The paper discusses the shrimp culture practices of Thailand, the top producing country of cultured shrimp. These shrimp culture techniques include the use of reservoirs for better waste management, utilization of undiluted seawater for culture, low-cost pumping systems, measures to neutralize acid sulfate soils, proper treatment of pond bottom, and solutions to disease problems.

The purpose of this study, Evaluation the effect of Rosmarinus officinalis and Thymus vulgaris extracts on the stability of poly unsaturated fatty acids in frozen Silver carp minced. Treatments include: Treatment 1 - Control: frozen meat packaged in conventional Treatment 2: Frozen Silver carp minced+Thyme 300 mg/kg in normal packaging Treatment 3: Frozen Silver carp minced+Rosemary 200 mg/kg in normal packaging Treatment 4: Frozen Silver carp minced+Rosemary compound (100 mg/kg) and Thyme (100 mg/kg) in normal packaging After rapid freezing of samples in the spiral freezer by individual quick freezing method, to maintain the cold temperature (-18) °C were transferred. Sampling and measurements to determine the fatty acid profile of the zero phase beginning in the first month and then every ten days, and 15 days in the second month of the third month after the monthly test. Identifying, defining and measuring the fatty acid profile by gas chromatography was performed. In this study, levels of both saturated and unsaturated fatty acids in three experimental and one control were identified as follows: A: saturated fatty acids: Meristic C14: 0/Palmitic C16: 0/Hepta decaenoic C17: 0/Stearic C18: 0/Arashidic C20: 0/B:Mono unsaturated fatty acids: palmitoleic C16: 1-W7/Oleic C18: 1-W9/Gadoleic C20: 1-W9 C:Poly unsaturated fatty acids: Linoleic C18: 2-W6/α-Linolenic C18: 3-W3 D:High unsaturated fatty acids: Arachidonic C20: 4-W6 Eicosapentaenoic acid C20: 5-EPA/W3 Docosahexaenoic C22: 6-DHA/W3 Results of this study was to determine, Thyme and rosemary extracts containing silver carp minced stored in freezing conditions, Stability of different types of fatty acids, monounsaturated fatty acids, poly-unsaturated fatty acids, omega-3 and omega-6 fatty acids are. So that none of the fatty acids measured were not significant 100% increase or decrease, While changes in the fatty acid oxidation during storage time is minimized. The results obtained from the fatty acid profiles and indicators of their and statistical tests show that treatment with rosemary extract More stable during storage (-18) ° C In comparison with the control and other treatments are shown; And at relatively low compared to other treatments and control samples oleic acid and linoleic acid, palmitic more. The objective of this study was to determine the effect of dietary vitamins A, D-3, E, and C on the gonad development, lipid peroxidation, and immune response of yearling rice field eel, Monopterus albus. A 6-wk feeding trial was designed according to an L-16(4(5)) orthogonal design, in which four vitamins, each at four supplementation levels, were arranged. Sixteen diets were mixed with the different vitamin levels and randomly assigned to 16 groups of fish. Increasing dietary vitamin E supplementation level significantly (P <= 0.05) increased the gonadosomatic index and lowered the serum content of malondialdehyde of rice field eel. Increasing dietary vitamin A and C levels also showed similar effect, but the differences were not statistically significant. Serum immunoglobulin M content increased significantly (P <= 0.01) as dietary vitamin C supplementation levels increased. The concentrations of calcium in bones showed significant (P <= 0.05) trend with vitamin D-3 and A supplementation levels, but the bone phosphorus content was not affected by the dietary vitamin levels.

A kind of optical pH sensor was demonstrated that is based on a pH-sensitive fluorescence dye-doped (eosin) cellulose acetate (CA) thin-film modified microstructured polymer optical fiber (MPOF). It was obtained by directly inhaling an eosin-CA-acetic acid mixed solution into array holes in a MPOF and then removing the solvent (acetic acid). The sensing film showed different fluorescence intensities to different pH solutions in a pH range of 2.5-4.5. Furthermore, the pH response range could be tailored through doping a surfactant, hexadecyl trimethyl ammonium bromide (CTAB), in the sensing film. (c) 2007 Optical Society of America.

