. Henkilöstön rooli ja erityispiirteiden vaikutukset tulee kuitenkin tarkistaa ennen mallin käyttöä. BSC-järjestelmän käyttöönoton arvioidaan selkeyttävän voimalaitoksen mittaristokokonaisuutta sekä parantavan syy-seuraussuhteiden hahmottamista ja alempien tasojen tavoitteiden kytkentää laitostason tavoitteisiin.

A variant upstream of human leukocyte antigen C (HLA-C) shows the most significant genome-wide effect on HIV control in European Americans and is also associated with the level of HLA-C expression. We characterized the differential cell surface expression levels of all common HLA-C allotypes and tested directly for effects of HLA-C expression on outcomes of HIV infection in 5243 individuals. Increasing HLA-C expression was associated with protection against multiple outcomes independently of individual HLA allelic effects in both African and European Americans, regardless of their distinct HLA-C frequencies and linkage relationships with HLA-B and HLA-A. Higher HLA-C expression was correlated with increased likelihood of cytotoxic T lymphocyte responses and frequency of viral escape mutation. In contrast, high HLA-C expression had a deleterious effect in Crohn's disease, suggesting a broader influence of HLA expression levels in human disease.

The production and use of false identity and travel documents in organized crime represent a serious and evolving threat. However, a case-by-case perspective, thus suffering from linkage blindness and a limited analysis capacity, essentially drives the present-day fight against this criminal problem. To assist in overcoming these limitations, a process model was developed using a forensic perspective. It guides the systematic analysis and management of seized false documents to generate forensic intelligence that supports strategic and tactical decision-making in an intelligence-led policing approach. The model is articulated on a three-level architecture that aims to assist in detecting and following-up on general trends, production methods and links between cases or series. Using analyses of a large dataset of counterfeit and forged identity and travel documents, it is possible to illustrate the model, its three levels and their contribution. Examples will point out how the proposed approach assists in detecting emerging trends, in evaluating the black market's degree of structure, in uncovering criminal networks, in monitoring the quality of false documents, and in identifying their weaknesses to orient the conception of more secured travel and identity documents. The process model proposed is thought to have a general application in forensic science and can readily be transposed to other fields of study.

The integration of information which can be gained from accessory [i.e. age (t)] and rock-forming minerals [i.e. temperature (T) and pressure (P)] requires a more profound understanding of the equilibration kinetics during metamorphic processes. This paper presents an approach comparing conventional P-T estimate from equilibrated assemblages of rock-forming minerals with temperature data derived from yttrium-garnet-monazite (YGM) and yttrium-garnet-xenotime (YGX) geothermometry. Such a comparison provides an initial indication on differences between equilibration of major and trace elements. Regarding this purpose, two migmatites, two polycyclic and one monocyclic gneiss from the Central Alps (Switzerland, northern Italy) were investigated. While the polycyclic samples exhibit trace-element equilibration between monazite and garnet grains assigned to the same metamorphic event, there are relics of monazite and garnet obviously surviving independent of their textural position. These observations suggest that surface processes dominate transport processes during equilibration of those samples. The monocyclic gneiss, on the contrary, displays rare isolated monazite with equilibration of all elements, despite comparably large transport distances. With a nearly linear crystal-size distribution of the garnet grain population, growth kinetics, related to the major elements, were likely surface-controlled in this sample. In contrast to these completely equilibrated examples, the migmatites indicate disequilibrium between garnet and monazite with a change in REE patterns on garnet transects. The cause for this disequilibrium may be related to a potential disequilibrium initiated by a changing bulk chemistry during melt segregation. While migmatite environments are expected to support high transport rates (i.e. high temperatures and melt presence), the evolution of equilibration in migmatites is additionaly related to change in chemistry. As a key finding, surface-controlled equilibration kinetics seem to dominate transport-controlled processes in the investigated samples. This may be decisive information towards the understanding of age data derived from monazite.

Diplomityön tavoitteena on tutkia mitä uusia tiedonhallinnallisia ongelmia ilmenee, kun massaräätälöidyn tuotteen tuotetieto hallitaan läpi tuotteen elinkaaren, sekä miten nämä ongelmat voitaisiin ratkaista. Ongelmat ja haasteet kerätään kirjallisuuslähteistä ja massaräätälöintiprosessi yhdistetään PLM-vaiheisiin. Ratkaisua tutkitaan testaamalla kuinka standardit STEP ja PLCS sekä standardeja tukeva PLM järjestelmä voisivat tukea massaräätälöidyn tuotteen elinkaaren tiedonhallintaa. MC tuotteiden ongelmia ovat tuoterakenteen monimutkaisuus, jäljitettävyys ja muutosten hallinta läpi elinkaaren. STEP ja PLCS pystyvät kummatkin tahollaan tukemaan tiedonhallintaa. MC-tuotteen geneerinen tuoterakenne on kuitenkin manuaalisesti liittettävä elinkaaritiedon tukemiseen. PLM-järjestelmä pystyy tukemaan MC-tuotteiden elinkaarta, mutta koska toiminto ei ole järjestelmään sisäänrakennettuna, MC-tuotteiden tukemisen parantamisessa on edelleen haasteita.

In the cerebral cortex, the activity levels of neuronal populations are continuously fluctuating. When neuronal activity, as measured using functional MRI (fMRI), is temporally coherent across 2 populations, those populations are said to be functionally connected. Functional connectivity has previously been shown to correlate with structural (anatomical) connectivity patterns at an aggregate level. In the present study we investigate, with the aid of computational modeling, whether systems-level properties of functional networks-including their spatial statistics and their persistence across time-can be accounted for by properties of the underlying anatomical network. We measured resting state functional connectivity (using fMRI) and structural connectivity (using diffusion spectrum imaging tractography) in the same individuals at high resolution. Structural connectivity then provided the couplings for a model of macroscopic cortical dynamics. In both model and data, we observed (i) that strong functional connections commonly exist between regions with no direct structural connection, rendering the inference of structural connectivity from functional connectivity impractical; (ii) that indirect connections and interregional distance accounted for some of the variance in functional connectivity that was unexplained by direct structural connectivity; and (iii) that resting-state functional connectivity exhibits variability within and across both scanning sessions and model runs. These empirical and modeling results demonstrate that although resting state functional connectivity is variable and is frequently present between regions without direct structural linkage, its strength, persistence, and spatial statistics are nevertheless constrained by the large-scale anatomical structure of the human cerebral cortex.