本文用中空纤维膜基萃取法研究了铈(IV)、钍和 RE(III)的膜基萃取;铈(IV)与 RE(III)、钍和 RE(III)的膜基萃取分离;铈(IV)与 RE(III)的界面化学反应动力学;膜基萃取中的流体动力学;萃取操作中乳化发生的机理;测定了中空纤维膜的孔率和孔径。在铈(IV)、钍和 RE(III)的膜基萃取过程中,研究了水相流量、油相流量、原料液中硫酸浓度、萃取剂 N1923浓度和水相溶质浓度对基于水相总传质系数的影响。钍的总传质系数受水相流量影响较大,不受油相流量影响,从而提出水相扩散层控制的膜基萃取传质机理;铈(IV)的传质系数受水相流量影响,油相流量影响较小,主要为水相扩散层控制的传质机理;RE(III)的传质系数受水相和油相的影响都比较小,为膜内传质过程的传质机理。酸浓度对钍和铈(IV)传质系数影响较小,是由于酸浓度影响分配系数,而对于水相扩散控制的传质过程,水相分传质系数与分配系数无关;酸浓度对 RE(III)的影响,是由于膜内传质阻力与分配系数在关;N1923 对 RE(III)萃取传质系数的影响进一步证明膜内过程控制的传质机理,同时对数曲线的斜率大于1 也说明界面反应为一复杂过程;水相溶质浓度不影响基于水相的总传质系数,进一步证实了传质速度与初始浓度无关,也从一个侧面反应了膜基萃取实验有较好的重复性。实验研究的结果,对铈(IV)的膜基萃取选择油相组成为 10%N1923+4%异辛醇+正庚烷(或煤油),水相酸浓度为 1~2mol/L,水相和油相流量可以在较大的范围内选择;钍的膜基萃取条件相似,只是油组成中萃取剂浓度为 1%N1923。在流体动力学部分,通过作用力分析,利用能量守恒原理,推导出了膜基萃取操作中的雷诺数,建立了层流流动模型;腔内外流体流速的径向分布模型和平均流速的计算式;腔内外压力沿管长度分布的数学模型,并得到了两相流体的压力差计算式。流体的动力学分析对于研究乳化发生的机理、传质的数学模型和扩散层厚度等皆有理论价值。通过流体动力学理论,结合膜破裂压和界面张力的实验方法研究了乳化发生的机理。水相进入有机相的乳化,对于非同级萃取,外压作用是乳化的主要原因,提出了类似于重力液滴形成的乳化模型;对于有机相进入水相的乳化,亲油膜的表面易形成油膜及界面张力形成的附加压力是乳化的基本原因,而随着有机相内溶质浓度增加界面张力降低使乳化在萃取操作的后期较易发生。膜破裂压和界面张力的研究方法使乳化的研究变得可以进行实验测定,这对于膜材的选择是很有价值的。界面反应动力学采用了上升单滴法,这种方法一般具有实验重复性较差的缺点。通过实验发现,影响重复性的因素主要是单滴形成速率的稳定性、聚结界面处油水界面位置的恒定和扩散传质的消除等。我们采用盘管式油加液管的设计,比较简单地解决了单滴形成速率稳定的问题,油水界面恒定是一个技术性问题,在实验中得到了较好的解决,通过传质时间与控制聚结界面的面积解决了消除扩散传质的难题。本论文的创新之处有如下几个方面:1.根据氟碳铈矿中钍、铈(IV)与稀土(III)分离的总目标,首次实现了伯胺 N1923对上述离子的中空纤维膜基萃取,提出了利用动力学差异的新型分离模式;2.对流量改变对传质系数的影响,所有文献报道的流量范围都没有达到流量增加使传质系数减小,而这一较高的流量揭示了油水界面随压力增加向膜内移动的事实,这对防止乳化时的压力控制是非常重要的。流量对扩散层厚度影响是对传质系数影响的主要原因,而混流的影响是次要的因素;3.铈(IV)和 RE(III)、钍和 RE(III)的分离表明,分离系数远远大于由总传质系数预计的结果,从而提出了动力学竞争萃取分离的机理;4.膜破裂压的测定方法是一个创新的设计。这种方法对于膜材选择和萃取时操作压力的控制是非常重要的。在现有文献中,都是直接在膜萃取操作时测定水相中的游离油或油相中游离水的体积的方法进行研究,这即不能研究乳化的机理,也不能测定准确的乳化时间。因此膜破裂压测定为乳化过程提供了一个新的研究方法;5.乳化机理的研究得到了较新的结论,尤其对于油相进入水相的乳化机理,较好地解释了在水相有超压存在下油相漏液的原因;6.界面反应动力学实验装置中盘管式油相加样解决了油滴形成稳定性的问题,提高了实验测定的可靠性的重复性。

在哺乳动物复杂的神经网络中,突触是信息传递的枢纽,其突触传递效能的持续性变化被称为突触可塑性(synaptic plasticity)。长时程增强(long-term potentiation,LTP)和长时程抑制(long-term depression,LTD)现象是两种经典的突触可塑性形式,被视作学习和记忆可能的物质基础,得到了广泛地关注。其中,海马CA1区谷氨酸能突触处的LTP和LTD目前研究得最为广泛。 α-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid(AMPA)受体作为介导兴奋性谷氨酸能突触基础传递的主要受体,是海马CA1区LTP和LTD正常表达的必要条件。近期的研究表明,AMPA受体通过胞吞、胞吐及侧向移动等方式在细胞膜和细胞内进行着持续地循环。因此,通过调节AMPA受体的上、下膜,进而影响突触后膜上AMPA受体的数量,便能对LTP和LTD产生影响。在本研究中,我们利用生物信息学的手段,以AMPA受体为靶点,设计出了旨在特异阻断LTP或LTD的多肽。运用离体脑片全细胞记录方式,在海马CA1区证明了干扰肽Pep-A2能够特异地阻断LTP而不影响LTD,Pep-A3能够特异地阻断LTD而不影响LTP。并初步探究了其关键的作用位点,为进一步理解LTP和LTD具体的分子机理打下了基础。成瘾作为异常的学习记忆过程,势必涉及到突触可塑性的变化。而特异性地阻断LTP和LTD,对药物成瘾效果的影响却鲜有报道(Wang YT,2007)。在另一部分工作中,我们采用穿膜肽Tat-A2和Tat-A3,在吗啡条件化位置偏爱(morphine conditioned place preference,morphine CPP)模型小鼠的测试前进行系统给药,结果发现两种干扰肽均能阻断或损伤其CPP的表达过程。这一现象,提示我们LTP和LTD在条件化位置偏爱的表达过程中都是不可或缺的,同时也为人们更好地理解成瘾过程的机理,及开发专一有效的治疗药物提供了新的思路。

Green)地上部分95%乙醇提取物中通过色谱分离得到20个化合物, 其中化合物20为新化合物。基于波谱数据它们被鉴定为(E)-阿魏酸二十烷基酯(1)、β-谷甾醇(2)、羽扇豆醇(3)、齐墩果酸(4)、7-oxo-β-sitosterol(5)、乙酰齐墩果酸(6)、(6′-O-palmitoyl)-sitosterol 3-O-β-D-glucoside(7)、rotundioic acid(8)、地榆糖甙Ⅱ(9)、27-O-(E)-对羟基肉桂酰-28-齐墩果酸(10)、27-O-(Z)-对羟基肉桂酰-28-齐墩果酸(11)、hycandinic acid ester(12)、绿原酸丁酯(13)、4,5-二咖啡酰奎尼酸丁酯(14)、4,5-dihydroxyprenyl caffeate(15)、28-O-β-D-glucopyranosyl rotundioic acid (16)、4-(6-O-caffeoyl-β-D-glucopyranosyloxy)-5-hydroxyprenyl caffeate (aohada-glycoside C, 17)、 4-β-D-glucopyranosyloxy-5-hydroxy-prenyl caffeate (aohada-glycoside A, 18)、β-胡萝卜甙(19)以及3-[O-β-D-(6-O-咖啡酰吡喃葡萄糖)]-甲基-2-烯-γ-内酯 (20)。化合物13、14、15和17有较强的α-葡萄糖甙酶抑制活性。当浓度为1 mg/ml时,它们对α-葡萄糖甙酶的抑制分别为61.5%、95.5%、72.1%、62.6%,活性高于阿卡波糖。 综述了木犀属植物化学成分及1993年以来苯丙素甙类化合物活性研究进展。 Twenty compounds were isolated from the 95% ethanol extract of the aerial parts of Osmanthus yunnanensis Fr. P. S. Green by chromatography for the first time. On the basis of spectral data, they were identified as (E)-ferulic acid eicosyl ester (1), β-sitosterol (2), lupenol (3), oleanolic acid (4), 7-oxo-β-sitosterol (5), acetyloleanolic acid (6), (6′-O-palmitoyl)-sitosterol 3-O-β-D-glucoside (7), rotundioic acid (8), ziyu glycosideⅡ (9), 3β-hydroxy-27-p-(E)-coumaroyloxy-olean-12-en-28-oic acid (10), 3β-hydroxy-27-p-(Z)-coumaroyloxyolean-12-en-28-oic acid (11), hycandinic acid ester (12), chlorogenic acid butyl ester (13), 4,5-di-O-caffeoylquinic acid butyl ester (14), 4,5-dihydroxyprenyl caffeate (15), 28-O-β-D-glucopyranosyl rotundioic acid (16), 4-(6-O-caffeoyl-β-D-glucopyranosyloxy)-5-hydroxyprenyl caffeate (aohada- glycoside C, 17), 4-β-D-glucopyranosyloxy-5-hydroxyprenyl caffeate (aohada- glycoside A, 18), β-daucosterol(19) and 3-[O-β-D-(6-O-caffeoylglucopyranosyl)]- methyl-2-en-γ-lactone (20). Compound 20 is a new one. Compounds 13, 14, 15 and 17 inhibit α-glucosidase with corresponding inhibitory rate of 61.5%, 95.5%, 72.1% and 62.6% at a concentration of 1 mg/ml, higher than acarbose. The chemical studies on Osmanthus genus and bioactivities of phenylpropanoid glycosides were summarized.

本论文由三章组成。第一章为综述,概述了植物中环烯醚萜类化合物的研究进展;第二和第三章为实验论文,分别报道了唇形科药用植物绵参和蔷薇科药用植物地榆的化学成分研究。 第一章概述了植物中环烯醚萜类化合物的研究成果,主要包括结构类型及药理活性等方面。 第二章包括两个部分。第一部分报道了藏药绵参(Eriophyton wallichii Benth)地上部分甲醇提取物的化学成分。采用正、反相硅胶柱层析等各种分离方法,从中共分离出7个化合物,有6个化合物为首次从该植物中分离得到,分别为β-谷甾醇(1),夏至草苦素(marrubiin,2),乌苏酸(3),cimigoside(4),5-deoxyantirrhinoside(5),8-表马钱子酸葡萄糖苷(8-epiloganic acid,6)和apigenin 7-(6''-p-coumaroyl)glucoside(7)。第二部分,采用高效液相色谱-质谱联用技术对绵参地上部分的甲醇提取物进行了分析,通过标准品对照紫、外光谱分析以及多级质谱分析与文献对照鉴定了8个成分,分别是:8-epiloganic acid(Ⅰ),quercitrin 3-glucoside-7-(6''-p-coumaroyl)glucoside(Ⅱ),ajugoside(I) (Ⅲ),chrysoeriol 7-O-E-p-coumaroyl-3-O-b-D-glucoside(Ⅳ),helichrysoside(Ⅴ),生物碱(Ⅵ),apigenin 2,3-dihydrogen-7-(6''-p-coumaroyl) glucoside(Ⅶ),apigenin 7-(6''-p-coumaroyl) glucoside(Ⅷ)。 第三章报道了中药地榆根部乙醇提取物正丁醇相的化学成分,通过正、反相硅胶柱层析等各种分离方法,从中分离得到8个化合物,分别为3,4¢- O-二甲基逆没食子酸(8),3,3¢,4¢-O-三甲基逆没食子酸(9)和3,4¢-O-二甲基逆没食子酸-4-O-b-D-木糖苷(10),19a-羟基-3-O-(a-L-阿拉伯糖)乌苏酸-28-O-b-D-葡萄糖苷(11), 3b-[(a-L-arabinopyranosyl)oxy]-urs-11,13(18)-dien-28-oic acid b-D- glucopyranosyl ester(13),3-O-a-L-arabinopyranosyl-urs-12,18(19)-dien-28-oic acid b-D-glucopyranosyl ester(14),儿茶素(15),还有一种可能是皂苷11的工作产物(12)。 This dissertation consisted of three chapters. The first chapter elaborated the progress of iridoids occurring in plants. The later two chapters respectively elaborated the chemical constituents of Eriophyton wallichii Benth. and Sanguisorba officinalis L. The first chapter is a review of the research progress of iridoids occurring in plants, which includes their structure and pharmacology. The second chapter consisted of two parts. The first part is about the chemical constituents of methanol extraction from the aerial parts of Eriophyton wallichii Benth. Seven compounds were isolated and identified. Among them, the compounds of marrubiin, ursolic acid, cimigoside, 5-deoxyantirrhinoside, 8-epiloganic acid,apigenin 7-(6''-p-coumaroyl)glucoside were firstly reported in this plant. A HPLC-MSn method was developed for rapid identification of major compounds of Eriophyton wallichii. A total of 8 peaks in the chromatograms were unequivocally determined (peaks 1, 8) or tentatively identified (peaks 2-7) based on the detailed UV and tandem mass spectra analysis. Seven components were identified as 8-epiloganic acid(Ⅰ),Quercitrin 3-glucoside-7-(6''-p-coumaroyl)glucoside(Ⅱ),ajugoside(I)(Ⅲ),Chrysoeriol 7-O-E-p-coumaroyl-3-O-b-D-glucoside(Ⅳ),helichrysoside(Ⅴ),apigenin 2,3-dihydrogen-7-(6''-p-coumaroyl) glucoside(Ⅵ),apigenin 7-(6''-p-coumaroyl) glucoside(Ⅶ)。 The third chapter elaborated the chemical constituents of methanol extraction from Sanguisorba officinalis L, eight compounds were isolated from this plant by repeat column chromatography over silica gel. These compounds were identified as 3,4′-O-dimethylellagic acid, 3,3′,4′-O-trimethylellagic acid, 3,4′-O-dimethylellagic acid-4-O-b-D-xyloside, 3b-O-a-L-arabinopyranosyl-19a- hydroxyl-urs-12-en-28-oic acid 28-b-D-glucopyranoside, 3b-[(a-L-arabinopyranosyl)oxy]-urs-11,13(18)-dien- 28-oic acid b-D-glucopyranosyl ester,3-O-a-L–arabinopyranosyl-urs-12,18(19) -dien-28-oic acid b-D-glucopyranosyl ester, catechin.

本文对禄春安息香(Styrax macranthus)种子和攀援孔药花(Porandra scandens)全草的化学成分进行了研究,共获得30个化合物,其中2个为新化合物。 从禄春安息香种子95%乙醇提取物中分离并鉴定了12个化合物,其中2个新化合物鉴定为3-[7-methoxy-2-(3,4-methylenedioxy phenyl) benzofuran-5-yl] propyl 3-[7-methoxy-2-(3,4-methylenedioxyphenyl)benzofuran-5-yl] propanoate (1) 和去甲氧基-egonol-龙胆双糖甙 (2);已知化合物分别为2-(3,4-二氧亚甲基苯基)-5-甲酰基-7-甲氧基-苯并呋喃 (3)、egonol (4)、去甲氧基-egonol (5)、去甲基-egonol (6)、egonol-葡萄糖甙 (7)、egonol-龙胆双糖甙 (8)、egonol-龙胆三糖甙 (9)、豆甾醇 (10)、二十四烷酸 1-甘油酯 (11) 和胡萝卜甙 (12)。生物活性测试发现,化合物2具有促进雌激素E2合成的作用。 从攀援孔药花全草95%乙醇提取物中分离并鉴定了19个化合物:(2S,3S,4R)-2-[(2R)-2-羟基-二十一烷酰基氨基]-二十一烷-1,3,4-三醇 (13)、(2S,3S,4R)–2–二十四烷酰基氨基-十八烷-1,3,4-三醇 (14)、胡萝卜甙 (12)、β-谷甾醇 (15)、(20S,22E,24R)-5α,8α-表二氧-麦角甾-6,22-二烯-3β-醇 (16)、6β-羟基-豆甾-4-烯-3-酮 (17)、十六烷酸 1-甘油酯 (18)、桦木酸 (19)、大黄素 (20)、二十二烷酸 1-甘油酯 (21)、对羟基苯甲醛 (22)、十七烷酸 1-甘油酯 (23)、金色酰胺醇乙酸酯(24)、十九烷酸 1-甘油酯 (25)、棕榈酸 (26)、(E)-p-香豆酸 (27)、(22E,24S)-24-麦角甾醇-7,22-二烯-3β,5α,6β-三醇 (28)、2-去氧-β-蜕皮激素 (29)和auranamide (30)。 综述了近十年来发现的2-芳基苯并呋喃类新木脂素的结构特征、来源、生物活性和化学全合成。 Phytochemical investigation on the seeds of Styrax macranthus and the whole plants of Porandra scandens led to the isolation of thirty compounds, two of which were new ones. Two new 2-aryl benzofuran derivatives, 3-[7-methoxy-2-(3,4-methylenedioxy phenyl) benzofuran-5-yl]propyl 3-[7-methoxy-2-(3,4-methylenedioxyphenyl)benzo furan-5-yl]propanoate (1) and demethoxy egonol gentiobioside (2), were isolated from the 95% aqueous ethanolic extract of the seeds of Styrax macranthus, together with 7-methoxy-2-(3,4-methylenedioxyphenyl) benzofuran-5-carbaldehyde (3), egonol (4), demethoxy egonol (5), demethyl egonol (6), egonol glucoside (7), egonol gentiobioside (8), egonol gentiotrioside (9), stigmasterol (10), 2,3-dihydroxypropyl tetracosoate (11), and daucosterol (12). In vitro test, compound 2 promote the synthesis of estrogen E2. Nineteen compounds were isolated from the 95% aqueous ethanolic extract of the whole plant of Porandra scandens for the first time. Their structures were identified as (2S,3S,4R)-2-[(2R)-2-hydroxy-heneicosanoylamino]-1,3,4- heneicosanetriol (13), (2S,3S,4R)-2-tetracosanoylamino-1,3,4-octadecanetriol (14), daucosterol (15), β-sitosterol (12), (20S,22E,24R)-5α,8α-epidioxy-ergosta-6,22-diene- 3β-ol (16), 6β-hydroxylstigmast-4-en-3-one (17), 1-glycerol-1-hexadecoate (18), betulinic acid (19), emodin (20), 1-glycerol-1-docosoate (21), p-hydroxybenzaldehyde (22), 1-glycerol-1-heptadecoate (23), aurantiamide acetate (24), 1-glycerol-1- nonadecoate (25), palmatic acid (26), (E)-p-coumaric acid (27), (22E,24S)- 24-metbylcbolesta-7,22-diene-3β,5α,6β-triol (28), 2-deoxycrustecdysone (29), and auranamide (30). The characteristic, natural resource, bioactivity, and the total synthesis of 2-aryl benzofurans were reviewed.

钮子瓜(Zehneria maysorensis Arn.)是一种常用的中草药,其性味苦、凉,主要功效为清热利湿、散风止痛,主治膀胱炎、头痛。体外活性筛选实验表明,袋花忍冬(Lonicera saccata Rehd.)95%乙醇提取物的乙酸乙酯部分对血管紧张素转化酶显示较强的抑制活性。为明确钮子瓜的药用物质基础和袋花忍冬中具有ACE抑制活性的成分,首次对两个植物的成分进行了研究。 1. 从钮子瓜95%乙醇提取物中主要通过色谱方法首次分离了14个化合物,通过波谱方法鉴定为(2S,3S,4R,10E)-2-[(2R)-2-羟基二十四烷酰基氨基]-10-十八烷-1,3,4-三醇(1)、(2S,3S,4R)-2-二十四烷酰基氨基-十八烷-1,3,4-三醇 (2)、胡萝卜苷(3)、swertish (4)、苯甲酸(5)、水杨酸(6)、loliolide (7)、胸腺嘧啶(8)、尿嘧啶(9)、(23Z)-9,19-环阿尔廷-23-烯-3β,25-二醇(10)、(20S,22E,24R)-5α,8α-表二氧-麦角甾-6,22-二烯-3β-醇(11)、十六烷酸 1-甘油酯(12)、大豆脑苷Ⅰ(13)和(22E,24S)-24-甲基-5α-胆甾-7,22-二烯-3β,5α,6β-三醇(14)。其中化合物4为一黄酮碳苷,具有旋转异构现象,有止痛作用;化合物6具有抗炎、镇痛、减热的活性,它们可能是钮子瓜药用物质基础的一部分。 2. 从袋花忍冬95%乙醇提取物中首次分离并鉴定了16个已知化合物:胡萝卜苷(3)、(20S,22E,24R)-5α,8α-表二氧-麦角甾-6,22-二烯-3β-醇(11)、十六烷酸 1-甘油酯(12)、E-p-coumaryl behenate (15)、谷甾醇(16)、2,6-dihydroxyhumula-3(12), 7(13),9(E)-triene (17)、环阿尔廷-25-烯-3β,24ξ-二醇 (18)、二十四烷酸 (19)、2,4-二羟基-3,6-二甲基苯甲酸甲酯 (20)、乌苏酸 (21)、柚皮素 (22)、木犀草素 (23)、柏双黄酮(24)咖啡酸 (25)、洋芹素(26)和木犀草素-7-O-β-D-葡萄糖苷 (27)。其中木犀草素(23)和咖啡酸(25)含量较高,它们为抑制ACE活性的成分。 3.综述了黄酮碳苷的旋转异构现象。 Zehneria maysorensis is a folk medicine for the treatment of cystitis and headache. The ethyl acetate soluble fraction of the 95% ethanol extract of Lonicera saccata showed obvious ACE inhibitory activity in vitro. To reveal their active constitutents, they were subjected to chemically study. From the 95% ethanol extract of the whole plants of Zehneria maysroensis fourteen compounds were isolated for the first time. On the basis of spectral data and/or by comparison with authentic samples, they were characterized to be (2S,3S,4R,10E)-2-[(2R)-2-hydroxytetracosanoylamino]-10-octadecene-1,3,4-triol (1), (2S,3S,4R)-2-tetracosanoylamino-1,3,4-octadecanetriol (2), daucosterol (3), swertish (4), benzoic acid (5), salicylic acid (6), loliolide (7), thymine (8), uracil (9), (23Z)-9,19-cycloart-23-ene-3β,25-diol (10), (20S,22E,24R)-5α,8α-epidioxy-ergosta- 6,22-diene-3β-ol (11), 2,3-dihydroxypropyl hexadecoate (12), soya-cerebroside (13) and (22E,24S)-24-methyl-5α-cholesta-7,22-diene-3β,5α,6β-triol (14). Compound 4, a C-glycosylflavone, showed a very interesting rotational isomerism. Compounds 4 and 6 may be the active constituents of Zehneria maysorensis considering their sedative and anti-inflammation activity, respectively. From the whole plants of Lonicera saccata, sixteen compounds were isolated for the first time. On the basis of spectral data and/or by comparison with authentic samples, they were identified to be daucosterol (3), (20S,22E,24R)-5α,8α-epidioxy- ergosta-6,22-diene-3β-ol (11), 2,3-dihydroxypropyl hexadecoate (12), E-p-coumaryl behenate (15), β-sitosterol (16), 2,6-dihydroxyhumula-3(12),7(13),9(E)-triene (17), cycloart-25-ene-3β,24ξ-diol (18), tetracosanoic acid (19), methyl 2,4-dihydroxy- 3,6-dimethylbenzoate (20), ursolic acid (21), naringenin (22), luteolin (23), cupressuflavone (24), caffeic acid (25), apigenin (26) and luteolin-7-O-β-D- glucopyranoside (27). Luteolin (23) and caffeic acid (25) were the ACE inhibitory active constituents. Rotational isomerism for C-glycosylflavonoid was reviewed.

1-甲基-2-甲氧羰基-3, 6, 8-三羟基-7-甲氧基蒽醌是从唐菖蒲干球茎中分离到的具有环氧化酶-2选择性抑制活性的多取代蒽醌类化合物。本文试图合成该化合物,实现了其类似物的合成,同时发现了几个未见报道的反应。 1.通过Diels-Alder 反应合成了关键中间体——3-甲基-5-羟基-1, 2, 4-苯三甲酸三甲酯,1-COOMe选择性水解产物与1, 2, 3-三甲氧基苯进行分子间Friedel-Crafts反应的产物再进行分子内Friedel-Crafts反应得到了目标产物的类似物1-甲基-2-甲氧羰基-3-羟基-6,7,8-三甲氧基蒽醌(路线1)。目标产物及其它类似物的合成正在进行中。 2.以乙酰乙酸甲酯和巴豆醛为原料,经过Michael加成、分子内的Aldol反应、芳香化、选择性甲酰化和还原反应,得到关键中间体2-甲基-3-羟甲基-6-甲氧基苯甲酸甲酯及其衍生物。通过该化合物与3,4,5-三甲氧基苯甲酸甲酯进行Friedel-Crafts烷基化反应得到了多取代的二苯基甲烷衍生物,拟进一步关环合成目标化合物(路线2)。 3.发现邻甲氧基苯甲酸甲酯中酯甲基可以被正丁基锂和仲丁基锂中烷基交换生成相应的酯,反应的机理不明确。当使用叔丁基锂时,得到的是邻甲氧基苯基叔丁酮,这个方法可以用来合成芳基叔丁酮类化合物。 4.以2-苄氧基-6-甲基苯甲酸甲酯为原料进行氯甲基化反应时,以苯和二氯乙烷作溶剂,发生了苄基的迁移和芳环的偶联,分别得到2,2'-二甲基-3,3'-二甲氧羰基-4,4'-二羟基联苯和2,2'-二甲基-3,3'-二甲氧羰基-4,4'-二羟基-5,5'-二苄基联苯。这是对称联苯合成的新方法。 5.水杨酸羟基邻对位的选择性甲酰化可以分别通过水杨酸和水杨酸甲酯用HMTA/CF3COOH来实现。 6.Lewis酸催化3,4,5-三甲氧基苄醇环化成1, 2, 3, 6, 7, 8, 11, 12, 13-nonamethoxyl-10,15-dihydro-5H-trbibenzo [a, d, g] cyclononene (NDTC),产率(54%)高于已有方法(12%)。 Methyl 3,6,8-trihydroxy-7-methoxy-1-methylanthraquinone-2-carboxylate is a new COX-2 selective inhibitor isolated from Gladiolus gandavensis. Two strategies were investigated to synthesis this compound, in which some important reactions were discovered. 1. The key intermediate 5-hydroxy-3-methylbenzene-1,2,4-tricarboxylic acid 2,4-dimethyl ester was prepared via Diels-Alder reaction followed by selective hydrolysis of 1-COOMe. This compound was coupled with 1,2,3-trimethoxybenzene and the product undergo intramolecular Friedel-Crafts reaction to give methyl 3-hydroxy-5,6,7-trimethoxy-1-methylanthraquinone-2-carboxylate (1st route). The target compound and other analogues are being prepared with the same procedure. 2. The key intermediates methyl 3-hydroxymethyl-6-methoxy-2-methylbenzoate and its derivatives were prepared starting from crotonaldehyde and methyl acetoacetate via Michael addition, intramolecular aldol reaction, aromatization, formylation and reduction. The intermediates were coupled respectively with derivatives of gallic acid to give polysubstituted diphenylmethane. However, attempts to cyclize these compounds to the target compounds and analogues were not successful (2nd route). 3. In the process for ortho-lithiation of methyl 2-methoxybenzoate, the substrate converted respectively to n-butyl 2-methoxybenzoate and sec-butyl 2-methoxybenzoate when n-BuLi and sec-BuLi were used. However, tert-BuLi reacted with methyl 2-methoxybenzoate afford 2-methoxyphenyl tert-butyl ketone, which could be used to synthesize aryl tert-butyl ketones. 4. The transformtion of methyl 2-benzoxy-6-methylbenzoate to dimethyl 4,4'-dihydroxy-2,2'-dimethylbiphenyl-3,3'-dicarboxylate in benzene, and dimethyl 5,5'-dibenzyl-4,4'-dihydroxy-2,2'-dimethylbiphenyl-3,3'-dicarboxylate in 1,2-dichloroethane in the presence of ZnCl2 provides a new method for the synthesis of symmetric biphenyl. 5. The formylation of salicylic acid at C-5 and methyl 2-hydroxybenzoate at C-3 could be regioselectively realized by using HMTA/CF3COOH. 6. Racemic 1, 2, 3, 6, 7, 8, 11, 12, 13-nonamethoxyl-10, 15-dihydro-5H-trbibenzo [a, d, g] cyclononene was prepared via Lewis acids catalyzed trimerization of 3, 4, 5-trimethoxylbenzyl alcohol with yield (54%) higher than the reported procesure (12%).

八月瓜属植物五枫藤(Holboellia latifolia Wall.)和驳骨草属植物小驳骨(Gendarussa vulgaris Nees)均为药用植物, 前者化学成分研究不深入, 后者的化学成分未见报道。川西茶藨(Ribes takare D. Don)为茶藨子属植物, 没有化学成分的报道。本论文对三个植物的化学成分和活性成分进行了研究, 主要通过色谱方法分离得到了48 个化合物, 采用波谱分析或与已知标准品对照等手段鉴定了它们的结构, 其中有1 个新的原小檗碱类化合物和3 个新的联苯类化合物,发现了具有细胞毒活性和α-葡萄糖苷酶抑制活性的化合物。1、从五枫藤地上部分的95%乙醇提取物中分离得到了12 个化合物: 五加苷K (1)、hederagenin 3-O- α-L-rhamnopyranosyl-(1→2)- α-L-arabinopyranoside (2)、β-萘乙酸(3) 、3-O-α-L-rhamnopyranosyl-(1→2)-[β-D-glucopyranosyl-(1→3)]-α-L-arabinopyranosyl oleanolic acid 28-O-α-L-rhamnopyranosyl-(1→4)-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl ester (4) 、3-O- α-L-rhamnopyranosyl-(1→2)-O- β- D-glucopyranosyl-(1→2)- α-L-arabinopyranosyl oleanolic acid (5) 、3-O-( β-D-glucopyranosiduronic acid)-oleanolic acid 28-O- β-D-glucopyranoside (6)、lup-20(29)-en-3-one (7)、lupeol (8)、β-谷甾醇(9)、齐墩果酸(10)、乌苏酸(11)、β-胡萝卜苷(12)。化合物1 对Lu-06、N-04 和Bre-04 癌细胞株的GI50 分别是0.77µg/mL、1.26 µg/mL 和1.55 µg/mL, 化合物2 对N-04 癌细胞株的GI50 为2.44 µg/mL。2、从小驳骨地上部分的95%乙醇提取物中分离得到了1 个原小檗碱类新化合物13-hydroxyl gusanlung A (25), β-谷甾醇(9)、齐敦果酸(10)、β-胡萝卜苷(12)、棕榈酸(1-)甘油酯(13)、棕榈酸(14)、阿苯哒唑(15)、阿苯哒唑砜(16)、阿苯哒唑亚砜(17)、aurantiamide acetate (18)、华良姜素(19)、芫花素(20)、(-)-丁香树酯醇(21)、gusanlung B (22) 、eupteleasaponinsⅤ acetate (23)、gusanlungA (24)、刺五加苷E (26)、岩白菜素(27)、咖啡酸(28)。化合物25 对肝癌细胞株(HepG2) 的GI50 为2.08 µg/mL。3、从川西茶藨地上部分的95%乙醇提取物中分离鉴定了22 个化合物: β-谷甾醇(9) 、β- 胡萝卜苷(12) 、O-acetyloleanolic aldehyde (29),4,7,8-trimethoxy-2,3-methylenedioxydibenzofuran (30) 、3', 5-dimethoxy-3, 4-methylenedioxybiphenyl (31) 、桦木醇(32) 、6,7-dimethoxy-1-methyl-3,4-dihydroquinolin-2-one (33)、3'-hydroxy-5-methoxy-3,4-methylenedioxybiphenyl (34) 、7-hydroxy-4,8-dimethoxy-2,3-methylenedioxydibenzofuran (35)、桦木醛(36)、没食子酸(37) 、6β- 羟基-4- 烯-3- 酮- 豆甾醇(38) 、5α, 8α-epidioxy-(22E,24R)-ergosta-6, 22-dien-3β-ol (39)、verrucofortine (40)、6-methoxycalpogoniumisoflavone A (41)、2-羟基二苯甲酮(42)、桦木酸(43), 3, 5-二甲氧基苯甲酸-4-O-β-D-吡喃葡萄糖苷(44)、洋芹素(45)、刺槐素(46)、水杨酸(47)、洋芹素-5-O- β-D-葡萄糖苷(48), 化合物30、31 和35 为新的联苯化合物。化合物30的α-葡萄糖苷酶抑制率为10.2% (1.00 mg/mL); 化合物35 的抑制率为17.2% (1.00mg/mL)。4、综述了1960 年以来原小檗碱类化合物药理活性研究进展。 Plants Holboellia latifolia Wall and Gendarussa vulgaris Nees, are used as folkmedicine. Ribes takare D. Don belongs to the genus Ribes. The three plants have notbeen chemically studied in detail. Chemical and bioactive study of three plants led tothe isolation of 48 compounds by chromatography. Their structures were elucidatedon the basis of spectroscopic evidence or comparison with authentic samples. Amongthe 48 componds isolated one protoberberine alkaloid and three biphenyls are newones. Cytotoxic and α-glucosidase inhibitory compounds had been found.1. Twelve compounds were isolated from the 95% ethanol extract of the aerial partof H. latifolia Wall. They were characterized as fellow: eleutheroside K (1),hederagenin-3-O- α-L-rhamnopyranosyl-(1→2)- α-L-arabinopyranoside (2),2-naphthyl acetic acid (3),3-O-α-L-rhamnopyranosyl-(1→2)-[β-D-glucopyranosyl-(1→3)]-α-L-arabinopyranosyl oleanolic acid 28-O-α-L-rhamnopyranosyl-(1→4)-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl ester (4), 3-O- α-L-rhamnopyranosyl-(1→2)-O- β- D-glucopyranosyl-(1→2)- α-L-arabinopyranosyl oleanolic acid (5),3-O-( β-D-glucopyranosiduronic acid)-oleanolic acid-28-O- β-D-glucopyranoside (6),lup-20(29)-en-3-one (7), lupeol (8), β-sitosterol (9), oleanolic acid (10), ursolicacid (11), and β-daucosterol (12). Compound 1 showed moderate cytotoxicity againstLu-06 (GI50, 0.77 µg/mL), N-04 (GI50, 1.26 µg/mL) and Bre0-4 (GI50=1.55 µg/mL)and compound 2 showed moderate cytotoxicity against N-04 (GI50=2.44 µg/mL).2. A new protoberberine alkaloid, 13-hydroxyl gusanlung A (25), was isolated fromthe aerial part of Gendarussa vulgaris Nees, together with β-sitosterol (9), oleanolicacid (10), β-daucosterol (12), glycerol monopalmitate (13), palmific acid (14),albendazole (15), albendazole sulphone (16), albendazole sufloxide (17), aurantiamideacetate (18), kumatakenin (19), genkwanin (20), (-)-syringaresinol (21), gusanlung B(22), eupteleasaponinsⅤ acetate (23), gusanlung A (24), eleutheroside E (26),bergenin (27) and caffeic acid (28). Compound 25 showed cytotoxicity against HepG2 cells (GI50, 2.08 µg/mL).3. Phytochemical study of the Ribes takare D. Don led to the isolation of three newbiphenyls, 4,7,8-trimethoxy-2,3-methylenedioxydibenzofuran (30), 3', 5-dimethoxy-3,4-methylenedioxybiphenyl (31) and 7-hydroxy-4,8-dimethoxy-2,3-methylenedioxydibenzofuran (35), along with nineteenknown compounds, β-sitosterol (9), β-daucosterol (12), O-acetyloleanolic aldehyde(29), betulin (32), 6,7-dimethoxy-1-methyl-3,4-dihydroquinolin-2-one (33),3'-hydroxy-5-methoxy-3, 4-methylenedioxybiphenyl (34), betulinic aldehyde (36),gallic acid (37), stigmast-4-en-6β-ol-3-one (38), 5α, 8α-epidioxy-(22E, 24R)-ergosta-6,22-dien-3β-ol (39), verrucofortine (40), 6-methoxycalpogonium isoflavone A (41),2-hydroxybenzophenone (42), betulinic acid (43), 3,5-dimethoxygallic acid-4-O- β-D-glucopryranoside (44), apigenin (45), acacetin (46), salicylic acid (47) andapigenin-5-O- β-D-glucopryranoside (48). α-Glucosidase inhibitory rates ofcompound 30 and 35 were respectively 10.2% and 17.2% at a concentration of 1.00 mg/mL).4. Pharmacological activities of protoberberines were summarized.

本论文由三部分共6 章组成。第一部分报道了余甘子、细叶草乌和土荆皮等三种药用植物的化学成分研究成果;第二部分报道了细叶草乌和土荆皮中分离得到的化合物的活性测试,以及这两种植物的质谱分析;第三部分概述了土荆皮的研究现状。第一部分包括1-3 章。在第1 章、第2 章和第3 章中分别报道了余甘子(Phyllanthus emblica L.) 、细叶草乌(Aconitum richardsonianum var.pseudosessiliflorum) 和土荆皮( pseudolarix kaempferi) 的化学成分。采用正、反相硅胶柱层析等各种分离方法,从余甘子中共分离出10 个化合物,其中1 个为新化合物,另外还有2 个为首次从该植物中分离得到。细叶草乌的化学成分研究尚未见报道,我们从该植物中共分离出15 个化合物,其中6 个为二萜生物碱,9个为非生物碱成分。从土荆皮中分离得到16 个化合物,其中8 个二萜、1 个三萜和7 个其它类型化合物,其中有4 个化合物为首次在该植物中分离得到;从土荆皮挥发油中分离鉴定出了22 个化合物,占挥发油总量的90%。第二部分包括4-5 章。第4 章报道了从细叶草乌和土荆皮中分离得到的13个化合物的药理活性研究,结果显示,展花乌头宁和土荆乙酸葡萄糖苷等表现出较高的组织蛋白酶K 抑制活性;土荆乙酸葡萄糖苷表现出较高的组织蛋白酶B抑制活性;8-去乙酰滇乌碱表现出较高的蛋白质酪氨酸磷酸酶抑制活性。第5 章报道了细叶草乌和土荆皮总浸膏的质谱( ESI-MS ) 分析,研究结果表明,ESI-MS 法可以简单快速地检测这两种植物的主要成分;通过ESI-MS2 分析初步探讨了一些化合物的裂解规律,尝试质谱在其结构测定中的具体应用。第三部分为第6 章。从化学成分、药理、构效关系、主要成分的定量分析、及其合成研究等方面概述了土荆皮的研究进展。 This dissertation consists of three parts. The first part elaborate thephytochemical investigation of three medicinal plants, Phyllanthus emblica L.,Aconitum richardsonianum var. pseudosessiliflorum and pseudolarix kaempferi. Thesecond part reported the bioassay of 13 constituents from Aconitum richardsonianumvar. pseudosessiliflorum and pseudolarix kaempferi, and ESI-MS analysis of these twoplant . The third part is a review on the research progress of pseudolarix kaempferi.The first part is composed of three chapters. Chapters 1-3 focus on the isolationand identification of chemical constituents from Phyllanthus emblica L., Aconitumrichardsonianum var. pseudosessiliflorum and pseudolarix kaempferi. 10 compoundsincluding a new tannin were isolated from the fruits of Phyllanthus emblica by repeatcolumn chromatography over normal and reversed phase silica gel, 2 of them werefirstly reported in this plant. The chemical constituents of Aconitum richardsonianumvar. pseudosessiliflorum never reported before, 15 compounds including 6 diterpenealkaloids were isolated and identified from the roots of this plant. 16 compoundsincluding 8 diterpenes , 1 triterpene and 7 other compounds were isolated from the bark of pseudolarix kaempferi, among them, 4 compounds were firstly reported fromthe EtOH extracts of this plant, and 22 compounds were identified from its essentialoil, representing 90% of the total essential oil.The second part includes chapters 4 and 5. Chapter 4 reported thepharmacological activities of 13 compounds isolated from Aconitum richardsonianumvar. pseudosessiliflorum and pseudolarix kaempferi. Results demonstrated that chasmanine and pseudolaric acid B-β-D-glucoside exhibit relatively high anti-Cathepsin K activities; pseudolaric acid B-β-D-glucoside exhibits relatively highanti-Cathepsin B activity; 8-deacetyl-yunaconitine exhibits relatively high anti-PTP1Bactivity. Chapter 5 reported the ESI-MS analysis of extractions from Aconitumrichardsonianum var. pseudosessiliflorum and pseudolarix kaempferi, it was showedthat ESI-MS can be used as an useful tool in analyzing the major constituents of thesetwo plant much quick and easy, in addition, the fragmentation rules of somecompounds were discussed, in order to find some applications of ESI-MS2 method in their structure determination.The third part is a review on the research progress of pseudolarix kaempferi,including the chemical constituents, pharmacology, structure-activity relationship(SAP), quantitative analysis and synthesis of the major constituents.

本学位论文报道了西藏产三种藏族传统植物药材的化学成分研究。论文由四章组成,前三章是实验部分,分别报道了尼泊尔黄堇(Corydalis hendersonii Hemsl.)、藏波罗花(Incarvillea younghusbandii Sprague)和全缘叶绿绒蒿(Meconopsis interifolia Franch.)的化学成分研究结果。从这三种青藏高原药用植物中共分离鉴定出33 个化合物,其中1 个是新化合物。第四章概述了罂粟科紫堇属植物的化学和药理研究进展。 第一章为尼泊尔黄堇的化学成分研究。通过正、反相硅胶柱色谱等分离方法从药用植物尼泊尔黄堇的地上部分共分离纯化得到12 个化合物。运用MS、1H-NMR、13C-NMR、DEPT、HMBC、NOESY 等现代波谱学方法将它们的结构鉴定为:刺罂粟碱(1) , 普托品(2) , 新那亭(3) , 斯可任(4) , tetrahydrothalifendine (5) ,9-methyl-decumbenine C (6),tetrahydroberberrubine (7),隐品碱(8),α-别隐品碱(9),6,7-methylenedioxy-1(2H)-oxoisoquinolinone (10),6-丙酮基-5,6 -二氢血根碱(11)和β-谷甾醇(12)。其中化合物6 为新化合物,为首次发现的分子骨架上C-9 位连有甲基的苯肽异喹啉类型生物碱。另外,除化合物1 和2 外,其它9 个生物碱(3~11)均为首次从该种植物中分离得到。同时,我们还对对尼泊尔黄堇中的总生物碱进行了串联质谱分析。 第二章为藏波罗花的化学成分研究。从该药用植物的地上部分共分离得到16个化合物,通过理化常数和波谱数据鉴定为:异佛手柑内酯(1),6-甲氧基当归素(2),欧前胡素(3),花椒毒内酯(4),珊瑚菜素(5),水合氧化前胡素(6),rivulobirin A (7),齐墩果酸甲酯(8),咖啡酸甲酯(9),银桦酸(10),(D)-boschniakinic acid (11),对羟基苯甲酸(12) , tert-O-β-D-glucopyranosyl-(R)-heraclenol (13) , 5-methoxy-8-O-β-D-glucopyranosyloxypsoralen (14),前胡苷V(15)和苯乙醇-O-β-D-吡喃葡萄糖-(1→2)-O-β-D-吡喃葡萄糖苷(16)。所有以上化合物均为首次从该种植物中分离得到。另外我们还首次对藏波罗花挥发油的化学成分进行了气相色谱-质谱(GC-MS)联用分析,共鉴定出39 个挥发性成分。 第三章为全缘叶绿绒蒿化学成分的分离鉴定。从传统藏药材全缘叶绿绒蒿地上部分共分离纯化出8 个化合物。通过理化常数和波谱数据将他们的结构分别鉴定为:去甲血根碱(1),β-谷甾醇(2),3-羟基-齐墩果烷-12(13)-烯-30-酸(3),6-丙酮基-5,6-二氢血根碱(4),木犀草素(5),胡萝卜苷(6),quercetin 3-O-β-D-glucopyranosyl-(1→6)-β-D-glucopyranoside (7)和普托品(8)。其中化合物1,4 和7 为首次从该种药用植物中分离得到。 第四章为综述,总结和归纳了近年来罂粟科紫堇属植物的化学和药理研究进展。 This dissertation consists of four parts. The first, second and third parts report the studies on the chemical constituents from the medicinal plants of Corydalis hendersonii, Incarvillea younghusbandii and Meconopsis interifolia. The forth part reviews the progress of the studies on Corydalis species. The first chapter is about the isolations and identifications of alkalids from the aerial parts of C. hendersonii which is a traditional Tibetan medicine to treat febrifuge, high blood pressure and hepatitis. A new isoquinoline alkaloid, 9-methyl-decumbenine C (6), together with ten known alkaloids, stylopine (1), protopine (2), canadine (3), scoulerine (4), tetrahydrothalifendine (5), tetrahydroberberrubine (7), cryptopine (8), α-allocryptopine (9), 6,7-methylenedioxy-1(2H)-isoquinolinone (10) and 6-acetonyl-5,6-dihydrosanguinarine (11), and β-sitosterol (12) were isolated. Their structures were elucidated by spectroscopic methods. Furthermore, the total alkaloids were analyzed by ESI-MSn. The second chapter is about the isolations and identifications of chemical constituents from the aerial parts of I. younghusbandii. Sixteen compounds were isolated and purified by normal and reversed phase silica gel column chromatography. By spectral analysis, there structures were identified as isobergapten (1), sphondin (2), imperatorin (3), xanthotoxin (4), phellopterin (5), heraclenol (6), rivulobirin A (7), methyl oleanolate (8), methyl caffeate (9), grevillic acid (10), (D)-boschniakinic acid (11), 4-hydroxybenzoic acid (12), tert-O-β-D-glucopyranosyl-(R)-heraclenol (13), 5-methoxy-8-O-β-D-glucopyranosyloxypsoralen (14), decuroside Ⅴ(15), and phenylethyl-O-β-Dglucopyranosyl-(1→2)-β-D-glucopyranoside (16). All of these compounds were isolated from this plant for the first time.By the way, the chemical components of the essential oil from I. younghusbandii were analyzed by GC-MS for the first time. The third chapter is about the the isolations and identifications of the chemical constituents of M. interifolia. Eight compounds were isolated and identified as norsanguinarine (1), β-sitosterol (2), 3-hydroxyolean-12(13)-en-30-oic acid (3), 6-acetonyl-5,6-dihydrosanguinarine (4), luteolin (5), daucosterol (6), quercetin 3-O-β-D-glucopyranosyl-(1→6)-β-D-glucopyranoside (7) and protopine (8). The compounds 1, 4 and 7 were isolated from this herb for the first time. The last chapter is a review of the research progress of the studies on Corydalis species, which includes the chemical constituents in this genus and their pharmacology.

青稞,是我国藏区居民对裸大麦的称谓,它不仅是藏民的主要食粮、燃料和牲畜饲料,而且也是啤酒、医药和保健品生产的原料;青稞不仅为藏区人民的健康和经济发展做出了很大的贡献,而且对人类健康和社会经济的可持续发展都有重要的意义。青藏高原是我国及世界上青稞分布和种植面积最大的地区,资源极其丰富。虽然从经典遗传直到分子标记对我国大麦遗传多样性都有研究,但研究手段、数量仍然不够深入,对我国大麦资源遗传多样性研究的信息非常有限,不能很好地满足大麦遗传研究和育种应用的需要,尤其是对西藏栽培大麦的遗传多样性的研究还只是刚刚开始,关于栽培青稞多态性的研究报道很少。本研究采用SSR标记和蛋白质电泳两类技术,从SSR标记位点、单体醇溶蛋白、B组醇溶蛋白和淀粉粒结合蛋白(SGP)等四个方面对我国青藏高原栽培青稞的遗传多样性进行了综合评价。 SSR标记具有基因组分布广泛、数量丰富、多态性高、容易检测、共显性、结果稳定可靠、实验重现性好、操作简单、经济、易于高通量分析等许多优点,被认为是用于遗传多样性、品种鉴定、物种的系统发育、亲缘关系及起源等研究的非常有效的分子标记。本研究采用SSR标记分析了64份青藏高原栽培青稞的遗传多样性,同时评估SSR标记在我国大麦育种和品种鉴定中的应用潜力。选择了30个已知作图位点SSR标记,其中25个标记与重要性状的控制位点连锁紧密。选择的30个SSR标记,5个未得到很好的扩增产物,3个无多态性。22个多态性SSR标记位点中,每位点检测出等位基因2~15个,共检测出等位基因132个,平均每位点6.0 个。各多态位点检测出基因型为2~11种,位点HVM33的基因型最多。各多态位点的多态信息指数为0.16~0.91, 平均为0.65。根据PIC值选择了13个SSR标记用于我国青藏高原栽培青稞基因型鉴定,这些标记的PIC值为0.6以上。结合PIC值和基因型差异,选择了8个多态信息含量高的SSR标记,构建了高效指纹图谱,此图谱能把64份材料完全区分。 贮藏蛋白电泳分析是研究相关编码蛋白基因多态性的非常有效的方法。大麦单体蛋白与小麦醇溶蛋白相对应,具有丰富的多态性,可用于大麦遗传多样性、品种鉴定和群体进化等研究。本研究通过A-PAGE电泳技术研究了84份青藏高原栽培青稞的单体醇溶蛋白多态性。大麦单体醇溶蛋白图谱与小麦醇溶蛋白电泳图谱类似,所分离的蛋白清晰地分为ω-,γ-,β-和α-四个部分。青藏高原栽培青稞单体醇溶蛋白具有丰富的多态性,84份青稞材料中存在43条不同的蛋白带,75种组合带谱;其中67种为单一材料所独有,另8种则分别包含了2-3份材料。每份材料中拥有醇溶蛋白带为6-16条,含有6-10条单体醇溶蛋白带材料较多。西藏和四川材料群体单体醇溶蛋白多态性不同,具有区域特异性。西藏材料中发现了40条不同蛋白带,3条特异带,46 种蛋白组合;四川材料中出现了40种不同蛋白带,26种条带组合, 3条特异带。基于单体蛋白多态性的聚类与材料的来源有一定的相关性。A-PAGE单体蛋白具有丰富的多态性,可作为遗传研究和品种鉴定的标记。 大麦醇溶蛋白(hordein)是大麦籽粒的主要贮藏蛋白,与大麦的营养品质和加工品质密切相关,而且具有丰富的多态性,广泛用于品种鉴定、种质筛选、遗传多样性和亲缘关系研究。B组醇溶蛋白是主要的醇溶蛋白组份,约占总醇溶蛋白的80%,而且具有丰富的多态性。本研究采用SDS-PAGE分析了72份青藏高原栽培青稞B组醇溶蛋白的遗传多样性。青藏高原栽培青稞B组醇溶蛋白具有丰富的多态性,72份青稞材料中存在15种蛋白带,30种组合带谱,其中15种为单一材料所独有,另15种则分别包含了2-10份材料。每份材料中B组醇溶蛋白条带数为4-8条,含5、6条的材料较常见。不同来源的群体材料间B组醇溶蛋白组成存在差异,西藏青稞含有26种蛋白组合带谱,其中有19种特异带谱;四川群体中共发现11种蛋白组合带型,其中有4种特有带谱。两群体中都存在稀有条带。聚类分析将材料分成三组,材料聚类与材料来源地没有明显的相关性。 淀粉粒蛋白(Starch granule proteins, SGPs)是一类与淀粉粒结合的微量蛋白,一些淀粉粒蛋白具有淀粉生化合成中主要的酶蛋白功能,其变异会影响淀粉含量和特性,从而影响淀粉的应用。关于我国大麦淀粉粒组成研究还未见报道。本实验首次开创了我国大麦淀粉粒结合蛋白的研究工作。采用SDS-PAGE电泳技术研究了青藏高原栽培青稞的SGP组成,并分析了不同SGP组合间淀粉含量的差异,初步探索了所分离的SGP蛋白与淀粉合成的关系。66份青稞材料中分离了10种主要的SGP,其表观分子量为40-100KD,低于60KD的SGP带有7条,共有16种组合带谱;各SGP蛋白和组合带谱出现的频率存在差异,青藏高原青稞的SGP组成存在多态性。西藏青稞和四川青稞的SGP组成有很大差异,SGP组成具有地域差异性,西藏青稞含有12种蛋白组合带谱,其中有9种特异带谱;四川群体中共发现7种蛋白组合带型,其中有4种特有带谱;两群体中仅有3种共同的蛋白组合带谱。SGP蛋白特性将66份青稞分为三组, 即Ⅰ、Ⅱ、Ⅲ,材料聚类与材料来源具有一定的相关性。不同组合带谱材料间淀粉含量差异显著性检验结果显示,不同带谱间材料的总淀粉含量、直链淀粉含量和支链淀粉含量有差异,带谱2(SGP1+3+7+9+10)和8(SGP1+2+4+6+8)的总淀粉含量及支链淀粉含量显著大于组合带谱3(SGP1+3+7+10)的总淀粉含量。组合带谱7(SGP1+2+6+8)的直链淀粉含量显著低于带谱11(SGP1+5+8)的直链淀粉。带谱SGP2、3、4、5、6、7、8、9、10可能参与淀粉合成,SGP9可能与高支链淀粉的合成相关。 SSR标记位点、单体醇溶蛋白、B组醇溶蛋白、淀粉结合蛋白等四个方面的研究结果表明青藏高原SSR标记多态性、单体醇溶蛋白多态性、B组醇溶蛋白多态性和SGP多态性都非常丰富,与青藏高原是栽培青稞的多样性分布中心的观点一致。 青藏高原栽培青稞的SSR标记、单体醇溶蛋白、B组醇溶蛋白和SGP多态性表现出很大差异。SSR标记覆盖了整个基因组,多态性非常高。单体蛋白、B组醇溶蛋白、SGP蛋白是育种中非常关注的性状,他们只是代表基因组中的某一区域或位点,多态性相对较低。但单体蛋白多态性很高,84份材料中检测出43条不同蛋白带,75种不同的组合带谱。SSR标记技术和单体蛋白技术都是遗传多样性研究的有力工具,但单体蛋白技术不仅多态性高,而且经济、操作简便,是种质鉴定的理想方法。 对不同标记的多态性材料数据进行聚类,聚类图能为我们提供各材料间的遗传相似信息,为材料选择提供参考。但材料聚类与材料来源的地理区域的相关性表现不一致。SSR聚类和B组醇溶蛋白聚类与材料的来源地无相关性,而单体醇溶蛋白和SGP聚类与材料来源地有一定相关性,即西藏群体和四川群体分别有集中类群,这可能是人为选择的附加效应。 不同来源的群体材料的遗传多样性不同,具有区域特异稀有基因,加强不同地区间资源的交换和配合使用,有利于增加群体遗传多样性和新品种培育。 青藏高原栽培青稞的麦芽浸提性状、淀粉性状、病虫及裸粒等重要农艺性状控制位点存在丰富的变异,遗传基础宽广,可能蕴藏着多种不同的等位基因,是研究重要性状遗传特性、基因资源挖掘和遗传育种的宝贵资源库。 Hulless barley, due to its favorable attributes such as high feed value, good human nutrition,rich dietary fiber and ease processing, attracts people,s attention . Hulless barley plays a very important role in Tibetan life, used as essential food crop, main animal feed and important fuel. In addition to tsampa (roasted barley flour), a main food for Tibetan, hulless barley is also made into cake, soup, porridge, recent naked barley liquor and cornmeal. Qinghai-Tibet Plateau is one of a few areas which plant naked barley widely in the world and also has a long growing history. Genetic diversity of the cultivated hulless barley in this region , however, has not been documented. The study of genetic diversity existing within this population is of particular interest in germplasm identification, preservation, and new cultivar development. This study analyzed the genetic diversity of the cultivated naked barley from Qinghai-Tibet plateau through the study of SSR marker loci and monomeric prolamins, B-horden and starch granule proteins. SSRs are present abundantly in genomes of higher organisms and have become a popular marker system in plant studies. SSRs offer a number of advantages, such as the high level of polymorphisms, locus specificity, co-dominance, reproducibility, ease of use through PCRand random distribution throughout the genome. In barley, several hundred SSRs have been developed and genetically mapped and can therefore be selected from specific genomic regions. The genetic diversity of 64 cultivated naked barley from Tibet and Sichuan was studied with 30 SSRs of known map location.Among the selected SSR markers, PCR products of 5 SSR markers were not obtained and 3 SSR marker loci were monomeric. A total of 132 alleles were identified at 22 polyomeric SSR loci. The number of alleles per locus ranged from 2 to 15, with an average of 6.0. The polymorphism information content values for the SSRs ranged from 0.08 to 0.94, with an average of 0.65. 13 SSR markers with the PIC value >0.6 have been selected for discrimination of Qinghai-Tibet naked barley genotypews. A finger Print map was developed through 7 SSR markers with the high PIC value. It could be used as an efficient tool for gene discovery and identification of gernplasm. Hordeins, the main storage proteins of the barley seed, are composed of momomeric and polymeric prolamins and divided into -A, B, C and D groups in order of decreasing electrophoretic mobility. Hordeins show high inter-genotypic variation and have been extensively used as markers for cultivar identification and analyzing the genetic diversity. This study analyzed the genetic diversity of B-hordein in 72 naked barley from Qinqhai-Tibet Plateau. Extensive diversity was observed. A total of 15 different bands and 30 distinct patterns were found. Jaccard's coefficient of similarity was calculated, and the accessions were divided into three main groups by cluster analysis using UPGMA. Differentiation among the populations from different collecting regions based on the polymorphism of B-hordein was investigated. Monomeric prolamins show high inter-genotypic variation and have been used as molecular markers for cultivar identification, analyzing the genetic diversity in collections and investigating the evolution processes and structure of populations However, the cultivated hulless accessions from Qinghai-Tibet Pateau in China have never been examined with respect to monomeric prolamins. This study analyzed the genetic diversity of monomeric prolamins (protein fraction corresponding to wheat gliadins) using the Acid -PAGE technique in eighty-four cultivated hulless barley from Qinqhai-Tibet Plateau in China. Extensive diversity was observed. A total of 43 different bands were found, of which 21 different bands were in the region of ω group, 8 in the region of γ, 8 in the region of β, and 6 in the region of α group. Among the 86 accessions, 75 distinct patterns were identified. The number of bands ranged from 6 to 16, depending on the variety. Jaccard’s coefficient of similarity was calculated, and the lines were grouped by cluster analysis using UPGMA. A dendrogram was obtained from the analysis of the groups and five main clusters were identified. No relationship between the distribution in the dendrogram and growth habits and origins of the cultivars could be detected. Starch is the major constituent of the cereal endosperm, comprising approximately 65% of the dry weight of the mature wheat grain. The starch formed in all organs of plants is packaged into starch granules, which vary widely between species and cultivars in size and shape. Wheat endosperm starch granules contain about corresponding to the main biosynthase of starch. This report firstly dealed with intraspecific variation of the major SGPs in cultivated naked barley from Qinghai-Tibet plateau. A total of 10 major SGPs were observed in the range of 40KD-100KD and 16 types of patterns were found. Based on the variation of SGPs, accessions studied were classified into 3 groups. A geographical cline of electrophoregram was observed. In addition, significance test of the difference of starch content among groups and types of patterns were done, and the results indicated those SGPs could be related to the content of starch. Diagram obtained through cluster analysis exhibited a structuration of diversity and genetic relationship among cultivated hulless accessions. In breeding program, parents with genetically distant relationship for hybridization will increase genetic diversity of progenies. In conclusion, cultivated naked barley from Qinghai-Tibet Plateau in China presents a high variability with respect to monomeric prolamins,SSR markers , B- hordeins and SGPs. The result of this study supports Qinghai-Tibet Plateau is the center of cultivated hulless barley and the cultivated naked barley is considered to be a gene pool with large diversity and could be applied to breeding for cereal.
